1. Uncovering spatiotemporal patterns of atrophy in progressive supranuclear palsy using unsupervised machine learning.

Abstract

To better understand the pathological and phenotypic heterogeneity of progressive supranuclear palsy and the links between the two, we applied a novel unsupervised machine learning algorithm (Subtype and Stage Inference) to the largest MRI data set to date of people with clinically diagnosed progressive supranuclear palsy (including progressive supranuclear palsy-Richardson and variant progressive supranuclear palsy syndromes). Our cohort is comprised of 426 progressive supranuclear palsy cases, of which 367 had at least one follow-up scan, and 290 controls. Of the progressive supranuclear palsy cases, 357 were clinically diagnosed with progressive supranuclear palsy-Richardson, 52 with a progressive supranuclear palsy-cortical variant (progressive supranuclear palsy-frontal, progressive supranuclear palsy-speech/language, or progressive supranuclear palsy-corticobasal), and 17 with a progressive supranuclear palsy-subcortical variant (progressive supranuclear palsy-parkinsonism or progressive supranuclear palsy-progressive gait freezing). Subtype and Stage Inference was applied to volumetric MRI features extracted from baseline structural (T1-weighted) MRI scans and then used to subtype and stage follow-up scans. The subtypes and stages at follow-up were used to validate the longitudinal consistency of subtype and stage assignments. We further compared the clinical phenotypes of each subtype to gain insight into the relationship between progressive supranuclear palsy pathology, atrophy patterns, and clinical presentation. The data supported two subtypes, each with a distinct progression of atrophy: a 'subcortical' subtype, in which early atrophy was most prominent in the brainstem, ventral diencephalon, superior cerebellar peduncles, and the dentate nucleus, and a 'cortical' subtype, in which there was early atrophy in the frontal lobes and the insula alongside brainstem atrophy. There was a strong association between clinical diagnosis and the Subtype and Stage Inference subtype with 82% of progressive supranuclear palsy-subcortical cases and 81% of progressive supranuclear palsy-Richardson cases assigned to the subcortical subtype and 82% of progressive supranuclear palsy-cortical cases assigned to the cortical subtype. The increasing stage was associated with worsening clinical scores, whilst the 'subcortical' subtype was associated with worse clinical severity scores compared to the 'cortical subtype' (progressive supranuclear palsy rating scale and Unified Parkinson's Disease Rating Scale). Validation experiments showed that subtype assignment was longitudinally stable (95% of scans were assigned to the same subtype at follow-up) and individual staging was longitudinally consistent with 90% remaining at the same stage or progressing to a later stage at follow-up. In summary, we applied Subtype and Stage Inference to structural MRI data and empirically identified two distinct subtypes of spatiotemporal atrophy in progressive supranuclear palsy. These image-based subtypes were differentially enriched for progressive supranuclear palsy clinical syndromes and showed different clinical characteristics. Being able to accurately subtype and stage progressive supranuclear palsy patients at baseline has important implications for screening patients on entry to clinical trials, as well as tracking disease progression.

Abstract

A joint determination of the reactor antineutrino spectra resulting from the fission of $^{235}$U and $^{239}$Pu has been carried out by the Daya Bay and PROSPECT collaborations. This Letter reports the level of consistency of $^{235}$U spectrum measurements from the two experiments and presents new results from a joint analysis of both data sets. The measurements are found to be consistent. The combined analysis reduces the degeneracy between the dominant $^{235}$U and $^{239}$Pu isotopes and improves the uncertainty of the $^{235}$U spectral shape to about 3\%. The ${}^{235}$U and $^{239}$Pu antineutrino energy spectra are unfolded from the jointly deconvolved reactor spectra using the Wiener-SVD unfolding method, providing a data-based reference for other reactor antineutrino experiments and other applications. Abstract

