Self-incompatibility (SI) is a genetic mechanism to prevent self-fertilization and thereby promote outcrossing in hermaphroditic plant species through discrimination of self and nonself-pollen by pistils. In many SI systems, recognition between pollen and pistils is controlled by a single multiallelic locus (called the S -locus), in which multiple alleles (called S -alleles) are segregating. Because of the extreme level of polymorphism of the S -locus, identification of S -alleles has been a major issue in many SI studies for decades. Here, we report an RNA-seq–based method to explore allelic diversity of the S -locus by employing the long-read sequencing technology of the Oxford Nanopore MinION and applied it for the gametophytic SI system of Petunia (Solanaceae), in which the female determinant is a secreted ribonuclease called S-RNase that inhibits the elongation of self-pollen tubes by degrading RNA. We developed a method to identify S -alleles by the search of S-RNase sequences, using the previously reported sequences as queries, and found in total 62 types of S-RNase including 45 novel types. We validated this method through Sanger sequencing and crossing experiments, confirming the sequencing accuracy and SI phenotypes corresponding to genotypes. Then, using the obtained sequence data together with polymerase chain reaction–based genotyping in a larger sample set of 187 plants, we investigated the diversity, frequency, and the level of shared polymorphism of S -alleles across populations and species. Iron (Fe) is an essential micronutrient for plants. Due to the low Fe bioavailability in cultivated soils, Fe deficiency is a widespread agricultural problem. In this study, we present the functional characterization of a petunia (Petunia hybrida) basic-helix-loop-helix transcription factor PhbHLH121 in response to Fe shortage. Real-time PCR revealed that the expression of PhbHLH121 in petunia roots and shoots was downregulated under Fe-limited conditions. CRISPR/Cas9-edited phbhlh121 mutant plants were generated to investigate the functions of PhbHLH121 in petunia. Loss-of-function of PhbHLH121 enhanced petunia tolerance to Fe deficiency. Further investigations revealed that the expression level of several structural genes involved in Fe uptake in petunia, such as IRT1 and FRO2, was higher in phbhlh121 mutants compared to that in wild-type under Fe-limited conditions, and the expression level of several genes involved in Fe storage and Fe transport, such as VTL2, FERs and ZIF1, was lower in phbhlh121 mutants compared to that in wild-type under Fe-deficient conditions. Yeast one-hybrid assays revealed that PhbHLH121 binds to the G-box element in the promoter of genes involved in Fe homeostasis. Yeast two-hybrid assays revealed that PhbHLH121 interacts with petunia bHLH IVc proteins. Taken together, PhbHLH121 plays an important role in the Fe deficiency response in petunia. ABSTRACT Low‐temperature stress limits plant growth, and reduces aesthetics of many ornamental plants. Plants have developed different adaptive mechanisms to cope with low‐temperature stress, in which NAC transcription factor family members playing an important role in low‐temperature tolerance. However, their roles in petunia in response to low temperature are still largely unknown. Here, we found that a NAC transcription factor, namely, PhNH10, negatively regulates low‐temperature response in petunia. PhNH10‐silenced and ‐CRISPR/Cas9 mutant plants displayed higher survival rate, anthocyanin content and abscisic acid concentration than PhNH10‐overexpression and wild‐type plants under low‐temperature condition. PhNH10 can directly bind to the PhABA8ox promoter to active its expression, which further promotes the abscisic acid catabolism, while silencing of PhABA8ox increased the ABA concentration and low‐temperature tolerance. In addition, PhNH10 interact with a low‐temperature‐related E2 ubiquitin‐conjugating enzyme, PhUBC2‐1, which in turn inhibited the binding capacity of PhNH10 on PhABA8ox promoter. Our research has elucidated an extensive mechanistic network underlying the PhNH10‐mediated regulation of low‐temperature response in petunia. This finding not only presents a new viewpoint in understanding the low‐temperature tolerance mechanisms but also delineates a promising pathway for transgenic petunia with improved low‐temperature resistance. The ARF gene family plays a vital role in regulating multiple aspects of plant growth and development. However, detailed research on the role of the ARF family in regulating flower development in petunia and other plants remains limited. This study investigates the distinct roles of PhARF5 and PhARF19a in Petunia hybrida flower development. Phylogenetic analysis identified 29 PhARFs, which were grouped into four clades. VIGS-mediated silencing of PhARF5 and PhARF19a led to notable phenotypic changes, highlighting their non-redundant functions. PhARF5 silencing resulted in reduced petal number and limb abnormalities, while PhARF19a silencing disrupted corolla tube formation and orientation. Both genes showed high expression in the roots, leaves, and corollas, with nuclear localization. The transcriptomic analysis revealed significant overlaps in DEGs between PhARF5 and PhARF19a silencing, indicating shared pathways in hormone metabolism, signal transduction, and stress responses. Phytohormone analysis confirmed their broad impact on phytohormone biosynthesis, suggesting involvement in complex feedback mechanisms. Silencing PhARF5 and PhARF19a led to differential transcription of numerous genes related to hormone signaling pathways beyond auxin signaling, indicating their direct or indirect crosstalk with other phytohormones. However, significant differences in the regulation of these signaling pathways were observed between PhARF5 and PhARF19a. These findings reveal the roles of ARF genes in regulating petunia flower development, as well as the phylogenetic distribution of the PhARFs involved in this process. In this work, we assessed the effects of increasing ozone (O3) on four petunia varieties with different floral pigmentation (pink, red, rose-red, and white). Plants were exposed, in open-top chambers located in China, to three O3 concentrations, i.e., (i) ambient air (AA), (ii) AA + 60 ppb O3 (AA + 60), and (iii) AA + 120 ppb O3 (AA + 120), for 85 days (9 h day−1). Flower diameter and duration were assessed, together with leaf chlorophyll and flavonoid contents. White petunia showed a reduced flower diameter and longevity under AA + 60 (−7 and −6%, respectively, in comparison to AA), whereas pink and red petunias only showed this under AA + 120 (−8 and −7%, on average, respectively). Chlorophyll loss occurred in all varieties under AA + 60 (−30%, on average), and at AA + 120 in white and red petunias (−54%, on average). The total flavonoid content in the pink and white varieties increased only under AA + 120 (around +85%), while it grew at both AA + 60 and AA + 120 (+92% and two-fold higher, respectively) in the red variety. Increasing O3 concentrations did not affect particularly the red-rose variety. The white variety showed the strongest correlations among flower and leaf properties, confirming a variety-related O3 response, as well as demonstrating that it had the highest O3 sensitivity. Petunia hybrida is an economically important ornamental plant species. Branching in ornamental plants is closely associated with their ornamental traits, and branching is a significant agronomic trait in petunia, which shapes plant architecture and production cost. Although there are few studies regarding the involvement of the SPEAR genes in lateral branch development, they are known to regulate the development of plant organs. The PhSPEAR1 gene in petunia, an ortholog of Arabidopsis SPEAR1, was isolated for study. According to the results of real-time quantitative PCR (qRT-PCR), PhSPEAR1 was primarily expressed in the roots. The fluorescence signal indicated that PhSPEAR1 was localized in the nucleus and cytoplasm. An increase in PhSPEAR1 expression was induced by cytokinin or decapitation. Overexpression of PhSPEAR1 increased lateral branches in Arabidopsis. Based on our findings, PhSPEAR1 participates significantly in the regulation of branch number in petunia. [ABSTRACT FROM AUTHOR]

