Extraction of a GC-amenable hydrocarbon fraction from oil by liquid-liquid diffusion across a laminar interface can be performed in a microfluidic format. Analysis of figures of merit, determined using standard analytical techniques, show this method to be an effective new tool for rapidly processing small quantities of oil and petroleum for GC analysis. Methods based upon similar microsystems devices could find widespread use in a variety of fields, including those associated with organic geochemistry and oil exploration and production, where the manipulation of petroleum constituents (greater than C14) is necessary for analytical purposes.

Raman spectroscopy is a form of vibrational spectroscopy that is well suited to the molecular identification of a variety of analytes, including both explosives and biological agents. The technique has been gaining more widespread interest due to improvements in instrumentation, sensitivity, and its ease of use, in comparison to other techniques. In this paper, we describe recent advances in Raman spectroscopy with respect to the detection of high-energy explosives and biological materials. In particular, emphasis is placed on the exploitation of enhancement factors that overcome traditional limitations on sensitivity, namely, surface enhancement and resonance enhancement, functionalization of target analytes, and the use of novel lab-on-a-chip technology.

SERRS has been used for the first time for the measurement of C-reactive protein (CRP) in an immunoassay. CRP, a biological marker for the diagnosis of infection and inflammation, is quantified in an ELISA using conventional reagents, but the usual colorimetric detection step is replaced by SERRS detection, offering improved sensitivity and potential for multiplexing analysis.

Lab-on-a-chip systems offer a versatile environment in which low numbers of cells and molecules can be manipulated, captured, detected and analysed. We describe here a microfluidic device that allows the isolation, electroporation and lysis of single cells. A431 human epithelial carcinoma cells, expressing a green fluorescent protein-labelled actin, were trapped by dielectrophoresis within an integrated lab-on-a-chip device containing saw-tooth microelectrodes. Using these same trapping electrodes, on-chip electroporation was performed, resulting in cell lysis. Protein release was monitored by confocal fluorescence microscopy.

A simple surface-enhanced Raman spectroscopy (SERS) microflow cell was developed to investigate distributions of scytonemin pigment within cyanobacteria from samples of rock collected from an arctic desert that contained endolithic cyanobacteria. The assay, which has future potential use in a variety of applications, including astrobiology and analysis of microorganisms in remote environments, involved studying SERS spectra of bacteria from within geological samples. By using a dispersed colloidal substrate in the microfluidic device, surface enhancement of the order >105 was obtained for the determination of the pigment in the microorganisms when compared to the native Raman spectra. The SERS assay, which had a nM sensitivity for scytonemin, showed that the concentration of pigment was highest in samples that had experienced the highest stress environments, as a result of high doses of UV irradiation.

This paper demonstrates the use of micron sized beads, modified with fluorescent dyes, as non-invasive sensors to probe the local changes in pH, within a microfluidic channel. To achieve this, amine modified polystyrene spheres (either 3 microm or 6 microm in diameter) were functionalised with the pH sensitive fluorochrome SNARF-1 to produce point sensors. The modified beads were trapped at defined positions close to a pair of integrated planar gold microelectrodes within the channel, using optical tweezers. Both transient and steady-state electrochemical potentials were applied to the microelectrode pair in order to generate changes in the local pH, associated with electrolysis. The functionalised beads indicated the pH changes in the channel, measured as a change in the fluorescence signal, generated by the immobilised pH sensitive dye. Responses were measured with temporal resolutions of between 1 and 200 ms, whilst the spatial resolution of the pH gradients was limited by the size of the beads to 3 microm.

SNARF-1 fluorochrome was used to functionalize 3μm diameter latex spheres making them sensitive to the pH of their environment, manifested as a change in their fluorescence. The fluorescence emission at 580nm was excited using a filtered xenon arc lamp at 515nm. A solution of functionalized latex spheres was placed between gold microelectrodes in a microfluidic channel. Optical tweezers were used to trap and manipulate the spheres in the vicinity of the microelectrodes, to map out the pH profile in the electrolyte solution, induced by passing 20 microsecond transient current pulses through the microelectrodes.

A microfluidic based device has been developed for the continuous separation of polymer microspheres, taking advantage of the flow characteristics of systems. The chip consists of an asymmetric cavity with variable channel width which enables continuous amplification of the particle separation for different size particles within the laminar flow profile. The process has been examined by varying the sample inlet position, the sample to media flow rate ratio, and the total flow rate. This technique can be applied for manipulating both microscale biological and colloidal particles within microfluidic systems.

The first simultaneous detection of three dye-labelled oligonucleotides in a microfluidics chip by SERRS is reported.
