Anthropomorphization is a key category for discussing the depiction of the animal world in the novel and TV series Four Tankmen and a Dog. This applies to both individual specimens of portrayed species as well as to the structure of, and divisions prevailing in, the animal world. In other words, relations in the animal world sometimes mirror those in the human world. Animals are not treated as autonomous entities. Both their behaviour in performing various tasks and the way they are talked about indicate that even a creature as brilliant as Szarik, whose uncanny intellect would have him easily fit in a fairy tale, is ultimately treated as subservient to man (or perhaps him especially, seeing that, as his master’s magical assistant, it is ultimately his calling). Dogs, horses, cows — they are there to serve humans in various ways, and their anthropomorphic representations only further this notion. The same principle applies to wild animals — killing them provides one with food and clothing, but it is also shown as a test of manhood, as well as a skill that is most useful for a prospective soldier. The only being in the novel and series that could be subjugated or defeated by humans (or dogs), and is equally reluctant towards all strangers entering its territory, is the cat.

4. Root-knot nematodes produce functional mimics of tyrosine-sulfated plant peptides.

Root-knot nematodes (Meloidogyne spp.) are highly evolved obligate parasites threatening global food security. These parasites have a remarkable ability to establish elaborate feeding sites in roots, which are their only source of nutrients throughout their life cycle. A wide range of nematode effectors have been implicated in modulation of host pathways for defense suppression and/or feeding site development. Plants produce a diverse array of peptide hormones including PLANT PEPTIDE CONTAINING SULFATED TYROSINE (PSY)-family peptides, which promote root growth via cell expansion and proliferation. A sulfated PSY-like peptide RaxX (required for activation of XA21 mediated immunity X) produced by the biotrophic bacterial pathogen (Xanthomonas oryzae pv. oryzae) has been previously shown to contribute to bacterial virulence. Here, we report the identification of genes from root-knot nematodes predicted to encode PSY-like peptides (MigPSYs) with high sequence similarity to both bacterial RaxX and plant PSYs. Synthetic sulfated peptides corresponding to predicted MigPSYs stimulate root growth in Arabidopsis. MigPSY transcript levels are highest early in the infection cycle. Downregulation of MigPSY gene expression reduces root galling and egg production, suggesting that the MigPSYs serve as nematode virulence factors. Together, these results indicate that nematodes and bacteria exploit similar sulfated peptides to hijack plant developmental signaling pathways to facilitate parasitism.

Abstract Background Small-secreted peptides are increasingly recognized as a novel class of intracellular signal molecules, playing crucial roles in plant growth and development. However, the precise role and mechanism governed by peptides containing Tyrosine Sulfation (PSY) are still under investigation. Currently, there is a lack of accessible information concerning the PSY gene family in wheat. Results Therefore, in this investigation, we identified 29 PSY genes in Triticum aestivum, with the aim of unraveling their significance in plant development processes and their response to a variety of stress conditions. Phylogenetic analysis showed that TaPSY genes clustered into five groups. Additionally, an analysis of the gene structure of TaPSYs displayed a conserved evolutionary path. The syntenic relationship demonstrated the 69 orthologous gene pairs in T. dicoccoides, Ae. tauschii, T. turgidum, and H. vulgare, respectively. Furthermore, the Ka/Ks analysis indicated that TaPSY genes have experienced purifying selection during their evolutionary processes. The promoters of TaPSY genes were found to contain numerous CAREs, and these elements are known to perform essential functions in various development processes, phytohormone responses, as well as defense and stress mechanisms. In addition, the identification of potential miRNAs targeting TaPSY genes was followed by an examination of their expression patterns across various tissues. Among the 29 TaPSY genes, twenty miRNAs were discovered to target eighteen of them. Moreover, TaPSY genes displayed a distinct expression across different tissues and stress conditions. Conclusions Hence, these discoveries offer a significant reference point for forthcoming molecular investigations and hold promise for bolstering wheat yield and stress resilience through targeted genetic enhancements and strategic breeding approaches. An empirical investigation was held regarding whether topological properties associated with point clouds formed by cryptocurrencies' prices could contain information on (locally) explosive dynamics of the processes involved. Those dynamics are associated with financial bubbles. The Phillips, Shi and Yu [33,34] (PSY) timestamping method as well as notions associated with the Topological Data Analysis (TDA) like persistent simplicial homology and landscapes were employed on a dataset consisting of the time series of daily closing prices of the Bitcoin, Ethereum, Ripple and Litecoin. The note provides some empirical evidence that TDA could be useful in detecting and timestamping financial bubbles. If robust, such an empirical conclusion opens some interesting paths of further research.

