Tropical pelagic predators are exploited by fisheries and their movements are influenced by factors including prey availability, temperature, and dissolved oxygen levels. As the biophysical parameters vary greatly within the range of circumtropical species, local studies are needed to define those species' habitat preference and model possible behavioral responses under different climate change scenarios. Here, we tagged yellowfin tuna Thunnus albacares in the Galápagos Marine Reserve and tracked the horizontal and vertical movements of eight individuals for 4–97 days. The tuna traveled a mean of 13.6 km day−1 horizontally and dispersed throughout the archipelago and in offshore waters inside the Galápagos Marine Reserve and in the surrounding Ecuadorian exclusive economic zone. Vertically, they traveled a mean of 2 km day−1, although high‐resolution data from a recovered tag suggested that transmitted data underestimated their vertical movement by a factor of 5.5. The tracked yellowfin tuna spent most of their time near the surface, with an overall mean swimming depth of 24.3 ± 46.6 m, and stayed shallower at night (11.1 ± 16.3 m) than during the day (37.7 ± 60.9 m), but on occasion dived to cold, oxygen‐poor waters below 200 m. Deep dives were commonly made during the day with a mean recovery period of 51 min between exposures to modeled oxygen‐limiting conditions <1.5 mL L−1, presumably to re‐oxygenate. The depth and frequency of dives were likely limited by dissolved oxygen levels, as oxygen‐depleted conditions reach shallow depths in this region. The main habitat of tracked yellowfin tunas was in the shallow mixed layer, which may leave them vulnerable to fishing. Vertical expansion of low‐oxygen waters under future climate change scenarios may further compress their habitat, increasing their vulnerability to surface fishing gear. [ABSTRACT FROM AUTHOR]

The Eocene deposits of Egypt provide a wide variety of shallow marine facies and fossil assemblages, allowing paleoenvironmental reconstructions in this warmhouse climate interval. Forty-three rock samples have been collected from two middle Eocene sections, exposed at southeast Beni-Suef area in northern Egypt. The studied outcrops are lithologicsally subdivided into two rock units named from base to top as follows: (1) the Qarara Formation (Lutetian) and (2) the El Fashn Formation (Bartonian). Most investigated rock samples showed richness in benthic foraminiferal assemblages and only rare occurrences of index planktonic foraminifera. They yielded 160 foraminifera species which belong to 4 suborders, 19 superfamilies, 34 families, and 59 genera. The stratigraphic distribution of the identified species allowed us to define four local benthic bio-ecozones: (1) Bolivina carinata Lowest Occurrence Zone (Lutetian), (2) Cibicides mabahethi/Cancris auriculus primitivus Concurrent-Range Zone, (3) Nonion scaphum Lowest Occurrence Zone, and (4) Brizalina cookei/Nonionella insecta Concurrent-Range Zone (Bartonian). These biozones are described and discussed in detail and correlated to stratigraphic equivalents in Egypt. The rareness of index planktonic foraminifera through the studied sections does not allow a precise biostratigraphic and chronostratigraphic correlation. The shale samples show low TOC values, which may be related to low productivity, high sediment influx, and/or prevailing oxic conditions. Bulk rock geochemistry, consistent with the benthic foraminifera ecological preferences, indicated that the studied sections were deposited in moderate to high oxygen levels and warm climatic conditions in a typical shelf marine setting. The identified species showed strong similarities with nearby southern Tethys areas, reflecting migration via the trans-Sahara seaway, and minor similarities with those identified from the northwestern Tethys and the North Atlantic province attributed to the change in the environmental and climatic conditions, such as cooler, latitudinal zoned climatic conditions along the northwestern Tethys, which was unsuitable for their biological demands. Theragnostics is a promising approach that integrates diagnostics and therapeutics into a single personalized strategy. To conduct effective theragnostic studies, it is essential to create an in vitro environment that accurately reflects the in vivo conditions. In this review, we discuss the importance of redox homeostasis and mitochondrial function in the context of personalized theragnostic approaches. Cells have several ways to respond to metabolic stress, including changes in protein localization, density, and degradation, which can promote cell survival. However, disruption of redox homeostasis can lead to oxidative stress and cellular damage, which are implicated in various diseases. Models of oxidative stress and mitochondrial dysfunction should be developed in metabolically conditioned cells to explore the underlying mechanisms of diseases and develop new therapies. By choosing an appropriate cellular model, adjusting cell culture conditions and validating the cellular model, it is possible to identify the most promising therapeutic options and tailor treatments to individual patients. Overall, we highlight the importance of precise and individualized approaches in theragnostics and the need to develop accurate in vitro models that reflect the in vivo conditions. Study questionDoes the oxygen concentration in the culture medium [either physiologic (5%) or atmospheric (20%)] affect mitochondrial ultrastructure and function in preimplantation mouse embryos generated by IVF?Summary answerEmbryos cultured in 20% oxygen show increased mitochondrial abnormalities compared to embryos cultured in 5% oxygen.What is known alreadyART are widely used and have resulted in the birth of more than 8 million children. A variety of media and oxygen concentrations are used to culture embryos. Embryos cultured under physiological O2 tension (5%) reach the blastocyst stage faster and have fewer alterations in gene expression when compared with embryos cultured under atmospheric oxygen conditions (20%). The mechanisms by which oxygen tension affects preimplantation development remain unclear, but mitochondria are believed to play an important role. The aim of this study was to evaluate how mitochondrial ultrastructure and function in IVF embryos were affected by culture under physiologic (5%) or atmospheric (20%) oxygen concentrations.Study design, size, durationZygotes, 2-cell, 4-cell, morula and blastocyst were flushed out of the uterus after natural fertilization and used as controls. Marine sponges are an ideal source for isolating as yet undiscovered microorganisms with some sponges having about 50% of their biomass composed of microbial symbionts. This study used a variety of approaches to investigate the culturable diversity of the sponge-associated bacterial community from samples collected from the South Australian marine environment. Twelve sponge samples were selected from two sites and their bacterial population cultivated using seven different agar media at two temperatures and three oxygen levels over 3 months. These isolates were identified using microscopic, macroscopic, and 16S rRNA gene analysis. A total of 1234 bacterial colonies were isolated which consisted of four phyla: Actinobacteria, Firmicutes, Proteobacteria, and Bacteroidetes, containing 21 genera. The diversity of the bacterial population was demonstrated to be influenced by the type of isolation medium, length of the incubation period and temperature, sponge type, and oxygen level. The findings of this study showed that marine sponges of South Australia can yield considerable bacterial culturable diversity if a comprehensive isolation strategy is implemented. Two sponges, with the highest and the lowest diversity of culturable isolates, were examined using next-generation sequencing to better profile the bacterial population. A marked difference in terms of phyla and genera was observed using culture-based and culture-independent approaches. This observed variation displays the importance of utilizing both methods to reflect a more complete picture of the microbial population of marine sponges. Ancient oxygenic photosynthetic prokaryotes produced oxygen as a waste product, but existed for a long time under an oxygen-free (anoxic) atmosphere, before an oxic atmosphere emerged. The change in oxygen levels in the atmosphere influenced the chemistry and structure of many enzymes that contained prosthetic groups that were inactivated by oxygen. In the genome of Acaryochloris marina, multiple gene copies exist for proteins that are normally encoded by a single gene copy in other cyanobacteria. Using high throughput RNA sequencing to profile transcriptome responses from cells grown under microoxic and hyperoxic conditions, we detected 8446 transcripts out of the 8462 annotated genes in the Cyanobase database. Two-thirds of the 50 most abundant transcripts are key proteins in photosynthesis. Microoxic conditions negatively affected the levels of expression of genes encoding photosynthetic complexes, with the exception of some subunits. In addition to the known regulation of the multiple copies of psbA, we detected a similar transcriptional pattern for psbJ and psbU, which might play a key role in the altered components of photosystem II. Furthermore, regulation of genes encoding proteins important for reactive oxygen species-scavenging is discussed at genome level, including, for the first time, specific small RNAs having possible regulatory roles under varying oxygen levels.

During follicle maturation, oxygen levels continuously decrease in the follicular fluid and reach lowest levels in the preovulatory follicle. The current study was designed to comprehensively understand effects of low oxygen levels on bovine granulosa cells (GC) using our established estrogen active GC culture model. As evident from flow cytometry analysis the viability of GC was not found to be affected at severely low oxygen condition (1% O2) compared to normal (atmospheric) oxygen condition (21% O2). Estimations of hormone concentrations using competitive radioimmunoassay revealed that the production of estradiol and progesterone was significantly reduced at low oxygen condition. To understand the genome-wide changes of gene expression, mRNA microarray analysis was performed using Affymetrix’s Bovine Gene 1.0 ST Arrays. This resulted in the identification of 1104 differentially regulated genes of which 505 were up- and 599 down-regulated under low oxygen conditions. Pathway analysis using Ingenuity pathway analyzer (IPA) identified 36 significantly affected (p < 0.05) canonical pathways. Importantly, pathways like “Estrogen-mediated S-phase Entry” and “Cyclins and Cell Cycle Regulation” were found to be greatly down-regulated at low oxygen levels. This was experimentally validated using flow cytometry based cell cycle analysis. Up-regulation of critical genes associated with angiogenesis, inflammation, and glucose metabolism, and down-regulation of FSH signaling, steroidogenesis and cell proliferation indicated that low oxygen levels induced early luteinization associated changes in granulosa cells. Identification of unmethylated CpG sites in the CYP19A1 promoter region suggests that granulosa cells were not completely transformed into luteal cells under the present low oxygen in vitro condition. In addition, the comparison with earlier published in vivo microarray data indicated that 1107 genes showed a similar expression pattern in granulosa cells at low oxygen levels (in vitro) as found in preovulatory follicles after the LH surge (in vivo). Overall, our findings demonstrate for the first time that low oxygen levels in preovulatory follicles may play an important role in supporting early events of luteinization in granulosa cells.

