Lung cancer (LC) is a foremost cause of cancer-associated mortalities worldwide and it causes 17 % of the total cancer incidences and 20 % of the cancer-associated mortalities annually. In this exploration, we intended to formulate the Tin oxide-Chitosan-Polyethylene glycol-Crocin nanocomposites (SCP-Cr-NCs) and examine its anticancer role against the LC in both in vitro and in vivo models. The formulated SCP-Cr-NCs were characterized using several methods. The LC was initiated to the BALB/c mice via injecting the 2 × 106 of A549 cells into the posterior flank and treated with the 10 and 20 µM of formulated SCP-Cr-NCs for 19 days. The tumor weight and xenobiotic dysfunction markers status was examined by standard methods. The status of pro-inflammatory markers was studied using ELISA kits. The lung histology was examined microscopically. The viability of SCP-Cr-NCs treated A549 cells were investigated. The ROS and MMP status and apoptosis in the SCP-Cr-NCs treated A549 cells were inspected by fluorescence staining techniques. The tumor weight and the status of xenobiotic dysfunction markers were remarkably reduced by the SCP-Cr-NCs. The SCP-Cr-NCs supplementation also diminished the IL-1β, TNF-α, IL-6, and CEA. The viability of SCP-Cr-NCs administered A549 cells were reduced drastically. The outcomes of fluorescence assay was demonstrated that the SCP-Cr-NCs were diminished the MMP and elevated the ROS status and apoptosis in the A549 cells. SCP-Cr-NCs also restrained the cell adhesion of A549 cells. Collectively, our findings provided the evidence that the formulated SCP-Cr-NCs demonstrated the anticancer potential and it could be a talented chemopreventive candidate to treat the LC. Background: Globally, iron-deficiency anemia (IDA) remains a major health obstacle. This health condition has been identified in 47% of pre-school students (aged 0 to 5 years), 42% of pregnant females, and 30% of non-pregnant females (aged 15 to 50 years) worldwide according to the WHO. Environmental and genetic factors play a crucial role in the development of IDA; genetic testing has revealed the association of a number of polymorphisms with iron status and serum ferritin. Aim: The current study aims to reveal the association of TMPRSS6 rs141312 and BMP2 rs235756 with the iron status of females in Saudi Arabia. Methods: A cohort of 108 female university students aged 18–25 years was randomly selected to participate: 50 healthy and 58 classified as iron deficient. A 3–5 mL sample of blood was collected from each one and analyzed based on hematological and biochemical iron status followed by genotyping by PCR. Results: The genotype distribution of TMPRSS6 rs141312 was 8% (TT), 88% (TC) and 4% (CC) in the healthy group compared with 3.45% (TT), 89.66% (TC) and 6.89% (CC) in the iron-deficient group (P = 0.492), an insignificant difference in the allelic distribution. The genotype distribution of BMP2 rs235756 was 8% (TT), 90% (TC) and 2% (CC) in the healthy group compared with 3.45% (TT), 82.76% (TC) and 13.79% (CC) in iron-deficient group (P = 0.050) and was significantly associated with decreased ferritin status (P = 0.050). In addition, TMPRSS6 rs141312 is significantly (P<0.001) associated with dominant genotypes (TC+CC) and increased risk of IDA while BMP2 rs235756 is significantly (P<0.026) associated with recessive homozygote CC genotypes and increased risk of IDA. The exposure to the electromagnetic fields is well known phenomenon. During the 20th century, environmental exposure to man-made electromagnetic fields has been gradually increasing as growing electricity demand. Every advance technology is posing the challenges in social behavior and created more artificial sources of exposure to the electromagnetic fields now a day. People are exposed to a complex mix of weak electric and magnetic fields, at home and at workplacesbyvarious ways, like as generation and transmission of electricity, domestic appliances, and industrial equipment, to telecommunications and broadcasting. In general, electromagnetic radiations generated as the byproduct of electricity running in electronic devices. An attempt has been made for the study of some valuable bio-effect of electromagnetic radiations with an application of Phantom model. An approach of specific absorption rate (SAR) distribution has been applied in this model. The present results have been complied with the European and USA