Polycyclic aromatic hydrocarbons (PAHs) are a class of persistent and toxic organic pollutants that are widely distributed in nature. Although catabolic pathways are essential for bacteria to effectively utilize PAHs, transporting PAHs and their intermediate metabolites across the bacterial cell envelope is a key factor in PAH catabolism and bacterial survival. In this study, we identified a major facilitator superfamily (MFS) transporter member, PhaT, in a PAH-degrading plasmid of Novosphingobium pentaromativorans US6–1. Evolutionary analyses indicate that PhaT analogs are widely distributed in PAH-degrading bacteria, including the genera Novosphingobium and Sphingobium. Interestingly, deletion of the phaT gene upregulated the levels of PAH-degrading enzymes and PAH-degrading gene expression, thereby accelerating phenanthrene biodegradation. Further studies demonstrated that a deficiency of PhaT impaired the efflux of phenanthrene and its benzene ring-containing intermediate metabolites. Furthermore, the deficiency of PhaT also promoted the biodegradation of naphthalene but not of benzo[ a ]pyrene. Molecular docking and site-directed mutagenesis revealed the essential roles of hydrophobic amino acids in the PhaT central cavity for binding to benzene ring-containing substrates. Collectively, these findings indicate that PhaT plays a pivotal role in facilitating the efflux of PAHs and their benzene ring-containing intermediate metabolites. [Display omitted] • The absence of PhaT accelerates phenanthrene biodegradation. • The deficiency of PhaT impaires the efflux of phenanthrene and its benzene ring-containing intermediate metabolites. • Hydrophobic amino acids in the central cavity of PhaT is crucial to PAH and their intermediate metabolite transport. • The PhaT analogs are widely distributed in the PAH-degrading bacteria, including Novosphingobium and Sphingobium. Interannual variations provide insight into the sensitivity of convective processes. Thus, CloudSat and ERA5 are used to explore the relationship among convective cores, outflows and environmental conditions during El Niño‐Southern Oscillation (ENSO) cycles. Results reveal greater upper‐tropospheric stability during El Niño, resulting in a lower level of neutral buoyancy compared to La Niña. However, outflow levels remain relatively consistent across ENSO cycles. This suggests that, despite less favorable conditions for deep convection during El Niño, stronger convective intensity is required to achieve outflow levels comparable to those in La Niña. Indeed, our results suggest that convection observed during El Niño tends to have broader cores and lower entrainment rates, translating to greater intensity compared to La Niña. These findings emphasize the importance of considering both large‐scale and convective‐scale processes, providing an update to the fixed anvil temperature (FAT) and the proportionately higher anvil temperature (PHAT) hypotheses as originally proposed. Plain Language Summary: Examining year‐to‐year variations provides unique insights into understanding how storms may change in a warmer climate. We use CloudSat and ERA5 reanalysis to examine variations in convective outflow (i.e., an airflow pushed out of storms), environmental factors, and cloud properties during ENSO cycles. Comparing El Niño to La Niña, we find that the atmosphere higher up in the troposphere tends to be more stable, which usually slows down the development of storms, during El Niño. However, the heights where convective outflow occurs do not change much during El Niño and La Niña events. This happens because during El Niño, the upward movement of air in storms is more powerful. This stronger upward movement offsets the stabilizing effect of the upper troposphere, so the overall outflow from the storms stays about the same. Our study aligns with the main idea of the FAT hypothesis, which postulates that in a warming climate, the temperature of the anvils (i.e., wide, flat clouds crawling out of the storm top) stays relatively constant primarily due to a thermodynamic constraint. However, our results show that convective‐scale processes with dynamic control play a key role as well, providing an update to the FAT as originally proposed. Key Points: LNB is lower during El Niño due to a greater UT stability, but convective outflow levels remain consistent across ENSO cyclesThe intensity of convective cores during El Niño has to be stronger than that during La NiñaOur finding necessitates a modification to FAT and the proportionately higher anvil temperature (PHAT) [ABSTRACT FROM AUTHOR]