To investigate the impact of the Bandar Abbas thermal power plant on the waters of the Persian Gulf coast, a combination of satellite images and ground data was utilized to determine the Sea Surface Temperature (SST) as a thermal index, Total Organic Carbon (TOC) and Chemical Oxygen Demand (COD) as biological indices. Additionally, measurements of SO 2 , O 3 , NO 2 , CO 2 , CO, and CH 4 values in the atmosphere were taken to determine the plant's impact on air pollution. Temperature values of the water for different months were predicted using Long Short-Term Memory (LSTM), Support Vector Regression (SVR), and Cascade neural networks. The results indicate that the waters near thermal power plants exhibit the highest temperatures in July and September, with temperatures reaching approximately 50 °C. Furthermore, the SST values were found to be strongly correlated with ecological indices. The Multiple Linear Regression (MLR) analysis revealed a strong correlation between the temperature and TOC, COD, and O 2 in water (R TOC 2 = 0.98), R O 2 2 = − 0.89 , R COD 2 = 0.87 and O 3 , NO 3 , CO 2 , and CO in the air (R O 3 2 = 0.99 , R NO 3 2 = 0.97 , R CO 2 2 = 0.95 , R CO 2 = 0.96). Brain is the major consumer of glucose in the human body, whose pattern of consumption changes through lifetime, decreasing during adolescence up to adulthood. This evidence leads to the hypothesis that, in cerebral developmental stages, glycolysis might be the driving force for the high-energy requirement. Furthermore, several studies claim that neurogenesis process is accompanied by a shift into mitochondrial oxidative metabolism. Much of what we know about the functioning of human T lymphocytes is based on the experiments carried out in atmospheric oxygen (O¿) concentrations, which are significantly higher than those maintained in blood. Interestingly, the gender differences in the activity of T cells and their susceptibility to apoptosis under different O¿ conditions have not yet been described. The aim of the study was to compare two main markers of lymphocyte function: proliferation capacity and ability to produce cytokines as well as their susceptibility to apoptosis under two different O¿ concentrations, between men and women.~Introduction~Background~25 healthy volunteers, both males (13) and females (12) were recruited to the study (mean age 25.48 ± 5.51). By using cytometry proliferation parameters of human CD4+ CD28+ cells or CD8+CD28+ cells in response to polyclonal stimulation of the TCR/CD3 complex at atmospheric (21%) and physiological (10%) O¿ concentrations using our modified dividing cell tracking technique (DCT) were analyzed as well as the percentages of apoptotic cells. We also determined the levels of IFN-¿, IL-2, IL-10 and IL-17A using Cytometric Bead Array Flex system in cell culture supernatants.~Materials and Methods~Methods~CD4+CD28+ and CD8+CD28+ cells from the whole study group were characterized by shorter time required to enter the first (G1) phase of the first cell cycle at 21% compared to 10% O¿. Both T cell populations performed significantly more divisions at 21% O¿. The percentages of dividing cells were also significantly higher at atmospheric O¿. Interestingly, data analysis by gender showed that male lymphocytes had similar proliferative parameters at both O¿ concentrations while female lymphocytes proliferate more efficiently (note from the author: we cannot say that lymphocytes proliferate faster, rather more effectively, because cells perform more divisions, which gives more percentage of offspring cells) at 21% oxygen. Compared to males, the female CD4+ cells showed increased susceptibility to apoptosis at both O¿ concentrations. No differences in the levels of cytokines regardless of gender and oxygen conditions were found.~Results~Results~We showed that in vitro female T cells (both CD4+ and CD8+ cells) are more sensitive than male lymphocytes to low O2 concentration as demonstrated by the decrease in their proliferation dynamics. Compost quality greatly depends on maturity and humification, and odor emissions can cause severe air pollution and nutrient losses during composting. In this study, a laboratory trial was carried out under different aeration conditions to explore the effects of boundary-oxygen content (microaerobic, aerobic, oxygen-rich) on maturity, humification, and odor emissions during chicken manure composting. Results showed that the aeration rate (AR) of 0.1 m3 h−1 was the minimum aeration requirement to realize the maturity and humification of chicken manure. The germination index (GI) and humification indexes were significantly higher in the aerobic and oxygen-rich treatments (AR ≥ 0.1 m3 h−1) than in the microaerobic treatments (AR < 0.1 m3 h−1). When comparing the maturity and humification of the final compost, the lowest NH 3 and H 2 S emissions occurred in the treatment with continuous aeration at 0.1 m3 h−1. In addition, a greater than 35% reduction in C/N could be used as an evaluation indicator of maturity and humification. Redundancy and correlation analysis indicated that maturity, humification, and odor emissions were significantly influenced