This study investigated the effects of substrates composed of various ratios of wood fiber and peat (0, 25, 50, 75, and 100% peat (v/v)) mixed with different amounts of lime (0, 2, 4, 6, and 8 g L−1) and start fertilizer (0, 2, and 4 g L−1 Multimix) on the growth and biomass accumulation of petunia (Petunia x hybrida Vilm 'Finity F1 Purple') and basil (Ocimum basilicum L. 'Marian') in an ebb-and-flow greenhouse system. Growth parameters included plant height, weight, canopy diameter, and chlorosis symptoms for petunia, along with substrate pH and EC measurements. Petunia showed optimal growth in substrates with higher peat content, while basil produced satisfactory biomass across a pH range of 5–7 regardless of substrate type. Optimal petunia cultivation in 100% wood fiber required a significant dose of start fertilizer without lime. Monitoring pH and EC using pour-through and press methods revealed a pH decrease in substrates with added start fertilizer, while substrates with higher wood fiber content were less acidic. Substrates with over 50% (v/v) wood fiber without lime showed a rapid pH increase over five weeks. The pour-through method generally underestimated EC values compared to the press method. Petunia hybrida, widely grown as a bedding plant, has reduced growth and flower quality at temperatures above 30 °C (heat stress), primarily due to heat stress‐induced ethylene (ET) production. The gene acdS encodes the 1‐aminocyclopropane‐1‐carboxylate (ACC) deaminase (ACCD) enzyme, which is known for its role in reducing ET production by breaking down the ET precursor, ACC, in plant tissues.This study investigated the impact of heat stress on both 'Mirage Rose' WT petunia and its acdS‐overexpressing transgenic lines.Heat stress‐induced growth inhibition was observed in WT plants but not in transgenic plants. The increased stress tolerance of transgenic plants over WT plants was associated with lower ET production, ROS accumulation, higher SPAD values, water content, and relative water content. Furthermore, higher sensitivity of the WT to heat stress than the transgenic plants was confirmed by analysing ET signalling genes, heat shock transcription factor genes, and antioxidant‐ and proline‐related genes, more strongly induced in WT than in transgenic plants.Overall, this study suggests the potential application of acdS overexpression in other floriculture plants as a viable strategy for developing heat stress‐tolerant varieties. Vein-associated pigmentation (venation) is a type of floral coloration adopted by plants to attract pollinators. Several petunia (Petunia hybrida) lines generate dorsoventrally asymmetric venation patterning of the corolla tube, in which venation is only present in the dorsal tube. The molecular mechanism underlying this trait is unknown. Here, we demonstrate that miR319 is preferentially expressed in the dorsal corolla tube, leading to dorsoventrally asymmetric expression of its target genes. Transgenic lines overexpressing phy-miR319a generated uniform venation patterning of the corolla tube. Knockout of TCP genes targeted by miR319 promoted venation patterning in the ventral and dorsal tube, while overexpression of the miR319 target gene, PhTCP6 , completely inhibited corolla tube venation patterning. In addition, miR319 -targeted TCPs negatively regulated venation patterning, probably by repressing the regulator of venation patterning, AN4. Together, our data demonstrate that asymmetric expression of miR319 promotes venation patterning in the petunia dorsal tube alone by repressing the expression of its target TCP genes, which negatively regulate corolla tube venation patterning. When natural days are short, photoperiodic lighting at the end or beginning of the day (day extension) or in the middle of the night (night break) promotes flowering of long-day plants. The objective of this study was to compare broad-spectrum warm-white light-emitting diodes (LEDs) and red (R) + far-red (FR) LEDs at flowering regulation when delivered at different timings in the night period. We performed a greenhouse experiment on four long-day ornamentals [coreopsis (Coreopsis grandiflora) 'Early Sunrise', snapdragon (Antirrhinum majus) 'Liberty Classic Yellow', petunia (Petunia ×hybrida) 'Easy Wave Burgundy Star', and petunia 'Wave Purple Improved']. We grew plants under a truncated 8-hour photoperiod with or without low-intensity (~2 lmol·m22·s21) nighttime lighting from warm-white or R+FR LEDs. For each light quality, we delivered four timings: 1) 8 hours after dusk; 2) 8 hours before dawn; 3) 4 hours after dusk + 4 hours before dawn; and 4) 4-hour night break. The effectiveness of floral promotion was determined by time from the treatment onset to the first open flower. Coreopsis flowered similarly under all lighting treatments, irrespective of light quality and timing, but did not flower under the short-day treatment by the end of the experiment. At flowering, coreopsis was 18% to 19% shorter under white than R+FR LEDs. In contrast, snapdragon flowered 9 to 20 days later under white than R+FR LEDs, when delivered for 8 hours at night, but flowered similarly under these two lamp types as a 4-hour night break. Compared with the shortday treatment, white and R+FR LEDs promoted flowering of both petunia cultivars, although flowering generally occurred later under white than R+FR LEDs. Snapdragon and petunia 'Easy Wave Burgundy Star' developed 30% to 122% more lateral branches under white than R+FR LEDs, when delivered for 8 hours at night. The effectiveness of warm-white LEDs was generally unaffected by timing, although it was most promotive of flowering in snapdragon when delivered for 8 hours before dawn. For R+FR LEDs, 8-hour day-extension lighting was generally more effective than 4-hour night-break lighting, irrespective of timing. We conclude when delivered for 8 hours at night, warm-white LEDs are generally less effective than R+FR LEDs at promoting flowering of long-day ornamentals but similarly effective as 4-hour night-break lighting. The effectiveness of day-extension lighting is generally independent of timing, although for R+FR LEDs, 8 hours after-dusk and/or before-dawn lighting was generally more effective than 4-hour night-break lighting. Self‐incompatibility (SI) is an intraspecific reproductive barrier widely present in angiosperms. The SI system with the broadest occurrence in angiosperms is based on an S‐RNase linked to a cluster of multiple S‐locus F‐box (SLF) genes found in the Solanaceae, Plantaginaceae, Rosaceae, and Rutaceae. Recent studies reveal that non‐self S‐RNase is degraded by the Skip Cullin F‐box (SCF)SLF‐mediated ubiquitin–proteasome system in a collaborative manner in Petunia, but how self‐RNase functions largely remains mysterious. Here, we show that S‐RNases form S‐RNase condensates (SRCs) in the self‐pollen tube cytoplasm through phase separation and the disruption of SRC formation breaks SI in self‐incompatible Petunia hybrida. We further find that the pistil SI factors of a small asparagine‐rich protein HT‐B and thioredoxin h together with a reduced state of the pollen tube all promote the expansion of SRCs, which then sequester several actin‐binding proteins, including the actin polymerization factor PhABRACL, the actin polymerization activity of which is reduced by S‐RNase in vitro. Meanwhile, we find that S‐RNase variants lacking condensation ability fail to recruit PhABRACL and are unable to induce actin foci formation required for pollen tube growth inhibition. Taken together, our results demonstrate that phase separation of S‐RNase promotes SI response in P. hybrida, revealing a new mode of S‐RNase action. [ABSTRACT FROM AUTHOR]