Pleomorphic hyalinizing angiectatic tumor (PHAT) is a very rare entity of soft tissue considered a “neoplasm of uncertain behaviour of connective or other soft tissue” by the World Health Organization (2020). It develops in subcutaneous tissue of the lower extremities, more frequently in the region of the ankle and foot, and rarely as a deep-seated soft tissue mass in locations such as the perineum, buttock, arms, head and neck, and viscera. Although inconsistent cytogenetic data have been reported on PHAT so far, there are potential morphological and genetic overlaps with hemosiderotic fibrolipomatous tumor (HFLT) and myxoinflammatory fibroblastic sarcoma (MIFS). Here we report a case of PHAT at the level of the upper third of the right thigh in a 48-year-old patient and we also focus on the differential diagnoses of these entities and conduct a literature review of reported cases.

- Abstract
Pleomorphic hyalinizing angiectatic tumor (PHAT) of soft parts and hemosiderotic fibrolipomatous tumor (HFLT) are two rare low-grade locally recurring neoplasms with predilection for the foot/ankle. Recent studies support a close link between the two entities, and origin of PHAT from HFLT and occurrence of hybrid HFLT/PHAT have been documented. Both lesions often harbor TGFBR3 or MGEA5 rearrangements. Rare sarcomas originating from HFLT/PHAT have been reported, typically resembling myxofibrosarcoma or myxoinflammatory fibroblastic sarcoma. We describe a novel SMARCA4-deficient undifferentiated sarcoma with rhabdoid features originating from hybrid HFLT/PHAT in the foot of a 54-year-old male. The tumor pursued a highly aggressive course with rapid regrowth after resection and multiple metastases resulting in patient's death within 5 months, despite systemic chemotherapy. Immunohistochemistry revealed SMARCA4 loss in the undifferentiated sarcoma, but not in the HFLT/PHAT. Molecular testing confirmed TGFBR3/MGEA5 rearrangements. This report expands the phenotypes of sarcomas developing from pre-existing PHAT/HFLT. [ABSTRACT FROM AUTHOR]

- Abstract
Background: Platelet-rich plasma (PRP) has long been used for the restoration of hair in conjunction with microneedling or on its own. Fat grafting to the scalp has also been utilized in the past to improve the quality of hair and the possibility of successful hair transplant. The novel therapy reported in this case series combines the natural progression of these two techniques and utilizes synergistic effects to improve the quality of hair, either in preparation for micrografting or without hair transplant. Objectives: To demonstrate the principles behind the novel approach to restoration of hair and the rationale for its use. Methods: A review of the evidence for PRP and fat transfer for non-scarring alopecia serves as the foundation for the combination treatment reported herein. Through presentation of three cases in this series, we provide examples of the utility of this approach for non-scarring alopecia. This report includes a female who suffered non-scarring alopecia following COVID-19 hospitalization and intensive care stay where she lost a large percentage of her hair, in addition to two male patients suffering from androgenic alopecia. Results: Platelet-rich plasma-hybridized adipose transplant hair was shown in these three cases to improve both the quality and density of hair. It improved the density of hair in all patients and was characterized first by a short period of transient hair loss followed by new hair growth which develops starting at 4 weeks and was readily apparent at 12-week follow-up. Results were maintained at 6-month and 1-year follow-up. Conclusions: PHAT hair offers a combination of beneficial effects—namely the unique healing properties and growth signaling provided by PRP, along with adipocyte angiogenic and growth signaling, which both work to improve scalp quality. The combination of these effects is better than previously characterized PRP injections alone in the hands of these individual practices. This may be due to synergistic interactions at a cellular level, but additional clinical studies are needed to better understand this novel treatment and the observed effects. - Abstract
Pleomorphic hyalinizing angiectatic tumor (PHAT) is a rare mesenchymal tumor of intermediate malignancy. PHAT, and the related hemosiderotic fibrolipomatous tumor, show a recurrent t(1;10)(p22;q24). Fluorescence in situ hybridization (FISH) and BAC (bacterial artificial chromosome) clones have previously identified TGFBR3 and MGEA5 as fusion partners. However, targeted RNA-sequencing allowed for the correct identification of FBXW4 and not MGEA5 as the fusion partner of TGFBR3 in a subcutaneous PHAT, a finding further confirmed by RT-PCR. FBXW4 and MGEA5 share a common cytogenetic location at 10q24.32, thereby suggesting that the use of less precise technology may have led to inaccurate gene identification. The study of additional cases is however required. [ABSTRACT FROM AUTHOR]