The most common clinical phenotype of progressive supranuclear palsy is Richardson syndrome, characterized by levodopa unresponsive symmetric parkinsonism, with a vertical supranuclear gaze palsy, early falls and cognitive impairment. There is currently no detailed understanding of the full sequence of disease pathophysiology in progressive supranuclear palsy. Determining the sequence of brain atrophy in progressive supranuclear palsy could provide important insights into the mechanisms of disease progression, as well as guide patient stratification and monitoring for clinical trials. We used a probabilistic event-based model applied to cross-sectional structural MRI scans in a large international cohort, to determine the sequence of brain atrophy in clinically diagnosed progressive supranuclear palsy Richardson syndrome. A total of 341 people with Richardson syndrome (of whom 255 had 12-month follow-up imaging) and 260 controls were included in the study. We used a combination of 12-month follow-up MRI scans, and a validated clinical rating score (progressive supranuclear palsy rating scale) to demonstrate the longitudinal consistency and utility of the event-based model's staging system. The event-based model estimated that the earliest atrophy occurs in the brainstem and subcortical regions followed by progression caudally into the superior cerebellar peduncle and deep cerebellar nuclei, and rostrally to the cortex. The sequence of cortical atrophy progresses in an anterior to posterior direction, beginning in the insula and then the frontal lobe before spreading to the temporal, parietal and finally the occipital lobe. This in vivo ordering accords with the post-mortem neuropathological staging of progressive supranuclear palsy and was robust under cross-validation. Using longitudinal information from 12-month follow-up scans, we demonstrate that subjects consistently move to later stages over this time interval, supporting the validity of the model. Abstract

The Precision Reactor Oscillation and Spectrum (PROSPECT) Experiment is a reactor neutrino experiment designed to search for sterile neutrinos with a mass on the order of 1 eV/c$^2$ and to measure the spectrum of electron antineutrinos from a highly-enriched $^{235}$U nuclear reactor. The PROSPECT detector consists of an 11 by 14 array of optical segments in $^{6}$Li-loaded liquid scintillator at the High Flux Isotope Reactor in Oak Ridge National Laboratory. Antineutrino events are identified via inverse beta decay and read out by photomultiplier tubes located at the ends of each segment. The detector response is characterized using a radioactive source calibration system. This paper describes the design, operation, and performance of the PROSPECT source calibration system.

Abstract

PROSPECT, the Precision Reactor Oscillation and SPECTrum experiment, is a short-baseline reactor antineutrino experiment designed to provide precision measurements of the $^{235}$U product $\overline{\nu}_e$ spectrum of utilizing an optically segmented 4-ton liquid scintillator detector. PROSPECT's segmentation system, the optical grid, plays a central role in reconstructing the position and energy of $\overline{\nu}_e$ interactions in the detector. This paper is the technical reference for this PROSPECT subsystem, describing its design, fabrication, quality assurance, transportation and assembly in detail. In addition, the dimensional, optical and mechanical characterizations of optical grid components and the assembled PROSPECT target are also presented. The technical information and characterizations detailed here will inform geometry-related inputs for PROSPECT physics analysis, and can guide a variety of future particle detection development efforts, such as those using optically reflecting materials or filament-based 3D printing., Comment: 31 pages, 28 figures

Abstract

This work reports the production and characterization of lithium-loaded liquid scintillator (LiLS) for the Precision Reactor Oscillation and Spectrum Experiment (PROSPECT). Fifty-nine 90 liter batches of LiLS (${}^6{\rm Li}$ mass fraction 0.082%$\pm$0.001%) were produced and samples from all batches were characterized by measuring their optical absorbance relative to air, light yield relative to a pure liquid scintillator reference, and pulse shape discrimination capability. Abstract

