AIM- The aim of the present investigation is to evaluate Plumbago zeylanica L. (Plumbaginaceae) for evaluation of memory enhancing activity. MATERIAL & METHODS- All the plant materials were dried under shade and subjected to coarse powder for extraction process. Accurately weighed quantity of roots of Plumbago zeylanica Linn. extracted using petroleum ether, chloroform, methanol, butanol and finally water by soxhlet apparatus for 72 h. Qualitative chemical tests of chloroform, methanol, butanol and water extracts were subjected to various chemical tests to detect various Phytoconstituents. Briefly, 6.0 mg ß-carotene was dissolved in 10 ml of chloroform, and then 1 ml of solution pipette to glass filled of 20 mg linoleic acid. 5 ml of mixture then pipette to reaction tube filled of extract in a range of concentration, mixed homogenously. Briefly, 1 mL of 0.3 mM of DPPH solution was added to 1 mL each of the test solutions, and was incubated in the dark at room temperature for 30 min. Escape latency (EL) was recorded 120 min after drug administration from 11th day to 14th day. On 15th day, time spent in target quadrant (TSTQ) was noted 120 min after the drug administration. In case of animals administered with physostigmine, EL and TSTQ was noted after 30 min of drug administration. 0.4 ml of brain homogenate was added into a test tube containing 2.6 ml of phosphate buffer. 5,5-dithiobis-2-nitrobenzoic acid reagent (0.1 ml) was added to the above mixture and absorbance was noted at 412 nm. RESULTS- The preliminary phytochemical analysis revealed that different active constituent present in different extracts such as carbohydrates, proteins, amino acids, fat, oils, steroids, terpenoids, glycosides, alkaloids, tannins and other phenolics compounds. Vitamin E as standard was used in this assay and 84% inhibition was found to be at 30 minutes. Chloroform extract also showed 70% inhibition at 30 minutes which was reduced to 45% at the time of 120 minutes. Hydroxyl radical scavenging ability calculated as IC50 reveals that chloroform; methanol, butanol and water extracts have IC50 values of 47 μg/mL, 100μg/mL, 200 μg/mL and 150μg/mL, respectively. Objectives: The objective of this study is to extract Phytochemical constituent from Cinnamon Bark and to design and develop the transdermal patch of the herbal drug along with modern medicine Glibenclamide using solvent casting method to reduce the dose required to obtain same pharmacological the effect, also to reduce the toxicity of the drug. Methods: The Glibenclamide Transdermal patches were formulated by using the solvent casting method. The physical and chemical similarity of the medication and the base of patches were studied by Infrared Spectroscopy (FTIR). The outcomes recommended no physical and chemical properties incongruence between the medication and the patch base. The formulated transdermal patches were assessed for the weight difference, fatness, folding endurance, wetness, moisture captivation, ex vivo drug release, ex vivo drug absorption. Results: The diffusion examines were performed by utilizing the Franz diffusion cell and everted gut sac method. F5 shows maximum bioenhancing action compared to all other patches which contains Glibenclamide along with the Ethanolic extract of the Cinnamon bark.

In recent days we have been using many devices equipped with LED in one or other way, as display or light sources. These sources have large contribution of UV emission in their total emission of light. This adds the amount of UV radiations and its effect which we have already faced during the exposure of sun. To protect not only the human body, skin and eyesight, but also some essential surfaces of exteriors, plastic roof, we need some protecting film/materials that can absorb UV part from light radiation. Already aware of the harm that UV-A and UV-B rays from sun cause to the biotic components of the ecosystem, researchers and scientists have started seriously working in developing some films using polymer nanocomposites or nanomaterials which have improved tranceparancy, flexibility, durability features. Mainly concentrating on UV shielding applications, the article talks about various nanoparticles prominently used in the said purpose, namely CeO2, TiO2, ZnO, MnO2 and graphene oxide. Along with their respective synthesis methods, particle size, band gap, other experimental results have been highlighted. Green synthesis of ZnO nanoparticles with the assistance of fungus and bacteria also finds mention which is significant in reducing our dependence on chemicals. Hexagon shaped pristine and Al doped ZnO nanodisks (NDs) with exposed ± [0001] polar facets successfully synthesized with absolute ethanol: water system as dual solvent mixture show exceptionally good absorbance of 99% in UV range upto 390 nm. It also emphasizes upon the importance of the synergistic effect of the incorporated nanoparticles & the matrix material used and the conversion of cellulose acetate matrix into cellulose acetate naphthoate in order to enhance its service life in the commercial anti-UV applications.