standards (EN 50361, IEEE 1528) due to an assessment of dosimetric. A gripping device, DUT (Device Under Test) and software Open SAR were used to measure the transmitted radiofrequency (R. F.) or energy in general absorbed by human tissues. The SAR is a function of the electrical conductivity (?), the induced electric field from the radiated energy (measured in Volt/meter) and the mass density of human tissue. Due to the high sensitivity of the probe, its output voltage is measured without amplification. The electric field probe corresponds to the recommendations of CENELEC (European Committee for the Coordination of Electrical Standards) and ICEEE (the International Committee of Electrical and Electronic Engineers) for measuring electromagnetic fields of cell phones, base stations, and various radiating devices. Prostate cancer (PC) is the most prevalent type of cancer in men worldwide. In Saudi Arabia, the rate of PC is increasing annually. The sex steroid hormones androgens and their receptors have critical roles in PC development and progression. Additionally, apoptosis-related proteins such as heat-shock proteins are vital molecules in PC development. Steroid hormone-deprivation therapies remain the essential treatment for patients with metastatic PCs; however, acquired resistance to hormone deprivation and the transition to PC androgen independence is a major health obstacle. COVID-19 caused by the SARS-CoV-2 virus is widespread in all regions, and it disturbs host immune system functioning leading to extreme inflammatory reaction and hyperactivation of the immune response. Kabasura Kudineer (KSK) is preventive medicine against viral infections and a potent immune booster for inflammation-related diseases. We hypothesize that KSK and KSK similar plant compounds, might prevent or control the COVID-19 infection in the human body. 1,207 KSK and KSK similar compounds were listed and screened via the Swiss ADME tool and PAINS Remover; 303 compounds were filtered including active and similar drug compounds. The targets were retrieved from similar drugs of the active compounds of KSK. Finally, 573 genes were listed after several screening steps. Next, network analysis was performed to finalize the potential target gene: construction of protein-protein interaction of 573 genes using STRING, identifying top hub genes in Cytoscape plug-ins (MCODE and cytoHubba). These ten hub genes play a crucial role in the inflammatory response. Target-miRNA interaction was also constructed using the miRNet tool to interpret miRNAs of the target genes and their functions. Functional annotation was done via DAVID to gain a complete insight into the mechanism of the enriched pathways and other diseases related to the given target genes. In Molecular Docking analysis, IL10 attained top rank in Target-miRNA interaction and also the gene formed prominent exchanges with an excellent binding score (> = -8.0) against 19 compounds. Among them, Guggulsterone has an acute affinity score of -8.8 for IL10 and exhibits anti-inflammatory and immunomodulatory properties. Simple Summary: The results of prior studies on the genotypes and allele variations of the antiapoptotic gene and the risk of lymphoma are unclear. To address this gap, this case–control study included 205 Saudi patients, 100 of whom had lymphoma, and 105 who were healthy controls. Antiapoptotic genes, including B-cell lymphoma-2 (BCL2-938 C > A), MCL1 (rs9803935 T > G), and survivin (BIRC5-rs17882312 G > C and BIRC5-rs9904341 G > C) were identified using the tetra amplification refractory mutation polymerase chain reaction (PCR). BIRC5-C, MCL1-G, and BIRC5-G alleles were identified using allele-specific PCR. In addition to BCL-2-A mutations, lymphoma patients are more likely to have antiapoptotic gene genotypes and allele variations than healthy volunteers. The common bean (Phaseolus vulgaris L.), whose annual production is 26 million tons worldwide, is one of the main sources of protein and is known as one of the most important food sources. In this study, the karyotype variations and the genome size of four common bean genotypes in Turkey were investigated to determine whether the geographic variables in these regions affected the genome size and the karyotype parameters. In addition, it is known that as that the cytological and chromosomal parameters change under the influence of the climatic conditions of each region, appropriate