This Letter reports the first measurement of the $^{235}$U $\overline{\nu_{e}}$ energy spectrum by PROSPECT, the Precision Reactor Oscillation and Spectrum experiment, operating 7.9m from the 85MW$_{\mathrm{th}}$ highly-enriched uranium (HEU) High Flux Isotope Reactor. With a surface-based, segmented detector, PROSPECT has observed 31678$\pm$304 (stat.) $\overline{\nu_{e}}$-induced inverse beta decays (IBD), the largest sample from HEU fission to date, 99% of which are attributed to $^{235}$U. Despite broad agreement, comparison of the Huber $^{235}$U model to the measured spectrum produces a $\chi^2/ndf = 51.4/31$, driven primarily by deviations in two localized energy regions. The measured $^{235}$U spectrum shape is consistent with a deviation relative to prediction equal in size to that observed at low-enriched uranium power reactors in the $\overline{\nu_{e}}$ energy region of 5-7MeV.

Abstract

The Precision Reactor Oscillation and Spectrum Experiment, PROSPECT, is designed to make both a precise measurement of the antineutrino spectrum from a highly-enriched uranium reactor and to probe eV-scale sterile neutrinos by searching for neutrino oscillations over meter-long baselines. PROSPECT utilizes a segmented $^6$Li-doped liquid scintillator detector for both efficient detection of reactor antineutrinos through the inverse beta decay reaction and excellent background discrimination. PROSPECT is a movable 4-ton antineutrino detector covering distances of 7m to 13m from the High Flux Isotope Reactor core. It will probe the best-fit point of the $\bar\nu_e$ disappearance experiments at 4$\sigma$ in 1 year and the favored regions of the sterile neutrino parameter space at more than 3$\sigma$ in 3 years. PROSPECT will test the origin of spectral deviations observed in recent $\theta_{13}$ experiments, search for sterile neutrinos, and address the hypothesis of sterile neutrinos as an explanation of the reactor anomaly. Abstract

Research reactors host a wide range of activities that make use of the intense neutron fluxes generated at these facilities. Recent interest in performing measurements with relatively low event rates, e.g. reactor antineutrino detection, at these facilities necessitates a detailed understanding of background radiation fields. Both reactor-correlated and naturally occurring background sources are potentially important, even at levels well below those of importance for typical activities. Here we describe a comprehensive series of background assessments at three high-power research reactors, including γ-ray, neutron, and muon measurements. For each facility we describe the characteristics and identify the sources of the background fields encountered. The general understanding gained of background production mechanisms and their relationship to facility features will prove valuable for the planning of any sensitive measurement conducted therein.

15. Growing an Innovative Workforce. Forward Looking Education and Training for Forward Looking Business.

Abstract

This report describes and presents experiences of a project that acted as a catalyst for accelerating the pace of change in vocational education and training in Britain. The project involved three colleges of further education (CFEs), three occupations, and eight employers in a test of a new learning approach that would anticipate changes in the world of work and respond to the widely expressed needs of businesses. Chapter I describes the need for change and gives an overview of what senior managers, training managers, and further education staff would have to do to achieve outcomes similar to those pursued in the project. Chapter II gives an overview of the key achievements in each of three locations--Nottingham, Solihull, and the Wirral, in each of which there was a cluster of a CFE and one or more companies. Chapter III describes what was learned and how an innovative work force program can best be developed. The chapter highlights the lessons--in the form of "Learning Points"--associated with each characteristic of such programs. Chapter IV offers practical proposals for making the framework known to, and adopted by, forward-looking companies, public services, and colleges. (YLB)

Abstract

We characterized the role of structural variants, a largely unexplored type of genetic variation, in two non-Alzheimer's dementias, namely Lewy body dementia (LBD) and frontotemporal dementia (FTD)/amyotrophic lateral sclerosis (ALS). To do this, we applied an advanced structural variant calling pipeline (GATK-SV) to short-read whole-genome sequence data from 5,213 European-ancestry cases and 4,132 controls. We discovered, replicated, and validated a deletion in TPCN1 as a novel risk locus for LBD and detected the known structural variants at the C9orf72 and MAPT loci as associated with FTD/ALS. We also identified rare pathogenic structural variants in both LBD and FTD/ALS. Finally, we assembled a catalog of structural variants that can be mined for new insights into the pathogenesis of these understudied forms of dementia.

