The Breadfruit Institute, a program at the National Tropical Botanical Garden in Hawaiʻi, is dedicated to conserving, studying, and utilizing breadfruit for food and reforestation purposes. Breadfruit is a versatile fruit that grows on a long-lived tree and provides energy, protein, and essential nutrients. The institute has been working to reintroduce breadfruit through research, propagation, and education, and has distributed over 10,000 trees throughout Hawaii since 2007. Their goal is to promote local food production, improve nutrition, and reduce reliance on imported food. The article also discusses the concept of regenerative agriculture and the challenges faced by breadfruit, such as saltwater intrusion and climate change. Electroporation is a complex, iterative, and nonlinear phenomenon often studied through numerical simulations. In recent years, simulations of tissue electroporation have been conducted with static models. However, the results of a static model simulation are restricted to a fixed protocol signature of the pulsed electric field. In this paper, we describe a novel dynamic model of tissue electroporation that also accounts for tissue dispersion and temperature to allow time-domain simulations. We have implemented the biological dispersion of potato tubers and thermal analysis in a commercial finite-element method software. A cell electroporation model was adapted to account for the increase in tissue conductivity. The model yielded 12 parameters divided into three dynamic states of electroporation. The thermal analysis describes the dependence of tissue conductivity on temperature. The model parameters were evaluated using experiments with vegetal tissue (Solanum tuberosum) under electrochemotherapy protocols. The proposed model can accurately predict the conductivity of tissue under electroporation from 100 to 1000 V/cm. A negligible thermal effect was observed at 1000 V/cm, with a temperature increase of 0.89 ° C. We believe that the proposed model is suitable to describe the electroporation at the tissue level and provides a hint of the effects on the cell membrane. [ABSTRACT FROM AUTHOR]

Potato is the most widely grown non-grain crop and ranks as the third most significant global food crop following rice and wheat. Despite its long history of cultivation over vast areas, slow breeding progress and environmental stress have led to a scarcity of high-yielding potato varieties. Enhancing the quality and yield of potato tubers remains the ultimate objective of potato breeding. However, conventional breeding has faced challenges due to tetrasomic inheritance, high genomic heterozygosity, and inbreeding depression. Recent advancements in molecular biology and functional genomic studies of potato have provided valuable insights into the regulatory network of physiological processes and facilitated trait improvement. In this review, we present a summary of identified factors and genes governing potato growth and development, along with progress in potato genomics and the adoption of new breeding technologies for improvement. Environmental concerns and climate protection are gaining increasing emphasis nowadays. A growing number of industries and scientific fields are involved in this trend. Sustainable electronics is an emerging research strand. Environmentally friendly and biodegradable or biobased raw materials can be used for the development of green flexible electronic devices, which may serve to reduce the pollution generated by plastics and electronics waste. In this work, we present cytocompatible, electrically conductive structures of nanocarbon water-soluble composites based on starch films. To accomplish this goal, potato starch-based films with glycerol as a plasticiser were developed along with a water-soluble vehicle for nanocarbon-based electroconductive pastes specifically dedicated to screen printing technology. Societal Impact Statement Summary Can modern breeding technologies, such as genome editing, contribute to reduced pesticide usage? This question has been accentuated by a recent legal proposal to exempt genome‐edited plants from the strict regulations applied to classical genetically modified (GM) crops within the European Union (EU). Using official statistics on crop cultivation and pesticide usage for two example crops commonly grown in Sweden, we calculate that cereal farmers collectively could save up to 70 million € in pesticide usage for wheat alone and that a late blight‐resistant potato could reduce pesticide usage by over 80% provided that the EU legislation is amended.The European Commission has set goals to reduce the use of chemical pesticides, and one way to meet these goals in the agricultural sector is to breed disease‐resistant crops. Here, we ask whether modern breeding technologies, for example, genome editing using site‐directed nucleases, can contribute to these goals. This question has been accentuated by recent legal proposals in the European Union (EU) and several other jurisdictions worldwide to exempt genome‐edited plants from the strict regulations often applied to classical genetically modified (GM) crops. Using official statistics on crop cultivation and pesticide usage for two example crops commonly grown in Sweden (wheat and potato), we show that cereal farmers collectively could potentially save up to 70 million € in pesticide usage for wheat alone and that a late blight‐resistant potato could reduce pesticide usage by over 80% provided that the EU legislation is amended. Summary Ethylene response factors (ERFs) have been associated with biotic stress in Arabidopsis, while their function in non‐model plants is still poorly understood. Here we investigated the role of potato ERF StPti5 in plant immunity. We show that StPti5 acts as a susceptibility factor. It negatively regulates potato immunity against potato virus Y and Ralstonia solanacearum, pathogens with completely different modes of action, and thereby has a different role than its orthologue in tomato. Remarkably, StPti5 is destabilised in healthy plants via the autophagy pathway and accumulates exclusively in the nucleus upon infection. We demonstrate that StEIN3 and StEIL1 directly bind the StPti5 promoter and activate its expression, while synergistic activity of the ethylene and salicylic acid pathways is required for regulated StPti expression. To gain further insight into the mode of StPti5 action in attenuating potato defence responses, we investigated transcriptional changes in salicylic acid deficient potato lines with silenced StPti5 expression. We show that StPti5 regulates the expression of other ERFs and downregulates the ubiquitin‐proteasome pathway as well as several proteases involved in directed proteolysis. This study adds a novel element to the complex puzzle of immune regulation, by deciphering a two‐level regulation of ERF transcription factor activity in response to pathogens. Abscisic acid (ABA) regulates plant development, seed germination, and stress responses. The PYR1-like (PYL) proteins are essential for ABA signalling. However, the evolution and expression of PYL genes in potato (Solanum tuberosum) remain poorly understood. Here, we analysed and identified 17 PYL genes in the potato genome, which were categorised into three groups based on phylogenetic analysis. These genes are distributed across nine chromosomes with predicted proteins subcellar localisation primarily in the cytoplasm. These StPYLs revealed conserved exon structures and domains among the groups. Promoter region analysis indicated hormone and stress-related elements in all StPYL s. Protein–protein interactions and microRNA networks predicted that the interactions of StPYLs are crucial components of ABA signalling, underlining their pivotal role in stress management and growth regulation in potato. Expression profiling across different tissues and under various stresses revealed their varied expression pattern. Further, we validated the expression pattern of selected StPYLs through reverse transcription quantitative PCR under drought, salt, and Phytophthora infestans stresses. This revealed consistent upregulation of StPYL6 in these stresses, while StPYL11 exhibited significant downregulation over time. Other genes showed downregulation under drought and salt stresses while upregulation under P. infestans. Overall, our results suggested the potential role of PYL genes in abiotic and biotic stresses. Starch-based thermoplastic polymer is a biopolymer that is being widely explored as a replacement for conventional polymers. Since thermoplastic starch suffers from mechanical defects, certain mechanical and thermal properties of starch-based polymers can be improved by incorporating fillers or reinforcements derived mainly from natural substances. This article reports the preparation, physicochemical, and mechanical characterization and biodegradation of starch-based bioplastics extracted from potato (Solanum tuberosum) peels using glycerol (G) as plasticizer and reinforced with carob powder, a readily growing plant in Mediterranean climates. The present study investigates the effect of incorporating different proportions (0, 2, 5, 10, and 15 wt.