Signalling peptides play a vital role in regulating a wide range of aspects, including plant growth, development, and responses to various biotic and abiotic conditions. In recent times, small post-translationally altered peptides have garnered significant attention from plant biologists due to their crucial role as signalling molecules in various biological processes. Until now, the biological function of peptides containing Tyrosine Sulfation (PSY) gene family members has only been elucidated in Arabidopsis. However, their functions in plant developmental processes and their responses to different biotic and abiotic stress conditions remain elusive in other crops. Therefore, in this study, we identified 109 PSY genes in the rice genome for the first time. However, our detailed in silico analysis focused specifically on 7 OsPSY genes of the O. sativa Japonica Group (Nippobare). Phylogenetic analysis of PSY genes from various plant species has classified these genes into eight distinct groups, designated as Group I to VIII. Interestingly, the 7 OsPSY genes identified in rice were found to be distributed across all four groups including I, V, VI and VIII. In addition, closely linked groups of PSY genes showed more similarities in terms of gene structures, conserved motif distributions, and corresponding subcellular locations. Further, the Ka/Ks score analysis revealed that OsPSY genes have undergone significant purifying selection throughout their evolutionary history. Furthermore, multiple CAREs were discovered in the OsPSY promoters, suggesting their potential importance in regulating plant growth, responses to plant hormones, and multifactorial stress conditions. Additionally, the potential miRNAs that specifically targeted to OsPSY genes were identified and also examined their expression kinetics in different tissues and developmental stage. Out of the seven OsPSY genes, six OsPSY genes were found to be targeted by twelve miRNAs. Moreover the expression profile of OsPSY genes showed a differential expression pattern in various tissues and multifactorial stress conditions. Therefore, these findings serve as a valuable reference for further molecular studies aimed at enhancing the yield and stress tolerance of rice. The understanding the functions and regulatory mechanisms of the OsPSY genes can potentially lead to the development of targeted approaches to improve crop performance under various environmental conditions, ultimately contributing to agricultural productivity and global food security.

10. Rethinking Psychology in Africa: From Decolonizing to Universalizing Knowledge in an Emerging Field.

- Abstract
Inspired by broader calls to decolonize knowledge, psychologists in Africa have recently started debating the necessity and feasibility of creating a distinctly African psychology as a new academic discipline and field of practice. Some view this idea, whether sceptically or enthusiastically, as a primarily political move; others are more concerned with philosophical questions regarding the possibilities, and boundaries, of universal psychological knowledge. While critics warn of the risks of exoticizing and further marginalizing 'African' psychology from what they see as a universal discipline, proponents argue that mainstream 'Western psychology' has so far been harmful, or at best irrelevant, for Africans. My article engages with these recent debates by drawing on my fieldwork among psychotherapists in Uganda. I question conventional framings of psychotherapy as something external and foreign to 'Africa' that has been imposed by outsiders. Instead, I show how Ugandan therapists consider themselves part of a universal field of knowledge and how, through their efforts to make this knowledge relevant in Uganda, they actively engage in the production and negotiation of psy's universality. Nevertheless, they are confronted by historical legacies and contemporary structural inequalities which limit how and where they can practice, and how their work is valued. [ABSTRACT FROM AUTHOR]

12. 菠菜 PSY 基因家族的鉴定与表达分析.

- Abstract
PSY (phytoene synthase, PSY) is the first rate‑limiting enzyme in the plant carotenoid synthesis pathway and plays an important role in plant growth and development. In order to understand the function of PSY gene family in spinach, the number, structure, promoter elements, amino acid characteristics and gene evolution of PSY gene family members in spinach were identified by bioinformatics method, and the expression pattern of PSY genes in spinach tissue and in response to the changes in the ratio of red to blue light was analyzed by real‑time fluorescence quantitative PCR (RT‑qPCR) . Meanwhile, the expressions of R1B3 (red∶blue light = 1∶3) and R3B1 (red light∶blue light = 3∶1) and the correlation between SoPSYs gene expression and total carotenoid content were analyzed. The results showed that there were 4 PSY genes in spinach, containing SQS_PSY conserved domain, and the SoPSY protein was a hydrophilic protein. PSY1 and PSY2 of spinach were closely related to beet in Amaranthaceae, and PSY3 and PSY4 were closely related to PSY3 in tomato. There were various light‑responsive and phytohormone‑related elements in the promoter sequence of spinach PSY genes. SoPSY genes had tissue expression specificity, SoPSY1, SoPSY2, and SoPSY3 were only expressed in the leaves, and SoPSY4 was mainly expressed in the roots. Spinach carotene content decreased after treatment with R1B3 and R3B1. Among spinach genotypes, the expression patterns of each SoPSY gene in response to the ratio of red to blue light were different. The expression of SoPSY1 was not significantly changed in cultivated spinach, the expression patterns of SoPSY2 were reversed in both spinach genotypes, and the relative expression patterns of SoPSY3 were consistent in the wild and cultivated spinach, and the relative expressions of these three genes was closely related to the change of total carotenoid content. This study lays the foundation for an in‑depth understanding of the biological function of the PSY genes in spinach and their regulatory mechanism in response to changes in the ratio of red to blue light. Leucine-rich repeat-receptor kinases (LRR-RKs) perceive various endogenous peptide hormones that control plant growth and development. However, the majority of corresponding ligands and their direct ligand-binding receptors have not been identified yet. A recent study demonstrated that three LRR-RK PLANT PEPTIDE CONTAINING SULFATED TYROSINE RECEPTORS (PSYRs) act as ligand-receptors of the PSY family peptides that mediate the trade-off between the optimal plant growth and stress tolerance responses. The genetic, biochemical, and transcriptome analyses suggested that PSYR1, PSYR2, and PSYR3 function as negative regulators of plant growth in the absence of PSY peptides and induce stress tolerance responses, whereas the PSY family peptides repress PSYR signaling, allowing plant growth. This trade-off mechanism between plant growth and stress responses mediated by the PSY–PSYR signaling module allows plants to survive under ever changing environmental stresses.