by the key physicochemical factors (O 2 , temperature, NH 4 +-N and C/N) and key bacterial communities. Finally, an operable and straightforward strategy was proposed to improve compost maturity and reduce odor emissions through controlling temperature, C/N, and NH 4 +-N to regulate the key bacterial communities. This article explores the role that oxygen levels in US spacecraft from 1961 to 1998 have on the development of cataracts induced by space radiation in astronauts and whether oxygen levels are well accounted for in experimental studies examining cataractogenesis. The first epidemiological report in 2001 linked an increased risk of the primary types of cataracts, and nuclear cataract alone, for astronauts with higher lens doses. However, later studies of US astronauts in 2009 and 2012 reported a higher risk of cortical cataract and posterior subcapsular cataract, but not for nuclear cataract. Firstly, it is postulated that the high oxygen level atmospheres of spacecraft employed before 1976 were a factor in promoting nuclear cataract. The high oxygen levels of hyperbaric oxygen therapy are reportedly associated with nuclear cataract, and the low intraocular oxygen levels of diabetic patients are possibly linked to their higher risk of posterior subcapsular cataract and cortical cataract. Secondly, it is hypothesized that the normal hypoxic environment of the lens and lens epithelial cells (LECs), and all stem/progenitor cells in general, have an optimal Goldilocks range of oxygen levels. Too high a lenticular oxygen level increases oxidative stress and radiosensitivity due to the oxygen effect. Whereas too low an oxygen tension also increases oxidative stress and disrupts LEC differentiation. Even so, a focused literature search of the PubMed database of in vitro experiments with LECs shows that studies rarely account for the hypoxic state of the normal lens, whether ionizing radiation is a factor or not. It is therefore recommended that ocular physioxic levels should therefore be considered when designing in vitro studies to better understand the progression of cataractogenesis on long-duration missions to the Moon and Mars. Fossil charcoal provides direct evidence for fire events that, in turn, have implications for the evolution of both terrestrial ecosystems and the atmosphere. Most of the ancient charcoal record is known from terrestrial or nearshore environments and indicates the earliest occurrences of fire in the Late Silurian. However, despite the rise in available fuel through the Devonian as vascular land plants became larger and trees and forests evolved, charcoal occurrences are very sparse until the Early Mississippian where extensive charcoal suggests well-established fire systems. We present data from the latest Devonian and Early Mississippian of North America from terrestrial and marine rocks indicating that fire became more widespread and significant at this time. This increase may be a function of rising O2 levels and the occurrence of fire itself may have contributed to this rise through positive feedback. Recent atmospheric modeling suggests an O2 low during the Middle Devonian (around 17.5%), with O2 rising steadily through the Late Devonian and Early Mississippian (to 21-22%) that allowed for widespread burning for the first time. In Devonian-Mississippian marine black shales, fossil charcoal (inertinite) steadily increases up-section suggesting the rise of widespread fire systems. There is a concomitant increase in the amount of vitrinite (preserved woody and other plant tissues) that also suggests increased sources of terrestrial organic matter. Even as end Devonian glaciation was experienced, fossil charcoal continued to be a source of organic matter being introduced into the Devonian oceans. Scanning electron and reflectance microscopy of charcoal from Late Devonian terrestrial sites indicate that the fires were moderately hot (typically 500-600 °C) and burnt mainly surface vegetation dominated by herbaceous zygopterid ferns and lycopsids, rather than being produced by forest crown fires. The occurrence and relative abundance of fossil charcoal in marine black shales are significant in that these shales may provide a more continuous record of fire than is preserved in terrestrial environments. Our data support the idea that major fires are not seen in the fossil record until there is both sufficient and connected fuel and a high enough atmospheric O2 content for it to burn. The current research evaluated the combined effect of oxygen concentration (20%, 50% and 80% O 2) and storage temperature (4°C or −1.5°C) on the fresh and internal cooked color of dark-cutting (DC) beef. Steaks were packaged in modified atmosphere packaging (MAP) over a 12-day storage period. Although the metmyoglobin reducing activity (MRA) of DC steaks decreased with increasing O 2 levels, lower oxygen consumption (OC) and deeper oxygen penetration depth (OPD) improved the surface color of 50%O 2 -MAP and 80%O 2 -MAP DC steaks. 