Exercise may increase the antioxidant capacity of plasma by stimulating antioxidant enzymes. The study aimed to measure the effect of three repetitions of acute exercise on arylesterase (ARE) activity of the paraoxonase 1 (PON1) enzyme. Eleven average-trained men (age 34.0 ± 5.2 years) completed three treadmill runs. ARE activity in plasma was evaluated spectrophotometrically and compared with PON1 concentration (PON1c), paraoxonase (PON) activity, and high-density lipoprotein cholesterol (HDL-C) at rest and after exercise. In all repetitions of the exercise, ARE activity remained stable, and ARE activity standardized for PON1c (ARE/PON1c) was lower post- than pre-exercise. The ARE/PON1c ratio changes returned to baseline levels during rest after each exercise session. Pre-exercise ARE activity correlated negatively with post-exercise C-reactive protein (CRP) (ρ = −0.35, p = 0.049), white blood cell count (WBC) (ρ = −0.35, p = 0.048), polymorphonuclear leukocytes (PMN) (ρ = −0.37, p = 0.037), and creatine kinase (CK) (ρ = −0.37, p = 0.036). ARE activity may be depleted under conditions of oxidative stress, as increases in PON1c during acute exercise did not result in parallel increases in ARE activity. No adaptation of the response of ARE activity to exercise was detected in subsequent exercise sessions. Individuals with lower pre-exercise ARE activity may develop a higher inflammatory response to strenuous exercise.

Objectives. Oxidative stress, induced by physical activity, may stimulate the expression, release, and activity of certain antioxidant enzymes. We investigated the effect of three repeated bouts of strenuous exercise on paraoxonase 1 concentration (PON1c) and paraoxonase activity (PON). Methods. Eleven average-trained healthy men (age 34.0 ± 5.2 years) performed three strenuous exercise tests on a treadmill separated by 72 hours periods of resting. PON1c, PON, ferric-reducing activity of plasma (FRAP), lipid profile, C-reactive protein concentration (CRP), and lactate concentration were determined in plasma. Results. Each exercise bout resulted in similar PON1c, PON, FRAP, and high-density lipoprotein concentration (HDL-C) increments, while PON/HDL-C ratio remained stable in all repetitions. Percentage increments at the bout of each exercise were higher for PON1c (by 64.82% at the first, by 92.9% at the second, and by 77.02% at the third exercise) than for PON (by 6.49% at the first, 10.06% at the second, and by 12.32% at the third exercise). Association was found between preexercise PON and PON1c ( r = 0.56 , p = 0.029 ), pre- ( r = 0.87 , p = 0.00003 ) and postexercise HDL-C ( r = 0.6 , p = 0.0002 ), preexercise PON and cardiovascular fitness level of participants measured as VO2max ( r = 0.39 , p = 0.026 ), and postexercise PON and lactate concentration ( r = 0.44 , p = 0.01 ). Conclusions. PON1c and PON increase during strenuous exercise, yet the effect of exercise on PON1 concentration is more pronounced. PON1 does not show tolerance to physical activity. The enzyme may provide short-term protection from oxidative stress in each exercise bout. PON may depend on exercise load. Cardiovascular fitness levels may be associated with PON1 activity.

This paper is a literature overview of the complex relationship between vitamin C and two opposing physiological states, physical activity and sleep. The evidence suggests a clinically important bidirectional association between these two phenomena mediated by different physiological mechanisms. With this in mind, and knowing that both states share a connection with oxidative stress, we discuss the existing body of evidence to answer the question of whether vitamin C supplementation can be beneficial in the context of sleep health and key aspects of physical activity, such as performance, metabolic changes, and antioxidant function. We analyze the effect of ascorbic acid on the main sleep components, sleep duration and quality, focusing on the most common disorders: insomnia, obstructive sleep apnea, and restless legs syndrome. Deeper understanding of those interactions has implications for both public health and clinical practice.