- Abstract
A systematic review of the cases documented in the literature regarding Pleomorphic hyalinizing angiectatic tumor of soft parts (PHAT) was performed in order to identify (1) location on presentation (2) surgical treatment modality (3) recurrence rate (4) any associations between location, age, histology, surgery type on recurrence. A systematic review of medical literature listed on PubMed was conducted identifying any prior case report and/or case series of diagnosed PHAT, with no exclusion based on language or time. Twenty-nine articles were identified removing any articles with duplicated cases yielding a total of 93 cases. Cases were broken down by gender, presenting location (UE/LE/axial), surgery type [wide local resection, non-wide local, wide local with radiation therapy (RT), non-wide local with RT], recurrence, and time to recurrence. The mean age at presentation was 54.5 ± 17.1 (range 10-89) with the 76% of cases appearing in the lower extremity (15% UE, 9% Axial). Of the 93 patients, 74 had a known surgical procedure, 31% WL, 40% NWL, 8% WL + RT, 1% NWL + RT. Of those treated surgically, 63 pts had documented follow-up and 18 (29%) had recurrence. A strong association was observed between surgery type and recurrence. Local recurrence was more common within the group undergoing NWLE in 52% (16/41) of cases (p = 0.002). Kaplan-Meier analysis showed an estimate mean time for recurrence of 43.87 months [95% confidence interval (CI) 24.52-63.22; and standard error (SE) 7.59] for the entire population. A trend was also seen toward males having a shorter disease-free survival than females (29.4 mos. vs. 69.5 mos.). No significant association seen between size, location, histology type and recurrence. PHAT has a characteristic presentation in the LE with a relatively high rate of local recurrence and slow-growing potential. Wide local excision appears to be superior in decreasing recurrence rates and a long-term follow-up period is needed. [ABSTRACT FROM AUTHOR]

Introduction Pleomorphic hyalinizing angiectatic tumors are a rare group of tumors that are currently classified as benign tumors of unknown differentiation. To our knowledge, less than 100 cases have been reported in literature. We report a case that presented in the groin – an uncommon location for this rare tumor. Case report A 75 year-old female presented with a seven-year history of painless right groin mass with rapid growth of 2 year duration. On physical examination, a firm and mobile mass was identified in the right groin. It measured 12 cm × 8 cm, with no clinically palpable lymph nodes. Microscopic and immunohistochemical features were consistent with pleomorphic hyalinizing angiectatic tumor. Discussion Pleomorphic hyalinizing angiectatic tumor is a rare soft tissue tumor usually diagnosed using microscopic and immuohistochemical analysis to allow for differentiation from other soft tissue tumors. Informal housing is an acute phenomenon in most of the megacities of the developing countries. With the fast pace of urbanization, Ho Chi Minh City (HCMC) is facing rampant development of unorthodox houses. However, HCMC has some distinct differences to other cities in the developing countries: (I) Living environment of informal sectors is better, and (II) most households are able to legalize their houses. This is the results of flexible policies, in which local governments often legalized the ownerships and regularly upgraded infrastructures and facilities in informal areas. Although local government has attempted to reduce the number of informal housing, transactions in informal housing have been steadily increased in the last few years. Thoi Tam Thon County (TTTC), gathering a large number of illegally constructed houses in Hoc Mon District, HCMC, was selected as a pilot study to demonstrate the above differences and analyze policy challenges for informal housing. The survey showed that the costs of houses in the informal sectors are more affordable for low-income people. Additionally, living conditions in the impoverished neighborhoods are still acceptable. Results also reflect the awareness of the risks in illegally constructing houses. However, the affordable land prices and the opportunities to be legitimized are two major reasons encouraging low-income people take high risks. From the work presented in this paper, a few recommendations have been made to the local governments in order to have appropriate strategies to sustain urban growth and reduce informal housing in HCMC. 16. Phase Transform Time Delay Estimation to Counteract Spectral Haystacking Effects in Jet Exhaust Flow Measurements