Drought is widely recognized as the most severe environmental danger to horticulture in almost all regions across the globe. The objective of this study was to determine if applying a foliar treatment of salicylic acid (SA) at 400 ppm could mitigate the negative effects of drought on Petunia grandiflora. Four experimental variants were implemented for this purpose, with regular watering or moderate drought conditions. These variants were observed for duration of 14 days. The level of photosynthetic pigments was markedly greater in the samples exposed to drought but treated with foliar SA, in comparison to the samples exposed to drought without any treatment (128.28±8.78 and 38.01±1.15 µg/g total chlorophyll (a+b), respectively). The total polyphenol content and antioxidant capacity of both the leaves and flowers of Petunia were assessed. The polyphenols content was significantly reduced by drought. However, following the application of SA, anthocyanin levels increased in comparison to the sample that had been subjected to drought. The results demonstrate that applying SA to the leaves mitigates the adverse impacts of drought, providing a solution to the issue of water scarcity in ornamental plants, particularly Petunia grandiflora. The use of biochar in soilless media is becoming more common in greenhouse production. This study was conducted to evaluate Eastern red cedar biochar as a soilless-media supplement for the growth of geranium (Pelargonium graveolens L.) and petunia (Petunia sp. J.) as potted plants in greenhousese. Eastern red cedar biomass was produced at three different temperature ranges 300–350 °C, 400–450 °C, and 500–550 °C and applied at 15%, 30%, 45%, and 60% v/v to a soilless media plus a control. Additionally, Eastern red cedar (ERC) bark was also applied at 15%, 30%, 45%, and 60% v/v to a soilless media. The growth characteristics of the plants along with the physical and nutrient properties of the growth media were recorded. For physical properties, 100% soilless media had the greatest bulk density, 60% biochar at 400–450 °C showed the greatest total porosity, and 60% ERC bark showed the greatest air porosity. For geranium, 15% biochar at 500–550 °C showed the greatest height, water use efficiency, and shoot dry weight, while 15% biochar at 300–350 °C showed the greatest width. The greatest number of flowers was at 45% biochar at 500–550 °C. For petunia, 15% biochar at 300–350 °C showed the greatest height, and 15% biochar at 500–550 °C showed the greatest width. The greatest number of flowers and flower diameter was achieved with 100% soilless media. The 15% biochar at 500–550 °C had the greatest shoot dry weight and water use efficiency, and 60% biochar at 400–450 °C had the greatest root dry weight. Biochar treatment outperforms the ERC bark treatment for both species. Within biochar treatment, 15% biochar outperforms dry biomass, while for flowering, 100% soilless media performs best for petunia, and biochar performs best at less than 45% for geranium. The European Commission requested the EFSA Panel on Plant Health to evaluate the probability of entry of pests (likelihood of pest freedom at entry), including both regulated and non‐regulated pests, associated with unrooted cuttings of the genera Petunia and Calibrachoa produced under physical isolation in Kenya. The relevance of any pest for this opinion was based on evidence following defined criteria, based on the methodology used for High‐Risk Plants adapted for the specificity of this assessment. Fourteen EU‐regulated pests (Bemisia tabaci, cowpea mild mottle virus, Liriomyza huidobrensis, Liriomyza sativae, Liriomyza trifolii, potato leafroll virus, potato spindle tuber viroid, Ralstonia pseudosolanacearum, R. solanacearum, Scirtothrips dorsalis, tomato mild mottle virus, tomato spotted wilt virus, tomato yellow leaf curl virus and Xanthomonas vesicatoria) and six EU non‐regulated pests (Aleurodicus dispersus, pepper veinal mottle virus, Nipaecoccus viridis, Phenacoccus solenopsis, Tetranychus neocaledonicus and tomato yellow ring virus) fulfilled all relevant criteria and were selected for further evaluation. For these pests, the risk mitigation measures proposed in the technical dossier from Kenya were evaluated, taking into account the possible limiting factors. Additionally, an expert judgement is given on the likelihood of pest freedom, taking into consideration the risk mitigation measures acting on the pest, including uncertainties associated with the assessment. The estimated degree of pest freedom varies among the pests evaluated, with T. neocaledonicus being the pest most frequently expected on the imported cuttings. The Expert Knowledge Elicitation indicated, with 95% certainty, that between 9942 and 10,000 bags containing unrooted cuttings of Petunia spp. and Calibrachoa spp. per 10,000 would be free of T. neocaledonicus. Agroinfiltration-mediated transient expression is widely used in plant gene functional studies. In this study, the functionality of a metabolic engineering construct containing multiple expression cassettes for betalain biosynthesis genes was validated via agroinfiltration in petunia flower petals. The effects of transcription factors that upregulate anthocyanin biosynthesis and a gene that regulates cellular pH were also demonstrated on petunia petals by agroinfiltration. We also showed that flower petal agroinfiltration is a quick approach to test gene editing components, e.g. different guide RNAs. In general, the agroinfiltration-mediated transient transformation in flower petals is a great tool for testing flowering colour–related trait genes, as well as for optimising gene editing components. As peat (P) demand increases throughout the horticultural industry, alternative fibers must be evaluated. Sugarcane bagasse (B), wood fiber (W), and coconut coir (C) have received interest as domestically available alternatives to P, with demonstrated success in producing greenhouse crops. However, there is limited research comparing these materials to peat. This research evaluated the substrate properties and productivity of Petunia Supertunia Mini Vista 'Indigo' in pine bark substrates amended with C, W, B, or P and fertigated weekly at 100, 200, or 300 parts per million (ppm) nitrogen (N) to account for possible N immobilization. The container capacity was lowest and air-filled porosity was highest in W and B substrates. Substrate pH increased in W and B substrates, and C substrates were fertigated at 100 ppm N. Increasing the N rate increased the growth index in all substrates, especially B and W substrates later in the production period. Higher fertilization increased shoot mass, chlorophyll content, and blooms across all substrates, demonstrating that fertilizer supplementation may offset possible N immobilization. While plant growth and quality parameters were greatest in the P blend, increasing N applications produced similar-quality plants using alternative substrates, demonstrating that modifying fertilizer management practices can make alternative fibers a viable horticultural substrate. Nuclear Factor Y (NF-Y) is a class of heterotrimeric transcription factors composed of three subunits: NF-A, NF-YB, and NF-YC. NF-YC family members play crucial roles in various developmental processes, particularly in the regulation of flowering time. However, their functions in petunia remain poorly understood. In this study, we isolated four PhNF-YC genes from petunia and confirmed their subcellular localization in both the nucleus and cytoplasm. We analyzed the transcript abundance of all four PhNF-YC genes and found that PhNF-YC2 and PhNF-YC4 were highly expressed in apical buds and leaves, with their transcript levels decreasing before flower bud differentiation. Silencing PhNF-YC2 using VIGS resulted in a delayed flowering time and reduced chlorophyll content, while PhNF-YC4-silenced plants only exhibited a delayed flowering time. Furthermore, we detected the transcript abundance of flowering-related genes involved in different signaling pathways and found that PhCO, PhGI, PhFBP21, PhGA20ox4, and PhSPL9b were regulated by both PhNF-YC2 and PhNF-YC4. Additionally, the transcript abundance of PhSPL2, PhSPL3, and PhSPL4 increased only in PhNF-YC2-silenced plants. Overall, these results provide evidence that PhNF-YC2 and PhNF-YC4 negatively regulate flowering time in petunia by modulating a series of flowering-related genes. [ABSTRACT FROM AUTHOR]