Introduction: Stroke is one of the leading causes of mortality and disability in the world. A wide knowledge of risk factors for cerebrovascular diseases is one of the main elements of primary prevention, while knowledge of stroke symptoms can significantly speed up treatment and reduce negative health effects. The aim: To assess the knowledge about stroke and risk factors of cerebrovascular diseases among the inhabitants of the randomly selected rural community (in Lodz Province).Material and methods: The study included 143 adult residents of Rzasnia. The research was carried out using a self-constructed questionnaire, prepared on the basis of available literature.Results: The average score obtained from the knowledge survey on stroke was 5.79 +/- 3.47 (maximum number of points - 14). The average number of points in people with a family history of stroke was statistically significantly higher (7.46±3.31) than in those with no stroke in the family (4.20±2.98 ) (p0.001). There was no statistically significant difference between the average number of points and gender (women - 6.13±3.55 points, men - 5.64±3.55, NS) and age (R = -0.1, NS ). However, the positive correlation between educational level and the score obtained in our questionnaire was found (R =0.4, p0.05). The average number of correctly listed risk factors was: 0.6 ± 1.6. At least one risk factor was reported by only 34% of respondents. Respondents most often mentioned: excessive alcohol consumption (18%), smoking (18%), hypertension (14%), atherosclerosis (11%). The average number of correctly listed symptoms of stroke was: 1.2±1.3. At least one symptom of stroke was reported by only 58% of respondents. The respondents most often reported: headache (29%), speech disorders (22%), gait disturbances (13%), and sensory disorders (13%). If a stroke was suspected, the majority of respondents would call an ambulance (94%).Conclusions: Knowledge of the residents of the chosen rural commune about the stroke is not satisfactory and should be supplemented with an educational program that would include as many people as possible.

Asymmetric synthesis, in which chiral organocatalysts or metal complexes with chiral ligands are used, has become the most valuable methodology for the preparation of enantiomerically pure organic compounds. Among such catalysts/ligands, a growing number constitute various organosulfur compounds. This Review provides comprehensive and critical information on the plethora of sulfur-based chiral ligands and organocatalysts used in asymmetric synthesis, which have been published within the last 15 years. However, it is confined to the presentation of only those chiral catalysts/ligands that possess a stereogenic sulfur atom and includes sulfoxides, sulfinamides, N-sulfinyl ureas, sulfoximines, and some related S-chiral derivatives.

The last few years have brought many discoveries concerning important modulatory effects of histamine and its receptors on basic mechanisms of the immunological processes. The role of histamine H1 and H2 receptors in immunomodulation has been established. The immunomodulatory function of a newly described histamine H4 receptor has been revealed. One of the most important modulatory effects of histamine currently studied is its influence on T lymphocyte differentiation and function. Our present knowledge suggests that histamine may have a wider influence on various immunological processes than is now accepted; therefore, we need further studies to fully clarify the role of histamine and its receptors. This knowledge can bring new therapeutic solutions in allergies, autoimmune diseases and malignancies.

Introduction The positive impact of properly performed exercise on the human body is well known. In this study, the effect of maximal exercise on the antioxidant activity of the blood enzyme, paraoxonase (PON), was determined. Aim The aim of this study was to determine the influence of a single bout of maximal physical exercise on PON activity and to investigate the correlation of this activity with chosen biometric parameters and physical activity levels. Materials and methods In total, 15 subjects participated in this study. The average age of the subjects was 18 years (±2.74 years). Participants were subjected to maximum efforts on a treadmill until complete exhaustion resulted. They had their blood taken for analysis at three time points – before, at the end, and 2 h following the end of exercise. PON activity was determined by the ability to dispose of paraoxon. The subjects also filled in questionnaires in which they determined the amount and form of training carried out. Basic biometric data were collected from the subjects. Results and discussion This study demonstrates that the enzyme activity at the maximal effort is higher than at rest; however, it does not depend on the level of physical activity. PON activity 2 h following exercise, though higher than at rest, was not statistically significant. No relationship was found between PON activity and chosen parameters such as age, weight, BMI, lean, body mass. Conclusions A single bout of maximal exercise increases PON activity. A positive trend was also observed with respect to the impact of the physical activity level on PON activity at rest.