The Child is always precious so their health too. The maintenance of child health is desired and positively taken so every society has goal of improved level of child health status. Neonatal care begins from premarital age which continues from conception, pregnancy, and childbirth and to the childhood through proper care during each stage. When primary neonatal care is inadequate, then it leads to unavoidable and unacceptable high neonatal morbidity and mortality occurs. Among the concerned care thermal protection of newborn is also one of them since hypothermia is considered silent killer among neonate. To make aware regarding this condition with the aim to find the effectiveness of video assisted teaching program on thermal protection of newborns among mothers this study was conducted. Researcher used an evaluatory approach with a pre experimental, one group pre-test post test design in the study. A self prepared knowledge questionnaire was used to assess the knowledge among mothers on thermal protection of newborns followed by video assisted teaching program and again followed by same questionnaire for post test. α-Synuclein (α-Syn) aggregation and amyloid formation is directly linked with Parkinson's disease pathogenesis. However, the early events involved in this process remain unclear. Here, using the in vitro reconstitution and cellular model, we show that liquid-liquid phase separation of α-Syn precedes its aggregation. In particular, in vitro generated α-Syn liquid-like droplets eventually undergo a liquid-to-solid transition and form an amyloid hydrogel that contains oligomers and fibrillar species. Factors known to aggravate α-Syn aggregation, such as low pH, phosphomimetic substitution and familial Parkinson's disease mutations, also promote α-Syn liquid-liquid phase separation and its subsequent maturation. We further demonstrate α-Syn liquid-droplet formation in cells. These cellular α-Syn droplets eventually transform into perinuclear aggresomes, the process regulated by microtubules. This work provides detailed insights into the phase-separation behaviour of natively unstructured α-Syn and its conversion to a disease-associated aggregated state, which is highly relevant in Parkinson's disease pathogenesis.

Discovery of deafness genes and elucidating their functions have substantially contributed to our understanding of hearing physiology and its pathologies. Here we report on DNA variants in MINAR2 , encoding membrane integral NOTCH2-associated receptor 2, in four families underlying autosomal recessive nonsyndromic deafness. Neurologic evaluation of affected individuals at ages ranging from 4 to 80 y old does not show additional abnormalities. MINAR2 is a recently annotated gene with limited functional understanding. We detected three MINAR2 variants, c.144G > A (p.Trp48*), c.412_419delCGGTTTTG (p.Arg138Valfs*10), and c.393G > T, in 13 individuals with congenital- or prelingual-onset severe-to-profound sensorineural hearing loss (HL). The c.393G > T variant is shown to disrupt a splice donor site. We show that Minar2 is expressed in the mouse inner ear, with the protein localizing mainly in the hair cells, spiral ganglia, the spiral limbus, and the stria vascularis. Mice with loss of function of the Minar2 protein ( Minar2 tm1b/tm1b ) present with rapidly progressive sensorineural HL associated with a reduction in outer hair cell stereocilia in the shortest row and degeneration of hair cells at a later age. We conclude that MINAR2 is essential for hearing in humans and mice and its disruption leads to sensorineural HL. Progressive HL observed in mice and in some affected individuals and as well as relative preservation of hair cells provides an opportunity to interfere with HL using genetic therapies.

The mechanism of how the aggregation of the same precursor protein results in the formation of fibril polymorphs remains elusive. Here, we demonstrate the structure-function relationship of two polymorphs, pre-matured fibrils (PMFs) and helix-matured fibrils (HMFs), based on α-Syn aggregation intermediates. These polymorphs display the structural differences as demonstrated by solid-state NMR and mass spectrometry studies and also possess different cellular activities such as seeding, internalization, and cell-to-cell transfer of aggregates. HMFs, with a compact core structure, exhibit low seeding potency but readily internalize and transfer from one cell to another. The less structured PMFs lack transcellular transfer ability but induce abundant α-Syn pathology and trigger the formation of aggresomes in cells. Overall, the study highlights that the conformational heterogeneity in the aggregation pathway may lead to fibril polymorphs with distinct prion-like behavior.