Abstract

The properties of the synthesized films of organic-inorganic perovskites CH3NH3PbI3 obtained at various ratios of starting reagents (PbI2 and CH3NH3I) have been studied. As a solvent, we used chemically pure dried dimethylformamide (DMF). Organic-inorganic perovskites are promising for photovoltaic applications. It has been shown that regardless of the ratio of the starting reagents, single-phase perovskites are formed, at the same time the microstructure of the films changes significantly. It has been reported photoelectric and optical properties of synthesized films, namely: experimental and theoretical spectral dependences of the low-signal surface photovoltage and transmission. The band gap and the Urbach parameter dependence on the ratio of precursors were determined. It has been found that the materials’ band gap depends on the ratio of precursors and equals to 1.59, 1.62 and 1.57 eV, while the characteristic Urbach energy equals to 18, 19 and 22 meV for the PbI2:CH3NH3I films with PbI2 ratio of 1:1, 1:2 and 1:3, respectively. It has been ascertained that the spectral dependences of the low-signal surface photovoltage are much more sensitive to the material microstructure and its electronic structure close to the absorption edge, while the optical transmission spectra are not so sensitive. The limiting value of the short-circuit current density for the films with different PbI2 and CH3NH3I ratios has been determined.

Abstract

The paper deals with transforming social systems and their reflection in the concepts and models of the corresponding processes. A new approach to modelling and research of large social systems was proposed. The author emphasis's and describes qualitatively some aspects of the approach that are important for considering the transformation of society. The main feature of such systems is the following properties. Firstly, these systems operate in a small number of reasonably stable states. (In our approach, this is interpreted as so-called associative memory). Secondly, the evolution of such systems (actually, history for long periods) has an analogue in the learning process in models, when connections are established between the system elements. Furthermore, thirdly, the proposed approach was able to consider individuals’ internal properties (mentality) – participants in large social systems. New models of socio-economic systems are described, and an interpretation of their behaviour as trajectories on certain surfaces in the space of variables is given. The ways of transforming systems are explained: revolutionary, evolutionary and with a unique trajectory of transition. The classes are considered, into which the internal (mental) variables of individuals are divided. The internal representations of the individual from some slice of reality (in psychology, for example, this is called “internal plans”) are represented as a network of objects, concepts and the like, depending on the “object” and purpose of modelling. There are three networks (patterns, drawings) of descriptions: the actual state of affairs, presented in the form of a network; the state of things in the imagination of a particular individual and the desired (ideal) state of affairs in the imagination of a particular individual. The dynamics of changes in the parameters that characterize the individual depends on the state of the environment “externally” and on the “internal” mental variables and the above three ideas about the external environment. Adequate consideration of the external environment by representing and internal representations of surrounding individuals is possible. Such an approach is proposed when considering archetypes in the transformation of the social system. The problem of sustainable development of education and science systems is considered.

