Piwi proteins are important for germ cell development in most animals. These proteins are guided to specific targets by small guide RNAs, referred to as piRNAs or 21U RNAs in Caenorhabditis elegans. In this organism, even though genetic screens have uncovered 21U RNA biogenesis factors, little is known about how these factors interact or what they do. Based on the previously identified 21U biogenesis factor PID-1 (piRNA-induced silencing-defective 1), we here define a novel protein complex, PETISCO (PID-3, ERH-2, TOFU-6, and IFE-3 small RNA complex), that is required for 21U RNA biogenesis. PETISCO contains both potential 5′ cap and 5′ phosphate RNA-binding domains and interacts with capped 21U precursor RNA. We resolved the architecture of PETISCO and revealed a second function for PETISCO in embryonic development. This essential function of PETISCO is mediated not by PID-1 but by the novel protein TOST-1 (twenty-one U pathway antagonist). In contrast, TOST-1 is not essential for 21U RNA biogenesis. Both PID-1 and TOST-1 interact directly with ERH-2 using a conserved sequence motif. Finally, our data suggest a role for TOST-1:PETISCO in SL1 homeostasis in the early embryo. Our work describes a key complex for 21U RNA processing in C. elegans and strengthens the view that 21U RNA biogenesis is built on an snRNA-related pathway.

Loss of plasticity‐related gene 1 (PRG‐1), which regulates synaptic phospholipid signaling, leads to hyperexcitability via increased glutamate release altering excitation/inhibition (E/I) balance in cortical networks. A recently reported SNP in prg‐1 (R345T/mutPRG‐1) affects ~5 million European and US citizens in a monoallelic variant. Our studies show that this mutation leads to a loss‐of‐PRG‐1 function at the synapse due to its inability to control lysophosphatidic acid (LPA) levels via a cellular uptake mechanism which appears to depend on proper glycosylation altered by this SNP. PRG‐1+/− mice, which are animal correlates of human PRG‐1+/mut carriers, showed an altered cortical network function and stress‐related behavioral changes indicating altered resilience against psychiatric disorders. These could be reversed by modulation of phospholipid signaling via pharmacological inhibition of the LPA‐synthesizing molecule autotaxin. In line, EEG recordings in a human population‐based cohort revealed an E/I balance shift in monoallelic mutPRG‐1 carriers and an impaired sensory gating, which is regarded as an endophenotype of stress‐related mental disorders. Intervention into bioactive lipid signaling is thus a promising strategy to interfere with glutamate‐dependent symptoms in psychiatric diseases.

In meiner Arbeit beschrieb ich immunhistochemisch die Expression des Plasticity-Related Gene-1 (PRG-1) in charakteristischen perinatalen und postnatalen Entwicklungsstadien der Maus. Dazu etablierte ich am Konfokalen- und am Lichtmikroskop die Färbung mit ßGal im heterozygoten PRG-1 Tier und entwickelte ein neues Protokoll für den PRG-1 Antikörper im Wildtyp Tier. Außerdem erstellte ich zur genauen Proteinlokalisation 3D Rekonstruktionen von konfokalen Schnittbildern. Grundlegende Erkenntnisse überprüfte ich exemplarisch an humanen Schnitten und fand Hinweise darauf, dass die Ergebnisse aus dem Mausmodell sich auf den Menschen übertragen lassen. PRG-1 wurde lediglich in glutamatergen Neuronen gefunden, in denen es die postsynaptische Membran glutamaterger Synapsen flankierte. Ich zeigte, dass auch das mutierte, funktionslose PRG-1 Molekül (H235K) bei einer Rekonstitution in PRG-1-/- Mäusen mittels in-utero Elektroporation in der Membran eingebaut wird. Darüber hinaus lokalisierte ich durch 3D Rekonstruktionen den mutmaßlichen Interaktionspartner von PRG-1, den LPA-2 Rezeptor, an der Präsynapse glutamaterger Neurone. In Kooperationen konnte ich dadurch dazu beitragen, morphologische Daten zu einem neuen post- präsynaptischen Mechanismus in der synaptischen Erregungsmodulation beizusteuern. Ultrastrukturelle Entwicklungsanalysen, die durch elektronenmikroskopische Aufnahmen bestätigt wurden, zeigen zudem, dass PRG-1 zu einem sehr frühen Zeitpunkt der glutamatergen Synapsenreifung exprimiert