and stable cytological methods for each plant facilitate and enable the determination of the chromosomal structure and the identification of specific chromosomes in the genotypes of the relevant region. Correct and valuable information such as this enables breeders and researchers to determine the correct shape and actual size of chromosomes. The genome size of the genotypes was measured with a flow cytometer, and chromosome analyses were performed with the squash method. For each genotype, the karyotype parameters, such as the number of somatic chromosomes, the Mean Total Chromosome Length (MTCL), the Mean Centromere Index (MCI), and the Mean Arm Ratio (MAR), were measured. The results showed that the highest and the lowest amounts of DNA per nucleus (3.28 pg and 1.49 pg) were observed in the Bitlis and Elaziğ genotypes. In addition, all genotype chromosome numbers were counted to be 2n = 2x = 22. The Mean Total Chromosome Length varied from 15.65 µm in Elaziğ to 34.24 µm in the Bitlis genotype. The Mean Chromosome Length ranged between 1.42 µm and 3.11 µm in the Elaziğ and Bitlis genotypes. The Hakkari and Van genotypes consist of eleven metacentric chromosomes, while the Bitlis and Elaziğ genotypes consist of ten metacentric chromosomes and one sub-metacentric chromosome. However, the Mean Centromere Index and Arm Ratio differed considerably among the genotypes. The highest (46.88) and the lowest (43.18) values of the Mean Centromere Index were observed in the Hakkari and Elaziğ genotypes, respectively. On the other hand, the lowest (1.15) and the highest (1.36) values of the Mean Arm Ratio were obtained in the Bitlis and Elaziğ genotypes, respectively. Eventually, intraspecies variations in genome size and chromosomal parameters were observed, and it was determined that the changes in nuclear DNA content and different chromosomal parameters among the four Phaseolus genotypes from four different regions of Turkey indicate the effect of climate change in the regions on these parameters. Previous studies reported disrupted hepatic function and structure following the administration of cyclosporine A (CsA) in humans and animals. Recently, we found that avocado seeds (AvS) ameliorated CsA-induced nephrotoxicity in rats. As a continuation, herein we checked whether AvS could also attenuate CsA-induced hepatotoxicity in rats. Subcutaneous injection of CsA (5 mg/kg) for 7 days triggered hepatotoxicity in rats, as indicated by liver dysfunction, redox imbalance, and histopathological changes. Oral administration of 5% AvS powder for 4 weeks ameliorated CsA-induced hepatotoxicity, as evidenced by (1) decreased levels of liver damage parameters (alanine aminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphatase (ALP), and bilirubin), (2) resumed redox balance in the liver (reduced malondialdehyde (MDA) and increased superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GPx)), (3) downregulated hepatic expression of endoplasmic reticulum (ER) stress-related genes (X-box binding protein 1 (XBP1), binding immunoglobulin protein (BIP), C/EBP homologous protein (CHOP)), and apoptosis-related genes (Bax and Casp3), (4) upregulated expression of the anti-apoptotic gene Bcl2, (5) reduced DNA damage, and (6) improved liver histology. In this study, bioinformatics analyses were conducted to identify possible pathogenic roles of CK2α, which is a protein encoded by CSNK2A1, in the progression and aggressiveness of CLL. Furthermore, various computational tools were used to search for a competent inhibitor of CK2α from fungal metabolites that could be proposed for CLL therapy. In CLL patients, high-expression of CSNK2A1 was associated with early need for therapy (n = 130, p < 0.0001) and short overall survival (OS; n = 107, p = 0.005). Consistently, bioinformatics analyses showed CSNK2A1 to associate with/play roles in CLL proliferation and survival-dependent pathways. Furthermore, PPI network analysis identified interaction partners of CK2α (PPI enrichment p value = 1 × 10–16) that associated with early need for therapy (n = 130, p < 0.003) and have been known to heavily impact on the progression of CLL. These findings constructed a rational for targeting CK2α for CLL therapy. Consequently, computational analyses reported 35 fungal metabolites out of 5820 (filtered from 19,967 metabolites) to have lower binding energy (ΔG: − 10.9 to − 11.7 kcal/mol) and better binding