Silicon (Si) application may affect the plant response to nitrogen (N), possibly by changing the uptake, concentration, and partitioning of nutrients in plant tissues; however, this has not yet been proven in Arabica coffee plants. The effects of Si application methods [no Si, soil-applied soluble Si (168 mg Si L−1), and foliar-applied soluble Si (two application of 2 mg Si plant−1)] and N levels (0 and 80 mg N L−1) on biomass production and partitioning and uptake, partitioning, and stoichiometry of nutrients and Si in young Arabica coffee plants grown under greenhouse conditions were evaluated. Nitrogen fertilization increased the biomass production and uptake of all nutrients; however, reduced the concentrations of K, Ca, Mg, S, Mn, and Si in the leaves, Si in the stems, and K, Mg, and S in the roots of coffee plants as a dilution effect. In the presence of N, soil-applied Si increased the concentrations of Zn in the leaves and Ca and Si in the stems, the uptake of K, S, and Si, and the Si:N ratio. Foliar-applied Si increased the concentrations of N, P, K, and Zn in the leaves and Ca and Si in the stems, as well as the total uptake of K and Si and the Si:N ratio in coffee plants, being more evident in the N fertilization presence. This study unraveled that, especially when it was soil-applied, Si altered the nutrient uptake, allocation, and stoichiometric ratios with N, with a consequent increase in biomass production of young coffee plants fertilized with N. Increasing wheat (Triticum aestivum L.) yield and grain protein concentration (GPC) without excessive nitrogen (N) inputs requires understanding the genotypic variations in N accumulation, partitioning, and utilization strategies. This study evaluated whether high protein genotypes exhibit increased N accumulation (herein also expressed as N nutrition index, NNI) and partitioning (including remobilization from vegetative organs) compared to low-protein genotypes under low and high N conditions. Four winter wheat genotypes with similar yields but contrasting GPC were examined under two N rates (0 and 120 kg N ha-1) across two environments and four growing seasons in Oklahoma, US. As expected, the high-protein genotypes Doublestop CL+ (Dob) and Green Hammer (Grn) had greater GPC than the medium- (Gallagher, Gal) and low-protein genotypes (Iba), without any difference in grain yield. Total plant N accumulation at maturity showed diminishing increases for greater grain yield, and low-protein genotype showed greater N utilization efficiency (NUtE) than high-protein genotypes. The high-protein genotype Grn tended to achieve higher GPC by increasing total N uptake, while Dob exhibited a tendency towards higher N partitioning to grain (NHI). The allometric relationship between total N accumulation and biomass remained unchanged for both high- and low-protein genotypes. The N remobilization patterns differed between high- and low-protein genotypes. As N conditions improved, the proportional contributions of remobilized N from leaves tended to increase, while contributions from stems and chaff tended to decrease or remained unchanged for high-protein genotypes. This study highlights the importance of both N uptake capacity and efficient N partitioning to the grain as critical traits for realizing wheat's dual goals of higher yield and protein. Leaf N remobilization plays a critical role during grain filling, sustaining plant N status and contributing to protein levels. The higher NUtE observed in the low-protein genotype Iba likely contributed to its lower GPC, emphasizing the trade-off between NUtE and GPC. The physiological strategies employed by high-protein genotypes, such as genotype Grn's tendency for increased N uptake and Dob's efficient N partitioning, provide a foundation for future breeding efforts aimed at developing resource-efficient and nutritionally superior wheat genotypes capable of achieving both increased yield and protein. Rice is the most important crop in India and additionally the hub of food security of the worldwide population. Integrated application of manures and fertilizers played a pivotal role for improvement of soil physico-chemical properties under different cropping systems as well as nutrient accumulation and grain yield in rice. The present investigation was conducted in sandy loam soil at Agronomy Research Farm of Odisha University of Agriculture and Technology, Bhubaneswar, Odisha, India during 2018–2019 having sub-humid climate. Field experiment was designed to evaluate the effect of two date of transplanting, which were allotted to main plots and three combinations of nutrient management options allotted for sub plots on rice crop (var. Naveen). Among all the plant parts, leaf showed higher accumulation of nitrogen (3.69, 1.92%), stem showed higher accumulation of both phosphorus (0.43, 0.35%) and potassium (2.59, 2.09%) than other plant parts during vegetative stages, while panicle showed higher accumulation of nitrogen (1.22%), phosphorus (0.28%) during harvesting stages of the rice crop. Significantly higher nitrogen accumulation in different plant parts were optimized under Soil Test Based Fertilizer Recommendation (STBFR) + Green Manure (GM; Sesbania rostrata) (0.99, 0.46, 1.28%) method of nutrient management, while phosphorus (0.10, 0.11, 0.28%) and potassium (1.00, 1.64, 0.33%) accumulation were found to be significantly higher in different plant parts under STBFR + Farm Yard Manure (FYM) method of nutrient management. ABSTRACT: This study aimed to assess the effect of bulk tank milk (BTM), waste milk (WM), and pasteurized waste milk (PWM) on nutrient digestibility, ruminal and cecal fermentation, gastrointestinal tract (GIT) development, and antimicrobial resistance of fecal Escherichia coli from dairy calves at 2 periods (30 and 60 d of age). Calves were grouped according to BW, serum protein levels, and breed composition. Three treatments were included: BTM (n = 21), WM from cows under antibiotic treatment (n = 21), and PWM (WM submitted to HTST pasteurization; n = 21). A total of 63 calves were used, of which 18 animals (n = 6 per treatment) were evaluated in the period of 4 to 30 d, and 45 (n = 15 per treatment) from 4 to 60 d. During the experimental period, a daily intake of 6 L of milk was divided into 2 equal meals, with ad libitum access to water and starter. Milk and feed intakes were recorded daily. Apparent total-tract digestibility and nitrogen balance were conducted from 25 to 29 d of age (n = 6) and from 53 to 57 d of age (n = 15). Animals were slaughtered at 30 ± 1 and 60 ± 1 d of age for the assessment of ruminal and cecal fermentation and GIT development. Antimicrobial susceptibility testing was conducted at 1, 30, and 60 d of age (n = 15/treatment). Statistical analysis used a linear mixed-effects model for continuous outcomes and generalized linear models for single measurements (R software). Treatments WM and PWM had lower rumen pH, higher ruminal acetate concentration, larger reticulorumen and liver, and a higher prevalence of fecal-resistant E. coli compared with BTM at both 30 and 60 d. Up to 60 d, both BTM and WM treatments exhibited higher digestibility of ether extract and gross energy compared with the PWM, whereas WM and PWM treatments showed increased nitrogen intake and retention compared with the BTM treatment. These findings suggest that pasteurization of WM negatively affects nutrient digestibility and calf performance, while also affecting rumen development. Additionally, the use of milk containing antibiotic residue leads to the selection of resistant E. coli in the GIT over time.

