Corais possuem uma relação de simbiose com zooxantelas, microalgas associadas ao tecido do animal, responsáveis pela maior parte de seu ganho energético diário. O aquecimento da água do oceano rompe a simbiose, fenômeno conhecido por branqueamento. Sem sua principal fonte nutricional, corais dependem da heterotrofia para sobreviver, o que pode levar a estagnação do ciclo reprodutivo. Há corais, porém, que conseguem manter a gametogênese mesmo quando branqueados, entretanto a fisiologia desses gametas em tal situação é desconhecida. Neste trabalho quantificamos e avaliamos o balanço oxidativo de oócitos liberados de colônias saudáveis de Mussismilia braziliensis; além de oócitos de três colônias de Mussismilia harttii com diferentes estados de saúde: uma branqueada, uma parcialmente branqueada e uma saudável. Comparamos também, o balanço oxidativo entre as colônias saudáveis das duas espécies. Os oócitos foram congelados ao longo de uma curva de tempo pós desova (0, 5 e 10 h) para avaliar o balanço oxidativo durante o tempo de vida celular e o esforço celular para se manter saudável. Mediram-se os níveis de proteínas totais, a atividade das enzimas antioxidantes superóxido dismutase (SOD) e catalase (CAT) e os níveis de lipoperoxidação (TBARS), um marcador de dano lipídico. Em M. braziliensis a atividade da SOD aumentou entre 5 e 10h pós desova, assim como a lipoperoxidação entre 0 e 10h. Ao se comparar os oócitos advindos de corais com diferentes estados de saúde em M. harttii, não foi encontrada diferença entre os parâmetros estudados durante o tempo pós desova, com exceção da SOD em 0h. Estes organismos parecem preservar a qualidade das células gaméticas femininas mesmo 10h pós desova. Esta resposta pode estar relacionada a intensificação da heterotrofia garantindo o aporte nutricional e assim, o esforço reprodutivo e a qualidade dos gametas mesmo em corais branqueados. In this work, we quantify and evaluate the oxidative balance of oocytes released from healthy colonies of Mussismilia braziliensis; in addition to oocytes from three colonies of Mussismilia harttii with different health conditions: one bleached, one partially bleached and one healthy. We also compared the oxidative balance between healthy colonies of the two species. Oocytes were frozen along a post-spawning time curve (0, 5 and 10 h) to assess oxidative balance during cell life and cell effort to remain healthy. Total protein levels, the activity of the antioxidant enzymes superoxide dismutase (SOD) and catalase (CAT) and the levels of lipoperoxidation (TBARS), a marker of lipid damage, were measured. In M. braziliensis, SOD activity increased between 5 and 10h after spawning, as well as lipoperoxidation between 0 and 10h. When comparing the oocytes from corals with different health conditions in M. harttii, no difference was found between the parameters studied during the post-spawning time, except for SOD at 0h. These organisms seem to preserve the quality of female gametic cells even after 10 hours of spawning. This response may be related to the intensification of heterotrophy, ensuring nutritional support and thus reproductive effort and quality of gametes even in bleached corals. O uso de agrotóxicos começou a ser disseminado na década de 60 com a Revolução Verde. Entretanto, não se sabia quais seriam as consequências desse uso em longo prazo. Ao ser aplicado, resíduos de agrotóxico acabam atingindo animais não-alvo, se infiltrando no solo e atingindo a água. Como consequência, os agrotóxicos podem bioacumular e/ou impactar organismos não-alvo causando danos bioquímicos e fisiológicos antes mesmo de alterações morfológicas visíveis. Quando se trata de ecotoxicidade, os répteis são os animais menos estudados e encontram-se atualmente em declínio global devido, principalmente, a degradação do hábitat ao qual ocupam. O lagarto Salvator merianae tem ampla distribuição na América do Sul e é encontrado frequentemente em áreas de cultivo agrícola, além de seu período reprodutivo coincidir com a aplicação de agrotóxico nas culturas de primavera/verão. Os objetivos deste estudo foram comparar possíveis alterações em parâmetros bioquímicos e fisiológicos de S. merianae coletados em uma área exposta a agrotóxicos e uma área de referência. Para tal, quantificamos diferentes marcadores sanguíneos: albumina (ALB), alanina aminotransferase (ALT), fosfatase alcalina (FAL), gama glutamil transpeptidase (GGT), glicose sérica (GLI), proteínas totais (Pt), ácido úrico (AU), triglicerídeos (TGL), VLDL, corticosterona, TBARS, glutationa-S-transferase (GST), e da atividade da superóxido dismutase (SOD) e catalase (CAT). Para isso, foram coletadas amostras de sangue de 28 lagartos da espécie S. merianae na Estação Experimental Agronômica da UFRGS, no ano de 2019 e 2020, e fora do período de hibernação. Foi utilizado plasma para quantificarmos os marcadores de dano hepático, metabolismo intermediário e endócrino através de kits comerciais da BioTécnica, e para as análises de estresse oxidativo utilizamos eritrócitos sendo estes quantificados através de métodos espectrofotométricos padronizados para lagartos em nosso laboratório. O estudo sobre o mimetismo de Heliconius sempre recebeu muita atenção. Novas relações entre as espécies do gênero são descobertas em primorosos estudos. No entanto, quanto a Fisiologia do grupo existem poucas investigações. A relação estabelecida entre as espécies de Heliconius formando anéis miméticos ocorre tanto entre espécies pertencentes ao mesmo clado, como em espécies distantes filogeneticamente. Nesta Tese estudamos três espécies de Heliconius do sul do Brasil relacionando a Fisiologia e a morfologia com o mimetismo. Buscamos saber se o mimetismo existente entre o par mimético, Heliconius besckei (clado silvaniforme) e Heliconius erato phyllis (clado de acasalamento pupal) permanece em outras características além do padrão de coloração da asa. Além disso, investigamos se H. besckei diverge nestes mesmos parâmetros fisiológicos e morfológicos da espécie H. ethilla narcaea, que pertence também ao clado silvaniforme e assim como H. e. phyllis é simpátrica com H. besckei no sul do Brasil. Foram utilizadas metodologias de análise ultraestrutural da musculatura do tórax das borboletas, assim como também analisamos marcadores bioquímicos ligados ao balanço oxidativo e ao metabolismo intermediário. Para análise morfológica, utilizamos morfometria geométrica nas asas das três espécies para avaliar a forma e o tamanho. Nossos resultados mostraram que a espécie H. besckei convergiu com seu par mimético H. erato phyllis, para além do padrão de coloração, no tipo de voo, no tamanho e forma das asas anteriores, na massa corporal (tórax e abdômen), além de alguns marcadores do balanço oxidativo (TBARS, SOD e GST) e do metabolismo intermediário (conteúdo de proteínas totais, lipídios totais, triglicerídeos, colesterol e glicogênio). No entanto, devido às restrições à ambientes de altitudes maiores que 700m, as exigências fisiológicas de H. besckei para viver em simpatria com a espécie mimética parecem dispendiosas. In this thesis we studied three species of Heliconius from southern Brazil relating Physiology and morphology with mimicry. We sought to know whether the existing mimicry between the mimetic pair Heliconius besckei (silvaniform clade) and Heliconius erato phyllis (pupal mating clade) remains in other characteristics beyond the wing coloration pattern. In addition, we investigated whether H. besckei diverges in these same physiological and morphological parameters of the species H. ethilla narcaea, which also belongs to the silvaniform clade and as well as H. e. phyllis is sympatric with H. besckei in southern Brazil. Ultrastructural analysis methods were used to investigate the butterflies’ musculature of the thorax, as well as the study of biochemical markers linked to the oxidative balance and to the intermediate metabolism. For morphological analysis, we used geometric morphometry on the wings of the three species to evaluate shape and size. Our results showed that the species H. besckei converged with its mimetic pair H. erato phyllis, beyond the coloration pattern, in flight type, size and shape of anterior wings, body mass (thorax and abdomen), besides some markers of oxidative balance (TBARS, SOD and GST) and intermediate metabolism (total protein content, total lipids, triglycerides, cholesterol and glycogen). Os peixes de água doce são considerados um dos grupos mais ameaçados do mundo, visto que os ecossistemas aquáticos atualmente estão enfrentando constantes interferências de ações antrópicas, principalmente, com a intensificação da agricultura que tem impactado e descaracterizado diversos habitats de muitas espécies. Frente a isto, para a conservação da diversidade biológica nesses ambientes aquáticos é de extrema importância obter informações sobre as espécies constituintes destes habitats. Desta forma, ferramentas fisiológicas podem ser utilizadas para caracterizar a diversidade