Abstract

Research shows that higher education can help veterans build a new sense of identity, purpose and belonging after leaving the military. There are critical gaps, however, in our understanding of student veterans in the Australian context. This article aims to map the experiences of Australian veterans across the student life cycle to improve our understanding of this unique group. We systematically address the higher education stages of pre-access, access, participation, and attainment. Fourteen autobiographical interviews were conducted to obtain detailed and nuanced personal reflections. Findings demonstrate that veterans often make exemplary students who are motivated to pursue altruistic careers. However, transitions between the military and higher education can be difficult to navigate. We argue the need for a structured pathway between the military and higher education and increased support for veterans across the student life cycle. We recommend a range of initiatives to prepare veterans to thrive in higher education. [ABSTRACT FROM AUTHOR]

Abstract

Most RNA molecules form internal base pairs, leading to a folded secondary structure. Some of these structures have been demonstrated to be functionally significant. High-throughput RNA structure chemical probing methods generate millions of sequencing reads to provide structural constraints for RNA secondary structure prediction. At present, processed data from these experiments are difficult to access without computational expertise. Here we present FoldAtlas, a web interface for accessing raw and processed structural data across thousands of transcripts. FoldAtlas allows a researcher to easily locate, view, and retrieve probing data for a given RNA molecule. We also provide in silico and in vivo secondary structure predictions for comparison, visualized in the browser as circle plots and topology diagrams. Data currently integrated into FoldAtlas are from a new high-depth Structure-seq data analysis in Arabidopsis thaliana, released with this work.Availability and implementationThe FoldAtlas website can be accessed at www.foldatlas.com Source code is freely available at github.com/mnori/foldatlas under the MIT license. 18. Characterising fitness effects of gene copy number variation in yeast

Abstract

Diploid organisms including yeast, most animals, and humans, typically carry two copies of each gene. Variation above or below two copies can however sometimes occur. When gene copy number reduction from two to one causes a disadvantage, that gene is considered haploinsufficient (HI). In the first part of my work, I identified associations between Saccharomyces cerevisiae gene properties and genome-scale HI phenotypes from earlier work. I compared HI profiles against 23 gene properties and found that genes with (i) greater numbers of protein interactions, (ii) greater numbers of genetic interactions, (iii) greater gene sequence conservation, and (iv) higher protein expression were significantly more likely to be HI. Additionally, HI showed negative relationships with (v) cell cycle regulation and (vi) promoter sequence conservation. I exploited the aforementioned associations using Linear Discriminant Analysis (LDA) to predict HI in existing data and guide experimental identification of 6 novel HI phenotypes, previously undetected in genome-scale screenings. I also found significant relationships between HI and two gene properties in Schizosaccharomyces pombe, relationships that hold despite the lack of conserved HI between S. cerevisiae and Sz. pombe orthologue gene pairs. These data suggest associations between HI and gene properties may be conserved in other organisms. The relationships and model presented here are a step towards understanding HI and its underlying mechanisms. Increases in copy number can occur through gene duplication. When duplication produces two functional gene copies, both experience relaxed selection and rapid mutation. This sometimes leads to interesting evolutionary events such as gain of novel function (neofunctionalisation). Previous work shows an ancient ancestor of S. cerevisiae underwent whole genome duplication (WGD) followed by massive redundant gene loss. Interestingly some duplicate pairs show retention of both copies, including the pair TUB1 and TUB3. Existing sequence data shows that TUB3 has experienced a very high rate of evolution post-WGD, suggesting neofunctionalisation. To characterise TUB3, I have carried out experiments measuring fitness effects of varying TUB1, TUB2 and TUB3 copy number across many environments. In ethanol media, some TUB1 and TUB3 null mutants interestingly show severe defects. Other data suggest stress response, ethanol tolerance, protein degradation and/or regulatory roles, which may involve the regulatory Snf1p protein kinase complex.

37. Investigating student approaches to scenario-based assessments of systems thinking

Abstract

The development of systems thinking is considered a critical skill set for addressing interdisciplinary problems. This skill set is particularly important in the field of engineering, where engineers are often tasked with solving socio-technical problems that often require knowledge beyond their original discipline and practice in unfamiliar contexts. However, existing assessments often fail to accurately measure teachable knowledge or skills that constitute systems thinking. To investigate this issue, we compared students' performance on two previously and independently peer-reviewed scenario-based assessments for systems thinking: The Village of Abeesee and the Lake Urmia Vignette. Twenty undergraduate engineering students participated in a multi-phase case study utilizing think aloud protocols and semi-structured interview methods to elicit the approaches students took thinking across the two instruments and past experiences that they felt prepared them to solve these ill-structured problems. We found that the way a scenario is presented to students impacts their subsequent problem-solving approach, which complicates assessment of systems thinking. Additionally, students identified only limited opportunities for the development of ill-structured problem-solving skills necessary for systems thinking. Our findings inform future work on improving systems thinking assessments and emphasize the importance of more intentionally supplying opportunities for students to practice solving ill-structured problems throughout the curriculum. Abstract

Commercial cropping farms in New Zealand (NZ) showed highly variable rates of nitrate-N leaching losses (13–148 kg/ha/year) across sites, cropping systems and seasons. Losses were measured at 1.2 m below the soil surface using a network passive-wick drainage fluxmeters (DFMs) which were installed across nine commercial farms between August 2014 and June 2016. Sites were located in the Canterbury, Manawatu, Hawke's Bay, Waikato and Auckland regions and monitored for a period 51 to 72 months. Twelve DFMs were installed at each site and drainage volumes were validated using an established soil water balance model. At seven sites, losses were calculated using measured drainage and measured concentrations, and at two sites losses were calculated using modelled drainage and measured concentrations. Nitrate-N was the predominant form of inorganic N (96.7–99.9%) in drainage water and annual losses averaged 52 kg/ha/yr for mixed cropping systems with livestock grazing (n = 6) and 101 kg/ha/yr for mixed cropping systems with a focus on vegetable production (n = 3). While not broadly representative of all NZ cropping land uses, results from this study do, nevertheless, suggest that when drainage occurs, nitrate losses may be considerable under the land uses represented here (> 70 kg/ha/y).