Fall-calving primiparous beef females [body weight (BW): 451 ± 28 (SD) kg; body condition score (BCS): 5.4 ± 0.7] were individually-fed 100% (control; CON; n  = 13) or 70% (nutrient restricted; NR; n  = 13) of estimated metabolizable energy and metabolizable protein requirements from day 160 of gestation to calving. Post-calving, all dams were individually-fed tall fescue hay supplemented to meet estimated nutrient requirements for maintenance, growth, and lactation until day 149 of lactation. Four-hour milk yields were collected on days 21, 42, 63, 84, 105, and 147 of lactation, and milk nutrient composition was determined. Doppler ultrasonography of both pudendoepigastric arterial trunks was conducted every 21 d from days 24 to 108 of lactation. Total mammary blood flow was calculated, and hemodynamics from both sides were averaged. Data were analyzed as repeated measures with nutritional plane, day of lactation, their interaction, calving date, and calf sex (if P  < 0.25) as fixed effects. We previously reported that post-calving, NR dams weighed 64 kg less and were 2.0 BCS lower than CON, but calf birth weight was not affected. Milk weight and volume were 15% less (P  = 0.04) for NR dams than CON. Milk protein concentration was lower (P  = 0.008) for NR dams than CON, but triglyceride and lactose concentrations were not affected (P  ≥ 0.20) by nutritional plane. Milk urea N concentration of NR dams tended to be greater (P  = 0.07) on day 42 but was lower (P  = 0.01) on day 147 of lactation than CON. Total milk protein, triglyceride, and lactose yields were less (P  ≤ 0.05) for NR dams than CON. Total milk urea N yield was less (P  ≤ 0.03) for NR dams than CON on days 21, 63, and 147 of lactation. Maternal heart rate was greater (P  = 0.008), but pudendoepigastric arterial trunk peak systolic velocity, resistance index, and cross-sectional area were less (P  ≤ 0.04) and pulsatility index tended to be less (P  = 0.06) for NR dams than CON. Mammary blood flow was 19% less (P  = 0.004) for NR dams than CON, but mammary blood flow relative to milk weight or dam BW was not affected (P  ≥ 0.14) by nutritional plane. Most milk yield, milk nutrient composition, and mammary blood flow variables were affected (P  ≤ 0.04) by day of lactation. In summary, first-parity beef females that were nutrient restricted during late gestation and then fed to meet estimated nutrient requirements during lactation had decreased milk nutrient yield and a similar reduction in mammary blood flow. ABSTRACT: Peripartum rumen-protected choline (RPC) supplementation is beneficial for cow health and production, yet the optimal dose is unknown. In vivo and in vitro supplementation of choline modulates hepatic lipid, glucose, and methyl donor metabolism. The objective of this experiment was to determine the effects of increasing the dose of prepartum RPC supplementation on milk production and blood biomarkers. Pregnant multiparous Holstein cows (n = 116) were randomly assigned to one of 4 prepartum choline treatments that were fed from −21 d relative to calving (DRTC) until calving. From calving until +21 DRTC, cows were fed diets targeting 0 g/d choline ion (control, CTL) or the recommended dose (15 g/d choline ion; RD) of the same RPC product that they were fed prepartum. The resulting treatments targeted: (1) 0 g/d pre- and postpartum [0.0 ± 0.000 choline ion, percent of dry matter (%DM); CTL]; (2) 15 g/d pre- and postpartum of choline ion from an established product (prepartum: 0.10 ± 0.004 choline ion, %DM; postpartum: 0.05 ± 0.004 choline ion, %DM; ReaShure, Balchem Corp.; RPC1RD▸RD); (3) 15 g/d pre- and postpartum of choline ion from a concentrated RPC prototype (prepartum: 0.09 ± 0.004 choline ion, %DM; postpartum: 0.05 ± 0.003 choline ion, %DM; RPC2, Balchem Corp.; RPC2RD▸RD); or (4) 22 g/d prepartum and 15 g/d postpartum from RPC2 [prepartum: 0.13 ± 0.005 choline ion, %DM; postpartum: 0.05 ± 0.003 choline ion, %DM; high prepartum dose (HD), RPC2HD▸RD]. Treatments were mixed into a total mixed ration, and cows had ad libitum access via a roughage intake control system (Hokofarm Group). From calving to +21 DRTC, all cows were fed a common base diet and treatments were mixed into the total mixed ration (supplementation period, SP). Thereafter, all cows were fed a common diet (0 g/d choline ion) until +100 DRTC (postsupplementation period, postSP). Milk yield was recorded daily and composition analyzed weekly. Blood samples were obtained via tail vessel upon enrollment, approximately every other day from −7 to +21 DRTC, and at +56 and +100 DRTC. Feeding any RPC treatment reduced prepartum dry matter intake compared with CTL. During the SP, no evidence for a treatment effect on energy-corrected milk (ECM) yield was found, but during the postSP, RPC1RD▸RD and RPC2RD▸RD treatments tended to increase ECM, protein, and fat yields. During the postSP, the RPC1RD▸RD and RPC2RD▸RD treatments tended to increase, and RPC2HD▸RD increased, the de novo proportion of total milk fatty acids. During the early lactation SP, RPC2HD▸RD tended to increase plasma fatty acids and β-hydroxybutyrate concentrations, and RPC1RD▸RD and RPC2RD▸RD reduced blood urea nitrogen concentrations compared with CTL. The RPC2HD▸RD treatment reduced early lactation serum lipopolysaccharide binding protein compared with CTL. Overall, peripartum RPC supplementation at the recommended dose tended to increase ECM yield postSP, but no evidence was seen of an additional benefit on milk production with an increased prepartum dose of choline ion. The effects of RPC on metabolic and inflammatory biomarkers support the potential for RPC supplementation to affect transition cow metabolism and health and may support the production gains observed.

The increase of the area cultivated with sugarcane has demanded information about the culture, mainly in relation to its nutritional needs. There are few studies that report the nutrient uptake in sugarcane, in addition, there are no reports of studies focused on the different periods of sugarcane growth and development. In view of the above, the objective of this research was to determine the macronutrient uptake and dry matter production in the shoots of two sugarcane varieties. The experiment was carried out in Vicentina, Mato Grosso do Sul, Brazil, during the 2017/2018 harvest, using two varieties of sugarcane (RB92579 and SP832847). The experimental design was in randomized blocks with two sugarcane varieties, nine collecting times (90, 120, 150, 180, 210, 240, 270, 300 and 330 days after planting (DAP) and 4 replications. Samples were collected from the shoots of 10 plants, which were divided into leaves, sheaths and stalks, then dried and weighed, and finally the samples were submitted to laboratory analysis to determine the nutrient concentrations in each organ. The accumulation of macronutrients in the shoots of both varieties followed the order: K > N > Ca > S>Mg > P. The export by the stalk followed the order K > N > Ca > S > P > Mg for RB92579 and K > N > Ca = P > S > Mg for SP832847. Maximum accumulation of nutrients and the maximum dry matter production occurred in the interval between 240 and 300 DAP. The variety SP832847 accumulated higher amounts of nutrients and dry matter in relation to RB92579, as well as the time required for the maximum accumulation was shorter. [ABSTRACT FROM AUTHOR]

In total, 69 bulls (on average 238 ± 11 days of age at start and 367 ± 25 kg of bodyweight) were assigned to three dietary treatments (n = 23/group): Positive control (CON; 13.7% CP; 2.11 g methionine/kg DM), negative control deficient in CP (RED; 9.04% CP; 1.56 g methionine/kg DM) and crude protein-deficient ration supplemented with RPMET (RED+RPMET; 9.04% CP; 2.54 g methionine/kg DM). At slaughter, samples of liver, muscle and blood serum were taken and underwent subsequent metabolomics profiling using a UHPLC-QTOF-MS system. A total of 6540 features could be detected. Twenty metabolites in the liver, five metabolites in muscle and thirty metabolites in blood serum were affected (p < 0.05) due to dietary treatments. In total, six metabolites could be reliably annotated and were thus subjected to subsequent univariate analysis. Reduction in dietary CP had minimal effect on metabolite abundance in target tissues of both RED and RED+RPMET bulls as compared to CON bulls. The addition of RPMET altered the hepatic anti-oxidant status in RED+RPMET bulls compared to both RED and CON bulls. Results exemplify nutrient partitioning in growing German Simmental bulls: bulls set maintenance as the prevailing metabolic priority (homeostasis) and nutrient trafficking as the second priority, which was directed toward special metabolic functions, such as anti-oxidant pathways. IntroductionSoybean breeding in southwestern China has vastly improved soybean yields with the increasing demand for nutrients such as phosphorus (P) and nitrogen (N). This study aimed to assess the impact of soybean breeding on P and N utilization efficiencies.MethodsField experiments with split-plot experimental designs were conducted at two locations [Dafang (DF) and Shiqian (SQ)] in the 2019 growing season to determine the agronomic efficiency of P fertilizer (AEp), P and N utilization efficiencies, and P and N accumulation and partitioning in different soybean organs under 0 (P0) and 35 (P35) kg ha−1 P supply.ResultsThe results showed that soybean breeding targeting high seed yield also improved AEp (p < 0.05) and P (p < 0.05) and N utilization efficiencies (p < 0.05), with the improvement in AEp associated with the high yield response to P supply. P and N accumulation significantly increased in pods (p < 0.05) and leaves (p < 0.05) but not in stems or roots with year of release, while P and N concentrations did not change in any organ with year of release. In addition, only pod dry weight significantly increased (p < 0.01) with year of release, and P and N partitioning increased to pods (p < 0.05) but decreased to stems (p < 0.05) with year of release. Correlation and PCA analyses revealed P and N utilization efficiencies positively correlated with P and N partitioning to pods but negatively correlated with P and N partitioning to stems. While P supply increased P and N accumulation, it reduced P utilization efficiency.DiscussionWe conclude that (1) soybean breeding improved AEp and P and N utilization efficiencies; (2) the increased P and N partitioning to pods but decreased partitioning to stems contributed to the high P and N utilization efficiencies in new soybean cultivars, reducing the demand for N and P; (3) P supply increased nutrient accumulation but reduced P utilization efficiency. These results highlight the significance of appropriate resource allocation among organs and efficient P management for enhancing nutrient utilization and reducing fertilizer requirements.