%) of carob powder (Cb) in the films thus prepared. These biopolymer films were fully characterized using analytical techniques including Fourier transform infrared spectroscopy with attenuated total reflection (FTIR/ATR), thermogravimetric analysis (TGA/DTG), X-ray diffraction (XRD), optical microscopy (OM), Scanning electron microscopy (SEM), mechanical evaluations, and biodegradability assessments. The biodegradability of the obtained bioplastic samples was evaluated. Scanning electron microscopy (SEM) revealed strong interfacial adhesion between the constituent filler and the polymer matrix. Potato (Solanum tuberosum L.) seed production using the aeroponic system has been widely implemented in Indonesia. However, aeroponic systems in tropical areas such as Indonesia faced obstacles in the form of high wilt attacks caused by the bacteria Ralstonia solanacearum. This research aimed to control R. solanacearum wilt disease in an aeroponic system using various microbes. The research was carried out from September 2022 to January 2023. This research consisted of four stages, namely exploration and isolation of microorganisms, identification of microbial antagonism, testing of in vitro potential microbial antagonism and testing of selected microbial antagonism in the aeroponic system. The nutrients used in the aeroponic system contained R. solanacearum with a concentration of 104 CFU mL-1, while the concentration of the microbial solution used was 108 CFU mL-1. Pseudomonas fluorescence was consistently the best microbe both in vitro and in aeroponics, whereas Bacillus subtilis could only overcome wilt in vitro and could not overcome wilt in aeroponic system. Pseudomonas fluorescence required 5 min soaking time and could increase wilt with increasing soaking time. The consortium between B. subtilis and P. fluorescence with 10 min soaking time could overcome wilt by 85% and increase the number of tubers and tuber weight by 79% and 85%, respectively, so this prospective microbial consortium could be applied to aeroponic systems environmentally friendly in tropical areas with high R. solanacearum opportunities. The tomato potato psyllid, Bactericera cockerelli Šulc, originating from North and Central America, poses a serious threat to Solanaceae crops in Australia. This study investigates the potential of the Australian native and commercially available green lacewing, Mallada signatus Schneider, as a biological control agent for B. cockerelli. The effect of feeding on B. cockerelli on the development rate and survival, of M. signatus were evaluated. Further, a greenhouse cage study was conducted to determine the optimal density of M. signatus larvae needed to effectively control an established B. cockerelli population. In our study, the third instar larvae of M. signatus consumed over 75 B. cockerelli nymphs in 24 h. Following the introduction of M. signatus larvae to caged tomato plants, eight M. signatus larvae per caged tomato plant decreased B. cockerelli population by 64% at the end of the sampling. These results indicated M. signatus, particularly at the larval stage, is an effective biological control option for B. cockerelli, especially in greenhouse tomato cultivation. This research offers valuable insights for the Australian horticultural industry, presenting a viable, eco‐friendly alternative to traditional, chemical pesticide‐reliant pest management strategies. Summary: Potato flour/gelatine films in different ratios (6:0, 5:1, 4:1, 3:1, 2:1 and 1:1) were prepared and their properties were examined in this study. Potato flour film‐forming dispersion without gelatine showed the highest particle size and apparent viscosity. As the gelatine concentration increased, the composite films became smoother and more transparent. Gelatine incorporated with potato flour films reduced the retrogradation enthalpy and glass transition temperature, leading to the low tensile strength and high elongation at break. Although water content and water solubility index increased with the higher gelatine concentration, water vapour permeability of composite films decreased from 14.59 × 10−13 gm−1s−1Pa−1 to 5.83 × 10−13 gm−1s−1Pa−1. In addition, it was noted that water contact angle increased from 24.50° to 102.33° as gelatine concentration increased from 0% (6:0) to 50% (1:1). The genus Clavibacter (family Microbacteriaceae) includes gram‐positive actinobacterial species infecting diverse agricultural crops such as alfalfa, maize, pepper, potato, tomato and wheat. Clavibacter tessellarius, causing bacterial mosaic of wheat, was reported for the first time in 1976 in Nebraska (United States). Since then, the pathogen was thought to be restricted to North America (Canada and the United States). While bacterial strains suspected to be C. tessellarius have occasionally been isolated from wheat samples outside North America, occurrence of the pathogen in the Old World has not yet been confirmed by the plant protection authorities. In spring 2020 (January to May) dozens of wheat fields with young plants showing leaf chlorosis and mild mosaic symptoms were surveyed and sampled in southern Iran. Among 192 surveyed wheat fields, gram‐positive, orange‐pigmented actinobacterial strains were isolated from samples of 53 fields. Altogether, 61 bacterial strains were identified as C. tessellarius based on the phenotypic features, PCR‐based molecular tests and pathogenicity assays. Multilocus sequence analysis of five housekeeping genes (atpD, dnaK, gyrB, ppk and rpoB) using 10 representative strains from diverse geographic areas in southern Iran confirmed the phylogenetic status of the strains within C. tessellarius. Our data indicate that the bacterial mosaic pathogen occurs in four southern Iranian provinces including Bushehr, Fars, Kerman and Khuzestan. Thus, occurrence of C. tessellarius outside North America is confirmed. Virus diseases represent important economic threats to seed potato production worldwide, yet relatively little is known of their epidemiology at the landscape‐scale. In this study, data was compiled from the Scottish national seed potato classification scheme on the incidence of 10 different potato viruses for the years 2009–2022. A co‐occurrence analysis identified that 12 virus species pairs occurred together more often than expected by chance, and potato blackleg was positively associated with eight potato viruses. ArcGIS was used to investigate spatial and spatiotemporal variation in incidence rates of the three most prevalent viruses (potato virus Y, potato leaf roll virus and potato virus A), and this revealed prominent geographic differences in long‐term disease outcomes. Focusing on potato virus Y as the most commonly occurring single infection, interpretable machine‐learning techniques were used to investigate the influence of key crop, management and environmental factors on patterns of incidence in space and time. The results showed that health characteristics of seed stocks were among the most important predictors of incidence, along with blackleg infection, several management features, cultivar resistance, distance to the nearest seed and ware crop, temperature variables and several soil features. This approach provides a comprehensive overview of potato viruses in Scotland, a deeper understanding of epidemiological risk factors at the landscape‐scale and a forecast model that could serve as the basis of a decision support tool for improved management of potato virus Y. Plasmopara viticola is controlled by fungicides with different modes of action, which include zoxamide. Zoxamide was developed in 1998 by Dow AgroSciences LIC (Indianapolis, IN) and has been commercialized since 2001 by Rohm and Haas Company. This fungicide is highly effective against oomycetes, used for foliar application in vine crops, potato, and other vegetables to control oomycete-induced diseases. Zoxamide acts by causing mitotic arrest by binding to β-tubulin, inhibiting tubulin polymerization and cell division of the pathogen. In the past three decades, the management of fungicide resistance has emerged as a significant concern for both growers and regulatory authorities; this is primarily due to the substantial increase in resistance, in