Abstract

The ESA space mission Euclid was launched on July 1st, 2023 and is undergoing its science verification phase. In this invited review we show that Euclid means a before and an after for our understanding of ultra-cool dwarfs and substellar-mass objects and their connections with stars, exoplanets and the Milky Way. Euclid enables the study with unprecedented statistical significance a very large ensemble of ultracool dwarfs, the identification of new types of substellar objects, and the determination of the substellar binary fraction and the Initial Mass Function (IMF) in diverse galactic environments from the nearest stellar nurseries to the ancient relics of Galactic formation., Comment: 8 pages

Abstract

The Tucana-Horologium association is one of the closest young stellar groups to the Sun and despite the close proximity its age is still debated in the literature. We take advantage of the state-of-the-art astrometry delivered by the third data release of the Gaia space mission combined with precise radial velocity measurements obtained from high-resolution spectroscopy to investigate the dynamical age of the association. We perform an extensive traceback analysis using a combination of different samples of cluster members, metrics to evaluate the minimum size of the association in the past and models for the galactic potential to integrate the stellar orbits back in time. The dynamical age of $38.5^{+1.6}_{-8.0}$ Myr that we derive in this paper is consistent with the various age estimates obtained from isochrone fitting in the literature (30-50 Myr) and reconciles, for the first time, the dynamical age of the Tucana-Horologium association with the age obtained from lithium depletion ($\sim40$ Myr). Our results are independent from stellar models and represent one more step towards constructing a self-consistent age scale for the young stellar groups of the Solar neighbourhood based on the 3D space motion of the stars., Comment: 13 pages, 6 figures, accepted for publication in MNRAS

Abstract

The flow of information in high-energy collisions has been recently investigated by various groups: this includes the entanglement entropy of the proton becoming classical information entropy of pdfs, jet splitting affecting entropy, or the entropy distribution in hadron decays. Here we examine fragmentation functions in this context, including their entropy as probability distributions, and propose it as one convenient number to characterize progress in their extraction. Several groups have recently investigated the flow of information in high-energy collisions, from the entanglement entropy of the proton yielding classical Shannon entropy of its parton distribution functions (pdfs), through jet splitting generating entropy, to the entropy distribution in hadron decays. Lacking in the literature is a discussion of the information entropy of fragmentation functions (FFs) in the instances where they can be considered as probability distributions, and we here provide it. We find that this entropy is a single, convenient number to characterize future progress in the extraction of fragmentation functions. We also deploy the related Kullback-Leibler divergence between two distributions to assess existing relations among FFs and parton distribution functions (pdfs) such as that of Barone, Drago and Ma. Abstract

The nature and origin of free-floating planets (FFPs) are still largely unconstrained because of a lack of large homogeneous samples to enable a statistical analysis of their properties. So far, most FFPs have been discovered using indirect methods; microlensing surveys have proved particularly successful to detect these objects down to a few Earth masses. However, the ephemeral nature of microlensing events prevents any follow-up observations and individual characterization. Several studies have identified FFPs in young stellar clusters and the Galactic field but their samples are small or heterogeneous in age and origin. Here we report the discovery of between 70 and 170 FFPs (depending on the assumed age) in the region encompassing Upper Scorpius and Ophiuchus, the closest young OB association to the Sun. We found an excess of FFPs by a factor of up to seven compared with core-collapse model predictions, demonstrating that other formation mechanisms may be at work. We estimate that ejection from planetary systems might have a contribution comparable to that of core-collapse in the formation of FFPs. 25. Ontogeny of morphological variations in the vertebral column: Prevalence and bony variability in young Spanish children

Abstract

Pre- and postnatal development and variability in discrete vertebral traits have been poorly described in embryonic studies. Numerous authors have reported that these variations are observable only from adolescence; scientific publications on the vertebrae of fetuses and infants are scarce. Thus, the aims of this study were to (1) describe the ontogeny and variability of anatomical variations in the vertebral column of a Spanish infant population and (2) analyze the frequency and relationship between sex, age, and intertrait variables. A total of 4728 vertebrae from 197 skeletons were studied. The age at death ranged from 22 intrauterine weeks to 8 years. Twenty morphological traits related to vertebral column development were analyzed. A descriptive statistical analysis was performed, and the chi-square test was used to measure the relationship between sex, age, and intertrait variables. We observed that 88.32% of skeletons expressed discrete traits along the spine. In fetuses, the double transverse foramen and unclosed transverse process of the axis were the most prevalent traits. In infants older than one year, the appearance of the L5 cleft neural arch, unclosed transverse process of the atlas, and craniocaudal shifts were frequent. A significant result was found between sex and the unclosed transverse process in the axis. The intertrait relationship was significant for all traits that shared the same embryonic structure. 36. Novel approaches for vaccination against equine viral arteritis