Purpose : Mycorrhizal fungi are critical for the growth and survival of trees although the knowledge on the extent of their association with different tree species in the boreal forest remains limited. Methods: We examined the vertical distribution and composition of the root mycorrhizal communities of black spruce (Picea mariana (Mill.) B.S.P) and trembling aspen (Populus tremuloides Michx) along three soil layers (organic, minerals top 0–15 cm and bottom 15–30 cm) in pure and mixed stands, using next generation sequencing. Results: We found that spruce and aspen differ in the composition of their mycorrhizal communities in respective pure stands. The difference was present also in mixed stands, despite a shift in the composition of species-specific mycorrhizal communities between pure and mixed stands. In mixed stands, the relative abundance of spruce-specialist mycorrhizae in the organic layer was higher than that of aspen-specialists. The opposite pattern was observed in the mineral soil. The mixed stands exhibited lower richness and abundance of generalist mycorrhizae in the organic and in the mineral soil layers. Conclusion: The results suggest that it is the soil chemistry that structure species-specific mycorrhizal communities between pure stands and along different soil depth within stands. However, in mixed stands, it is the identity of tree species that determines the structure of mycorrhizae communities within soil layers. We speculate that the differences in the richness and abundance of individual mycorrhizal communities of spruce and aspen along the soil profile would likely contribute to stronger partitioning of tree nutrient uptake between these two species in mixed stands. ‘Honeycrisp’ apple is susceptible to bitter pit, which is associated with fruit mineral nutrient composition. Rootstock genotypes can affect nutrient acquisition, distribution, and fruit yields, which all affect fruit nutrient composition and bitter pit susceptibility. However, the changes of these traits among different rootstock genotypes in response to abiotic stress under semiarid conditions are relatively unknown. The objective of this study was to evaluate the influence of different rootstocks and irrigation on nutrient uptake and partitioning with ‘Honeycrisp’ apple grown in an irrigated, semiarid environment. ‘Honeycrisp’ apple trees were grafted on four different rootstocks, Geneva 41 (‘G.41’), Geneva 890 (‘G.890’), M.9-T337 (‘M.9’), and Budagovsky 9 (‘B.9’), and these were planted at high density (3000 trees/ha). Irrigation was applied as either a water-limited treatment where volumetric soil water content was maintained near 50% field capacity (FC) and a well-watered control where soil water content was maintained near 100% FC. ‘G.890’, the most vigorous rootstock, had lower nitrogen and higher potassium content in leaves, while ‘B.9’, the least vigorous rootstock, had lower potassium and higher nitrogen content. Rootstock genotype did not affect calcium uptake. Interestingly, water-limited conditions increased the nutrient content in root and stems but not in leaves. Water-limited trees partitioned more nitrogen and calcium to roots, while well-watered trees in the control partitioned more nutrients to the stems. Fruit size was the largest for ‘G.890’ and smallest for ‘B.9’. Both ‘G.41’ and ‘G.890’ had higher bitter pit incidence, which was associated with higher potassium content in leaves and fruit. These results suggest that rootstock-induced vigor and irrigation can both contribute to nutrient imbalances in leaves and fruit that could affect the development of physiological disorders in ‘Honeycrisp’ apple.

Long non-coding RNAs (lncRNAs) hold gene regulatory potential, but require substantial further functional annotation in livestock. Applying two metabogenomic approaches by combining transcriptomic and metabolomic analyses, we aimed to identify lncRNAs with potential regulatory function for divergent nutrient partitioning of lactating crossbred cows and to establish metabogenomic interaction networks comprising metabolites, genes and lncRNAs. Through correlation analysis of lncRNA expression with transcriptomic and metabolomic data, we unraveled lncRNAs that have a putative regulatory role in energy and lipid metabolism, the urea and tricarboxylic acid cycles, and gluconeogenesis. Especially FGF21, which correlated with a plentitude of differentially expressed genes, differentially abundant metabolites, as well as lncRNAs, suggested itself as a key metabolic regulator. Our objective was to determine whether abomasal infusions of increasing doses of oleic acid (cis -9 C18:1; OA) improved fatty acid (FA) digestibility and milk production of lactating dairy cows. Eight rumen-cannulated multiparous Holstein cows (138 d in milk ± 52) were randomly assigned to treatment sequence in a replicated 4 × 4 Latin square design with 18-d periods consisting of 7 d of washout and 11 d of infusion. Production and digestibility data were collected during the last 4 d of each infusion period. Treatments were 0, 20, 40, or 60 g/d of OA. We dissolved OA in ethanol before infusions. The infusate solution was divided into 4 equal infusions per day, occurring every 6 h, delivering the daily cis -9 C18:1 for each treatment. Animals received the same diet throughout the study, which contained (percent diet dry matter) 28% neutral detergent fiber, 17% crude protein, 27% starch, and 3.3% FA (including 1.8% FA from a saturated FA supplement containing 32% C16:0 and 52% C18:0). Infusion of OA did not affect intake or digestibility of dry matter and neutral detergent fiber. Increasing OA from 0 to 60 g/d linearly increased the digestibility of total FA (8.40 percentage units), 16-carbon FA (8.30 percentage units), and 18-carbon FA (8.60 percentage units). Therefore, increasing OA linearly increased absorbed total FA (162 g/d), 16-carbon FA (26.0 g/d), and 18-carbon FA (127 g/d). Increasing OA linearly increased milk yield (4.30 kg/d), milk fat yield (0.10 kg/d), milk lactose yield (0.22 kg/d), 3.5% fat-corrected milk (3.90 kg/d), and energy-corrected milk (3.70 kg/d) and tended to increase milk protein yield. Increasing OA did not affect the yield of mixed milk FA but increased yield of preformed milk FA (65.0 g/d) and tended to increase the yield of de novo milk FA. Increasing OA quadratically increased plasma insulin concentration with an increase of 0.18 μg/L at 40 g/d OA, and linearly increased the content of cis -9 C18:1 in plasma triglycerides by 2.82 g/100 g. In conclusion, OA infusion increased FA digestibility and absorption, milk fat yield, and circulating insulin without negatively affecting dry matter intake. In our short-term infusion study, most of the digestion and production measurements responded linearly, indicating that 60 g/d OA was the best dose. Lignin-based microcapsules are extremely attractive for their biodegradability and photolysis resistance. However, the water-soluble all-lignin shells were unsatisfactory in terms of rainfall and foliar retention, and lacked the test of agricultural production practices. Herein, a novel microcapsule based on a flexible skeleton formed by interfacial polymerization and absorbed with lignin particles (LPMCs) was prepared in this study. Further analysis demonstrated that the shell was formed by cross-linking the two materials in layers and showed excellent flexibility and photolysis resistance. The pesticide loaded LPMCs showed about 98.68 % and 73.00 % improvement in scour resistance and photolysis resistance, respectively, as compared to the bare active ingredient. The foliar retention performance of LPMCs was tested in peanut plantations during the rainy season. LPMCs loaded with pyraclostrobin (Pyr) and tebuconazole (Teb) exhibited the best foliar disease control and optimum plant architecture, resulting in an increase in yield of about 5.36 %. LPMCs have a promising application prospect in the efficient pesticide utilization, by controlling its deformation, adhesion and release, an effective strategy for controlling diseases and managing plant growth was developed. Tirzepatide, a glucose-dependent insulinotropic polypeptide/glucagon-like peptide 1 receptor (GIPR/GLP-1R) agonist, has, in clinical trials, demonstrated greater reductions in glucose, body weight, and triglyceride levels compared with selective GLP-1R agonists in people with type 2 diabetes (T2D). However, cellular mechanisms by which GIPR agonism may contribute to these improved efficacy outcomes have not been fully defined. Using human adipocyte and mouse models, we investigated how long-acting GIPR agonists regulate fasted and fed adipocyte functions. In functional assays, GIPR agonism enhanced insulin signaling, augmented glucose uptake, and increased the conversion of glucose to glycerol in a cooperative manner with insulin; however, in the absence of insulin, GIPR agonists increased lipolysis. In diet-induced obese mice treated with a long-acting GIPR agonist, circulating triglyceride levels were reduced during oral lipid challenge, and lipoprotein-derived fatty acid uptake into adipose tissue was increased. Our findings support a model for long-acting GIPR agonists to modulate both fasted and fed adipose tissue function differentially by cooperating with insulin to augment glucose and lipid clearance in the fed state while enhancing lipid release when insulin levels are reduced in the fasted state. As milk production in dairy cattle continues to increase, so do the energetic and nutrient demands on the dairy cow. Difficulties making the necessary metabolic adjustments for lactation can impair lactation performance and increase the risk of metabolic disorders. The physiological adaptations to lactation involve the mammary gland and extramammary tissues that coordinately enhance the availability of precursors for milk synthesis. Changes in