John Lanchester's 'crunch lit' novel Capital (2012) draws attention to what I call "experiences of decorporation": the vulnerable ageing body and the medical system both become victims of a market-logic which invisibilises the cared for, the carer, and the caring. Taking place on a single street in London, the metonymic construction of the novel calls for an analysis of how the illness and death of Petunia Howe, the oldest resident on Pepys Road, is symptomatic of the ethical effacement of the body politic under the clout of neoliberal politics. Humic substances are components of soil organic matter that influence soil structure and fertility. Humic and fulvic acids can be extracted from soil and other organic sources, and are used as biostimulants to promote plant growth and increase nutrient availability and uptake. The goal of this study was to determine whether selected humic and fulvic acid-based commercial products would promote growth and flowering of petunia (Petunia ×hybrida) 'Picobella Blue' grown in soilless media with low or optimal fertilizer rates. Plants were grown in 11.4-cm pots filled with peat-based media [80:20 peat:perlite (v/v); pH 5.4]. Three biostimulant products were evaluated at different rates and application frequencies: Huma Pro, a liquid humic acid biostimulant; Fulvi Pro, a liquid fulvic acid biostimulant; andMicromate, a powder containing both humic and fulvic acids. In Expt. 1, Huma Pro and Fulvi Pro were drenched weekly onto the growing media at a rate of 5, 10, or 20 mL·L-1; Micromate was drenched weekly at a rate of 5, 10, 20, or 40 g·L-1. Plants were fertilized with either 50 mg·L-1 nitrogen (N) (low fertility) or 100 mg·L-1 N (optimal fertility) from Jack's Professional 20N-1.3P-15.7K Petunia FeED each irrigation. Control plants received fertilizer but no biostimulant treatments. In Expt. 2, biostimulant treatments were drenched once at transplant, biweekly, or weekly at a rate of 1.25, 2.5, 5, or 10 mL·L-1 for Huma Pro and Fulvi Pro; and at 5, 10, 20, or 40 g·L-1 for Micromate. All plants received constant liquid feed at the lower fertilizer rate of 50 mg·L-1 N. In Expt. 1, plants fertilized with 100mg·L-1 N and treated with 20 g·L-1 Micromate had the best performance. The average shoot dry weight was 32% greater than that of the control plants. Micromate (20 g·L-1)-treated plants had an average of five more flowers per plant, and they flowered 4 days earlier than untreated control plants. In Expt. 2, plants treated with 40 g·L-1 of Micromate weekly had the greatest shoot dry weight compared with the other treatments. Weekly Micromate treatments (40 g·L-1) resulted in plants with an average of 13 more flowers per plant, which flowered 7 days earlier than control plants. Plants treated with Fulvi Pro and Huma Pro at 20 mL·L-1 had a significantly greater concentration of potassium in shoot tissue, whereas Micromate treatments at 20 and 40 g·L-1 resulted in a greater concentration of phosphorous in the shoots. Floral homeotic MADS-box transcription factors ensure the correct morphogenesis of floral organs, which are organized in different cell layers deriving from distinct meristematic layers. How cells from these distinct layers acquire their respective identities and coordinate their growth to ensure normal floral organ morphogenesis is unresolved. Here, we studied petunia (Petunia × hybrida) petals that form a limb and tube through congenital fusion. We identified petunia mutants (periclinal chimeras) expressing the B-class MADS-box gene DEFICIENS in the petal epidermis or in the petal mesophyll, called wico and star, respectively. Strikingly, wico flowers form a strongly reduced tube while their limbs are almost normal, while star flowers form a normal tube but greatly reduced and unpigmented limbs, showing that petunia petal morphogenesis is highly modular. These mutants highlight the layer-specific roles of PhDEF during petal development. We explored the link between PhDEF and petal pigmentation, a well-characterized limb epidermal trait. The anthocyanin biosynthesis pathway was strongly downregulated in star petals, including its major regulator ANTHOCYANIN2 (AN2). We established that PhDEF directly binds to the AN2 terminator in vitro and in vivo, suggesting that PhDEF might regulate AN2 expression and therefore petal epidermis pigmentation. Altogether, we show that cell layer–specific homeotic activity in petunia petals differently impacts tube and limb development, revealing the relative importance of the different cell layers in the modular architecture of petunia petals. Electric lighting is often necessary to achieve a target daily light integral (DLI) for the production of high-quality young annual bedding plants (plugs). Early in production, plugs have a low leaf area index that limits light interception and likely results in wasted radiation supplied by electric sources. Previous research has shown that the addition of far-red radiation (700-780 nm) to the radiation spectrum in sole-source lighting experiments or the use of end-of-day far-red (EOD-FR) radiation treatments can promote an increase in leaf expansion and leaf area for many species. However, leaf expansion in response to far-red radiation may depend on other factors such as the ratio of red (600-699 nm) to far-red radiation (R:FR) and air temperature. Thus, the objectives of this work were to examine the effects of far-red radiation applied throughout the photoperiod and as an end-of-day radiation treatment on the morphology of petunia 'Dreams Midnight' seedlings grown under different temperature conditions. Specifically, petunia seed was sown in 128-cell trays and moved to one of two growth chambers set at 16 or 21 °C when cotyledons unfolded. Seedlings received an equal total photon flux density (400-780 nm) of 164 mmol·m22·s21 for a 17.25-hour photoperiod, and either a high (~10.7) or low R:FR (0.5). Low R:FR-treated seedlings were grown at a constant temperature of either 16 or 21 °C and placed under blackout conditions at the end of the photoperiod. High R:FRgrown seedlings received either a 1-hour end-of-day white (EOD-W) or EOD-FR treatment at the end of the photoperiod, and were grown at a constant 16 or 21 °C; one EOD-FR treatment was also shifted from the 21 °C chamber to the 16°C at the end of the photoperiod for both the EOD-FR treatment and subsequent dark period. Seedlings were harvested at 21 and 28 days after treatment initiation. For petunia seedlings grown at 21 °C, EOD-FR treatments had minimal effect on morphology or dry mass as all measured parameters, including total and average leaf area and stem length, were similar to EOD-W treatments. In contrast, low R:FR-treated seedlings showed responses characteristic of plants grown under shade, including significant stem elongation, an increase in total and average leaf area, and a reduction in leaf mass per unit area. As expected, production at 16 °C slowed the growth of petunia seedlings resulting in much smaller plants compared with the 21 °C grown plants, but shade responses such as elongated leaves and stems under a low R:FR were apparent. The EOD-FR-treated seedlings that received the diurnal temperature shift also showed reduced leaf area and dry mass compared with their constant 21 °C counterparts. Shade responses were observable at both 16 and 21 °C for low R:FR-grown plants, but the quantifiable impact of temperature on far-red responses could not be fully determined in the present study. Further research is warranted investigating crop responses to far-red radiation as well as potential interacting environmental factors as the promotion of morphological responses, such as leaf expansion, early in production may prove a useful strategy. The European Commission requested the EFSA Panel on Plant Health to evaluate the probability of entry of pests (likelihood of pest freedom at entry), including both, regulated and non‐regulated pests, associated with unrooted cuttings of the genera Petunia and Calibrachoa produced under physical isolation in Guatemala. The relevance of any pest for this opinion was based on evidence following defined criteria, based on the methodology used for high‐risk plants adapted for the specificity of this assessment. Nineteen EU regulated pests (Bemisia tabaci, pepper golden mosaic virus, pepper huasteco yellow vein virus, tomato severe leaf curl virus, tomato yellow leaf curl virus, tomato spotted wilt virus, Liriomyza huidobrensis, Liriomyza sativae, Liriomyza trifolii, Bactericera cockerelli, Eotetranichus lewisi, Epitrix subcrinita, Epitrix cucumeris, Helicoverpa zea, Chloridea virescens, Spodoptera ornithogalli, Ralstonia solanacearum, Ralstonia pseudosolanacearum, Xanthomonas vesicatoria) and one EU non‐regulated (Phenacoccus solenopsis) pest fulfilled all relevant criteria and were selected for further evaluation. For these pests, the risk mitigation measures proposed in the technical dossier from Guatemala were evaluated taking into account the possible limiting factors, and an expert judgement is given on the likelihood of pest freedom taking into consideration the risk mitigation measures acting on the pest, including uncertainties associated with the assessment. The limited and partially conflicting information provided in the dossier contributes to the wide estimates of pest freedom. The estimated degree of pest freedom varies among the pests evaluated, with Ralstonia spp. (R. solanacearum and R. pseudosolanacearum) being the pest most frequently expected on the imported cuttings. The