Abstract

BACKGROUND: Acute coronary syndromes most commonly arise from thrombosis of lipid-rich coronary atheromas that have large plaque burden despite angiographically appearing mild.OBJECTIVES: We sought to examine the outcomes of percutaneous coronary intervention (PCI) of non-flow-limiting vulnerable plaques.METHODS: Three-vessel imaging was performed with a combination intravascular ultrasound (IVUS) and near-infrared spectroscopy (NIRS) catheter after successful PCI of all flow-limiting coronary lesions in 898 patients presenting with myocardial infarction (MI). Patients with an angiographically non-obstructive stenosis not intended for PCI but with IVUS plaque burden ≥65% were randomized to treatment of the lesion with a bioresorbable vascular scaffold (BVS) plus guideline-directed medical therapy (GDMT) vs. GDMT alone. The primary powered effectiveness endpoint was the IVUS-derived minimum lumen area (MLA) at protocol-driven 25-month follow-up. The primary (non-powered) safety endpoint was randomized target lesion failure (TLF; cardiac death, target vessel-related MI or clinically-driven target lesion revascularization) at 24 months. The secondary (non-powered) clinical effectiveness endpoint was randomized lesion-related major adverse cardiac events (MACE; cardiac death, MI, unstable angina, or progressive angina) at latest follow-up.RESULTS: A total of 182 patients were randomized (93 BVS, 89 GDMT alone) at 15 centers. The median angiographic diameter stenosis of the randomized lesions was 41.6%; by NIRS-IVUS median plaque burden was 73.7%, median MLA was 2.9 mm2, and median maximum lipid plaque content was 33.4%. Angiographic follow-up at 25 months was completed in 167 patients (91.8%), and median clinical follow-up was 4.1 years. The follow-up MLA in BVS-treated lesions was 6.9±2.6 mm2 compared with 3.0±1.0 mm2 in GDMT alone-treated lesions (least square means difference 3.9 mm2, 95% CI 3.3-4.5, PCONCLUSIONS: PCI of angiographically mild lesions with large plaque burden was safe, substantially enlarged the follow-up MLA and was associated with favorable long-term clinical outcomes, warranting the performance of an adequately powered randomized trial.

25. Joint Determination of Reactor Antineutrino Spectra from 235U and 239Pu Fission by Daya Bay and PROSPECT

Abstract

A joint determination of the reactor antineutrino spectra resulting from the fission of 235U and 239Pu has been carried out by the Daya Bay and PROSPECT collaborations. This Letter reports the level of consistency of 235U spectrum measurements from the two experiments and presents new results from a joint analysis of both data sets. The measurements are found to be consistent. The combined analysis reduces the degeneracy between the dominant 235U and 239Pu isotopes and improves the uncertainty of the 235U spectral shape to about 3%. The 235U and 239Pu antineutrino energy spectra are unfolded from the jointly deconvolved reactor spectra using the Wiener-SVD unfolding method, providing a data-based reference for other reactor antineutrino experiments and other applications. This is the first measurement of the 235U and 239Pu spectra based on the combination of experiments at low- and highly enriched uranium reactors.

A comprehensive program of services for working parents who are employees of a consortium of companies is described and evaluated. With the support of the Prospect Hill Executive Office Park in Waltham, Massachusetts, the Prospect Hill Parents' and Children's Center offers parent support services, such as information and counseling, information resources, workplace child care, sick child care, and financial support. The center also provides management information services, including employer seminars, newsletters, reports, and consultation. This booklet includes: (1) background information on the setting and initiation of the model; (2) discussion of options in parent support services and the identification of service components that meet parents' needs; and (3) discussion of dimensions of the consortium model, including design, implementation, insurance and liability, marketing, and the initial organizational structure. Each chapter of the booklet begins with general considerations of issues that emerged during evaluation. Chapters also include illustrative case histories. It is believed that the experience gained in the development of the Prospect Hill Parents' and Children's Center can be effectively used in the development of similar employer-supported programs. Appendices include a glossary; a list of programs for reference with succinct information in several categories; and a selected bibliography. Abstract