whole-body metabolism and nutrient partitioning are accomplished, in part, through the bioenergetic and biosynthetic capacity of the mitochondria, providing energy and diverting important substrates, such as AA and fatty acids, to the mammary gland in support of lactation. With increased oxidative capacity and ATP production, reactive oxygen species production in mitochondria may be altered. Imbalances between oxidant production and antioxidant activity can lead to oxidative damage to cellular structures and contribute to disease. Thus, mitochondria are tasked with meeting the energy needs of the cell and minimizing oxidative stress. Mitochondrial function is regulated in concert with cellular metabolism by the nucleus. With only a small number of genes present within the mitochondrial genome, many genes regulating mitochondrial function are housed in nuclear DNA. This review describes the involvement of mitochondria in coordinating tissue-specific metabolic adaptations across lactation in dairy cattle and the current state of knowledge regarding mitochondrial-nuclear signaling pathways that regulate mitochondrial proliferation and function in response to shifting cellular energy need. The objective of our study was to determine the effects of altering the ratio of stearic (C18:0; SA) and oleic (cis -9 C18:1; OA) acids in supplemental fatty acid (FA) blends on FA digestibility and milk yield of dairy cows. Eight multiparous Holstein cows (mean ± SD; 157 ± 11.8 d in milk) were randomly assigned to treatment sequence in a replicated 4 × 4 Latin square design with 14-d periods. Digestibility and production data were collected during the last 4 d of each period. The treatments were an unsupplemented control diet (CON), and 3 diets incorporating FA supplement blends at 1.4% of diet dry matter (DM) containing (as a % of total FA) 50% SA and 10% OA, 40% SA and 20% OA, or 30% SA and 30% OA. The FA blends were balanced to contain 33% palmitic, 5% linoleic, and <0.5% linolenic acids. The FA supplements replaced soyhulls in the CON diet. Preplanned contrasts were as follows: (1) overall effect of FA treatments [CON vs. the average of the FA-supplemented diets; (50:10 + 40:20 + 30:30)/3], (2) the linear effect of OA inclusion in the supplemental FA blend, and (3) the quadratic effect of OA inclusion in the supplemental FA blend. There was no effect of treatment on DM intake, but the replacement of soyhulls in the FA treatments decreased neutral detergent fiber intake. Overall, compared with CON, FA treatments increased DM and neutral detergent fiber digestibility, and increasing OA within FA treatments quadratically increased digestibility of DM and neutral detergent fiber. Overall, FA treatments increased the intake of total, 16-carbon, and 18-carbon FA, decreased the digestibility of total and 18-carbon FA, but increased absorption of total, 16-carbon, and 18-carbon FA. Within FA treatments, increasing OA linearly increased the digestibility of total, 16-carbon, and 18-carbon FA, as well as the absorption of total, 16-carbon, and 18-carbon FA. Overall, FA treatments increased the yields of milk, energy-corrected milk, and milk fat, and tended to increase milk protein yield. Compared with CON, FA treatments had no effect on the yield of de novo milk FA and increased the yields of mixed and preformed milk FA. Within FA treatments, increasing OA did not affect the yields of milk or milk components, linearly decreased the yield of de novo FA, and quadratically affected the yield of mixed and preformed milk FA. Overall, FA treatments increased plasma nonesterified fatty acids but did not affect β-hydroxybutyrate or insulin. Within FA treatments, increasing OA quadratically affected plasma nonesterified fatty acids, and tended to linearly increase β-hydroxybutyrate and quadratically affect insulin. In conclusion, supplemental FA blends containing different ratios of SA and OA did not affect DM intake but increased the yields of milk and milk components. Supplemental FA blends also increased digestibility of DM and neutral detergent fiber and decreased digestibility of total and 18-carbon FA compared with CON. Although increasing OA within FA supplements did not alter milk production, increasing OA within FA supplements increased total, 16-carbon, and 18-carbon FA digestibility and FA absorption. Further research is required to determine longer term effects of SA and OA on nutrient digestion and partitioning and opportunities for maintaining or improving FA digestibility with increasing SA intake and availability in the small intestine. Sugarcane being a heavy biomass producer needs enormous quantity of nutrients. Soil has to be replenished with the quantities of nutrients utilized by the crop to maintain the productivity as well as soil quality. Partitioning of nutrients (N, P and K) in the aerial parts of sugarcane genotypes entered in the advanced varietal trial (AVT) during 2015–2016, and their nutrient use efficiencies were assessed. Among Early genotypes, Co 10006 accumulated the highest proportion of N (53%) and K (60%) in tops and trash. In Mid-late category, Co 86032 accumulated the highest proportion of N and P in tops and trash, while Co 10017 accumulated the lowest proportion of N, P and K in tops and trash. P and K accumulation in foliage of Mid-late genotypes was more than 70%. The nutrient-rich tops and trash can be effectively utilized/recycled in the farm to reduce nutrient loss. Among Early genotypes, Co 10027 was N- and K-efficient utilizer and required 1.27 and 1.47 kg of N and K, respectively, for one tonne cane production and for P, Co 10026 was the most efficient genotype which required 0.158 kg P for one tonne cane. Among Mid-late genotypes, the genotype Co 10017 was found to be efficient in terms of NPK use and required 1.73, 0.147 and 1.39 kg of N, P and K, respectively, for one tonne cane production. Hence, cultivation of nutrient-efficient advanced generation sugarcane genotypes will reduce the pressure on the resources or they may be used as parents in breeding to develop efficient varieties. Key Message: Nutrient uptake in the biomass of healthy and root (wilt) disease affected coconut palms are different. Root (wilt) is a predominant phytoplasmal disease affecting coconut (Cocos nucifera L.) palms and to sustain the palm health and productivity, better nutrient management practices are required. The objective of this research was to understand the uptake pattern and the partitioning of nutrients in the total biomass of healthy and root (wilt) diseased palms grown in humid tropical Entisol of Kerala. 5 coconut palms each were selected from diseased palms viz., Early (DE), Middle (DM) and Advanced (DA) category along with the control, which is Apparently Healthy palm (AH), without any diseases. The total biomass component with respect to each part viz., leaf (L), petiole (P), spadix (S), spathe (Sp), stem (St), leaf sheath fiber (LSF), husk (H) and nuts (copra:C + shell (Sh)) (N) were estimated. Nutrient uptake by the entire biomass uptake is classified as 1. Recyclable portion (L, P, S, Sp, H, LSF and Sh) 2. Removable portion (C) 3. Reserve biomass (St) and then computed the nutrient partitioning by standard analytical procedures. For all nutrients except Mg, reserve biomass had the greatest nutrient uptake followed by recyclable biomass and removable biomass components. Among the nutrients, potassium recorded the highest total uptake. In AH palm, the total uptake of potassium was 1075 g per palm, whereas it was 407.3 g per palm in the diseased palms. The total uptake of nutrients which showed a significant relation with disease index followed the order K > N > Ca > S > P > Mg in apparently healthy palms and N > K > Ca > S > P > Mg in diseased palms. The percentage reduction in total uptake of N, P, K, Ca, Mg, S and B by diseased palms in comparison with healthy palms was 36.5, 37.6, 57.5, 44.1 23.3, 43.7 and 48.9. The results of the study indicated that the pattern of nutrient uptake in the different biomass components of apparently healthy and root (wilt) disease affected palms are significantly different. Reductions in insulin sensitivity in periparturient dairy cows develop as a means to support lactation; however, excessive mobilization of fatty acids (FA) increases the risk for peripartal metabolic disorders. Our objectives were to investigate the effect of prepartum body condition score (BCS) on systemic glucose and insulin tolerance, and to compare direct and indirect measurements of insulin sensitivity in peripartal lean and overweight dairy cows. Fourteen multiparous Holstein cows were allocated into two groups according to their BCS at day −28 prepartum: lean (n = 7; BCS ≤ 3.0) or overweight; (n = 7; BCS ≥ 4.0). Liver biopsies were performed on day −27, −14 and 4, relative to expected parturition. Intravenous insulin or glucose tolerances tests were performed following each liver biopsy. Relative to lean cows, overweight cows exhibited lower dry matter intake, lost more BCS and displayed increased plasma FA and β-hydroxybutyrate concentrations and elevated liver lipid content during peripartum. Glucose clearance rate was lower for all cows postpartum. Prepartum BCS had minimal effects on insulin and glucose tolerance; however, the ability of the cow to restore blood glucose levels following an insulin challenge was suppressed by increased BCS. Glucose-dependent parameters of insulin and glucose tolerance were not correlated with surrogate indices of insulin sensitivity. We conclude that prepartum BCS had minimal effect on systemic insulin sensitivity following parturition. The observed inconsistency between surrogate indices of insulin sensitivity and direct measurements of insulin and glucose tolerance adds support to growing concerns regarding their usefulness as tools to estimate systemic insulin action in periparturient cows.