Eriobotrya is an evergreen fruit tree native to South-West China and adjacent countries. There are more than 26 loquat species known in this genus, while E. japonica is the only species yet domesticated to produce fresh fruits from late spring to early summer. Fruits of cultivated loquat are usually orange colored, in contrast to the red color of fruits of wild E. henryi (EH). However, the mechanisms of fruit pigment formation during loquat evolution are yet to be elucidated. To understand these, targeted carotenoid and anthocyanin metabolomics as well as transcriptomics analyses were carried out in this study. The results showed that b-carotene, violaxanthin palmitate and rubixanthin laurate, totally accounted for over 60% of the colored carotenoids, were the major carotenoids in peel of the orange colored ‘Jiefangzhong’ (JFZ) fruits. Total carotenoids content in JFZ is about 10 times to that of EH, and the expression levels of PSY, ZDS and ZEP in JFZ were 10.69 to 23.26 folds to that in EH at ripen stage. Cyanidin-3-O-galactoside and pelargonidin-3-O-galactoside were the predominant anthocyanins enriched in EH peel. On the contrary, both of them were almost undetectable in JFZ, and the transcript levels of F3H, F3’H, ANS, CHS and CHI in EH were 4.39 to 73.12 folds higher than that in JFZ during fruit pigmentation. In summary, abundant carotenoid deposition in JFZ peel is well correlated with the strong expression of PSY, ZDS and ZEP, while the accumulation of anthocyanin metabolites in EH peel is tightly associated with the notably upregulated expressions of F3H, F3’H, ANS, CHS and CHI. This study was the first to demonstrate the metabolic background of how fruit pigmentations evolved from wild to cultivated loquat species, and provided gene targets for further breeding of more colorful loquat fruits via manipulation of carotenoids and anthocyanin biosynthesis. Chlorophylls and carotenoids are essential photosynthetic pigments. Plants spatiotemporally coordinate the needs of chlorophylls and carotenoids for optimal photosynthesis and fitness in response to diverse environmental and developmental cues. However, how the biosynthesis pathways of these two pigments are coordinated, particularly at posttranslational level to allow rapid control, remains largely unknown. Here, we report that the highly conserved ORANGE (OR) family proteins coordinate both pathways via posttranslationally mediating the first committed enzyme in each pathway. We demonstrate that OR family proteins physically interact with magnesium chelatase subunit I (CHLI) in chlorophyll biosynthesis pathway in addition to phytoene synthase (PSY) in carotenoid biosynthesis pathway and concurrently stabilize CHLI and PSY enzymes. We show that loss of OR genes hinders both chlorophyll and carotenoid biosynthesis, limits light-harvesting complex assembly, and impairs thylakoid grana stacking in chloroplasts. Overexpression of OR safeguards photosynthetic pigment biosynthesis and enhances thermotolerance in both Arabidopsis and tomato plants. Our findings establish a novel mechanism by which plants coordinate chlorophyll and carotenoid biosynthesis and provide a potential genetic target to generate climate-resilient crops. Chlorophylls and carotenoids are essential photosynthetic pigments, but how their biosynthesis pathways in plants are coordinated remains largely unclear. This study reveals that the highly conserved OR family proteins coordinate photosynthetic pigment synthesis via co-chaperoning the first committed enzymes magnesium chelatase subunit I of chlorophyll biosynthesis and phytoene synthase of carotenoid biosynthesis. 17. Machine learning based personalized promotion strategy of piglets weaned per sow per year in large-scale pig farms

