Despite substantial reductions in anthropogenic emissions, ozone (O3) pollution remains a severe environmental problem in urban China. These reductions affect ozone formation by altering levels of O3 precursors, intermediates, and the oxidation capacity of the atmosphere. However, the underlying mechanisms driving O3 changes are still not fully understood. Here, we employ a regional chemical transport model to quantify ozone changes due to a specified emission reduction (50 %) for winter and summer conditions in 2018. Our results indicate that reductions in nitrogen oxide (NOx) emissions increase surface O3 concentrations by 15 %–33 % on average across China in winter and by up to 17 % in volatile organic compound (VOC)-limited areas during summer. These ozone increases are associated with a reduced NOx titration effect and higher levels of OH radicals. Reducing NOx emissions significantly decreases the concentration of particulate nitrate, which enhances ozone formation through increased HO2 radical levels due to reduced aerosol uptake and diminished aerosol extinction. Additionally, an enhanced atmospheric oxidative capacity, driven by larger contributions from the photolysis of oxidized VOCs (OVOCs) and OH-related reactions, also favors urban ozone formation. With additional reductions in anthropogenic VOC emissions, increases in summertime ozone (VOC-limited areas) can be offset by reduced production of radicals from VOC oxidations. To effectively mitigate ozone pollution, a simultaneous reduction in the emission of NOx and specific VOC species should be applied, especially regarding alkenes, aromatics, and unsaturated OVOCs, including methanol and ethanol.

A multiscale modeling ensemble chain has been assembled as a first step towards an air quality analysis and forecasting (AQF) system for Latin America. Two global and three regional models were tested and compared in retrospective mode over a shared domain (120–28° W, 60° S–30° N) for the months of January and July 2015. The objective of this experiment was to understand their performance and characterize their errors. Observations from local air quality monitoring networks in Colombia, Chile, Brazil, Mexico, Ecuador and Peru were used for model evaluation. The models generally agreed with observations in large cities such as Mexico City and São Paulo, whereas representing smaller urban areas, such as Bogotá and Santiago, was more challenging. For instance, in Santiago during wintertime, the simulations showed large discrepancies with observations. No single model demonstrated superior performance over others or among pollutants and sites available. In general, ozone and NO2 exhibited the lowest bias and errors, especially in São Paulo and Mexico City. For SO2, the bias and error were close to 200 %, except for Bogotá. The ensemble, created from the median value of all models, was evaluated as well. In some cases, the ensemble outperformed the individual models and mitigated extreme over- or underestimation. However, more research is needed before concluding that the ensemble is the path for an AQF system in Latin America. This study identified certain limitations in the models and global emission inventories, which should be addressed with the involvement and experience of local researchers.

Cloud properties are strongly influenced by ice formation; hence, we need to understand the sources of ice-nucleating particles (INPs) around the globe. Boreal forests are known as sources of bioaerosol, and recent work indicates that these dominate the INP spectra above −24 °C. To quantify the INP population at temperatures below −24 °C, we deployed a portable cloud expansion chamber (PINE) in a Finnish boreal forest from 13 March 2018 to 11 May 2018. Using the 6 min time resolution PINE data, we present several lines of evidence that INPs below −24 °C in this location are also from biological sources: (i) an INP parameterization developed for a pine forest site in Colorado, where many INPs were shown to be biological, produced a good fit to our measurements; a moderate correlation of INPs with aerosol concentration larger than 0.5 µm and the fluorescent bioaerosol concentration; (ii) a negative correlation with relative humidity that may relate to enhanced release of bioaerosol at low humidity from local sources such as the prolific lichen population in boreal forests; and (iii) the absence of correlation with ultra-fine particles (3.5 to 50 nm), indicating that new particle formation events are not sources of INPs. This study should motivate further work to establish whether the commonality in bioaerosol ice-nucleating properties between spring in Finland and summer in Colorado is more generally applicable to different coniferous forest locations and times and also to determine to what extent these bioaerosols are transported to locations where they may affect clouds.