80%O 2 -MAP resulted in the highest (P < 0.05) percentage myoglobin denaturation and alleviated the phenomenon of persistent pink (PP) observed in DC beef. Compared with normal chilling, superchilling significantly increased the redness values of DC steaks through inhibition of lipid oxidation and enhancement of MRA, while it had no effects on PP due to the limited OPD. The results suggest that 80%O 2 -MAP combined with superchilling can be used to improve fresh color and minimize PP of DC beef. • 50%O 2 -MAP and 80%O 2 -MAP effectively improved the surface color of DC steaks. • 80%O 2 -MAP relieved the development of persistent pinking (PP) in cooked DC steaks. • Superchilled beef had higher MRA and redness, but lower TBARS than chilled beef. • Superchilling did not improve the PP of DC steaks due to the limited OPD. The reconstruction of palaeo-oxygenation levels in marine deposits from the Cretaceous has obtained a huge interest all around the world in recent years. This fascinating topic is here pointed out for the first time in the Austral Basin using the information provided by the black shales of the Río Mayer Formation, Patagonia, Argentina. The combination of sedimentology, ichnology and geochemistry (TOC, Ce anomaly and MnO content) allow the identification of three major intervals respecting the oxygen content. During the Berriasian and early Valanginian anoxic conditions prevail in the outer shelf. After that, between the late Valanginian and Hauterivian dysoxic palaeoenvironments were developed. Finally, a more oxygenated palaeoenvironment occurred since Aptian associated with a progradation of a proximal deltaic system. The identification of anoxic conditions is of much interest for the hydrocarbon research in this stratigraphical unit, which represents the most significant source rock of the Austral Basin. Aquatic animals are naturally exposed simultaneously to environments with different concentrations of humic acid (HA) and seasonal or daily variations of dissolved oxygen (DO) levels. This study investigated the effects of simultaneous exposure to different HA and DO levels on plasma ion levels and some hematological and oxidative parameters in different tissues of silver catfish ( Rhamdia quelen). The fish were exposed to 0, 2.5 or 5 mg L HA for 120 h. After this period, each group was divided into two groups: normoxia and hypoxia. Exposure to the different DO levels lasted 96 h, totaling 216 h of experimentation. At the end of the experimental period, blood sampling was performed, and the fish were euthanized prior to the excision of the gills and the brain to evaluate hematological and oxidative parameters. To verify the antioxidant capacity of HA, total phenolic compounds were measured. In general, all tissues of silver catfish exposed simultaneously to hypoxia and different HA concentrations showed a reduction in lipid peroxidation levels, as well as a modulation of the antioxidant system. These effects occurred in an HA concentration-dependent manner. Thus, HA is beneficial to silver catfish exposed to hypoxia. These beneficial effects can be attributed, most likely, to the action of the different HA constituents and functional groups, including phenolic compounds, which have antioxidant properties. In their natural distribution range and in on land-based mariculture facilities, Haliotis midae may be exposed to a relatively wide range of temperatures and oxygen levels, that can induce differential degrees of stress with concomitant metabolic responses. In the current study juvenile H. midae were exposed for one month to two constant temperatures (14°C and 19°C), with three oxygen levels (82%, 98% and 126% oxygen saturation) at each temperature. One additional treatment was included at 19°C, 126% oxygen, where feed was supplemented with l-proline. The results presented here suggest that long-term 19°C exposure is more stressful to H. midae juveniles than a 14°C exposure of the same duration, since exposure to higher temperature generally resulted in higher activities of the antioxidant enzymes measured but lower total antioxidant capacities. Haemolymph glucose levels responded to oxygen treatment at the 14°C exposure, but the same response was not initiated in the 19°C treatment, possibly because the animals exposed to the 19°C treatment relied on proteins to a larger extent. Increased oxygen levels at the 19°C treatment did not cause more DNA damage or protein carbonylation compared to lower oxygen levels. Supplementing feed with l-proline at the high temperature/high oxygen treatment caused a decrease in haemolymph haemocyanin (which may exhibit catalase function) content and a slight decrease in some of the antioxidant enzyme activities, while maintaining the same total antioxidant capacity compared with the same treatment where feed was not supplemented. The overall DNA integrity profile also changed with proline-supplementation, as did the induction level of hsp70. When projecting tissue growth over a 12month period under these conditions, we estimate that H. midae juveniles in this study may exhibit a significant increase in mass when their feed was supplemented with l-proline, indicating that l-proline supplementation provides some benefit to the individual. In this work, pure Nb processed by Powder Metallurgy Hot