During synchronous fracturing of horizontal well clusters, the induced stresses generated by multiple wells affect the formation of complex fractures. By using linear superposition and vector representation, the productivity model for the horizontal stress field between wells was constructed. The results demonstrated simultaneous fracturing can produce stronger and a wider range of stress interference variable impact on induced stress field, the most influential factor is the row spacing, accounting for 32.09% and the engineering factors with the least impact is fracture conductivity, with weight of 0.08%. The research results were used to YA gas field. After the preliminary implementation in the field, the initial production of a well in the reservoir increased by 20.48%, the decline rate slowed down by 13.16% and the output of well increased by 43.75% in the stable production period. In addition, these results might also provide an acceptable explanation for the high productivity of synchronous fracturing operations in horizontal well clusters. Generally speaking, a clear understanding of changes in induced stress fields in horizontal clusters might provide are liable foundation for the determination of optimal well spacing and a fracturing program for horizontal well clusters to develop unconventional formations. Background: Mounting evidence has demonstrated that circular RNAs (circRNAs) play indispensable roles in the progression of bladder cancer. Public database mining showed that hsa_circRNA_100146 (circRNA_100146) was highly expressed in bladder cancer. This study aimed to characterize the biological role of circRNA_100146 and clarify the underlying mechanism in bladder cancer. Methods: We evaluated the relationship between circRNA_100146 expression and clinicopathological features. Furthermore, gain- and loss-of-function studies were conducted in bladder cancer cells via transfection with gene-carrying plasmids (over-expression) or specific short hairpin RNAs (knockdown). Moreover, computational algorithms and dual-luciferase reporter assays were performed to explore the possible mechanisms of action. Additionally, in vivo xenograft experiments were performed to further analyze the effect of circRNA_100146 on tumor growth. Results: Our data showed that circRNA_100146 expression was increased in bladder cancer tissues and cell lines, and that high expression of circRNA_100146 was correlated with poor patient prognosis. Upregulation of circRNA_100146 promoted cell proliferation, migration, and invasion, and inhibited cell apoptosis, whereas knockdown of circRNA_100146 displayed opposite effects on bladder cancer cells. Notably, circRNA_100146 could combine with miR-149-5p and promote ring finger protein 2 (RNF2) expression, thereby facilitating the progression of bladder cancer. Furthermore, overexpression of RNF2 reversed the effects of circRNA_100146 knockdown on the biological behaviors of bladder cancer cells. The in vivo experiments revealed that downregulation of circRNA_100146 dramatically delayed tumor growth. Conclusion: Our findings indicate that circRNA_100146 functions as a sponge of miR-149-5p in promoting bladder cancer progression by regulating RNF2 expression and that circRNA_100146 may serve as a novel biomarker in human bladder cancer. Background: Bladder cancer (BC) is a common and deadly disease. Over the past decade, a number of genetic alterations have been reported in BC. Bladder urothelium expresses abundant urea transporter UT-B encoded by Slc14a1 gene at 18q12.3 locus, which plays an important role in preventing high concentrated urea-caused cell injury. Early genome-wide association studies (GWAS) showed that UT-B gene mutations are genetically linked to the urothelial bladder carcinoma (UBC). In this study, we examined whether Slc14a1 gene has been changed in UBC, which has never been reported. Case presentation: A 59-year-old male was admitted to a hospital with the complaint of gross hematuria for 6 days. Ultrasonography revealed a size of 2.8 × 1.7 cm mass lesion located on the rear wall and dome of the bladder. In cystoscopic examination, papillary tumoral lesions 3.0-cm in total diameter were seen on the left wall of the bladder and 2 cm to the left ureteric orifice. Transurethral resection of bladder tumor (TURBT) was performed. Histology showed high-grade non-muscle invasive UBC. Immunostaining was negative for Syn, CK7, CK20, Villin, and positive for HER2, BRCA1, GATA3. Using a fluorescence in situ hybridization (FISH), Slc14a1 gene rearrangement was identified by a pair of break-apart DNA probes. Understanding the flow processes that