3. Fun30 nucleosome remodeller regulates white-to-opaque switching in Candida albicans

Abstract

Candida albicans ( C. albicans) is an opportunistic pathogen in humans and possesses a white-opaque heritable switching system. Wor1 is a master regulator of white-opaque switching and is essential for opaque cell formation in C. albicans. However, the regulatory network of Wor1 in white-opaque switching is still vague. In this study, we obtain a series of Wor1-interacting proteins using LexA-Wor1 as bait. Among these proteins, function unknown now 30 (Fun30) interacts with Wor1 in vitro and in vivo. Fun30 expression is upregulated in opaque cells at the transcriptional and protein levels. Loss of FUN30 attenuates white-to-opaque switching, while ectopic expression of FUN30 significantly increases white-to-opaque switching in an ATPase activity-dependent manner. Furthermore, FUN30 upregulation is dependent on CO 2; loss of FLO8, a key CO 2-sensing transcriptional regulator, abolishes FUN30 upregulation. Interestingly, deletion of FUN30 affects the WOR1 expression regulation feedback loop. Thus, our results indicate that the chromatin remodeller Fun30 interacts with Wor1 and is required for WOR1 expression and opaque cell formation.

Abstract

This paper presents a novel three-coil reconnection electromagnetic launcher (TCREL). Analytical calculations and simulation results indicated that not only does this model possess a higher structural strength, but also the launching efficiency is improved effectively compared with that of the traditional reconnection electromagnetic launcher (REL). In addition, take the TCREL model as an independent module; a toroidal REL (TREL) model can be obtained after several modules are uniformly placed and fixed around the circumference. The TREL model can reduce the magnetic flux leakage among driving coils, and further improves the launching efficiency. To verify the conceptual feasibility, two-stage TCREL and REL models were built and tested. The experiments demonstrate that the TCREL model possesses better launching performance than does the REL model; the former can obtain the desired launching efficiency of 15.818% but the latter only 8.248% in the contrast experiment. The experimental results are also in accordance with the simulation results.

Abstract

A streamlined approach toward the rapid fabrication of streptavidin-biotin-based protein microarrays was investigated. First, using our engineered versatile plasmid (pBADcM-tBirA) and an optimal coexpression strategy for biotin ligase and biotin acceptor peptide (BAP) chimeric recombinant protein, an autogeneration system for biotinylated probes was developed. This system permitted an advantageous biotinylation of BAP chimeric recombinant proteins, providing a strategy for the high-throughput synthesis of biotinylated probes. Then, to bypass the conventional rate-limiting steps, we employed an on-chip purification process to immobilize the biotinylated probes with high-throughput recombinant lysates. The integration of the autogeneration of probes and on-chip purification not only contributed to the effective and reliable fabrication of the protein microarray, but also enabled simplification of the process and an automated throughput format. This labor- and cost-effective approach may facilitate the use of protein microarrays for diagnosis, pharmacology, proteomics, and other laboratory initiatives.

18. Bistable expression of WOR1, a master regulator of white--opaque switching in Candida albicans

Abstract

Candida albicans, a commensal organism and a pathogen of humans, can switch stochastically between a white phase and an opaque phase without an intermediate phase. The white and opaque phases have distinct cell shapes and gene expression programs. Once switched, each phase is stable for many cell divisions. White--opaque switching is under a1-[alpha]2 repression and therefore only happens in a or [alpha] cells. Mechanisms that control the switching are unknown. Here, we identify Wor1 (white--opaque regulator 1) as a master regulator of white--opaque switching. The deletion of WOR1 blocks opaque cell formation. The ectopic expression of WOR1 converts all cells to stable opaque cells in a or cells. In addition, the ectopic expression of WOR1 in a/[alpha] cells is sufficient to induce opaque cell formation. Importantly, WOR1 expression displays an all-or-none pattern. It is undetectable in white cells, and it is highly expressed in opaque cells. The ectopic expression of Wor1 induces the transcription of WOR1 from the WOR1 locus, which correlates with the switch to opaque phase. We present genetic evidence for feedback regulation of WOR1 transcription. 19. Fast millimeter-wave three-dimensional holographic reconstruction based on target accurate distance.

Abstract

In millimeter-wave imaging systems, the inversion efficiency of both the narrowband and broadband systems is affected by the target's distribution distance. The mismatched distances cause inefficiency of the system, and the recovered images suffer from displacement blur. We analyze in detail the reasons for this phenomenon and propose a simple method to overcome it. Minimizing the sharpness metric of images is a key strategy to solve this problem. Using this method, the exact range information of the target can be obtained from the minimum sharpness of the unknown object, and then, the region of interest of the inversion can be located. Both synthetic and full-wave simulation data demonstrate the superiority of the method. Finally, the efficiency of the method is verified by the in-house developed test system. 50. Regulation of cervical cancer via G15-mediated inhibition of G protein-coupled estrogen receptor.

Abstract

Cervical cancer is among the most common gynecological malignancies. G protein-coupled estrogen receptor (GPER) is involved in the development of various tumors; however, its role in cervical cancer remains unclear. We investigated whether G15, an inhibitor of GPER, can regulate its expression and affect cervical cancer progression. We examined the biological behaviors of G15-treated SiHa and HeLa cells using Cell Counting Kit-8, monoclonal proliferation, plate scratching, and Transwell invasion experiments. Western blotting was used to detect the expression of GPER, E-cadherin, N-cadherin, vimentin, Bcl-2, Bax, phosphatidylinositol-3-kinase (PI3K)/AKT, and programmed death ligand 1 (PD-L1). The expression of GPER, E-cadherin, vimentin, and PD-L1 in cervical cancer and adjacent tissues was detected using immunohistochemistry. The correlation between GPER expression and clinicopathological characteristics was analyzed. The expression of GPER in cervical cancer tissues was significantly higher than that in paracancerous tissues, and it was detected in the membrane and cytoplasm of SiHa and HeLa cells. The proliferation, migration, and invasion abilities of SiHa and HeLa cells were reduced after G15 treatment. The G15-treated groups exhibited higher expression of E-cadherin and Bax and lower expression of N-cadherin, vimentin, Bcl-2, GPER, p-PI3K, p-AKT, and PD-L1 than the control group. The expression of E-cadherin was lower and that of vimentin was higher in cancer tissues than in paracancerous tissues; PD-L1 was highly expressed in tumor and stromal cells in cancer tissues but not in paracancerous tissues. G15 functions by regulating the GPER/PI3K/AKT/PD-L1 signaling pathway and may serve as a new immunotherapy for treating patients with cervical cancer., (Copyright © 2024 Wolters Kluwer Health, Inc.
