Whether and how intramolecular crosslinks in polymeric materials contribute to mechanical properties is debated in both experimental and theoretical arenas. The tethering threads of Octopus bimaculoides egg cases provide a rare window to investigate this question in a biomaterial. The only detectable component of the load-bearing fibers in octopus threads is a 135 kDa protein, octovafibrin, comprising 29 tandem repeats of epidermal growth factor (EGF) each of which contains 3 intramolecular disulfide linkages. The N- and C-terminal C-type lectins mediate linear end-to-end octovafibrin self-assembly. Mechanical testing of threads shows that the regularly spaced disulfide linkages result in improved stiffness, toughness, and energy dissipation. In response to applied loads, molecular dynamics and X-ray scattering show that EGF-like domains deform by recruiting two hidden length β-sheet structures nested between the disulfides. The results of this study further the understanding of intramolecular crosslinking in polymers and provide a foundation for the mechanical contributions of EGF domains to the extracellular matrix.

The evolution of new traits enables expansion into new ecological and behavioural niches. Nonetheless, demonstrated connections between divergence in protein structure, function and lineage-specific behaviours remain rare. Here we show that both octopus and squid use cephalopod-specific chemotactile receptors (CRs) to sense their respective marine environments, but structural adaptations in these receptors support the sensation of specific molecules suited to distinct physiological roles. We find that squid express ancient CRs that more closely resemble related nicotinic acetylcholine receptors, whereas octopuses exhibit a more recent expansion in CRs consistent with their elaborated taste by touch sensory system. Using a combination of genetic profiling, physiology and behavioural analyses, we identify the founding member of squid CRs that detects soluble bitter molecules that are relevant in ambush predation. We present the cryo-electron microscopy structure of a squid CR and compare this with octopus CRs1 and nicotinic receptors2. These analyses demonstrate an evolutionary transition from an ancestral aromatic cage that coordinates soluble neurotransmitters or tastants to a more recent octopus CR hydrophobic binding pocket that traps insoluble molecules to mediate contact-dependent chemosensation. Thus, our study provides a foundation for understanding how adaptation of protein structure drives the diversification of organismal traits and behaviour.

Chemotactile receptors (CRs) are a cephalopod-specific innovation that allow octopuses to explore the seafloor via taste by touch1. CRs diverged from nicotinic acetylcholine receptors to mediate contact-dependent chemosensation of insoluble molecules that do not readily diffuse in marine environments. Here we exploit octopus CRs to probe the structural basis of sensory receptor evolution. We present the cryo-electron microscopy structure of an octopus CR and compare it with nicotinic receptors to determine features that enable environmental sensation versus neurotransmission. Evolutionary, structural and biophysical analyses show that the channel architecture involved in cation permeation and signal transduction is conserved. By contrast, the orthosteric ligand-binding site is subject to diversifying selection, thereby mediating the detection of new molecules. Serendipitous findings in the cryo-electron microscopy structure reveal that the octopus CR ligand-binding pocket is exceptionally hydrophobic, enabling sensation of greasy compounds versus the small polar molecules detected by canonical neurotransmitter receptors. These discoveries provide a structural framework for understanding connections between evolutionary adaptations at the atomic level and the emergence of new organismal behaviour.

Biology is entering a new era in which techniques honed in model systems can be applied to the expanding array of organisms with sequenced genomes. In this issue of Cell, van Giesen et al. (2020) characterize the molecular foundation of the touch-taste sensory system in octopus suckers.

For many animals, evolution has selected for complex visual systems despite the high energetic demands associated with maintaining eyes and their processing structures. Therefore, the metabolic demands of visual systems make them highly sensitive to fluctuations in available oxygen. In the marine environment, oxygen changes over daily, seasonal and inter-annual time scales, and there are large gradients of oxygen with depth. Vision is linked to survival in many marine animals, particularly among the crustaceans, cephalopods and fish, and early life stages of these groups rely on vision for prey capture, predator detection and their distribution in the water column. Using in vivo electroretinogram recordings, we show that there is a decrease in retinal sensitivity to light in marine invertebrates when exposed to reduced oxygen availability. We found a 60-100% reduction in retinal responses in the larvae of cephalopods and crustaceans: the market squid (Doryteuthis opalescens), the two-spot octopus (Octopus bimaculatus), the tuna crab (Pleuroncodes planipes) and the graceful rock crab (Metacarcinus gracilis). A decline in oxygen also decreases the temporal resolution of vision in D. opalescens These results are the first demonstration that vision in marine invertebrates is highly sensitive to oxygen availability and that the thresholds for visual impairment from reduced oxygen are species-specific. Oxygen-impaired retinal function may change the visual behaviors crucial to survival in these marine larvae. These findings may impact our understanding of species' vulnerability to ocean oxygen loss and suggest that researchers conducting electrophysiology experiments should monitor oxygen levels, as even small changes in oxygen may affect the results.