epidermal cells of petunia (Petunia × hybrida) flowers are the main site of volatile emission. However, the mechanisms underlying the release of volatiles into the environment are still being explored. Here, using cell-layer-specific transcriptomic analysis, reverse genetics by virus-induced gene silencing and clustered regularly interspaced short palindromic repeat (CRISPR), and metabolomics, we identified EPIDERMIS VOLATILE EMISSION REGULATOR (EVER)—a petal adaxial epidermis-specific MYB activator that affects the emission of volatiles. To generate ever knockout lines, we developed a viral-based CRISPR/Cas9 system for efficient gene editing in plants. These knockout lines, together with transient-suppression assays, revealed EVER's involvement in the repression of low-vapor-pressure volatiles. Internal pools and annotated scent-related genes involved in volatile production and emission were not affected by EVER. RNA-Seq analyses of petals of ever knockout lines and EVER-overexpressing flowers revealed enrichment in wax-related biosynthesis genes. Liquid chromatography/gas chromatography–MS analyses of petal epicuticular waxes revealed substantial reductions in wax loads in ever petals, particularly of monomers of fatty acids and wax esters. These results implicate EVER in the emission of volatiles by fine-tuning the composition of petal epicuticular waxes. We reveal a petunia MYB regulator that interlinks epicuticular wax composition and volatile emission, thus unraveling a regulatory layer in the scent-emission machinery in petunia flowers. The art of pressed flowers is a method of artistic expression involving the pressing of flowers, leaves, and other plant organs for artistic creative purposes. However, the pressing process often results in color variation of the plant material, which significantly diminishes the quality of artistic works and must be solved using appropriate techniques. During this research, phenylalanine (10 mmol.L-1) was used to treat the petals of postharvest Petunia flowers to investigate the impact of phenylalanine on mitigating color variation, and the effect of phenylalanine on inhibiting the color variation of Petunia petals during the pressing process was evaluated by color measurement, physicochemical indices, and gene expression level analyses. Using the CIEL*a*b* color measurements, the samples from the test group had significantly higher brightness (L*) and red coloration (a*) at the final stage (S4) than the control group. In addition, phenylalanine had a significant inhibitory effect on malondialdehyde and superoxide anion accumulations in Petunia petals during pressing and reduced the enzymatic activities of superoxide dismutase, polyphenol oxidase, and catalase. The quantitative reverse-transcription polymerase chain reaction analysis showed that the transcript levels of CHS, DFR, F3050H, and UFGT genes in the petals of the treatment group continued to increase during the pressing process, and the transcript levels of key genes in the anthocyanin metabolic pathway of the treated samples were higher than those of the control group at the final stage (S4). These results indicated that phenylalanine can effectively diminish the color variation of Petunia petals in the pressing process, which could serve as a theoretical basis for the development of a comprehensive technology system aimed at preserving the color of pressed horticultural plants. The production of transgenic petunia plants with heterologous ZRNase II gene and the evaluation of resistance to Tobacco mosaic virus in the resultant plants was the aim of our research. We have generated transgenic petunia plants with ZRNase gene via Agrobacterium-mediated transformation. The presence of ZRNase II gene was confirmed by PCR-analysis. Orcin method revealed that RNase activity in transgenic petunia expressing ZRNase II gene was 1.34-2.10 fold higher compared to wild-type plants. RNase activity varied among different transgenic lines. Both petunia varieties used for production of transgenic lines differed in their natural resistance to Tobacco mosaic virus.The symptoms of the viral infection in the transgenic plants of the susceptible variety inoculated with the virus were less severe. Indirect ELISA confirmed 3.3 -- 4.0 fold lower accumulation of viral antigenes in the infected transformed plants compared to the infected wild type ones. The in vitro growth of the pollen tube (PT), an object of comprehensive and intensive research, is a model for studying the mechanisms of sexual reproduction in higher plants. We have studied the potential role of reactive oxygen species (ROS) in the in vitro germination and growth maintenance of the petunia (Petunia hybrida E. Vilm.) male gametophyte. The exogenous treatment with H2O2 influences the PT germination and polar growth in vitro. The addition of H2O2 to culture medium increases both the percentage of pollen grain germination and the PT length in the case of long cultivation, but inhibits both processes during the first hour of cultivation. This suggests that endogenous ROS play a decisive role in the early stages of pollen germination, with the sensitivity to endogenous ROS emerging later over the course of their growth. The addition of diphenylene iodonium chloride (DPI), a NADPH oxidase inhibitor, considerably decreases both the germination and the growth of the petunia male gametophyte at low concentrations (0.1 μM), and completely arrests the growth at high concentrations (1 μM). ROS are necessary for polar growth of the petunia male gametophyte; they are secreted in the early stages of pollen grain activation and are further localized to the initiation of the PT, mainly in the PT apical part, during polar growth, as confirmed with the help of intravital fluorescence microscopy. The important feature of petunia in tissue culture is its unpredictable and genotype-dependent callogenesis, posing challenges for efficient regeneration and biotechnology applications. To address this issue, machine learning (ML) can be considered a powerful tool to analyze callogenesis data, extract key parameters, and predict optimal conditions for petunia callogenesis, facilitating more controlled and productive tissue culture processes. The study aimed to develop a predictive model for callogenesis in petunia using ML algorithms and to optimize the concentrations of phytohormones to enhance callus formation rate (CFR) and callus fresh weight (CFW). The inputs for the model were BAP, KIN, IBA, and NAA, while the outputs were CFR and CFW. Three ML algorithms, namely MLP, RBF, and GRNN, were compared, and the results revealed that GRNN (R2≥83) outperformed MLP and RBF in terms of accuracy. Furthermore, a sensitivity analysis was conducted to determine the relative importance of the four phytohormones. IBA exhibited the highest importance, followed by NAA, BAP, and KIN. Leveraging the superior performance of the GRNN model, a genetic algorithm (GA) was integrated to optimize the concentration of phytohormones for maximizing CFR and CFW. The genetic algorithm identified an optimized combination of phytohormones consisting of 1.31 mg/L BAP, 1.02 mg/L KIN, 1.44 mg/L NAA, and 1.70 mg/L IBA, resulting in 95.83% CFR. To validate the reliability of the predicted results, optimized combinations of phytohormones were tested in a laboratory experiment. The results of the validation experiment indicated no significant difference between the experimental and optimized results obtained through the GA. This study presents a novel approach combining ML, sensitivity analysis, and GA for modeling and predicting callogenesis in petunia. The findings offer valuable insights into the optimization of phytohormone concentrations, facilitating improved callus formation and potential applications in plant tissue culture and genetic engineering. Petunia (Petunia × hybrida Hort. Vilm.