Wiseman et al. show that triplication of genes other than APP is sufficient to exacerbate Aβ deposition and associated cognitive changes in a mouse model of Down syndrome – Alzheimer’s disease. This occurs independently of changes to γ-secretase but results from a novel mechanism that lowers the soluble Aβ40/42 ratio., Down syndrome, caused by trisomy of chromosome 21, is the single most common risk factor for early-onset Alzheimer’s disease. Worldwide approximately 6 million people have Down syndrome, and all these individuals will develop the hallmark amyloid plaques and neurofibrillary tangles of Alzheimer’s disease by the age of 40 and the vast majority will go on to develop dementia. Triplication of APP, a gene on chromosome 21, is sufficient to cause early-onset Alzheimer’s disease in the absence of Down syndrome. However, whether triplication of other chromosome 21 genes influences disease pathogenesis in the context of Down syndrome is unclear. Here we show, in a mouse model, that triplication of chromosome 21 genes other than APP increases amyloid-β aggregation, deposition of amyloid-β plaques and worsens associated cognitive deficits. This indicates that triplication of chromosome 21 genes other than APP is likely to have an important role to play in Alzheimer’s disease pathogenesis in individuals who have Down syndrome. We go on to show that the effect of trisomy of chromosome 21 on amyloid-β aggregation correlates with an unexpected shift in soluble amyloid-β 40/42 ratio. This alteration in amyloid-β isoform ratio occurs independently of a change in the carboxypeptidase activity of the γ-secretase complex, which cleaves the peptide from APP, or the rate of extracellular clearance of amyloid-β. These new mechanistic insights into the role of triplication of genes on chromosome 21, other than APP, in the development of Alzheimer’s disease in individuals who have Down syndrome may have implications for the treatment of this common cause of neurodegeneration.

The basic research dilemma of the present study relates to the assumption that every individual can learn, not based on a personal innate intelligence but what and how a person learns to acquire intelligence. The investigation involved developing an instrument to detect the trait of basic knowledge through knowing factors that form a student’s knowledge preference. Furthermore, the study attempted to analyze the association between knowledge preference and the student’s nurturing culture and environmental influential factors. The assessment of tacit knowledge is to gain information that has been influent student’s learning formally or informally. Student early characteristics of the knowledge learning preference can be derived from different sources of knowing, for instance, the biological heredity procession in family, language, thinking process, perception, and frames of thought. A test named the Knowledge Preference Test was developed, and it consisted of 50 items. It statistically received a reliability coefficient of 0.91. The study participants included 2 017 secondary school students in Thailand. The study found significant association patterns among knowledge preference, student biological and cultural status, and some areas of student achievement. The purpose of this study was to examine the performance of the Thai-version of WHOQOL-BREF in assessing the quality of life (QoL) among Thai college students. The psychometric properties of WHOQOL-BREF were assessed in this study. The self-administered WHOQOL-BREF questionnaire was applied. A total of 407 Thai college students (male age = 20.5 ± 1.2; female age = 20.5 ± 1.2) participated in this study. Item-response distributions, internal consistency reliability, discriminant validity, criterion-related validity and construct validity through confirmatory analysis were analyzed. The findings indicate that the WHOQOL-BREF had acceptable internal consistency (α = 0.73–0.83 across four domains), all items highly correlated with corresponding domain scores (r = 0.53–0.80), the indices of a two-order confirmatory factor analysis (CFA) demonstrate that the data fit the model well with allowing covary of error variances of some items, all items had good property of criterion-related validity and item discrimination and, all three domain scores except the social relationship domain had significant associations with overall QoL or general health. The results suggest that the WHOQOL-BREF was reliable and valid to health professionals in the assessment of the QoL of college-based Thai youth, but some unsuitable items may be deleted in future studies.

Suicide of an individual could leave devastating consequences for family, friends, relatives, and society. Suicide could be considered a serious concern and issue to public health, especially among adolescents. The purpose of the study was to examine associations of suicide ideation with attitudes toward suicide (ATS), quality of life (QOL), and attitudes toward death and dying (ADD) in high school seniors. The participants were conveniently sampled and consisted of 1,247 high school senior students from China, South Korea, Thailand, and Vietnam. The survey found that 21.7% of the Chinese, 20.9% of the Korean, 16.8% of the Thai, and 26.3% of the Vietnamese participants exhibited suicide ideation. The analysis revealed that student suicide ideation was significantly associated with ATS, QOL, and sustained elements of depression during the past 12 months in all four countries. In addition, there were statistically differences among the Chinese, Korean, Thai, and Vietnamese students in QOL, ATS, and ADD. The analysis determined that there were certain predictive factors for suicide ideation in the participants. The impact of these findings could extend to professional roles within the school system including curricula and services. Administrators, faculty, or social workers could gain a better understanding of the current suicide ideation phenomenon and help raise awareness to reduce potential suicide attempts among high school students in these Asian countries.

PURPOSE: The 21 Critical National Health Objectives (CNHOs) for Adolescents and Young Adults derived from Healthy People 2010 addressed the most significant threats to the health of individuals aged 10-24 years. This study assessed trends in the 21 CNHOs between 1991 and 2009, and from baseline years for which 2010 targets were established to 2009, and the extent to which targets were achieved. METHODS: For one CNHO (new HIV diagnoses), national data were not available. For CNHOs measured by census systems, the percentage of change in each health outcome was calculated between 1991 and 2009 and between baseline years and 2009. Any change ≥5% was considered as an improvement or deterioration. For CNHOs measured by national probability-based surveillance surveys, multivariate logistic regression was conducted using Stata Version 10.0 (StataCorp, College Station, TX) to calculate odds ratios for each outcome from 1991, and from baseline years to 2009, controlling for gender, race/ethnicity, and age or school grade-level. To calculate the percentage of targets being achieved, the difference between baseline data and 2009 data was divided by that between baseline data and target. RESULTS: Adolescents and young adults achieved two targets (rode with a driver who had drunk alcohol, physical fighting), improved for 12 CNHOs, made mixed progress by sub-objective for two, showed no progress in four, and regressed in achieving two (Chlamydia infections; overweight). Progress varied by demographic variables. CONCLUSION: Although encouraging trends were seen in young people's health, the United States achieved only two CNHOs. Objective To examine the relationship between the total volume of leisure-time physical activity (LTPA) and obesity among African American adults in Indianapolis. Methods Logistic regression analysis with 649 African American adults. Results The data show an inverse graded relationship between the total volume of LTPA and obesity for African American women, but not for men. Conclusions African American women who accumulate a high volume of LTPA (ie, 300 minutes or more per week) are less likely to be obese. Further research is needed to investigate the gender difference in the effect of LTPA on obesity.