This article follows an increased interest in the octopus in both popular science and fiction. Octopuses have long held fascination and are commonly tied to processes of aging: Even though their life expectancy tends to be lower than that of humans, they are often framed as "old", not only by appearing as mythical creatures from an unknown past but also by appearing wise and intelligent. Whereas the octopus has been framed as Other, prominently by inspiring the aesthetics of alien life forms, recent examples have underlined the possibility of inter-species contact and communication. This article traces these moments of contact and investigates the role of aging in such fictional encounters. By focusing on two recent examples, Shelby Van Pelt's Remarkably Bright Creatures (2022) and Gina Chung's Sea Change (2023), it illustrates the ways that contemporary fiction narratively links the octopus to older age and discusses forms of non-human aging., (Copyright © 2024. Soft-bodied aquatic organisms have exhibited remarkable capabilities in navigating and moving within liquid environments serving as a profound inspiration for the development of bionic robots with intricate movements. Traditional rigid components are being replaced by stimulus-responsive soft materials such as hydrogels and shape memory polymers, leading to the creation of dynamically responsive soft robots. In this study, the development of a bionic robot inspired by the shape of an octopus and the adsorptive properties of its tentacles, specifically tailored for targeted stimulation and pH sensing in the cervix, are presented. This approach involves the design of a soft, water-based Janus adhesive hydrogel patch that adheres to specific parts of the cervix and responds to pH changes through external stimuli. The hydrogel patch incorporates inverse opal microstructures mimicking the legs of an octopus, to facilitate efficient and stable locomotion, unidirectional transport of biofluids, and pH-responsive behavior. This miniature bionic robot showcases controlled adhesion and precise unidirectional fluid transport highlighting its potential for targeted stimulus response and pH sensing in the uterine cervical tract. An essential requirement for biomedical devices is the capability of conformal adaptability on diverse irregular 3D (three-dimensional) nonflat surfaces in the human body that may be covered with liquids such as mucus or sweat. However, the development of reversible adhesive interface materials for biodevices that function on complex biological surfaces is challenging due to the wet, slippery, smooth, and curved surface properties. Herein, we present an ultra-adaptive bioadhesive for irregular 3D oral cavities covered with saliva by integrating a kirigami-metastructure and vertically self-aligning suction cups. The flared suction cup, inspired by octopus tentacles, allows adhesion to moist surfaces. Additionally, the kirigami-based auxetic metastructure with a negative Poisson's ratio relieves the stress caused by tensile strain, thereby mitigating the stress caused by curved surfaces and enabling conformal contact with the surface. As a result, the adhesive strength of the proposed auxetic adhesive is twice that of adhesives with a flat backbone on highly curved porcine palates. For potential application, the proposed auxetic adhesive is mounted on a denture and performs successfully in human subject feasibility evaluations. An integrated design of these two structures may provide functionality and potential for biomedical applications.

Regeneration is a process that restores structure and function of tissues damaged by injury or disease. In mammals complete regeneration is often unsuccessful, while most of the low phyla animals can re-grow many parts of their body after amputation. Cephalopod molluscs, and in particular Octopus vulgaris, are well known for their capacity to regenerate their arms and other body parts, including central and peripheral nervous system. To better understand the mechanism of recovery following nerve injury in this species we investigated the process of axon regrowth and nerve regeneration after complete transection of the Octopus pallial nerves. This injury induces scar formation and activates the proliferation of hemocytes which invade the lesion site. Hemocytes appear involved in debris removal and seem to produce factors that foster axon re-growth. Connective tissue is involved in driving regenerating fibers in a single direction, outlining for them a well-defined pathway. Injured axons are able to quickly re-grow thus to restoring structure and function.