biológica e suas implicações ecológicas, e abranger também, as respostas dos organismos às alterações ambientais e agentes estressores. Os peixes anuais são organismos que vivem em áreas úmidas temporárias que secam sazonalmente. Devido a isto, apresentam um ciclo de vida curto e rápido, mostrando um grande potencial para serem utilizados como modelos biológicos para diversas pesquisas, principalmente, aquelas ligadas ao estudo de envelhecimento. O objetivo do presente trabalho foi analisar marcadores fisiológicos relacionados ao balanço redox do peixe anual Cynopoecilus fulgens, endêmico da região sul do Brasil, com variáveis ambientais e estimativas do período reprodutivo. Foram coletados 443 indivíduos compreendendo juvenis, adultos e senis, entre o mês de junho e novembro de 2016, nos arredores do Parque Nacional da Lagoa do Peixe (PNLP), localizado na Planície Costeira Externa do Rio Grande do Sul, em três diferentes unidades amostrais (P1, P2 e P3). Estes animais foram eutanasiados em campo e em laboratório foram medidos (comprimento padrão) e pesados (massa corporal). Após, foram homogeneizados para a realização dos ensaios da Superóxido Dismutase, Catalase, Glutationa Peroxidase e Glutationa S-transferase, e para determinação dos níveis de Lipoperoxidação, através do TBARS, Proteínas Carboniladas e Proteínas Totais. Os resultados mostraram que machos e fêmeas de C. fulgens empregam, Freshwater fish are considered one of the most threatened groups in the world, as aquatic ecosystems are currently facing constant interference from anthropic actions, mainly with the intensification of agriculture that has impacted and de-characterized diverse habitats of many species. To conserve biological diversity in these aquatic environments, it is extremely important to obtain information about the species that constitute these habitats. In this way, physiological tools can be used to characterized biological diversity and its ecological implications, and also include organisms responses to environmental changes and stressors. Annual fish are organisms that live in temporary wetlands that dry out seasonally. Due to this, the life cycle is considered short and fast presenting great potential as biological models for several researches, mainly those related to the study of aging. The aim of the present study was to analyze physiological markers related to the redox balance of the annual fish Cynopoecilus fulgens, endemic to southern Brazil, with environmental variables and estimates of the reproductive period. A total of 443 individuals were collected, including juveniles, adults and senile, between June to November 2016, around the Parque Nacional da Lagoa do Peixe, located in the Coastal Plain of Rio Grande do Sul, in three sampling units (P1, P2 and P3). These animals were euthanized in the field and in the laboratory were measured and weighed. O incremento no uso de pesticidas vem ocasionando a degradação de diferentes ecossistemas, sobretudo aquáticos. Apesar de possuírem aplicações específicas a determinados organismos, sabe-se que os agrotóxicos são capazes de ocasionar alterações metabólicas e funcionais em diversos organismos não alvo. Os anfíbios constituem a Classe de vertebrados mais ameaçada globalmente, sendo que os declínios populacionais observados no mundo todo são decorrentes da ação sinérgica de diversos fatores, incluindo a fragmentação e degradação de habitat, principalmente para fins agrícolas, a exposição a contaminantes derivados destas atividades, dentre outros. Neste contexto, Melanophryniscus admirabilis é uma espécie de sapo bufonídeo micro endêmica, descrita atualmente para apenas uma localidade no estado do Rio Grande do Sul (Brasil), e avaliada como Criticamente em Perigo de extinção, em nível regional, nacional e global. O alto grau de ameaça desta espécie deve-se a um conjunto de fatores, como a fragmentação e perda de qualidade do habitat, principalmente devido à conversão de áreas para agricultura e à larga utilização de agrotóxicos no entorno da área de ocorrência da espécie.Com isso, o objetivo do presente estudo foi analisar possíveis alterações em parâmetros metabólicos e do balanço oxidativo em homogeneizado total de girinos de M. admirabilis expostos a duas concentrações de formulação comercial contendo Sulfentrazone (Boral® 500 SC) e outras duas contendo Glifosato (Roundup® Original). Foram analisados os níveis totais de glicogênio, proteínas e ácido úrico; a possível ocorrência de lipoperoxidação (LPO) através