wird. Sie legen seine potentiell wichtige Rolle in diesem Prozess nahe. Letztendlich zeigten meine Färbungen zur neuronalen Ausreifung im Gyrus Dentatus Veränderungen, die bei Abwesenheit von PRG-1 auftreten und auf seine Beteiligung hinweisen könnten. In einem weiteren Projektteil untersuchte ich das intrakranielle und entwicklungsspezifische PRG-1 Vorkommen. Dafür erstellte ich einen „Atlas“ der PRG-1 Expression von der perinatalen Entwicklung bis zum erwachsenen Tier und beschrieb darin die Stärke der PRG-1-Expression in unterschiedlichen kortikalen und subkortikalen Arealen. Kurz zusammengefasst durchläuft PRG-1 peri- und postnatal eine dynamische Expression, die für die einzelnen Hirnregionen charakteristisch und spezifisch ist. In “Input” Neuronen des Kortex, speziell in der Schicht IV, im Hippokampus und im olfaktorischen System, welches aufeinanderfolgend glutamaterge Projektionen auf glutamaterge Neurone besitzt, wies ich ein durchgehend starkes und sehr früh einsetzendes PRG-1 Signal nach. Die dynamische PRG-1 Expression in kortikalen Regionen, die an der sensorischen Wahrnehmung beteiligt sind, spiegelt die synaptische Organisation wider. Im erwachsenen Tier beschränkt sich das Signal schließlich auf die Haupt-Input- Schichten/Neurone des Kortex. Bei Abwesenheit von PRG-1 zeigten Mäuse in einer Kooperation mit dem Institut für Anatomie in Magdeburg signifikante Verhaltensänderungen bei Angstversuchen.) Diese Versuche warfen die Frage auf, ob in den PRG-1-/- Mäusen die primäre sensorische Sinneswahrnehmung oder die höhere kortikale Verarbeitung der aufgenommenen Sinnesreize gestört ist. Insgesamt konnte ich einige Fragen zur PRG-1 Expression beantworten, seine Rolle im ZNS beleuchten und zudem Anhaltspunkte für zukünftige Untersuchungen schaffen., In my dissertation I described the expression of plasticity-related Gene -1 (PRG-1) in characteristic perinatal and postnatal development of the mouse by immunhistochemistry. Therefore I established in heterozygote animals the ßGal coloration at the confocal and the light microscope and developed a new protocol for the PRG-1 antibody in the wildtyp animal. Furthermore for localization of the protein I produced 3D reconstructions of confocal slices. Basic results I screened on human slices and found hints that the consequences may be transferred to the situation in humans. PRG-1 could only be found in glutamatergic neurons, where it is situated at the postsynaptic membrane of glutamatergic synapses. I showed that the mutated protein with loss of function (H235) is also correctly placed in the membrane in PRG-1 knockout mice. Furthermore I localized the presumably partner of PRG-1, the LAP2 Receptor, at the presynaps of glutamatergic neurons. In cooperations I could help to detect a new post-presynaptic mechanism for modulation of glutamatergic excitation. Ultra structural analyses of development in mice could show that PRG-1 is early present in synaptic maturation. This is proved by electronic microscope results. Probably the protein plays an important role in this process. I could also show changes in neuronal maturation in PRG-1 knockout mice. This could also be a hint of its participation in this process. In another project I analyzed the appearance of PRG-1 intracranial and during development. I described the intensity of PRG-1 expression from prenatal to adult animal. PRG-1 has a dynamic expression that is specific and characteristic for each part of the brain. In cortical, hippocampal and olfactory input neurons exists a strong signal that appears early in development. The dynamic cortical sensoric expression of PRG-1 reflects the synaptic organization. In the adult animal PRG-1 is limited to main input neurons. A cooperation with the anatomic institute of Magdeburg showed, when PRG-1 is missing, the animals present significant changes in behavior and fear experiments. This questions if in PRG-1 -/- mice the primer sensory detection or the cortical translation could be deranged. To sum up I could answer some questions about PRG-1 expression, find hints about its possible role in the nervous system and provide information for future analysis.

