Florida Administrative Code (FAC) 5E-2.0311, "Performance Standards and Acceptable Test Conditions for Preventive Termite Treatments for New Construction," approved in 2003, currently remains without change as originally written. This Code requires broad revision to include updating existing and new termite management methods and technologies, describing acceptable testing protocols that objectively evaluate termite management product efficacy, and clearly and accurately define product performance standards required for approval and use in Florida, USA. This manuscript provides modification and expansion of the existing Code, first in outline form, then followed by a proposed complete revision including detailed explanations for each outline heading and sub-heading. Efficacy requirements, acceptable test conditions, and performance standards requirements are described for termite management products, materials, and non-chemical physical exclusion barriers. We also include testing protocols for field plot and structure efficacy tests. Minimum performance standards also are described for field plots and structure tests to include: (1) termiticide applications to soil, (2) termite baiting systems, (3) pesticides applied directly to wood and building components, (4) pesticide combination mixtures, and (5) new application technologies, products, and methodologies. Although this manuscript addresses revision of a specific Florida Administrative Code, it also is appropriate for consideration by the United States Environmental Protection Agency (USEPA) as well as regulatory entities in other states responsible for pesticide regulation and consumer protection services when writing or revising their code requirements for termite management. Nylanderia fulva (Mayr) is an invasive ant that is a serious pest in the southern United States. Pest control operators and homeowners are challenged to manage pest populations below acceptable thresholds. Contact and bait insecticides are key components of an Integrated Pest Management (IPM) strategy, however, little is known about their efficacy. In repellency and efficacy bioassays, N. fulva were not completely repelled by any insecticide tested, although fewer ants crossed a surface treated with Temprid®. Few insecticides provided rapid control. Termidor® and Temprid® were the best performing with mean mortality of 100% in 13.4 and 19.0 days, respectively. In no-choice bait acceptance studies, it was shown that N. fulva generally had greater acceptance of carbohydrate-based ant baits (Advion®, InTiceTM (gel), and InTiceTM (granular)). However, mortality was low for the InTiceTM baits in a 7-day bioassay. Maxforce® Ant Killer Bait Gel and Advance® 375A in the spring and Maxforce® Complete in the summer and fall required the fewest days to reach 100% mortality. Bait active ingredients that resulted in the highest mortality were hydramethylnon and fipronil. Nylanderia fulva (Mayr), the tawny crazy ant, is an invasive pest established in Florida and several other Gulf Coast states. In their invasive ranges in the Southeastern USA, large N. fulva populations have reduced species abundance, even displacing another invasive ant, Solenopsis invicta (Buren). In North Florida, N. fulva populations survive winter temperatures that reach below freezing for extended periods. However, the shallow littoral debris used by N. fulva for nest construction offers little insulation to brood and reproductives when exposed to freezing temperatures. Field populations of N. fulva in North Florida were observed tunneling below ground, a previously undescribed behavior. Other invasive ants exhibit similar subterranean tunneling behavior as a means of thermoregulation. To test the hypothesis that N. fulva has the capacity to construct subterranean tunnels across a range of ecologically relevant temperatures, tunneling performance for N. fulva and S. invicta, another invasive ant that tunnels extensively, were compared at four temperatures (15.0, 18.0, 20.0, and 22.0 °C). Overall, N. fulva tunneled significantly less than S. invicta. Nylanderia fulva tunneled furthest at warmer temperatures whereas S. invicta tunneled furthest at cooler temperatures. However, N. fulva constructed subterranean tunnels at all temperatures evaluated. These data support the hypothesis that N. fulva is capable of tunneling in temperatures as low as 15.0 °C, confirming that this ant can also perform a behavior that is used by other ants for cold avoidance. Simple Summary: Termites are structurally destructive pests that can infest a consumer's most important investment, their home. The evolution of termite control is complex, involving people's understanding of termite biology and behavior, building construction, the use of soil termiticides, baits, wood treatments, and physical barriers, as well as regulations from different industries (pest control and building construction). Soil termiticides have been the standard method of treatment for decades, but the concern for human and environmental health has driven the development of alternatives. This article discusses the evolution of termite control methods that were alternatives to the