IMPORTANCE: Growing interest in microbial dysbiosis during critical illness has raised questions about the therapeutic potential of microbiome modification with probiotics. Prior randomized trials in this population suggest that probiotics reduce infection, particularly ventilator-associated pneumonia (VAP), although probiotic-associated infections have also been reported. OBJECTIVE: To evaluate the effect of Lactobacillus rhamnosus GG on preventing VAP, additional infections, and other clinically important outcomes in the intensive care unit (ICU). DESIGN, SETTING, AND PARTICIPANTS: Randomized placebo-controlled trial in 44 ICUs in Canada, the United States, and Saudi Arabia enrolling adults predicted to require mechanical ventilation for at least 72 hours. A total of 2653 patients were enrolled from October 2013 to March 2019 (final follow-up, October 2020). INTERVENTIONS: Enteral L rhamnosus GG (1 × 10(10) colony-forming units) (n = 1321) or placebo (n = 1332) twice daily in the ICU. MAIN OUTCOMES AND MEASURES: The primary outcome was VAP determined by duplicate blinded central adjudication. Secondary outcomes were other ICU-acquired infections including Clostridioides difficile infection, diarrhea, antimicrobial use, ICU and hospital length of stay, and mortality. RESULTS: Among 2653 randomized patients (mean age, 59.8 years [SD], 16.5 years), 2650 (99.9%) completed the trial (mean age, 59.8 years [SD], 16.5 years; 1063 women [40.1%.] with a mean Acute Physiology and Chronic Health Evaluation II score of 22.0 (SD, 7.8) and received the study product for a median of 9 days (IQR, 5-15 days). VAP developed among 289 of 1318 patients (21.9%) receiving probiotics vs 284 of 1332 controls (21.3%; hazard ratio [HR], 1.03 (95% CI, 0.87-1.22; P = .73, absolute difference, 0.6%, 95% CI, –2.5% to 3.7%). None of the 20 prespecified secondary outcomes, including other ICU-acquired infections, diarrhea, antimicrobial use, mortality, or length of stay showed a significant difference. Abstract

Study and implementation of innovative systems of environmentally friendly and energy-efficient transport based on magnetic levitation, the principle of operation of which is based on the use of new solid-state magnetic materials based on compounds of rare earth materials, in particular materials with high-temperature superconductivity based on Y, permanent magnets based on Nd and Sm and magnetocaloric alloys based on Dy, Tb are of great interest throughout the world. In this work, the basic principles of magneto-levitation transport with the most economical principle of acceleration and deceleration - gravitational - are studied experimentally on mock-ups. The strength characteristics were measured: the levitation force and the lateral stabilization force, as well as losses during periodic translational motion of a cryostat with high-temperature superconducting elements made of ceramic material Y-Ba-Cu-O over the paths of permanent magnets made of the rare-earth compound Nd-Fe-B. A system for measuring the speed and compensation of losses for the implementation of continuous motion has been created and tested. The presented results indicate the possibility of scaling the layout project. It is concluded that the investigated scheme may be of practical interest for intracity and local transport communication with high comfort, environmental friendliness and record economy in the case of a successful solution of the problem of cooling HTSC elements to the temperature of the phase transition to the superconducting state, for example, using new principles of solid-state magnetic cooling based on compounds Dy-N, Tb-Ni, etc.

Abstract

The precision reactor oscillation and spectrum experiment, PROSPECT, has made world-leading measurements of reactor antineutrinos at short baselines. In its first phase, conducted at the high flux isotope reactor (HFIR) at Oak Ridge National Laboratory, PROSPECT produced some of the strongest limits on eV-scale sterile neutrinos, made a precision measurement of the reactor antineutrino spectrum from 235U, and demonstrated the observation of reactor antineutrinos in an aboveground detector with good energy resolution and well-controlled backgrounds. The PROSPECT collaboration is now preparing an upgraded detector, PROSPECT-II, to probe yet unexplored parameter space for sterile neutrinos and contribute to a full resolution of the reactor antineutrino anomaly, a longstanding puzzle in neutrino physics. By pressing forward on the world’s most precise measurement of the 235U antineutrino spectrum and measuring the absolute flux of antineutrinos from 235U, PROSPECT-II will sharpen a tool with potential value for basic neutrino science, nuclear data validation, and nuclear security applications. Following a two-year deployment at HFIR, an additional PROSPECT-II deployment at a low enriched uranium reactor could make complementary measurements of the neutrino yield from other fission isotopes. PROSPECT-II provides a unique opportunity to continue the study of reactor antineutrinos at short baselines, taking advantage of demonstrated elements of the original PROSPECT design and close access to a highly enriched uranium reactor core.