Rice, a staple food worldwide, contains varying amounts of nutrients in different grain tissues. The underlying molecular mechanism of such distinct nutrient partitioning remains poorly investigated. Here, an optimized rapid laser capture microdissection (LCM) approach was used to individually collect pericarp, aleurone, embryo and endosperm from grains 10 days after fertilization. Subsequent RNA-Seq analysis in these tissues identified 7760 differentially expressed genes. Analysis of promoter sequences of tissue-specific genes identified many known and novel cis -elements important for grain filling and seed development. Using the identified differentially expressed genes, comprehensive spatial gene expression pathways were built for accumulation of starch, proteins, lipids, and iron. The extensive transcriptomic analysis provided novel insights about nutrient partitioning mechanisms; for example, it revealed a gradient in seed storage protein accumulation across the four tissue types analysed. The analysis also revealed that the partitioning of various minerals, such as iron, is most likely regulated through transcriptional control of their transporters. We present the extensive analysis from this study as an interactive online tool that provides a much-needed resource for future functional genomics studies aimed to improve grain quality and seed development. The objective of this study was to determinate dry matter production and macronutrients accumulation in the shoots of soybean plants of determinate and indeterminate types of growth. The experiment was carried out in Dourados, Mato Grosso do Sul, Brazil, in the 2016/17 season. The experimental design was a random block, with two soybean cultivars (BRS 317 determinate and BRS 1001 indeterminate), seven collection times (15, 30, 45, 60, 75, 90 and 105 days after emergence) and four replicates. At each collection, the plants were divided into leaves, stems, branches, pods and grains, and then submitted to laboratory analysis to determine the nutrients contents in each organ. Macronutrient accumulation in both cultivars followed the order: N > K > Ca > P>Mg > S. The indeterminate cultivar produced more dry matter, and had a longer time required for maximum production, than the determinate cultivar. The indeterminate cultivar exported more nutrients than did the determinate cultivar. Thus, the former demanded more attention to replacement of nutrients to the soil. [ABSTRACT FROM AUTHOR]

This experiment aimed to examine endocrine and metabolic responses to glucose, insulin, and adrenocorticotropin (ACTH) infusions in early-lactation dairy goats of different levels of milk production (LMP). Goats were grouped as either high (HY; 4.0 L/d, n = 13) or low milk yield (LY; 2.4 L/d, n = 13). Individual milk yield (L/d) and dry matter intake (DMI; kg/d) were measured daily. Concentration (m M) of glucose, fatty acids, and β-hydroxybutyrate, percent of milk fat and protein, body weight (BW; kg), and body condition score (BCS) were assessed weekly (from 2–6 wk postpartum). An intravenous glucose tolerance test (IVGTT), an insulin tolerance test (ITT), and an ACTH stimulation test were carried out at 43, 44, and 45 ± 0.7 d in milk, respectively. The HY goats had greater milk yield (+67%), energy-corrected milk (ECM; +70%), DMI (+28%), ratio of ECM output to metabolic BW (+67%), and feed efficiency (+25%), but lesser BCS than LY goats (2.4 vs. 2.6). The DMI (% of BW) was moderately correlated with ECM (r = 0.70) and negatively correlated with BCS (r = −0.57). At the time of the IVGTT, HY goats had lesser basal insulin and glucose than LY goats. However, results from IVGTT and ITT indicate that the sensitivity of peripheral tissues to insulin was unaffected by LMP. Compared with LY, HY goats had lesser insulin secretion (−52%) and greater insulin clearance rate (+47%) after glucose infusion. The ITT and ACTH stimulation test results show that both the growth hormone response to insulin and the cortisol response to ACTH were unaffected by LMP. Also, basal plasma concentrations of GH and cortisol were not correlated with glucose and fatty acids concentrations or any performance traits. Collectively, our results suggest that differences between HY and LY goats, concerning milk yield and feed efficiency, were probably more closely related to differences in insulin secretion and clearance than to differences in peripheral tissue responsiveness to the effects of catabolic and anabolic hormones. Pregnancy is a remarkable physiological state, during which the metabolic system of the mother adapts to ensure that nutrients are made available for transfer to the fetus for growth and development. Adaptations of maternal metabolism during pregnancy are influenced by the metabolic and nutritional status of the mother and the production of endocrine factors by the placenta that exert metabolic effects. Insufficient or inappropriate adaptations in maternal metabolism during pregnancy may lead to pregnancy complications with important short- and long-term effects for both the health of the child and mother. This is very evident in gestational diabetes, which is marked by greater glucose intolerance and insulin resistance above that expected of a normal pregnancy. Gestational diabetes is associated with increased fetal weight and/or increased adiposity, higher instrumented delivery rates and greater risks for both mother and child of developing type 2 diabetes in the long-term. However, despite the negative health impacts of such metabolic imbalances during pregnancy, the precise mechanisms responsible for orchestrating these changes remain largely unknown. The present review describes the dynamic pregnancy-specific changes that occur in the metabolic system of the mother during pregnancy. It also discusses findings using surgical, pharmacological, genetic and dietary methods in experimental animals that highlight the role of pathways in maternal tissues that lead to metabolic dysfunction, with a particular focus on gestational diabetes. Finally, it summarises the work largely employing gene targeting and hormone administration in rodents that have illuminated the involvement of placental endocrine function in driving maternal metabolic adaptations. While current animal models may not fully replicate what is observed in humans, these have been instrumental in showing that there is a dynamic interplay between changes in maternal metabolic physiology and the placental production of endocrine factors that govern the availability of nutrients to the growing fetus. However, more work is required to specifically identify the placenta-driven changes in maternal metabolic physiology that ensure the appropriate level of insulin production and action during pregnancy. In doing so, these studies may pave the way to understanding the development of pregnancy complications like gestational diabetes, as well as further our understanding of type-2 diabetes and the control of metabolic physiology more broadly. The current nutrient recommendations focus on pigs fed ad libitum up to 140 kg in body weight (BW). It remains unclear whether this applies to pigs weighing above 140 kg in BW under different rearing conditions. This study aimed to estimate protein (Pd) and lipid (Ld) depositions and the metabolizable energy (ME), standardized ileal digestible lysine (SID lysine) requirement and partitioning in 224 C21 Goland pigs (90–200 kg in BW). The control pigs (C) received diets limiting ME up to 170 kg in slaughter weight (SW) at 9 months of age (SA); older (OA) pigs had restricted diets limiting ME and SID lysine up to 170 kg in SW at >9 months SA; younger (YA) pigs were fed nonlimited amounts of ME and SID lysine up to 170 kg in SW at 170 kg in SW. The estimated MEm averaged 1.03 MJ/kg0.60. An 11% increase in MEm was observed in OA pigs compared to the controls. Energy restriction had negligible effects on the estimated MEm. The marginal efficiency of SID lysine utilization for Pd averaged 0.725, corresponding to a SID lysine requirement of 9.8 g/100 g Pd.

Recent advances in oil-palm breeding and agronomic practices may have influenced the dry-matter production and nutrient demand of tenera oil palm since the last comprehensive studies published some three decades ago. This has raised concerns, since updated knowledge of nutrient requirements at various stages of oil palm growth is essential for formulating fertilizer recommendations that meet agronomic, economic, and environmental objectives. We therefore filled in this knowledge gap with the objective to quantify the standing biomass, dry-matter production, and nutrient demand of tenera oil palm of different ages, grown under current agronomic practices. Tenera palms were sampled at 12, 29, 48, 88, 133, 209, and 238 months since nursery planting and separated into spear leaves, leaflets, rachis, petiole, leaf bases, cabbage, trunk, bole, and roots attached to bole for determination of dry weights and nutrient contents. Fruit bunches were harvested, with dry weights and nutrient contents of ripe bunches determined. Annual dry-matter production was calculated from dry weights of different plant parts, and together with nutrient concentrations determined in these plant parts and fruit bunches, annual nutrient demand was calculated. Excluding roots, the total standing vegetative biomass increased almost linearly from 1.3 kg palm−1 at 12 months old to 808.6 kg palm−1 at 238 months old. Whole-palm dry-matter production increased with palm age but at a decreasing rate, from 1.5 kg palm−1 year−1 at 12 months old to 285.5 kg palm−1 year−1 after 133 months, with little increase thereafter. The maximum rates of nutrient demand occurred after 209 months, coinciding with peak production of fruit bunches—except for K, which occurred at month 88 when vegetative growth demand for K peaked. Annual gross amounts of nutrients required to produce 25 t fresh fruit bunches ha−1 were 1.99 kg N, 0.28 kg P, 3.94 kg K, 0.42 kg Mg, 0.99 kg Ca, and 2.5 g B per palm. The net amounts of nutrients required to meet similar production level of fruit bunches were 1.04 kg N, 0.16 kg P, 1.85 kg K, 0.26 kg Mg, 0.47 kg Ca, and 1.3 g B per palm per year if all nutrients contained in the pruned leaves were recycled. These results provide much-needed updated data on the dry-matter production and nutrient demand of tenera oil palm grown under current agronomic practices and also serve as a general yardstick for practitioners to further refine fertilizer recommendations.

