The use of vaccines have resulted in a remarkable improvement in global health. It has saved several lives, reduced treatment costs and raised the quality of animal and human lives. Current traditional vaccines came empirically with either vague or completely no knowledge of how they modulate our immune system. Even at the face of potential vaccine design advance, immune-related concerns (as seen with specific vulnerable populations, cases of emerging/re-emerging infectious disease, pathogens with complex lifecycle and antigenic variability, need for personalized vaccinations, and concerns for vaccines' immunological safety -specifically vaccine likelihood to trigger non-antigen-specific responses that may cause autoimmunity and vaccine allergy) are being raised. And these concerns have driven immunologists toward research for a better approach to vaccine design that will consider these challenges. Currently, immunoinformatics has paved the way for a better understanding of some infectious disease pathogenesis, diagnosis, immune system response and computational vaccinology. The importance of this immunoinformatics in the study of infectious diseases is diverse in terms of computational approaches used, but is united by common qualities related to host–pathogen relationship. Bioinformatics methods are also used to assign functions to uncharacterized genes which can be targeted as a candidate in vaccine design and can be a better approach toward the inclusion of women that are pregnant into vaccine trials and programs. The essence of this review is to give insight into the need to focus on novel computational, experimental and computation-driven experimental approaches for studying of host–pathogen interactions and thus making a case for its use in vaccine development.

This study was carried out to investigate the potability of vendor waters mostly used by the urban dwellers in Idi-Araba, Lagos State, Nigeria. Sixteen water samples were analyzed, eleven from water vendors and four from source (Pumped Wells). One water sample from Nigerian bottling company was used as a control standard. Chemical analysis of the various water samples showed that they contained little and insignificant amount of toxic metals such as cadmium, nickel, lead and silver. Microbiological studies showed that vendor water was more contaminated than the source water and the contamination involved indicator and pathogenic organisms. All the organisms found in source water and more were found in vendor water. Susceptibility studies showed that of all the antibiotics used, the quinolones (Ofloxacin and Ciprofloxacin) and also aminoglycoside (Gentamycin) were the most effective antibiotics against the isolated microorganisms while Cloxacillin was the least active. The results of this investigation therefore suggests that the use of vendor and source waters in Idi-Araba appear unsafe for human consumption and domestic use. There is therefore the need for government and relevant authorities/organizations to provide potable water, stage awareness campaign and mass education on hygienic principles amongst the inhabitants of Surulere and the water vendors in particular.

The entire world is now experiencing the challenge of COVID-19 infection. However, still very few evidence-based treatment options are available for the prevention and treatment of COVID-19 disease. The present review aims to summarize the publicly available information to give a comprehensive yet balanced scientific overview of all the fat-soluble vitamins concerning their role in SARS-CoV-2 virus infection. The roles of different fat-soluble vitamins and micronutrients in combating SARS-CoV-2 infection have been recently explored in several studies. There are various hypotheses to suggest their use to minimize the severity of COVID-19 infection. These vitamins are pivotal in the maintenance and modulation of innate and cell-mediated, and antibody-mediated immune responses. The data reported in recent literature demonstrate that deficiency in one or more of these vitamins compromises the patients’ immune response and makes them more vulnerable to viral infections and perhaps worse disease prognosis. Vitamins A, D, E, and K boost the body’s defense mechanism against COVID-19 infection and specifically prevent its complications such as cytokine storm and other inflammatory processes, leading to increased morbidity and mortality overemphasis. However, more detailed randomized double-blind clinical pieces of evidence are required to define the use of these supplements in preventing or reducing the severity of the COVID-19 infection.Keywords: SARS-CoV-2, COVID-19, fat-soluble vitamins, micronutrients, immune response