Abstract

This thesis explores the potential of two prototype ('marker') vaccines with differential diagnostic capability for improved control of equine viral arteritis. The rational for using a sub-unit adjuvanted vaccine and a recombinant live virus vaccine, analysis of immune responses they induce and assessment of the protection against infection in experimental conditions is described and discussed in this thesis. Also, the development of new methods for measuring cytotoxic T-lymphocyte responses to EAV is described, increasing the repertoire of immunological assays for EAV infections. Finally, attempts to determine the antigenic specificity of the cytotoxic T-lymphocyte response have been taken. The sub-unit vaccination approach developed in this project derives from the findings of Dr. Ewan Chirnside et al (1995a). This strategy has been developed further. A recombinant protein expressed in E.coli comprising the putative ectodomain of the large envelope glycoprotein (GL) of equine arteritis virus (EAV) was used as a vaccine in ponies resulting in the induction of virus neutralising antibody (VNAb) responses which are comparable in titre to those induced by inactivated whole virus vaccines. The protection afforded by this vaccine against virulent challenge, which correlated with the pre-challenge VNAh titres, was characterised by reduction of nasal virus excretion and viraemia. A second 'marker' vaccination strategy for EAV was explored. A recombinant live virus containing a deletion of a neutralisation domain on GL was constructed by manipulations of the cDNA full length clone of EAV. A synthetic peptide (Peptide-1) whose sequence is contained within the deleted fragment is the basis of an already developed antibody capture diagnostic ELISA test for EAV. The deletion mutant virus (EAV-GlA) showed in vitro slightly slower growth kinetics than wild type EAV and when administered by the nasopharyngeal route to ponies these experienced an asymptomatic infection, although virus was recovered from blood and nasal secretions. Both inoculated ponies developed a discriminating antibody response which lacked the Peptide-1 antigen specificity. The serum VNAb response was of high titre against EAV-GlA but of low titre against wild type EAV. Yet, the EAV-AGl inoculated ponies were highly protected against virulent challenge. This novel approach shows great potential as a marker vaccine. Safety and duration of immunity need to be investigated. The structure of VNAb epitopes and effector mechanisms of immunity stimulated by administration of EAV-AGl deserve also attention. The serum VNAb response that invariably develops in the infected animal lasts for many months or years and is believed to play an important role in virus clearance. However, very little was known about the cellular immunity against EAV due the lack of methods to evaluate these immune responses. Methods to detect cytotoxic T-lymphocyte (CTL) precursors in the peripheral blood of EAV convalescent ponies are described using a 51Cr release cytolysis assay. Primary equine dermal cells, used as CTL targets, were shown to express MHC-I but not MHC-II, retain 51Cr efficiently and support EAV replication. Peripheral blood mononuclear cells (PBMC) collected from EAV convalescent ponies that have been incubated with or without live EAV were used as effectors. EAV induced PBMC cultures showed evidence for expansion and activation of lymphoblasts, with an increase in the CD8+/CD4+ ratio in comparison to mock-induced PBMC. The cytotoxicity induced by EAV stimulated PBMC was virus specific, showed genetic restriction, was mediated by CD8+ T lymphocytes and could be detected for periods between 4 months to more than 1 year post-infection. These findings and methods will hopefully contribute to understanding of virus-host interactions in horses, in particular the mechanisms of virus clearance occurring during EAV infection. The last part of this project described efforts to try to identify proteins serving as targets for EAV specific CTL's. Target cells transiently expressing EAV0RF7 were used in the standard CTL assay developed during the course of these studies (Chapter 4). Cloning of EAV0RF7 into the mammalian expression vector pcDNA3 for subsequent expression in different cell lines is described. Whilst, expression of EAV N protein in C0S7 and RK-13 cells using plasmid transfection techniques was achieved, EDC's failed to express N efficiently. Unlike plasmid transfection, baculovirus transduction appeared to be a suitable method to achieve protein expression in a high proportion of EDC. The levels of expression of EAV N antigen in transduced EDC, which occurred without the loss of their original morphology, suggested a priori that this method was adequate to identify CTL targets. Time and resources constraints limited the number of experiments performed but sufficient progress was made to indicate that this expression technique has good potential for examining EAV CTL targets. In this research project two novel approaches have been used for the generation of marker vaccines against EAV and methods to evaluate cytotoxic T lymphocyte responses developed. The essential findings of the thesis provide new and interesting information about EAV vaccinology/immunology that now provide the basis for rational approaches to be undertaken to disease prevention.