Die Informationsübertragung im ZNS verläuft über synaptische Verbindungen und kann durch diese moduliert werden. Plasticity related gene 1 (PRG-1) wurde kürzlich als Modulator der exzitatorischen Informationsübertragung an Synapsen beschrieben. Gemäß des derzeit postulierten Mechanismus verändert das postsynaptisch lokalisierte PRG-1 den Gehalt an bioaktiven Lipiden im synaptischen Spalt und moduliert auf diese Weise die Signalweiterleitung über die präsynaptisch lokalisierten LPA2-Rezeptoren. In der vorliegenden Arbeit wurde ein Mausmodell erzeugt, welches das Reportergen „yellow fluoresent protein“ (YFP) unter Kontrolle von Elementen des nativen PRG-1 Promoters exprimiert. Die Expression des Reporters im erzeugten Mausmodell wird durch 200 kb kotransferierte genomische Sequenz, die den Transkriptionsstart von PRG-1 umschließt, gesteuert. Nachfolgende immunhistologische Untersuchungen dokumentierten, daß die gewählten 200 kb Sequenz ausreichen, um sowohl die gehirnspezifische Expression von YFP, als auch die PRG-1 spezifische Expression des Reporterproteins in diversen Neuronentypen zu steuern. Ein weiteres Ziel dieser Arbeit war die Untersuchung der transkriptionellen Regulation von prg1. Mittels RLM-Race wurde erstmals dokumentiert, daß die Transkription von PRG-1 über multiple Transkriptionsstartorte initiiert wird. Ein weiter stromaufwärts gelegener Transkriptionsstart, der vorab publiziert wurde, konnte mittels PCR-Analyse bestätigt werden. Mittels Reportergenassays von Deletionskonstrukten in primären kultivierten Neuronen bzw. Astrozyten, konnte die Sequenz eines Minimalpromoters identifiziert werden, der eine ca. vierzigfache transkriptionelle Stimulation in Neuronen erzeugt. Die Funktionalität des identifizierten Minimalpromoters konnte durch Elektroporation eines Reportergenkonstrukts in organotypische Slicekulturen dokumentiert werden. Mittels EMSA wurde der identifizierte Minimalpromoter untersucht und Sequenzabschnitte identifiziert, an denen DNS-Proteinkomplexe nachgewiesen werden konnten. In silico Analysen ermittelten konservierte Bindemotive für die Transkriptionsfaktoren SP1 und CREB. Im Anschluß konnte die nachgewiesenen DNS/Proteinkomplexe spezifisch durch Konsensusoligonukleotide der Sequenz von SP1 und CREB kompetetiert werden. Eine Nex1 mediierte transkriptionelle Regulation von PRG-1 war zu Beginn dieser Studie bekannt. Mittels Kotransfektionen eines Nex1-Expressionsvektors wurde nachgewiesen, daß kein signifikanter Einfluß von Nex1 auf den identifizierten Minimalpromoter vorliegt, so daß ein neuartiger Regulationsmechanismus von PRG-1 beschrieben wurde. Mittels Western Blot Analysen konnte keine Veränderung des PRG-1 Gehalts in Gesamthirnhomogenaten von Nex1 defizienten Mäusen festgestellt werden. Lokalisationsveränderungen von PRG-1 konnten durch Untersuchungen der im Rahmen dieser Arbeit erzeugten Reportermaus ausgeschlossen werden. Eine weitere Zielstellung dieser Arbeit war die Identifikation möglicher Interaktionspartner von PRG-1. Durch den „Yeast Two-Hybrid Sreening Service at DKFZ“ wurde ein Two-Hybrid-Screen durchgeführt, der NSF als möglichen Interaktionspartner prognostizierte. Diese Interaktion konnte durch Immunpräzipitationen von in HEK 293-Zellen überexprimierten affinitätsmarkierten Proteinen bestätigt werden. Nachfolgend konnte NSF auch aus Gesamthirnhomogenaten durch an Protein A-Sepharose gekoppelte PRG-1 Antikörper präzipitiert werden, die im Rahmen dieser Arbeit hergestellt wurden., Transmission of information in the CNS is processed via synaptic connections and is modulated. The protein plasticity related gene 1 (PRG-1) was recently described as modulator of excitatory synaptic transmission. According to the current postulated mechanism, the postsynaptic expressed PRG-1 modifies the concentration of bioactive lipids in the synaptic cleft and modulates thereby the signal transmission via the presynaptic localized LPA2-receptors. In the present study a mouse model was generated expressing the reporter gene yellow flourescent protein (YFP) under control of elements from the native PRG-1 promoter. Expression of the reporter was controlled by a 200 kb