affinity (Kd: 9.77 × 107 M−1 to 3.77 × 108 M−1) compared with the native ligand (ΔG: − 10.8, Kd: 8.3 × 107 M−−1). Furthermore, molecular dynamics simulation study established that Butyl Xanalterate-CK2α complex continuously remained stable throughout the simulation time (100 ns). Moreover, Butyl Xanalterate interacted with most of the catalytic residues, where complex was stabilized by more than 65% hydrogen bond interactions, and a significant hydrophobic interaction with residue Phe113. Here, high-expression of CSNK2A1 was implicated in the progression and poor prognosis of CLL, making it a potential therapeutic target in the disease. A technique to predict crucial clinical prostate cancer (PC) is desperately required to prevent diagnostic errors and overdiagnosis. To create a multimodal model that incorporates long-established messenger RNA (mRNA) indicators and conventional risk variables for identifying individuals with severe PC on prostatic biopsies. Urinary has gathered for mRNA analysis following a DRE and before a prostatic examination in two prospective multimodal investigations. A first group (n = 489) generated the multimodal risk score, which was then medically verified in a second group (n = 283). The full-length BRCA1-associated RING domain 1 (BARD1) gene encodes a 777-aa protein. BARD1 displays a dual role in cancer development and progression as it acts as a tumor suppressor and an oncogene. Structurally, BARD1 has homologous domains to BRCA1 that aid their heterodimer interaction to inhibit the progression of different cancers such as breast and ovarian cancers following the BRCA1-dependant pathway. In addition, BARD1 was shown to be involved in other pathways that are involved in tumor suppression (BRCA1-independent pathway) such as the TP53-dependent apoptotic signaling pathway. However, there are abundant BARD1 isoforms exist that are different from the full-length BARD1 due to nonsense and frameshift mutations, or deletions were found to be associated with susceptibility to various cancers including neuroblastoma, lung, breast, and cervical cancers. This article reviews the spectrum of BARD1 full-length genes and its different isoforms and their anticipated associated risk. Additionally, the study also highlights the role of BARD1 as an oncogene in breast cancer patients and its potential uses as a prognostic/diagnostic biomarker and as a therapeutic target for cancer susceptibility testing and treatment. In this article we report a novel Ag NPs fabricated chitosan-agarose composite functionalized core-shell type Fe 3 O 4 nanoparticle (Ag/CS-Agar@Fe 3 O 4). The biogenic material was analyzed over a number of physicochemical methods like, FT-IR, FE-SEM, TEM, EDX, XRD, VSM and ICP-OES. In catalytic exploration we aimed the synthesis of diverse 2 H -indazolo0- b ]phthalazine-trione derivatives via one-pot three component coupling of phathalalhydrazide, dimedone and different aldehydes. It afforded good to excellent yields under solvent-less conditions. Robustness of the catalyst was justified by catalyst recyclability for consecutive 10 times, hot filtration and leaching tests. Again, biological activity of the material was evaluated by studying the antioxidant and cytotoxicity properties over lung and liver cancer cell lines. Antioxidant potential of Ag/CS-Agar@Fe 3 O 4 was assessed by DPPH radical scavenging studies and the corresponding IC50 was found to be 96.57 μg/mL. Liver and lung cancer studies over Ag/CS-Agar@Fe 3 O 4 was carried out by MTT assay against HepG2 and A549 cell lines. The corresponding IC50 values were found as 192.35 and 365.28 μg/mL respectively. % Cell viability of the nanomaterial decreased dose dependently over both the cell lines without any cytotoxicity on normal cell line. Perception of hub genes engaged in metastatic gastric cancer (mGC) promotes novel ways to diagnose and treat the illness. The goal of this investigation is to recognize the hub genes and reveal its molecular mechanism. In order to explore the potential facts for gastric cancer, the expression profiles of two different datasets were used (GSE161533 and GSE54129). The genes were confirmed to be part of the PPI network for gastric cancer pathogenesis and prognosis. In Cytoscape, the CytoHubba module was used to discover the hub genes. Responsible hub genes were identified. Data from Kaplan–Meier plotter confirmed the predictive value of these distinct