Social network data offer interesting opportunities in urban studies. In this study, we used Twitter data to analyse city dynamics over the course of the day. Users of this social network were grouped according to city zone and time slot in order to analyse the daily dynamics of the city and the relationship between this and land use. First, daytime activity in each zone was compared with activity at night in order to determine which zones showed increased activity in each of the time slots. Then, typical Twitter activity profiles were obtained based on the predominant land use in each zone, indicating how land uses linked to activities were activated during the day, but at different rates depending on the type of land use. Lastly, a multiple regression analysis was performed to determine the influence of the different land uses on each of the major time slots (morning, afternoon, evening and night) through their changing coefficients. Activity tended to decrease throughout the day for most land uses (e.g. offices, education, health and transport), but remained constant in parks and increased in retail and residential zones. Our results show that social network data can be used to improve our understanding of the link between land use and urban dynamics., Comment: 15 pages, 6 figures

There is little knowledge available on the spatial behaviour of urban tourists, and yet tourists generate an enormous quantity of data (Big Data) when they visit cities. These data sources can be used to track their presence through their activities. The aim of this paper is to analyse the digital footprint of urban tourists through Big Data. Unlike other papers that use a single data source, this article examines three sources of data to reflect different tourism activities in cities: Panoramio (sightseeing), Foursquare (consumption), and Twitter (being connected). Tourist density in the three data sources is compared via maps, correlation analysis (OLS) and spatial self-correlation analysis (Global Moran's I statistic and LISA). Finally the data are integrated using cluster analysis and combining the spatial clusters identified in the LISA analysis in the different data sources. The results show that the data from the three activities are partly spatially redundant and partly complementary, and allow the characterisation of multifunction tourist spaces (with several activities) and spaces specialising in one or various activities (for example, sightseeing and consumption). The case study analysed (Madrid) reveals a significant presence of tourists in the city centre, and increasing specialisation from the centre outwards towards the periphery. The main conclusion of the paper is that it is not sufficient to use one data source to analyse the presence of tourists in cities; several must be used in a complementary manner., Comment: 20 pages,8 figures

As a consequence of the accelerated globalization process, today major cities all over the world are characterized by an increasing multiculturalism. The integration of immigrant communities may be affected by social polarization and spatial segregation. How are these dynamics evolving over time? To what extent the different policies launched to tackle these problems are working? These are critical questions traditionally addressed by studies based on surveys and census data. Such sources are safe to avoid spurious biases, but the data collection becomes an intensive and rather expensive work. Here, we conduct a comprehensive study on immigrant integration in 53 world cities by introducing an innovative approach: an analysis of the spatio-temporal communication patterns of immigrant and local communities based on language detection in Twitter and on novel metrics of spatial integration. This research evidences the usefulness of open big data to map mobility patterns in a medium-sized city. Motivated by the novel analysis that big data allow worldwide and in large metropolitan areas, we developed a methodology aiming to complement origin-destination surveys with \`a la carte spatial boundaries and updated data at a minimum cost. This paper validates the use of Twitter data to map the impact of public spaces on the different parts of the metropolitan area of Concepci\'on, Chile. Results have been validated by local experts and evidence the main mobility patterns towards spaces of social interaction like malls, leisure areas, parks and so on. In recent years, what has become known as collaborative consumption has undergone rapid expansion through peer-to-peer (P2P) platforms. In the field of tourism, a particularly notable example is that of Airbnb. This article analyses the spatial patterns of Airbnb in Barcelona and compares them with hotels and sightseeing spots. New sources of data, such as Airbnb listings and geolocated photographs are used. Analysis of bivariate spatial autocorrelation reveals a close spatial relationship between Airbnb and hotels, with a marked centre-periphery pattern, although Airbnb predominates around the main hotel axis and hotels predominate in some peripheral areas of the city. Another interesting finding is that Airbnb capitalises more on the advantages of proximity to the main tourist attractions of the city than does the hotel sector. Finally, it was possible to detect those parts of the city that have seen the greatest increase in pressure from tourism related to Airbnb's recent expansion., Comment: 17 pages, 11 figures, 11 tables

A common immune-related adverse event (irAE) with immune checkpoint inhibitors (ICIs) is thyroid dysfunction (TD-irAEs). The clinical presentation can be varied, and its association with prognosis remains unclear. We investigated the characteristics of TD-irAEs and their association with clinical outcomes among cancer patients treated with ICIs in a real-life setting. Response to treatment was assessed using RECIST v1.1. We calculated the probability of recurrence and survival associated with TD-irAEs using multivariable-adjusted regression and Cox proportional hazards models. In this single-center retrospective analysis, we included 238 patients (72% male) with a median age of 69.5 years. Primary tumors were melanoma (23.1%), lung (60.5%), or urothelial cancer (16.4%), treated with atezolizumab (23.1%), pembrolizumab (44.5%), ipilimumab (0.4%), and/or nivolumab (25.6%). Seventy (29%) patients developed TD-irAEs in a median time of 69 days (41-181). The incidence of TD-irAEs with combination therapy was higher than with monotherapy (67% vs 6.3%, P = 0.011). TD-irAE patients showed a higher objective response rate (ORR) than those without TD-irAEs (60% vs 42.3%, P = 0.013) and longer overall survival (OS) 45 vs 16 months, P < 0.006. Patients who developed TD-irAEs had a relative reduction of 77% (OR 0.23, 95% CI 0.11-0.47) in the risk of progression and of 47% in the risk of mortality (HR 0.53, 95% CI 0.36-0.80), independent of age, sex, primary tumor, or ICI regimen. TD-irAEs occur in nearly 30% of our patients receiving ICIs. In our analysis, TD-irAEs appeared to be associated with higher ORR and longer OS and showed a reduction in the risk of progression and mortality. [ABSTRACT FROM AUTHOR]

