The infamous chytrid fungus Batrachochytrium salamandrivorans (Bsal) recently led to the collapse of European fire salamander populations (Salamandra salamandra) in The Netherlands. Currently, the pathogen has been rapidly expanding its range and threatens salamander populations throughout Europe, including Germany. Here, Bsal is known from wild and captive amphibians and has mostly been reported from the federal state of Northrhine-Westphalia. Due to the geographical proximity of infected areas, its dispersal into neighbouring states is possible. A Bsal taskforce was therefore recently formed in the state of Hesse that aims to implement preparative measures for Bsal mitigation at different levels. Based on the known Bsal susceptibility of salamanders in captivity and their inherent threat potential towards natural populations, an epidemiological screening for Bsal prevalence in private amphibian collections throughout the state of Hesse was conducted. We analysed a total of 174 samples from nine private collections of different urodelan species via qPCR and did not detect Bsal. We discuss our results and their implications for salamander conservation relative to other surveys of this kind and underscore the importance of tight cooperation between private keepers and conservation scientists in order to protect wild amphibians from the lethal Bsal fungus. Investigation into antemortem semen collection methods in reptiles is required to counteract anthropogenic induced reptile biodiversity loss and to further develop assisted reproductive technologies in reptiles. The purpose of this study was to evaluate whether repeated electroejaculation under anesthesia could be done safely in the veiled chameleon (Chamaeleo calyptratus). Electroejaculation under anesthesia (alfaxalone 15 mg/kg intravenously) was performed once weekly over a 4-wk period in 12 Chamaeleo calyptratus. External vent appearance and cloacoscopy was performed prior to and after electroejaculation to assess the effect of the electroejaculation on the external and internal anatomy. Hematological and chemistry values were evaluated prior to the study and afterwards. Semen collection was successful in 50% of the electroejaculation events. Cloacoscopy images scores were significantly higher between the pre- and postelectroejaculation events for each week. No significant difference was observed between study week 1 prior to electroejaculation and study week 4 after electroejaculation in vent external morphology (P = 0.157), and internal cloacal scoring (P = 0.058). A significant increase in the white blood cell counts (P = 0.015), absolute heterophil counts (P = 0.02), absolute monocyte counts (P = 0.003), and albumin (P = 0.038) was observed between the pre- and postejaculation sampling periods. Aspartate aminotransferase (P = 0.001) and potassium (P = 0.036) decreased significantly between the pre- and postsampling periods. Vent image scores were significantly higher (P = 0.046) between the pre- and postsampling images collected during week 1; however, there were no significant differences noted between pre- and postsample images for weeks 2 (P= 0.063), 3(P= 0.083), or 4 (P= 0.083). Serial electroejaculation procedures under alfaxalone anesthesia can be performed safely in veiled chameleons. Although changes were observed in the complete blood count and biochemistry results, these changes were determined to not be clinically significant. The identification of avian Mycoplasma spp. by conventional immunologic, phenotypic, and molecular methods can be demanding and time-consuming. We evaluated MALDI-TOF MS for its suitability to identify avian mycoplasmas at the species level. We generated a mycoplasma spectral database of 36 main spectrum profiles (MSPs) representing 23 avian Mycoplasma spp. using 23 type and reference strains, 1 live vaccine strain, and 8 clinical isolates. We then used 112 avian Mycoplasma clinical isolates of different avian mycoplasmas, 4 Mycoplasma live vaccine strains, and 1 Mycoplasma type strain, previously cultured and identified to the species level by molecular methods, to evaluate the MSP database. Protein extraction and MALDI-TOF MS analysis were performed with a maximum of 3 repetitions per isolate. MALDI-TOF MS resulted in accurate species-level identification with a score of ≥2.0 for 112 of 117 (96%) isolates. The MALDI-TOF MS analysis of 4 of 5 isolates that did not yield a score of ≥2.0 resulted in best-match identifications that were still concordant at species level with the molecular method used for previous identification. An indirect immunofluorescence serologic assay, PCR assay, and histopathology were used to screen for psittaciform orthobornaviruses (PaBV) in wild Cacatuidae in Victoria, Australia. Anti-PaBV antibodies were detected, but PCR and histopathology did not detect PaBV. This study presents the first evidence of PaBV in wild birds in Australia. [ABSTRACT FROM AUTHOR]

