A new species of the genus Oreolalax Myers & Leviton, 1962 is described from Sichuan Province, southwest China. Molecular phylogenetic analyses based on mitochondrial gene sequences clustered the new species as an independent clade nested with O. rugosus, O. liangbeiensis, and O. major. The new species could be distinguished from its congeners by a combination of the following characters: body size moderate (39.8–52.8 mm in male); head broad; tympanum absent; interorbital region with dark triangular pattern; 1/3 toes webbed, with broad lateral fringes, belly smooth, brown yellow or medium yellow scattered variable brown spots; skin on dorsum relatively rough with fine tiny and large warts granules; middle pectoral glands are evident in males; flanks with dark-brown warts granules; upper surface of limbs with dark bars; and iris orange above and creamy-white below. The new species inhabits subtropical alpine scrub and swamp.

Abstract Deoxyribonucleotide (DNA) is uniquely programmable and biocompatible, and exhibits unique appeal as a biomaterial as it can be precisely designed and programmed to construct arbitrary shapes. DNA hydrogels are polymer networks comprising cross-linked DNA strands. As DNA hydrogels present programmability, biocompatibility, and stimulus responsiveness, they are extensively explored in the field of biomedicine. In this study, we provide an overview of recent advancements in DNA hydrogel technology. We outline the different design philosophies and methods of DNA hydrogel preparation, discuss its special physicochemical characteristics, and highlight the various uses of DNA hydrogels in biomedical domains, such as drug delivery, biosensing, tissue engineering, and cell culture. Finally, we discuss the current difficulties facing DNA hydrogels and their potential future development. Graphical Abstract

Abstract Background A stent with characteristics of a hybrid design may have advantages in improving the patency of symptomatic iliofemoral vein obstruction. This study assessed the safety and effectiveness of the V-Mixtent Venous Stent in treating symptomatic iliofemoral outflow obstruction. Methods Eligible patients had a Clinical-Etiologic-Anatomic-Physiologic (CEAP) C classification of ≥ 3 or a Venous Clinical Severity Score (VCSS) pain score of ≥ 2. The primary safety endpoint was the rate of major adverse events within 30 days. The primary effectiveness endpoint was the 12-month primary patency rate. Secondary endpoints included changes in VCSS from baseline to 6 and 12 months, alterations in CEAP C classification, Chronic Venous Disease Quality of Life Questionnaire (CIVIQ-14) scores at 12 months, and stent durability measures. Results Between December 2020 and November 2021, 171 patients were enrolled across 15 institutions. A total of 185 endovenous stents were placed, with 91.81% of subjects receiving one stent and 8.19% receiving 2 stents. CO2 geological storage is a guaranteed technology for achieving carbon neutrality. The offshore deposit basin is far away from the human activity area, and the implementation of a CO2 geological storage is more advantageous than the land sedimentary basin. To find out the CO2 storage potential in the Weixinan sag of the Beibu Gulf Basin, based on the 8 indicators such as caprock burial depth, the maximum thickness of single layer mudstone, mud-soil ratio, fault density and reservoir porosity, formation thickness, sand-soil ratio, permeability, etc., it has established a reservoir evaluation system for the secondary structure unit of the basin. Combined with the core physical property data of lays and 3D geological models, the reservoir and caprock of the Weixinan sag are optimized, and the effective storage potential is evaluated with the US-DOE method. The results show that within 800−3000 meters below the sea floor, in the Weixinan sag, there are two sets of reservoir-cap combinations suitable for CO2 storage. Combination 1: in Neogene, the neritic mudstone of the first member of the Jiaowei Formation is the cap rock, and the neritic sandstone of the second member of the Jiaowei Formation and the Xiayang Formation is the reservoir, and the effective storage capacity is 0.749 billion tons. Combination 2: in the Paleogene, the deep lake mudstone in the second member of the Weizhou Formation is the cap rock, and the sandstone in the third member of the Weizhou Formation is the reservoir, and the effective storage capacity is 0.374 billion tons. In the Weixinan sag, two sets of reservoir-cap assemblages suitable for CO2 storage in the saline aquifers are selected, with an effective storage capacity of 1.123 billion tons. According to the evaluation of the annual CO2 emission of 70 million tons in the west of Guangdong Province, it can meet the CO2 emission reduction needs of the region for more than 16 years. Offshore geological storage is a potential path for the green and sustainable development of coastal heavy chemical clustering areas and to achieve the goal of carbon neutrality.

Tissue repair and regeneration have long been a focus of research, with growing evidence highlighting the role played by immunomodulation. The repair of tissue cells can be significantly promoted through the localized immunomodulation of tissues. Hydrogels are increasingly used in this field due to their biocompatibility, versatility, and relative ease of fabrication. This Review highlights the critical role of inflammation and immune development in the complex process of tissue repair after injury, highlighting the dynamic interactions between immune cells that ultimately influence the outcome of tissue healing. We also summarize the precise functions of immune cells and their responses in various tissue injury scenarios, which are critical for harnessing the full potential of immunomodulatory strategies to promote tissue repair. We discuss recent advances in drug-carrying hydrogels, which offer promising avenues for tissue regeneration with precise control of the immune environment for constructive tissue repair. Further advances in our understanding of the mechanisms driving inflammation and immune cell behavior during tissue injury are expected to foster the development of increasingly effective hydrogel-based immunomodulatory strategies to promote tissue repair and regeneration.