Abstract Background The purpose of this study was to analyze the relationship between different productive factors and piglets weaned per sow per year (PSY) in 291 large-scale pig farms and analyze the impact of the changes in different factors on PSY. We chose nine different algorithm models based on machine learning to calculate the influence of each variable on every farm according to its current situation, leading to personalize the improvement of the impact in the specific circumstances of each farm, proposing a production guidance plan of PSY improvement for every farm. According to the comparison of mean absolute error (MAE), 95% confidence interval (CI) and R2, the optimal solution was conducted to calculate the influence of 17 production factors of each pig farm on PSY improvement, finding out the bottleneck corresponding to each pig farm. The level of PSY was further analyzed when the bottleneck factor of each pig farm changed by 0.5 standard deviation (SD). Results 17 production factors were non-linearly related to PSY. The top five production factors with the highest correlation with PSY were the number of weaned piglets per litter (WPL) (0.6694), mating rate within 7 days after weaning (MR7DW) (0.6606), number of piglets born alive per litter (PBAL) (0.6517), the total number of piglets per litter (TPL) (0.5706) and non-productive days (NPD) (− 0.5308). Among nine algorithm models, the gradient boosting regressor model had the highest R2, smallest MAE and 95% CI, applied for personalized analysis. When one of 17 production factors of 291 large-scale pig farms changed by 0.5 SD, 101 pig farms (34.7%) can increase 1.41 PSY (compared to its original value) on average by adding the production days, and 60 pig farms (20.6%) can increase 1.14 PSY on average by improving WPL, 45 pig farms (15.5%) can increase 1.63 PSY by lifting MR7DW. Conclusions The main productive factors related to PSY included WPL, MR7DW, PBAL, TPL and NPD. The gradient boosting regressor model was the optimal method to individually analyze productive factors that are non-linearly related to PSY.

20. Study on the influence of different production factors on PSY and its correlation

Abstract Background Finding out the key reproductive performance factors, affecting piglets weaned per sow per year (PSY) can improve the production efficiency and profitability of pig farms. The objective was to understand the actual distribution of different production factors and PSY of breeding pig farms, analyze the correlation to find the main production factors affecting PSY, and formulating a Production Efficiency Improvement Plan in practice. Data included 603 breeding pig farms from September 28, 2020 to September 26, 2021. Regression analysis was used to evaluate the relationship between PSY and key production factors, and the characteristics of total pig farms versus high performance (HP) pig farms (the production performance was in the top 10%) or top 5% pig farms were compared. Spearman’s rank correlation coefficient was used to analyze the correlation between production factors and find the factors related to PSY. Non-linear support vector regression (NL-SVR) was used to analyze the personalized PSY improvement through a various change of the four key factors. Results The median distribution of 15 production factors and PSY in total pig farms were different from those of HP farms. All of data were distributed nonlinearly. Mating rate within 7 days after weaning (MR7DW), farrowing rate (FR), number of piglets born alive per litter (PBAL) and number of weaned piglets per litter (WPL) were moderately correlated with PSY, and the correlation coefficients were 0.5058, 0.4427, 0.3929 and 0.3839, respectively. When the four factors in NL-SVR changed in medium (0.5 piglet or 5%) or high level (1.0 piglet or 10%), PSY can be increased by more than 0.5. Conclusion NL-SVR model can be used to analyze the impact of changes in key production factors on PSY. By taking measures to improve MR7DW, FR, PBAL and WPL, it may effectively improve the current PSY and fully develop the reproductive potential of sows.

21. Anti-Cancer Effects of a New Herbal Medicine PSY by Inhibiting the STAT3 Signaling Pathway in Colorectal Cancer Cells and Its Phytochemical Analysis.

Colorectal cancer (CRC) is an inflammation-associated common cancer worldwide. Paejang-san and Mori Cortex Radicis have been traditionally used for treating intestinal inflammatory diseases in Korea and China. In the present study, we developed a new herbal formula as an alternative to CRC treatments, which is composed of two main components of Paejangsan (Patriniae Radix (Paejang in Korean) and Coix Seed (Yiyiin in Korean)), and Mori Cortex Radicis (Sangbekpi in Korean) based on the addition and subtraction theory in traditional medicine, hence the name PSY, and explored the potential therapeutic effects of the new formula PSY in human CRC cells by analyzing viability, cell cycle and apoptosis. We found that PSY ethanol extract (EtOH-Ex), but not water extract, significantly suppressed the viability of human CRC cells, and synergistically decreased the cell proliferation compared to each treatment of Patriniae Radix and Coix Seed extract (PY) or Mori Cortex Radicis extract (S), suggesting the combination of PY and S in a 10-to-3 ratio for the formula PSY. PSY EtOH-Ex in the combination ratio reduced cell viability but induced cell cycle arrest at the G2/M and sub-G1 phases as well as apoptosis in CRC cells. In addition, the experimental results of Western blotting, immunofluorescence staining and reporter assays showed that PSY also inhibited STAT3 by reducing its phosphorylation and nuclear localization, which resulted in lowering STAT3-mediated transcriptional activation. In addition, PSY regulated upstream signaling molecules of STAT3 by inactivating JAK2 and Src and increasing SHP1. Moreover, the chemical profiles of PSY from UPLC-ESI-QTOF MS/MS analysis revealed 38 phytochemicals, including seven organic acids, eight iridoids, two lignans, twelve prenylflavonoids, eight fatty acids, and one carbohydrate. Furthermore, 21 potentially bioactive compounds were highly enriched in the PSY EtOH-Ex compared to the water extract. Together, these results indicate that PSY suppresses the proliferation of CRC cells by inhibiting the STAT3 signaling pathway, suggesting PSY as a potential therapeutic agent for treating CRC and 21 EtOH-Ex-enriched phytochemicals as anti-cancer drug candidates which may act by inhibiting STAT3. 22. Transcript expression level analysis of phytoene synthase and phytoene desaturase associated with β-carotene content in selected Kenyan Bitter melon.