Shallow waters are characterized by fluctuating environmental conditions, modulating marine life cycles and biological phenomena. Multiple variations in water temperature could affect eggs and embryos during spawning events of many marine invertebrate species, yet most of the findings on embryonic development in invertebrates come from experiments based on the constant temperature. In this study, to examine the effects of temperature variation on octopus embryos, Amphioctopus fangsiao, a common shallow-water octopus along the coast of China, was exposed to the constant temperature (18 °C, in situ temperature of the seawater in Lianyungang), ramping temperatures (from 18 to 24 °C), diel oscillating temperatures (18 °C and 20 °C for 12 h each day), and acute increasing temperatures (the temperature increased sharply from 18 °C to 24 °C at embryonic development stage XIX) for 47 days (from embryogenesis to settlement). The results demonstrated that the temperature variations accelerated the development time of A. fangsiao embryos. Temperature fluctuations could cause embryonic oxidative damage and disorder of glycolipid metabolism, thereby affecting the growth performance of embryos and the survival rate of hatchings. Through transcriptome sequencing, the mechanistic adaption of the embryo to environmental temperature variations was revealed. The pathways involved in the TCA cycle, DNA replication and repair, protein synthesis, cell signaling, and nervous system damage repair were significantly enriched, indicating that the embryo could improve heat tolerance to thermal stress by regulating gene expression. Moreover, acute warming temperatures posed the most detrimental effects on A. fangsiao embryos, which could cause embryos to hatch prematurely from the vegetal pole, further reducing the survival of hatchings. Meanwhile, the diel oscillating temperature was observed to affect the normal morphology of the embryo, resulting in embryo deformities. Thus, the constant temperature is critical for balanced growth and defense status in octopuses by maintaining metabolism homeostasis. Global warming is one of the most significant and widespread effects of climate change. While early life stages are particularly vulnerable to increasing temperatures, little is known about the molecular processes that underpin their capacity to adapt to temperature change during early development. Using a quantitative proteomics approach, we investigated the effects of thermal stress on octopus embryos. We exposed Octopus berrima embryos to different temperature treatments (control 19°C, current summer temperature 22°C, or future projected summer temperature 25°C) until hatching. By comparing their protein expression levels, we found that future projected temperatures significantly reduced levels of key eye proteins such as S-crystallin and retinol dehydrogenase 12, suggesting the embryonic octopuses had impaired vision at elevated temperature. We also found that this was coupled with a cellular stress response that included a significant elevation of proteins involved in molecular chaperoning and redox regulation. Energy resources were also redirected away from non-essential processes such as growth and digestion. These findings, taken together with the high embryonic mortality observed under the highest temperature, identify critical physiological functions of embryonic octopuses that may be impaired under future warming conditions. Dicyemids (phylum Dicyemida) are the most common and most characteristic endosymbionts in the renal sacs of benthic cephalopod molluscs: octopuses and cuttlefishes. Typically, 2 or 3 dicyemid species are found in a single specimen of the host, and most dicyemids have high host specificity. Host-specific parasites are restricted to a limited range of host species by ecological barriers that impede dispersal and successful establishment; therefore, phylogenies of interacting groups are often congruent due to repeated co-speciation. Most frequently, however, host and parasite phylogenies are not congruent, which can be explained by processes such as host switching and other macro-evolutionary events. Here, the history of dicyemids and their host cephalopod associations were studied by comparing their phylogenies. Dicyemid species were collected from 8 decapodiform species and 12 octopodiform species in Japanese waters. Using whole mitochondrial cytochrome c oxidase subunit 1 (COI) sequences, a phylogeny of 37 dicyemid species, including 4 genera representing the family Dicyemidae, was reconstructed. Phylogenetic trees derived from analyses of COI genes consistently suggested that dicyemid species should be separated into 3 major clades