Post-translational modifications play crucial roles in regulating protein functions and stabilities. PTEN is a critical tumor suppressor involved in regulating cellular proliferation, survival, and migration processes. However, dysregulation of PTEN is common in various human cancers. PTEN stability and activation/suppression have been extensively studied in the context of tumorigenesis through inhibition of the PI3K/AKT signaling pathway. PTEN undergoes various post-translational modifications, primarily including phosphorylation, acetylation, ubiquitination, SUMOylation, neddylation, and oxidation, which finely tune its activity and stability. Generally, phosphorylation modulates PTEN activity through its lipid phosphatase function, leading to altered power of the signaling pathways. Acetylation influences PTEN protein stability and degradation rate. SUMOylation has been implicated in PTEN localization and interactions with other proteins, affecting its overall function. Neddylation, as a novel modification of PTEN, is a key regulatory mechanism in the loss of tumor suppressor function of PTEN. Although current therapeutic approaches focus primarily on inhibiting PI3 kinase, understanding the post-translational modifications of PTEN could help provide new therapeutic strategies that can restore PTEN’s role in PIP3-dependent tumors. The present review summarizes the major recent developments in the regulation of PTEN protein level and activity. We expect that these insights will contribute to better understanding of this critical tumor suppressor and its potential implications for cancer therapy in the future.

In this paper, we study the fixed-time group consensus problem for second-order multi-agent systems under the influence of actuator faults and external disturbances. First, we design a finite-time observer to quickly and accurately evaluate the effects of these disturbances and faults. Then a new non-singular terminal sliding mode surface is constructed, and a fixed-time fault-tolerant controller is designed based on the introduction of a dynamic event-triggering mechanism. The designed control protocol can effectively avoid singular phenomena and compensate and suppress the effects of these faults and disturbance information on the system to achieve fault tolerance. The Lyapunov function is applied to prove that the convergence time of the system is not influenced by the initial value setting, and the Zeno phenomenon is excluded. Finally, the performance of the proposed method is compared with previous methods through numerical simulations to prove its superiority.

This dataset presents perceived values and socioeconomic indicators collected in Siaya, a rural county in Kenya in 2022. The data was obtained from 300 household surveys and group interviews conducted in six sub-counties across eleven villages. Socioeconomic data were collected with a special focus on climate change vulnerability. Information on housing, health, water accessibility and usage, electricity accessibility and usage, extreme weather events, community service, and information accessibility were mapped across survey questions. The user-perceived value (UPV) game – a perception-based surveying approach – was used to elicit local communities’ needs and perceptions of climate change challenges. The UPV game involves asking interviewees to select which graphically depicted items would be most necessary in different situations and probing them for the reasons behind their choices (why-probing). The data was collected in two languages (Dholuo and English) and then translated into English. These surveys and interviews were conducted to better understand the needs of rural Kenyan communities and their perceptions of climate change, with the aim to identify ways to build resilience. Kenyan policymakers can use the dataset to inform county-level energy and development plans, while researchers and development practitioners can use the dataset to better design their research and programmes to reflect local needs and values.

AbstractContext The Tongmai Yangxin pill (TMYX) has potential clinical effects on no-reflow (NR); however, the effective substances and mechanisms remain unclear.Objective This study evaluates the cardioprotective effects and molecular mechanisms of TMYX against NR.Materials and methods We used a myocardial NR rat model to confirm the effect and mechanism of action of TMYX in alleviating NR. Sprague-Dawley (SD) rats were divided into Control (Con), sham, NR, TMYX (4.0 g/kg), and sodium nitroprusside (SNP, 5.0 mg/kg), and received their treatments once a day for one week. In vitro studies in isolated coronary microvasculature of NR rats and in silico network pharmacology analyses were performed to reveal the underlying mechanisms of TMYX and determine the main components, targets, and pathways of TMYX, respectively.Results TMYX (4.0 g/kg) showed therapeutic effects on NR by improving the cardiac structure and function, reducing NR, ischemic areas, and cardiomyocyte injury, and decreasing the expression of cardiac troponin I (cTnI). Moreover, the mechanism of TMYX predicted by network pharmacology is related to the HIF-1, NF-κB, and TNF signaling pathways. In vivo, TMYX decreased the expression of MPO, NF-κB, and TNF-α and increased the expression of GPER, p-ERK, and HIF-1α. In vitro, TMYX enhanced the diastolic function of coronary microvascular cells; however, this effect was inhibited by G-15, H-89, L-NAME, ODQ and four K+ channel inhibitors.Conclusions TMYX exerts its pharmacological effects in the treatment of NR via multiple targets. However, the contribution of each pathway was not detected, and the mechanisms should be further investigated.

Abstract Background and aims The increasing prevalence of metabolic and cardiovascular diseases poses a significant challenge to global healthcare systems. Regular physical activity (PA) is recognized for its positive impact on cardiovascular risk factors. This study aimed to investigate the relationship between moderate-to-vigorous physical activity (MVPA), sedentary behavior (SB), and abdominal aortic calcification (AAC) using data from the National Health and Nutrition Examination Survey (NHANES). Methods The study used data from NHANES participants aged 40 and above during the 2013–2014 cycle. AAC scores were assessed using the Kauppila scoring system, and MVPA and SB were self-reported. Sociodemographic variables were considered, and multivariable linear regression models were used to analyze associations between MVPA, SB, and AAC scores. Subgroup analyses were conducted based on age, sex, BMI, hypertension, and diabetes. Results The study included 2843 participants. AAC prevalence was higher in older age groups, smokers, and those with diabetes or hypertension. Lower socioeconomic status was associated with higher AAC prevalence. With the increase in the proportion of photovoltaic and wind power access, the scale and form of distribution networks are becoming more and more complex. The traditional single distribution network vulnerability assessment method is difficult to use to identify the vulnerable links in the distribution network. Therefore, this paper proposes a method for identifying and evaluating vulnerable links in distribution networks based on minimum discriminant information. First, considering the influence of distributed grid connection, an improved probabilistic power flow calculation method is proposed, which improves the calculation efficiency and accuracy. Second, considering the correlation degree, transmission capacity, and voltage stability of branches in the distribution network, the identification index of vulnerable lines is defined. Based on power quality and operating state, the identification index of vulnerable nodes in a distribution network is defined. Finally, based on the indicators of vulnerable nodes and vulnerable lines, the vulnerable links in the distribution network are comprehensively evaluated based on the principle of minimum discriminant information, and the vulnerable links of the entire distribution network are evaluated according to different degrees of vulnerability. The rationality and effectiveness of the proposed method are verified via an example analysis of actual power grid data.

