13. A new FRET biosensor enlightens cAMP signalling in budding yeast

Abstract

The cAMP-PKA signalling cascade in budding yeast regulates adaptation to changing environments. Many questions remain about the function of cAMP dynamics, largely because no robust method for in vivo cAMP measurements exists for yeast. Here we developed yEPAC, a FRET-based biosensor for cAMP measurements in yeast. We show that this biosensor can be used in flow cytometry, allowing for high-throughput single cell-level quantification that complements microscopy measurements. We quantified both steady-state cAMP levels and dynamic changes in response to sudden nutrient transitions. Steady-state cAMP levels were found to correlate with growth rate (rather than glycolytic flux) and were independent of extracellular sensing. During transitions, a cAMP-peak differentiates between different carbon source transitions, and is generated only when switching from non-or slowly-fermentable sugars to rapidly fermentable ones. Furthermore, generation of the cAMP peak is mediated by a combination of extracellular sensing and metabolism; deficits in either results in a largely diminished response. Moreover, the cAMP peak follows Weber’s law; its height scales with the relative, and not the absolute, change in glucose. This means that cells respond to small changes in extracellular glucose when the initial level is low, but ignore these changes when this level is high. Lastly, we speculate that the cAMP peak height conveys information about prospective growth rates, especially for fermentable sugars. In conclusion, our yEPAC-sensor makes possible new avenues for understanding yeast physiology, signalling and metabolic adaptation.

Abstract

Sialylated glycan structures are known for their immunomodulatory capacities and their contribution to tumor immune evasion. However, the role of aberrant sialylation in colorectal cancer and the consequences of complete tumor desialylation on anti-tumor immunity remain unstudied. Here, we report that CIRSPR/Cas9-mediated knock out of the CMAS gene, encoding a key enzyme in the sialylation pathway, in the mouse colorectal cancer MC38 cell line completely abrogated cell surface expression of sialic acids (MC38-Sianull ) and, unexpectedly, significantly increased in vivo tumor growth compared to the control MC38-MOCK cells. This enhanced tumor growth of MC38-Sianull cells could be attributed to decreased CD8+ T cell frequencies in the tumor microenvironment only, as immune cell frequencies in tumor-draining lymph nodes remained unaffected. In addition, MC38-Sianull cells were able to induce CD8+ T cell apoptosis in an antigen-independent manner. Moreover, low CMAS gene expression correlated with reduced recurrence-free survival in a human colorectal cancer cohort, supporting the clinical relevance of our work. Together, these results demonstrate for the first time a detrimental effect of complete tumor desialylation on colorectal cancer tumor growth, which greatly impacts the design of novel cancer therapeutics aimed at altering the tumor glycosylation profile. Abstract

Priming of CD8+ T cells by dendritic cells (DCs) is crucial for the generation of effective antitumor immune responses. Here, we describe a liposomal vaccine carrier that delivers tumor antigens to human CD169/Siglec-1+ antigen-presenting cells using gangliosides as targeting ligands. Ganglioside-liposomes specifically bound to CD169 and were internalized by in vitro-generated monocyte-derived DCs (moDCs) and macrophages and by ex vivo-isolated splenic macrophages in a CD169-dependent manner. In blood, high-dimensional reduction analysis revealed that ganglioside-liposomes specifically targeted CD14+ CD169+ monocytes and Axl+ CD169+ DCs. Liposomal codelivery of tumor antigen and Toll-like receptor ligand to CD169+ moDCs and Axl+ CD169+ DCs led to cytokine production and robust cross-presentation and activation of tumor antigen-specific CD8+ T cells. Finally, Axl+ CD169+ DCs were present in cancer patients and efficiently captured ganglioside-liposomes. 27. Culture and Development: From Romantic Relativism to Reality.

Abstract

Using case studies of French-speaking West Africa and Southern Appalachia, this paper examines the role of traditional culture in developing regions by challenging current theories on that subject. The capitalistic and Marxist approaches that dominated well into the 1960s maintained that elimination of traditional social relations and values was necessary for progress in underdeveloped areas. The strategy was to introduce modernizing influences which would transform local independence and self-sufficiency by integration into a market economy. Residents would internalize this perception and see progress in relation to urbanization. The romantic approach toward development which appeared in the 1960s and 1970s blamed external forces rather than native residents for the lack of development. This theory attributed underdevelopment to imperialist expansion by industrialized societies. However, it is important to note that underdevelopment would exist even without external exploitation. A synthesis of the capitalistic, Marxist, and romantic ideologies seems to be the best approach to understanding the relationship between traditional values and modernizing influences in developing areas. Balanced, humane development goals will be achieved by people who recognize, respect, and work with existing cultural values, but do not over-glamorize them. (Author/KC)

Abstract

Managing and implementing supplier relationships is an interest shared by both academics and practitioner. A growing focus of attention is on the outcomes of these relationships since they have been shown to deliver enhanced performance. However, variations in performance across different relationship types remains less understood. To rectify this, in our study, buyer-supplier relationship outcomes are measured by a performance metric that includes both financial and non-financial elements. The results are drawn from a survey of UK buyers in the engineering, electronics and telecommunications sectors and show that differences can be found in performance outcomes for a variety of governance structures. Abstract

New product development (NPD) is increasingly being viewed as a dynamic interplay between two or more actors. The basic tenet of this relationship is that linkages with other actors help firms create value by combining resources, sharing knowledge, increasing speed to market, reducing risk and in general creating a competitive advantage that is likely to be sustainable. Despite the increasing importance attributed to the involvement of external actors such as users and suppliers in NPD, little is known about current practice in the Irish context. Based on empirical evidence collected from 572 Irish manufacturers, external cooperation remains latent and relatively unused as a mechanism for Irish firms to develop new products. Implications of these findings for managers and researchers are discussed.
