1. Research Note: Oxidative stress and immune response following the administration of live attenuated Mycoplasma gallisepticum vaccination in backyard chicken

Abstract

ABSTRACT: In the present study, we investigated a possible relationship between the immune response and the oxidative stress (OS) state trend in a group of 12 chickens after intraocular administration of an attenuated Mycoplasma gallisepticum (MG) vaccine. Blood samples were collected at the vaccination time (T0), after 14 (T1) and 21 d (T2). White blood cell count (WBC), differential leucocyte count, and anti-MG antibodies titer (S/P) were studied as immune response indexes. As plasmatic OS biomarkers levels, we considered malondialdehyde (MDA), nitric oxide (NO), superoxide dismutase (SOD), reactive oxygen metabolites derived compounds (d-ROMs), the ferric reducing ability of plasma (FRAP), and superoxide anion (O2−). After antigenic stimulation, it was observed a significant decrease in monocythemia and a significant increase in thrombocythemia, S/P, MDA, and SOD. Furthermore, subjects with high d-ROMs levels at T0 tended to develop higher cellular mobilization with increases in WBC and lymphocytes accompanied by lower antibody release. It was also observed that the antioxidant components FRAP and SOD were moderately positively correlated to the entity of antibody response.

6. Twisted cohomology of arrangements of lines and Milnor fibers

Abstract

Let $\A$ be an arrangement of affine lines in $\C^2,$ with complement $\M(\A).$ The (co)homo-logy of $\M(\A)$ with twisted coefficients is strictly related to the cohomology of the Milnor fibre associated to the conified arrangement, endowed with the geometric monodromy. Although several partial results are known, even the first Betti number of the Milnor fiber is not understood. We give here a vanishing conjecture for the first homology, which is of a different nature with respect to the known results. Let $\Gamma$ be the graph of \emph{double points} of $\A:$ we conjecture that if $\Gamma$ is connected then the geometric monodromy acts trivially on the first homology of the Milnor fiber (so the first Betti number is combinatorially determined in this case). This conjecture depends only on the combinatorics of $\A.$ We prove it in some cases with stronger hypotheses. In the final parts, we introduce a new description in terms of the group given by the quotient ot the commutator subgroup of $\pi_1(\M(\A))$ by the commutator of its \emph{length zero subgroup.} We use that to deduce some new interesting cases of a-monodromicity, including a proof of the conjecture under some extra conditions., Comment: 2 m pages, 7 figures

Abstract

Given a subspace arrangement, there are several De Concini-Procesi models associated to it, depending on distinct sets of initial combinatorial data (building sets). The first goal of this paper is to describe, for the root arrangements of types A_n, B_n (=C_n), D_n, the poset of all the building sets which are invariant with respect to the Weyl group action, and therefore to classify all the wonderful models which are obtained by adding to the complement of the arrangement an equivariant divisor. Then we point out, for every fixed n, a family of models which includes the minimal model and the maximal model; we call these models `regular models' and we compute, in the complex case, their Poincar\'e polynomials., Comment: 27 pages, 6 figures

Abstract

We have measured the transverse field muon spin precession in the flux lattice (FL) state of the two gap superconductor MgB2 and of the electron doped compounds Mg1-xAlxB2 in magnetic fields up to 2.8T. We show the effect of the two gaps on the internal field distribution in the FL, from which we determine two coherence length parameters and the doping dependence of the London penetration depth. This is an independent determination of the complex vortex structure already suggested by the STM observation of large vortices in a MgB2 single crystal. Our data agrees quantitatively with STM and we thus validate a new phenomenological model for the internal fields., Comment: now in press Phys. Rev. Lett., small modifications required by the editor

41. Generic Behavioural Criteria of Managerial Effectiveness: An Empirical and Comparative Case Study of UK Local Government

Abstract

Purpose: The purpose of this paper is to present the findings of a "partnership-research" study of effective and ineffective managerial behaviour within the "local government" setting of the Wolverhampton City Council Social Care Department, and to describe how the research supports and challenges the organisation's existing "leadership and management behavioural competency framework". Additionally, it reveals and discusses the extent to which the results are consistent with equivalent and comparable findings from an equivalent study within a "central government" department. Design/methodology/approach: Concrete examples of effective and ineffective managerial behaviour were collected using the Critical Incident Technique (CIT) of Flanagan, and the obtained data were analysed using content and thematic analytic methods. Findings: The paper finds that from a total of 218 usable critical incidents 50 discrete behavioural items were identified, of which 25 were examples of "effective" and 25 of "ineffective" behaviour. A comparison against equivalent findings from the "central government" study revealed high degrees of overlap with 92 per cent of the "effective" and 96 per cent of the "ineffective" behavioural items being the same as, similar to, or containing some congruence of meaning. Research limitations/implications: Although the number of CIT informants (n = 40) falls at the top end of the typical sample range for qualitative research, it is possible that data collection "saturation" has not been reached. Whereas the subject of the present "local government" study was first line and middle managers, the focus of the compared "central government" study also included senior managers. Originality/value: The results of this replica research lend additional empirical support to those who believe in "generic" and "universalistic" explanations of managerial and leadership effectiveness. Abstract