terms of occurrence and spread, towards most fungicide groups. The monitoring of the effectiveness of fungicides is the only way to accurately identify as soon as possible the onset of resistance. In this study, we were interested in tracking the changes in the sensitivity of zoxamide to P. viticola populations collected during a 6-year monitoring (2017–2022) in two Italian locations: the autonomous province of Trento and the autonomous region of Friuli-Venezia Giulia. Bioassays on leaf discs were carried out, and EC50 and MIC values were elaborated. From our results, zoxamide showed for a long period of time high sensitivity, but over the last years we observed a change, as in 2022 most of the samples tested in these two regions showed MIC > 100 mg L−1. Phytopathogenic bacteria of the genus Pectobacterium are responsible for several diseases that affect potato (Solanum tuberosum L.) production worldwide, including blackleg and tuber soft rot. These bacteria are highly diverse, with over 17 different species currently identified. However, some of the recently described species, such as Pectobacterium punjabense, are still poorly understood. In this study, we focused on P. punjabense isolates collected from diseased potato tubers in Russia in 2021. Whole-genome sequencing was used to characterise the genomic diversity of the pathogen and determine the biochemical profiles of the isolated bacteria. The ability of these isolates to cause soft rot symptoms was tested. A comparative assessment of the potential pathogenicity of the Pectobacterium isolates was conducted by infecting potato tubers and measuring the accumulation of biomass in a liquid medium during cultivation at different temperatures. A TaqMan qPCR assay was developed for the highly sensitive and specific characterisation of P. punjabense strains, which can be used in diagnostic systems. NHX proteins are transmembrane antiporters belonging to the cation/proton antiporter gene family, with a conserved Na+ (K+)/H+ exchange (PF00999) protein domain. NHXs play a prominent role in plant growth, development, and defense. However, the role of NHX gene family in potato (Solanum tuberosum L.) is yet to be known. In this study, we conducted a genome-wide analysis of the potatoNHX gene family. A total of 25StNHX family memberswere identified to be unevenly distributed on 10 chromosomes. The proteins ranged in length from 252 to 1,153 amino acids, with molecular masses ranging from 27516.32 to 127860.87 kD, and isoelectric points (pI) ranging from 4.96 to 9.3. Analyses of gene structures and conserved motifs indicated that StNHX genes in the same phylogenetic cluster are conserved. Phylogenetic analysis divided the StNHX genes into three subfamilies (Classes I, II, and III). Synteny analysis indicated that StNHX gene family Class III of NHX and all Arabidopsis thaliana NHXs shared a close evolutionary relationship. Analysis of cisacting elements in the upstream 1,500 bp promoter region of potato NHX genes showed that these genes could be regulated by light, stress, and hormones such as abscisic acid and gibberellic acid. Protein-protein interaction network analysis indicated that StNHX proteins may participate in the regulation of potato growth and stress response. Besides, To determine a potential role of these genes in tissue development and drought response, we analyzed the RNA-seq data of different DM potato tissues. The results showed that NHX genes exhibited distinct tissue-specific expression patterns. We further examined the expression patterns of StNHX in different tissues (leaves, roots, shoots, tubers, stolons, and flowers) during the flowering stage in 'Jizhangshu NO.8.' potato. The qRT-PCR results further confirmed the importance of StNHX genes in potato plant growth and development. We further analyzed the RNA-seq data (DM potato) under different abiotic stresses (salt, drought, and heat), and found that the expression of StNHX genes was induced under abiotic stress. qRT-PCR analysis of shoots and roots of 'Jizhangshu NO.8.' potato treated for 0, 6, 12, and 24 hwith 15% PEG6000 confirmed that the 25 StNHX genes are involved in the response to drought stress in potato. The results of this study may be useful for selecting appropriate candidate genes for the breeding of new drought-tolerant potato varieties. The study assesses the gender implications of a target product profile for table potato in Kenya. Breeding programs mostly emphasize farmers’ trait requirements and rarely pay attention to other value chain actors’ preferences. This partially contributes to the low uptake of improved varieties. Therefore, efforts are required to assess the gender implications of crop product profile proposals during development, testing, and dissemination. In this paper, we assess the gender implications of traits targeted by a potato breeding program in Kenya using the G+ tools. The study applies the G+ product profile tool to examine the instances the selected traits may exacerbate gender disparity along four domains: unpaid labor, access to employment opportunities, requiring extra inputs and control over benefits. We use mixed methods including a review by social scientists, key informant interviews and a multistakeholder workshop to gather insights from female and male farmers, breeders, and other stakeholders. Findings show that pest, disease resistance, and shelf-life traits benefit women and men in the target customer segments. On the other hand, earliness, dry matter, and yield could increase drudgery for women. The traits increase demand for women’s unpaid labor during harvesting, sorting and food preparation time while also displacing women from profitable nodes. We recommend that gender-responsive strategies accompany the release of the target variety to mitigate inequities and enhance adoption. [ABSTRACT FROM AUTHOR]

Plants are sessile by nature, and as such they have evolved to sense changes in seasonality and their surrounding environment, and adapt to these changes. One prime example of this is the regulation of flowering time in angiosperms, which is precisely timed by the coordinated action of two proteins: FLOWERING LOCUS T (FT) and TERMINAL FLOWER 1 (TFL1). Both of these regulators are members of the PHOSPHATIDYLETHANOLAMINE BINDING PROTEIN (PEBP) family of proteins. These regulatory proteins do not interact with DNA themselves, but instead interact with transcriptional regulators, such as FLOWERING LOCUS D (FD). FT and TFL1 were initially identified as key regulators of flowering time, acting through binding with FD; however, PEBP family members are also involved in shaping plant architecture and development. In addition, PEBPs can interact with TCP transcriptional regulators, such as TEOSINTE BRANCHED 1 (TB1), a well-known regulator of plant architecture, and key domestication-related genes in many crops. Here, we review the role of PEBPs in flowering time, plant architecture, and development. As these are also key yield-related traits, we highlight examples from the model plant Arabidopsis as well as important food and feed crops such as, rice, barley, wheat, tomato, and potato. Planting potatoes through plastic film with incomplete or excessive soil coverage over seed holes significantly impairs yield. Existing covering methods rely solely on mechanical transmissions, leading to bulky and inconsistent soil coverage of the seed holes. This paper reports an innovative method using a precise soil covering device based on the YOLOv4-tiny real-time object detection system to accurately identify potato plastic film holes and cover them with soil. The system adopts a lightweight and high-precision detection scheme, balancing increased network depth with reduced computation. It can identify holes in the plastic film in real-time and with high accuracy. To verify the effectiveness of YOLOv4-tiny real-time object detection system, a precise soil covering device based on this detection system has been designed and applied to a double crank multi-rod hill-drop planter. Field tests revealed that the system's average accuracy rate for detecting holes is approximately 98%, with an average processing time of 15.15 ms per frame. This fast and accurate performance, combined with the device's robust real-time operation and anti-interference capabilities during soil covering, effectively reduce the problems of soil cover omission and repeated covering caused by existing mechanical transmission methods. The CONSTANS-like (COL) gene plays important roles in plant growth, development, and abiotic stress. A total of 15 COL genes are unevenly distributed on eight chromosomes in the potato genome. The amino acid length of the family members was 