- Abstract
This study determined a superior data processing technique for correlating an acoustic signal passing through a subsonic jet engine exhaust in order to estimate the traversal time of the signal. Thrust measurement is possible with enough time delay estimates across different portions of the exhaust. This preliminary study did not take the full array of data necessary to measure thrust, but did validate key aspects of the measurement process. The turbulent shear layers of the exhaust spectrally broaden the signal, creating the appearance of spectral "haystacks", making traditional correlation methods unworkable. An experiment was performed to evaluate the ability of a novel sound source to produce a signal from which a reliable and precise time delay estimate could be found. The test apparatus was installed on either side of a Honeywell TFE731-2 turbofan research engine exhaust cone, with the source and receivers placed near the jet exit plane. The signal was then directed across the jet exhaust. This flow environment is considered an extreme challenge for accurate acoustic signal propagation. A key contribution of this paper is the determination that the Phase Transform processor of the Generalized Cross-Correlation (GCC) method produces the most reliable time delay estimates, for the given signal and flow conditions. Several alternative time delay estimators and GCC processors were examined and evaluated on this data. A proposed explanation is provided for why this time delay estimation technique produces the most accurate results, as well as explanations for why the technique became less reliable as the flow environment became more challenging, with an observed 22% anomalous TDE selection rate for the N1Corr = 60% and N1Corr = 70% conditions combined, versus only 6% for the idle and N1Corr = 50% conditions combined. This paper also details the development and first use of a novel acoustic source that produces a two-tone narrowband signal emanating from a single po

Pleomorphic hyalinizing angiectatic tumor (PHAT) is a rare soft tissue tumor of intermediate malignancy and uncertain cellular origin and lineage of differentiation. Although PHAT is still poorly characterized at the genetic level, there is a potential genetic overlap with two other soft tissue tumors: myxoinflammatory fibroblastic sarcoma (MIFS) and hemosiderotic fibrolipomatous tumor (HFLT); MIFS and HFLT share a characteristic t(1;10)(p22;q24) with breakpoints in the TGFBR3 locus on chromosome 1 and near the MGEA5 locus on chromosome 10. Recently, a PHAT with a similar t(1;10) was reported, suggesting a genetic link between MIFS/HFLT and PHAT. To ascertain whether PHAT is also associated with this translocation, two cases were subjected to single nucleotide polymorphism (SNP) array and fluorescence in situ hybridization analyses. Neither PHAT showed a t(1;10) or other types of rearrangement of the TGFBR3 or MGEA5 loci. Both tumors showed imbalances in the SNP array analysis, but none was shared. Thus, the results indicate that PHAT is genetically distinguishable from MIFS and HFLT, but further studies are needed to identify the salient genetic pathways involved in PHAT development. Pleomorphic hyalinizing angiectatic tumor (PHAT) is a rare soft tissue tumor of low malignant potential, which most often arises in the lower extremities. To the best of our knowledge, there have been no prior descriptions of the imaging features of this neoplasm. In this case series, we report the imaging findings in three patients. Two patients had a lower extremity subcutaneous PHAT, while the third patient had an intramuscular upper extremity PHAT. While imaging features are variable, a diagnosis of PHAT should be considered when encountering an enhancing, subcutaneous tumor with ill-defined margins, particularly in the extremity. Abstract: Pleomorphic hyalinizing angiectatic tumor (PHAT) is a recently described rare entity. The tumor histogenesis is proposed to be of primitive undifferentiated mesenchyme. The tumor has a predilection for the subcutaneous soft tissue especially of lower extremity, although other locations have been well documented. We report a case of PHAT arising in the hilum of the kidney, clinically mimicking an infiltrating malignant neoplasm of renal pelvis. The tumor was discovered during workup for unrelated gastrointestinal tract symptoms. Because of the location of the lesion, excision of mass and radical nephrectomy were performed. The tumor had strong immunohistochemical expression of vimentin, CD34, CD99, and vascular endothelial growth factor (VEGF). No additional lesions were documented during 3 years of follow-up. This is consistent with the current thinking that PHAT is a benign neoplasm with increased incidence of recurrence. We document the unique retroperitoneal location of this rare tumor and suggest that PHAT should be considered among the list of unusual lesions at this site. [Copyright &y& Elsevier]