Skeletal muscle satellite cells (SMSCs), a type of myogenic stem cell, play a pivotal role in postnatal muscle regeneration and repair in animals. Circular RNAs (circRNAs) are a distinct class of non-coding RNA molecules capable of regulating muscle development by modulating gene expression, acting as microRNAs, or serving as protein decoys. In this study, we identified circ_14820, an exonic transcript derived from adenosine triphosphatase family protein 2 (ATAD2), through initial RNA-Seq analysis. Importantly, overexpression of circ_14820 markedly enhanced the proliferation of goat SMSCs while concomitantly suppressing their differentiation. Moreover, circ_14820 exhibited predominant localization in the cytoplasm of SMSCs. Subsequent small RNA and mRNA sequencing of circ_14820-overexpressing SMSCs systematically elucidated the molecular regulatory mechanisms associated with circ_14820. Our preliminary findings suggest that the circ_14820-miR-206-CCND2 regulatory axis may govern the development of goat SMSCs. These discoveries contribute to a deeper understanding of circRNA-mediated mechanisms in regulating skeletal muscle development, thereby advancing our knowledge of muscle biology.

Green roof systems are regarded as a viable solution for mitigating urban environmental challenges and offering a multitude of environmental benefits. Currently, green roofs are increasingly being utilized for the management of rainwater runoff and wastewater. The integration of decentralized rainwater and sewage on-site treatment technology with urban green buildings is being gradually promoted. Green roofs can also be considered as a form of decentralized rainwater and sewage on-site technology, which holds great potential for widespread adoption in the future. Several studies have suggested that green roofs may serve as a potential source of pollutants; however, there are also studies that clearly demonstrate the efficient removal of nutrients and organic pollutants by green roofs. This article critically examines the existing literature on water treatment aspects associated with green roofs and elucidates their classification and operational mechanisms. Through an analysis of previous research cases, it becomes evident that both substrate and vegetation play a significant role in influencing the treatment performance of green roofs. By designing and configuring appropriate substrate and vegetation, green roofs can play a pivotal role in the purification of water quality. Finally, a brief outlook is presented for the future research directions of green roofs, with the anticipation that green roofs will feature more innovative and environmentally friendly designs, as well as expanded prospects for application.

Background and aims: Thromboangiitis obliterans (TAO), also known as Buerger’s disease, is a rare vasculitis. Observational epidemiology studies have suggested a relationship between the gut microbiota and TAO. However, due to confounding factors and reverse causality, the causal relationship remains unclear. Based on the assumption of their association, this study sought specific gut microbiota causally linked to TAO. Methods: The case–control study was conducted at the Xiangya Hospital of Central South University from November 2022 to January 2023 including twelve TAO patients and nine healthy controls. We conducted a Mendelian randomization (MR) study using summary statistics from a genome-wide association study (GWAS) of gut microbiota and TAO. Considering the scale and accessibility of the data, the MiBioGen consortium served as the exposure, whereas the FinnGen consortium GWAS study served as the outcome. Finally, we compared the results of the MR with those of the case–control studies. Results: The inverse variance weighted (IVW) (OR = 0.119, 95% CI: 0.021–0.688, p = 0.017) and maximum likelihood (ML) (OR = 0.121, 95% CI: 0.020–0.742, p = 0.022) estimates suggest that Ruminiclostridium 5 has a suggestive protective effect on TAO while the IVW (OR = 5.383, 95% CI: 1.128–25.693, p = 0.035) and ML (OR = 5.658, 95% CI: 1.142–28.021, p = 0.034) estimates suggest that Eubacterium (xylanophilum group) has a suggestive risk effect on TAO, and the ML (OR = 0.055, 95% CI: 0.004–0.755, p = 0.030) estimates suggest that Lachnospira has a suggestive protective effect on TAO. No significant heterogeneity of instrumental variables or horizontal pleiotropy was found. The results of the case–control study showed that the TAO had a lower relative abundance of Ruminiclostridium 5 (p = 0.015) and Lachnospira (p = 0.048), and a higher relative abundance of Eubacterium (xylanophilum group) (p = 0.029) than the healthy controls. These results were consistent with the MR analysis. Conclusions: Our study demonstrates that Ruminiclostridium 5, Lachnospira, and Eubacterium (xylanophilum group) are causally related to TAO, suggesting their potential significance for the prevention and treatment of TAO.

BackgroundRupture of abdominal aortic aneurysm (rAAA) is a fatal event in the elderly. Elevated blood pressure and weakening of vessel wall strength are major risk factors for this devastating event. This present study examined whether the expression profile of mechanosensitive genes correlates with the phenotype and outcome, thus, serving as a biomarker for AAA development.MethodsIn this study, we identified mechanosensitive genes involved in AAA development using general bioinformatics methods and machine learning with six human datasets publicly available from the GEO database. Differentially expressed mechanosensitive genes (DEMGs) in AAAs were identified by differential expression analysis. Molecular biological functions of genes were explored using functional clustering, Protein-protein interaction (PPI), and weighted gene co-expression network analysis (WGCNA). According to the datasets (GSE98278, GSE205071 and GSE165470), the changes of diameter and aortic wall strength of AAA induced by DEMGs were verified by consensus clustering analysis, machine learning models, and statistical analysis. In addition, a model for identifying AAA subtypes was built using machine learning methods.Results38 DEMGs clustered in pathways regulating 'Smooth muscle cell biology' and 'Cell or Tissue connectivity'. By analyzing the GSE205071 and GSE165470 datasets, DEMGs were found to respond to differences in aneurysm diameter and vessel wall strength. Thus, in the merged datasets, we formally created subgroups of AAAs and found differences in immune characteristics between the subgroups. Finally, a model that accurately predicts the AAA subtype that is more likely to rupture was successfully developed.ConclusionWe identified 38 DEMGs that may be involved in AAA. This gene cluster is involved in regulating the maximum vessel diameter, degree of immunoinflammatory infiltration, and strength of the local vessel wall in AAA. The prognostic model we developed can accurately identify the AAA subtypes that tend to rupture.