Matching fertilization with crop needs is important for maximizing yields and reducing fertilizer losses. Seasonal variation in nutrient uptake dynamics is poorly understood and thus, the ability to optimize fertilization strategies is limited. This study aims to investigate the effects of planting dates on macronutrient uptake dynamics in cassava genotypes with full irrigation. The performance of cassava genotypes, i.e., CMR38-125-77, Kasetsart 50 and Rayong 11, were evaluated in the early rainy (ERS) and post rainy seasons (PRS) for two years using a randomized complete block design with four replicates. The plants were harvested at 1, 3, 6, 9 and 12 months. Planting dates had significant effects on the accumulation of dry matter and storage roots as well as nutrient uptakes and partitioning. On average, the total nutrient uptake per plant to produce 2831–3279 g of biomass with 1244–1810 g of storage roots in the ERS varied among cassava genotypes, ranging from 21.1–24.3 g N, 5.1–5.9 g P, 26.5–29.5 g K, 14.1–22.2 g Ca, 6.1–7.6 g Mg and 2.0–2.3 g S. The total nutrient uptake per plant to produce 3353–3824 g of biomass with 1604–2253 g of storage roots in the PRS ranged from 27.1–32.4 g N, 5.2–6.0 g P, 29.1–31.3 g K, 11.9–20.3 g Ca, 7.3–9.9 g Mg and 1.2–1.5 g S. In the ERS, the majority of the total nutrient uptake occurred at the early growth stages, whereas in the PRS, this occurred at the mid- to late growth stages. At final harvest, the percentages of nutrient removal by the storage roots for ERS were 24.7–36.0% N, 26.0–32.3% P, 43.4–51.5% K, 12.4–17.6% Ca, 22.2–31.5% Mg and 27.2–31.5% S, whereas in the PRS the percentages were 30.4–44.4% N, 33.3–41.6% P, 44.7–57.3% K, 12.0–15.1% Ca, 20.2–28.1% Mg and 12.0–25.4% S. CMR38-125-77 exhibited satisfactory performance in nutrient uptake, nutrient use efficiency and storage roots yield across the planting dates. The evidence obtained from this study would greatly facilitate more efficient adoption of precision agriculture in cassava production by applying recommended fertilizers, e.g., rates, kinds and timings, according to crop demand in each growing season in Thailand and for choosing superior cassava genotypes.

ABSTRACT African mahogany is a species preferred by many entrepreneurs as an investment in the forestry sector. However, there are few studies or research on the species, especially in Brazil. The aim of this work was to study the efficiency of nutrient uptake, accumulation, distribution and use in Khaya ivorensis and Khaya senegalensis, in addition to aspects of growth variables and of dry matter partitioning as a function of mineral fertilisation. Morphological, physiological and nutritional parameters were determined in the African mahogany plants 140 days after transplanting (DAT). The greatest zinc (Zn) content was detected in the shoots of both the Khaya ivorensis and Khaya senegalensis plants, accounting for more than 50% of the accumulated total of the plants with added nutrient solution. Maintaining the plants of Khaya senegalensis and Khaya ivorensis with the use of nutrient solution provides the best conditions for nutrient use, and allows plants with excellent morphological and nutritional characteristics to be ready for the field within five months after transplanting in a shaded nursery. The most efficient uptake, transport and use of macro and micronutrients is found in the species Khaya senegalensis with the addition of nutrient solution.

The objective was to investigate effects of feeding lambs fresh versus dried Puccinellia. tenuiflora (Griesb.) Scribn. & Merr. on nutrient and water intake and apparent digestibility. Twenty‐four lambs were offered fresh or dried (48 hr) grass. The dry matter (DM) content of fresh grass was 65% of dried grass and the neutral detergent fiber exclude ash (aNDFom) content of fresh grass was 87% of dried grass, whereas the metabolizable energy concentration of two forages was similar. Lambs eating fresh grass had higher (p <.05) DM intake (944 versus 837 g DM day−1 sheep−1), metabolizable energy intake (9.1 versus 7.9 MJ day‐1sheep−1), and crude protein intake (86 versus 75 g day‐1 sheep−1) than those eating dried grass. However, there was no significant difference between treatments in drinking water intake, fecal nutrient output or nutrient digestibility. Due to insufficient dry matter intake and heightened nutrient requirements in early lactation, periparturient dairy cows mobilize adipose and muscle tissues to bridge energy and amino acid gaps, respectively. Our objective was to evaluate the relationship between the relative muscle thickness of late pregnant cows and their early lactation performance. At 35 d before expected calving (BEC), longissimus dorsi muscle thickness (LDT) was measured in forty-one multiparous Holstein cows via ultrasound. Tissue mobilization was evaluated via ultrasound images of LDT and backfat thickness (BFT) at 21 and 7 d BEC as well as at 0, 10, 30, and 60 DIM. Plasma concentrations of 3-methylhistidine (3-MH), creatinine (CRE), non-esterified fatty acids (NEFA), and β-hydroxybutyrate (BHB) were evaluated weekly. Milk yield and milk component data were collected through 60 DIM. Cattle were assigned post hoc to high-muscle (HM; n = 20; LDT > 4.49 cm) or low-muscle (LM; n = 21; ≤4.37 cm) groups, with mean LDT at 35 d BEC greater in HM (5.05 ± 0.49) than in LM (3.52 ± 0.65) animals. Between 35 and 21 d BEC, LM cows gained LDT, whereas HM cows gained BFT. HM cows mobilized more muscle from 21 d BEC to 30 DIM, as reflected by a greater loss of LDT, greater 3-MH concentrations (532 vs. 438 ± 30 ng/mL), and a greater 3-MH:CRE ratio (0.164 vs. 0.131 ± 0.008) in the first three weeks postpartum. The LDT and BFT at 21 d BEC were related to the amount of respective tissue mobilized through 30 DIM (R2 = 0.37 and 0.88, respectively). Although calves born to HM cattle were larger (45.2 vs. 41.8 ± 0.7 kg), HM cows produced less milk (38.8 vs. 41.6 ± 0.8 kg/d) with a tendency towards higher fat content (4.33 vs. 4.05 ± 0.12%), likely related to the mobilization of more backfat from 0 to 60 DIM (1.78 vs. 0.68 ± 0.34 mm), compared to LM cattle. These findings suggest that a cow’s metabolic status, as measured by LDT and BFT prepartum, may influence the metabolic strategy the animal uses to meet energy and amino acid requirements in late gestation and early lactation.