Abstract

OBJECTIVE: To develop a patient/care-giver reported scale capable of easily and reliably assessing functional disability in 4 repeat tauopathies (4RTs). BACKGROUND: 4R tauopathies including progressive supranuclear palsy, corticobasal degeneration and a subset of frontotemporal dementias manifest a range of overlapping clinical phenotypes. No available rating scale is capable of evaluating the functional impact of these complex disorders. METHODS: A multi-staged modified Delphi process was used to propose, evaluate and rank potential scale items providing content validity ratios. Staged cognitive pretesting involving input from examiners, patients and caregivers was followed by validation testing in patients participating in the 4R Tauopathy Neuroimaging Initiative or the PROgressive Supranuclear Palsy CorTico-Basal Syndrome MSA Longitudinal Study. Clinimetric properties were examined using classical test theory and item response methods, assessing data quality, reliability, construct validity, convergent validity and known-group validity. RESULTS: The resultant Cortical Basal ganglia Functional Scale (CBFS) included questions on Motor Experiences in Daily Living (14 items) and Non-Motor Experiences of Daily Living (17 items). Reliability was acceptable for internal consistency, test-retest stability, item discrimination, item-scaling thresholds and item-fit. Examination of construct validity revealed a parsimonious two-factor solution, and concurrent validity demonstrated significant correlations between the CBFS and other measures of disease severity and functional impairment. The CBFS significantly discriminated between all diagnostic groups and controls (all AUCs>90). The CBFS scores demonstrated sensitivity to change over a 12 month follow-up in patients with probable 4RTs. CONCLUSIONS: The CBFS is a patient/care-giver reported outcome measure with excellent clinimetric properties that captures disability correlated with motor, cognitive and psychiatric impairments.

38. Joint Determination of Reactor Antineutrino Spectra from 235U and 239Pu Fission by Daya Bay and PROSPECT

Abstract

A joint determination of the reactor antineutrino spectra resulting from the fission of 235U and 239Pu has been carried out by the Daya Bay and PROSPECT collaborations. This Letter reports the level of consistency of 235U spectrum measurements from the two experiments and presents new results from a joint analysis of both data sets. The measurements are found to be consistent. The combined analysis reduces the degeneracy between the dominant 235U and 239Pu isotopes and improves the uncertainty of the 235U spectral shape to about 3%. The 235U and 239Pu antineutrino energy spectra are unfolded from the jointly deconvolved reactor spectra using the Wiener-SVD unfolding method, providing a data-based reference for other reactor antineutrino experiments and other applications. This is the first measurement of the 235U and 239Pu spectra based on the combination of experiments at low- and highly enriched uranium reactors.

Abstract

We examined the role of repeat expansions in the pathogenesis of frontotemporal dementia (FTD) and amyotrophic lateral sclerosis (ALS) by analyzing whole-genome sequence data from 2,442 FTD/ALS patients, 2,599 Lewy body dementia (LBD) patients, and 3,158 neurologically healthy subjects. Pathogenic expansions (range, 40-64 CAG repeats) in the huntingtin (HTT) gene were found in three (0.12%) patients diagnosed with pure FTD/ALS syndromes but were not present in the LBD or healthy cohorts. We replicated our findings in an independent collection of 3,674 FTD/ALS patients. Postmortem evaluations of two patients revealed the classical TDP-43 pathology of FTD/ALS, as well as huntingtin-positive, ubiquitin-positive aggregates in the frontal cortex. The neostriatal atrophy that pathologically defines Huntington's disease was absent in both cases. Our findings reveal an etiological relationship between HTT repeat expansions and FTD/ALS syndromes and indicate that genetic screening of FTD/ALS patients for HTT repeat expansions should be considered.