Ice nucleating particles (INPs) play a crucial role in initiating ice crystal formation in clouds, influencing the dynamics and optical properties of clouds and their impacts on precipitation and the climate system. Despite their importance, there is limited knowledge about the vertical distribution of INPs. This study focuses on aircraft measurements conducted during spring 2018 above the boreal forest of Hyytiälä, Finland. Similarities between INP concentrations, activated fractions, particle concentrations, and size distributions observed at ground level and in the boundary layer aloft indicate that surface particles and INPs are efficiently transported and mixed within the boundary layer. INP concentrations observed in the boundary layer are best predicted by a parameterization describing near-surface INP concentrations driven by the abundance of biogenic aerosol in the Finnish boreal forest, suggesting that biogenic INPs are dominant in the boundary layer above the same environment. Most of the INP concentrations and activated fractions observed in the free troposphere are notably lower than in the boundary layer, and the distinct particle size distributions suggest that different aerosol populations, likely resulting from long-range transport, are present in the free troposphere. However, we show one case where higher INP concentrations are observed in the free troposphere and where a homogeneous particle population exists from the surface to the free troposphere. This indicates that surface particles and INPs from the boreal forest can occasionally reach the free troposphere, which is particularly important as the INPs in the free troposphere can further travel horizontally and/or vertically and impact cloud formation.

Abstract Monoterpenoid indole alkaloid (MIA) biosynthesis in Catharanthus roseus is a paragon of the spatiotemporal complexity achievable by plant specialized metabolism. Spanning a range of tissues, four cell types, and five cellular organelles, MIA metabolism is intricately regulated and organized. This high degree of metabolic differentiation requires inter-cellular and organellar transport, which remains understudied. Here, we have characterized a vacuolar importer of secologanin belonging to the multidrug and toxic compound extrusion (MATE) family, named CrMATE1. Phylogenetic analyses of MATEs suggested a role in alkaloid transport for CrMATE1, and in planta silencing in two varieties of C. roseus resulted in a shift in the secoiridoid and MIA profiles. Subcellular localization of CrMATE1 confirmed tonoplast localization. Biochemical characterization was conducted using the Xenopus laevis oocyte expression system to determine substrate range, directionality, and rate. We can confirm that CrMATE1 is a vacuolar importer of secologanin, translocating 1 mM of substrate within 25 min. The transporter displayed strict directionality and specificity for secologanin and did not accept other secoiridoid substrates. The unique substrate-specific activity of CrMATE1 showcases the utility of transporters as gatekeepers of pathway flux, mediating the balance between a defense arsenal and cellular homeostasis.

Abstract The ocean surface mixed layer plays a crucial role as an entry or exit point for heat, salt, momentum, and nutrients from the surface to the deep ocean. In this study, we introduce a framework to assess the evolution of the mixed layer depth (MLD) for realistic forcings and preconditioning conditions. Our approach involves a physically‐based parameter space defined by three dimensionless numbers: λs representing the relative contribution of the buoyancy flux and the wind stress at the air‐sea interface, Rh the Richardson number which characterizes the stability of the water column relative to the wind shear, and f/Nh which characterizes the importance of the Earth's rotation (ratio of the Coriolis frequency f and the pycnocline stratification Nh). Four MLD evolution regimes (“restratification,” “stable,” “deepening,” and “strong deepening”) are defined based on the values of the normalized temporal evolution of the MLD. We evaluate the 3D parameter space in the context of 1D simulations and we find that considering only the two dimensions (λs, Rh) is the best choice of 2D projection of this 3D parameter space. We then demonstrate the utility of this two‐dimensional λs − Rh parameter space to compare 3D realistic ocean simulations: we discuss the impact of the horizontal resolution (1°, 1/12°, or 1/60°) and the Gent‐McWilliams parameterization on MLD evolution regimes. Finally, a proof of concept of using observational data as a truth indicates how the parameter space could be used for model calibration.