form gravity-flow deposits is vital for modeling and predicting sandstone bodies in the subsurface, which is of great significance for conventional and unconventional oil and gas exploration and development in lacustrine basins. This study analyses gravity-flow deposits of the Late Triassic Chang 7 oil member of the Yanchang Formation in the Ordos Basin, using a combination of well-log analysis, core observations, thin-section analysis, and laboratory measurements. The sedimentary facies, formation mechanisms, distribution patterns, and depositional models of gravity-flow deposits are investigated. Thirteen facies and eight bed-types are recognized in the gravity-flow deposits. Bed types represent deposits of sandy slides, sandy slumps, debrites, high-density turbidites, hybrid event beds, transitional flow deposits, surge-like low-density turbidites, and quasi-steady low-density turbidites. Gravity-flow deposits, particularly those caused by sediment failure, are composed of slides, slumps, debrites, high-density turbidites, hybrid event beds, transitional flow deposits, and surge-like low-density turbidites. These deposits form as isolated lenticular sand bodies, with retrogradational internal stacking patterns. In contrast, gravity-flow deposits caused by flooding rivers are composed of high-density turbidites, hybrid event beds, transitional flow deposits, and quasi-steady low-density turbidites. These deposits exhibit elongate morphologies with progradation stacking patterns internally. The collision between the North China Block and the South China Block, which occurred during the closing of the Qinling Ocean, lead to frequent volcanic eruptions and earthquakes. These activities promoted the development of gravity-flow deposits associated with sediment failure from delta fronts in both the northeastern and southwestern parts of the basin. Concurrently, extremely humid climatic conditions promoted increased fluvial drainage, leading to enhanced hyperpycnal flows into the deep-lacustrine basin depocenters in the Late Triassic. The present-day in-situ stress field has many important and practical applications during the exploration and development of hydrocarbon resources. The Ordos Basin is a significant petroliferous basin in China, within which, the Yanchang Formation serves as the most significant tight oil reservoir. However, an overview of previous studies indicates that little was known as to the present-day stress state in the Yanchang Formation. Therefore, the present-day stress field in the Yanchang Formation tight oil reservoir of the Ordos Basin is systematically analyzed in this study. The orientation of horizontal maximum principal stress (S Hmax) within the Yanchang Formation of Ordos Basin ranges between north-northeast-south-southwest-trending (NNE-SSW) and east-northeast-west-southwest-trending (ENE-WSW) based on interpretations of borehole breakouts and drilling-induced fractures from borehole imaging logs. The S Hmax orientation shows various tendencies in different wells, which may result from differential development in lithology, natural fracture and bedding plane over various regions. The estimated magnitudes of vertical stress (S v), S Hmax and horizontal minimum principal stress (S hmin) in the studied wells indicate that the normal faulting stress regime is dominant within the Yanchang Formation. In addition, the geomechanical assessments of wellbore stability and natural fracture reactivation are also performed. Natural fractures in the Yanchang Formation Chang 7 tight oil reservoir of Ordos Basin are developed striking mainly in the northeast-southwest-trending (NE-SW) and west-northwest-east-southeast-trending (WNW-ESE). Among them, those fractures with NE-SW-trending strikes are more conductive with reference to the present-day stress state, contributing more to tight oil production in the Ordos Basin. In addition, within the Yanchang Formation, if horizontal wells are drilled towards the NNE-SSW-trending to ENE-WSW-trending, they are more likely to have wellbore instability issues. "Sweet sections" in giant shale oil provinces are preferential fields that primarily support China to increase the reserves and production of continental shale oil. Based on the study of the geological conditions of shale oil in the continental basins in China, it was found that the shale stratum in major oil generation windows generally has higher degrees of oil and gas accumulation, and mostly contains oil. Hydrocarbon generation and reservoir capacities are the two key parameters for evaluating and optimizing favorable shale oil provinces. The evaluation index (volume of shale stratum multiplied by total organic carbon (TOC) multiplied by total porosity) for the giant continental shale oil provinces is also proposed. It is optimized that the Upper Triassic Chang 7 Member in the south-central Ordos Basin, Lower Cretaceous Qing 1 Member in the Gulong-Changling Sag in the Songliao Basin, Middle — Lower Permian in the Junggar Basin, Da'anzhai Member of the Ziliujing Formation of Lower Jurassic in the central and northern Sichuan Basin, and Paleogene oil-rich sag in the Bohai Bay Basin are the five giant continental shale oil provinces. The word "geological sweet sections" in continental shale oil provinces of China refers to favorable shale intervals which are relatively rich in oil, with superior physical properties, and more easily modified and developed commercially under applicable economic and technological conditions. After evaluation, there are mainly two types of "geological sweet sections" of giant continental shale oil developed onshore in China. One type of "geological sweet sections" is generally mudstone with optimal physical properties or a thin tight reservoir, to which the shale oil migrates a short distance. They are medium-to-high-mature zones with a thin sandy shale stratum in the Chang 7 Member in the Ordos Basin, mixed shale stratum in the medium-mature Lucaogou Formation in the Jimsar Sag, and multi-layered mixed Paleogene shale stratum in the Bohai Bay Basin. The other type of "geological sweet sections" is generally shale oil residing in various shale reservoir spaces. This type was developed in the Qing 1 Member in the Gulong Sag and Da'anzhai Member in the north-central Sichuan Basin. Free shale oil mainly occurs in shale, sandy-carbonate lamina, micro-lamella structure, and micro-fractures. Layers with lamina, lamination, and micro-fractures are generally shale oil "geological sweet sections." Starting from field tests and the construction of the "geological sweet sections" in giant continental shale oil provinces, the shale oil industry has been rapidly developing and will become an important supplement to domestic oil production in China. Purpose: A series of new 68Ga-labeled tracers based on [68Ga]Ga-PSMA-617 were developed to augment the tumor-to-kidney ratio and reduce the activity accumulation in bladder, ultimately minimize radiation toxicity to the urinary system.We introduced quinoline group, phenylalanine and decanoic acid into different tracers to enhance their lipophilicity, strategically limiting their metabolic pathway through the urinary system. Their binding affinity onto LNCaP cells was determined through in vitro saturation assays and competition binding assays. In vivo metabolic study, PET imaging and biodistribution experiment were performed in LNCaP tumor-bearing B-NSG male mice. The most promising tracer was selected for first-in-human study.Four radiotracers were synthesized with radiochemical purity (RCP) > 95% and molar activity in a range of 20.0-25.5 GBq/μmol. The binding affinities (Ki) of TWS01, TWS02 to PSMA were in the low nanomolar range (< 10 nM), while TWS03 and TWS04 exhibited binding affinities with Ki > 20 nM (59.42 nM for TWS03 and 37.14 nM for TWS04). All radiotracers exhibited high stability in vivo except [68Ga]Ga-TWS03. Micro PET/CT imaging and biodistribution analysis revealed that [68Ga]Ga-TWS02 enabled clear tumor visualization in PET images at 1.5 h post-injection, with higher tumor-to-kidney ratio (T/K, 0.93) and tumor-to-muscle ratio (T/M, 107.62) compared with [68Ga]Ga-PSMA-617 (T/K: 0.39, T/M: 15.01) and [68Ga]Ga-PSMA-11 (T/K: 0.15, T/M: 24.00). The Carnian Pluvial Event (CPE) induced an intense carbon-cycle disturbance and biological crisis at a global scale. The eruption of the Wrangellia Large Igneous Province (W-LIP) is considered a possible triggering mechanism; however, this cannot fully explain the significant differences in environmental perturbations across different regions. In this paper, we present detailed records of petrography, mercury geochemistry, carbon and sulfur isotopes, trace elements, and provenance data for the Triassic lacustrine facies of the Ordos Basin in the North China Plate, which provide insights into the important effects of volcanic activities, other than the W-LIP, on palaeoenvironment during the CPE interval. Massive emissions of SO 2 , CO 2 , and pyroclastic deposits resulting from these volcanic activities triggered four humid episodes, each characterized by lake-level rise, negative δ13C org excursion, a transition from mudstones to organic-rich shales facies, as well as algal bloom within a time span of 180 kyrs. The establishment