genes in various stages of gastric malignancy. Upregulated and downregulated genes were identified to utilize for further analysis. Positive regulation by a host of viral process, positive regulation of granulocyte differentiation, negative regulation of histone H3–K9 methylation were found in DEGs analysis. In addition, five KEGG pathways were identified as an essential enhancer that include nucleotide excision repair; base excision repair; DNA replication; homologous recombination; and complement and coagulation cascades. POLE, BUB1B, POLD4, C3, BLM, CCT7, PRPF31, APEX1, PSMA7, and CDC45 were chosen as hub genes after combining the PPI results. Our study recommends that BUB1B, CCT7, APEX1, PSMA7, and CDC45 might be potential biomarkers for gastric cancer. These biomarkers are upregulated genes. In the current investigation, we studied the potential anti‐ulcerative activity of PAs against acidified ethanol (HCl/ethanol)‐caused gastric ulceration. Fifty male albino Wistar rats were allocated into five equal groups: control, HCl/ethanol (3 mL/kg), lansoprazole (LPZ, 30 mg/kg) + HCl/ethanol, and PAs (100 and 250 mg/kg) + HCl/ethanol. LPZ and PAs were applied one week before gastric ulcer induction. PAs pretreatment notably reduced gastric mucosal macroscopic and microscopic pathological changes in a dose‐dependent manner. Additionally, PAs activated the innate antioxidant molecules including glutathione and its derived antioxidants (glutathione peroxidase and glutathione reductase), along with superoxide dismutase and catalase, while attenuating pro‐oxidant formation, including malondialdehyde and nitric oxide. Interestingly, PAs supplementation at a higher dose suppressed gastric inflammatory and apoptotic responses, as demonstrated by the reduced levels of interleukin‐1β, interleukin‐6, tumor necrosis factor alpha, high‐mobility group box 1, cyclooxygenase 2, prostaglandin E2, nuclear factor kappa‐B, Bcl‐2‐associated X protein, and caspase‐3, while B cell lymphoma 2 was elevated. Hence, PAs could exhibit antiulcer activity by protecting gastric tissue from the development of oxidative damage, inflammatory responses, and apoptosis events associated with ulceration. Practical implications: Gastric ulcer is a lesion in the gastric mucosal layer associated with excessive inflammatory response, apoptotic events, oxidative damage, and irritation, and may develop into cancer with about 5%–10% morbidity rate. However, medications commonly used in GU treatment cannot normalize gastric mucosa, while causing several adverse effects. Therefore, new therapeutic approaches are needed to treat or prevent gastric ulceration. Proanthocyanidins (PAs, condensed tannins) are dietary flavonoids found in abundance in different plant species, including their fruits, bark, and seeds. Due to their potent antioxidative activity, PAs have been applied to prevent or treat oxidative stress‐related diseases, including cancer, as well as metabolic, neurodegenerative, cardiovascular, and inflammatory disorders. The CLEC-2 receptor protein belongs to the C-type lectin superfamily of transmembrane receptors that have one or more C-type lectin-like domains. CLEC-2 is a physiological binding receptor of podoplanin (PDPN), which is expressed on specific tumour cell types and involved in tumour cell-induced platelet aggregation and tumour metastasis. CLEC-2 and podoplanin-expressing tumour cells interact to increase angiogenesis, tumour development, and metastasis. CLEC-2 is a hemi-immunoreceptor tyrosine-based activation motif (hemi-ITAM) receptor located on platelets and a subset of dendritic cells that are expressed constitutively. This molecule is secreted by activated platelets around tumours and has been shown to inhibit platelet aggregation and tumour metastasis in colon carcinoma by binding to the surface of tumour cells. Pharmacokinetic studies were carried using a DrugLiTo, and molecular docking was performed using AutoDock Tools 1.5.6 (ADT). Twenty-nine bioactive compounds were included in the study, and four of them, namely, piperine, dihydrocurcumin, bisdemethoxycurcumin, and demothoxycurcumin, showed potential antagonist properties against the target. The resultant best bioactive was compared with commercially available standard drugs. Further, validation of respective compounds with an intensive molecular dynamics simulation was performed using Schrödinger software. To the best of