347-453 aa, the molecular weight was 38.65-49.92 kD, and the isoelectric point was 5.13-6.09. The StCOL family can be divided into three subfamilies by evolutionary tree analysis, with conserved motifs and similar gene structure positions in each subfamily. The analysis of promoter cis-acting elements showed 17 cis-acting elements related to plant hormones, stress, and light response. Collinearity analysis of COL genes of tomato, potato, and Arabidopsis showed that 13 StCOL genes in the different species may have a common ancestor. A total of 10 conserved motifs and six kinds of post-translational modifications in the 15 StCOL proteins were identified. The 15 StCOL genes exhibit a genomic structure consisting of exons and introns, typically ranging from two to four in number. The results showed that 10 genes displayed significant expression across all potato tissues, while the remaining five genes were down-expressed in potato transcriptome data. The quantitative reverse transcription polymerase chain reaction (qRT-PCR) analysis exhibited differential expression of 8 StCOL genes in the potato leaves and tubers at different growth stages, as well as 7 StCOL genes under 2°C treatment conditions. Background: Cassava is one of three major potato crops and the sixth most important food crop globally. Improving yield remains a primary aim in cassava breeding. Notably, plant height significantly impacts the yield and quality of crops; however, the mechanisms underlying cassava plant height development are yet to be elucidated. Results: In this study, we investigated the mechanisms responsible for cassava plant height development using phenotypic, anatomical, and transcriptomic analyses. Phenotypic and anatomical analysis revealed that compared to the high-stem cassava cultivar, the dwarf-stem cassava cultivar exhibited a significant reduction in plant height and a notable increase in internode tissue xylem area. Meanwhile, physiological analysis demonstrated that the lignin content of dwarf cassava was significantly higher than that of high cassava. Notably, transcriptome analysis of internode tissues identified several differentially expressed genes involved in cell wall synthesis and expansion, plant hormone signal transduction, phenylpropanoid biosynthesis, and flavonoid biosynthesis between the two cassava cultivars. Conclusions: Our findings suggest that internode tissue cell division, secondary wall lignification, and hormone-related gene expression play important roles in cassava plant height development. Climate change is predicted to increase the occurrence of extreme weather events such as heatwaves, which may thereby impact the outcome of plant-herbivore interactions. While elevated temperature is known to directly affect herbivore growth, it remains largely unclear if it indirectly influences herbivore performance by affecting the host plant they feed on. In this study, we investigated how transient exposure to high temperature influences plant herbivory-induced defenses at the transcript and metabolic level. To this end, we studied the interaction between potato (Solanum tuberosum) plants and the larvae of the potato tuber moth (Phthorimaea operculella) under different temperature regimes. We found that P. operculella larvae grew heavier on leaves co-stressed by high temperature and insect herbivory than on leaves pre-stressed by herbivory alone. We also observed that high temperature treatments altered phylotranscriptomic patterns upon herbivory, which changed from an evolutionary hourglass pattern, in which transcriptomic responses at early and late time points after elicitation are more variable than the ones in the middle, to a vase pattern. Specifically, transcripts of many herbivory-induced genes in the early and late defense stage were suppressed by HT treatment, whereas those in the intermediate stage peaked earlier. Additionally, we observed that high temperature impaired the induction of jasmonates and defense compounds upon herbivory. Moreover, using jasmonate-reduced (JA-reduced, irAOC) and -elevated (JA-Ile-elevated, irCYP94B3s) potato plants, we showed that high temperature suppresses JA signaling mediated plant-induced defense to herbivore attack. Research on potato diseases had been widely reported, but a systematic review of potato diseases was lacking. Here, bibliometrics was used to systematically analyze the progress of potato disease. The publications related to "potato" and "disease" were searched in the Web of Science (WOS) from 2014 to 2023. The results showed that a total of 2095 publications on potato diseases were retrieved, with the annual publication output increasing year by year at a growth rate of 8.52%. The main countries where publications were issued were the United States, China, and India. There was relatively close cooperation observed between China, the United States, and the United Kingdom in terms of international collaboration, while international cooperation by India was less extensive. Based on citation analysis and trending topics, potential future research directions include nanoparticles, which provides highly effective carriers for biologically active substances due to their small dimensions, extensive surface area, and numerous binding sites; machine learning, which facilitates rapid identification of relevant targets in extensive datasets, thereby accelerating the process of disease diagnosis and fungicide innovation; and synthetic communities composed of various functional microorganisms, which demonstrate more stable effects in disease prevention and control. One of the factors precluding potato juice application in the food industry is its propensity for enzymatic browning. The addition of fruit and vegetable concentrates rich in organic acids can reduce unfavorable changes in the color of potato juices and influence the content of biologically active compounds. This study aimed to investigate the effect of the addition of natural fruit and vegetable concentrates to potato juices on their color and on the composition and contents of anthocyanin pigments isolated from them. The color, composition and amount of anthocyanins in potato juices and dyes were determined using HPLC-DAD and UHPLC MS/MS liquid chromatography. It was found that the juices without concentrate addition had, on average, 61% lower total polyphenol content and 63% lower anthocyanin content compared with the juices with added concentrates. The isolated pigments contained, on average, 30–40 times more anthocyanins compared with those isolated from the non-purified juices. Furthermore, the addition of concentrates enabled ca. 3–5 times more effective isolation of anthocyanins compared with the samples without these additives. This study evaluated the residue behavior and dissipation dynamics of a new imidacloprid FS 600 seed treatment in potato cultivation systems in Shandong and Jilin, China. Sensitive and accurate UPLC-MS/MS methods were established to quantify imidacloprid residues in potatoes, potato plants, and soil. Results showed that imidacloprid dissipation followed a first-order kinetic model, with half-lives ranging from 6.9 to 26.7 days in plants and 19.8 to 28.9 days in soil. At harvest, the highest average residues in potatoes and soil were 0.778 mg/kg and 0.149 mg/kg, respectively. The dietary risk assessment indicated a chronic risk quotient (CRQ) of 39.73% for adults, indicating minimal risk to human consumers, while the ecological risk quotient (ERQ) and ecotoxicity exposure ratio (TER) revealed low to moderate toxicity to earthworms, warranting caution in the use of this formulation. AbstractProcessed plant-based foods, particularly high carbohydrate-containing foods, are among the greatest contributors to dietary acrylamide, a probable human carcinogen, uptake. Between 2009 and 2020, five surveys were conducted to determine acrylamide in high carbohydrate-containing foods in Canada. These surveys included sampling of potato and sweet potato chips, French fries, and frozen potato/sweet potato products, as a follow-up to our earlier surveys from 2002 - 2008. Samples were analyzed using isotope dilution (13C3-acrylamide) with LC-MS/MS. The highest mean acrylamide levels were found in sweet potato chips. Among potato chips (57 to 4660 ng g−1), one brand consistently showed the highest concentrations with wide variability. Acrylamide concentrations decreased over time in ready-to-eat French fries (from 480 to 358 ng g−1), and one brand showed a clear reduction temporally. Wide variations were observed among brands, among lots/outlets of same brands, and among different food chains. Acrylamide levels in potato chips decreased between 2009 and 2016 (504.3 ng