dos níveis de TBARS; e o nível de atividade das enzimas Superóxido Dismutase (SOD), Catalase (CAT) e Glutationa S-Transferase (GST). Despite specific targets, agrochemicals are able to cause metabolic and functional alterations including in non-target organisms. Amphibians constitute the most globally threatened Class of vertebrates, and the population declines observed around the world are due to a synergy of different factors, including habitat degradation and fragmentation, mostly to agricultural purposes, and the exposure to contaminants derived of these activities, among others. Within this context, Melanophryniscus admirabilis is a micro endemic bufonid toad species, with occurrence registered to only one area in Rio Grande do Sul state (Brazil). This species has a critically endangered (CR) status, at regional, national and global levels. The great danger of extinction is due to several factors, including habitat fragmentation (especially for agricultural purposes) and the decreased habitat quality, mainly because of extensive use of agrochemicals in areas near of the natural habitat of this species. A pesca é uma das principais ameaças enfrentadas pelas tartarugas marinhas, porém os efeitos fisiológicos causados durante a captura nos diferentes artefatos de pesca são pouco estudados. O objetivo deste trabalho é estabelecer valores sanguíneos de referência para uma população de Chelonia mydas juvenis e compará-los com animais capturados incidentalmente pela pesca na mesma área, levando em conta a variação sazonal. Para tanto, coletou-se sangue de 96 tartarugas saudáveis, durante o verão e o inverno, nos três métodos de captura (mergulho livre, cerco flutuante e rede de emalhe). As amostras de sangue foram analisadas quanto as concentrações de glicose, lactato, proteínas totais, ácido úrico, albumina, lipídios totais, triglicerídeos, colesterol VLDL, colesterol total e corticosterona. Tartarugas capturadas no verão tiveram níveis mais elevados de glicose, proteínas totais, ácido úrico e albumina, e concentrações mais baixas de lipídios totais, em relação ao inverno. Apesar do cerco flutuante ser um mecanismo passivo de captura, seis dos dez parâmetros avaliados foram significativamente diferentes dos valores basais obtidos para espécie. A captura em redes de emalhe causou maiores perturbações que o cerco flutuante e todos os parâmetros avaliados diferiram-se significativamente dos valores obtidos para os animais capturados em mergulho livre (valores de referência). Esses resultados indicam que a captura incidental causa distúrbios nos parâmetros fisiológicos, ao quais podem ser utilizados como importantes marcadores em programas que visem auxiliar na reabilitação de tartarugas após a captura. Além disso, os valores de referência obtidos também podem ser utilizados para futuras comparações com populações da mesma espécie e com indivíduos afetados pelo estresse ou doenças., Fisheries is one of the main threats faced by sea turtles. However, literature relating the physiological effects of incidental capture by fishing gears is scarce. The aim of this study is to establish blood reference values for a juvenile population of Chelonia mydas and compare them with animals incidental captured by fisheries in the same area, taking account seasonal variation. Blood was collected from 96 healthy green sea turtles, during summer and winter, by three different methods (dive, pound net and gillnet). Blood samples were analyzed for glucose, lactate, total protein, uric acid, albumin, total lipids, triglycerides, VLDL cholesterol, total cholesterol and corticosterone concentrations. Turtles captured in summer had a significant increase in blood glucose, total protein, uric acid and albumin, and a decreased in total lipids compared with turtles captured in winter. Although pound net is a passive capture mechanism, six of ten parameters differed significantly from base-line values established for the species. Entanglement in gillnets caused greater perturbations than pound net and all parameters analyzed were significantly different from the values obtained for animals captured by diving (reference values). These data indicate that incidental capture causes significant physiological disturbance in sea turtles and the results obtained in this study are important to help in the rehabilitation of turtles after capture. Reference values can be used for future comparisons with populations of the same species and with unhealthy and stressed individuals.
