Abstract

Root parasitic weeds of the Orobanchaceae, such as witchweeds (Striga spp.) and broomrapes (Orobanche and Phelipanche spp.), cause serious losses in agriculture worldwide. No practical method to control these parasitic weeds has been developed to date. Understanding the characteristic physiological processes in the life cycles of root parasitic weeds is particularly important to identify specific targets for growth modulators. In our previous study, planteose metabolism was revealed to be activated soon after the perception of strigolactones in germinating seeds of O. minor. Nojirimycin inhibited planteose metabolism and impeded seed germination of O. minor, indicating that planteose metabolism is a possible target for root parasitic weed control. In the present study, we investigated the distribution of planteose in dry seeds of O. minor by matrix-assisted laser desorption/ionization–mass spectrometry imaging. Planteose was detected in tissues surrounding—but not within—the embryo, supporting its suggested role as a storage carbohydrate. Biochemical assays and molecular characterization of an α-galactosidase family member, OmAGAL2, indicated the enzyme is involved in planteose hydrolysis in the apoplast around the embryo after the perception of strigolactones to provide the embryo with essential hexoses for germination. These results indicated that OmAGAL2 is a potential molecular target for root parasitic weed control.HighlightPlanteose accumulated in tissues surrounding the embryo in Orobanche minor dry seeds and was indicated to be hydrolyzed in the apoplast around the embryo by α-galactosidase during germination.

Contains fulltext : 219632.pdf (publisher's version ) (Open Access), Preclinical applications of resting-state functional magnetic resonance imaging (rsfMRI) offer the possibility to non-invasively probe whole-brain network dynamics and to investigate the determinants of altered network signatures observed in human studies. Mouse rsfMRI has been increasingly adopted by numerous laboratories worldwide. Here we describe a multi-centre comparison of 17 mouse rsfMRI datasets via a common image processing and analysis pipeline. Despite prominent cross-laboratory differences in equipment and imaging procedures, we report the reproducible identification of several large-scale resting-state networks (RSN), including a mouse default-mode network, in the majority of datasets. A combination of factors was associated with enhanced reproducibility in functional connectivity parameter estimation, including animal handling procedures and equipment performance. RSN spatial specificity was enhanced in datasets acquired at higher field strength, with cryoprobes, in ventilated animals, and under medetomidine-isoflurane combination sedation. Our work describes a set of representative RSNs in the mouse brain and highlights key experimental parameters that can critically guide the design and analysis of future rodent rsfMRI investigations.

Abstract

MRI has potential as a translational approach from rodents to humans. However, given that mouse functional MRI (fMRI) uses anesthetics for suppression of motion, it has been difficult to directly compare the result of fMRI in "unconsciousness" disease model mice with that in "consciousness" patients. We develop awake fMRI to investigate brain function in 15q dup mice, a copy number variation model of autism. Compared to wild-type mice, we find that 15q dup is associated with whole-brain functional hypoconnectivity and diminished fMRI responses to odors of stranger mice. Ex vivo diffusion MRI reveals widespread anomalies in white matter ultrastructure in 15q dup mice, suggesting a putative anatomical substrate for these functional hypoconnectivity. We show that d-cycloserine (DCS) treatment partially normalizes these anormalies in the frontal cortex of 15q dup mice and rescues some social behaviors. Our results demonstrate the utility of awake rodent fMRI and provide a rationale for further investigation of DCS therapy.

47. Finishing Line Scheduling in the steel industry

Abstract

A new solution for large-scale scheduling in the steelmaking industry, called Finishing Line Scheduling (FLS), is described. FLS in a major steel mill is a task to create production campaigns (specific production runs) for steel coils on four continuous processes for a one-month horizon. Two process flows are involved in FLS, and the balancing of the two process flows requires resolving conflicts of due dates. There are also various constraints along the timeline for each process with respect to sequences of campaigns and coils. The two types of constraints--along process flows and timelines--make the FLS problem very complex. We have developed a high-performance solution for this problem as follows: Input coils are clustered by two clustering algorithms to reduce the complexity and size of the problem: Campaigns are created for each process from downstream to upstream processes, while propagating upward the process timings of the clusters. Timing inconsistencies along the process flows are then repaired by scheduling downward. Finally, coils are sequenced within each campaign. The FLS system enabled a steel mill to expand its scheduling horizon from a few days to one month, and to improve decision frequency from monthly to daily.

Abstract

The investigation of the surface morphology changes for Si surfaces treated with CF4/O2 reveals that the rough Si surfaces can be smoothened and Si trench corners can be rounded using CF4/O2 down-flow etching. Down-flow discharged CF4/O2 gas mixture in high O2 concentration etches the Si surface on which the SiFxOy forms. Fluorine atoms are prevented from reacting with Si by the formation of a thick SiFxOy layer at the concave part of the surface.

49. Damage-free selective etching of Si native oxides using NH3/NF3 and SF6/H2O down-flow etching

Abstract

NH3/NF3 and SF6/H2O down-flow etching are used in obtaining damage-free selective etching of Si native oxides against Si. The wafer is covered with a film in the NH3/NF3 etching, and after its removal by heating above 100 degrees centigrade, only SiO2 with an extremely high selectivity with respect to Si is found to be etched. For SF6/H2O microwave discharge, selective etching of SiO2 is obtained. On the water surface, a film of liquid solution that has HF and H2SOX appeared. As wet etching occurs by an HF solution, the selective etching of SiO2 occurs by the dissolved HF.