In the past decade the number of patients with type 1 diabetes treated with continuous subcutaneous insulin infusion (CSII) has increased rapidly. Treatment of the disease is focused on proper physical development and the prevention of complications. Aim of the study was to analyze changes in the treatment and clinical picture of type 1 diabetes in children over the years 2000 to 2010 with particular emphasis on the presence of autoimmune diseases and microangiopathy.The study included 567 children diagnosed with type 1 diabetes under the care of outpatient diabetes clinic. We compared 251 children, diabetes outpatient clinic patients in 2000, with 316 children in 2010. Data were obtained from the outpatient and hospital records. We compared baseline demographic, anthropometric data, treatment regimen, type of insulin, metabolic control, prevalence of autoimmune diseases and microangipathy.In 2010 there was a reduction in the age of diagnosis of diabetes from 10 to 8 years (p=0.039). Significantly increased the proportion of children treated with CSII (up to 60.1%) and decreased the percentage of children using conventional insulin for the benefit of insulin analogs. The increase in HbA1c from 7.4 to 8.0% (p0.001) has been shown and increase in proportion of patients with HbA1c7.5% in 2010. The percentage of children with obesity increased from 5.2 to 13.7% (p=0.004) and there was a significant increase in SDS-BMI. The percentage of children with autoimmune diseases such as celiac (from 0,4 to 7,3%, p0,001) and thyroid (from 6.9 to 21.3%, p0.001) has increased. The incidence of retinopathy decreased from 6 to 1% (p=0.04), and albuminuria decreased insignificantly.Over the last decade, a significant change in the method of treatment in children diagnosed with type 1 diabetes has occurred. The deterioration of metabolic control, despite the frequent use in the treatment of CSII, may be due to increased frequency of obesity and additional autoimmune diseases in today´s patients. More similar to physiologic way of insulin infusion in nowadays treatment may influence the decrease in the prevalence of retinopathy.

Objectives There are conflicting data on the influence of physical activity on paraoxonase1 (PON1) activity. The purpose of this study is to investigate the effect of maximal exercise (ME) on plasma paraoxonase (PON) and arylesterase (ARE) activity in elite rugby players. In addition, the influence of training experience and PON1 Q192R polymorphism on PON1 activity changes at ME was evaluated. Methods Twenty-five elite rugby players ages 22.0 ± 3.71 years, consisting of 11 juniors – J (2–7 years of training) and 14 seniors – S (8–15 years of training), completed ME on a cycle ergometer. PON and ARE activity, ferric reducing activity of plasma (FRAP), uric acid (UA), and total bilirubin concentration, as well as thiobarbituric acid reactive substances and lipid profile were investigated in the plasma before, at the bout, and 30 minutes after ME. Results At the bout of ME we found an increase in PON1 activities, ARE/high-density lipoprotein C ratio, TBil, and TChol. However, ARE activity changes were not observed in the group of rugby players training for ≤7 years. FRAP and UA increased later – 30 minutes after ME. Conclusion We conclude that in rugby players PON1 changes during ME depend on age, body composition, and training experience. The influence of PON1 Q192R polymorphism on PON1 changes at ME remains debatable.

The purpose of this study was to elucidate the participation of plasma PON1 (paraoxonase activity [PON] and arylesterase activity [ARE]) in antioxidant defense in response to a single bout of maximal exercise. PON, ARE, lipid profile, lipid peroxidation (thiobarbituric acid reactive substances [TBARS]), total antioxidant status (ferric reducing ability of plasma [FRAP]), concentration of uric acid [UA], and total bilirubin (TBil) were determined in the plasma before, at the bout and 2 h after maximal exercise on a treadmill in young sportsmen. Chosen physiological parameters also were controlled during maximal exercise. Following maximal exercise, the unaltered level of TBARS and increased FRAP were registered. ARE increment was the highest (37.6%) of all measured variables but lasted for a short time. UA increment was lower than ARE but long-lasting and correlated with FRAP. PON activity increment was associated with the combined effect of body weight, lean, body mass index (BMI) and basal metabolic rate (BMR). We conclude that PON1 is a co-factor of the first line of antioxidant defense during maximal exercise. Its activity is associated with body composition and not the physical fitness of the subjects.

