Abstract

The PI3K/AKT/mTOR axis promotes survival and is a frequently mutated pathway in cancer. Yet, inhibitors targeting this pathway are insufficient to induce cancer cell death as single agents in some contexts, including diffuse large B cell lymphoma (DLBCL). In these situations, combinations with inhibitors targeting BCL-2 survival proteins (ABT-199 and ABT-263) may hold potential. Indeed, studies have demonstrated marked synergy in contexts where PI3K/mTOR inhibitors suppress expression of the pro-survival protein, MCL-1. In this study, we use BH3 profiling to confirm that BCL-2 and BCL-XL support survival following PI3K pathway inhibition, and that the dual PI3K/mTOR inhibitor BEZ235 strongly synergizes with BCL-2 antagonists in DLBCL. However, we identify an alternative mechanism of synergy between PI3K/mTOR and BCL-2 inhibitors, independent of MCL-1 down-regulation. Instead, we show that suppression of AKT activation by BEZ235 can induce the mitochondrial accumulation of pro-apoptotic BAD and BIM, and that expression of a constitutively active form of AKT prevents sensitization to BCL-2 antagonism. Thus, our work identifies an additional mechanism of synergy between PI3K pathway inhibitors and BCL-2 antagonists that strengthens the rationale for testing this combination in DLBCL.

Abstract

Abstract The PI3K/AKT/mTOR axis is one of the most commonly mutated pathways in cancer, where aberrant activation promotes cell growth, proliferation, and survival. However, selective inhibitors targeting PI3K/AKT/mTOR have been hindered by their inability to effectively induce cell death in certain cancers. With the introduction of inhibitors that directly inhibit key pro-survival factors such as BCL-2 and BCL-XL (ABT-263 and ABT-199), the potential to achieve cancer cell death using combinations of targeted inhibitors has become a realizable goal. Here we show that selective inhibition of key components in the PI3K pathway (PI3K, AKT, or mTOR) potently sensitized a panel of DLBCL cell lines to ABT-263-induced apoptosis. While the degree of sensitization varied according to which PI3K pathway component was targeted, dual inhibition of both PI3K and mTOR consistently elicited the most potent sensitization across several cell lines. Previous work in other cancer types has linked the potency of this combination to the capacity of PI3K pathway inhibitors to reduce MCL-1 in an mTORC1-dependent manner. However, we found that this was not the case for DLBCL. For example, ABT-263 resistance induced by over-expression of MCL-1 was overcome when cells were co-treated with a dual-PI3K/mTOR inhibitor NVP-BEZ235 despite maintained expression of MCL-1. Instead, inhibition of the PI3K pathway led to a general increase in mitochondrial priming as measured by BH3 profiling. This occurred through distinct effects from both AKT and mTORC1 on the abundance of multiple BCL-2 family proteins at the mitochondria including reductions in the pro-survival proteins BCL-2, MCL-1, and BCL-XL as well as increases in Bim and Bad. In addition to these direct effects on BCL-2 family proteins, we found that inhibition of the PI3K pathway reduced expression of cancer-relevant proteins c-Myc and eIF4G as determined by reverse phase protein array. Thus, this project highlights the broad effects of dual-PI3K/mTOR inhibitors that sensitize DLBCL to apoptosis, and suggests that inhibition of the PI3K pathway has distinct effects on cell survival signaling among different cancer cell types. Abstract

This report provides an overview of EuroSea's initiatives focused on engaging the next generation of ocean observing and forecasting stakeholders. Many activities took place, including delivering workshops, presenting the EuroSea itinerant exhibition, collaborating with the WASCAL Floating University and the SEA-EU inter-university initiative, supporting the international Ocean Observers Initiative, and much more. Engaging the next generation of stakeholders in meaningful discussions and innovative projects is essential to ensure future-oriented intergenerational collaboration. Yet, this is often an overlooked aspect of public engagement within the Horizon 2020 landscape as it requires engagement techniques specifically tailored to reach the young generation. Within the framework of Horizon 2020, the European Union's research and innovation funding program, public engagement traditionally targets a diverse range of stakeholders, including researchers, policymakers, industry representatives, civil society organizations, NGOs, and citizens. While these efforts are crucial for fostering inclusive and transparent dialogue, targeted initiatives directed towards the younger generation and early-career ocean professionals need to be expanded. It is important to recognize the concerns, unique perspectives, and aspirations of young individuals who will inherit the outcomes of today's research and innovation. Dedicating resources to engage with the next generation is vital to ensure their active involvement in shaping their future and addressing global challenges such as the sustainability of ocean observing, monitoring, and forecasting. EuroSea has recognized the importance of fostering a deeper understanding of ocean observing and forecasting among the younger generation. This deliverable and the many activities feeding into it are a testament to EuroSea's commitment to this cause. Abstract

Background: Prior trials suggest that intravenous racemic ketamine is a highly effective for treatment-resistant depression (TRD), but phase 3 trials of racemic ketamine are needed. Aims: To assess the acute efficacy and safety of a 4-week course of subcutaneous racemic ketamine in participants with TRD. Trial registration: ACTRN12616001096448 at www.anzctr.org.au. Method: This phase 3, double-blind, randomised, active-controlled multicentre trial was conducted at seven mood disorders centres in Australia and New Zealand. Participants received twice-weekly subcutaneous racemic ketamine or midazolam for 4 weeks. Initially, the trial tested fixed-dose ketamine 0.5 mg/kg versus midazolam 0.025 mg/kg (cohort 1). Dosing was revised, after a Data Safety Monitoring Board recommendation, to flexible-dose ketamine 0.5–0.9 mg/kg or midazolam 0.025–0.045 mg/kg, with response-guided dosing increments (cohort 2). The primary outcome was remission (Montgomery-Åsberg Rating Scale for Depression score ≤10) at the end of week 4. Results: The final analysis (those who received at least one treatment) comprised 68 in cohort 1 (fixed-dose), 106 in cohort 2 (flexible-dose). Ketamine was more efficacious than midazolam in cohort 2 (remission rate 19.6% v. 2.0%; OR = 12.1, 95% CI 2.1–69.2, P = 0.005), but not different in cohort 1 (remission rate 6.3% v. 8.8%; OR = 1.3, 95% CI 0.2–8.2, P = 0.76). Ketamine was well tolerated. Acute adverse effects (psychotomimetic, blood pressure increases) resolved within 2 h. Conclusions: Adequately dosed subcutaneous racemic ketamine was efficacious and safe in treating TRD over a 4-week treatment period. The subcutaneous route is practical and feasible. Abstract

El auge de la movilidad vehicular durante el siglo XX significó el reemplazo progresivo de la movilidad peatonal y de una acción primaria del individuo: el caminar. Sin embargo, los conflictos generados por el crecimiento urbano y el deterioro ambiental han traído cuestionamientos acerca de las formas de pensar e intervenir nuestras ciudades dando lugar a una transformación gradual de paradigmas de movilidad. Hoy, la movilidad peatonal y la movilidad no motorizada se han vuelto temas recurrentes, tanto en agendas públicas como en la construcción del espacio urbano, visibilizando una importante variedad de reflexiones sobre el diseño, la calidad y la experiencia espacial. Para evidenciar la importancia de los entornos peatonales como espacio clave de las practicas cotidianas de movilidad sostenible y partir de una propuesta metodológica desarrollada como parte de un trabajo de maestría, esta ponencia presenta una propuesta de diseño aplicada al corredor Bolívar, entre calle 44 San Juan y Calle, en el centro de Medellín., The rise of vehicular mobility during the 20th century led to a progressive decline of pedestrian mobility and one of the primary activities of human beings: walking.