Objectives To investigate if patients with early rheumatoid arthritis responding insufficiently to initial methotrexate (MTX) and bridging glucocorticoids (GCs) could benefit from early but temporary etanercept introduction as a second remission-induction attempt.Methods CareRA2020 (NCT03649061) was a 2-year, open-label, multicentre, pragmatic randomised controlled trial. Treatment-naïve patients started MTX and GC bridging (COBRA-Slim: CS). Within a time window from week (W) 8 until W32, early insufficient responders (28-joint Disease Activity Score – C-reactive Protein (DAS28-CRP) >3.2 between W8 and W32 or ≥2.6 at W32) were randomised to a Standard-CS strategy (adding leflunomide first) or Bio-induction-CS strategy (adding etanercept for 24 weeks). Additional treatment adaptations followed the treat-to-target principle. Longitudinal disease activity (DAS28-CRP) over 104 weeks (primary outcome), achievement of DAS28-CRP

Abstract Geological evidence indicates that a supernova within 100 parsecs of Earth occurs around once per million years. Such nearby supernovas can produce an intense gamma-ray burst and a 100-fold increase of cosmic rays, lasting several centuries. We find that the effect of a short burst of gamma rays is small since they are strongly attenuated before reaching the lower stratosphere. Intense cosmic radiation affects stratospheric ozone but, due to compensating effects in catalytic chemical cycles, ozone depletion is moderate and comparable to that from current anthropogenic emissions. This also holds for the low-oxygen atmosphere during early evolution of terrestrial life. We estimate the increase in aerosol and clouds from a 100-fold increase of cosmic rays exerts a radiative forcing comparable in magnitude but opposite in sign to current anthropogenic climate forcing. We conclude that Earth’s atmosphere is effective at shielding the biosphere from nearby supernovae.

To manage Earth in the Anthropocene, new tools, new institutions, and new forms of international cooperation will be required. Earth Virtualization Engines is proposed as an international federation of centers of excellence to empower all people to respond to the immense and urgent challenges posed by climate change.

Abstract A key hallmark of Parkinson’s disease (PD) is Lewy pathology. Composed of α-synuclein, Lewy pathology is found both in dopaminergic neurons that modulate motor function, and cortical regions that control cognitive function. Recent work has established the molecular identity of dopaminergic neurons susceptible to death, but little is known about cortical neurons susceptible to Lewy pathology or molecular changes induced by aggregates. In the current study, we use spatial transcriptomics to capture whole transcriptome signatures from cortical neurons with α-synuclein pathology compared to neurons without pathology. We find, both in PD and related PD dementia, dementia with Lewy bodies and in the pre-formed fibril α-synucleinopathy mouse model, that specific classes of excitatory neurons are vulnerable to developing Lewy pathology. Further, we identify conserved gene expression changes in aggregate-bearing neurons that we designate the Lewy-associated molecular dysfunction from aggregates (LAMDA) signature. Neurons with aggregates downregulate synaptic, mitochondrial, ubiquitin-proteasome, endo-lysosomal, and cytoskeletal genes and upregulate DNA repair and complement/cytokine genes. Our results identify neurons vulnerable to Lewy pathology in the PD cortex and describe a conserved signature of molecular dysfunction in both mice and humans.

This study is anchored in the H2020 SEAMLESS project (https://www.seamlessproject.org, last access: 29 January 2024), which aims to develop ensemble assimilation methods to be implemented in Copernicus Marine Service monitoring and forecasting systems, in order to operationally estimate a set of targeted ecosystem indicators in various regions, including uncertainty estimates. In this paper, a simplified approach is introduced to perform a 4D (space–time) ensemble analysis describing the evolution of the ocean ecosystem. An example application is provided, which covers a limited time period in a limited subregion of the North Atlantic (between 31 and 21∘ W, between 44 and 50.5∘ N, between 15 March and 15 June 2019, at a 1/4∘ and a 1 d resolution). The ensemble analysis is based on prior ensemble statistics from a stochastic NEMO (Nucleus for European Modelling of the Ocean)–PISCES simulator. Ocean colour observations are used as constraints to condition the 4D prior probability distribution. As compared to classic data assimilation, the simplification comes from the decoupling between the forward simulation using the complex modelling system and the update of the 4D ensemble to account for the observation constraint. The shortcomings and possible advantages of this approach for biogeochemical applications are discussed in the paper. The results show that it is possible to produce a multivariate ensemble analysis continuous in time and consistent with the observations. Furthermore, we study how the method can be used to extrapolate analyses calculated from past observations into the future. The resulting 4D ensemble statistical forecast is shown to contain valuable information about the evolution of the ecosystem for a few days after the last observation. However, as a result of the short decorrelation timescale in the prior ensemble, the spread of the ensemble forecast increases quickly with time. Throughout the paper, a special emphasis is given to discussing the statistical reliability of the solution. Two different methods have been applied to perform this 4D statistical analysis and forecast: the analysis step of the ensemble transform Kalman filter (with domain localization) and a Monte Carlo Markov chain (MCMC) sampler (with covariance localization), both enhanced by the application of anamorphosis to the original variables. Despite being very different, the two algorithms produce very similar results, thus providing support to each other's estimates. As shown in the paper, the decoupling of the statistical analysis from the dynamical model allows us to restrict the analysis to a few selected variables and, at the same time, to produce estimates of additional ecological indicators (in our example: phenology, trophic efficiency, downward flux of particulate organic matter). This approach can easily be appended to existing operational systems to focus on dedicated users' requirements, at a small additional cost, as long as a reliable prior ensemble simulation is available. It can also serve as a baseline to compare with the dynamical ensemble forecast and as a possible substitute whenever useful.