Background: Bitter melon (Momordica charantia L.) is a widely cultivated food and medicinal plant native to the world's subtropics and tropics. Constraints affecting cultivation of Bitter melon affect productivity of β-carotene. Knowing the mechanism that controls the transcription of the β-carotene biosynthesis genes in Bitter melon will be of great value in improving the yield of this important metabolite. Methods and Results: The expressions of β-carotene biosynthetic genes such as Phytoene Desaturase (PDS) and Phytoene Synthase (PSY) were evaluated in Bitter melon accessions 'GBK027049', 'NS1026', 'Mahy-ventura', '453B' and 'Sibuka532'. Transcript expression level analysis of PSY and PDS, and amount of β-carotene in leaf, stem, and fruit, were determined using quantitative polymerase chain reaction and high-performance liquid chromatography (HPLC). Root transcript expression was used as a negative control for determining relative fold change in other tissues. Expression of PSY in fruit (6 to 27-fold compared to the control) was higher than in the other organs for all accessions. This was also the case of PDS expression (10 to 29-fold compared to the control). Leaves had the highest β-carotene concentration (17.92–45.35 µg∙g
−1 ); there was no difference between stems (5.67–12.75 µg∙g−1 ) and fruit (6.18–12.53 µg∙g−1 ). The highest β-carotene content was in accessions 'GBK027049' (12.53–45.35 µg∙g−1 ) and '453B' (6.18–32.09 µg∙g−1 ). The PSY and PDS expressions were positively correlated with amount of β-carotene in leaves, stems, and fruits. Conclusion: Bitter melon leaves, especially those of 'GBK027049' and '453B' accessions, are an alternative to alleviate the β-carotene deficiencies in the world and especially in Africa. The aim of this paper is to expand upon previous research into the country of origin effect through the use of empirical data by formulating an analysis method which allows the measurement of the extent of country of origin effect by the proxy of K-pop musicians associated with country of origin image and to propose an alternative framework which provides an explanation as to the discrepancies between empirical data and prior research. Our analysis results reveal that the impact of country of origin associated with ‘PSY’ and ‘BTS’ on the automobile sales of ‘Hyundai Sonata’ was significant in the U.S. market. An asymmetric country of origin effect was found vis-à-vis American and Japanese brands.

BackgroundPathogens often secrete molecules that mimic those present in the plant host. Recent studies indicate that some of these molecules mimic plant hormones required for development and immunity.Scope and conclusionThis Viewpoint reviews the literature on microbial molecules produced by plant pathogens that functionally mimic molecules present in the plant host. This article includes examples from nematodes, bacteria and fungi with emphasis on RaxX, a microbial protein produced by the bacterial pathogen Xanthomonas oryzae pv. oryzae. RaxX mimics a plant peptide hormone, PSY (plant peptide containing sulphated tyrosine). The rice immune receptor XA21 detects sulphated RaxX but not the endogenous peptide PSY. Studies of the RaxX/XA21 system have provided insight into both host and pathogen biology and offered a framework for future work directed at understanding how XA21 and the PSY receptor(s) can be differentially activated by RaxX and endogenous PSY peptides.

Phytoene synthase (PSY) catalyzes the first committed step in the carotenoid biosynthesis pathway and is a major rate-limiting enzyme of carotenogenesis. PSY is highly regulated by various regulators and factors to modulate carotenoid biosynthesis in response to diverse developmental and environmental cues. Because of its critical role in controlling the total amount of synthesized carotenoids, PSY has been extensively investigated and engineered in plant species. However, much remains to be learned on its multifaceted regulatory control and its catalytic efficiency for carotenoid enrichment in crops. Here, we present current knowledge on the basic biology, the functional evolution, the dynamic regulation, and the metabolic engineering of PSY. Prezentowany artykuł zajmuje się analizą motywu psa w Ap 22,15 w kontekście kulturowym starożytnej Grecji. Analiza stanu badań pokazała, że egzegeci w interpretacji terminu οἱ κύνες odwołują się przede wszystkim do tekstów paralelnych znajdujących się w samej Biblii (oraz literaturze pozabiblijnej). Taka interpretacja nie wyczerpuje jednak bogactwa znaczeniowego metafory psa. Ten sam tekst czytany przez chrześcijan, którzy mieli za sobą doświadczenie kultury hellenistycznej otwiera się na nowe znaczenia. Zdaniem autora artykułu chrześcijanie wywodzący się z kultury hellenistycznej mogli widzieć motyw psa w powiązaniu z zalotami o charakterze pederastycznym, gdzie stanowił on uosobienie starszego partnera w tej relacji – tzw. erastesa. Takie rozumienie metafory psa ma u swoich fundamentów odwołanie się do polowania, które stanowiło ważny element życia antycznych Greków. W literaturze greckiej oraz w malarstwie wazowym, przedstawiając homoseksualne zaloty starszych – erastesów względem młodszych – eromenosów odwoływano się do idei polowania. Widoczne jest to w ikonografii, gdzie sceny ukazujące pederastię często zawierają motyw psa, oraz literaturze, gdzie wysiłki erastesa ukazane są na podobieństwo polowania. Autor Apokalipsy umieszczając „psy” pośród kategorii ludzi wykluczonych z udziału w Mieście Świętym pokazał, że praktyki pederastyczne zamykają dostęp do wspólnoty nowego ludu Bożego.