Isostatic Pressing (PM HIP) was investigated as a potential alternative to C-103 Nb-based alloy for the manufacture of near-net-shape thruster combustion chambers. Three Nb powders with varied particle size range (fine, mid-range and coarse) and C-103 powder were investigated to understand the differences in particle size distribution (PSD), morphology and oxygen (O) content present in the alloy chemistry. The as-HIPed microstructures of pure Nb and C-103 are characterised by a near to fully dense microstructure and the absence of PPBs. Microstructural analyses performed on pure Nb highlighted the influence of particle size on the average as-HIPed grain size. Additionally, it was observed that O content plays a crucial role in the microhardness of pure Nb. Tensile tests performed on Nb mid-range and on Nb mid-range sieved showed that a simple sieve operation was effective in increasing the strength of the material while maintaining good levels of elongation. Alternatively, it was also demonstrated that pure Nb powder heat treatment can be regarded as an effective way to increase the O levels in pure Nb powders as witnessed by the high hardness levels. On the other hand, as-HIPed C-103 powder showed superior tensile properties if compared to the minimum specifications for wrought C-103. Finally, to summarise, the work performed on pure Nb, a simple structure-property relation model was developed to predict the YS of pure Nb based on the O levels and grain size of the as-HIPed microstructure. Abstract: This study examined the effect of dissolved oxygen (DO) level on critical thermal maximum (CTMax) in diploid and triploid brook charr (Salvelinus fontinalis) exposed to a temperature increase of 3°C/h. Because gas solubility is inversely proportional to temperature, DO declines during standard CTMax tests. With this treatment as a baseline, oxygen or nitrogen injection was used to provide three other DO conditions during CTMax tests: two hyperoxic (maintenance at initial 10mg/L and increase from 10mg/L at 2mg/L/h) and one hypoxic (decrease from 10mg/L at 2mg/L/h). Hyperoxia had no effect on temperature at CTMax or time taken to reach CTMax. Hypoxia, on the other hand, resulted in a significantly lower CTMax and shorter time to CTMax than under standard or hyperoxic conditions, with both indices affected by triploidy but not in a consistent fashion: in one experiment triploids had a lower CTMax and shorter time to CTMax than diploids and in a second experiment they had a higher CTMax and longer time to CTMax than diploids. Indices of the secondary stress response (plasma glucose and ions) during CTMax tests under hypoxia responded as would be predicted for an acute stress, with no difference between triploids and diploids. Adaptation to lowering oxygen levels (hypoxia) requires coordinated downregulation of metabolic demand and supply to prevent a mismatch in ATP utilization and production that might culminate in a bioenergetic collapse. Hypoxia diminishes ATP utilization by downregulating protein translation and the activity of the Na-K-ATPase. Hypoxia diminishes ATP production in part by lowering the activity of the electron transport chain through activation of the transcription factor hypoxia-inducible factor-1. The decrease in electron transport limits the overproduction of reactove oxygen species during hypoxia and slows the rate of oxygen depletion to prevent anoxia. • Real time oxygen availability measurement is clinically available with the cellular oxygen metabolism (COMET) monitor. • With COMET we were able to reliably measure mitochondrial oxygen concentrations during photodynamic therapy. • Technique could potentially be used for oxygen concentration feedback dosimetry in PDT. During photodynamic therapy (PDT) oxygen is transformed into reactive oxygen species (ROS) to induce cellular apoptosis in (pre)malignant cells. Real time oxygen availability measurement is clinically available with the Cellular Oxygen Metabolism (COMET) monitor. Primary objective is to show that mitochondrial oxygen availability (mitoPO 2) measurement is possible during clinical ALA-PDT. The secondary aim was to determine the pain sensation, because it is the most commonly reported side effect of PDT. Before and after the two fraction PDT treatment, with a 2-hour dark period, mitoPO 2 was measured and reported pain was documented with a visual analog scale (VAS) 0–100. Nine patients were included. Before the first PDT session the median signal quality was [IQR] 55.0% [34.2–68.0], which decreased after session one to 0% [0.0–10.0]. MitoPO 2 was 40.0 [17.7–53.8] mmHg and increased afterwards to 61.8 [38.2–64.8] mmHg. This likely the result of the delay time between the illumination stop and the mitoPO 2 measurements in a vasodilated, visibly red lesion. Before session two signal quality was 10.4% [0–20.15], 40% lower than at the start. In 5 patients the signal quality after session 2 was too low because of photobleaching and insufficient regeneration of PpIX, median 0% [0–10]. Subjects reported low median VAS scores, all below 3, directly after the mitoPO 2 measurements. With COMET we were able to reliably