This review emphasizes the changing aspects of the COVID-19 disease, featuring the cytokine storm’s pathological processes. Furthermore, we briefly reviewed potential therapeutic agents that may modulate and alleviate cytokine storms. The literature exploration was made using PubMed, Embase, MEDLINE, Google scholar, and China National Knowledge Infrastructure databases to retrieve the most recent literature on the etiology, diagnostic markers, and the possible prophylactic and therapeutic options for the management of cytokine storm in patients hospitalized with COVID-19 disease. The causative agent, severe acute respiratory coronavirus-2 (SARS-CoV-2), continually threatens the efficiency of the immune system of the infected individuals. As the first responder, the innate immune system provides primary protection against COVID-19, affecting the disease’s progression, clinical outcome, and prognosis. Evidence suggests that the fatalities associated with COVID-19 are primarily due to hyper-inflammation and an aberrant immune function. Accordingly, the magnitude of the release of pro-inflammatory cytokines such as interleukin (IL)-1, (IL-6), and tumor necrosis alpha (TNF-α) significantly differentiate between mild and severe cases of COVID-19. The early prediction of a cytokine storm is made possible by several serum chemistry and hematological markers. The prompt use of these markers for diagnosis and the aggressive prevention and management of a cytokine release syndrome is critical in determining the level of morbidity and fatality associated with COVID-19. The prophylaxis and the rapid treatment of cytokine storm by clinicians will significantly enhance the fight against the dreaded COVID-19 disease.Keywords: COVID-19, SARS-CoV-2, cytokine storm, hyper-inflammation, fatality

Then, proximate and phytochemical analyses of the extracts were carried out using several methods previously reported in the literatures. The biochemical studies were also carried out as reported in previous literatures.Results: The ethanolic leaves extract contains appreciable quantities of phytonutrients and micronutrients as well as phytochemical constituents. The LD50 of the extract was determined to be 3800mg/kg body weight. There was a dose-dependent elevation of the blood sugar in comparison with the control. There was no significant increase on the bilirubin and liver enzymes levels or on the haematological parameters of the lab animals. The extract significantly elevated the lipid profile (P value < 0.0001), the glomerular filtration rate (increased creatinine and blood urea levels – P value < 0.0001), the serum electrolytes and the animals’ weight. There was a significant decrease in the anion gap (P value < 0.01).Conclusion: The ethanol leaf extract of Justicia secunda has negative cardiac and renal effects on Wistar rats, causing increased lipid profile values, creatinine and blood urea levels in the experimental animals compared with control. The LD50 is below the safety level. Caution should be exercised as the biochemical profiles of cardiac and renal effects do not seem to be promising and the LD50 is below the safety level.Keywords: liver function, kidney function, cardiac function, serum electrolytes, Justicia secunda

The active pharmaceutical ingredients quantitative assay showed that all the brands of ciprofloxacin infusion were between the 95% and 105% limit of label claim while one metronidazole brand has < 95– 110% limit label claim. Six brands each of the two drugs evaluated fall below the acceptable pH range [ciprofloxacin (3.5– 4.6) and metronidazole (4.8– 5.2)], while the other two brands of both drugs passed the test. In the antibacterial study, Pseudomonas aeruginosa and Escherichia coli were susceptible to the ciprofloxacin (5 μg). However, Salmonella typhi recorded inhibition zone diameters within resistant and intermediate range. Peptostrepococcus spp was susceptible (at minimum inhibitory concentrations of 100 μg/mL) to all the brands of metronidazole, while none of the brands were effective on Lactobacillus spp. All the brands passed the test for particulate contamination. The particles size range was < 10μm.Conclusion: About eighty-one percent (81.25%) of the infusions have acceptable good microbiological quality. However, 18.75% that failed the tests is a concern knowing that these are lifesaving products.Keywords: intravenous infusions, quality assessment, ciprofloxacin, metronidazole, quality medicines

Endophytic microorganisms are the storehouse of novel secondary metabolites that can serve as an excellent source of drugs for different ailment. In this study, we isolated endophytic fungi associated withAzadirachta indicacollected from Agulu, Awka, Anambra State, South Eastern Nigeria, identified some of their bioactive secondary metabolites and evaluated their antimicrobial properties. The endophytic fungi were isolated following standard method and the pure culture of the isolates identified morphologically and molecularly. The fungi were subjected to solid-state fermentation on rice medium and the metabolites were extracted using ethyl acetate. The fungal crude extracts were screened for antimicrobial activity. The extracts were further subjected to high-performance liquid chromatography-diode-array detection (HPLC-DAD) analysis for the identification of the bioactive compounds. A total of four endophytic fungi identified asAureobasidiumsp. (AIL1), Sodariasp. (AIL3),Aspergillussp. (AIS1) andPenicilliumsp. (AIS2) were isolated from theAzadirachta indica. The crude extracts at 1 mg/mL, showed varying degrees of antimicrobial activity against the test fungi and bacteria Iinhibtion zone diameter (IZD) ranged from 2 to 11 mm. The minimum inhibitory concentrations (MICs) of the fungal extracts ranged from 0.0625 to 0.25 mg/mL. The HPLC-DAD analysis revealed the presence Glucobrassicin, Ferulic acid, 4-methoxybenzaldehyde, 12-Hydroxy-16-scalaren, 12-O-deacetyl-12-epi-scalarin, Ixoside, Citreodrimene F, and Cytosporin D, some of which has been reported to have shown antimicrobial activity.Azadirachta indicapossess sundry groups of fungal endophytes that contain secondary metabolites with interesting antimicrobial properties that could be explored in drug discovery.