-Andr.) is a well-suited plant for sustainable landscape issues in borderline areas with irrigation with saline water. Salicylic acid (SA) as a modulator performs an imperative function in modulating plant salt tolerance. However, there are a few reports on the effect of SA on petunia plants irrigated with saline water. During the 2022/2023 season, a factorial pot experiment in a randomized complete block design was carried out in Riyadh, Saudi Arabia, to assess the effect of SA concentration (0, 500, 1000, 2000 mgL−1) on petunia plant growth, flowering, ion content, chlorophyll level, and proline concentration under irrigation with salty water (230, 1500, 3000 mgL−1). Saline water up to 3000 mgL−1 dramatically reduced plant growth, chlorophyll, ions, and flowering attributes, while the contrary was observed in proline and sodium concentrations as compared to the control treatments (irrigation with tap water). Foliar spraying with 1000 mgL−1 SA considerably boosted plant growth and flowering as well as chlorophyll, proline, and ion content compared to untreated plants under such salinity levels. Alternatively, the application of 1000 mgL−1 to normal or salinized water significantly decreased the Na content in non-treated plants under such a salinity level. Accordingly, using 1000 mgL−1 of SA under salt stress conditions could be a useful technique to lessen the mutilation induced by the use of salinized water in the era of climate change. The Succineidae Beck, 1837 are a family of terrestrial mollusks that are increasingly transported worldwide in horticultural trade. Succineids are extremely difficult to identify morphologically, especially if collected from locations beyond their native range and in the absence of knowledge of their native provenance. In this study, we report an unidentified species of the Succineidae in a flower bed of Petunia sp., Ariake, Tokyo, Japan. A fragment of the mitochondrial cytochrome oxidase subunit 1 (COI) gene was sequenced, and the shell and genital morphologies were compared with those of other similar species. The action of the petunia strigolactone (SL) hormone receptor DAD2 is dependent not only on its interaction with the PhMAX2A and PhD53A proteins, but also on its expression patterns within the plant. Previously, in a yeast-2-hybrid system, we showed that a series of a single and double amino acid mutants of DAD2 had altered interactions with these binding partners. In this study, we tested the mutants in two plant systems, Arabidopsis and petunia. Testing in Arabidopsis was enabled by creating a CRISPR-Cas9 knockout mutant of the Arabidopsis strigolactone receptor (AtD14). We produced SL receptor activity in both systems using wild type and mutant genes; however, the mutants had functions largely indistinguishable from those of the wild type. The expression of the wild type DAD2 from the CaMV 35S promoter in dad2 petunia produced plants neither quite like the dad2 mutant nor the V26 wild type. These plants had greater height and leaf size although branch number and the plant shape remained more like those of the mutant. Introduction: The garden petunia, Petunia hybrida (Solanaceae) is a fertile, diploid, annual hybrid species (2n=14) originating from P. axillaris and P. inflata 200 years ago. To understand the recent evolution of the P. hybrida genome, we examined tandemly repeated or satellite sequences using bioinformatic and molecular cytogenetic analysis. Methods: Raw reads from available genomic assemblies and survey sequences of P. axillaris N (PaxiN), P. inflata S6, (PinfS6), P. hybrida (PhybR27) and the here sequenced P. parodii S7 (PparS7) were used for graph and k-mer based cluster analysis of TAREAN and RepeatExplorer. Analysis of repeat specific monomer lengths and sequence heterogeneity of the major tandem repeat families with more than 0.01% genome proportion were complemented by fluorescent in situ hybridization (FISH) using consensus sequences as probes to chromosomes of all four species. Results: Seven repeat families, PSAT1, PSAT3, PSAT4, PSAT5 PSAT6, PSAT7 and PSAT8, shared high consensus sequence similarity and organisation between the four genomes. Additionally, many degenerate copies were present. FISH in P. hybrida and in the three wild petunias confirmed the bioinformatics data and gave corresponding signals on all or some chromosomes. PSAT1 is located at the ends of all chromosomes except the 45S rDNA bearing short arms of chromosomes II and III, and we classify it as a telomere associated sequence (TAS). It is the most abundant satellite repeat with over 300,000 copies, 0.2% of the genomes. PSAT3 and the variant PSAT7 are located adjacent to the centromere or mid-arm of one to three chromosome pairs. PSAT5 has a strong signal at the end of the short arm of chromosome III in P. axillaris and P.inflata, while in P. hybrida additional interstitial sites were present. PSAT6 is located at the centromeres of chromosomes II and III. PSAT4 and PSAT8 were found with only short arrays. Discussion: These results demonstrate that (i) repeat families occupy distinct niches within chromosomes, (ii) they differ in the copy number, cluster organization and homogenization events, and that (iii) the recent genome hybridization in breeding P. hybrida preserved the chromosomal position of repeats but affected the copy number of repetitive DNA. Background: Shoot branching of flowering plants exhibits phenotypic plasticity and variability. This plasticity is determined by the activity of axillary meristems, which in turn is influenced by endogenous and exogenous cues such as nutrients and light. In many species, not all buds on the main shoot develop into branches despite favorable growing conditions. In petunia, basal axillary buds (buds 1–3) typically do not grow out to form branches, while more apical axillary buds (buds 6 and 7) are competent to grow. Results: The genetic regulation of buds was explored using transcriptome analyses of petunia axillary buds at different positions on the main stem. To suppress or promote bud outgrowth, we grew the plants in media with differing phosphate (P) levels. Using RNA-seq, we found many (> 5000) differentially expressed genes between bud 6 or 7, and bud 2. In addition, more genes were differentially expressed when we transferred the plants from low P to high P medium, compared with shifting from high P to low P medium. Buds 6 and 7 had increased transcript abundance of cytokinin and auxin-related genes, whereas the basal non-growing buds (bud 2 and to a lesser extent bud 3) had higher expression of strigolactone, abscisic acid, and dormancy-related genes, suggesting the outgrowth of these basal buds was actively suppressed. Consistent with this, the expression of ABA associated genes decreased significantly in apical buds after stimulating growth by switching the medium from low P to high P. Furthermore, comparisons between our data and transcriptome data from other species suggest that the suppression of outgrowth of bud 2 was correlated with a limited supply of carbon to these axillary buds. Candidate genes that might repress bud outgrowth were identified by co-expression analysis. Conclusions: Plants need to balance growth of axillary buds into branches to fit with available resources while allowing some buds to remain dormant to grow after the loss of plant parts or in response to a change in environmental conditions. Mildew Locus O (MLO), which encodes a seven-transmembrane domain protein, is responsible for the development of powdery mildew disease symptoms in many plant species. Hence, we knocked out PhMLO1 from Petunia hybrida cv. Mirage Rose at the genomic level using the Clustered regularly interspaced short palindromic repeat/CRISPR-associated nuclease 9 (CRISPR/Cas9) system and investigated involvement of the gene in powdery mildew disease development in the petunia. The MLO1 transcript levels observed in all mlo1 mutants were significantly lower than that in the wild type (WT). Specifically, the transcript level was the lowest in the mutants (line no. 1, 25, 33, and 85), followed by the mutants (line no. 14, 17, and 81), and line no. 6. Disease development was positively linked to the higher transcript levels of PhMLO1, because the disease severity observed in WT was greater than that observed in the mutants (line no. 6,14, 17, and 81), and disease symptoms were almost not observed in the mutants (line no. 1, 25, 33, and 85). In addition, the transmission of the edited alleles from the T0 to T1 generation was also confirmed, and the disease severity results were identical to those observed in the T0 mutant lines. Overall, the editing of PhMLO1 completely or partially prevented the development of this disease in this petunia, depending on MLO1 transcript level of the mutants. This study highlights the role of PhMLO1 in the development of powdery mildew disease in petunia and points to the need to edit the MLO genes in other ornamental plants to improve ornamental quality against powdery mildew disease. Summary: In angiosperms, the timely delivery of sperm cell nuclei by pollen tube (PT) to