Pancreatic ductal adenocarcinoma (PDAC) is characterized by its poor prognosis. The current challenge remains the absence of predictive biomarkers. Cytokines are crucial factors in the pathogenesis and prognosis of PDAC. Furthermore, there is growing interest in differentiating between molecular subtypes of PDAC. The aim of our study is to evaluate the association between the analyzed cytokines and the molecular subtypes of PDAC and to determine their prognostic value. Cytokine levels were measured in 73 patients, and molecular subtypes were analyzed in 34 of these patients. Transforming Growth Factor Beta 2 (TGF-β2) levels were independently associated with the basal-like and null subtypes. In patients with locally advanced and metastatic PDAC, elevated levels of interleukin (IL)-1α, IL-1β, IL-6, IL-8, IL-9, and IL-15 were associated with a higher risk of progression during first-line treatment, and increased levels of IL-1β, IL-6, IL-8, IL-9, and IL-15 were related to increased mortality. Furthermore, a significant association was observed between higher percentiles of IL-6 and IL-8 and shorter progression-free survival (PFS) during first-line treatment, and between higher percentiles of IL-8 and shorter overall survival (OS). In the multivariate analysis, only elevated levels of IL-8 were independently associated with a higher risk of progression during first-line treatment and mortality. In conclusion, the results of our study suggest that cytokine expression varies according to the molecular subtype of PDAC and that cytokines also play a relevant role in patient prognosis. Plants and fungi use light and other signals to regulate development, growth, and metabolism. The fruiting bodies of the fungus Phycomyces blakesleeanus are single cells that react to environmental cues, including light, but the mechanisms are largely unknown [1]. The related fungus Mucor circinelloides is an opportunistic human pathogen that changes its mode of growth upon receipt of signals from the environment to facilitate pathogenesis [2]. Understanding how these organisms respond to environmental cues should provide insights into the mechanisms of sensory perception and signal transduction by a single eukaryotic cell, and their role in pathogenesis. We sequenced the genomes of P. blakesleeanus and M. circinelloides and show that they have been shaped by an extensive genome duplication or, most likely, a whole-genome duplication (WGD), which is rarely observed in fungi [3-6]. We show that the genome duplication has expanded gene families, including those involved in signal transduction, and that duplicated genes have specialized, as evidenced by differences in their regulation by light. The transcriptional response to light varies with the developmental stage and is still observed in a photoreceptor mutant of P. blakesleeanus. A phototropic mutant of P. blakesleeanus with a heterozygous mutation in the photoreceptor gene madA demonstrates that photosensor dosage is important for the magnitude of signal transduction. We conclude that the genome duplication provided the means to improve signal transduction for enhanced perception of environmental signals. Our results will help to understand the role of genome dynamics in the evolution of sensory perception in eukaryotes.

In recent years, what has become known as collaborative consumption has undergone rapid expansion through peer-to-peer (P2P) platforms. In the field of tourism, a particularly notable example is that of Airbnb. This article analyses the spatial patterns of Airbnb in Barcelona and compares them with hotels and sightseeing spots. New sources of data, such as Airbnb listings and geolocated photographs are used. Analysis of bivariate spatial autocorrelation reveals a close spatial relationship between Airbnb and hotels, with a marked centre-periphery pattern, although Airbnb predominates around the city's main hotel axis and hotels predominate in some peripheral areas of the city. Another interesting finding is that Airbnb capitalises more on the advantages of proximity to the city's main tourist attractions than does the hotel sector. Multiple regression analysis shows that the factors explaining location are also different for hotels and Airbnb. There is little knowledge available on the spatial behaviour of urban tourists, and yet tourists generate an enormous quantity of data when they visit cities. These data sources can be used to track their presence through their activities. The aim of this paper is to analyse the digital footprint of urban tourists through Big Data. Unlike other papers that use a single data source, this article examines three sources of data to reflect different tourism activities in cities: Panoramio (sightseeing), Foursquare (consumption), and Twitter (being connected-accommodation). The results show that the data from the three activities are partly spatially redundant and partly complementary, and allow the characterisation of multifunction tourist spaces and spaces specialising in one or various activities. - Abstract
As a consequence of the accelerated globalization process, today major cities all over the world are characterized by an increasing multiculturalism. The integration of immigrant communities may be affected by social polarization and spatial segregation. How are these dynamics evolving over time? To what extent the different policies launched to tackle these problems are working? These are critical questions traditionally addressed by studies based on surveys and census data. Such sources are safe to avoid spurious biases, but the data collection becomes an intensive and rather expensive work. Here, we conduct a comprehensive study on immigrant integration in 53 world cities by introducing an innovative approach: an analysis of the spatio-temporal communication patterns of immigrant and local communities based on language detection in Twitter and on novel metrics of spatial integration. Accessibility is essentially a dynamic concept. However, most studies on urban accessibility take a static approach, overlooking the fact that accessibility conditions change dramatically throughout the day. Due to their high spatial and temporal resolution, the new data sources (Big Data) offer new possibilities for the study of accessibility. The aim of this paper is to analyse urban accessibility considering its two components –the performance of the transport network and the attractiveness of the destinations– using a dynamic approach using data from TomTom and Twitter respectively. This allows us to obtain profiles that highlight the daily variations in accessibility in the city of Madrid, and identify the influence of congestion and the changes in location of the population. These profiles reveal significant variations according to transport zones.
