3. Bridging the Gap Between Tolerogenic Dendritic Cells In Vitro and In Vivo: Analysis of Siglec Genes and Pathways Associated with Immune Modulation and Evasion.

Abstract

Background/Objectives: Dendritic cells (DCs) are master regulators of the adaptive immune response. Inflammatory DCs (inflamDCs) can prime inflammatory T cells in, for instance, cancer and infection. In contrast, tolerogenic DCs (tolDCs) can suppress the immune system through a plethora of regulatory mechanisms in the context of autoimmunity. We successfully generated tolDCs in vitro to durably restore immune tolerance to an islet autoantigen in type 1 diabetes patients in a clinical trial. However, cancers can induce inhibitory DCs in vivo that impair anti-tumor immunity through Siglec signaling. Methods: To connect in vivo and in vitro tolDC properties, we tested whether tolDCs generated in vitro may also employ the Siglec pathway to regulate autoimmunity by comparing the transcriptomes and protein expression of immature and mature inflamDCs and tolDCs, generated from monocytes. Results: Both immature DC types expressed most Siglec genes. The expression of these genes declined significantly in mature inflamDCs compared to mature tolDCs. Surface expression of Siglec proteins by DCs followed the same pattern. The majority of genes involved in the different Siglec pathways were differentially expressed by mature tolDCs, as opposed to inflamDCs, and in inhibitory pathways in particular. Conclusions: Our results show that tolDCs generated in vitro mimic tumor-resident inhibitory DCs in vivo regarding Siglec expression. [ABSTRACT FROM AUTHOR]

Abstract

Increased immune–inflammatory activation has been repeatedly linked to etiopathogenesis and the progression of both major depressive disorder (MDD) and bipolar depression (BD). We explore the role of soluble intercellular cell adhesion molecule-1 (sICAM-1) and soluble vascular cell adhesion molecule-1 (sVCAM-1) in diagnostic differentiation and disorder progression in patients with MDD and BD. Serum levels of sICAM-1 and sVCAM-1 were measured in 137 patients (MDD = 93 and BD = 44) and compared with 73 healthy controls. The severity of psychopathology was assessed using the Hamilton Depression Rating Scale and Clinical Global Impression Scale. After adjustment for multiple confounders, we noticed significant downregulation of sVCAM-1 and upregulation of sICAM-1 levels in both patient groups. Decreased sVCAM-1 levels were detected in patients with acute episodes of BD when compared to MDD. Immune mediators were related to indicators of progression in both mood disorders. They also followed different post-treatment normalization patterns in MDD and BD and in relation to the stage of each disorder. Adhesion molecules could potentially be useful in discriminating between patients with MDD and BD and determining the possible progression of the disorders. 10. Defining Human Regulatory T Cells beyond FOXP3: The Need to Combine Phenotype with Function.

Abstract

Regulatory T cells (Tregs) are essential to maintain immune homeostasis by promoting self-tolerance. Reduced Treg numbers or functionality can lead to a loss of tolerance, increasing the risk of developing autoimmune diseases. An overwhelming variety of human Tregs has been described, based on either specific phenotype, tissue compartment, or pathological condition, yet the bulk of the literature only addresses CD25-positive and CD127-negative cells, coined by naturally occurring Tregs (nTregs), most of which express the transcription factor Forkhead box protein 3 (FOXP3). While the discovery of FOXP3 was seminal to understanding the origin and biology of nTregs, there is evidence in humans that not all T cells expressing FOXP3 are regulatory, and that not all Tregs express FOXP3. Namely, the activation of human T cells induces the transient expression of FOXP3, irrespective of whether they are regulatory or inflammatory effectors, while some induced T cells that may be broadly defined as Tregs (e.g., Tr1 cells) typically lack demethylation and do not express FOXP3. Furthermore, it is unknown whether and how many nTregs exist without FOXP3 expression. Several other candidate regulatory molecules, such as GITR, Lag-3, GARP, GPA33, Helios, and Neuropilin, have been identified but subsequently discarded as Treg-specific markers. Multiparametric analyses have uncovered a plethora of Treg phenotypes, and neither single markers nor combinations thereof can define all and only Tregs. To date, only the functional capacity to inhibit immune responses defines a Treg and distinguishes Tregs from inflammatory T cells (Teffs) in humans. This review revisits current knowledge of the Treg universe with respect to their heterogeneity in phenotype and function. We propose that it is unavoidable to characterize human Tregs by their phenotype in combination with their function, since phenotype alone does not unambiguously define Tregs. There is an unmet need to align the expression of specific markers or combinations thereof with a particular suppressive function to coin functional Treg entities and categorize Treg diversity. Abstract