- Abstract
This paper investigates wideband sound source localization in outdoor cases. In such cases, time difference of arrival (TDOA)-based methods are commonly used for 2-D and 3-D wideband sound source localization. These methods have lower accuracy in comparison with direction of arrival-based approaches. However, they feature fewer microphones and less computation time. High accuracy sound source localization using these methods needs highly accurate time delay measurement, and therefore, high frequency signal sampling rates. Moreover, the need to use numerical analysis methods for local calculations (solving nonlinear equations of closed-form methods) will increase computation time while the calculations may still not converge. Also, a good initial guess close to the true solution is needed to avoid local minima. In this paper, a simple, fast (real time) and accurate pure geometric phase transform-based exact location calculation approach for 3-D localization of wideband sound sources in outdoor far-field and low degree reverberation cases, using only four microphones, is proposed. Based on the proposed method, a simple arrangement of microphones is implemented. Experimental results show that the proposed method has more accuracy and less computation time simultaneously, in comparison with previous closed-form hyperbolic intersection and other TDOA-based state-of-the-art location calculation methods, overcoming their major weaknesses. Also, as the nonlinear closed-form equations are linearized, no initial guess is required. It features less than 0.2^\circ error for angle of arrival, less than 5% error for 3-D location finding, and computation times as low as 250 ms for the localization of a typical wideband sound source such as a flying object (helicopter). This thesis studies the design and implementation of an ultra-sonic water depth sounder. The depth sounder is implemented in a hand-held smart console used by divers. Since the idea of echo sounding is to measure the flight time between transmitting the signal and receiving the echo, the main challenge of this task is to find a time-of-flight (ToF) estimation for a signal in noise. It should be suitable for this specific application and robust when implemented in the device. The thesis contains an investigation of suitable ToF methods. More detailed evaluations of the matched filter, also known as the correlation method, and the linear phase approach are done. Aspects like pulse frequency and duration, speed of sound in water and underwater noise are taken into account. The ToF-methods are evaluated through simulation and experiments. The matched filter approach is found suitable based on these simulations and tests with signals recorded by the console. This verification leads to the implementation of the algorithm on the device. The algorithm is tested in real time, the results are evaluated and improvements suggested.

- Abstract
The algorithm is tested in real time, the results are evaluated and improvements suggested., Denna rapport behandlar skattning av vattendjup med hjälp av ultraljudspulser och implementation av detta. Time delay estimation (TDE)-based algorithms for estimation of direction of arrival (DOA) have been most popular for use with speech signals. This is due to their simplicity and low computational requirements. Though other algorithms, like the steered response power with phase transform (SRP-PHAT), are available that perform better than TDE based algorithms, the huge computational load required for this algorithm makes it unsuitable for applications that require fast refresh rates using short frames. In addition, the estimation errors that do occur with SRP-PHAT tend to be large. This kind of performance is unsuitable for an application such as video camera steering, which is much less tolerant to large errors than it is to small errors. We propose an improved TDE-based DOA estimation algorithm called time delay selection (TIDES) based on either minimizing the weighted least squares error (MWLSE) or minimizing the time delay separation (MWTDS). In the TIDES algorithm, we consider not only the maximum likelihood (ML) TDEs for each pair of microphones, but also other secondary delays corresponding to smaller peaks in the generalized cross-correlation (GCC). From these multiple candidate delays for each microphone pair, we form all possible combinations of time delay sets. From among these we pick one set based on one of the two criteria mentioned above and perform least squares DOA estimation using the selected set of time delays. The MWLSE criterion selects that set of time delays that minimizes the least squares error. The MWTDS criterion selects that set of time delays that has minimum distance from a statistically averaged set of time delays from previously selected time delays. Both TIDES algorithms are shown to out-perform the ML-TDE algorithm in moderate signal to reverberation ratios. In fact, TIDES-MWTDS gives fewer large errors than even the SRP-PHAT algorithm, which makes it very suitable for video camera steering applications. Under small signal to reverberation ratio environments, TIDES-MWTDS breaks down, but TIDES-MWLSE is still shown to out-perform the algorithm based on ML-TDE. Master of Science