and that the most common genera, Dicyema and Dicyemennea, are not monophyletic. Thus, morphological classification does not reflect the phylogenetic relationships of these 2 genera. Divergence (speciation) of dicyemid species seems to have occurred within a single host species. Possible host-switching events may have occurred between the Octopodiformes and Decapodiformes or within the Octopodiformes or the Decapodiformes. Therefore, the mechanism of dicyemid speciation may be a mixture of host switching and intra-host speciation. Constructing high-density contact-separation sites on conductive materials highly determines the sensitivity of flexible resistance-type sensors relying on the crack microstructures. Herein, inspired from the multiple-tentacle structures on octopus, we demonstrated a sort of novel carbonized ZIF-8@loofah (CZL) as conductive material to develop ultrasensitivity flexible sensor, in which the carbonized ZIF-8 nanoparticles (~100 nm) served as tentacles. Originating from the formation of high-density contact-separation sites, the fabricated CZL-based strain sensor delivered ultrahigh sensitivity of GF max  = 15,901, short response time of 22 ms and excellent durability over 10,000 cycles. These features enable the sensor with efficient monitoring capacity for complex human activities, such as pulse rate and phonation. Moreover, when CZL was assembled into triboelectric nanogenerator (TENG), CZL-based TENG can effectively convert the irregular biomechanical energy into electric energy, providing sustainable power supply for the continuous operation of the sensing micro-system. This study is considered the first report on the digestive tract pH of the pygmy octopus (Paroctopus digueti). Adult octopuses obtained from the wild (mean ± SD) (42.1 ± 15.1 g), and those acclimated to captivity in a fed (25.4 ± 9.0 g, n = 15) or fasted (23.1 ± 6.1 g, n = 15) state, were studied. The digestive tract regions of buccal mass (BMA), anterior salivary glands (ASG), posterior salivary glands (PSG), crop (CRO), stomach (STO), caecum (CAE), digestive gland (DGL) and intestine (INT) were dissected. The pH of the internal part of the digestive tract regions was measured. Food intake (dry weight) per octopus was 53.8 ± 35.1 mg to 214.9 ± 157.6 mg at 15 min and 8 h, respectively. The apparent food transit time was approximately 8 h for the appearance of feces in the posterior intestine. In all cases, the pH of the digestive tract regions was lower than pH 7.0. No statistical difference was found when comparing the pH by digestive tract regions between wild octopuses and octopuses in captivity (fasting and feeding). In acclimatized octopuses, the average pH was 6.41 ± 0.22 and 6.41 ± 0.23 for fasting and fed octopuses, respectively. Although DGL had the lowest pH values relative to other digestive tract tissues (p < 0.05), pH was always >5.0 (6.04 ± 0.12 in the wild and 5.97 ± 0.17 in feeding octopuses). The systematics of the New Zealand octopods have only been reviewed twice in the last 100 years. In these revisions many species have been provisionally classified in the genus Octopus. Recent genetic studies have synonymized some New Zealand species with octopuses from other regions. The present study investigates the systematics and phylogeny of octopuses from New Zealand using eighty eight specimens, three mitochondrial genes (16S rRNA, cytochrome c oxidase subunit I, and cytochrome c oxidase subunit III) and one nuclear gene (Rhodopsin). Forty-four new octopod DNA sequences (belonging to 13 species) were included, adding to the 83 existing sequences from GenBank. All sequences were used to generate phylogenetic trees based on Maximum Likelihood (ML) and Bayesian inference (BI), with a data set composed by 97 species, including octopod sister groups and Vampyroteuthis infernalis as an outgroup. Gene tree and species delimitation analyses revealed a distinct genetic difference between two sympatric Graneledone subspecies, which we propose as valid species. Muusoctopus tangaroa is a sister species of M. thielei from Kerguelen; while Enteroctopus zealandicus forms a clade with E. megalocyathus from South America and E. dofleini from the North Pacific. Similarly, Octopus campbelli, O. huttoni, and O. mernoo form a monophyletic group with Robsonella fontaniana from South America, Scaeurgus unicirrhus from the Atlantic and O. pallidus from Australia. Pinnoctopus cordiformis is close to Grimpella thaumastocheir and several species of Octopus sensu lato as in previous phylogenetic studies. This study suggests that octopuses from New Zealand have different phylogenetic and biogeographic origins and represent independent radiations into this region.