the ovule is vital for double fertilization. Penetration of PT into maternal stigma tissue is a critical step for sperm cell nuclei delivery, yet little is known about the process. Here, a male‐specific and sporophytic mutant xt6, where PTs are able to germinate but unable to penetrate the stigma tissue, is reported in Oryza sativa. Through genetic study, the causative gene was identified as Chalcone synthase (OsCHS1), encoding the first enzyme in flavonoid biosynthesis. Indeed, flavonols were undetected in mutant pollen grains and PTs, indicating that the mutation abolished flavonoid biosynthesis. Nevertheless, the phenotype cannot be rescued by exogenous application of quercetin and kaempferol as reported in maize and petunia, suggesting a different mechanism exists in rice. Further analysis showed that loss of OsCHS1 function disrupted the homeostasis of flavonoid and triterpenoid metabolism and led to the accumulation of triterpenoid, which inhibits significantly α‐amylase activity, amyloplast hydrolysis and monosaccharide content in xt6, these ultimately impaired tricarboxylic acid (TCA) cycle, reduced ATP content and lowered the turgor pressure as well. Our findings reveal a new mechanism that OsCHS1 modulates starch hydrolysis and glycometabolism through modulating the metabolic homeostasis of flavonoids and triterpenoids which affects α‐amylase activity to maintain PT penetration in rice, which contributes to a better understanding of the function of CHS1 in crop fertility and breeding. Anthocyanins are responsible for the color spectrum of both ornamental and natural flowers. However, not all plant species produce all colors. For example, roses are not blue because they do not naturally possess a hydroxylase that opens the pathway for delphinidin and its derivatives. It is more intriguing why some plants do not carry orange or scarlet red flowers with anthocyanins based on pelargonidin, because the precursor for these anthocyanins should be available if anthocyanins are made at all. The key to this is the substrate specificity of dihydroflavonol 4-reductase (DFR), an enzyme located at the branch point between flavonols and anthocyanins. The most common example is petunia, which does not bear orange flowers unless the enzyme is complemented by biotechnology. We changed a few amino acids in the active site of the enzyme and showed that the mutated petunia DFR started to favor dihydrokaempferol, the precursor to orange pelargonidin, in vitro. An experiment was conducted at Banda University of Agriculture & Technology, Banda to evaluate the effect of different growing media compositions containing cocopeat, Vermicompost, FYM, Leaf Mold, Perlite, and vermiculite in different ratios on vegetative growth and flowering of petunia plant. The results revealed that, the maximum root length and root spread were observed in media composition T5 [Cocopeat + Vermicompost + Vermiculite (2:1:1)] whereas, vegetative growth with no of leaves per plants, no of branches per plant and plant spread in T4 [Cocopeat + Perlite + Vermicompost (2:1:1)]. The better flower quality parameters like flower diameter, flower bud diameter, stalk length of flower were observed in T5 media containing Cocopeat + Vermicompost + Vermiculite (2:1:1) while minimum days taken to bud initiation (60.75) and first flower opening (66.66) in T2 [Soil + Sand + Vermicompost (2:1:1)]. However, plants showed the significant root growth, vegetative growth with better flowering time and quality was highest in media containing Cocopeat + Vermicompost + Vermiculite (2:1:1). Therefore, the findings suggest that for quality flower production of potted petunia growing media composition of T5 [Cocopeat + Vermicompost + Vermiculite (2:1:1)] was ideal with better plant morphological development and sustained flowering for display. Petal size is a key indicator of the ornamental value of plants, such as Petunia hybrida L., which is a popular ornamental species worldwide. Our previous study identified a flower-specific expression pattern of a DNA-binding one finger (Dof)-type transcription factor (TF) PhDof28, in the semi-flowering and full-flowering stages of petunia. In this study, subcellular localization and activation assays showed that PhDof28 was localized in the cell nucleus and could undergo in vitro self-activation. The expression levels of PhDof28 tended to be significantly up-regulated at the top parts of petals during petunia flower opening. Transgenic petunia 'W115' and tobacco plants overexpressing PhDof28 showed similar larger petal phenotypes. The cell sizes at the middle and top parts of transgenic petunia petals were significantly increased, along with higher levels of endogenous indole-3-acetic acid (IAA) hormone. Interestingly, the expression levels of two TFs, PhNAC100 and PhBPEp, which were reported as negative regulators for flower development, were dramatically increased, while the accumulation of jasmonic acid (JA), which induces PhBPEp expression, was also significantly enhanced in the transgenic petals. These results indicated that PhDof28 overexpression could increase petal size by enhancing the synthesis of endogenous IAA in petunias. Moreover, a JA-related feedback regulation mechanism was potentially activated to prevent overgrowth of petals in transgenic plants. Research into molecular mechanisms of self-incompatibility (SI) in plants can be observed in representatives of various families, including Solanaceae. Earlier studies of the mechanisms of S-RNase-based SI in petunia (Petunia hybrida E. Vilm.) demonstrate that programmed cell death (PCD) is an SI factor. These studies suggest that the phytohormon cytokinin (CK) is putative activator of caspase-like proteases (CLPs). In this work, data confirming this hypothesis were obtained in two model objects—petunia and tomato (six Solanaceae representatives). The exogenous zeatin treatment of tomato and petunia stigmas before a compatible pollination activates CLPs in the pollen tubes in vivo, as shown via the intravital imaging of CLP activities. CK at any concentration slows down the germination and growth of petunia and tomato male gametophytes both in vitro and in vivo; shifts the pH of the cytoplasm (PHc) to the acid region, thereby creating the optimal conditions for CLP to function and inhibiting the F-actin formation and/or destructing the cytoskeleton in pollen tubes to point foci during SI-induced PCD; and accumulates in style tissues during SI response. The activity of the ISOPENTENYLTRANSFERASE 5 (IPT5) gene at this moment exceeds its activity in a cross-compatible pollination, and the levels of expression of the CKX1 and CKX2 genes (CK OXIDASE/DEHYDROGENASE) are significantly lower in self-incompatible pollination. In moth-pollinated petunias, production of floral volatiles initiates when the flower opens and occurs rhythmically during the day, for optimal flower-pollinator interaction. To characterize the developmental transcriptomic response to time of day, we generated RNA-Seq databases for corollas of floral buds and mature flowers in the morning and in the evening. Around 70% of transcripts accumulating in petals demonstrated significant changes in expression levels in response to the flowers' transition from a 4.5-cm bud to a flower 1 day postanthesis (1DPA). Overall, 44% of the petal transcripts were differentially expressed in the morning vs. evening. Morning/evening changes were affected by flower developmental stage, with a 2.5-fold larger transcriptomic response to daytime in 1DPA flowers compared to buds. Analyzed genes known to encode enzymes in volatile organic compound biosynthesis were upregulated in 1DPA flowers vs. buds--in parallel with the activation of scent production. Based on analysis of global changes in the petal transcriptome, PhWD2 was identified as a putative scent-related factor. PhWD2 is a protein that is uniquely present in plants and has a three-domain structure: RING-kinase-WD40. Suppression of PhWD2 (termed UPPER - Unique Plant PhEnylpropanoid Regulator) resulted in a significant increase in the levels of volatiles emitted from and accumulated in internal pools, suggesting that it is a negative regulator of petunia floral scent production. Key message: TFL1-like genes of the basal eudicot Platanus acerifolia have conserved roles in maintaining vegetative growth and inhibiting flowering, but may act through distinct regulatory mechanism. Three TERMINAL FLOWER 1 (TFL1)-like genes were isolated and characterized