In this study, foxtail millet was used as raw material. The purpose of research was to investigate the changes of the properties and structures of four foxtail millet proteins, including albumin, glutelin, gliadin and globulin, after treatment by extrudating with α-amylase. The results demonstrated the increased water holding capacities, decreased foaming abilities, as well as the loss of the contents of several amino acids of these foxtail millet proteins under all extrusion conditions. Additionally, after extruding with enzyme, the contents of these proteins decreased, whereas the emulsification abilities increased. Among these proteins, the emulsification ability of gliadin was increased, while the foam stabilities of gliadin and glutelin were enhanced. Additionally, extruding with amylase resulted in less protein bands of albumin and globulin on gel. Furthermore, the secondary and tertiary structures of these foxtail millet proteins were comparable along extruding with enzyme, with minor blue shift. In conclusion, extruding with enzyme could improve the properties of foxtail millet proteins, with little influence on the secondary and tertiary structures. These findings indicate that the pregelatinization foxtail millet flour obtained with enzymatic extrusion processing technology could be used for an ideal functional food resource.

Abstract Background Hull colour is an important morphological marker for selection in seed production of foxtail millet. However, the molecular mechanisms underlying hull colour variation remain unknown. Results An F7 recombinant inbred line (RIL) population containing 215 lines derived from Hongjiugu × Yugu18 was used to analyze inheritance and detect the quantitative trait loci (QTL) for four hull colour traits using major gene plus polygene mixed inheritance analysis and composite interval mapping (CIM) in four environments. Genetic analysis revealed that the hull colour L* value (HCL*) was controlled by two major genes plus additive polygenes, the hull colour a* value (HCa*) was controlled by three major genes, the hull colour b* value (HCb*) was controlled by two major genes plus polygenes, and the hull colour C* value (HCC*) was controlled by four major genes. A high-density genetic linkage map covering 1227.383 cM of the foxtail millet genome, with an average interval of 0.879 cM between adjacent bin markers, was constructed using 1420 bin markers. Based on the genetic linkage map and the phenotypic data, a total of 39 QTL were detected for these four hull colour traits across four environments, each explaining 1.50%–49.20% of the phenotypic variation. Of these, six environmentally stable major QTL were co-localized to regions on chromosomes 1 and 9, playing a major role in hull colour. There were 556 annotated genes within the two QTL regions. Based on the functions of homologous genes in Arabidopsis and the Kyoto Encyclopedia of Genes and Genomes (KEGG) and Gene Ontology (GO) gene annotations, five genes were predicted as candidate genes for further studies. Conclusions This is the first study to use an inheritance model and QTL mapping to determine the genetic mechanisms of hull colour trait in foxtail millet. We identified six major environmentally stable QTL and predicted five potential candidate genes to be associated with hull colour. These results advance the current understanding of the genetic mechanisms underlying hull colour traits in foxtail millet and provide additional resources for application in genomics-assisted breeding and potential isolation and functional characterization of the candidate genes.

Quaternary Xiashu loess deposits are widely distributed in the middle and lower reaches of the Yangtze River. The Xiashu loess deposits provide an important archive for studying the area's paleoclimate. We analyzed the magnetic susceptibility of 239 bulk samples from two newly excavated Xiashu loess profiles: the Puwulu and Pancheng. Based on a comparison with previously studied profiles in this region, our new record shows a good correlation in the stratigraphy, with three well-developed paleosols (layers S1, S2, and S3) as the marked layers. This correlation was observed in the Taishanxincun, Laohushan, and Dagang profiles. Despite the good spatial correlation in stratigraphy, the well-developed paleosol layers were thicker than the underlying loess layers. The average sediment accumulation rates in the paleosol layers (6.2 cm/ka, n=19) were higher than those in the loess layers (5.1 cm/ka, n=17). This stratigraphic feature of the Xiashu loess deposits differs from that of the Chinese loess plateau in North China. Combined with previous studies on the origin of the Xiashu loess deposits, the new records suggest a proximal origin; however, the dominant contribution of sediment materials from the floodplains at the foot of Laoshan Mountain. Increased material flux through the primary stream runoff on the floodplain resulted in higher sediment accumulation rates in the paleosols. Based on sedimentary model analysis, the paleosol and loess layers in the Xiashu deposits are continuous sedimentary records on glacial-interglacial scales. After adjusting for fluctuating sediment deposition rates, the revised magnetic susceptibility displayed a clear cyclic variation pattern, which was interpreted as the intensity of pedogenesis. Power spectral analysis suggests that the cyclic pattern of variations shows a strong frequency in the 100 kyr band for both the Pancheng and Puwulu profiles. This may explain the paleoclimatic dynamics of the paleosols that formed. The most developed paleosol layers (S1, S2, S3, S4, and S5) correlated well with the orbital parameter eccentricity with clear 100-kyr ice cycles. Ice age terminations suggest sea-level changes and a far-reaching influence on precipitation in the middle and lower reaches of the Yangtze River. These observations indicate the relationship between the glacial cycle and soil development is closer than previously believed. The coincidence with eccentricity may largely support the idea that changes in global ice volume have promoted the formation of strongly developed paleosols along the incised valleys of the Yangtze River.

