Abstract

The effectiveness of most cancer targeted therapies is short-lived. Tumors often develop resistance that might be overcome with drug combinations. However, the number of possible combinations is vast, necessitating data-driven approaches to find optimal patient-specific treatments. Here we report AstraZeneca's large drug combination dataset, consisting of 11,576 experiments from 910 combinations across 85 molecularly characterized cancer cell lines, and results of a DREAM Challenge to evaluate computational strategies for predicting synergistic drug pairs and biomarkers. 160 teams participated to provide a comprehensive methodological development and benchmarking. Winning methods incorporate prior knowledge of drug-target interactions. Synergy is predicted with an accuracy matching biological replicates for >60% of combinations. However, 20% of drug combinations are poorly predicted by all methods. 3. Pain after hysterectomy: a comparison between four currently available procedures

Abstract

Objective To compare the postoperative pain of four types of hysterectomy. Design Prospective study. Setting Cairo University Faculty of Medicine Hospital and the authors' private practice. Subjects A total of 427 patients underwent hysterectomies for different indications. Interventions Patients were assigned to either total abdominal hysterectomy, laparoscopic-assisted vaginal hysterectomy, subtotal laparoscopic hysterectomy, or vaginal hysterectomy. Measures Linear pain score 12 and 24 h after surgery, dose of meperidine required in the first 24 h, multidimensional pain score 8 days after surgery, and dyspareunia assessed after 3 months of surgery. Results Both laparoscopic procedures were followed by the least postoperative pain and earliest return to work. Subtotal laparoscopic hysterectomy was followed by the least dyspareunia. Abdominal hysterectomy was followed by the highest pain score and latest return to normal physical activity. Conclusion Laparoscopic hysterectomy had the best patient acceptance, with the least postoperative pain.

4. Hydraulic Conductivity: Comparison Between Field Testing and Indirect Techniques

Abstract

Pumping and -field-permeability tests were conducted at four sites to determine soil permeability that is used in designing groundwater control systems. Laboratory tests, on soil samples extracted during site investigation, were performed. Test results were used to check the applicability of the indirect techniques relating permeability to grading characteristics. Confined, unconfined and mixed-aquifer conditions were characterized based on field measurements. It was found that it is more realistic to estimate permeability using field testing. If only data of indirect methods are available, its use sholud be limitted to tender or prelimenary studies. In this case, while implementing some justifications, Hazen's formula can give a good estimate.

Abstract

Analysis of raft foundation and superstruc ture is usually performed considering only soil-raft interaction. In this work, an extensive parametric study, using field tests and finite element analysis, is conducted to study and quantify the effect of raft thickness, column and beam rigidities and so il behavior, on the interaction process. Results of both experimental and numerical techniques are analyzed then input to an artificial neural network , ANN, to be capable of predicting redistributed column loads. A simplified analysis procedure is recommen ded and its accuracy is verified .

Abstract

The authors report a radionuclide renal study in a case of obstructive uropathy secondary to an aneurysm of the internal iliac artery. The aneurysm, which appeared as a vascular blush on the radionuclide angiogram, was diagnosed by contrast angiography. The authors are reporting this case for two reasons: (a) an aneurysm of the internal iliac artery is rare and (b) it is even more rare that such an aneurysm will cause obstructive uropathy. The combination of a vascular blush in the radionuclide an-giographic phase of the renal studies associated with obstructive uropathy should raise the possibility of an aneurysm as the cause of this obstruction.