our knowledge, this is the first report on major bioactive found on clove as natural antagonists for CLEC-2 computationally. A series of sequential accumulation of genetic and epigenetic changes are responsible for the initiation and progression of diseases via the normal > adenoma > carcinoma sequence. Genetic variants in crucial cancer-causing genes are known to mediate the risk of cancer. Objective. In this case-control study, we examined single nucleotide polymorphism (SNP) in HER1 (rs763317 and rs3752651) and HER2 (rs1136201 and rs1058808) genes to assess their role in the susceptibility of CRC in a Saudi population. Methods. TaqMan allelic discrimination assay was utilized to identify the genotypes in 163 normal and 143 CRC patients. Results. In the overall analysis, the rs3752651 and rs1136201 were significantly associated with the risk of CRC. Although none of the examined SNPs had any impact on the age at which CRC was diagnosed, interestingly, three SNPs showed a significant association based on gender. The rs3752651 conferred significant protection only in men, whereas rs1136201 diminished the risk and rs1058808 considerably increased the susceptibility of CRC only in women. Conclusions. Our result suggests that these SNPs in HER1 and HER2 after validation in larger cohorts of different ethnicities may be utilized as genetic screening markers for predicting colorectal cancer predisposition. In the clinical setting, iron deficiency anaemia (IDA) represents a major global health concern. This health condition is reported in 30% of non-pregnant women, 42% of pregnant women (aged 15–50 years), 12.7% of men (15 years or older) and in 47% of preschool children (aged 0 to 5 years). Several genetic polymorphisms associated with iron status have been identified by using genome-wide association studies. This study aimed to identify the functional polymorphisms rs855791 and rs2111833 in the transmembrane serine protease 6 (TMPRSS6) gene in female university students with IDA in the Kingdom of Saudi Arabia. About 108 female students, aged from 18 to 25 years, were randomly selected and included to this study. Fifty-eight participants were iron deficient, and fifty participants were healthy. Blood samples were collected from all participants and assessed based on their haematological and biochemical iron status indices. Genotyping was carried out by using PCR. The genotype distribution of the TMPRSS6 rs855791 region in female students from Tabuk University, northern Saudi Arabia, was 0% (CC), 77.6% (CT) and 22.4% (TT) in the iron-deficient students compared to 2% (CC), 96% (CT) and 2% (TT) in the healthy students, indicating significant differences in the allelic distribution between the iron-deficient group and the healthy group. The genotype distribution of the TMPRSS6 rs2111833 polymorphism was 8.6% (GG), 89.7% (GA) and 1.7% (AA) in the iron-deficient students compared to 6% (GG), 92% (GA) and 2% (AA) in the healthy students, respectively, showing no differences between the iron-deficient group and the healthy group in allelic distribution. Our data demonstrated that the TMPRSS6 polymorphism rs855791 is significantly associated with decreased iron status, whereas TMPRSS6 polymorphism rs2111833 is not linked with iron deficiency status in female university students in northern Saudi Arabia. Varumin (1and 2)is known as commercially produced drugsin the Republic of Macedonia. It contains different herbal medicines such as mistletoe, elecampane, corneltree bark, marigold, yarrow, and cynodon and St John’s wort. In addition, Varumin (1 and 2)has been designed to increase the defending strength (immunity) of different parts of the human body infected by acute and chronic diseases. Since herbal drugs profiles of Varumin (1 and 2), the present study aimed to assessing the chemical profile of potent compounds in order to give a better understanding of possible therapeutical mechanisms in the occurrence of diseases via detecting compounds present in Varumin (1 and 2). The obtained results revealed that the amount of Rutin and Quercetin ranged between 63.04μg/mL to 64.09μg/mL and 65.93μg/mL to 66.26μg/mL respectively. And, the total flavonoids were found between the range of 1.901±0.08μg/mL to 2.094±0.08μg/mL. Finally, the Non Enzymatic Antioxidant properties (NEAOP) have been studied well and the obtained results were in the range of 85.91±1.63 μg/mL to 97.95 ±0.24 μg/mL.