The objective of our study was to evaluate the effects of altering the dietary ratio of palmitic (C16:0) and oleic (cis -9 C18:1) acids on nutrient digestibility, energy partitioning, and production responses of lactating dairy cows. Cows were blocked by milk yield and assigned to 3 groups (12 cows per group) in a main plot: low (45.2 ± 1.7 kg/d), medium (53.0 ± 1.6 kg/d), and high (60.0 ± 1.9 kg/d). Within each production group, a truncated Latin square arrangement of fatty acid (FA) treatments was used in 2 consecutive 35-d periods. The FA treatments supplemented at 1.5% of diet dry matter were (1) 80:10 (80% C16:0 + 10% cis -9 C18:1), (2) 73:17 (73% C16:0 + 17% cis -9 C18:1), (3) 66:24 (66% C16:0 + 24% cis -9 C18:1), and (4) 60:30 (60% C16:0 + 30% cis -9 C18:1). Treatment × production group interactions were observed for yields of milk, fat-corrected milk, energy-corrected milk, milk fat, milk protein, and milk lactose and energy partitioned to milk. Increasing cis -9 C18:1 in FA treatments reduced fat-corrected milk, energy-corrected milk, and milk energy output in low-producing cows but increased these in high-producing cows. Increasing cis -9 C18:1 in FA treatments did not affect milk yield, milk protein yield, and milk lactose yield in low- and medium-producing cows but increased these in high-producing cows. Regardless of production level, there was no effect of treatments on dry matter intake; however, increasing cis -9 C18:1 in FA treatments increased body weight change and body condition score change. Increasing cis -9 C18:1 in FA treatments increased total FA digestibility due to a linear increase in 16- and 18-carbon FA digestibilities. Interactions between FA treatments and production level were observed for the yield of milk fat and milk FA sources. In low-producing cows, increasing cis -9 C18:1 in FA treatments decreased milk fat yield due to a decrease in de novo and mixed milk FA without changes in preformed milk FA. In contrast, in high-producing cows, increasing cis -9 C18:1 in FA treatments increased milk fat yield due to an increase in de novo and preformed milk FA. Our results indicate that high-producing dairy cows (averaging 60 kg/d) responded better to a fat supplement containing more cis -9 C18:1, whereas low-producing cows (averaging 45 kg/d) responded better to a supplement containing more C16:0. Reductions in insulin sensitivity in periparturient dairy cows develop as a means to support lactation; however, excessive mobilization of fatty acids (FA) increases the risk for peripartal metabolic disorders. Our objectives were to investigate the effect of prepartum body condition score (BCS) on systemic glucose and insulin tolerance, and to compare direct and indirect measurements of insulin sensitivity in peripartal lean and overweight dairy cows. Fourteen multiparous Holstein cows were allocated into two groups according to their BCS at day −28 prepartum: lean (n = 7; BCS ≤ 3.0) or overweight; (n = 7; BCS ≥ 4.0). Liver biopsies were performed on day −27, −14 and 4, relative to expected parturition. Intravenous insulin or glucose tolerances tests were performed following each liver biopsy. Relative to lean cows, overweight cows exhibited lower dry matter intake, lost more BCS and displayed increased plasma FA and β-hydroxybutyrate concentrations and elevated liver lipid content during peripartum. Glucose clearance rate was lower for all cows postpartum. Prepartum BCS had minimal effects on insulin and glucose tolerance; however, the ability of the cow to restore blood glucose levels following an insulin challenge was suppressed by increased BCS. Glucose-dependent parameters of insulin and glucose tolerance were not correlated with surrogate indices of insulin sensitivity. We conclude that prepartum BCS had minimal effect on systemic insulin sensitivity following parturition. The observed inconsistency between surrogate indices of insulin sensitivity and direct measurements of insulin and glucose tolerance adds support to growing concerns regarding their usefulness as tools to estimate systemic insulin action in periparturient cows. Water stress and sulfur (S) deficiency are two constraints increasingly faced by crops due to climate change and low-input agricultural practices. To investigate their interaction in the grain legume pea (Pisum sativum), sulfate was depleted at the mid-vegetative stage and a moderate 9-d water stress period was imposed during the early reproductive phase. The combination of the stresses impeded reproductive processes in a synergistic manner, reducing seed weight and seed number, and inducing seed abortion, which highlighted the paramount importance of sulfur for maintaining seed yield components under water stress. On the other hand, the moderate water stress mitigated the negative effect of sulfur deficiency on the accumulation of S-rich globulins (11S) in seeds, probably due to a lower seed sink strength for nitrogen, enabling a readjustment of the ratio of S-poor (7S) to 11S globulins. Transcriptome analysis of developing seeds at the end of the combined stress period indicated that similar biological processes were regulated in response to sulfur deficiency and to the combined stress, but that the extent of the transcriptional regulation was greater under sulfur deficiency. Seeds from plants subjected to the combined stresses showed a specific up-regulation of a set of transcription factor and SUMO ligase genes, indicating the establishment of unique regulatory processes when sulfur deficiency is combined with water stress. The California Net Energy System (CNES) has been successfully used for many years to generate estimates of grazing animal energy requirements, supplemental needs, and energy value of grazed forage diets. Compared to pen feeding situations, validation of feed nutritive value estimates or animal performance projections are extremely difficult in grazing animals because many of the system inputs are constantly changing. A major difficulty in applying this or any energy accounting system in the field is acquiring accurate estimates of forage intake. We discuss the various equations available to estimate forage intake for grazing animals with emphasis on beef cows. Progress has been made in recent years although there remains substantial discrepancy among various equations, particularly in the upper range of forage digestibility. Validation work and further development is needed in this area. For lactating cows, our conclusion is that the adjustment of intake for milk production (0.2 kg increase in forage intake per kg of milk produced) needs to be increased to a minimum of 0.35. A particular challenge with the CNES for grazing beef cows is the dramatic interaction that can occur between genetic potential for production traits and nutrient availability. Examples from literature are provided and a case study is presented demonstrating that energy requirements are dynamic and depend on nutrients available in grazing systems. The CNES is a useful tool in grazing beef cattle management although there remains substantial opportunity and need to improve inputs and validate the system in grazing situations. Responses of dairy cows with high or low milk yield (MY) beyond 450 d in milk (DIM) to 3 metabolic challenges were investigated. Twelve multiparous Holstein-Friesian cows that calved in late winter in a pasture-based system were managed for a 670-d lactation by delaying re-breeding. Cows were selected for either high MY (18.9 ± 1.69 L/cow per d; n = 6) or low MY (12.3 ± 3.85 L/cow per d; n = 6) at 450 DIM. Cows were housed indoors for 2 periods of 12 d at approximately 460 and 580 DIM. Each cow was fed freshly cut pasture (460 DIM) or pasture silage (580 DIM) plus 6.0 kg of DM barley grain daily (approximately 200 MJ of total metabolizable energy/cow per day). At all other times, cows were managed as a single herd and grazed pasture supplemented with cereal grain to an estimated intake of 180 MJ of metabolizable energy/cow per d. Cows were fitted with a jugular catheter during the final week of each experimental period. Over a period of 3 d, each cow underwent an intravenous glucose tolerance test (0.3 g/kg of body weight), an insulin tolerance test (0.12 IU of insulin/kg of body weight), and a 2-dose epinephrine challenge (0.1 and 1.6 µg/kg of body weight). Cows selected for high MY had greater milk and milk solids yields between 450 and 580 DIM than low MY cows (17.3 vs. 10.8 ± 1.49 kg of milk/d and 2.4 vs. 1.5 ± 0.23 kg of milk solids/d). The results indicated that whole body and peripheral tissue responsiveness to insulin may vary between cows of high and low MY. Following the glucose tolerance test, high MY cows had a lower plasma insulin response with a greater glucose area under the curve than low MY cows. Further, high MY cows had slower plasma glucose clearance compared with low MY cows during an insulin tolerance test. The plasma nonesterified fatty acid (NEFA) responses to the IVGTT and the ITT were similar between cows of high and low MY, but the clearance of NEFA from the plasma following both the IVGTT and ITT were slower at 580 compared with 460 DIM. The sensitivity to epinephrine was greater in high MY cows compared with low MY cows as the glucose and NEFA area under the curve and the percentage change in NEFA were greater in high MY after the low dose epinephrine challenge. However, the lipolytic but not the glucose appearance in response to epinephrine was greater in high MY cows than low MY cows. Following the high dose of epinephrine, the glucose response was lower, but the NEFA response was greater in high MY compared with low MY cows. Cows able to sustain greater MY to 580 DIM had a greater propensity for lipid mobilization, possibly enhancing nutrient partitioning to the mammary gland during the late stages of an extended lactation. Cotton is a vital crop worldwide, and as increases in population and changes in climate occur, it is critical for breeders to continue to improve lint yields. One area to further investigate is how nutrient uptake and nutrient use efficiencies have changed through time to develop future management and breeding strategies. To evaluate the change in nutrient uptake, nutrient use efficiency and yield of 20 cotton cultivars released in the USA between 1953 and 2018. A field experiment in 2020 and 2021 was carried out to evaluate the nutrient uptake, internal nutrient use efficiency and partitioning of assimilated nutrients among different components of biomass for the twenty cotton cultivars released between 1953 and 2018 in the USA. Linear regression analysis was performed to estimate the rate of gain for lint yield, crop biomass, total nutrient uptake and use efficiency. The rate of genetic gain for crop biomass increased 4.3 times faster than the rate of gain for lint yield. The total nutrient uptake and internal use efficiency were significantly different among cultivars, with a consistent increase measured across the span of years of release for the 20 cultivars. Nutrient concentration in vegetative growth either decreased or remained unchanged over time, while concentration in the seed increased. Modern cultivars demonstrate a 27% increase in efficiency, producing more lint per unit of nutrient uptake compared to those from the 1950 s, owing to improved nutrient availability with limited seed partitioning. Partitioning of total assimilated nutrients of N, P and K into the different biomass components were inconsistent over the years. Modern cotton cultivars have shown an increased total nutrient uptake but a slower improvement in nutrient use efficiency. The limited changes observed in the partitioning of primary nutrients for reproductive growth, along with the increased concentration of nutrients in seed, suggest potential focus for future advancements in breeding strategies. This research offers valuable perspectives on nutrient uptake, partitioning and use efficiency of cotton cultivars released between 1953 and 2018. Differences in nutrient use efficiency among cultivars were observed, highlighting the potential for enhanced nutrient uptake traits in cotton. Reproductive success is a key component of lifetime efficiency – which is the ratio of energy in milk (MJ) to energy intake (MJ) over the lifespan, of cows. At the animal level, breeding and feeding management can substantially impact milk yield, body condition and energy balance of cows, which are known as major contributors to reproductive failure in dairy cattle. This study extended an existing lifetime performance model to incorporate the impacts that performance changes due to changing breeding and feeding strategies have on the probability of reproducing and thereby on the productive lifespan, and thus allow the prediction of a cow’s lifetime efficiency. The model is dynamic and stochastic, with an individual cow being the unit modelled and one day being the unit of time. To evaluate the model, data from a French study including Holstein and Normande cows fed high-concentrate diets and data from a Scottish study including Holstein cows selected for high and average genetic merit for fat plus protein that were fed high- v. low-concentrate diets were used. Generally, the model consistently simulated productive and reproductive performance of various genotypes of cows across feeding systems. In the French data, the model adequately simulated the reproductive performance of Holsteins but significantly under-predicted that of Normande cows. In the Scottish data, conception to first service was comparably simulated, whereas interval traits were slightly under-predicted. Selection for greater milk production impaired the reproductive performance and lifespan but not lifetime efficiency. The definition of lifetime efficiency used in this model did not include associated costs or herd-level effects. Further works should include such economic indicators to allow more accurate simulation of lifetime profitability in different production scenarios.