standard of the time and the regulatory structure that provides a level of consumer protection for some products. The global economic impact of termites is estimated to be approximately USD 40 billion annually, and subterranean termites are responsible for about 80% of the total impact. Twenty-eight species of termites have been described as invasive, and these termites are spreading, partially due to global trade, making effective control methods essential. Termite control is complex, as is the biology and behavior of this social insect group. In the U.S., termite prevention and control (with claims of structural protection) is regulated by more than one industry (pest control and building construction), and at the federal and state levels. Termite prevention has historically relied on building construction practices that do not create conducive conditions for termite infestations, but as soil termiticides developed, heavy reliance on pesticides became the standard for termite control. The concern for human and environmental health has driven the development of termite control alternatives and regulation for products claiming structural protection. Product development has also provided unprecedented opportunities to study the biology and behavior of cryptobiotic termites. Technological advances have allowed for the re-examination of questions about termite behavior. Lower termites express a unique form of eusocial polyphenism in that totipotent workers can differentiate into either soldier or reproductive caste phenotypes. In this initial effort using RNA interference in termites, we found that two hexamerin genes, Hex-1 and Hex-2, participate in the regulation of caste polyphenism. Our methodology involved a dual gene-silencing approach that used a single short-interfering RNA fragment to silence the two homologous hexamerin genes. We performed validation studies that evaluated effects on nontarget housekeeping genes, silencing of a nonhousekeeping control gene, and effects at the protein level. We found that the two hexamerin proteins, which are inducible by the morphogenetic juvenile hormone and which constitute a significant proportion of total termite protein, suppress juvenile-hormone-dependent worker differentiation to the soldier caste phenotype. This mechanism allows termite colonies to retain high proportions of altruistic worker caste members, thus apparently enhancing colony-inclusive fitness. These findings demonstrate a unique status quo regulatory mechanism for termite worker caste retention and provide an example of previously undescribed preadult developmental/caste-regulatory genes from any social insect. Abstract: RNA interference (RNAi) represents a breakthrough technology for conducting functional genomics research in non-model organisms and for the highly targeted control of insect pests. This study investigated RNAi via voluntary feeding in the economically important pest termite, Reticulitermes flavipes. We used a high-dose double-stranded (ds) RNA feeding approach to silence two termite genes: one encoding an endogenous digestive cellulase enzyme and the other a caste-regulatory hexamerin storage protein. Contrary to results from previous low-dose studies that examined injection-based RNAi, high-dose silencing of either gene through dsRNA feeding led to significantly reduced group fitness and mortality. Hexamerin silencing in combination with ectopic juvenile hormone treatments additionally led to lethal molting impacts and increased differentiation of presoldier caste phenotypes (a phenotype that is not capable of feeding). These results provide the first examples of insecticidal effects from dsRNA feeding in a termite. Additionally, these results validate a high-throughput bioassay approach for use in (i) termite functional genomics research, and (ii) characterizing target sites of conventional and novel RNAi-based termiticides. Abstract: Efficacy of three prototype termite cellulase inhibitors, cellobioimidazole (CBI), fluoromethylcellobiose (FMCB) and fluoromethylglucose (FMG) was investigated using biochemical and feeding assays. Optimal conditions for measuring endoglucanase, exoglucanase and β-glucosidase activities were first determined. The three inhibitors were then tested under optimal conditions against enzyme fractions that represented endogenous (foregut/salivary gland/midgut) and symbiotic (hindgut) cellulases. In vitro, CBI and FMCB both inhibited exoglucanase and β-glucosidase activity (I50s in nM and mM range, respectively). Feeding assays showed significant impacts on both survivorship and feeding stimulation by FMCB and CBI. Enzymatic measurements on feeding assay survivors showed impacts on all three cellulase activities by CBI and lesser impacts by FMG and FMCB. Validative bioassays with the sugars glucose, maltose and cellobiose showed no feeding stimulation or mortality as occurred in feeding inhibitor bioassays. These results indicate efficacy for two cellobiose-based inhibitors, FMCB and CBI, suggesting potential for these inhibitors as novel termite control agents. [Copyright &y& Elsevier]