g−1) relative to the period 2002 - 2008 (1096.9 ng g−1). Summary: Potato (Solanum tuberosum) is the fourth largest food crop in the world. Late blight, caused by oomycete Phytophthora infestans, is the most devastating disease threatening potato production. Previous research has shown that StRFP1, a potato Arabidopsis Tóxicos en Levadura (ATL) family protein, positively regulates late blight resistance via its E3 ligase activity. However, the underlying mechanism is unknown.Here, we reveal that StRFP1 is associated with the plasma membrane (PM) and undergoes constitutive endocytic trafficking. Its PM localization is essential for inhibiting P. infestans colonization. Through in vivo and in vitro assays, we investigated that StRFP1 interacts with two sugar transporters StSWEET10c and StSWEET11 at the PM.Overexpression (OE) of StSWEET10c or StSWEET11 enhances P. infestans colonization. Both StSWEET10c and StSWEET11 exhibit sucrose transport ability in yeast, and OE of StSWEET10c leads to an increased sucrose content in the apoplastic fluid of potato leaves. StRFP1 ubiquitinates StSWEET10c and StSWEET11 to promote their degradation.We illustrate a novel mechanism by which a potato ATL protein enhances disease resistance by degrading susceptibility (S) factors, such as Sugars Will Eventually be Exported Transporters (SWEETs). As one of the most important food crops, the potato is widely planted in the oasis agricultural region of Northwest China. To ascertain the impact of regulated deficit irrigation (RDI) on various facets including dry matter accumulation, tuber yield, quality and water use efficiency (WUE) of potato plants, a two-growth season field experiment under mulched drip irrigation was conducted in the desert oasis region of Northwest China. Water deficits, applied at the seedling, tuber formation, tuber expansion and starch accumulation stages, encompassed two distinctive levels: mild (55–65% of field capacity, FC) and moderate (45–55% FC) deficit, with full irrigation (65–75% FC) throughout the growing season as the control (CK). The results showed that water deficit significantly reduced (p < 0.05) above-ground dry matter, water consumption and tuber yield compared to CK, and the reduction increased with the increasing water deficit. A mild water deficit at the tuber formation stage, without significantly reducing (p > 0.05) yield, could significantly increase WUE and irrigation water use efficiency (IWUE), with two-year average increases of 25.55% and 32.33%, respectively, compared to CK. Water deficit at the tuber formation stage increased starch content, whereas water deficit at tuber expansion stage significantly reduced starch, protein and reducing sugar content. Additionally, a comprehensive evaluation showed that a mild water deficit at the tuber formation stage is the optimal RDI strategy for potato production, providing a good balance between yield, quality and WUE. Groundwater resources serve as the primary source of water in the agro-pastoral ecotone of northern China, where scarcity of water resources constrains the development of agriculture and animal husbandry. As a typical rainfed agricultural area, the agro-pastoral ecotone in Inner Mongolia is entirely dependent on groundwater for agricultural irrigation. Due to the substantial groundwater consumption of irrigated farmland, groundwater levels have been progressively declining. To obtain a sustainable irrigation pattern that significantly conserves water, this study faces the challenge of unclear water transport relationships among water, soil, and crops, undefined water cycle mechanism in typical irrigation units, and water use efficiency, which was not assessed. Therefore, this paper, based on in situ experimental observations and daily meteorological data in 2022–2023, utilized the DSSAT model to explore the growth processes of potato, oat, alfalfa, and sunflower, the soil water dynamics, the water balance, and water use efficiency, analyzed over a typical irrigation area. The results indicated that the simulation accuracy of the DSSAT model was ARE < 10%, nRMSE/% < 10%, and R2 ≥ 0.85. The consumption of the soil moisture during the rapid growth stage for the potatoes, oats, alfalfa, and sunflower was 7–13% more than that during the other periods, and the yield was 67,170, 3345, 6529, and 4020 kg/ha, respectively. The soil evaporation of oat, potato, alfalfa, and sunflower accounted for 18–22%, 78–82%; 57–68%, and 32–43%, and transpiration accounted for 40–44%, 56–60%, 45–47%, and 53–55% of ETa (333.8 mm–369.2 mm, 375.2 mm–414.2 mm, 415.7 mm–453.7 mm, and 355.0 mm–385.6 mm), respectively. It was advised that irrigation water could be appropriately reduced to decrease ineffective water consumption. The water use efficiency and irrigation water use efficiency for potatoes was at the maximum amount, ranging from 16.22 to 16.62 kg/m3 and 8.61 to 10.81 kg/m3, respectively, followed by alfalfa, sunflowers, and oats. For the perspective of water productivity, it was recommended that potatoes could be extensively cultivated, alfalfa planted appropriately, and oats and sunflowers planted less. Hyperspectral imaging has emerged as a pivotal technology in agricultural research, offering a powerful means to non-invasively monitor stress factors, such as drought, in crops like potato plants. In this context, the integration of attention-based deep learning models presents a promising avenue for enhancing the efficiency of stress detection, by enabling the identification of meaningful spectral channels. This study assesses the performance of deep learning models on two potato plant cultivars exposed to water-deficient conditions. It explores how various sampling strategies and biases impact the classification metrics by using a dual-sensor hyperspectral imaging systems (VNIR -Visible and Near-Infrared and SWIR—Short-Wave Infrared). Moreover, it focuses on pinpointing crucial wavelengths within the concatenated images indicative of water-deficient conditions. The proposed deep learning model yields encouraging results. In the context of binary classification, it achieved an area under the receiver operating characteristic curve (AUC-ROC—Area Under the Receiver Operating Characteristic Curve) of 0.74 (95% CI: 0.70, 0.78) and 0.64 (95% CI: 0.56, 0.69) for the KIS Krka and KIS Savinja varieties, respectively. Moreover, the corresponding F1 scores were 0.67 (95% CI: 0.64, 0.71) and 0.63 (95% CI: 0.56, 0.68). An evaluation of the performance of the datasets with deliberately introduced biases consistently demonstrated superior results in comparison to their non-biased equivalents. Notably, the ROC-AUC values exhibited significant improvements, registering a maximum increase of 10.8% for KIS Krka and 18.9% for KIS Savinja. The wavelengths of greatest significance were observed in the ranges of 475–580 nm, 660–730 nm, 940–970 nm, 1420–1510 nm, 1875–2040 nm, and 2350–2480 nm. These findings suggest that discerning between the two treatments is attainable, despite the absence of prominently manifested symptoms of drought stress in either cultivar through visual observation. Introduction: Potatoes are one of the world's most important agricultural crops, with potential for making a major contribution to global food security. This study shows how a biostimulant derived from a plant extract can improve potato crop yield and global food supply. Successful potato production currently requires significant levels of inputs including fertiliser, pesticides and irrigation, however non-microbial plant biostimulants or mixtures of biostimulants with synergistic actions, have the capacity to reduce inputs and improve the sustainability of intensive agriculture. Methods: A complex biostimulant containing a number of flavonoids including protocatechuic acid, quercetin, chlorogenic acid, coumaroyl quinic acid and gentistic acid was tested against three potato varieties for its efficacy in improving plant growth characteristics and tuber production in controlled and field environments. Results: In containers, complex biostimulant treatment enhanced photosynthetic ability, with elevated levels of chlorophyll, higher specific leaf areas and significantly larger leaf assimilation areas. Treatment also significantly increased tuber yield by an average of 33% in tuber weight across three potato varieties and shifted tuber production toward larger sized tubers. The biostimulant derived from flavonoids was also assessed in 6 commercial potato crops and consistently increased total yield (average 5.2%) and marketable yield, representing an increase in margins of UK£700 per hectare across the 6 crops. Similar increases in yield were seen when comparing chitted