Abstract

The most common approaches for assigning weights to observations in minimum L1-norm (ML1) is to introduce weights of p or p\sqrt{p}, p being the weights vector of observations given by the inverse of variances. Hence, they do not take covariances into consideration, being appropriated only to independent observations. To work around this limitation, methods for decorrelation/unit-weight reduction of observations originally developed in the context of least squares (LS) have been applied for ML1, although this adaptation still requires further investigations. In this article, we presented a deeper investigation into the mentioned adaptation and proposed the new ML1 expressions that introduce weights for both independent and correlated observations; and compared their results with the usual approaches that ignore covariances. Experiments were performed in a leveling network geometry by means of Monte Carlo simulations considering three different scenarios: independent observations, observations with “weak” correlations, and observations with “strong” correlations. The main conclusions are: (1) in ML1 adjustment of independent observations, adaptation of LS techniques introduces weights proportional to p\sqrt{p} (but not p); (2) proposed formulations allowed covariances to influence parameters estimation, which is unfeasible with usual ML1 formulations; (3) introducing weighs of p provided the closest ML1 parameters estimation compared to that of LS in networks free of outliers; (4) weighs of p\sqrt{p} provided the highest successful rate in outlier identification with ML1. Conclusions (3) and (4) imply that introducing covariances in ML1 may adversely affect its performance in these two practical applications.

Abstract

Evolution has equipped poxvirus genomes with the coding capacity for several virus-host interaction products which interfere with host cell gene expression and protein function, creating an adequate intracellular environment for a productive infection. We show here that Vaccinia virus (VACV) induces the expression of the cellular transcription factor EGR-1 (early growth response-1) in Mouse Embryonic Fibroblasts (MEFs) through the MEK (mitogen-activated protein kinase (MAPK)/ERK)/ERK (extracellular signal-regulated kinases) pathway, from 3 to 12 h post infection (h.p.i.). By using starved egr-1 knockout (egr-1−/−) MEFs, we demonstrate that VACV replication is reduced by ~1 log in this cell line. Although western blotting and electron microscopy analyses revealed no difference in VACV gene expression or morphogenesis, the specific infectivity of VACV propagated in egr-1−/− MEFs was lower than virus propagated in wild type (WT) cells. This lower infectivity was due to decreased VACV DNA replication during the next cycle of infection. Taken together, these results revealed that EGR-1 appears to facilitate VACV replication in starved fibroblasts by affecting viral particles infectivity. [ABSTRACT FROM AUTHOR]

Abstract

Cabruca is an agroforest of cacao trees shaded by native forest trees. It is the predominant vegetation type throughout eastern part of the range of the golden-headed lion tamarins, Leontopithecus chrysomelas, an endangered primate endemic to Atlantic Forest. Understanding how lion tamarins use this agroforest is a conservation priority. To address this question, we documented the diet, home range size, group sizes and composition, density, number of litters and body condition of lion tamarins living in cabruca, and other habitats. Jackfruit, Artocarpus heterophyllus, was the most used species used by lion tamarins in cabruca and was widely available and used throughout the year. In cabruca, home range size was the smallest (22-28 ha) and density of lion tamarins was the highest (1.7 ind/ha) reported for the species. Group size averaged 7.4 individuals and was not significantly different among the vegetation types. In cabruca, groups produced one or two litters a year, and all litters were twins. Adult males in cabruca were significantly heavier than males in primary forest. Our study is the first to demonstrate that breeding groups of golden-headed lion tamarins can survive and reproduce entirely within cabruca agroforest. Jackfruit proved to be a keystone resource for lion tamarins in cabruca, and bromeliads were important as an animal prey foraging microhabitat. In cases where cabruca contains concentrated resources, such as jackfruit and bromeliads, lion tamarins may not only survive and reproduce but may fare better than in other forest types, at least for body condition and reproduction.