Aside from being one of the most fascinating groups of marine organisms, cephalopods play a major role in marine food webs, both as predators and as prey, while representing key living economic assets, namely for artisanal and subsistence fisheries worldwide. Recent research suggests that cephalopods are benefitting from ongoing environmental changes and the overfishing of certain fish stocks (i.e., of their predators and/or competitors), putting forward the hypothesis that this group may be one of the few "winners" of climate change. While many meta-analyses have demonstrated negative and overwhelming consequences of ocean warming (OW), acidification (OA), and their combination for a variety of marine taxa, such a comprehensive analysis is lacking for cephalopod molluscs. In this context, the existing literature was surveyed for peer-reviewed articles featuring the sustained (≥24 h) and controlled exposure of cephalopod species (Cephalopoda class) to these factors, applying a comparative framework of mixed-model meta-analyses (784 control-treatment comparisons, from 47 suitable articles). Impacts on a wide set of biological categories at the individual level (e.g., survival, metabolism, behavior, cell stress, growth) were evaluated and contrasted across different ecological attributes (i.e., taxonomic lineages, climates, and ontogenetic stages). Contrary to what is commonly assumed, OW arises as a clear threat to cephalopods, while OA exhibited more restricted impacts. In fact, OW impacts were ubiquitous across different stages of ontogeny, taxonomical lineages (i.e., octopuses, squids, and cuttlefish). These results challenge the assumption that cephalopods benefit from novel ocean conditions, revealing an overarching negative impact of OW in this group. Importantly, we also identify lingering literature gaps, showing that most studies to date focus on OW and early life stages of mainly temperate species. Our results raise the need to consolidate experimental efforts in a wider variety of taxa, climate regions, life stages, and other key environmental stressors, such as deoxygenation and hypoxia, to better understand how cephalopods will cope with future climate change., (© The Author(s) 2023. Histopathology associated with the apicomplexan Aggregata valdessensis in the wild population of Octopus tehuelchus was studied. Moreover, to analyze the impact of the prevalence and infection intensity on the dorsal mantle length, sex, maturity stages, condition index and total hemocyte counts on wild O. tehuelchus, Generalized Linear Models (GLMs) were applied. Kruskal-Wallis (KW) test was used to compare growth parameter (RNA/DNA ratios) in infected and uninfected octopuses. Lesions including hypertrophy of host cells and destruction of the tissue organ architecture as well as the replacement of host tissue by parasites were associated with infection. The infection caused an inflammatory defensive response in form of hemocytic infiltration.. The prevalence of apicomplexan increased with octopus dorsal mantle length (GLM, t = 2.68; p = 0.007). Infection intensity was significantly higher in male octopus (GLM, t = 2.69; p = 0.009) and decreased at higher temperatures in both sexes (GLM, t = -2.42; p = 0.018). The digestive gland condition index did not significantly vary with infection intensity, and no significant relationship between total hemocyte counts and coccidian infection was found (Lineal Model, t = 0.46; p = 0.64). Benzophenone-3 (BP-3) is an active ingredient in sunscreen lotions and personal-care products that protects against the damaging effects of ultraviolet rays. Given its worldwide dissemination, it has been linked with harmful effects on aquatic biota; however, its impact is not fully understood calling for further studies. To understand the impacts on an important economically and ecologically species, we evaluated the toxicity of BP-3 during the embryonic development of Octopus maya. Embryos were exposed to increasing concentrations of up to 500 µg BP-3/L until hatching. Antioxidant enzyme activities, oxidative-stress indicators, and B-esterases activities were measured at different developmental phases (organogenesis, activation, and growth). There were no significant differences between treatments, suggesting the lack of production of toxic metabolites that may be related to a protective chorion, an underdeveloped detoxification system, and the experimental conditions that limited phototoxicity., (© 2023. In Die Another Day, James Bond receives an Aston Martin that can render itself invisible by dynamically reproducing the surroundings on the car's "polymer skin". In what is widely regarded as the worst Bond movie ever, the invisible car scene is cited as the moment the plot plunges into the truly absurd. But what if nature had actually invented such a technology, and did so hundreds of millions of years ago? The coleoid cephalopods - octopus, cuttlefish and squid - are living examples of dynamic camouflage. Their skin is covered with a high-resolution array of 'cellular pixels' (chromatophores) that are controlled by the brain. To disappear into their surroundings, cephalopods recreate an approximation of their environment on their skin by activating different combinations of colored chromatophores. However, unlike the fictional Bond car, whose surface is