and unchitted seed potatoes and both types of seed responded positively to biostimulant application. Biostimulant treatment had no significant effects on tuber specific gravity, dry matter percentage and starch content, except at one location where these quality characteristics were higher in the control plants. Conclusion: The flavonoid based complex biostimulant produced significant effects on potato yield and quality in both container experiments and in field trials indicating its potential for contributing to sustainable potato production. A total of 3,860 accessions from the global in trust clonal potato germplasm collection w3ere genotyped with the Illumina Infinium SolCAP V2 12K potato SNP array to evaluate genetic diversity and population structure within the potato germplasm collection. Diploid, triploid, tetraploid, and pentaploid accessions were included representing the cultivated potato taxa. Heterozygosity ranged from 9.7% to 66.6% increasing with ploidy level with an average heterozygosity of 33.5%. Identity, relatedness, and ancestry were evaluated using hierarchal clustering and model-based Bayesian admixture analyses. Errors in genetic identity were revealed in a side-by-side comparison of in vitro clonal material with the original mother plants revealing mistakes putatively occurring during decades of processing and handling. A phylogeny was constructed to evaluate inter- and intraspecific relationships which together with a STRUCTURE analysis supported both commonly used treatments of potato taxonomy. Accessions generally clustered based on taxonomic and ploidy classifications with some exceptions but did not consistently cluster by geographic origin. STRUCTURE analysis identified putative hybrids and suggested six genetic clusters in the cultivated potato collection with extensive gene flow occurring among the potato populations, implying most populations readily shared alleles and that introgression is common in potato. Solanum tuberosum subsp. andigena (ADG) and S. curtilobum (CUR) displayed significant admixture. ADG likely has extensive admixture due to its broad geographic distribution. Solanum phureja (PHU), Solanum chaucha (CHA)/Solanum stenotomum subsp. stenotomum (STN), and Solanum tuberosum subsp. tuberosum (TBR) populations had less admixture from an accession/population perspective relative to the species evaluated. A core and mini core subset from the genebank material was also constructed. SNP genotyping was also carried out on 745 accessions from the Seed Savers potato collection which confirmed no genetic duplication between the two potato collections, suggesting that the collections hold very different genetic resources of potato. AbstractPotato (Solanum tuberosum L.) is one of the most important food crops in Korea. In July 2021 and 2022, dark black-rot symptoms with pink tinges were observed on field-grown potato tubers in Hongsung and Chuncheon, Korea, respectively. We obtained four isolates (HSv05 and HSv10 from Hongsung, and CCp03 and CCp05 from Chuncheon) from diseased tubers and identified these isolates as Pythium aphanidermatum by analyzing the sequences of internal transcribed spacer rDNA region and mitochondrial cytochrome c oxidase subunit II (COX2) mtDNA gene. Additionally, we compared the cultural and morphological characteristics of these four isolates with those of the reference isolate KACC 48066 of P. aphanidermatum and the literature. Further, we tested the pathogenicity of all these isolates against potato tubers. The cultural and morphological characteristics of the four test isolates were similar to those of the reference isolate and the literature; all four test isolates proved pathogenic to potato tubers. Therefore, we concluded that P. aphanidermatum is the causal agent of potato leak and this is the first report of the disease on potato in Korea. [ABSTRACT FROM AUTHOR]

Bacterial soft rot caused by coinfection with Dickeya spp. and Pectobacterium spp. in hosts can cause successive changes in fields, and it is difficult to prevent the spread of and control the infection. Pectobacterium spp. are prevalent in the growing areas of tuberous crops, including taro and potato. Recently, Dickeya fangzhongdai has emerged as a virulent pathogen in taro. To determine the prevalence status of the causal agents and evaluate the potential spreading risks of D. fangzhongdai, screening and taxonomic classification were performed on phytopathogenic bacteria collected from different taro-growing areas in Guangdong Province, China, and biological and genomic characteristics were further compared among typical strains from all defined species. The causative agents were verified to be phytobacterial strains of D. fangzhongdai, Pectobacterium aroidearum and Pectobacterium colocasium. P. aroidearum and P. colocasium were found to form a complex preferring Araceae plants and show intensive genomic differentiation, indicating their ancestor had adapted to taro a long time prior. Compared with Pectobacterium spp., D. fangzhongdai was more virulent to taro corms under conditions of exogenous infection and more adaptable at elevated temperatures. D. fangzhongdai strains isolated from taro possessed genomic components of additional T4SSs, which were accompanied by additional copies of the hcp-vgrG genes of the T6SS, and these contributed to the expansion of their genomes. More gene clusters encoding secondary metabolites were found within the D. fangzhongdai strains than within the Pectobacterium complex; interestingly, distinct gene clusters encoding zeamine and arylpolyene were both most similar to those in D. solani that caused potato soft rot. These comparisons provided genomic evidences for that the newly emerging pathogen was potentially equipped to compete with other pathogens. Diagnostic qPCR verified that D. fangzhongdai was prevalent in most of the taro-growing areas and coexisted with the Pectobacterium complex, while the plants enriching D. fangzhongdai were frequently symptomatic at developing corms and adjacent pseudostems and caused severe symptoms. Thus, the emerging need for intensive monitoring on D. fangzhongdai to prevent it from spreading to other taro-growing areas and to other tuberous crops like potato; the adjustment of control strategies based on different pathopoiesis characteristics is recommended. Background: The dirigent (DIR) genes encode proteins that act as crucial regulators of plant lignin biosynthesis. In Solanaceae species, members of the DIR gene family are intricately related to plant growth and development, playing a key role in responding to various biotic and abiotic stresses. It will be of great application significance to analyze the DIR gene family and expression profile under various pathogen stresses in Solanaceae species. Results: A total of 57 tobacco NtDIRs and 33 potato StDIRs were identified based on their respective genome sequences. Phylogenetic analysis of DIR genes in tobacco, potato, eggplant and Arabidopsis thaliana revealed three distinct subgroups (DIR-a, DIR-b/d and DIR-e). Gene structure and conserved motif analysis showed that a high degree of conservation in both exon/intron organization and protein motifs among tobacco and potato DIR genes, especially within members of the same subfamily. Total 8 pairs of tandem duplication genes (3 pairs in tobacco, 5 pairs in potato) and 13 pairs of segmental duplication genes (6 pairs in tobacco, 7 pairs in potato) were identified based on the analysis of gene duplication events. Cis-regulatory elements of the DIR promoters participated in hormone response, stress responses, circadian control, endosperm expression, and meristem expression. Transcriptomic data analysis under biotic stress revealed diverse response patterns among DIR gene family members to pathogens, indicating their functional divergence. After 96 h post-inoculation with Ralstonia solanacearum L. (Ras), tobacco seedlings exhibited typical symptoms of tobacco bacterial wilt. The qRT-PCR analysis of 11 selected NtDIR genes displayed differential expression pattern in response to the bacterial pathogen Ras infection. Using line 392278 of potato as material, typical symptoms of potato late blight manifested on the seedling leaves under Phytophthora infestans infection. The qRT-PCR analysis of 5 selected StDIR genes showed up-regulation in response to pathogen infection. Notably, three clustered genes (NtDIR2, NtDIR4, StDIR3) exhibited a robust response to pathogen infection, highlighting their essential roles in disease resistance. Conclusion: The genome-wide identification, evolutionary analysis, and expression profiling of DIR genes in response to various pathogen infection in tobacco and