Abstract Background Chronic inflammatory pain (CIP) frequently coincides with depression among patients. The onset and development of pain and depression are associated with altered neural synaptic plasticity. Electroacupuncture (EA) can effectively relieve CIP and depression. However, the underlying mechanisms have not been fully illustrated. Objective To explore whether EA can relieve CIP and depression by regulating hippocampal synaptic plasticity, and the present study offers foundational evidence for the efficacy of EA in treating CIP‐related depression (CIPD). Methods Rats were divided into four groups: 0.9% normal saline group, complete Freund's adjuvant (CFA) group, CFA + duloxetine group, and CFA + EA group. Pain hypersensitivity was detected by mechanical withdrawal threshold and thermal paw withdrawal latency, and the depression level was gauged using the open field test, the sucrose preference test, and the forced swimming test. The morphology of the hippocampal neurons was observed using Nissl staining. The protein expression levels of synuclein (Syn), postsynaptic density protein‐95 (PSD‐95), brain‐derived neurotrophic factors (BDNFs), tyrosine‐protein kinase B (TrKB), p‐TrkB, cAMP response element binding protein (CREB), and p‐CREB were measured by western blotting and immunofluorescence staining. BDNF and TrkB mRNA expression were detected using quantitative real‐time polymerase chain reaction (PCR) (qRT‐PCR). The content of 5‐hydroxytryptamine (5‐HT) and γ‐aminobutyric acid (GABA) was detected using enzyme‐linked immunosorbent assay, and the glutamic acid (Glu) content was determined using the ultraviolet colorimetry method. The hippocampal neuron ultrastructure was observed using transmission electron microscopy. Results EA could alleviate CIP and related depressive behaviors as well as protect the hippocampal neuronal structure from damage and regulate 5‐HT/GABA/Glu levels in the hippocampus. Additionally, EA could significantly increase the expression of synapse‐associated proteins such as PSD‐95 and Syn by activating the BDNF/TrKB/CREB signaling pathway. Conclusion EA improves pain and depressive behaviors in CIPD rats, and the mechanism may be related to synaptic plasticity mediated by the BDNF/TrKB/CREB signaling pathway.

Complete and rapid healing of infected skin wounds remains a challenge in current clinical treatment. In this study, we prepared a self-healing injectable CK hydrogel by crosslinking two natural polysaccharides, carboxymethyl chitosan and oxidized konjac glucomannan, based on the Schiff base bond. To enhance the biological function of the hydrogel, we multi-functionalized hydrogen by loading it with berberine (BBR) and stem cell-derived exosomes (Exo), forming a composite hydrogel, CK@BBR&Exo, which could be injected directly into the wound through a needle and adhered to the wound. Furthermore, the self-healing properties of CK@BBR&Exo increased its usefulness and service life. Additionally, the drug-loaded CK@BBR&Exo hydrogel was versatile, inhibiting bacterial growth, regulating the inflammatory response, and promoting neovascularization in infected skin wounds, thus achieving the rapid healing of infected skin wounds. These results suggest that the CK@BBR&Exo-injectable self-healing hydrogel is an ideal dressing for treating infected skin wounds.

Abstract Background Central sensitization is one of the important mechanisms underlying neuropathic and radicular pain due to cervical spondylotic radiculopathy (CSR). Recent studies have shown that the calmodulin‐dependent protein kinase II (CaMKII)/cAMP‐response element binding protein (CREB)/brain‐derived neurotrophic factor (BDNF) signaling pathway mediates central sensitization through its involvement in spinal cord synaptic plasticity. Our group has previously found that electroacupuncture (EA) has a good analgesic effect on CSR. However, the central analgesic mechanism of EA for CSR is not yet clear. Methods The rats were randomly divided into Blank group, Sham‐operated group, CSR group, and EA group. We prepared the CSR rat model using the fish wire extrusion method. The behavioral and mechanical pain thresholds of the rats in each group were measured 5 days after successful modeling and 7 days after the intervention. The first intervention was started 5 days after successful modeling, and the EA group was treated by acupuncture at the bilateral LI4 and LR3 points on the same side as one group, connected to a G6805‐I electroacupuncture apparatus with continuous waves at 1.5 Hz. The remaining groups were not subjected to EA intervention. The treatment was administered once a day for 7 consecutive days and then executed. We used WB, immunofluorescence, and qRT‐PCR to detect the expression of CaMKII/CREB/BDNF signaling pathway‐related factors in the synaptic of rat spinal cord in each group. Results EA improved pain threshold and motor function in CSR rats, inhibited the expression of BDNF, P‐TrkB, CAMKII, and P‐CREB in spinal cord synapses, reduced the expression of pain factor c‐fos and postsynaptic membrane protein molecule neuroligin2, exerted a modulating effect on spinal cord synaptic plasticity in CSR rats, and suppressed the overactive synaptic efficacy. Conclusion EA mediates central sensitization and exerts analgesic effects on CSR by modulating spinal synaptic plasticity, which may be related to the inhibition of CaMKII/CREB/BDNF signaling pathway.

Abstract Background Renal excretion is one of the major routes of nanomaterial elimination from the body. Many previous studies have found that graphene oxide nanosheets are excreted in bulk through the kidneys. However, how the lateral size affects GO disposition in the kidneys including glomerular filtration, active tubular secretion and tubular reabsorption is still unknown. Results The thin, two-dimensional graphene oxide nanosheets (GOs) was observed to excrete in urine through the kidneys, but the lateral dimension of GOs affects their renal clearance pathway and renal injury. The s-GOs could be renal excreted via the glomerular filtration, while the l-GOs were predominately excreted via proximal tubular secretion at a much faster renal clearance rate than the s-GOs. For the tubular secretion of l-GOs, the mRNA level of basolateral organic anion transporters Oat1 and Oat2 in the kidney presented dose dependent increase, while no obvious alterations of the efflux transporters such as Mdr1 and Mrp4 mRNA expression levels were observed, suggesting the accumulation of l-GOs. During the GO renal elimination, mostly the high dose of 15 mg/kg s-GO and l-GO treatment showed obvious kidney injuries but at different renal compartment, i.e., the s-GOs induced obvious glomerular changes in podocytes, while the l-GOs induced more obvious tubular injuries including necrosis of renal tubular epithelial cells, loss of brush border, cast formation and tubular dilatation. The specifically tubular injury biomarkers KIM1 and NGAL were shown slight increase with mRNA levels in l-GO administrated mice. Conclusions This study shows that the lateral size of GOs affected their interactions with different renal compartments, renal excretion pathways and potential kidney injuries.

