Abstract

According to the WHO, about 3 million people die each year due to ambient air pollution. Most of the in vivo studies on the PM2.5 effects have been done on respiratory and cardiovascular tissues. However, little is known about the effects on the tissues involved on xenobiotic removal, such as kidneys. In the present study we assess the harmful effects of sub-chronic exposure to PM2.5 on the kidney, by investigating histologic and serum alterations in healthy and hypertensive rat models. Mean PM2.5 concentrations during exposures were slightly above the daily WHO standard. Exposed animals showed fibrosis, mesangial expansion, decrease glomerular and tubular lumen volumes in kidneys, with an elevated BUN. Hypertensive animals also exhibited much more severe alterations than healthy animals. We conclude that PM2.5 induces minimal or small-scale abnormalities that can be determinant for renal health preservation. Keywords: PM2.5, Hypertension, Elemental composition, Kidney histology, BUN

Abstract

Introduction: Type 2 diabetes mellitus (DM2) is characterized by being an inflammatory disease, with systemic, chronic, multimetabolic involvement. Its physiopathogenic mechanism is characterized by glyco-lipotoxicity, chronic oxidative stress, which cause hypoinsulinemia due to the death of beta cells or their transdifferentiation. Objective: To determine the effect of an enriched environment on metabolic markers and pancreatic histology in an experimental model of type 2 diabetes mellitus. Methodology: Metabolic markers were measured during the 16-month trial. In older adult Wistar rats at the end of the experiment, the animals were two years old. At the end of the experiment, the pancreas of the animals were studied histologically. Data were analyzed by ANOVA and paired t test (p≤0.05).

Abstract

Introducción: la diabetes mellitus tipo 2 (DM2) se caracteriza por ser una enfermedad inflamatoria, con afectación sistémica, crónica, plurimetabólica. Su mecanismo fisiopatogénico se caracteriza por glico-lipotoxicidad, estrés oxidativo crónico, que ocasionan hipoinsulinemia debido a la muerte de las células beta o su transdiferenciación.Objetivos: determinar el efecto de un ambiente enriquecido sobre marcadores metabólicos e histología pancreática en un modelo experimental de DM2.Materiales y métodos: se utilizaron 39 ratas Wistar macho de 12 meses edad al inicio del ensayo divididas en 5 grupos.Para alterar su metabolismo se utilizó una dieta con 30% de grasas saturadas (HFD) y/o consumo moderado (0,42 g/kg peso/día) de alcohol (Alc). El grupo con ambiente enriquecido (HFD+Alc+AE) se alojó en una jaula de mayor tamaño (250 dm3), con ruedas de correr y rampas, los otros animales en jaulas estándar de bioterio. El grupo control (C) se alimentó con dieta Chow. Se midieron marcadores metabólicos durante el ensayo de 16 meses de duración. Al finalizar el experimento se estudiaron histopatológicamente los páncreas de los animales. Los datos se analizaron mediante ANOVA y test t apareado (p=0,05).