of an age framework in the Ordos Basin enables comparative analyses of environmental evolution during the Carnian age across the North China Plate, the western Tethys domain, and the South China Plate. During the period of global climate perturbation driven by the W-LIP eruption, we conclude that short-term localized volcanic activities may have further induced high frequentcy environmental perturbations, which influenced biological shifts and increase organic carbon burial. Purpose: To investigate a new method and its effect on the procedure of dilating the ejaculatory duct and flushing the seminal vesicle with an F9 seminal vesicle scope in patients with chronic and recurrent seminal vesiculitis. Patients and Methods: Twenty-six patients with a diagnosis based to signs, laboratory detection, digital rectal examination, and transrectal ultrasonography were involved in present study. The patients underwent a surgical procedure of dilating the ejaculatory duct and flushing the seminal vesicles with an F9 seminal vesicle endoscope. All patients were followed for 3 months to 1 year after treatment. Results: There were significant reductions in symptoms, signs, white blood cell and red blood cell counts on microscopic examination, seminal vesicles size, improvement of inner walls echo in transrectal ultrasonography, and semen culture positive rate. Moreover, all patients showed improvement. Conclusions: The present study provides a new transurethral seminal tract endoscopic technique with seminal vesicle scope through the normal anatomic tract to treat patients with chronic seminal vesiculitis. Background: Prostate cancer often shows the over-activation of beta-catenin/t-cell factor (TCF) signaling. It remains largely unknown how the beta-catenin/TCF transcriptional machinery is tightly controlled. Methods: The ZNF433 mRNA and protein levels in the clinical tissues were examined using q-PCR, Western blot and immunohistochemistry. The phenotypes of prostate cancer cells were examined using MTT assay, Boyden chamber assay and anchorage-independent assay. The interaction between ZNF433 and beta-catenin was evaluated by immunoprecipitation. Results : In the present study, ZNF433 was upregulated in prostate cancer samples, and promoted the growth and migration of prostate cancer cells. Furthermore, ZNF433 was the binding partner of beta-catenin and activated beta-catenin/TCF signaling in prostate cancer. Moreover, ZNF433 enhanced the binding between beta-catenin and TCF4. In addition, NC043, small antagonist for beta-catenin/TCF complex, inhibited the malignant behaviors of prostate cancer cells driven by ZNF433. Conclusion: In summary, these studies demonstrate the tumor-promoting roles of ZNF433 in prostate cancer, and suggesting that ZNF433 was a potential target for the treatment. [ABSTRACT FROM AUTHOR]

The lacustrine organic‐rich shale of Triassic Chang 7 Member is an unconventional shale oil reservoir that is a major hydrocarbon target in the Ordos Basin, northern China. This study uses major, trace, and rare earth element (REE) abundance documented through a 115‐m‐thick core obtained from the western part of the Ordos Basin to assess dominant controls on temporal variations in total organic carbon in the Chang 7 shale. Our results suggest that total organic carbon trends in the Chang 7 can be directly linked with the tectonic and magmatic evolution of Qinling orogen. The Chang 73 and Chang 72 submembers of the lower part of the Chang 7 were likely deposited in association with heighted tectonic and magmatic activity of the Qinling Orogeny. Sediments derived from these submembers display light REE and trace element compositions similar to the Tianshui rhyolites suggesting deposition contemporaneous with magmatic and/or volcanic activity of the Qinling Orogeny. Higher nutrient concentrations introduced by volcanic eruptions would have stimulated primary productivity in the surface water enhancing the organic carbon flux to the lake bottom. In addition, higher rates of mountain building in the Qinling Orogeny may have been responsible for increasing subsidence rate of the southern Ordos Basin, also favourable for the establishment of anoxic bottom‐water conditions. In contrast, lower light REE and trace element abundances of the Chang 71 submember at the top of the Chang 7 suggest deposition during a tectonically and magmatic quiescent period of the Qinling Orogeny. The diminished volcanic flux could have lowered the rate of primary productivity. Moreover, a reduced rate of mountain building would have decreased the rate of foredeep subsidence favouring shallower water and