coated in tiny cameras to detect the environment, cephalopods don't see the world with their skin. Instead, the visual world is detected by the eyes, processed in the brain, and then used to activate motor commands that direct the skin's camouflage pattern. Thus, cephalopod skin patterns are an external manifestation of their internal perception of the world. How do cephalopods approximate the world with their skin? What can this teach us about how brains work? Coleoid cephalopods (octopuses, squids and cuttlefishes) are the only branch of the animal kingdom outside of vertebrates to have evolved both a large brain and camera-type eyes. They are highly dependent on vision, with the majority of their brain devoted to visual processing. Their excellent vision supports a range of advanced visually guided behaviors, from navigation and prey capture, to the ability to camouflage based on their surroundings. However, their brain organization is radically different from that of vertebrates, as well as other invertebrates, providing a unique opportunity to explore how a novel neural architecture for vision is organized and functions. Relatively few studies have examined the cephalopod visual system using current neuroscience approaches, to the extent that there has not even been a measurement of single-cell receptive fields in their central visual system. Therefore, there remains a tremendous amount that is unknown about the neural basis of vision in these extraordinary animals. Dicyemid mesozoans usually consist of 10 to 40 cells. They are characterized by 2 distinct embryos, vermiform and infusoriform, that develop within the axial cell of the adult. The means of escape of each embryo from the parent body was studied in Dicyema japonicum and Dicyema misakiense, parasites of Octopus sinensis. There were no differences in means of escape between species or embryo type, apparently due to morphological constraints whereby the parents (nematogen or rhombogen) share a similar body organization. Escapes were effected through the gap between adjacent peripheral cells of the adult, rupturing the axial cell membrane and the membrane that envelopes the embryo. After the embryo escaped, the path was closed by the enveloping membrane left behind by the embryo. Vermiform embryos can escape from any region of the body, although more embryos were observed to escape from anterior regions than from posterior regions. Infusoriform embryos escaped from both anterior and posterior regions in the axial cell, with more embryos observed to escape from the posterior regions. This retrospective study examines pathologic findings in 593 captive cephalopods (340 octopuses, 130 cuttlefish, 33 squid, and 90 nautiluses; 22 species in total) submitted to International Zoo Veterinary Group Pathology between May 2003 and August 2022. Common octopus, European common cuttlefish, hummingbird bobtail squid, and chambered nautilus were the most numerous species from the included orders of Octopoda, Sepiida, Sepiolida, and Nautilida, respectively. Commonly identified conditions included coccidiosis and renal dicyemid mesozoan infection in octopuses, amebiasis in squid, bacterial infections in cuttlefish, and idiopathic multisystemic inflammatory disease in nautiluses. Coccidiosis was most frequent in common octopuses, giant Pacific octopuses, and California 2-spot octopuses (present in 68.4%, 46.3%, and 23.8% of these species, respectively) and was the attributed cause of death in 32.3%, 36.0%, and 60.0% of such cases, respectively. Ulcerative dermatitis (UD) was common, affecting squid, cuttlefish, and octopuses, while many nautiluses exhibited black shell disease and/or UD. Notable differences in the prevalence of UD were detected between laboratory and aquaria-housed decapodiforms; 52.2% for laboratory squid versus 20.0% in aquaria; 51.3% in laboratory cuttlefish versus 11.0% in aquaria. All octopuses and nautiluses in the study were derived from aquaria. Semelparity-associated death in Coleoidea species was identified in 22.4% of octopuses, 11.5% of cuttlefish, and 6.1% of squid. This report aims to provide an overview and reassessment of species-specific disease patterns under aquarium and laboratory management as a starting point for future developments in husbandry and disease investigation.

Does warmth from hydrothermal springs play a vital role in the biology and ecology of abyssal animals? Deep off central California, thousands of octopus ( Muusoctopus robustus ) migrate through cold dark waters to hydrothermal springs near an extinct volcano to mate, nest, and die, forming the largest known aggregation of octopus on Earth. Warmth from the springs plays a key role by raising metabolic rates, speeding embryonic development, and presumably increasing reproductive success; we show that brood times for females are ~1.8 years, far faster than expected for abyssal octopods. Using a high-resolution subsea mapping system, we created landscape-scale maps and image mosaics that reveal 6000 octopus in a 2.5-ha area. Because octopuses die after reproducing, hydrothermal springs indirectly provide a food supplement to the local energy budget. Although localized deep-sea heat sources may be essential to octopuses and other warm-tolerant species, most of these unique and often cryptic habitats remain undiscovered and unexplored.