cotransfected sequence, encompassing the transcription start site of prg1. Immunohistologic analysis of the generated reporter mouse documented that the choosen 200 kb sequence stretch was effectual to confer strict neuronal expression of YFP. Additional all regulation elements necessary for the intraneuronal expression pattern of PRG-1 seem to be present on the tranferred sequence stretch. Another aim was the analysis of prg1 transcriptional regulation. Using RLM- race it could be shown that transcription of prg-1 is initated at multiple start points. Another transcription start point localized further upstream which was previously published from our group could be confirmed with PCR- analysis. Reporter gene-assays of deletion constructs in primary cultivated neurons and astrocytes identified the sequence of a minimal promotor, which stimulates specific transcription in neurons forty-fold. The functionality of the identified minimal promotor could be documented via electroporation of a reporter gene construct in organotypic slice constructs. Investigation of the identified minimal promoter via EMSA determined sequence fragments with DNA- protein-complexes. In silico analysis revealed conserved binding sites for the transcription factors SP1 and CREB in the rodent and human minimal promoter sequence. The detected DNA-protein-complexes could be specifically competed with SP1 and CREB consensus oligonucleotides. The direct regulation of PRG-1 transcription through the transcription factor Nex1 was published prior to this study. The question emerged, if transcriptional regulation of the identified minimal promoter was influenced by Nex1. No significant transcriptional stimulation could be detected in cotransfection experiments with a Nex1 expression vector, indicating a new Nex1 independent regulation mechanism. No alteration regarding the PRG-1 protein content in brain homogenates was detected in Nex1 deficient mice via western blot analysis. No variation regarding the spatial expression of PRG-1 could be detected via analysis of reporter gene expression in the generated PRG-1 reporter mouse. Another aim of this study was the identification of putative interaction partners of PRG-1. The cytoplasmatic C-terminus of PRG-1 was subcloned in an expression vector, which was used in a two-hybrid assay performed by “Yeast Two-Hybrid Sreening Service at DKFZ”. NSF was identified as putative interaction partner. This interaction was confirmed by immunoprecipitation of affinity tagged proteins expressed in HEK 293 cells. Furthermore this interaction was corroborated by co-precipitation of NSF via protein A-sepharose coupled PRG-1 antibodies, which were generated as part of this study.

Plasticity Related Gene-1 (PRG-1) is a brain-specific membrane protein and the first identified member of the Plasticity Related Gene family (PRG-1-5). PRGs belong to the Lipid Phosphate Phosphatase (LPP) superfamily whose members have an extracellular ectoenzymatic activity known to dephosphorylate Lysophosphatidic Acid (LPA) into its inactive monomers (Monoacylglycerol (MAG) and phosphate). PRG-1 is also known to act specifically at the excitatory synapse on hippocampal neurons and has been recently proposed as an important player in the modulatory control of hippocampal excitability by means of non- enzymatic control of extracellular LPA concentration. The non-enzymatic control occurs at the synaptic level but its exact process is not clarified yet. The interaction between PRG-1 and the Ras-specific exchange factor: Ras Guanosine Release Factor-2 (Ras GRF-2) has been identified. PRG-1/Ras GRF-2 interaction takes place not only after overexpression of both proteins in mammalian cells, but also in cortical primary neuronal cultures when analysing endogenous PRG-1/Ras GRF-2 interaction. Furthermore, the endogenous PRG- 1/Ras GRF-2 interaction is disrupted after extracellular LPA application and not after the application of the Thrombin activator: Thrombin Receptors Peptide (TRP) known to have a similar effect on cell fate but through distinct receptors. It has also been found that PRG-1 protein-protein interaction controls the