High temperature requirement protease A2 (HtrA2) is a mitochondrial serine protease that demonstrates multifaceted roles including protein quality control and proapoptotic properties in humans, making it a potential therapeutic target. Current literature suggests involvement of flexible regulatory loops in governing the allosteric propagation within the trimeric HtrA2 ensemble. Here, we have identified three important residues - R147, P148 (L3 loop) and F131 (LD loop) surrounding the catalytic-site that play crucial roles in stabilizing HtrA2 active conformation during its multimodal activation. Although mutagenesis of these residues does not affect the structural integrity, it renders the protease inactive by affecting the regulatory inter-subunit PDZ-protease crosstalk. This is further emphasized by the inactivity observed during N-terminal mediated activation of the HtrA2 loop mutants via BIR2 domain of the antiapoptotic protein XIAP. Overall, our results demonstrate the importance of L3 loop dynamics in mediating the inter-molecular allostery via R147-P148 residues. Understanding the on-off switch that regulates HtrA2 activation might help in designing HtrA2 modulators for therapeutic applications.

In this paper the antipyretic effect of the Hydroalcoholic extract of leaves of Colocasia esculenta against Brewer yeast induced pyrexia model in Wistar rats of either sex was investigated. Colocasia esculenta (Apiaceae) is a tropical perennial plant. It contain flavonoids, alkaloids, apigenin, anthocyanin, carbohydrates, fiber, minerals, protein, fat, calcium and iron, Fever induced in the animal (rats) by the injection 15%w/v of brewer’s yeast suspension (10mg/kg according to body weight of rats) subcutaneously in the back below the nape of the neck. . After 18 hr. of Brewer’s yeast injection and rise in rectal temperature was recorded. The temperature of animal was recorded at 0, 1, 2, 3 and 4hr after drug administration. Paracetamol (100 mg/kg p. o.) was used as standard drug. The group received Hydroalcoholic extract 200mg/kg and 400mg/kg showed significant decrease in rectal temperature respectively as compared with the group received standard drug. All the values are expressed as mean ± standard deviation and analysed for ANOVA. Differences between controls, standard ant test groups were considered significant at P < 0.001 levels. . The Hydroalcoholic extract of Colocasia esculenta leaves (200mg/kg and 400mg/kg) possesses dose dependent, significant antipyretic activity against Brewer yeast induced pyrexia. Dy3+ -doped CaAl12 O19 phosphors were synthesized utilizing a combustion method. Crystal structure and morphological examinations were performed respectively using X-ray diffraction (XRD) and scanning electron microscopy (SEM) techniques to identify the phase and morphology of the synthesized samples. Fourier transform infrared spectroscopy (FTIR) estimations were carried out using the KBr method. Photoluminescence properties (excitation and emission) were recorded at room temperature. CaAl12 O19 :Dy3+ phosphor showed two emission peaks respectively under a 350-nm excitation wavelength, centered at 477 nm and 573 nm. Dipole-dipole interaction via nonradiative energy shifting has been considered as the major cause of concentration quenching when Dy3+ concentration was more than 3 mol%. The CIE chromaticity coordinates positioned at (0.3185, 0.3580) for the CaAl12 O19 :0.03Dy3+ phosphor had a correlated color temperature (CCT) of 6057 K, which is situated in the cool white area. Existing results point out that the CaAl12 O19 :0.03Dy3+ phosphor could be a favorable candidate for use in white light-emitting diodes (WLEDs).

In this study, pristine and tin-doped vanadium pentoxide nanoparticles were synthesized via hydrothermal method and characterized using X-ray diffraction (XRD), Raman, transmission electron microscopy (TEM) and photoluminescence spectroscopy confirmed the orthorhombic structure, single phase and chain-layered structure. Microstructural analysis revealed the morphology of V2O5 as chain-layered-like structure. The crystallite size with microstrain with the peak-broadening of doped and undoped V2O5 nanoparticles was analyzed by William–Hall (W-H) method. The effect of Sn dopant on the crystallite size, morphology and luminescence property showed that the crystallite size reduces to 53.45 nm in the presence of Sn dopant. Optical sensing study on pure and Sn-doped V2O5 at room temperature showed the maximum 79.94% sensitivity for 45 ppm ammonia.

