Purpose: Radiolabeled graphene oxide (GO) nanosheets has been one of the most extensively studied nanoplatform for in vivo radioisotope delivery. Herein, we describe the functionalization of the surface of GO nanosheets with Fe3O4 magnetic nanoparticles, cysteine amino acid as an interface ligand, and cadmium telluride quantum dots. Materials and Methods: To enable In vivo PET imaging, the GO@Fe3O4-cys-CdTe QDs were labeled with 68Ga to yield [68Ga] Ga-Go@ Fe3O4-Cys-CdTe QDs. Furthermore, serum stability tests were performed and the biological behavior of the nanocomposite was evaluated in rats bearing fibrosarcoma tumor. Results: Liver, blood and tumor were the most accumulated sites at 1 h after the injection, and the radiolabeled nanocomposite as a PET/MRI imaging agent showed fast depletion from body and acceptable tumor uptake. Conclusion: Magnetic (Fe3O4) and fluorescent components (CdTe QDs) along with a positron-emitting radionuclide will help to design a multimodal imaging system (PET/MRI/OI) which will offer the advantages of combined imaging techniques and further possible used in localized radionuclide therapy. Stroke is a devastating disease. Endovascular mechanical thrombectomy is dramatically changing the management of acute ischemic stroke, raising new challenges regarding brain outcome and opening up new avenues for brain protection. In this context, relevant experiment models are required for testing new therapies and addressing important questions about infarct progression despite successful recanalization, reversibility of ischemic lesions, blood–brain barrier disruption and reperfusion damage. Here, we developed a minimally invasive non-human primate model of cerebral ischemia (Macaca fascicularis) based on an endovascular transient occlusion and recanalization of the middle cerebral artery (MCA). We evaluated per-occlusion and post-recanalization impairment on PET-MRI, in addition to acute and chronic neuro-functional assessment. Voxel-based analyses between per-occlusion PET-MRI and day-7 MRI showed two different patterns of lesion evolution: "symptomatic salvaged tissue" (SST) and "asymptomatic infarcted tissue" (AIT). Extended SST was present in all cases. AIT, remote from the area at risk, represented 45% of the final lesion. This model also expresses both worsening of fine motor skills and dysexecutive behavior over the chronic post-stroke period, a result in agreement with cortical-subcortical lesions. We thus fully characterized an original translational model of ischemia–reperfusion damage after stroke, with consistent ischemia time, and thrombus retrieval for effective recanalization. Abstract Background Meaningful translational large animal models for cardiac diseases are indispensable for studying disease mechanisms, development of novel therapeutic strategies, and evaluation of potential drugs. Methods For induction of heart failure, cardiac hypertrophy and fibrosis, a bare metal stent was implanted in the descending aorta of growing pigs (n = 7), inducing pressure stress on the left ventricle (group HYPI). The constant stent size in growing pigs resulted in antegrade partial obstruction of the aortic flow with a gradual increase in afterload. Five pigs with sham intervention served as control. Serial haemodynamic, pressure–volume loop measurements and transthoracic echocardiography (TTE) were performed to detect developing pressure overload of the LV and cardiac MRI with late enhancement for measuring LV and RV mass and ejection fraction. Results At 5-month follow-up, CT and contrast aortography, and intraluminal echocardiography confirmed aortic isthmus stenosis with a mean trans-stenotic gradient of 64 ± 13.9 mmHg. In this study, magnetic multimodal nanoparticles with potential applications in magnetic- and nuclear-medicine imaging, magnetic resonance imaging, hyperthermia, and theranostic (therapeutic and diagnostic), applications were prepared by coating iron oxide nanoparticles with silica (core-shell), functionalizing with aminopropyltriethoxy silane and coupling with diethylenetriamine pentaacetic acid ligand (DTPA). Radiolabeling of core-shell-DTPA particles with 68Ga radiometal was carried out through chelation of 68Ga(III) ions by DTPA and was used for positron emission tomography. The biodistribution of the 68Ga-radiolabeled magnetic nanoparticles compared to free 68Ga(III) was checked in normal Balb/c mice up to 2 h. [ABSTRACT FROM AUTHOR]

We assessed glucose uptake in different tissues in type 2 diabetes (T2D), prediabetes, and control subjects to elucidate its impact in the development of whole-body insulin resistance and T2D. Thirteen T2D, 12 prediabetes, and 10 control subjects, matched for age and BMI, underwent OGTT and abdominal subcutaneous adipose tissue (SAT) biopsies. Integrated whole-body 18F-FDG PET and MRI were performed during a hyperinsulinemic euglycemic clamp to asses glucose uptake rate (MRglu) in several tissues. MRglu in skeletal muscle, SAT, visceral adipose tissue (VAT), and liver was significantly reduced in T2D subjects and correlated positively with M-values (r = 0.884, r = 0.574, r = 0.707 and r = 0.403, respectively). Brain MRglu was significantly higher in T2D and prediabetes subjects and had a significant inverse correlation with M-values (r = -0.616). Myocardial MRglu did not differ between groups and did not correlate with the M-values. A multivariate model including skeletal muscle, brain and VAT MRglu best predicted the M-values (adjusted r² = 0.85). In addition, SAT MRglu correlated with SAT glucose uptake ex vivo (r = 0.491). In different stages of the development of T2D, glucose uptake during hyperinsulinemia is elevated in the brain in parallel with an impairment in peripheral organs. Impaired glucose uptake in skeletal muscle and VAT together with elevated glucose uptake in brain were independently associated with whole-body insulin resistance, and these tissue-specific alterations may contribute to T2D development. TTE and ex vivo analyses confirmed severe concentric LV hypertrophy (mean circumferential wall thickness, 19.4 ± 3.1, n = 7 vs 11.4 ± 1.0 mm, n = 5, HYPI vs controls, p < 0.05). The LV and RV mass increased significantly, paralleled by increased isovolumic relaxation constant (tau). Histological analyses confirmed substantial fibrosis and myocyte hypertrophy in both LV and RV. Expressions of ANP, BNP, and miRNA-29a were up-regulated, while SERCA2a and miRNA-1 were down-regulated. Plasma NGAL levels increased gradually, while the elevation of NT-proBNP was detected only at the 5-month FUP.Conclusion: These data prove that percutaneous artificial aortic stenosis in pigs is useful for inducing clinically relevant progredient heart failure based on myocardial hypertrophy and fibrosis.