Abstract

Background: Immunodeficient patients (IDPs) are at higher risk of contracting severe COVID-19 disease. Targeted vaccination strategies have been implemented to enhance vaccine-induced protection. In this population however, clinical effectiveness is variable and duration of protection unknown. Objective: To understand the cellular and humoral immune responses to mRNA and adenoviral vectored COVID-19 vaccines in patients with immunodeficiency. Methods: Immune responses to SARS-COV-2 spike were assessed after two doses of homologous ChAdOx1-nCoV-19 or BNT162b2 vaccines in 112 infection-naïve IDPs and 131 healthy health care workers (HCWs) as controls. Predictors of vaccine responsiveness were investigated. Results: Immune responses to vaccination were low, and viral neutralisation by antibody not detected despite high titre binding responses in many IDPs. In those responding, the frequency of specific T-cell responses in IDPs was similar to controls whilst antibody responses were lower. Sustained vaccine specific differences were identified: T-cell responses were greater in ChAdOx1-nCoV-19 compared with BNT162b2 immunised IDPs and antibody binding and neutralisation was greater in all cohorts immunised with BNT162b2. The positive correlation between T-cell and antibody responses was weak and increased with subsequent vaccination. Conclusion: Immunodeficient patients have impaired immune responses to mRNA and viral vector COVID-19 vaccines that appear influenced by vaccine formulation. Understanding the relative roles of T-cell and antibody mediated protection and potential of heterologous prime and boost immunization protocols is needed to optimise the vaccination approach in these high-risk groups.

Abstract

Background: The virus neutralization assay is a principal method to assess the efficacy of antibodies in blocking viral entry. Due to biosafety handling requirements of viruses classified as hazard group 3 or 4, pseudotyped viruses can be used as a safer alternative. However, it is often queried how well the results derived from pseudotyped viruses correlate with authentic virus. This systematic review and meta-analysis was designed to comprehensively evaluate the correlation between the two assays. Methods: Using PubMed and Google Scholar, reports that incorporated neutralisation assays with both pseudotyped virus, authentic virus, and the application of a mathematical formula to assess the relationship between the results, were selected for review. Our searches identified 67 reports, of which 22 underwent a three-level meta-analysis. Results: The three-level meta-analysis revealed a high level of correlation between pseudotyped viruses and authentic viruses when used in an neutralisation assay. Reports that were not included in the meta-analysis also showed a high degree of correlation, with the exception of lentiviral-based pseudotyped Ebola viruses. Conclusion: Pseudotyped viruses identified in this report can be used as a surrogate for authentic virus, though care must be taken in considering which pseudotype core to use when generating new uncharacterised pseudotyped viruses.

Abstract

Despite the success of COVID-19 vaccination programmes in reducing morbidity and mortality, a substantial number of individuals in the general population respond poorly to SARS-CoV-2 vaccination. Furthermore, only about 20% of individuals throughout low-income countries have received their first dose of the SARS-CoV-2 vaccine. Neutralising antibodies are a key correlate to protection from COVID-19, with booster vaccines offered to increase protection from new variants. However, poor responsiveness to immunisation increases the risk of infection and disease. The CDC reported that patients with compromised immune systems accounted for about 12% of adult COVID-19 hospital admissions and had higher rates of intensive care admissions and in-hospital deaths compared with non-immunocompromised inpatients, in both vaccinated and unvaccinated populations. Although a heterogeneous group, the majority of patients with immunodeficiency show variable and weaker antibody-mediated responses post-vaccination to SARS-CoV-2 than individuals without immunodeficiency. In March, 2022, a UK report revealed 45·5% of immunocompromised individuals received a booster vaccination dose. However, without immune monitoring it remains unclear which individuals within this high-risk population have sufficient immunity.

Abstract

Corona-Australis is one of the nearest regions to the Sun with active star formation, but despite the close proximity (d~150 pc) our current picture of this starburst is not yet complete. Our team has recently revised the census of Corona-Australis stars using Gaia- DR2 data and discovered a distributed population of young stars located in the Galactic north of the dark clouds that is twice as numerous as the historically known stellar population concentrated around the densest cores of the region. Here, we build on the current census of stars in this region by combining the Gaia-EDR3 catalogue with astrometric and photometric ground-based data delivered by the Cosmic-DANCe project. Abstract

The 4MOST Stellar Clusters Survey will target essentially all the Galactic globular and open clusters and star-forming regions accessible to 4MOST (about 120 globulars, 1800 open clusters and 80 star-forming regions). This will: shed light on how clusters form, evolve, dissolve, and populate the Milky Way; calibrate complex physics that affects stellar evolution, on which our ability to measure accurate ages ultimately rests; and evaluate the contribution of star clusters to the formation and evolution of the individual Galactic components with unparalleled statistics., Published in The Messenger vol. 190, pp. 13-16, March 2023.