Purpose: To investigate the clinical utility of transperineal template-guided saturation prostate biopsy (TPB) for risk stratification after transrectal ultrasound (TRUS)-guided biopsy. Materials and Methods: We retrospectively reviewed 155 patients who underwent TPB after previously negative results on TRUS-guided biopsy (n=58) or who were candidates for active surveillance (n=97) fulfilling the PRIAS criteria between May 2017 and November 2018. The patients' clinicopathologic data were reviewed, and the detection of clinically significant cancer (CSC) and upgrading of Gleason grade were identified. Results: The patients' median age and pre-TPB prostate-specific antigen (PSA) value were 65.0 years and 5.74 ng/mL, respectively. A median of 36 biopsy cores was obtained in each patient, with a median TPB core density of 0.88 cores/cm3. Of the 58 males with a previous negative result on TRUS-guided biopsy, prostate cancer (PCa) was detected in 17 males (29.3%), including 8 with CSC. Of the 97 patient candidates for active surveillance, upgrading of the Gleason grade was identified in 31 males (32.0%), 20 with a Gleason grade of 7 (3+4), 6 with a Gleason grade of 7 (4+3), and 5 with a Gleason grade of 8 (4+4). The overall complication rate was 14.8% (23/155), and there were no Clavien–Dindo grade 3 to 5 complications. Conclusions: TPB helps to stratify the risk of PCa that was previously missed or underdiagnosed by TRUS-guided biopsy. TPB might be used as a diagnostic tool to determine risk classification and to help counsel patients with regard to treatment decisions.

Abstract Background Farnesol is an acyclic sesquiterpene alcohol present in the essential oils of various plants in nature. It has been reported to be valuable in medical applications, such as alleviation of allergic asthma, gliosis, and edema as well as anti-cancerous and anti-inflammatory effects. Coenzyme Q10 (CoQ10), an essential cofactor in the aerobic respiratory electron transport chain, has attracted growing interest owing to its clinical benefits and important applications in the pharmaceutical, food, and health industries. In this work, co-production of (E,E)-farnesol (FOH) and CoQ10 was achieved by combining 3 different exogenous terpenes or sesquiterpene synthase with the RNA interference of psy (responsible for phytoene synthesis in Rhodobacter sphaeroides GY-2). Results FOH production was significantly increased by overexpressing exogenous terpene synthase (TPS), phosphatidylglycerophosphatase B (PgpB), and sesquiterpene synthase (ATPS), as well as RNAi-mediated silencing of psy coding phytoene synthase (PSY) in R. sphaeroides strains. Rs-TPS, Rs-ATPS, and Rs-PgpB respectively produced 68.2%, 43.4%, and 21.9% higher FOH titers than that of the control strain. Interestingly, the CoQ10 production of these 3 recombinant R. sphaeroides strains was exactly opposite to that of FOH. However, CoQ10 production was almost unaffected in R. sphaeroides strains modified by psy RNA interference. The highest FOH production of 40.45 mg/L, which was twice as high as that of the control, was obtained from the TPS-PSYi strain, where the exogenous TPS was combined with the weakening of the phytoene synthesis pathway via psy RNA interference. CoQ10 production in TPS-PSYi, ATPS-PSYi, and PgpB-PSYi was decreased and lower than that of the control strain. Conclusions The original flux that contributed to phytoene synthesis was effectively redirected to provide precursors toward FOH or CoQ10 synthesis via psy RNA interference, which led to weakened carotenoid synthesis. The improved flux that was originally involved in CoQ10 production and phytoene synthesis was redirected toward FOH synthesis via metabolic modification. This is the first reported instance of FOH and CoQ10 co-production in R. sphaeroides using a metabolic engineering strategy.