Abstract This paper studies the rating lives of wind turbine main bearings, as determined by the IEC 61400‐1 and ISO 281 standards. A critical review of relevant bearing life theory and turbine design requirements is provided, including discussion on possible shortcomings such as the existence (or not) of the bearing fatigue load limit and the validity of assuming linear damage accumulation. A detailed exploratory case study is then undertaken to determine rating lives for two models of main bearing in a 1.5 MW wind turbine. Rating life assessment is carried out under different conditions, including various combinations of main bearing temperature, wind field characteristics, lubricant viscosity, and contamination levels. Rating lives are found to be sufficiently above the desired 20‐year design life for both bearing models under expected operating conditions. For the larger bearing, operational loads are shown to be below or close to the bearing fatigue load limit a vast majority of the time. Key sensitivities for rating life values are temperature and contamination. Overall, the results of this study suggest that an ISO 281 rating life assessment does not account for reported rates of main bearing failures in 1 to 3 MW wind turbines. It is recommended that a similar analysis be undertaken for ISO/TS 16281 rating lives, along with further efforts to identify principal root causes of main bearing failures in future work, possibly leading to a new application standard specific to this component. It is also recommended that the impacts of partial wake impingement on main bearing rating lives are investigated.

Purpose: This study aimed to investigate the impact of a university's abrupt closure on the lives of graduate students and examined the role of grit, social support and school identification as they transferred to a new institution. Design/methodology/approach: An online survey was administered to a diverse sample of 130 psychology graduate students after their university unexpectedly closed and the students transferred to another institution to complete their degrees. The survey included both qualitative and quantitative measures of the impact on different areas of their lives, anxiety and depression, life satisfaction, social support, grit and school identification. Findings: Students reported experiencing financial loss, family and marital strain, anger, fear, uncertainty and loss of progress toward professional goals. Nearly 40% of participants had scores indicating elevated anxiety and depression. Students with higher anxiety and depression and lower life satisfaction tended to identify less with their new institution. Both social support and grit were related to better adjustment and life satisfaction. Originality/value: To the best of the authors' knowledge, this is the first study to examine the impact on graduate students following the closure of an institution of higher education. The findings have implications for educational policy and graduate student support services.

Abstract Beyond ecological and health impacts, invasive alien plant species can generate indirect and direct costs, notably through reduced agricultural yields, restoration, and management of the invaded environment. Acacia dealbata and Ailanthus altissima are invasive plant species that cause particularly significant damage to the railway network in the Mediterranean area. The allelopathic properties of Mediterranean plant species could be used as nature‐based solutions to slow down the spread of such invasive plant species along railway borders. In this context, a mesocosm experiment was set‐up: (i) to test the potential allelopathic effects of Cistus ladanifer, Cistus albidus, and Cotinus coggygria leaf aqueous extracts on seed germination and seedling growth of A. dealbata and A. altissima; (ii) to evaluate whether these effects depend on the extract dose; and finally, (iii) to estimate whether these effects are modified by soil amendment. Leaf aqueous extracts of the three native plant species showed negative effects on both seed germination and seedling growth of the two invasive species. Our results show that the presence of allelochemicals induces a delay in seed germination (e.g., A. dealbata germination lasted up to 269% longer in the presence of high‐dose leaf aqueous extracts of C. coggygria), which can lead to a decrease in individual recruitment. They also highlight a decrease in seedling growth (e.g., high‐dose C. coggygria leaf aqueous extracts induced a 26% decrease in A. dealbata radicle growth), which can alter the competitiveness of invasive species for resource access. Our results also highlight that compost addition limits the inhibitory effect of native Mediterranean plants on the germination of invasive alien plants, suggesting that soil organic matter content can counteract allelopathic effects on invasive alien plants. Thus, our findings revealed that the allelopathic potential of certain Mediterranean plant species could be a useful tool to manage invasive plant species.