This experiment investigated the metabolic response to a 2-dose epinephrine challenge of dairy cows undergoing an extended lactation. Twelve multiparous Holstein-Friesian cows that calved in late winter in a seasonally calving pasture-based dairying system were managed for a 670-d lactation by delaying rebreeding. In each of four 40-d experimental periods commencing at 73, 217, 422, and 520 (±9.1) d in milk (DIM), cows were offered a diet of perennial ryegrass (73 and 422 DIM) or pasture hay and silage (217 and 520 DIM), supplemented with 1 (CON; n = 6) or 6 kg of grain (GRN; n = 6) as a ration. Daily energy intake was approximately 160 and 215 MJ of metabolizable energy/cow for the CON and GRN treatments, respectively. At all other times, cows were managed as a single herd and grazed pasture supplemented with grain to an estimated daily total intake of 180 MJ of metabolizable energy/cow. Cows were fitted with a jugular catheter during the final week of each experimental period. Two doses of epinephrine (0.1 and 1.6 µg/kg of body weight) were infused via the catheter 2 h apart to each cow at approximately 100, 250, 460, and 560 DIM. Blood plasma concentrations of glucose and nonesterified fatty acids (NEFA) were measured before and after infusions. Cows in the GRN treatment had greater milk yield, milk fat and protein yields, and body weight than cows in the CON treatment. The maximum plasma glucose concentration was observed at 100 DIM for both the low and high doses of epinephrine. Thus, sensitivity and responsiveness to exogenous epinephrine were greater during early lactation, coinciding with increased priority of milk synthesis. Both the sensitivity and responsiveness to epinephrine decreased with decreasing milk yield, as measured by the acute appearance of NEFA in the plasma. Increased plasma glucose and NEFA clearance rates before 300 DIM indicated greater uptake of these substrates by the mammary gland for milk synthesis in early and mid lactation. These results support previous findings that major changes occur in terms of adipose tissue metabolism during extended lactations. Overall, sensitivity to epinephrine was not affected by diet, but responsiveness was greater in cows fed the GRN diet. The endocrine regulation of nutrient partitioning throughout traditional and extended lactations is complex, with many interactions between stage of lactation, diet, and milk yield potential. ABSTRACT For the development of nutrient budget models to recommend lime and fertilizers for agricultural and forestry crops, curves of plant growth and nutrient accumulation are required. Information about how nutrients are partitioned between the different plant organs is also necessary, but still scarce for pine in Brazil. This study evaluated the growth, biomass partitioning, and nutrient dynamics in pine forests in southern Brazil. To this end, we assessed unthinned 2, 4, 6, and 8-year-old stands of Pinus taeda L. Three plots of 20 × 30 m per stand were delimited, in which three trees of different classes of diameter at breast height (DBH) were chosen. These trees were measured, felled, and the weight of their fresh components (leaves, branches, bark, and wood) was evaluated. Samples of each tree compartment and from the forest litter were taken to determine dry weight and nutrient content. From trees of the mean DBH class, the roots were also collected and the dry weight and nutrient contents determined. The same sampling procedure was carried out with soil for physical and chemical characterization. Regression models were adjusted to estimate growth, nutrient uptake, and nutrient use efficiency of pine trees, based on data collected in this and previous studies. The equations developed in this research can be used in nutrient budget models as well as in other simulation models, to establish recommendations of lime and fertilizers for Pinus taeda stands in southern Brazil.

The objective of this study was to evaluate the effects of varying the ratio of dietary palmitic (C16:0), stearic (C18:0), and oleic (cis-9 C18:1) acids in basal diets containing soyhulls or whole cottonseed on nutrient digestibility, energy partitioning, and production response of lactating dairy cows. Twenty-four mid-lactation multiparous Holstein cows were used in a split-plot Latin square design. Cows were allocated to a main plot receiving either a basal diet with soyhulls (SH, n = 12) or a basal diet with whole cottonseed (CS, n = 12) that was fed throughout the experiment. Within each plot a 4 × 4 Latin square arrangement of treatments was used in 4 consecutive 21-d periods. Treatments were (1) control (CON; no supplemental fat), (2) high C16:0 supplement [PA; fatty acid (FA) supplement blend provided ∼80% C16:0], (3) C16:0 and C18:0 supplement (PA+SA; FA supplement blend provided ∼40% C16:0 + ∼40% C18:0), and (4) C16:0 and cis-9 C18:1 supplement (PA+OA; FA supplement blend provided ∼45% C16:0 + ∼35% cis-9 C18:1). Interactions between basal diets and FA treatments were observed for dry matter intake (DMI) and milk yield. Among the SH diets, PA and PA+SA increased DMI compared with CON and PA+OA treatments, whereas in the CS diets PA+OA decreased DMI compared with CON. The PA, PA+SA, and PA+OA treatments increased milk yield compared with CON in the SH diets. The CS diets increased milk fat yield compared with the SH diets due to the greater yield of de novo and preformed milk FA. The PA treatment increased milk fat yield compared with CON, PA+SA, and PA+OA due to the greater yield of mixed-source (16-carbon) milk FA. The PA treatment increased 3.5% fat-corrected milk compared with CON and tended to increase it compared with PA+SA and PA+OA. The CS diets increased body weight (BW) change compared with the SH diets. Additionally, PA+OA tended to increase BW change compared with CON and PA and increased it in comparison with PA+SA. The PA and PA+OA treatments increased dry matter and neutral detergent fiber digestibility compared with PA+SA and tended to increase them compared with CON. The PA+SA treatment reduced 16-carbon, 18-carbon, and total FA digestibility compared with the other treatments. The CS diets increased energy partitioning toward body reserves compared with the SH diets. The PA treatment increased energy partitioning toward milk compared with CON and PA+OA and tended to increase it compared with PA+SA. In contrast, PA+OA increased energy partitioned to body reserves compared with PA and PA+SA and tended to increase it compared with CON. In conclusion, milk yield responses to different combinations of FA were affected by the addition of whole cottonseed in the diet. Among the combinations of C16:0, C18:0, and cis-9 C18:1 evaluated, fat supplements with more C16:0 increased energy output in milk, whereas fat supplements with more cis-9 C18:1 increased energy storage in BW. The effect of soil amendment with degraded granite or perlite on plant growth of Ornithogalum thyrsoides was investigated in Central Chile. Bulbs were planted in April in raised soil beds under greenhouse. Three different substrates were compared: control (unamended soil), decomposed granite soil (DG) and soil in proportion 1:2, and perlite and soil in proportion 1:2. Soil nutrient availability in all treatments was considered higher than the plant requirements. The plants in the unamended soil had the greatest flower stem diameter, total fresh weight, and aboveground fresh weight. In terms of stem length, inflorescence length, belowground structures fresh weight, and inflorescence fresh weight, perlite with soil had similar results as unamended soil, but never better. Our results indicate that, under the conditions of this experiment, DG or perlite is not recommended for growing O. thyrsoides, since they did not improve either vegetative or reproductive growth. However, the impacts of a HFHS diet during pregnancy on maternal insulin sensitivity and signalling in relation to feto-placental growth and glucose utilization are unknown. The present study examined the effects of a HFHS diet during mouse pregnancy on maternal glucose tolerance and insulin resistance, as well as, on feto-placental glucose metabolism. Female mice were fed a control or HFHS diet from day (D) 1 of pregnancy (term = D20.5). At D16 or D19, dams were assessed for body composition, metabolite and hormone concentrations, tissue abundance of growth and metabolic signalling pathways, glucose tolerance and utilization and insulin sensitivity. HFHS feeding perturbed maternal insulin sensitivity in late pregnancy; hepatic insulin sensitivity was higher, whereas sensitivity of the skeletal muscle and white adipose tissue was lower in HFHS than control dams. These changes were accompanied by increased adiposity and reduced glucose production and glucose tolerance of HFHS dams. The HFHS diet also disturbed the hormone and metabolite milieu and altered expression of growth and metabolic signalling pathways in maternal tissues. Furthermore, HFHS feeding was associated with impaired feto-placental glucose metabolism and growth. A HFHS diet during pregnancy therefore causes maternal metabolic dysfunction with consequences for maternal nutrient allocation for fetal growth.