Plastic is one of the main sources of marine and terrestrial pollution. This material can fragment into micro- (-5 mm) and nanoplastics (NPs) (100 nm) following degradation. Animals are exposed to these particles by ingesting contaminated food, respiration or filtration, and transdermally. In organisms, NPs can cross biological membranes, and cause oxidative stress, cell damage, apoptosis, and endocrine interference. We previously demonstrated that polystyrene - NPs interfered with ovarian cell functions. Since reproduction involves a high energy expenditure and a crucial role is played by adipose tissue, the aim of the present study was to evaluate the effects of NPs on primary adipose stromal cells (ASCs) isolated from swine adipose tissues. In particular, the effects on cell viability, proliferation, metabolic activity, inflammatory process mediators and oxidative stress markers were assessed. The obtained results did not reveal a significant variation in cell proliferation, metabolic activity was increased (P0.01) but only at the lowest concentration, while viability showed a significant decrease after prolonged exposure to NPs (P0.01). Abstract

Background Combination doxycycline/macrocyclic lactone (ML) protocols have been shown to provide a more rapid adulticidal and microfilaricidal effect than either MLs or doxycycline alone, although female worms were reported to have a higher tolerance to treatments compared to male worms. The present study aimed to evaluate how ABC transporters may be involved in the synergic effect of the combination treatment. Adult worms of D. immitis were treated in vitro for 24 hours with doxycycline (DOXY), ivermectin (IVM) and a combination of both, and changes in the modulation of ABC transporter genes were measured. Levels of doxycycline inside different treatment media, post-treatment, were determined through HPLC analysis. Results Quantitative RT-PCR analysis showed the presence of changes in the modulation of ABC transporter genes evaluated in this study. In particular, in female worms, the combination treatment induced a substantial increase in gene expressions, especially of Dim-pgp-10 and Dim-haf-4; whereas in male worms, the greatest increase in gene expression was observed for Dim-pgp-10 and Dim-pgp-11 when treated with DMSO + IVM and DMSO + DOXY/IVM. HPLC analysis of the DOXY concentrations in the media after in vitro treatments of male worms showed a slight difference between the DMSO + DOXY samples and the combination (DMSO + DOXY + IVM), while no difference was observed among females. Conclusions Further studies are required to explain whether the modulation of cellular efflux plays a role, even partially, in the adulticide effect of doxycycline/macrocyclic lactone combinations in heartworm-infected dogs. To the authors’ knowledge, this is the first study to evaluate P-gp expression in adult D. immitis.

Abstract

A physiological equilibrium exists between pro- and antioxidant factors. When the oxidant factors exceed the capacity of their removal or inactivation, oxidative stress (OS) occurs. The OS levels were assayed in plasma obtained from 2 bird species. Blood samples were collected from 20 healthy domestic chicken hens, 10 living in an intensive farming environment and 10 free-range, and from 18 healthy Eurasian magpies (Pica pica; 7 females and 11 males, with an estimated age of >1 year of age). For OS biomarker assessment, the determinable reactive oxygen metabolites (d-ROMs) were measured, and the plasmatic antioxidant test (PAT) was performed; the OS index (OSI) was then calculated (d-ROMs/PAT × 1000) as a parameter of overall oxidative stress. Moreover, lipid peroxidation was assessed by measuring plasmatic malondialdehyde (MDA) levels. A hematological evaluation was also performed on each bird with a hemocytometer, on which a blood sample was placed to obtain both a total and differential white blood cell (WBC) count. In hens, OSI and MDA levels were significantly higher (P = .04, and P = .004) in subjects from intensive farming (14.7 ± 7.1 and 27.2 ± 10.4 nmol/mL) than in those bred in rural conditions (5.6 ± 10.3 and 8.2 ± 13.3 nmol/mL). In magpies, a positive correlation between the total WBC count and OS was found, and both d-ROMs and OSI were significantly higher (P = .03) in subjects with a total WBC count greater than the median value (20.4 × 103 cells/µL) with respect to those with a total WBC count less than the median value. The results generated from this study indicate that higher OS levels occurred in hens bred in an intensive indoor farm environment compared with outdoor free-range conditions. Possibly the higher OS levels could be related to the higher stocking density and dust levels found in the indoor facility. Additionally, the correlation between OS biomarker levels in magpies and total WBC count suggests that OS level is influenced by immune response, in agreement with previous studies. Collectively, present data seem to be promising for the application of OS measurement in avian medicine for health and animal welfare monitoring.

Abstract

In the present study, the Eurasian magpie (Pica pica), was evaluated as a possible bioindicator of environmental pollution by heavy metals (HMs). Levels of Ni, Pb, Cd, and Hg in feathers of 64 magpies (31 males and 33 females) were measured by ICP-MS technique. Plasmatic biomarkers of oxidative stress (OS) were also assessed. The birds were captured in the province of Parma (Italy), in different capture sites within 1 km from urban area (UZ), and farther than 5 km from urban area (RZ). Median HM levels were 0.68 mg/kg (0.18–2.27), 2.80 mg/kg (0.41–17.7), &lt, limit of detection (LOD) mg/kg (&lt, LOD–0.25), 3.90 mg/kg (1.35–85.9) for Ni, Pb, Cd and Hg, respectively. No significant differences in HM levels were found according to sex, while Ni and Pb were significantly higher in adult compared to young birds (p = 0.047, p = 0.004). Conversely, Cd and Hg levels in young magpies resulted higher than those of adults (p = 0.001 and p = 0.004). No correlation was found between OS biomarkers and HM levels. No differences were found in HM levels according to capture area, except for Hg level, which resulted higher in magpies of RZ (4.05 mg/kg (1.35–12.7)) compared to UZ (2.99 mg/kg (1.54–85.9)). Further experiments are needed to establish whether magpie feathers could represent a suitable non-invasive tool for biomonitoring HMs in the environment.