Objective: To check the effects of the vaccines on the hematopoietic system and weight of mice after immunization. Methods: The study was done with the Expanded Programme on Immunization vaccines donated by the Ministries of Health of Abia and Imo States of Nigeria. The vaccines were collected from the cold-chain stores and transported in vaccine carriers to the cold-chain facility in Nnamdi Azikiwe University Teaching Hospital within 3 hours of collection. They were used to immunize a total of 160 mice. The Ethics Committee of Nnamdi Azikiwe University Teaching Hospital, Nnewi of Anambra State, Nigeria approved the protocol. Results: Mice body weight changes test showed that the mice all had increased body weight at Days 3 and 7 post-immunization and none died during the 7 d post-immunization observation. The percentage weight gains of the mice compared with the control were 69%, 70%, 64%, 63%, 65% and 68% for oral polio vaccine, diphtheria-pertussis-tetanus, bacillus Calmette–Guerin, measles, yellow fever and hepatitis B vaccines respectively collected from Imo State. The mice immunized with oral polio vaccine, pentavalent, bacillus Calmette–Guerin, measles, yellow fever and hepatitis B vaccines collected from Abia State had 123%, 114%, 121%, 116%, 142% and 119% weight gain respectively compared with the control. Leukocytosis promoting toxicity test showed that none of the vaccines was able to induce proliferation of leukocytes up to ten folds. Leukopenic toxicity test showed that all the vaccines had an leukopenic toxicity test value higher than 80% of the control (physiological saline). Conclusions: The vaccine samples tested were safe and did not affect the hematopoietic system adversely. The storage conditions of the vaccines in the States’ cold-chain stores had not compromised the safety of the vaccines.

Background: There has been a recent increase in the number of human Onchocerciasis cases in hospitals in Ebonyi Central. This study therefore sets out to determine the prevalence of the disease in Ebonyi North and Central Senatorial Zones of Ebonyi State as well as determine the age groups and sex most affected. Methods: This study was carried out in Ebonyi North and Central Senatorial Zones of Ebonyi State between January 2011 and April 2012, among subjects of 20-59 years. Diagnosis was by skin examination for onchocercal lesions, ocular examination for ocular lesion and parasitological examination for the presence of microfilariae of Onchocerca volvulus in skin biopsies and adult filariae in excised nodules. Results: Out of the 250 persons randomly examined in each zone, 82 (32.8%) persons in Ebonyi Central zone comprising of 60 (38.7%) males and 22 (23.1%) females were found positive for Onchocerca volvulus while 73 (29.2%) persons in Ebonyi North Zone comprising 48 (32.0%) males and 25 (25.0%) females. Chi-square test shows that male subjects had significantly higher diseases prevalence (p < 0.05) than their female counterparts in the two zones but combined prevalence among male and female do not differ significantly in the two zones. The age group most affected was 50-59 years with the prevalence of 40.0% in Ebonyi Central and 38.6% in Ebonyi North Zones. Out of the 155 (30.8%) persons infected in the two zones, 28 (5.6%) males and 14 (2.8%) females had Onchocercal skin lesions, 57 (11.4%) males and 22 (4.4%) females had low vision. Partial blindness was observed in 16 (3.2%) males and 8 (1.6%) females. However, 7 (1.4%) males and 3 (0.6%) females had complete blindness. Farmers were mostly affected than any other occupational group. Conclusion: The findings obtained from this work suggested that Ebonyi Central and North Senatorial Zones are Onchocerciasis hypo-endemic Zones having the prevalence rate of 32.8% and 29.2% respectively. Sex, age and occupation are related to Onchocerciasis infections in both Zones. Males are more infected than females. Massive distribution of insecticides to destroy the vector Simulium damnosum (black fly), provision of adequate antiflarial drugs - ivermectin and mass health education targeted on preventive measures are highly recommended.