oxic/suboxic bottom‐water conditions. Thus, the hydrography of the lake during accumulation of the Chang 71 was not conducive for organic carbon accumulation. Together, the correlation of the Qinling mountain building and magmatism and deposition of the Chang 7 shale provide evidence for the role of tectonism in the accumulation of organic carbon enriched sediment. The Upper Triassic Chang 7 Member lacustrine organic-rich shale of the Yanchang Formation of the Ordos Basin is a significant hydrocarbon source rock containing abundant of tuff intervals ranging from millimeters to tens of centimeters thick. The present study relies upon petrographic and geochemical data of Chang 7 tuff intervals and organic-rich shale to consider fertilization of the lake surface by volcanic debris as a triggering mechanism of black shale sedimentation. Paleoproductivity proxies, including P/Al, Ni/Al, and Cu/Al, display elevated values in association with tuff intervals, suggesting increased productivity induced by volcanic nutrient seeding of the lake surface. Moreover, vertical variations of TOC mimic trends of paleoproductivity proxies, indicating that primary productivity was critical to organic carbon enrichment of the Chang 7 Member. Previous studies have postulated that lake bottom redox conditions were predominantly oxic–suboxic during deposition of the Chang 7 and that these deposits accumulated slowly, neither of which would have favored the export and preservation of organic matter in the absence of enhanced productivity. Accumulation of the organic-rich Chang 7 shale demonstrates the critical role that intensified primary productivity triggered by volcanism can play in the accumulation of carbonaceous sediment. Abstract Overactivation of beta-catenin/TCF signaling in prostate cancer is very common. However, how the beta-catenin/TCF complex is regulated in the nucleus remains largely unknown. In this study, we have shown that NOL8, a binding protein of beta-catenin, enhanced the interaction between beta-catenin and TCF4, and activated beta-catenin/TCF signaling. NOL8 is up-regulated in the prostate cancer, and promoted the growth, migration and colony formation of cancer cells. Knocking down the expression of NOL8 inhibited the growth, migration and colony formation of prostate cancer cells. The molecular mechanism study demonstrated that NOL8 promoted the migration and colony formation of cancer cells by activating beta-catenin/TCF signaling. Taken together, this study demonstrated the oncogenic roles of NOL8 in prostate cancer and suggested that NOL8 might be an important therapeutic target for prostate cancer. The concentration and distribution of organic matter, phosphate nodules, and framboidal pyrite preserved in shales are controlled by organic matter decomposition and the cycles of phosphorus, sulfur, and iron, which, in turn, are controlled by bottom-water redox conditions. The organic-rich shale in the 7th member of the Upper Triassic Yanchang Formation (Ch7) contains high organic carbon concentrations and abundant phosphate nodules and pyrite framboids, providing an opportunity to better understand the redox conditions at the time of deposition. Framboidal pyrite in the Ch7 organic-rich shales are composed of octahedral, pyritohedral, and spherulitic microcrystals with uniform crystal size. SEM photographs and statistical data suggest that pyrite framboids have large mean diameters (10.0 to 18.1 μm) and standard deviations (2.8 to 5.9 μm). This may indicate that the organic-rich shale was deposited under oxic-suboxic bottom-water conditions. The C org :P ratios of the organic-rich shale exhibit considerable variation, ranging from 7.2 to 216, but the majority of them (83.2%) belong to the lower-intermediate range (< 130), suggesting that the redox conditions were predominantly oxic-suboxic and intermittently anoxic. The C org :P ratios also show periodic vertical variation, which suggests that redox conditions fluctuated between oxic-suboxic and anoxic. Additional support for the trends based on C org :P ratios is provided by DOP T values, which show similar vertical variation and redox patterns. All evidence shows that the redox conditions during deposition of the Ch7 organic-rich shale was predominantly oxic-suboxic accompanied by intermittent anoxic environments. Compared to conventional reservoirs, pore structure and diagenetic alterations of unconventional tight sand oil reservoirs are highly heterogeneous. The Upper Triassic Yanchang Formation is a major tight-oil-bearing formation in the Ordos Basin, providing an opportunity to study the factors that control reservoir heterogeneity