from London plane tree (Platanus acerifolia). All genes have conserved genomic organization and characteristic of the phosphatidylethanolamine-binding protein (PEBP) family. Sequence alignment and phylogenetic analysis indicated that two genes belong to the TFL1 clade, designated as PlacTFL1a and PlacTFL1b, while another one was grouped in the BFT clade, named as PlacBFT. qRT-PCR analysis showed that all three genes primarily expressed in vegetative phase, but the expression of PlacTFL1a was much higher and wider than that of PlacTFL1b, with the latter only detected at relatively low expression levels in apical and lateral buds in April. PlacBFT was mainly expressed in young stems of adult trees followed by juvenile tissues. Ectopic expression of any TFL1-like gene in Arabidopsis showed phenotypes of delayed or repressed flowering. Furthermore, overexpression of PlacTFL1a gene in petunia also resulted in extremely delayed flowering. In non-flowering 35:PlacTFL1a transgenic petunia plants, the FT-like gene (PhFT) gene was significantly upregulated and AP1 homologues PFG, FBP26 and FBP29 were significantly down-regulated in leaves. Yeast two-hybrid analysis indicated that only weak interactions were detected between PlacTFL1a and PlacFDL, and PlacTFL1a showed no interaction with PhFDL1/2. Flower breeders are continually refining their methods for producing high-quality flowers. Phalaenopsis species are considered the most important commercially grown orchids. Advances in genetic engineering technology have provided researchers with new tools that can be used along with traditional breeding methods to enhance floral traits and quality. However, the application of molecular techniques for the breeding of new Phalaenopsis species has been relatively rare. In this study, we constructed recombinant plasmids carrying flower color-related genes, Phalaenopsis Chalcone synthase (PhCHS5) and/or Flavonoid 3′,5′-hydroxylase (PhF3′5′H). These genes were transformed into both Petunia and Phalaenopsis plants using a gene gun or an Agrobacterium tumefaciens-based method. Compared with WT, 35S::PhCHS5 and 35S::PhF3′5′H both had deeper color and higher anthocyanin content in Petunia plants. Additionally, a phenotypic comparison with wild-type controls indicated the PhCHS5 or PhF3′5′H-transgenic Phalaenopsis produced more branches, petals, and labial petals. Moreover, PhCHS5 or PhF3′5′H-transgenic Phalaenopsis both showed deepened lip color, compared with the control. However, the intensity of the coloration of the Phalaenopsis lips decreased when protocorms were co-transformed with both PhCHS5 and PhF3′5′H. The results of this study confirm that PhCHS5 and PhF3′5′H affect flower color in Phalaenopsis and may be relevant for the breeding of new orchid varieties with desirable flowering traits. Petunia hybrida Hort. "Easy Wave Pink", a qualitative long-day plant (LDP), was investigated to study the effects of the night interruption light (NIL) provided by light-emitting diodes (LEDs) quality shifting on the morphogenesis, blooming, and transcription of photoreceptor genes. Plants were grown in a closed-type plant factory employing white (W) LEDs at an intensity of 180 μmol·m−2·s−1 PPFD provided for short day (SD, 10 h light, 14 h dark), long day (LD, 16 h light, 8 h dark), or SD with 4 h night interruption (NI) with LEDs at an intensity of 10 μmol·m−2·s−1 PPFD. The NIL quality was shifted from one light spectrum to another after the first 2 h of NI. Light treatments consisting of all possible pairings of W, far-red (Fr), red (R), and blue (B) light were tested. The SD and LD were referenced as the control, while 12 NI treatments involved altering LED NIL qualities, as follows: from R to B (NI-RB), from B to R (NI-BR), from Fr to R (NI-FrR), from R to Fr (NI-RFr), from Fr to B (NI-FrB), from B to Fr (NI-BFr), from B to W (NI-BW), from W to B (NI-WB), from W to Fr (NI-WFr), from Fr to W (NI-FrW), from W to R (NI-WR), and from R to W (NI-RW). The NI-RFr resulted in the longest shoots, while the NI-WR and NI-RW resulted in the shortest shoots. NI-WR, NI-RW, NI-BW, NI-WB, NI-RFr, NI-RB, NI-BR, and LD all exhibited flowering. High-level expressions of photoreceptor genes were confirmed in the NI-RFr, NI-FrR, NI-BFr, NI-RW, and NI-WR treatments. Morphogenesis and blooming were both impacted by the photoperiod. The process of optimizing in vitro seed sterilization and germination is a complicated task since this process is influenced by interactions of many factors (e.g., genotype, disinfectants, pH of the media, temperature, light, immersion time). This study investigated the role of various types and concentrations of disinfectants (i.e., NaOCl, Ca(ClO)2, HgCl2, H2O2, NWCN-Fe, MWCNT) as well as immersion time in successful in vitro seed sterilization and germination of petunia. Also, the utility of three artificial neural networks (ANNs) (e.g., multilayer perceptron (MLP), radial basis function (RBF), and generalized regression neural network (GRNN)) as modeling tools were evaluated to analyze the effect of disinfectants and immersion time on in vitro seed sterilization and germination. Moreover, non‑dominated sorting genetic algorithm‑II (NSGA‑II) was employed for optimizing the selected prediction model. The GRNN algorithm displayed superior predictive accuracy in comparison to MLP and RBF models. Also, the results showed that NSGA‑II can be considered as a reliable multi-objective optimization algorithm for finding the optimal level of disinfectants and immersion time to simultaneously minimize contamination rate and maximize germination percentage. Multidrug and toxic compound extrusion (MATE) transporter proteins are a class of secondary transporter proteins that can transport flavonoids. Anthocyanins, a kind of flavonoid, are important secondary metabolites widely found in higher plants; they determine the flower color of most angiosperms. TT12 in Arabidopsis was the first MATE protein identified to be involved in flavonoid transport. Petunia (Petunia hybrida) is an important ornamental plant and is one of the ideal plants for studying plant flower color. However, there are few reports on anthocyanin transport in petunia. In this study, we characterized a homolog of Arabidopsis TT12 in the petunia genome, PhMATE1, that shares the highest amino acid sequence identity with Arabidopsis TT12. PhMATE1 protein contained 11 transmembrane helices. PhMATE1 showed a high transcription level in corollas. The silencing of PhMATE1 mediated by both virus‐induced gene silence and RNA interference changed flower color and reduced anthocyanin content in petunia, suggesting that PhMATE1 is involved in anthocyanin transport in petunia. Furthermore, PhMATE1 silencing downregulated the expression of the structural genes of the anthocyanin synthesis pathway. Ethylene-responsive element binding factors (ERFs) are widely involved in the regulation of plant responses to different abiotic stresses. In petunia (Petunia Ã--hybrida), PhERF2 belonging to the subfamily â...? of ERF transcription factors participates in the response to waterlogging stress. In this study, we investigated waterlogging tolerance variation of WT and transgenic petunia plants with RNAi silencing and overexpression of PhERF2 through photosynthetic and physiological performance. Chlorophyll content and root vigor declined continuously in both WT and PhERF2 transgenic lines under waterlogging stress, but the extent of the fall in PhERF2-overexpressing lines was less than that in WT and PhERF2-RNAi lines. At the end of waterlogging treatment, soluble protein levels in PhERF2-overexpressing lines were significantly higher than those in WT and PhERF2-RNAi lines, while the latter showed a higher malondialdehyde content overall. Different degrees of reductions in Pn, Gs, and Tr levels occurred in both WT and PhERF2 transgenic lines upon exposure to waterlogging. The Ci levels of PhERF2-overexpressing lines decreased after 3 hours of waterlogging treatment, and the Ci levels of WT and PhERF2-RNAi lines gradually increased from 6 to 72 hours of waterlogging treatment. These data suggested that non-stomatal factors were the primary limiting factors for Pn in WT and PhERF2-RNAi lines under severe stress, while the stomatal opening was the main factor limiting Pn in PhERF2-overexpressing lines. Our results demonstrated that the contribution of PhERF2 to the waterlogging tolerance of petunia appears to depend on the regulation of physiological and photosynthetic responses.