Statins are drugs with the unquestionable effectiveness in the reduction of low density lipoprotein cholesterol (LDL-C) and the cardiovascular risk with the acceptable safety profile. On the basis of the above statins are the most common used drugs worldwide. The present review is aimed to discuss the potential mechanisms of statins leading to occurrence of glucose metabolism disturbances through the influence on insulin secretion by the beta-cells of pancreatic islets and the cells' sensitivity on insulin. It might be a results of disadvantageous statin properties connected to the intensification of inflammation and oxidation within the pancreatic islets, and the influence on adipokines secretion by the fat tissue cells. However, it should be emphasized that despite the recommendations of US Food and Drug Administration suggesting to keep caution in connection to potentially prodiabetic statins' properties, this data need to be confirmed in large multicenter clinical trials with properly designed main endpoints.

Urolithiasis is increasingly being diagnosed in children. Some of the major risk factors for kidney stones are hipercalciuria, hiperoxaluria, hipocitraturia and hydrogen ion content measured by pH. Recently, more and more attention is being paid to the impact of diabetes type 1 and 2 on the development of nephrolithiasis especially in periods of poor metabolic control.was to evaluate the BRI (bonn risk index - rate of spontaneous crystallization of calcium oxalate), the concentration of oxalate, citrate and urine pH in children with acid-base balance disturbances occurring in patients with newly diagnosed type 1 diabetes (DMT1).The study group included 40 patients aged 6 to 17 years (average age ± SD: 12,58 ± 3,33) with newly diagnosed DMT1. The study was performed twice: at the beginning of the disease, immediately after the treatment of diabetic ketoacidosis (study I) and after obtaining satisfactory metabolic control (study II), that is after about two weeks. The control group consisted of 100 children (6-17 years, average age 12,34 ± 3,96) without symptoms suggestive of urolithiasis. In every child, a 24-hour urine sample was collected. BRI was implemented in the urine by his own semi-micro method.Ionized calcium level was significantly higher immediately after the diagnosis of type 1 diabetes compared to the control group (0,68±0,67 vs. 0,40 ± 0,18 mmol/l; p0,001) and to study II (0,68 ± 0,67 vs. 0,28 ± 0,23 mmol/l; p=0,008). BRI value was significantly higher in early onset compared to the control group and the study II (3,18 ± 5,54 vs. 0,66 ± 0,52, p0,001; 3,18 ± 5,54 vs. 0,64 ± 1,56; p=0,034). BRI correlated inversely with pH at admission to the hospital (r=-0,53, p=0,0023).Metabolic alterations occurring during diabetic ketoacidosis at diagnosis of new type 1 diabetes may predispose to the development of the urinary stones, and thereby to the kidney damage.

Experience with use of real-time continuous glucose monitoring systems (RT-CGMS) in teenagers with diabetes type 1 is limited, and in unselected groups of young patients did not show improvement in metabolic control.The objective of this study was to assess short-term RT-CGMS usage in teenagers with type 1 diabetes, in terms of possibility to improve metabolic control and acceptance of the system.40 subjects, aged 14.5±2 years on insulin pump therapy were included in the study. Mean diabetes duration was 6±3 years and HbA1c level before the study was 8.4±1.5%. The analysis was based on single 5-6 days long sensor usage, connected with education of the family. We analysed several parameters of glycaemic variability during the study, and HbA(1)c level before and 2 months after the study. Patients' satisfaction was assessed on the basis of a questionnaire.HbA(1)c level in the whole teenagers group decreased insignificantly by 0.3%; from 8.4±1,% to 8.1±1.6%. In 24 (60%) patients we showed improvement in HbA(1)c by at least 0.5% (mean 0.9%, from 8.1±1.3% to 7.2±1.2%; p=0.03). HbA(1)c level was slightly higher in girls than in boys at the beginning of the study and a greater reduction in HbA(1)c was shown for boys. After two months the difference was significant: 8.6±1.9% in girls vs. 7.6±1.3% in boys, p=0.03. In the group with HbA(1)c decrease and in boys we demonstrated improvement in mean glycaemia and glycaemic variability parameters on the last day of the sensor usage, compared to the first day. In boys however, increased AUC70 mg/dl/ min was noticed. The patients from the group with HbA(1)c decrease reported fewer problems with system calibration: 3.2 vs. 2.6 score, p=0.03. This group also reported higher satisfaction score connected with new knowledge: 4.0 vs. 3.5, and with quality of life: 4.1 vs. 3.6.Short-term usage of RT-CGMS, combined with satisfaction questionnaire performed in teenagers with diabetes type 1 can be useful in defining the group of young patients who can benefit from RT-CGMS usage in long-term metabolic control improvement.