Abstract Objective Studies using the estrogen receptor alpha (ERα) knock-out (αERKO) mice have demonstrated that ERα plays a crucial role in various estrogen-mediated metabolic regulations. ERα is a ligand dependent transcription regulator and its activity is regulated by estrogenic compounds. ERα consists of two transcriptional activation domains, AF-1 and AF-2. The activities of these domains are regulated through different mechanisms; however, the specific physiological role in metabolic regulation by these domains is still unclear. Methods We utilized an ERα AF-2 mutant knock-in mouse (AF2ERKI) to evaluate the physiological functionality of ERα transactivation domains. Due to the estrogen insensitive AF-2 mutation, the phenotypes of AF2ERKI mice are seemingly identical to the global αERKO including obesity in the females. Distinct from the αERKO, the AF-1 function of AF2ERKI mice can be activated by tamoxifen (Tam). Ovariectomized (OVX) AF2ERKI and WT females were treated with Tam and fed a high-fat diet (HFD) for 10 weeks. Additionally, indirect calorimetric analysis was performed using metabolic chambers with food intake and locomotor activity recorded for Tam-treated AF2ERKI and αERKO females. Results Obesity in HFD-fed AF2ERKI females was prevented by Tam treatment; particularly, inguinal fat accumulation was strongly blocked by Tam treatment. Alterations in fat metabolism genes, however, were not found in either inguinal fat nor visceral fat to be Tam-regulated, even though fat accumulation was strongly reduced by Tam treatment. Indirect calorimetric analysis revealed that without alteration of food intake and locomotor activity Tam treatment increased energy expenditure in AF2ERKI but not αERKO females. Conclusions These results suggest that the activation of ERα AF-1 prevents fat accumulation. The prevention of obesity through AF-1 is mediated by induction of energy expenditure rather than ERα AF-1 functionality of lipid metabolism gene regulation in fat tissues. Between June and November 2015, 25 woodpeckers (Picidae) with neurologic signs or unknown cause of death were admitted to a veterinary clinic. Alive birds were clinically examined. Birds that were found dead or died despite intensive care treatment were forwarded to a pathologic examination. Necropsy and subsequent tests included screening for several infectious agents and toxins. Three birds tested positive for Sarcocystis calchasi. Toxoplasma gondii was detected in one bird demonstrating intracerebral cysts. Mycoplasma gypis was detected in one woodpecker in the absence of respiratory signs. Several microbial pathogens (eg, Aspergillus fumigatus, Clostridium perfringens, and Escherichia coli) were isolated from single individuals. However, there was no consistent finding in all birds that could explain nervous signs and mortality of the woodpeckers examined. To the authors' knowledge, M. gypis and S. calchasi were detected in a woodpecker for the first time in this study. [ABSTRACT FROM AUTHOR]

Several Mycoplasma spp. are well-known pathogens in poultry. In birds of prey, White Storks ( Ciconia ciconia), and some waterfowl (Anatidae, Pelecanidae) species, mycoplasmas occur commonly and seem to be apathogenic or commensal and most likely belong to the physiologic microbial flora of the respiratory tract. In other bird species, such as Common Nightingales ( Luscinia megarhynchos) and tits (Paridae), Mycoplasma spp. are absent in healthy birds. In corvids, the prevalence and role of Mycoplasma spp. in disease remains unclear. In previous studies, Mycoplasma sturni was detected in diseased corvids; however, those studies included only a limited sample size or preselected individuals. We collected tracheal swabs of 97 free-ranging Corvidae, including 68 randomly selected individuals from hunting bags and 29 birds that had been admitted to a veterinary clinic. Tracheal swabs were examined for Mycoplasma spp. using culture and genus-specific PCR. If Mycoplasma spp. were detected, the species were identified by sequencing the 16S ribosomal (r) RNA gene and 16-23S rRNA intergenic transcribed spacer region. Five of 68 (7%) of the hunted birds and nine of 29 (31%) of the birds admitted to the veterinary clinic were PCR positive. In 13 of 14 PCR-positive samples, mycoplasmas were cultured and M. sturni was the only mycoplasmal species identified. None of the positive corvids from the hunting bags had clinical signs, whereas five of nine birds admitted to the veterinary clinic showed apathy, lameness, injuries, or fractures, which may not be associated with mycoplasmal infections. These data support the notion that M. sturni is the Mycoplasma sp. most frequently found in corvids, though its prevalence and ability to cause disease may involve interaction with other aspects of bird health.