The conservation and management of marine ecosystems hinge on a comprehensive understanding of the status and trends of top predators. This review delves into the ecological significance of marine top predators, examining their roles in maintaining ecosystem stability and functioning through an integrated analysis of current scientific literature. We first assess the efficacy of various monitoring methods, ranging from traditional field observations to cutting-edge technologies like satellite tracking and environmental DNA (eDNA) analysis and evaluating their strengths and limitations in terms of accuracy, spatial coverage, and cost-effectiveness, providing resource managers with essential insights for informed decision-making. Then, by synthesizing data from diverse marine ecosystems, this study offers a comprehensive overview of the trends affecting top predator populations worldwide. We explore the multifaceted impacts of human activities, climate change, and habitat degradation on the abundance and distribution of these key species. In doing so, we shed light on the broader implications of declining top predator populations, such as trophic cascades and altered community structures. Following a thorough assessment of successful strategies for reversing the decline of top predators, a compilation of recommendations is presented, encompassing effective governance interventions. A crucial aspect of effective ecosystem-based management is the implementation of robust monitoring strategies. Mitigation measures are imperative to reverse the adverse impacts on marine top predators. We present a comprehensive array of mitigation options based on successful case studies. These include the establishment of marine protected areas, the enforcement of fisheries regulations, and the promotion of sustainable fishing practices. We deepen the synergies between these strategies and their potential to mitigate human-induced stressors on top predator populations to safeguard their pivotal role in maintaining marine ecosystem structure and function. By examining marine top predators’ ecological significance, analyzing population trends, discussing monitoring techniques, and outlining effective mitigation strategies, we provide a comprehensive resource for researchers, policymakers, and stakeholders engaged in fostering ecosystem-based management approaches. We conclude that integrating these insights into current management frameworks will be essential to safeguard both top predators and the broader marine environment for future generations.

Marling (limestone alkaline amendment) agrarian practices have a plurimillennial influence on soil pH and on soil-associated ecosystems. Although the earliest written records in Europe date back to antiquity, the origin of this agrarian practice is not well known. In order to trace the evolution of this practice in the early agrarian societies of Western Europe, we searched for topographic anomalies of ancient limestone quarries that may have been used for marling. Four hundred circular closed topographic depressions (CDs) were identified under two areas of ancient forest in northern France with Lidar digital elevation models (DEMs). Soils, morphology and archaeological context were studied and filling sediments were dated in several dozen CDs.The gaps in the carbonate rocks, soils with calcaric properties (scattered limestone fragments), unstratified fills, the absence of buried paleosols, and dates (OSL, C-14) later than the first local agricultural societies made it possible to recognise the CDs that may have served as limestone quarries. This was the case for most of the 30 CDs studied, leaving a minority of natural origin (suffosion, depressions of thermokarstic origins). Amongst the quarries identified, the preferential excavation of soft (friable) limestone and the location of these quarries in fossil agrarian plots led us to hypothesize that they were used for alkaline amendments of the cultivated fields. The quarries were filled from the Middle Neolithic to Late Antiquity, shortly before the period of afforestation of these former agrarian territories.