The accurate, quantitative, and objective prediction of the brain age for premature infants will contribute to the exploration of brain maturity and catch-up growth. Traditional approaches rely heavily on a pediatrician’s clinical experience, which makes the whole process time-consuming and labor-intensive. To solve this problem, we propose a deep learning-based brain age prediction model for preterm infants via neonatal MRI for this purpose, and it is called as BAPNET for short. First of all, we collected a specific dataset including MR images of 281 preterm infants. Then, a pretraining model (DeepBrainNet) is applied as the main backbone, and transfer learning is utilized to enhance the baseline model by making knowledge transfer from the ImageNet dataset. The proposal can be viewed as a specific prediction model by absorbing knowledge enhancement from peripheral visual features. On a test set of 70 preterm infants held out from the original dataset, 2D-BPANET achieved results with an mean square error (MAE) of 1.15 and the 95% - 95% content tolerance interval for a difference (prediction and ground truth) of [−3.82, 3.39], whereas 3D-BPANET achieved better results with an MAE of 1.8 and a difference of [0.51, 3.09]. Meanwhile, we leverage heatmaps to verify the consistency between hindbrain regions and cortical fold regions outputted by our model and the latest studies of brain development in preterm infants. In conclusion, BPANET demonstrates that deep learning can estimate brain maturity in preterm infants and provides a reference standard for preterm infant brain development, which could be applied as a promising tool.

Abstract Plastic surgery is a discipline that uses surgical methods or tissue transplantation to repair, reconstruct and beautify the defects and deformities of human tissues and organs. Three-dimensional (3D) bioprinting has gained widespread attention because it enables fine customization of the implants in the patient's surgical area preoperatively while avoiding some of the adverse reactions and complications of traditional surgical approaches. In this paper, we review the recent research advances in the application of 3D bioprinting in plastic surgery. We first introduce the printing process and basic principles of 3D bioprinting technology, revealing the advantages and disadvantages of different bioprinting technologies. Then, we describe the currently available bioprinting materials, and dissect the rationale for special dynamic 3D bioprinting (4D bioprinting) that is achieved by varying the combination strategy of bioprinting materials. Later, we focus on the viable clinical applications and effects of 3D bioprinting in plastic surgery. Finally, we summarize and discuss the challenges and prospects for the application of 3D bioprinting in plastic surgery. We believe that this review can contribute to further development of 3D bioprinting in plastic surgery and provide lessons for related research. Graphical Abstract

Fenugreek (Trigonella foenum-graecum L.) is a traditional medicinal plant for treating human diseases that is widely cultivated in many countries. However, the component and related metabolic pathways are still unclear. To understand the changes in expression of the component and related genes during seed development, this study employed metabolomic and transcriptomic analyses and integrative analysis to explore the metabolites and pathways involved in the growth of fenugreek. The antifungal activity of the fenugreek seeds was also analyzed. A total of 9499 metabolites were identified in the positive ion mode, and 8043 metabolites were identified in the negative ion mode. Among them, the main components were fatty acyls, prenol lipids, steroids, steroid derivatives, flavonoids, and isoflavonoids. Among these enriched pathways, the top 20 pathways were “flavone and flavonol biosynthesis”, “isoflavonoid biosynthesis”, and “flavonoid biosynthesis”. 3,7-Di-O-methylquercetin, flavonoids, pseudobaptigenin, isoflavonoids, methylecgonine, alkaloids, and derivatives were the most significantly upregulated metabolites. There were 38,137 differentially expressed genes (DEGs) identified via transcriptomic analysis. According to the KEGG pathway enrichment analysis, 147 DEGs were significantly enriched in “flavonoid biosynthesis”. Ten DEGs of the six key enzymes were found to be involved in three pathways related to flavonoid and alkaloid synthesis in fenugreek. The antifungal activity test revealed the inhibitory effect of the ethanol extract of fenugreek seeds on Alternaria tenuissima (Kunze)Wiltshire and Magnaporthe oryzae. These findings further prove that the use of botanical pesticides in fenugreek fruit has research value.

In this article, an observer-based adaptive non-singular fast-reaching terminal sliding mode control strategy is proposed to tackle the problem of actuator faults and uncertain disturbance in aerial robot systems. Firstly, a model of an aerial robot system is established through dynamic analysis. Next, an adaptive observer, combined with a fast adaptive fault estimation (FAFE) algorithm, is proposed to estimate system states and actuator failure and compensate for faults in a precise and prompt manner. In addition, a non-singular fast terminal sliding surface is defined, taking into account the fast convergence of the tracking errors in order to provide appropriate trajectory tracking results. Since the upper bounds of the disturbances caused by the manipulator of the system in practice are unknown, the control approach may benefit from the addition of an adaptive control strategy that can suppress the influence of uncertain disturbances. The Lyapunov stability theory demonstrates that tracking errors are able to converge stably and quickly. In the end, the contrast experiment is conducted to exhibit the effectiveness of the proposed control strategy. The results demonstrate quicker convergence and improved estimating accuracy.

Carbon dioxide removal (CDR) is the essential ‘net’ in net zero. However, a thriving CDR industry will not come into being without government intervention. As governments start to devise CDR support policies, this paper solicits the views of market participants in two of the most prominent CDR methods: bioenergy with carbon capture and storage (BECCS) and direct air carbon capture and storage (DACCS). We survey 47 BECCS and DACCS project developers and financiers active in Europe, conducting in-depth interviews with 27 of them to identify their key challenges and preferred policy interventions to address them. We find that participants prefer compliance markets, such as links to emissions trading systems, to generate demand but seek government support to cushion early market risks. They acknowledge the need for stringent monitoring and regulation to ensure environmental integrity. Bearing industry expectations in mind, policymakers face five key challenges in developing CDR: reaching scale, striking a balance with emissions cuts, safeguarding integrity, ensuring fairness and accelerating the speed of deployment.