Proteomics, the temporal study of proteins expressed by an organism, is a powerful technique that can reveal how organisms respond to biological perturbations, such as disease and environmental stress. Yet, the use of proteomics for addressing ecological questions has been limited, partly due to inadequate protocols for the sampling and preparation of animal tissues from the field. Although RNAlater is an ideal alternative to freezing for tissue preservation in transcriptomics studies, its suitability for the field could be more broadly examined. Moreover, existing protocols require samples to be preserved immediately to maintain protein integrity, yet the effects of delays in preservation on proteomic analyses have not been thoroughly tested. Hence, we optimised a proteomic workflow for wild-caught samples. First, we conducted a preliminary in-lab test using SDS-PAGE analysis on aquaria-reared Octopus berrima confirming that RNAlater can effectively preserve proteins up to 6 h after incubation, supporting its use in the field. Subsequently, we collected arm tips from wild-caught Octopus berrima and preserved them in homemade RNAlater immediately, 3 h, and 6 h after euthanasia. Processed tissue samples were analysed by liquid chromatography tandem mass spectrometry to ascertain protein differences between time delay in tissue preservation, as well as the influence of sex, tissue type, and tissue homogenisation methods. Over 3500 proteins were identified from all tissues, with bioinformatic analysis revealing protein abundances were largely consistent regardless of sample treatment. However, nearly 10% additional proteins were detected from tissues homogenised with metal beads compared to liquid nitrogen methods, indicating the beads were more efficient at extracting proteins. Cephalopods are highly visual animals with camera-type eyes, large brains, and a rich repertoire of visually guided behaviors. However, the cephalopod brain evolved independently from those of other highly visual species, such as vertebrates; therefore, the neural circuits that process sensory information are profoundly different. It is largely unknown how their powerful but unique visual system functions, as there have been no direct neural measurements of visual responses in the cephalopod brain. In this study, we used two-photon calcium imaging to record visually evoked responses in the primary visual processing center of the octopus central brain, the optic lobe, to determine how basic features of the visual scene are represented and organized. We found spatially localized receptive fields for light (ON) and dark (OFF) stimuli, which were retinotopically organized across the optic lobe, demonstrating a hallmark of visual system organization shared across many species. An examination of these responses revealed transformations of the visual representation across the layers of the optic lobe, including the emergence of the OFF pathway and increased size selectivity. We also identified asymmetries in the spatial processing of ON and OFF stimuli, which suggest unique circuit mechanisms for form processing that may have evolved to suit the specific demands of processing an underwater visual scene. This research aimed to chemically synthesize and evaluate the antiproliferative and anti-inflammatory potential of ozopromide (OPC), a novel compound recently isolated from O. vulgaris ink. After chemical synthesis, OPC structural characterization was confirmed by COSY2D, FTIR, and C-/H-NMR. OPC inhibited the growth of human breast (MDA-MB-231), prostate (22Rv1), cervix (HeLa), and lung (A549) cancerous cells, being the highest effect on the latter (IC 50 : 53.70 μM). As confirmed by flow cytometry, OPC induced typical apoptosis-derived morphological features on A549 cells, mostly at early and late apoptosis stages. OPC generated a dose-dependent effect inhibiting IL-6 and IL-8 on LPS-stimulated peripheral mononuclear cells (PBMCs). A major affinity of OPC to Akt-1 and Bcl-2 proteins in silico agreed with the observed pro-apoptotic mechanisms. Results suggested that OPC has the potential to alleviate inflammation and be further studied for anticancer activity. This baseline focuses on the octopus pot, a litter item found on the North Atlantic Iberian coast. Octopus pots are deployed from vessels in ropes, with several hundred units, and placed on the seabed, to capture mostly Octopus Vulgaris. The loss of gears due to extreme seas state, bad weather and/or fishing-related unforeseen circumstances, cause the octopus pots contaminating beaches and dunes, where they are transported by sea current, waves and wind actions. This work i) gives an overview of the use of octopus pot on fisheries, ii) analyses the spatial distribution of this item on the coast, and iii) discusses the potential measures for tackling the octopus pot plague on the North Atlantic Iberian coast. Wearable electronics demand high adhesion properties through various skin conditions. Here, 3D-printed porous skin patches with octopus-like suckers of different geometries are presented. Experimental and theoretical studies are investigated to show an enhanced, low-cost 3D-printed bioinspired patches that successfully obtain biosignals comparable to commercial electrodes.Clinical Relevance- This work establishes low-cost, highly-adhesive skin patches that are irritation- and contamination-free with effortless peel-off technique for biosignal measurement.