intracellular levels of the active protooncogene N-Ras and indeed the N-Ras activation is regulated depending on the extracellular concentration of LPA. To assess the intracellular signalling cascade, phosphorylation of Mitogen Activated Protein Kinase (MAPK) has been analysed; enhanced MEK/ERK activation but no p38 phosphorylation could be detected in cell culture after LPA application. And finally, significant axon elongation could be demonstrated after increasing the protein level of active N-Ras in primary neurons. Presented results will conclude showing PRG-1 as a Ras-cascade controller, during brain development mainly through Ras GRF-2 and depending on extracellular LPA presence. In the final schematic representation will be showed how extracellular LPA controls, in turn, Ras GRF-2 resulting in a decreased amount of active N-Ras protein known to induce axon growth during neuronal differentiation as well as to be deregulated in tumors. Newly presented data are the first results on PRG-1 function after its discovery on 2003 and the obtention of the PRG-1-/- animals on 2009. They represent a significant step further towards the understanding of the protein, its function during embryonal brain development but also its role in pathologies such as brain injury or cancer., Plasticity Related Gene-1 weist eine weitgehend gehirnspezifische Expression auf. Das PRG-1 Protein wurde als erstes Mitglied der Plasticity Related Genes Familie (PRG-1-5) identifiziert. Die PRG-Familie gehört zu den Lipid Phosphatase/Phosphotransferasen (LPP), die an der Regulation des LPA-Spiegels beteiligt sind. LPP Familienmitglieder haben eine ektoenzymatische Aktivität, die LPA zu zwei inaktiven Monomere, Monoacylglicerol (MAG) und Phosphat (P), degradiert. PRG-1 ist ein Protein, das die hippocampale Erregbarkeit über die Regulation des LPA-Spiegels moduliert. Es wird vermutet, dass PRG-1 einen nicht-enzymatischen Einfluss auf Synapsen hat, welcher abhängig von der extrazellulären LPA-Konzentration ist. Im Rahmen dieser Arbeit wurde die Interaktion zwischen PRG-1 und Ras-specific Exchange Factor: Ras Guanosine Release Factor-2 (Ras GRF-2) untersucht. DieseWechselbeziehung wurde sowohl mittels Überexpression beider Proteine in Säugerzellen, als auch durch eine endogene Expression in kortikalen Neuronen analysiert. Darüber hinaus konnte hierbei LPA als spezifischer extrinsischer Faktor, der die Interaktion unterbricht, nachgewiesen werden. Anders als Thrombin, welches einen vergleichbaren Effekt auf Zellen hat wie LPA, jedoch nur LPA die Interaktion unterbindet und Thrombin keinen Einfluss auf diese Interaktion nimmt. Des Weiteren wurde bewiesen, dass PRG-1 Protein-Protein Interaktionen durch extrazellulläres LPA reguliert werden und gleichzeitig die Expression des intrazellulär onkogen aktiven N-Ras beeinflussen. Um zu kontrollieren ob PRG-1 als Membranprotein auf extrinsische Faktoren, wie zum Beispiel LPA, reagiert und somit eine intrazelluläre Signalkaskade auslöst, wurde die Phosphorylierung der Mitogen Aktivated Protein Kinase (MAPK) untersucht. Dabei konnte gezeigt werden, dass eine MEK/ERK Aktivierung, in Gegenzätz zur p38 Phosphorylierung, nach LPA Behandlung vorhanden ist. Detaillierte morphologische Analysen an N-Ras überexprimierenden primären Neuronen zeigten eine Verlängerung der Axone. Die im Verlauf dieser Arbeit generierten Daten zeigen PRG-1 als Regulator der Ras-Kaskade in Abhängigkeit von extrazellulärem LPA und Ras GRF-2. Aufgrund der Zusammenfassung ergibt sich die Hypothese, dass extrazelluläres LPA indirekt Ras GRF-2 beeinflusst. Dieses führt zu einer Abnahme von aktivem N-Ras, welches das Auswachsen der Axone während der neuronalen Differenzierung begünstigt und die Entwicklung von Tumoren negativ beeinflussen kann. Diese zum ersten Mal präsentierten Daten sind die ersten Resultate zur Funktion von PRG- 1 seit seiner Entdeckung im Jahre 2003 und der Herstellung von PRG-1-/- im Jahre 2009. Die Daten liefern einen Fortschritt im Verständnis der Funktion von PRG-1 während der embryonalen Gehirnentwicklung und dessen Rolle bei pathologischen Veränderungen, wie zum Beispel Gehirnverletzungen oder Krebs.