A field experiment was conducted on the Loess Plateau in 2021 and 2022, with three planting densities (600,000 plants/ha, 900,000 plants/ha, and 1200,000 plants/ha) and four phosphorus (P) levels (0 kg/ha, 15 kg/ha, 75 kg/ha and 135 kg/ha) designed to study the effects of planting density and phosphate fertilizer management on the growth and resource utilization of common buckwheat. Results showed that optimizing P application (75 kg/ha) and planting density (900,000 plants/ha) effectively improved the resource use efficiency and yield of common buckwheat, as a result of significantly improving photosynthetic capacity and dry matter accumulation. The ability of the buckwheat population to absorb soil moisture and nutrients, as well as intercept IPAR, was maximized through the modification of planting density, thereby further promoting photosynthesis and productivity. The integration of combining moderate planting density with P fertilizer increased yield by boosting buckwheat growth and enhancing resource use efficiency based on the optimization of crop productivity without additional P input. In conclusion, future agriculture should focus on the SPAD value, 1000-grain weight, and illuminance of buckwheat population to effectively increase grain yield. This cultivation system of combining planting density and P application would be a promising alternative agricultural practice. Implementing a planting density of 900,000 plants/ha and a P application rate of 75 kg/ha provide opportunities to improve productivity and sustainability on the Loess Plateau in agriculture.

Abstract Background Following lung cancer surgery, patients often experience severe symptoms which are not properly assessed at discharge. The aim of this study was to identify the clinical presentation at discharge and the influencing factors of postoperative symptoms in patients who have undergone lung cancer surgery. Methods This cross-sectional study analysed data from patients who participated in a prospective cohort study that enrolled patients who underwent lung cancer surgery at six tertiary hospitals in the People’s Republic of China, from November 2017 to January 2020. Patient symptoms at discharge were measured using the MD Anderson Symptom Inventory Lung Cancer module. The five core symptoms were defined according to ratings of moderate to severe symptoms (≥ 4 on a 0–10 scale). A multivariate linear regression model was used to identify the influencing factors of each symptom at discharge. Results Among the 366 participants, 51.9% were male and the mean (SD) age was 55.81 (10.43) years. At discharge, the core symptoms were cough (36.4%), pain (28.2%), disturbed sleep (26.3%), shortness of breath (25.8%), and fatigue (24.3%), and more than half of the participants (54.6%) had one to five of the core symptoms, with moderate to severe severity. A low annual income and the use of two chest tubes were significantly associated (P = 0.030 and 0.014, respectively) with higher mean scores of the core symptoms. Conclusion Though clinically eligible for discharge, more than half of the participants had severe symptoms at discharge after lung cancer surgery. Special attention should be given to patients who have two chest tubes after surgery and those who have a low annual income.

Abstract Gene therapy, a medical approach that involves the correction or replacement of defective and abnormal genes, plays an essential role in the treatment of complex and refractory diseases, such as hereditary diseases, cancer, and rheumatic immune diseases. Nucleic acids alone do not easily enter the target cells due to their easy degradation in vivo and the structure of the target cell membranes. The introduction of genes into biological cells is often dependent on gene delivery vectors, such as adenoviral vectors, which are commonly used in gene therapy. However, traditional viral vectors have strong immunogenicity while also presenting a potential infection risk. Recently, biomaterials have attracted attention for use as efficient gene delivery vehicles, because they can avoid the drawbacks associated with viral vectors. Biomaterials can improve the biological stability of nucleic acids and the efficiency of intracellular gene delivery. This review is focused on biomaterial‐based delivery systems in gene therapy and disease treatment. Herein, we review the recent developments and modalities of gene therapy. Additionally, we discuss nucleic acid delivery strategies, with a focus on biomaterial‐based gene delivery systems. Furthermore, the current applications of biomaterial‐based gene therapy are summarized.

The structural, functional properties of protein isolated from small-seeded soybeans were investigated and characteristics of tofu were studied. Small-seeded soybean protein had obvious α', α, β, acidic and basic subunits bands and two endothermic peaks (76.02–76.63℃ and 91.94–94.25℃). Small-seeded black soybean protein isolates (SBSPI) had more β-sheet (31.90–33.54%) structure, while small-seeded yellow soybean protein isolates (SYSPI) had more α-helix (18.89–20.72%) structure. SYSPI had higher fluorescence intensity (839.10–847.80) than SBSPI (482.70–565.10). SBSPI exhibited higher surface hydrophobicity (939.51–1252.75) and water absorption capacity (8.07–8.50 g/g). Tofu made from small-seeded yellow soybeans had higher yield (549.46–560.23 g/100 g soybean) and was brighter (L*, 74.61–77.48) and more yellowish (b*, 14.83–14.95) in color. Tofu made from Fugu small-seeded black soybean (FGSBS) had the highest hardness (178.52 g), adhesiveness (-25.77 g.sec), chewiness (87.45 g) and resilience (0.26), signifying a more compact structure.

As the core component of rotating machinery, the fault diagnosis of rolling bearing has important engineering practical significance. Most of the current intelligent fault diagnosis methods are based on the premise that the training data and test data have similar probability distributions. However, in practical scenarios, there will inevitably be discrepancies in the distribution of vibration signals due to internal and external factors such as changes in working conditions, which will significantly affect the diagnostic performance of the intelligent diagnostic model. Aiming at problems that the vibration signal characteristic distribution of rolling bearings is inconsistent under different working conditions and the labels of the samples to be diagnosed are difficult to obtain, a new domain-adaptive fault diagnosis method is proposed in this paper. Firstly, the multi-scale feature extraction module is used to extract the features of the input signals, and the residual network structure is used to avoid the degradation of the model performance. Then, the APReLU activation function is used to make the vibration signals perform different nonlinear transformations according to their own characteristics through adaptive learning. Finally, the Joint Maximum Mean Discrepancy (JMMD) is used to reduce the displacement of both conditional and edge distributions between different domains. Therefore, this method can extract domain-invariant feature information and align the source and target domains, which can be used for cross-domain intelligent fault diagnosis. Six transfer fault diagnosis tasks based on the rolling bearing experimental platform are designed to evaluate the performance and effectiveness of the proposed method. At the same time, four popular methods are selected for comprehensive analysis and comparison. The results show that the method has good robustness and superiority under various diagnostic tasks.