potato have provided valuable insights into the roles of these genes under various stress conditions. Root-knot nematodes (RKNs) are a vital pest that causes significant yield losses and economic damage to potato plants. The use of chemical pesticides to control these nematodes has led to environmental concerns and the development of resistance in the nematode populations. Endophytic fungi offer an eco-friendly alternative to control these pests and produce secondary metabolites that have nematicidal activity against RKNs. The objective of this study is to assess the efficacy of Aspergillus flavus (ON146363), an entophyte fungus isolated from Trigonella foenum-graecum seeds, against Meloidogyne incognita in filtered culture broth using GC–MS analysis. Among them, various nematicidal secondary metabolites were produced: Gadoleic acid, Oleic acid di-ethanolamide, Oleic acid, and Palmitic acid. In addition, biochemical compounds such as Gallic acid, Catechin, Protocatechuic acid, Esculatin, Vanillic acid, Pyrocatechol, Coumarine, Cinnamic acid, 4, 3-indol butyl acetic acid and Naphthyl acetic acid by HPLC. The fungus was identified through morphological and molecular analysis, including ITS 1–4 regions of ribosomal DNA. In vitro experiments showed that culture filtrate of A. flavus had a variable effect on reducing the number of egg hatchings and larval mortality, with higher concentrations showing greater efficacy than Abamectin. The fungus inhibited the development and multiplication of M. incognita in potato plants, reducing the number of galls and eggs by 90% and 89%, respectively. A. flavus increased the activity of defense-related enzymes Chitinas, Catalyse, and Peroxidase after 15, 45, and 60 days. Leaching of the concentrated culture significantly reduced the second juveniles' stage to 97% /250 g soil and decreased the penetration of nematodes into the roots. A. flavus cultural filtrates via soil spraying improved seedling growth and reduced nematode propagation, resulting in systemic resistance to nematode infection. Therefore, A. flavus can be an effective biological control agent for root-knot nematodes in potato plants. Late blight is a serious disease of potato worldwide. Our study aimed to unveil genes involved in late blight resistance in potato by RNA-seq analysis after artificial inoculation under controlled conditions. In this study, two potato somatic hybrids (P7 and Crd6) and three varieties such as Kufri Girdhari, Kufri Jyoti and Kufri Bahar (control) were used. Transcriptiome analysis revealed statistically significant (p < 0.05) differentially expressed genes (DEGs), which were analysed into up-regulated and down-regulated genes. Further, DEGs were functionally characterized by the Gene Ontology annotations and the Kyoto Encyclopedia of Genes and Genomes pathways. Overall, some of the up-regulated genes in resistant genotypes were disease resistance proteins such as CC-NBS-LRR resistance protein, ankyrin repeat family protein, cytochrome P450, leucine-rich repeat family protein/protein kinase family, and MYB transcription factor. Sequence diversity analysis based on 38 peptide sequences representing 18 genes showed distinct variation and the presence of three motifs in 15 amino acid sequences. Selected genes were also validated by real-time quantitative polymerase chain reaction analysis. Interestingly, gene expression markers were developed for late blight resistant genotypes. Growth-regulating factors (GRFs) are transcription factors that play a pivotal role in plant growth and development. This study identifies 12 Solanum tuberosum GRF transcription factors (StGRFs) and analyzes their physicochemical properties, phylogenetic relationships, gene structures and gene expression patterns using bioinformatics. The StGRFs exhibit a length range of 266 to 599 amino acids, with a molecular weight of 26.02 to 64.52 kDa. The majority of StGRFs possess three introns. The promoter regions contain a plethora of cis-acting elements related to plant growth and development, as well as environmental stress and hormone response. All the members of the StGRF family contain conserved WRC and QLQ domains, with the sequences of these two conserved domain modules exhibiting high levels of conservation. Transcriptomic data indicates that StGRFs play a significant role in the growth and development of stamens, roots, young tubers, and other tissues or organs in potatoes. Furthermore, a few StGRFs exhibit differential expression patterns in response to Phytophthora infestans, chemical elicitors, heat, salt, and drought stresses, as well as multiple hormone treatments. The results of the expression analysis indicate that StGRF1, StGRF2, StGRF5, StGRF7, StGRF10 and StGRF12 are involved in the process of tuber sprouting, while StGRF4 and StGRF9 may play a role in tuber dormancy. Phosphorus deficiency and aluminum toxicity in acidic soils are important factors that limit crop yield. To further explore this issue, we identified 18 members of the StPHR gene family in the potato genome in this study. Through bioinformatics analysis, we found that the StPHR1 gene, an important member of this family, exhibited high expression levels in potato roots, particularly under conditions of phosphorus deficiency and aluminum toxicity stress. This suggested that the StPHR1 gene may play a crucial regulatory role in potato's resistance to phosphorus deficiency and aluminum toxicity. To validate this hypothesis, we conducted a series of experiments on the StPHR1 gene, including subcellular localization, GUS staining for tissue expression, heterologous overexpression, yeast two-hybrid hybridization, and bimolecular fluorescence complementation (BiFC). The results demonstrated that the StPHR1 gene is highly conserved in plants and is localized in the nucleus of potato cells. The heterologous overexpression of the gene in Arabidopsis plants resulted in a growth phenotype that exhibited resistance to both aluminum toxicity and phosphorus deficiency. Moreover, the heterologous overexpressing plants showed reduced aluminum content in the root system compared to the control group. Furthermore, we also identified an interaction between StPHR1 and StALMT6. These results highlight the potential application of regulating the expression of the StPHR1 gene in potato production to enhance its adaptation to the dual stress of phosphorus deficiency and high aluminum toxicity in acidic soils. The fungal pathogen Calonectria pseudonaviculata causes boxwood blight and is a significant threat to the boxwood industry, as well as historic boxwood gardens. The pathogen produces conidia in sticky masses that are splash dispersed, which germinate and infect through stomata on the leaves or stems, causing leaf spots and stem lesions. Despite its ability to cause severe infections on boxwood plants, the pathogen often has a low germination rate on artificial media under lab conditions. To identify cues that stimulate germination, we explored whether host factors could induce high germination rates. In this study, we demonstrate that C. pseudonaviculata spores achieve high germination rales when they are placed on detached leaves of boxwood and other known hosts, compared to potato dextrose agar and glass coverslips. We also demonstrate that gennination is induced by volatiles from detached leaves of boxwood, as well as the nonhost Berberis tlumbergii. When C. pseudonaviculata spores were exposed to volatiles from boxwood leaves in the presence of ethylene scrubber packs that contained potassium permanganate, the stimulatory effect on spore germination was reduced. However, ethylene, a regulator of leaf senescence, did not stimulate germination of C. pseudonaviculata spores. This suggests that the pathogen may have evolved to recognize one or more host volatiles, other than ethylene to induce germination, thus limiting its growth until it senses the presence of a host plant. The unculturable bacterium 'Candidatus Liberibacter soianacearum' (CLso) is responsible for a growing number of emerging crop diseases. However, we know little about the diversity and ecology of CLso and its psyllid vectors outside of agricultural systems, which limits our ability to manage crop disease and understand the impacts this pathogen may have on wild plants in natural ecosystems. In North America, CLso is transmitted to crops by the native potato psyllid (Bactericera cockerelli). However, the geographic and host plant range of the potato psyllid and CLso beyond the borders of agriculture are not well understood. A recent study of historic herbarium specimens revealed that a unique haplotype of CLso was present infecting populations of the native perennial Solanum