The combinatorial space of an enzyme sequence has astronomical possibilities and exploring it with contemporary experimental techniques is arduous and often ineffective. Multi-target objectives such as concomitantly achieving improved selectivity, solubility and activity of an enzyme have narrow plausibility under approaches of restricted mutagenesis and combinatorial search. Traditional enzyme engineering approaches have a limited scope for complex optimization due to the requirement of a priori knowledge or experimental burden of screening huge protein libraries. The recent surge in high-throughput experimental methods including Next Generation Sequencing and automated screening has flooded the field of molecular biology with big-data, which requires us to re-think our concurrent approaches towards enzyme engineering. Artificial Intelligence (AI) and Machine Learning (ML) have great potential to revolutionize smart enzyme engineering without the explicit need for a complete understanding of the underlying molecular system. Here, we portray the role and position of AI techniques in the field of enzyme engineering along with their scope and limitations. In addition, we explain how the traditional approaches of directed evolution and rational design can be extended through AI tools. Recent successful examples of AI-assisted enzyme engineering projects and their deviation from traditional approaches are highlighted. A comprehensive picture of current challenges and future avenues for AI in enzyme engineering are also discussed.

Background: To study the morbi-mortality related to ileostomy or colostomy closure at tertiary care hospital Bikaner Material and Method: prospective hospital based study. 30 patients reporting to the General Surgery dept. within study duration and eligible as per inclusion criteria will be included in the study. Results: Out of 30 Cases ,(30.00%) patients had hypertension ,6(20.00%) had diabetes, 5 (16.67%) had renal dysfunction and 2(6.67%) cases had COPD. 11 (36.67%) patients developed complications post closure of ileostomy or colostomy. Medical complications accounted for a large proportion of complications (n=5), while major (n=4) and minor complications (n=2) were present. Conclusion: ileostomy is effective and feasible as a diversion procedure and has reduced morbidity and complication rates. Keywords: Ileostomy, Colostomy. Complications.

Hydrogen production through PEC provides a green and sustainable source of energy and addresses the solar intermittency problem. While considerable efforts have been made over the past several decades but till date, there is no solar water splitting materials that simultaneously fulfills the high efficiency, long-term stability and low cost. In PEC, the semiconductor materials must exhibit sufficient voltage on irradiation to split water, long-term stability against corrosion in the aqueous electrolyte and the band edge potentials at the surfaces. The charge transfer from surface of the semiconductor to the electrolyte must be facile to minimize energy loss due to kinetic overpotential and selective for the oxygen and hydrogen evolution reaction. The photocatalysts such as TiO2, WO3, CuGaSe2, InGaN, Fe2O3, Cu2O etc. are the most promising semiconductor material due to its suitable band gap and valance band structure. But, the major concern in PEC water splitting is charge separation and transportation. Various strategies such as controlling the morphology, introducing defects, constructing a heterojunction and loading a co-catalyst are used raise the efficiency of PEC process.

However, how a single protein can form different fibril polymorphs in various synucleinopathies is not known. Here, we demonstrate the structure-function relationship of two distinct α-Syn fibril polymorphs, the pre-matured fibrils (PMF) and helix-matured fibrils (HMF) based on α-Syn aggregation intermediates. Herein, we present the synthesis, characterization and optical ammonia gas sensing of pure and Sn-doped vanadium oxide (V2O5) nanoparticles prepared by the sol-gel process. The V2O5 nanoparticles were doped with Sn concentrations of 2 wt%–6 wt%, and characterized using X-ray diffraction, atomic force microscopy, UV–vis spectroscopy and photoluminescence spectroscopy which confirmed the orthorhombic crystal structure. The crystalline size was found to decrease with enhancing the doping concentrations of Sn. The lattice-strain and crystalline size with the peak-broadening of pure and Sn doped V2O5 nanoparticles were analyzed by William-Hall (W-H) method and size-strain plot. The UV–vis absorption showed a decrease in the energy band gap (3.27 eV–3.07 eV) with an increase in the Sn doping concentration. The Sn-doped V2O5 nanoparticles were used to detect ammonia gas (5–50 ppm) through photoluminescence based detection method. Interestingly, it was observed that the optical response for the 2 wt% Sn doped V2O5 nanoparticles was maximum (77.84%) towards 50 ppm ammonia compared to other V2O5nanoparticles based samples.