Gesamthabilitationsschrift, Das ZNS ist ein komplex verschaltetes System, bei dem die sinnvollen Verbindungen zwischen den Neuronen untereinander entscheidende Voraussetzung für das Funktionieren und die höheren Leistungen sind. Eine akute Schädigung führt zum Verlust von neuronalen Verbindungen und kann schliesslich zu neuronalem Zelltod führen. Nach Abklingen der akuten Phase kommt es im ZNS nur zum spärlichen Wiederauswachsen von Axonen und damit zum dauerhaften Verlust von Funktionen. Daher ist das therapeutische Ziel, Neurone vor dem Zelltod zu bewahren und die neuronale Regeneration mit dem Ziel der Restitutio ad integrum zu fördern, und damit die verlorengegangenen Funktionen wiederzuerlangen. So ist der Gegenstand dieser Schrift die Aufklärung der molekularen Mechanismen der Zellprotektion von Selen und der Regeneration durch die neu identifizierten Plasticity-Related Genes (PRGs). Es konnte in den vorgestellten Arbeiten das Spurenelement Selen als neuroprotektive Substanz identifiziert werden, dass Neurone vor dem exzitatorischen und oxidativen Zelltod bewahrt. Selen erweist sich dabei als hochpotent. So sind schon nanomolare Konzentrationen an Selen effektiv. Weiterhin konnte gezeigt werden, dass Selen nicht einfach als Sauerstoffradikalfänger wirkt, sondern es zur Neuroprotektion der de novo Proteinsynthese bedarf. Da die neuroprotektiven Effekte von Selen in die ‘Todesstrecke’ vieler neuropathologischer Prozesse (wie Ischämie und epileptischen Krampfanfälle) eingreifen, ist die adequate Selenzufuhr eine wichtige Strategie zur Prävention von neuronalem Zellschaden. Darüber hinaus bietet Selen eine neue Alternative zu den Antioxidantien und könnte durch die hohe Potenz bei exzitatorischen Schäden klinische Anwendung finden. Mit der PRG Familie wurde eine neue Proteinfamilie im ZNS mit auswachsfördernden Effekten entdeckt. Dabei zeigte sich, dass PRG-1 im sich entwickelnden und in regenerierenden Neuronen exprimiert wird und vor dem LPA-induzierten Axonkollaps schützt. LPA selbst ist im ZNS nach einer Schädigung vorhanden und inhibiert axonales Wachstum. PRG-1 ist in der Lage, die LPA-Rezeptoraktivierung zu regulieren und damit in die RhoA Signalkaskade einzugreifen. Somit können PRG-1 exprimierende Neurone die auswachsinhibitorische Umgebung überkommen und in ihr Zielgebiet wieder einwachsen. Darüber hinaus induziert PRG-3, ein weiteres Mitglied der PRG Familie, das Neuritenwachstum und steigert das Längenwachstum. Weitere Untersuchungen mit genetisch veränderter PRG-1 Expression werden in Zukunft die Rolle von PRG-1 in vivo aufklären und die Möglichkeit einer klinischen Anwendung prüfen., The central nervous system is a highly wired structure, in which functional connections are necessary for proper functioning. Acute brain damage leads to a loss of neuronal connections and eventually to neuronal cell death. However, only weak axonal regeneration occurs following the acute phase of degeneration which leads to permanent functional loss. Thus, it is a therapeutic aim to prevent neuronal cell death and to improve neuronal regeneration to regain proper brain function. Therefore, the goal of this thesis is to investigate the molecular mechanisms of cell protection mediated by the trace element selenium and axonal regenerative growth induced by Plasticity-Related Genes (PRGs). Selenium is highly neuroprotective in nano molar range and requires the de novo protein synthesis. Thus, selenium does not act simply as a radical scavenger rather than induces translation of proteins which protect from oxidative stress and excitotoxicity. Since selenium acts within the common death pathway of many neuropathological processes, it is conceivable to use selenium in clinical settings. Second, we discovered a novel gene family, named Plasticity-Related Genes (PRGs, PRG1-PRG-5) with regenerative potential. As prototypic representatives, PRG-1 and PRG-3 are regulated during brain development and show upregulation following brain lesion. Furthermore, PRG-1 and PRG-3 induces neurite growth and prevent from LPA-induced axon collapse. Further studies include the analysis of transgenic mice overexpressing GFP- tagged PRG-1 and PRG-3 as well as gene disruption mutants, which will provide a valuable tool to prove the regenerative capacity of PRGs in vivo.