and the heterogeneity of oil accumulation in tight oil sandstones. The Chang 8 tight oil sandstone in the study area is comprised of fine-to medium-grained, moderately to well-sorted lithic arkose and feldspathic litharenite. The reservoir quality is extremely heterogeneous due to large heterogeneities in the depositional facies, pore structures and diagenetic alterations. Small throat size is believed to be responsible for the ultra-low permeability in tight oil reservoirs. Most reservoirs with good reservoir quality, larger pore-throat size, lower pore-throat radius ratio and well pore connectivity were deposited in high-energy environments, such as distributary channels and mouth bars. For a given depositional facies, reservoir quality varies with the bedding structures. Massive- or parallel-bedded sandstones are more favorable for the development of porosity and permeability sweet zones for oil charging and accumulation than cross-bedded sandstones. Authigenic chlorite rim cementation and dissolution of unstable detrital grains are two major diagenetic processes that preserve porosity and permeability sweet zones in oil-bearing intervals. Nevertheless, chlorite rims cannot effectively preserve porosity-permeability when the chlorite content is greater than a threshold value of 7%, and compaction played a minor role in porosity destruction in the situation. Intensive cementation of pore-lining chlorites significantly reduces reservoir permeability by obstructing the pore-throats and reducing their connectivity. Stratigraphically, sandstones within 1 m from adjacent sandstone-mudstone contacts are usually tightly cemented (carbonate cement > 10%) with low porosity and permeability (lower than 10% and 0.1 mD, respectively). The carbonate cement most likely originates from external sources, probably derived from the surrounding mudstone. Most late carbonate cements filled the previously dissolved intra-feldspar pores and the residual intergranular pores, and finally formed the tight reservoirs. The petrophysical properties significantly control the fluid flow capability and the oil charging/accumulation capability of the Chang 8 tight sandstones. Oil layers usually have oil saturation greater than 40%. A pore-throat radius of less than 0.4 μm is not effective for producible oil to flow, and the cut off of porosity and permeability for the net pay are 7% and 0.1 mD, respectively. Abstract: β-Microseminoprotein (MSMB) is one of the most abundant proteins in human seminal plasma. It has been identified that MSMB increased significantly in oligoasthenoteratozoospermic patients compared with fertile controls. We hypothesized that the functional polymorphism (rs10993994) of MSMB gene could be a risk factor for spermatogenic failure. For this study, 338 patients with idiopathic oligozoospermia or azoospermia and 382 fertile controls were recruited from an infertility clinic. Semen analysis was performed by computer-assisted semen analysis system. The functional polymorphism of MSMB gene was genotyped using TaqMan method. Sixty three seminal plasma samples were used to test the expression of MSMB by enzyme-linked immunosorbent assay (ELISA). The TT genotype and T allele were associated with an increased risk of idiopathic infertility with azoospermia (TT genotype: OR, 1.75; 95% CI, 1.03–2.95; T allele: OR, 1.34; 95% CI, 1.03–1.75). However, no differences were found in risk for the TT genotype or T allele among men with oligozoospermia. In addition, idiopathic infertile males have significantly higher MSMB expression levels than fertile controls. We present the first epidemiologic evidence supporting the involvement of common genetic polymorphism in MSMB gene in spermatogenic failure. These results suggest that men carrying the variant have an increased risk of spermatogenic failure associated with male infertility. Further studies are needed to confirm the roles of the polymorphism in idiopathic azoospermia and investigate the biological mechanism of elevated MSMB expression in infertile males. Authigenic chlorite plays an important controlling role in reservoir quality, and is widely distributed in the tight sandstone reservoirs of the Triassic Yanchang Formation in the Southwestern Ordos Basin, China. The occurrence and origins of authigenic chlorite were investigated using an integrated approach of optical microscopy, scanning electron microscopy, X-ray diffraction, electron probe microanalysis, and fluid inclusion analysis. The sandstones are fine-grained lithic arkoses and feldspathic litharenite with 11.5% volcanic rock fragments. Authigenic chlorite occurs in two