measure mitochondrial oxygen concentrations during photodynamic therapy. This study assessed the effects of two artificial photoperiods (LD 24:0 and LD 12:12) and three temperature regimes (9, 11, and 18°C) for 30 days on haematological parameters of trout ( Oncorhynchus mykiss) kept in freshwater. Samples were taken at days 7, 14, and 30 during exposure to treatments. A higher mortality (22%) and lower oxygen concentration (<8.0 mg/l) were associated with the combination of photoperiod LD 24:0 and 18°C. The LD 24:0 photoperiod (independently of temperature) increased the haematocrit and the number of erythrocytes at days 7, 14, and 30 ( P < 0.01). A temperature of 18°C (independently of photoperiod regimes) diminished the number of total leucocytes, lymphocytes and thrombocytes ( P < 0.01). The LD 24:0 photoperiod (also independently of temperature) lowered the number of lymphocytes only after 14 days of experimentation ( P < 0.01). Interaction between artificial photoperiod and temperature was only observed at day 14 for polychromatophils ( P < 0.01). These results resemble the effects of stress caused by elevated temperatures and the application of continuous light photoperiods, indicating that survival risks may develop in trout farming when this combination is met. Fossil charcoal, as the direct evidence of wildfires, is present in most of the plant-bearing deposits from the Central European Zechstein (Upper Permian). The charcoal content of Upper Permian sediments collected at the locality Frankenberg–Geismar (NW-Hesse, Germany) is allochthonous and the shape of the fragments, and the rounded edges, indicate transport over relatively long distances. Charcoalified conifer needles are very rare and usually highly fragmented. The anatomical diversity of charred wood is higher than that of the permineralised woods from the Zechstein that have been described previously. It is not yet clear whether this represents a higher diversity of natural taxa or anatomicaly different parts belonging to only a few taxa. Until now, virtually nothing is known about the fire ecology of Late Palaeozoic, conifer-dominated, upland vegetation. The presence of charcoal indicates that these vegetation, which grew under mesic to xeric conditions, may have experienced more or less regular wildfires. Wildfires may have been relatively frequent during the Late Permian, as compared to climatically comparable modern ecosystems. [Copyright &y& Elsevier]

The influence of environmental parameters (water temperature and dissolved oxygen content) on the haemoglobin content of a naturally occurring population of Daphnia carinata was studied in a population resident in an intermittently flowing, shallow body of water. It was found that the Hb content of the animals was influenced by a combination of both the water temperature and dissolved oxygen profiles of the water body. In Summer, stratification, as indicated by sampling at three depths, constrained thermoregulatory behaviour; Hb-poor animals from the upper levels of the water column chose a lower temperature in hypoxia than their Hb-rich counterparts from greater depth. In Winter, the water column was mixed and remained moderately hypoxic at all times. Due to the low temperature of the water, Hb-contents in winter were much lower than those found in Summer. Overall, it was found that thermoregulatory behaviour in D. carinata is dependent upon Hb-content, which is strongly influenced by the animals’ environment. [Copyright &y& Elsevier]

Paleolimnological analyses were used to infer limnological changes during the past ~ 300 yrs in the west basin of Peninsula Lake, a small (853 ha) Precambrian Shield lake in Ontario, Canada, that has been subjected to moderate cultural disturbances (forest clearance, cottage and resort development). This study represents a pioneering attempt to use sedimentary chironomid assemblages and weighted-averaging models to quantify past hypolimnetic anoxia (expressed as the anoxic factor, AF). Impacts of forest clearance and human land-use on deepwater oxygen availability and surface water quality were assessed by comparing chironomid-inferred AF and diatom-inferred total phosphorus concentration ([TP]) to changes in terrestrial pollen and historical data. This study also discusses the ability of chironomids to quantitatively infer changes in AF.Pre-disturbance chironomid assemblages were stable and dominated by taxa indicative of oxygen-rich hypolimnetic conditions (e.g., Protanypus, Heterotrissocladius, Micropsectra type), while diatoms indicated oligotrophic lake status (diatom inferred [TP] = 5-7 μg·l-1). Chironomids characteristic of lower oxygen availability (e.g., Chironomus, Procladius) increased following land-clearance, road construction, establishment of a grist mill and lakeshore development beginning ca. 1870. Increased abundances of Tanytarsus s. lat., a multigeneric group of mainly littoral chironomids, since 1900, indicated that littoral chironomids may have comprised a greater proportion of fossil assemblages during periods of eutrophication and prolonged anoxia. Abundances of meso-eutrophic diatom taxa (e.g., Fragilaria