Abstract

Background/Aims: An epithelial to mesenchymal transition of retinal pigment epithelium (RPE) cells plays an important role in the formation of proliferative fibrocellular membranes in individuals with proliferative retinopathy. Prostaglandin E 2 (PGE 2 ) exhibits various physiological actions including both pro- and anti-inflammatory effects. We have now investigated the effect of PGE 2 on the epithelial-mesenchymal transition (EMT) of mouse RPE cells and fibrotic reactions in a type I collagen gel. Methods: Mouse RPE cells were incubated in three dimentional type 1 collagen gel culture system with transforming growth factor (TGF)-β2 (1ng/ml) in the absence or presence of PGE 2 at various concentration. The expression of α-smooth muscle actin as well as matrix metalloproteinase (MMP)-3, the phosphorylation of Smad2, paxillin were examined by immunoblot analysis. The release of MMP-2 and MMP-9 was examined by gelatin zymography. Results: PGE 2 inhibited the TGF-β2–induced contraction of collagen gels mediated by RPE cells in concentration dependent manner. The expression of α–smooth muscle actin and fibronectin as mesenchymal markers was inhibited. The release of MMP–2 and MMP-9 as well as the phosphorylation of Smad2 and paxillin induced by TGF-β2 in RPE cells were also attenuated by PGE 2 . Conclusion: PGE 2 may inhibit the contraction of proliferative fibrocellular membranes that can lead to retinal detachment in individuals with progressive vitreoretinal diseases.

Abstract

The time over threshold (TOT) method has several advantages over direct pulse height analysis based on analog to digital converters (ADCs). A key advantage is the simplicity of the conversion circuit which leads to a high level of integration and a low power consumption. The TOT technique is well suited to build multi-channel readout systems for pixelated detectors as described in our previous work that also exploits the Pulse Width Modulation (PWM) method. The main limitation of the TOT technique is that the relation between the input charge to be measured and the width of the encoded pulse is strongly non-linear. Dynamic range limitation is also an issue. To address these aspects, we propose a new time over threshold conversion circuit where the threshold of the comparator is dynamically changed instead of being constant. We call this scheme the “dynamic TOT method”. We show that it improves linearity and dynamic range. It also shortens the duration of measured pulses leading to higher counting rates. We present a short analysis that explains how the ideal linear input charge to TOT transfer function can theoretically be obtained. We describe the results obtained with a test circuit built from discrete components and present several of the spectrums obtained with crystal detectors and a radioactive source. A case of haematoma in the pineal region is reported in a patient whose initial symptom was subarachnoid haemorrhage. The encapsulated haematoma and surrounding blood clots were removed surgically. Abstract

The expression of the myeloperoxidase (MPO) gene is restricted to cells of the myeloid cell lineage and is induced by granulocyte colony-stimulating factor (G-CSF). In this study, a series of deletion mutations was introduced in the promoter of the human MPO gene, which was then fused to the chloramphenicol acetyltransferase gene. The G-CSF-induced promoter activity was examined in mouse myeloid precursor FDC-P1 transformants that constitutively express the G-CSF receptor. A G-CSF-responsive element (GRE) in the MPO gene was found approximately 800 base pairs upstream from the transcription initiation site. When the 5'-flanking region of the human MPO gene contained this element, it yielded promoter activity in cells cultured with G-CSF but not in cells cultured with interleukin 3. Gel shift assays with the element showed that a specific nuclear factor(s) (NF/G-CSF) binds to the element. The NF/G-CSF was purified by affinity chromatography using an oligonucleotide of GRE. Protein sequence analysis of the purified NF/G-CSF indicated that NF/G-CSF is a ubiquitous transcription factor, NF-Y, which is composed of three subunits. The recombinant NF-Y was then shown to bind to GRE in a combination of the three subunits.

50. Butyrate activates the WAF1/Cip1 gene promoter through Sp1 sites in a p53-negative human colon cancer cell line.

Abstract

Butyrate is a well known colonic luminal short chain fatty acid, which arrests cell growth and induces differentiation in various cell types. We examined the effect of butyrate on the expression of WAF1/Cip1, a potent inhibitor of cyclin-dependent kinases, and its relation to growth arrest in a p53-mutated human colon cancer cell line WiDr. Five millimolar butyrate completely inhibited the growth of WiDr and caused G1-phase arrest. WAF1/Cip1 mRNA was rapidly induced within 3 h by treatment with 5.0 mM butyrate, and drastic WAF1/Cip1 protein induction was detected. Using several mutant WAF1/Cip1 promoter fragments, we found that the butyrate-responsive elements are two Sp1 sites at -82 and -69 relative to the transcription start site. We also found that a TATA element at -46 and two overlapping consensus Sp1 sites at -60 and -55 are essential for the basal promoter activity of WAF1/Cip1. These findings suggest that butyrate arrests the growth of WiDr by activating the WAF1/Cip1 promoter through specific Sp1 sites in a p53-independent fashion.