morphotypes, including pore-lining and pore-filling chlorite. Pore-lining chlorite occurs as coatings on the framework grains and covers the entire fining-upward sandstones. Pore-filling chlorite is restricted to the middle to upper part of the fining-upward sandstones and appears as discrete flaky plates or rosettes in the pores or attaches to the surfaces of pore-lining chlorite. Pore-lining chlorite originated from smectite precursors that were derived from volcanic material alteration in the early stage of diagenesis, with temperatures primarily about 90 °C. In contrast, pore-filling chlorite was precipitated in pore spaces with higher concentrations of Mg2+ and Fe2+ released from volcanic rock fragments and mica debris dissolution. Moreover, the chemical composition of pore-filling chlorite was affected by early calcite cementation which consumed some ions at temperatures lower than about 110 °C. The thin and continuous pore-lining chlorite is the only chlorite type with a positive effect on reservoir quality, whereas the very thick pore-lining chlorite (>6.7 μm) or pore-filling chlorite significantly reduce the porosity and permeability. Abstract: Studies of the relationship between male infertility and polymorphisms in the regions of FSHR G-29A (rs1394205), 919A>G (Thr307Ala, rs6165) and 2039A>G (Asn680Ser, rs6166) have reported inconsistent results. To assess the association between them, a meta-analysis was conducted. PubMed and CBMdisc literature search were conducted to identify all eligible studies investigating such a relationship. The pooled ORs were performed for co-dominant model, dominant model and recessive model in FSHR G-29A, Thr307Ala and Asn680Ser respectively to assess the strength of association. A total of 1644 male infertility cases and 1748 controls were collected from seven case–control studies. In the overall analysis, no significant association between the three polymorphisms and risk of male infertility was observed. Stratified analysis showed that there were no significantly increased risks of azoospermia and oligoasthenoteratozoospermia (OAT) in any of the genetic models. This meta-analysis supports that FSHR G-29A, Thr307Ala and Asn680Ser polymorphisms may not be capable of causing male infertility susceptibility. [Copyright &y& Elsevier]

Purpose: To analyze the abundance and difference of voltage-dependent anion channel (VDAC) mRNA in ejaculated spermatozoa from normozoospermic fertile donors and infertile patients with idiopathic asthenozoospermia. Methods: High motile and low motile spermatozoa were separated respectively from ejaculates of 36 donors and 40 patients using a discontinuous Percoll gradient centrifugation. Real-Time PCR was performed to detect mRNA abundance and difference of three VDAC subtypes between two groups with different sperm motility. Results: Real-Time PCR demonstrated that three VDAC mRNAs were present in mature spermatozoa. The VDAC2 mRNA level in ejaculated spermatozoa of patients was significantly higher than that of donors. No significant differences of VDAC1 and VDAC3 mRNA levels were found between two groups. Conclusion: The high abundance of VDAC2 mRNA seemed to have a positive correlation with low sperm motility. Objective:Epididymal protease inhibitor (Eppin) was located on the surface of spermatozoa and modulates the liquefaction of human semen. Here, we identify the correlative protein partner of Eppin to explore the molecular mechanism of liquefaction of human semen. Methods:(1) Human seminal vesicle proteins were transferred on the membrane by Western blotting and separated by 2-D electrophoresis and incubated in recombinant Eppin. The correlative protein was identified by Mass Spectrometry (MS) (2). Western blotting was used to determine the relation of rEppin and rFibronectin(Fn); (3) Co-localization in spermatozoa were detected using immunofluorescence; (4) Correalation of Eppin and Fn was proved by co-immunoprecipitation. Results:Fn was identified as the binding partner of recombinant Eppin by MS. Recombinant of Eppin was made and demonstrated that the Eppin fragment binds the fn 607-1265 fragment. The Eppin-Fn complex presents on the sperm tail and particularly in the midpiece region of human ejaculated spermatozoa. Immunoprecipitation indicated that Eppin in the spermatozoa lysates was complexed with Fn. Conclusions:Our study demonstrates that Eppin and Fn bind to each other in human semen and on human ejaculated spermatozoa. Eppin-Fn complex may involve in semen coagulation, liquefaction and the survival and preparation of spermatozoa for fertility in the female reproductive tract. [ABSTRACT FROM AUTHOR]