umbelliferum in California decades before CLso was first detected in crops. We hypothesized that this haplotype and other potentially novel CLso variants arc still present in S. umbelliferum populations. To test this, we surveyed populations of S'. umbelliferum in Southern California for CLso and potato psyllid vectors. We found multiple haplotypes of CLso and the potato psyllid associated with these populations, with none of these genetic variants having been previously reported in California crops. These results suggest that CLso and its psyllid vectors are much more widespread and diverse in North American natural plant communities than suggested by data collected solely from crops and weeds in agricultural fields. Further characterization of these apparently asymptomatic haplotypes will facilitate comparison with disease-causing variants and provide insights into the continued emergence and spread of CLso. Late blight, caused by the notorious pathogen Phytophthora infestons, poses a significant threat to potato (Solanum tuberosum) crops worldwide, impacting their quality as well as yield. Here, we aimed to investigate the potential use of cinnamaldéhyde, carvacrol, and eugenol as control agents against P. infestons and to elucidate their underlying mechanisms of action. To determine the pathogen-inhibiting concentrations of these three plant essential oils (PEOs), a comprehensive evaluation of their effects using gradient dilution, mycelial growth rate, and spore germination methods was carried out. Cinnamaldéhyde, carvacrol, and eugenol were capable of significantly inhibiting P. infestons by hindering its mycelial radial growth, zoospore release, and sporangium germination; the median effective inhibitory concentration of the three PEOs was 23.87. 8.66, and 89.65 pl/liter, respectively. Scanning electron microscopy revealed that PEOs caused the irreversible deformation of P. infestons, resulting in hyphal shrinkage, distortion, and breakage. Moreover, propidium iodide staining and extracellular conductivity measurements demonstrated that all three PEOs significantly impaired the integrity and permeability of the pathogen's ceil membrane in a time- and dose-dependent manner. In vivo experiments confirmed the dose-dependent efficacy of PEOs in reducing the lesion diameter of potato late blight. Altogether, these findings provide valuable insight into the antifungal mechanisms of PEOs vis-à-vis late blight-causing P. infestons. By utilizing the inherent capabilities of these natural compounds, we could effectively limit the harmful impacts of late blight on potato crops, thereby enhancing agricultural practices and ensuring the resilience of global potato food production. Summary: Potato production in East Africa is seriously impacted by the potato cyst nematode (PCN), Globodera rostochiensis , where it has been recorded in at least three countries. In Kenya, it is widespread in all major potato-growing regions, often at very high densities. Consecutive cropping of potato on the same land and a sub-tropical climate have influenced PCN biology. For example, unusually large cysts have been regularly recovered. We have analysed the biological properties of these 'giant cysts'. The giant cysts contained more eggs than those recovered from UK fields. Egg size did not differ from UK populations and there was no difference in overall lipid content or lipid profile in J2 from giant cysts, compared to control samples. The nematodes in giant cysts were also genetically indistinguishable from any other G. rostochiensis sampled. When grown under UK glasshouse conditions, the offspring of nematodes from giant cysts were no different in size from those grown from control cysts, indicating that gigantism is not a heritable trait and may simply reflect favourable conditions for PCN under Kenyan farming systems. To date, all the PCN tested from Kenya, including those from giant cysts, are avirulent on potato cultivars containing the H 1 resistance gene. [ABSTRACT FROM AUTHOR]

Altitude is an important ecological factor affecting plant physiology and ecology, material metabolism and gene expression. Tuber color changes were observed in purple and red potatoes growing at four different elevations ranging from 1,800 ± 50 to 3,300 ± 50 m in the Tiger Leaping Gorge area of Yunnan Province. The results showed that the total phenol content, total flavone content, total anthocyanin content and biological yield of anthocyanin increased with increasing altitude until 2,800 ± 50 m, and the highest anthocyanin content were detected in the purple potato Huaxinyangyu and the red potato Jianchuanhong at the flowering stage and budding stage, respectively. Combined transcriptomic and metabolomic analyses revealed that the content and diversity of flavonoids are associated with genes expression via the promotion of propane metabolism to improve potato adaptation to different altitudes. Potato production systems present various phytosanitary problems. Among these, potato early dying (PED) caused by Verticillium spp. is a disease that is difficult to detect in its early stages and whose expression occurs in critical growing phases of the crop, such as tuber filling, generating a high economic impact. The objective of this work was to use spectral data to classify potato plants and identify the degree of severity of PED using spectral signatures and multispectral images captured on potato plants under greenhouse and commercial production conditions. Methods such as principal component analysis (PCA), random forest (RF), support vector machine (SVM), and artificial neural network (ANN) algorithms were implemented. All algorithms performed well; however, the RF was more accurate after iteration. The RF had a good capacity for indirect detection of PED, with an average accuracy of 60.9%. The wavelengths related to the red and red edges, especially from 710 to 735 nm, proved to be highly informative. As a result of the congruence between field and greenhouse data, the RECI, NDRE, VWI, and GRVI spectral indices were consistent with the discrimination of symptoms and PED severity levels. Identified wavelengths can be applied in the design of optical sensors that, together with the use of ML algorithms, can be implemented in the remote detection of early death in potato crops. Potatoes are widely planted worldwide and are the third most important food crop. Mutation breeding involves the artificial use of various physical and chemical factors to induce plants to produce new genotypes. In this study, we established a potato radiation mutation system and analysed the features of a radiation mutant. The LD50 of the potato callus was 24.8 Gy after linear regression analysis. The radiation mutant 1 (RM1) showed significant dwarfism and strong growth; RM1 plants decreased in height by 31%. The root and leaf fresh weights of the RM1 increased 1.8-fold compared to the wild-type (WT, Chuanyu 10) cultivar. Leaf microstructure results showed that the thicknesses of the upper epidermis and lower epidermis of RM1 plants were greater than those of the WT cultivar. Transcriptome analysis of seeding revealed 1179 upregulated and 1641 downregulated differentially expressed genes (DEGs) in RM1 plants. Further analysis showed that the expression levels of WRKY and MYB transcription factors, CIPK and MAPK protein kinases, ABC transporters, hormones, and ROS pathway genes were altered. The aim of this study was to compare Fe, Zn, Mn, Al and Li contents in table potato tubers, obtained from seven potato cultivars with different flesh colour, and one cultivar characterised by light-coloured flesh. The study material consisted of potato tubers produced in a one-factor field experiment carried out at the Agricultural Experimental Station in Zawady belonging to the University of Siedlce, Poland (52°03′ N, 22°33′ E) in 2021–2023. The following cultivars were tested: Eurostar (light yellow flesh), Rote Emmalie and Herbie 26 (red flesh), Provita, Salad Blue, Blaue Annelise, Vitelotte Noire and Bora Valley (purple and blue flesh). The study showed that the genetic characteristics of the cultivars determined the nutrient content and uptake with tuber yield. Of the tested minerals which are important for human nutrition (Fe, Zn, Mn), red and purple cultivars accumulated more Fe than the light yellow-fleshed Eurostar. Zn content was the highest in red-fleshed cultivars compared with light and purple-fleshed cultivars. Mn content was the highest in the tubers of the light yellow-fleshed Eurostar and lowest in red Rote Emmalie. Fe, Zn, Mn and Al contents and uptake with tuber yield were influenced by weather conditions during the growing season. [ABSTRACT FROM AUTHOR]