crotonensis, Asterionella formosa, Aulacoseira ambigua, A. subarctica) increased concurrent with European settlement (ca. 1870) and diatom-inferred [TP] doubled (~ 6-12 μg·l-1), further indicating that naturally-oligotrophic Precambrian Shield lakes were extremely sensitive to initial land-clearance activities.Recent increases in oligotrophic diatom taxa (e.g., Cyclotella stelligera) indicate a shift to more oligotrophic conditions since ca. mid-1960s, with greatest changes since ca. 1980. The chironomids Heterotrissocladius and Micropsectra type also increased at this time suggesting greater deepwater oxygen availability. These recent water-quality improvements, possibly in response to enhanced nutrient removal from detergents and sewage, climate-related reductions in external phosphorus loads, and catchment (but not lake) acidification and reforestation, suggest that habitat for commercially-valuable cold-water fishes has improved in recent decades despite greater recreational lake-use.Paleolimnological assessment of trophic status changes in Peninsula Lake using fossil diatom and chironomid assemblages were in good agreement. Diatom inferences of [TP] and chironomid inferences of AF both suggest that Peninsula Lake was historically oligotrophic, became oligo-mesotrophic after European settlement, and returned to oligotrophy in recent yrs. Chironomid inferences of [TP] consistently underestimated the trophic status of Peninsula Lake, possibly due to its relatively large hypolimnion. • The effect of collapse of river-and lake-dominated algae on CH 4 production was estimated. • Anaerobic and reductive conditions were induced by lake-dominated algae collapses. • Lake-dominated algae played a more vital role in CH 4 production than river-dominated algae. • The distinct labile carbon fraction regulated CH 4 production. • Higher mcrA and lower pmoA were found in lake-dominated algae collapse than river-dominated one. The impoundment of dammed rivers accelerates phytoplankton succession from river-dominated to lake-dominated species. Little is known about the role of phytoplankton succession in methane (CH 4) production. In this study, we performed a 61-day microcosm investigation to simulate the collapse processes of Cyclotella meneghiniana (river-dominated algae) and Chlorella pyrenoidosa and Microcystis aeruginosa (lake-dominated algae). The results suggested that different methanogenic conditions were induced by the collapse of river-and lake-dominated algae. The rapid settlement of C. meneghiniana induced aerobic conditions in the water that inhibited anaerobic CH 4 production and intensified CH 4 oxidation as a result of an increase in pmoA. However, the decomposition of C. pyrenoidosa and M. aeruginosa depleted dissolved oxygen and provided abundant labile organic matter, which jointly elevated mcrA and the mcrA/pmoA ratio. Under this condition, anaerobic CH 4 production was the dominant pathway for the mineralization of algae-derived carbon. Finally, the CH 4 produced per unit of particulate total carbon (identified as the carbon content of the algal biomass) by C. pyrenoidosa and M. aeruginosa was 16.29-fold and 8.56-fold higher, respectively, than that produced by C. meneghiniana. These observations provided evidence that lake-dominated algae played a more vital role in CH 4 production than river-dominated algae when algal succession occurred. This discovery might be a new and vital, yet largely underestimated CH 4 emission pathway in river-reservoir systems, that should be considered when evaluating the effect of hydraulic projects on greenhouse gas emissions. Atovaquone (AV) acts on the malaria parasite by competing with ubiquinol (UQH 2 ) for its union to the mitochondrial bc 1 complex, preventing the ubiquinone-8 and ubiquinone-9 (UQ-8 and UQ-9) redox recycling, which is a necessary step in pyrimidine biosynthesis. This study focused on UQ biosynthesis in Plasmodium falciparum and adopted proof-of-concept research to better elucidate the mechanism of action of AV and improve its efficacy. Initially, UQ biosynthesis was evaluated using several radioactive precursors and chromatographic techniques. This methodology was suitable for studying the biosynthesis of both UQ homologs and its redox state. Additionally, the composition of UQ was investigated in parasites cultivated at different oxygen saturations or in the presence of AV. AV affected the redox states of both UQ-8 and UQ-9 homologs by increasing the levels of the respective reduced forms. Conversely, low-oxygen environments specifically inhibited UQ-9 biosynthesis and increased the antimalarial efficacy of AV. These findings encouraged us to investigate the biological importance and the potential of UQ biosynthesis as a drug target based on its inhibition by 4-nitrobenzoate (4-NB), a 4-hydroxybenzoate (4-HB) analog. 4-NB effectively inhibits UQ biosynthesis and enhances the effects of AV on parasitic growth and respiration rate. Although 4-NB itself exhibits poor antimalarial activity, its 50% inhibitory concentration (IC 50 ) value increased significantly in the presence of a soluble UQ analog, p -aminobenzoic acid (pABA), or 4-HB.