Analysis of the acylcarnitines' (ACs) is the mainstay for screening for fatty acid oxidation disorders (FAOD). Data about the ACs profile in the dried blood spot samples of healthy newborns in Serbia are not at disposal. Therefore, we determined the ACs levels and established the cut-offs. Between August 2018 and August 2019 a total of 1771 samples had been analysed. Cut-offs, established using a non-parametric approach, were verified in comparison with the worldwide target ranges and the data for several Caucasian populations. The majority of ACs had comparable distribution in Serbian and the worldwide population. In case of discrepancy, the individual alterations had a frequency of less than 10%. Seventeen out of 25 established cutoffs were in the worldwide target range. Reliability of the cut-offs positioning out of the target ranges is not jeopardized, since alterations are negligible or similar findings were reported for other Caucasian populations. The established and verified set of cut-offs can be used in the future screening for carnitine uptake/transport defect, medium-chain acyl-CoA dehydrogenase deficiency, very long-chain acyl-CoA dehydrogenase deficiency, long-chain L-3 hydroxyacyl-CoA dehydrogenase deficiency, trifunctional protein deficiency, carnitine palmitoyltransferase deficiency Ia and II, as well as carnitine:acylcarnitine translocase deficiency. 23. Philosophieren mit Kindern: Projektbericht

Abstract

Im Rahmen dieses Projektes haben vier Studierende der Sozialen Arbeit an der Hochschule Luzern ihre Idee eines Lehrmittels zum Philosophieren mit Kindern der ersten und zweiten Primarschulklasse umgesetzt; entstanden sind einerseits Anleitungen für nicht philosophiedidaktisch ausgebildete Lehrpersonen, andererseits auch für Bezugspersonen zum Philosophieren zu Hause. Ebenfalls erarbeitet wurde das Lehrmittel, bestehend aus situativ verwendbaren Sequenzen. Die erarbeiteten Produkte wurden der Auftraggeberin, einer Lehrperson der Volksschule St. Karli, zum regelmässigen Gebrauch übergeben. Das Lehrmittel verfügt über Anleitungen zu einer prozessorientierten und ergebnisoffenen, philosophischen Gesprächsführung. Angelehnt an Wissen aus der Philosophie sowie bereits bestehenden Lehrmitteln entstand ein Produkt, welches professionelles, theoretisches Wissen aus der Sozialen Arbeit mit neuen, kritisch reflektierten pädagogischen Haltungen verbindet, um die Förderung der sozialen, emotionalen, personalen und metakognitiven Fähigkeiten von Schüler:innen zu fördern. Abstract

Here we explore the contemporary practices of female participation in the music scenes in Serbia, and to an extent, in the Balkans. This research identified and described the possibilities, gendered constraints, and acts of transgressions that together weave a complex dynamic of female participation in popular music, in relation to the changing field of dominant gender ideologies in Serbia and the surrounding region. This work provides a critical analysis of gender issues in music-making and performing and of the topic of gendered labour in music, by relying on multiple case studies grounded in local contexts. We consider the mores and demands of the music market and everyday culture, their link to personal experiences, and the reach of the social institutions regarding music. Either as role models or cherished leaders, female musicians employ different tactics to fight stereotypes, strengthen communities, and ensure female participation. This work maps the strategies and tools they have been putting in place in order to sustain their audience, income, and presence to the best possible extent. Bearing in mind the imperative of transforming a standard way of working, communicating with audiences, and maintaining earning potential, this text singles out practices that could be recognised as (female) leadership in contemporary circumstances. We also consider a wider spectrum of roles that female musicians and music professionals have taken on or were awarded within their professional circles, local scenes, communities, or wider society.