As demanded by Argentinian cartoonist Quino's (2014) character Libertad (meaning freedom), the Latin American tradition of Liberation Psychology has explicitly called for a decolonising praxis since the 1970s. Such call has implications for training courses in psychology and other fields related to health and wellbeing, such as psychiatrists or social workers. However, there are various challenges in translating a decolonial rhetoric into concrete practice within the classroom, research and practice placements. This paper presents an account of experience and dialogues between two liberatory and critical community psychology educators in universities from the UK and Ecuador. Based on an international participatory roundtable on decolonisation and a pedagogical exchange between their students, the authors discuss the limitations and possibilities of engaging in a 'new praxis' that is both decolonial and liberatory. In previous studies, people paid more attention to the influence of news on stock price, but neglected the role of investors in stock trading. News influences investors as they make investment decisions. In this article, we look at how news affects investor sentiment. We selected PSY (psychology line), commonly used in stock investment, as an indicator to measure investor sentiment. Based on the SVM model, we establish a model that USES news texts to predict changes in investor sentiment. Our model can achieve high accuracy, which is helpful for guiding the prediction of investor sentiment in the stock market and making correct decisions. After comparing several indicators and models, we found that the investor sentiment prediction model based on SVM has a high accuracy, which can analyze the investor sentiment of the day according to the stock related news and guide investors to make correct investment decisions. Krabbe disease (KD) is a childhood leukodystrophy with no cure currently available. KD is due to a deficiency of a lysosomal enzyme called galactosyl-ceramidase (GALC) and is characterized by the accumulation in the nervous system of the sphingolipid psychosine (PSY), whose cytotoxic molecular mechanism is not fully known yet. Here, we study the expression of some fundamental autophagy markers (LC3, p62, and Beclin-1) in a KD murine model [the twitcher (TWI) mouse] by immunohistochemistry and Western blot. Moreover, the autophagy molecular process is also shown in primary fibroblasts from TWI and WT mice, with and without PSY treatment. Data demonstrate that large p62 cytoplasmic aggregates are present in the brain of both early and late symptomatic TWI mice. p62 expression is also upregulated in TWI sciatic nerves compared to that measured for WT nerves. In vitro data suggest that this effect might not be fully PSY-driven. Finally, we investigate in vitro the capability of autophagy inducers (Rapamycin, RAP and Resveratrol, RESV) to reinstate the WT phenotype in TWI cells. We show that RAP administration can partially restore the autophagy markers levels, while RESV cannot, indicating a line along which new therapeutic approaches can be developed. • Autophagy is dysregulated in Twitcher mouse (TWI) model of Krabbe disease. • Large p62 cytoplasmic aggregates are present in TWI brains and p62 is upregulated in TWI sciatic nerves. • In vitro data suggest that this autophagy dysregulation might not be fully psychosine driven. • Rapamycin can partially restore the autophagy marker levels in TWI cells. Psy's Gangnam Style was the first YouTube video ever to reach one and then two billion hits. This paper examines how and why it succeeded in speaking to and for so many in its time. Psy's Gangnam Style is best understood as a multi-media, digital representation of the metropolitan experience, a cityscape. If traditional pastoral represents a yearning to flee to the country to escape the alienation of the city, Psy's song and dance is an invitation to recreate an alternative pastoral in the city. Daily life is transposed into leisure as festivity becomes possible in city spaces planned and developed for entirely different purposes. In the tradition of the modernist flaneur, Psy narrates the city from the perspective of the scavenger or ragpicker making his way through the glorious detritus of contemporary commodity culture. Carnival, mass ornament, flaneur--these are the tropes I borrow from classical cultural studies to examine Gangnam Styles different facets of engagement with urban issues. In the conclusion, I lean on Jacques Ranciere on the distribution of the sensible to argue that it is ultimately through the expansion of the aesthetic through play that Gangnam Style seeks to recuperate the right to the city. Carotenoids exert beneficial effects on human health through their excellent antioxidant activity. To increase carotenoid productivity in the marine Pennales Phaeodactylum tricornutum, we genetically engineered the phytoene synthase gene (psy) to improve expression because RNA-sequencing analysis has suggested that the expression level of psy is lower than other enzyme-encoding genes that are involved in the carotenoid biosynthetic pathway. We isolated psy from P. tricornutum, and this gene was fused with the enhanced green fluorescent protein gene to detect psy expression. After transformation using the microparticle bombardment technique, we obtained several P. tricornutum transformants and confirmed psy expression in their plastids. We investigated the amounts of PSY mRNA and carotenoids, such as fucoxanthin and β-carotene, at different growth phases. The introduction of psy increased the fucoxanthin content of a transformants by approximately 1.45-fold relative to the levels in the wild-type diatom. However, some transformants failed to show a significant increase in the carotenoid content relative to that of the wild-type diatom. We also found that the amount of PSY mRNA at log phase might contribute to the increase in carotenoids in the transformants at stationary phase.