The natural history of type 1 diabetes is concerned with the appearance of autoantibodies against antigens of pancreatic beta cells. The last decade revealed some evidence of the participation of T regulatory lymphocytes - cells which suppress immune response - in the pathogenesis of type 1 diabetes and prediabetes.was the assessment of T regulatory cells in the blood of children at risk for developing type 1 the diabetes mellitus.85 subjects, siblings of children with type 1 diabetes, were enrolled into the study. The presence of anti-GAD65 antibodies was assessed. With the use of flow cytometry the following cell subpopulations were noted: CD4+, CD4+CD25high and CD4+CD25highCD127low with the coexpression of: CD28, CD45RO, CD54, CD62L and CD134 molecules.We did not observe any differences in white blood cell count, lymphocyte (including CD4+) count and the percentage between the examined and control groups. We noted higher percentages of T regulatory cells: CD4+CD25high, CD4+CD127low and CD4+CD25highCD127low in children with the presence of anti-GAD65 antibodies as compared to the control children.Higher percentages of T regulatory cells in the blood of children with the presence of anti-GAD65 antibodies may suggest an intensive regulatory response present in patients at risk for developing type 1 diabetes.

Paraoxonase 1 ([i]PON1[/i]) is a member of a three-gene family ([i]PON1, PON2[/i], and [i]PON3[/i]). PON1 activity dominates in human plasma. It is secreted from hepatic cells and is found in the circulation bound to high-density lipoproteins (HDLs). For many years it has been known only for its ability to hydrolyze organophosphate derivatives. More recently, PON1’s antioxidant activity draws attention as the enzyme was described to prevent oxidation of lipoproteins by reactive oxygen species formed during oxidative stress. PON1 was also shown to hydrolyze atherogenic products of oxidative lipid modification such as phospholipid peroxides and cholesterol ester hydroperoxides. Some studies indicate that the enzyme presents a lipolactonase activity and hydrolyzes homocysteine thiolactone (HCTL). There is growing evidence as to PON1’s protective role in atherosclerosis. Genetic (PON1 polymorphism) and environmental factors and lifestyle may influence PON1 blood concentration and biological activity. Among the many recognized factors accounting for lifestyle, physical activity plays an important role. Various, often opposite, effects on PON1 status are observed in regular training and single physical activities. The results of different studies are often contradictory. It may depend on the time, intensity, and frequency of physical activity. Additionally, it seems that the effects of physical activity on [i]PON1[/i] blood concentration and activity are modified by environmental and lifestyle factors as well as [i]PON1[/i] polymorphism.

The problem of insurance crimes has existed since the birth of insurance companies. At present it is a serious problem that the insurance companies are fighting. The ingenuity of the insurance frauds seems to be unlimited and new methods of swindling compensation are appearing again and again. The size of this occurrence is constantly increasing which is a result of the existance of organised crime groups, which often comprise of insurance agents, loss handlers, technical experts and police officers. Therefore battling this problem by taking action which aims to restrain, and eventually stop this occurrence is very important.