Abstract: Termites have developed cellulose digestion capabilities that allow them to obtain energy and nutrition from nutritionally poor food sources, such as lignocellulosic plant material and residues derived from it (e.g., wood and humus). Lower termites, which are equipped with both endogenous (i.e., of termite origin) and symbiotic cellulases, feed primarily on wood and wood-related materials. This study investigated cellulase gene diversity, structure, and activity in the lower termite, Reticulitermes flavipes (Kollar). We initially used a metagenomics approach to identify four genes encoding one endogenous and three symbiotic cellulases, which we refer to as Cell-1, -2, -3 and -4. These four genes encode proteins that share significant sequence similarity with known endoglucanases, exoglucanases and xylanases. Phylogenetic analyses further supported these inferred relationships by showing that each of the four cellulase proteins clusters tightly with respective termite, protozoan or fungal cellulases. Gene structure studies revealed that Cell-1, -3 and -4 are intron-free, while Cell-2 contains the first intron sequence to be identified from a termite symbiont cellulase. Quantitative real-time PCR (qRT-PCR) revealed that the endogenous Cell-1 gene is expressed exclusively in the salivary gland/foregut, whereas symbiotic Cell-2, -3, and -4 are highly expressed in the hindgut (where cellulolytic protists are harbored). Cellulase activity assays mapped the distribution pattern of endoglucanase, exoglucanase and xylanase activity throughout the R. flavipes digestive tract. Cellulase gene expression correlated well with the specific types of cellulolytic activities observed in each gut region (foregut + salivary gland, midgut and hindgut). These results suggest the presence of a single unified cellulose digestion system, whereby endogenous and symbiotic cellulases work sequentially and collaboratively across the entire digestive tract of R. flavipes. Integrated pest management and outdoor water conservation comprise key practices encouraged by nonformal educators who work with the public to address significant issues related to water quality and availability. Disconnects among diverse audiences and educational programs pose challenges in achieving the goals of these types of programs. To discern a potential nonformal educational programming strategy, we conducted audience segmentation research incorporating residential environmental landscape best management practices and compared resulting subgroups' characteristics. We explored this paradigm through the context of Extension education and used a nationally-distributed electronic survey instrument to measure well-being and landscape management perceptions and practices in the United States. We collected 2601 complete survey responses and employed cluster analysis to divide the respondents into homogenous subgroups, followed by analysis of variance and chi-square analyses to identify differences among the subgroups. The group that indicated the highest level of engagement with water conservation and IPM practices also had significantly higher well-being, demonstrating the potential to align IPM education with more commonly promoted conservation practices. Membership in the highest IPM and water conservation group was related to several socio-demographic characteristics, including age, homeownership, and homeowners' association membership. This group was also most likely to engage with educational programs. Understanding these indicators of engagement in landscape best management practices and nonformal education, along with higher well-being, may be useful in determining target audiences and potential engagement with nonformal education programs. • Audience segmentation can inform more impactful and targeted nonformal education. • Integrated pest management (IPM) and water conservation rarely considered together. • 2601 survey responses were collected from households across the United States. • High engagement in IPM corresponds to greater water conservation. • Those most engaged in IPM and water conservation have highest levels of personal well-being. Abstract: Lower termites rely on cellulolytic protozoa to aid in the digestion of their wood-based diet. However, despite the major contribution of protozoa to the lower termite digestive system, few techniques have been developed to monitor shifts in protozoan populations. This study investigated whether quantitative real-time PCR (qRT-PCR) and/or cellulase enzyme assays can be used to monitor changes of cellulolytic protozoan populations in the lower termite, Reticulitermes flavipes (Kollar). Previously developed cellulase primer sets were used to test for changes in cellulase gene expression, while three different cellulase enzyme assays were used to assess changes in cellulase enzyme activity. The results from this study indicate that qRT-PCR is a reliable method to monitor shifts in cellulolytic protozoan populations. Specifically, qRT-PCR can serve as a useful monitoring technique during high-throughput screening of novel termite control agents such as cellulase inhibitors, and help to answer questions relating to whether or not such control agents impact cellulolytic protozoan populations.