Atmospheric oxidation capacity (AOC) characterizes the ability of the atmosphere to scavenge air pollutants. However, the processes involved in China, where anthropogenic emissions have changed dramatically in the past decade, are not fully understood. A detailed analysis of different parameters that determine the AOC in China is presented on the basis of numerical simulations performed with the regional chemical–meteorological Weather Research and Forecasting model with Chemistry (WRF-Chem). The model shows that the aerosol effects related to extinction and heterogeneous processes produce a decrease in surface ozone of approximately 8–10 ppbv in NOx-limited rural areas and an increase of 5–10 ppbv in VOC-limited urban areas. In this latter case, the ozone increase is noticeable for aerosol concentrations ranging from 20 to 45 µg m−3 in July 2018. The ozone reduction in NOx-sensitive regions is due to the combined effect of nitrogen dioxide and peroxy radical uptake on particles and of the light extinction by aerosols, which affects the photodissociation rates. The ozone increase in VOC-sensitive areas is attributed to the uptake of NO2 by aerosols, which is offset by the reduced ozone formation associated with HO2 uptake and with aerosol extinction. Our study concludes that more than 90 % of the daytime AOC is due to the reaction of the hydroxyl radical with VOCs and carbon monoxide. In urban areas, during summertime, the main contributions to daytime AOC are the reactions of OH with alkene (30 %–50 %), oxidized volatile organic compounds (OVOCs) (33 %–45 %), and carbon monoxide (20 %–45 %). In rural areas, the largest contribution results from the reaction of OH with alkenes (60 %). Nocturnal AOC is dominantly attributed to the reactions with the nitrate radical (50 %–70 %). Our results shed light on the contribution of aerosol-related NOx loss and the high reactivity of alkenes for photochemical pollution. With the reduction in aerosols and anthropogenic ozone precursors, the chemistry of nitrogen and temperature-sensitive VOCs will become increasingly important. More attention needs to be paid to the role of photodegradable OVOCs and nocturnal oxidants in the formation of secondary pollutants.

Abstract The Arctic environment is transforming rapidly due to climate change. Aerosols’ abundance and physicochemical characteristics play a crucial, yet uncertain, role in these changes due to their influence on the surface energy budget through direct interaction with solar radiation and indirectly via cloud formation. Importantly, Arctic aerosol properties are also changing in response to climate change. Despite their importance, year-round measurements of their characteristics are sparse in the Arctic and often confined to lower latitudes at Arctic land-based stations and/or short high-latitude summertime campaigns. Here, we present unique aerosol microphysics and chemical composition datasets collected during the year-long Multidisciplinary drifting Observatory for the Study of Arctic Climate (MOSAiC) expedition, in the central Arctic. These datasets, which include aerosol particle number concentrations, size distributions, cloud condensation nuclei concentrations, fluorescent aerosol concentrations and properties, and aerosol bulk chemical composition (black carbon, sulfate, nitrate, ammonium, chloride, and organics) will serve to improve our understanding of high-Arctic aerosol processes, with relevance towards improved modelling of the future Arctic (and global) climate.

Abstract Background Freshwater ecosystem degradation and biodiversity decline are strongly associated with intensive agricultural practices. Simultaneously occurring agricultural stressors can interact in complex ways, preventing an accurate prediction of their combined effects on aquatic biota. Here, we address the limited mechanistic understanding of multiple stressor effects of two globally important stressors, an insecticide (chlorantraniliprole), and increased fine sediment load and assessed their impact on the transcriptomic profile of two stream macroinvertebrates: the amphipod Gammarus pulex and the caddisfly Lepidostoma basale. Results We identified mainly antagonistic stressor interactions at the transcriptional level, presumably because the insecticide adsorbed to fine sediment particles. L. basale, which is phylogenetically more closely related to the insecticide’s target taxon Lepidoptera, exhibited strong transcriptional changes when the insecticide stressor was applied, whereas no clear response patterns were observed in the amphipod G. pulex. These differences in species vulnerability can presumably be attributed to molecular mechanisms determining the cellular affinity toward a stressor as well as differential exposure patterns resulting from varying ecological requirements between L. basale and G. pulex. Interestingly, the transcriptional response induced by insecticide exposure in L. basale was not associated with a disruption of the calcium homeostasis, which is the described mode of action for chlorantraniliprole. Instead, immune responses and alterations of the developmental program appear to play a more significant role. Conclusions Our study shows how transcriptomic data can be used to identify multiple stressor effects and to explore the molecular mechanisms underlying stressor-induced physiological responses. As such, stressor effects assessed at the molecular level can inform about modes of action of chemicals and their interplay with non-chemical stressors. We demonstrated that stressor effects vary between different organismic groups and that insecticide effects are not necessarily covered by their described mode of action, which has important implications for environmental risk assessment of insecticides in non-target organisms.