Abstract

Ací explorem les pràctiques contemporànies de la participació femenina en l’escena musical de Sèrbia i, en certa manera, dels Balcans. Aquesta investigació va identificar i descobrir les possibilitats, les restriccions de gènere i els actes de transgressió que, en conjunt, teixien una dinàmica complexa de participació femenina en la música popular, en relació amb el camp canviant de les ideologies de gènere dominants a Sèrbia i la regió circumdant. Aquest treball proporciona una anàlisi crítica de les qüestions de gènere en la creació i execució de música i del tema del treball de gènere en la música, basant-se en casos connectats a contextos locals. Per a fer-ho, considerem els costums i les demandes del mercat musical i la cultura quotidiana, el seu vincle amb les experiències personals i l’envergadura de les institucions socials al voltant de la música. Siga com a models a seguir o com a líders apreciades, les dones músiques empren diferents tàctiques per a combatre els estereotips, enfortir les comunitats i garantir la participació femenina. Aquest treball recull les estratègies i eines que han estat implementant per a mantenir la seua audiència, els ingressos i la presència de la millor manera possible. Tenint en compte l’imperatiu de transformar una manera estàndard de treballar, comunicar-se amb les audiències i mantenir el potencial d’ingressos, aquest text destaca pràctiques que es podrien reconéixer com a lideratge (lideratge femení) en les circumstàncies contemporànies. Contains fulltext : 220591.pdf (Publisher’s version ) (Open Access) BACKGROUND AIMS: Recent technical and clinical advances with cell-based therapies (CBTs) hold great promise in the treatment of patients with rare diseases and those with high unmet medical need. Currently the majority of CBTs are developed and manufactured in specialized academic facilities. Due to small scale, unique characteristics and specific supply chain, CBT manufacturing is considered costly compared to more conventional medicinal products. As a result, biomedical researchers and clinicians are increasingly faced with cost considerations in CBT development. The objective of this research was to develop a costing framework and methodology for academic and other small-scale facilities that manufacture cell-based therapies. METHODS: We conducted an international multi-center costing study in four facilities in Europe using eight CBTs as case studies. This study includes costs from cell or tissue procurement to release of final product for clinical use. First, via interviews with research scientists, clinicians, biomedical scientists, pharmacists and technicians, we designed a high-level costing framework. Next, we developed a more detailed uniform methodology to allocate cost items. Costs were divided into steps (tissue procurement, manufacturing and fill-finish). The steps were each subdivided into cost categories (materials, equipment, personnel and facility), and each category was broken down into facility running (fixed) costs and operational (variable) costs. The methodology was tested via the case studies and validated in developer interviews. Costs are expressed in 2018 euros (€). RESULTS: The framework and methodology were applicable across facilities and proved sensitive to differences in product and facility characteristics. Case study cost estimates ranged between €23 033 and €190 799 Euros per batch, with batch yield varying between 1 and 88 doses. The cost estimations revealed hidden costs to developers and provided insights into cost drivers to help design manufacturing best practices. CONCLUSIONS: This framework and methodology provide step-by-step guidance to estimate manufacturing costs specifically for cell-based therapies manufactured in academic and other small-scale enterprises. Abstract

Induction of antigen-specific regulatory T cells (Tregs) in vivo is the holy grail of current immune-regulating therapies in autoimmune diseases, such as type 1 diabetes. Tolerogenic dendritic cells (tolDCs) generated from monocytes by a combined treatment with vitamin D and dexamethasone (marked by CD52hi and CD86lo expression) induce antigen-specific Tregs. We evaluated the phenotypes of these Tregs using high-dimensional mass cytometry to identify a surface-based T cell signature of tolerogenic modulation. Naïve CD4+ T cells were stimulated with tolDCs or mature inflammatory DCs pulsed with proinsulin peptide, after which the suppressive capacity, cytokine production and phenotype of stimulated T cells were analysed. TolDCs induced suppressive T cell lines that were dominated by a naïve phenotype (CD45RA+CCR7+). These naïve T cells, however, did not show suppressive capacity, but were arrested in their naïve status. T cell cultures stimulated by tolDC further contained memory-like (CD45RA-CCR7-) T cells expressing regulatory markers Lag-3, CD161 and ICOS. T cells expressing CD25lo or CD25hi were most prominent and suppressed CD4+ proliferation, while CD25hi Tregs also effectively supressed effector CD8+ T cells. We conclude that tolDCs induce antigen-specific Tregs with various phenotypes. This extends our earlier findings pointing to a functionally diverse pool of antigen-induced and specific Tregs and provides the basis for immune-monitoring in clinical trials with tolDC., Comp Graphics & Visualisation