Over the last decade, Western publics have gradually caught on to the Kpop phenomenon; the Korean Wave has arrived on European and North American shores and shows no signs of receding. Heightened interest has corresponded with increased mainstream media coverage, both among news and entertainment outlets. This article analyzes mainstream media coverage of the Korean Wave from 2009 to 2019, including an examination of overall trends in K-pop framing over time. This analysis suggests that coverage of K-pop in Western media has proceeded through four distinct stages of development: 1) Introductory Stage, 2) Gangnam Style Stage, 3) Korean Wave Stage, and 4) Mainstreaming Stage. This article also examines how the changing portrayal of K-pop for general audiences has corresponded with a similar evolution in portrayals of South Korea and Korean society as a whole. This essay explores the political implications of the flash mob dance in Dhaka, Bangladesh performed in response to the 2012 global viral sensation of South Korean rapper PSY’s “Gangnam Style” music video. The global fame of “Gangnam Style” has much to do with its success online and in the U.S. popular music industry. It, however, also solicited suspicion from popular culture critics that the images of comical PSY worked successfully thanks to unchecked consumption of the racial stereotypes of Asian men. While recognizing these problems as more than valid, this essay simultaneously calls for a more transnational and inter-Asian understanding of the material to argue for a productive quality of PSY’s performance. Using “refraction” as a mode of thinking about inter-Asian circulation of pop culture, this essay considers the flash mob performed in Dhaka, Bangladesh as an important yet underexplored case study that shows different performative practices associated with “Gangnam Style” deeply rooted in historicity of colonialism and nationalism. The case study shows that the circulation of “Gangnam Style” materialized through a performance in Dhaka enlarged contemporary discourse among young urban Bangladeshi spectators around Bangladeshiness and its cultural identity. This complicated an easy assumption about “Gangnam Style” and its success in the U.S. mainstream pop culture, while simultaneously displacing the Bangladeshi cultural subjects from the immobile position of “the Other.” [ABSTRACT FROM AUTHOR]

Während Frank am Stil als "Handschrift des stilschöpferischen Individuums" gleichwohl festhält, sieht sich Elisa Ronzheimer anhand ihres Materials gezwungen, sowohl diesen Topos wie die Logik von Norm und Abweichung zu verabschieden. Was bei Frank Kombinatorik heißt, muss radikalisiert und neu gedacht werden, um dem international erfolgreichen südkoreanischen Musik-Video "Gangnam Style" gerecht zu werden, weil in diesem Fall das Original selbst schon parodistisch ist. Ronzheimer schlägt vor, das Individualitätsparadigma zu ersetzen durch ein Verständnis von Stil als kollektiver und partizipatorischer Praxis der Figuration und Defiguration. Damit scheint aber auch die Möglichkeit einer Art Wiederkehr der rhetorischen Regelpoetik am oder nach dem 'Ende' des Stils auf. Die Aussicht, dass heute Bedingungen herrschen, unter denen es eine Praxis, ein 'Üben' geben könnte, das nicht mehr automatisch Abweichungen produziert, sondern zur Homogenisierung tendiert, hat Moritz Baßler jüngst unter dem Titel "Der Neue Midcult" entfaltet.

Cassava (Manihot esculenta Crantz) is a predominant food security crop in several developing countries. Its storage roots, rich in carbohydrate, are deficient in essential micronutrients, including provitamin A carotenoids. Increasing carotenoid content in cassava storage roots is important to reduce the incidence of vitamin A deficiency, a public health problem in sub-Saharan Africa. However, cassava improvement advances slowly, mainly due to limited information on the molecular factors influencing β-carotene accumulation in cassava. To address this problem, we performed comparative transcriptomic and untargeted metabolic analyses of roots and leaves of eleven African cassava landraces ranging from white to deep yellow colour, to uncover regulators of carotenoid biosynthesis and accumulation with conserved function in yellow cassava roots. Sequence analysis confirmed the presence of a mutation, known to influence β-carotene content, in PSY transcripts of deep yellow but not of pale yellow genotypes. We identified genes and metabolites with expression and accumulation levels significantly associated with β-carotene content. Particularly an increased activity of the abscisic acid catabolism pathway together with a reduced amount of L-carnitine, may be related to the carotenoid pathway flux, higher in yellow than in white storage roots. In fact, NCED_3.1 was specifically expressed at a lower level in all yellow genotypes suggesting that it could be a potential target for increasing carotenoid accumulation in cassava. These results expand the knowledge on metabolite compositions and molecular mechanisms influencing carotenoid biosynthesis and accumulation in cassava and provide novel information for biotechnological applications and genetic improvement of cassava with high nutritional values. • Abscisic acid catabolism activity is higher in carotenoid-rich cassava storage roots. • Low NCED_3.1 and L-carnitine content is associated to carotenoid accumulation in cassava roots. • The psy gene is mutated in deep yellow but not in pale yellow cassava genotypes. In this article I provide a brief history of Hallyu, noting its mix of cultural and economic characteristics, and then analyse the reception of the phenomenon in the Arab Middle East by considering fan activity on social media platforms. I then conclude by discussing the cultural, political and economic benefits of Hallyu to Korea and indeed the wider world. For the sake of convenience, I will be using the term Hallyu (or Neo-Hallyu) rather than the Korean Wave throughout my paper.

The ‘psy’ sciences emerged from the tangled roots of philosophy, physiology, biology and medicine, and these origins have produced heterogeneous fields. Scientists in these areas work in a complex, overlapping ecology of fields that results in the constant co-presence of dissonant theories, methods and research objects. This raises questions regarding how conceptual clarity is maintained. Using the optical metaphor ‘depth of field’, I show how researchers in all fields marginalize potential threats to routine scientific work by framing them as either too broad and imprecise or too narrow and technical.