The Portable Ice Nucleation Chamber 2 (PINCii) is a newly developed continuous flow diffusion chamber (CFDC) for measuring ice nucleating particles (INPs). PINCii is a vertically oriented parallel-plate CFDC that has been engineered to improve upon the limitations of previous generations of CFDCs. This work presents a detailed description of the PINCii instrument and the upgrades that make it unique compared with other operational CFDCs. The PINCii design offers several possibilities for improved INP measurements. Notably, a specific icing procedure results in low background particle counts, which demonstrates the potential for PINCii to measure INPs at low concentrations ( L−1). High-spatial-resolution wall-temperature mapping enables the identification of temperature inhomogeneities on the chamber walls. This feature is used to introduce and discuss a new method for analyzing CFDC data based on the most extreme lamina conditions present within the chamber, which represent conditions most likely to trigger ice nucleation. A temperature gradient can be maintained throughout the evaporation section in addition to the main chamber, which enables PINCii to be used to study droplet activation processes or to extend ice crystal growth. A series of both liquid droplet activation and ice nucleation experiments were conducted at temperature and saturation conditions that span the spectrum of PINCii's operational conditions (-50≤ temperature ≤-15 ∘C and 100 ≤ relative humidity with respect to ice ≤160 %) to demonstrate the instrument's capabilities. In addition, typical sources of uncertainty in CFDCs, including particle background, particle loss, and variations in aerosol lamina temperature and relative humidity, are quantified and discussed for PINCii.

Understanding the role of transport and photochemistry is essential to mitigate tropospheric ozone (O3) pollution within a region. In previous studies, the O3 concentration budget has been widely used to determine the contributions of two processes to the variations of O3 concentrations. These studies often conclude that local photochemistry is the main cause of regional O3 pollution; however, they fail to explain why O3 in a targeted region is often primarily derived from O3 and/or its precursors transported from the outside regions, as reported by many studies of O3 source apportionment. Here, we present a method to calculate the hourly contributions of O3-related processes to the variations of not only the mean O3 concentration but also the total O3 mass (the corresponding budgets are noted as the O3 concentration and mass budget, respectively) within the atmospheric boundary layer (ABL) of the concerned region. Based on the modelling results of WRF-CMAQ (Weather Research and Forecasting and Community Multiscale Air Quality), the two O3 budgets were applied to comprehensively understand the effects of transport and photochemistry on the O3 pollution over the Pearl River Delta (PRD) region in China. Quantified results demonstrate the different role of transport and photochemistry when comparing the two O3 budgets: photochemistry drives the rapid increase of O3 concentrations during the day, whereas transport, especially vertical exchange through the ABL top, controls both rapid O3 mass increase in the morning and decrease in the afternoon. The diurnal changes of the transport contributions in the two O3 budgets highlight the influences of the ABL diurnal cycle and regional wind fields on regional O3 pollution. Through high contributions to the O3 mass increase in the morning, transport determines that most O3 in the PRD originates from the global background and emissions outside the region. However, due to the simultaneous rapid increase of ABL volumes, this process only has a relatively limited effect on O3 concentration increase compared to photochemistry, and transport effect on the regional sources of O3 cannot be illustrated by the O3 concentration budget. For future studies targeting O3 and other secondary pollutants with moderately long atmospheric lifetimes (e.g. fine particulate matter and some of its components), insights from both concentration and mass budgets are required to fully understand the role of transport, chemistry and other related processes.

This analysis describes the successes, challenges and opportunities to improve global vaccine safety surveillance as observed by the Vaccine Safety Working Group from its role as a platform of exchange for stakeholders responsible for monitoring the safety of vaccines distributed through the COVAX mechanism. Three key elements considered to be essential for ongoing and future pandemic preparedness for vaccine developers in their interaction with other members of the vaccine safety ecosystem are (1) the availability of infrastructure and capacity for active vaccine safety surveillance in low-income and middle-income countries (LMICs), including the advancement of concepts of safety surveillance and risk management to vaccine developers and manufacturers from LMICs; (2) more comprehensive mechanisms to ensure timely exchange of vaccine safety data and/or knowledge gaps between public health authorities and vaccine developers and manufacturers; and (3) further implementation of the concept of regulatory reliance in pharmacovigilance. These aims would both conserve valuable resources and allow for more equitable access to vaccine safety information and for benefit/risk decision-making.