Horns are an important breeding trait for sheep. However, no widely recognized viewpoint on the regulatory genes and mechanisms of horns is available, and the genetic basis of the four-horn phenotype (FHP) is unclear. This work conducted a genome-wide association study with 100 sheep genomes from multiple breeds to investigate the genetic basis of the FHP. The results revealed three significant associations (corrected as p < 1.64 × 10−8) of the InDels (CHR2: g.133,742,709delA, g.133,743,215insC, and g.133,743,940delT) for FHP in the intergenic sequence (IGS) between the MTX2 and the LOC105609047 of CHR2. Moreover, 14 significant associations (corrected as p < 1.42 × 10−9) of SNPs with the FHP phenotype were identified in CHR2 and CHR16, including five (e.g., CHR16: g.40,351,378G > A and g.40,352,577G > A) located in the intron of the ADAMTS12 gene, eight (e.g., CHR2: g.133,727,513C > T and g.133,732,145T > G) in the IGS between MTX2 and LOC105609047, and only one (CHR2: g.133,930,761A > G) in the IGS between HOXD1 and MTX2. Obvious divergence was also observed in genotype patterns between the FHP and others (two horns and hornless) in the HOXD1 and ADAMTS12 gene regions. An extremely significant linkage also occurred between Loci I and Loci II within 100 individuals (LD = −156.02186, p < 0.00001). In summary, our study indicated that the genomic sequences from CHR2 and CHR16 contributed to the FHP in sheep, specifically the key candidate genes HOXD1 and ADAMTS12. These results improved our understanding of the Mendelian genetic basis of the FHP in sheep.

The heavy metal ion Cu2+ was used as the research object in this study. Cu2+ was detected using a cysteine- and glutathione-modified cadmium telluride quantum dot solution. Compounds of Cu2+ and modified quantum dots on the surface caused the fluorescence quenching of quantum dots. A linear relationship existed between the concentration and the intensity of the change in fluorescence quenching. Using this linear relationship, the quantitative detection of the Cu2+ content in the solution by quantum dots was achieved; the detection band was 476-800 nm. Under optimized conditions, the detection limit of the method was 3.58×10-5 mol/L (S/N=3, n=11), and the linear range was 3×10-7 to 8×10-4 mol/L. In a certain experimental environment, standard normal variate preprocessing of the characteristic band and partial least squares modeling and analysis yielded good self-prediction and actual prediction abilities. The correction coefficient was 0.9006. The RMSEC was 8.83084 µmol/L, the determination coefficient of the validation set was 0.8686, and the RMSEP was 5.4320 µmol/L. The results showed that the modeling effect was good in the experiment, the operation was simple and convenient, and the experiment was fast, reliable, and nonpolluting, indicating that the method of detecting Cu2+ using a quantum dot fluorescent probe was feasible.

PurposeTo review the association between central obesity and abdominal aortic aneurysm (AAA).Materials and methodsThe PubMed, Web of Sciences, Embase, The China national knowledge infrastructure (CNKI), and Cochrane Library were searched up to April 30, 2022. Researches includes investigation of the relationship between central obesity markers and AAA. Included studies must use recognized measures of central obesity, i.e., waist circumference (WC) and waist-to-hip ratio (WHR), or use imaging techniques to calculate abdominal fat distribution, such as computed tomography (CT) imaging.ResultsEleven clinical researches were identified of which eight discussed the association between physical examination and AAA, and three studies mainly focused on abdominal fat volume (AFV). Seven researches concluded that there was a positive correlation between markers of central obesity and AAA. Three studies found no significant link between markers of central obesity and AAA. One of the remaining studies reported different results for each sex. Three studies pooled in a meta-analysis identified correlation between central obesity and AAA presence (RR = 1.29; 95% confidence interval, 1.14–1.46).ConclusionCentral obesity plays a role in the risk of AAA. Standardized central obesity markers may be predictors of AAA. However, there was no association between abdominal fat volume and AAA. Extracellular vesicles (EVs) are a collective term for nanoscale or microscale vesicles secreted by cells that play important biological roles. Mesenchymal stem cells are a class of cells with the potential for self-healing and multidirectional differentiation. In recent years, numerous studies have shown that EVs, especially those secreted by mesenchymal stem cells, can promote the repair and regeneration of various tissues and, thus, have significant potential in regenerative medicine. However, due to the rapid clearance capacity of the circulatory system, EVs are barely able to act persistently at specific sites for repair of target tissues. Hydrogels have good biocompatibility and loose and porous structural properties that allow them to serve as EV carriers, thereby prolonging the retention in certain specific areas and slowing the release of EVs. When EVs are needed to function at specific sites, the EV-loaded hydrogels can stand as an excellent approach. In this review, we first introduce the sources, roles, and extraction and characterization methods of EVs and describe their current application status. We then review the different types of hydrogels and discuss factors influencing their abilities to carry and release EVs. We summarize several strategies for loading EVs into hydrogels and characterizing EV-loaded hydrogels. Furthermore, we discuss application strategies for EV-loaded hydrogels and review their specific applications in tissue regeneration and repair. This article concludes with a summary of the current state of research on EV-loaded hydrogels and an outlook on future research directions, which we hope will provide promising ideas for researchers.

Lysobacter species have attracted increasing attention in recent years due to their capacities to produce diverse secondary metabolites against phytopathogens. In this research, we analyzed the genomic and transcriptomic patterns of Lysobacter capsici CK09. Our data showed that L. capsici CK09 harbored various contact-independent biocontrol traits, such as fungal cell wall lytic enzymes and HSAF/WAP-8294A2 biosynthesis, as well as several contact-dependent machineries, including type 2/4/6 secretion systems. Additionally, a variety of hydrolytic enzymes, particularly extracellular enzymes, were found in the L. capsici CK09 genome and predicted to improve its adaption in soil. Furthermore, several systems, including type 4 pili, type 3 secretion system and polysaccharide biosynthesis, can provide a selective advantage to L. capsici CK09, enabling the species to live on the surface in soil. The expression of these genes was then confirmed via transcriptomic analysis, indicating the activities of these genes. Collectively, our research provides a comprehensive understanding of the biocontrol potential and soil adaption of L. capsici CK09 and implies the potential of this strain for application in the future.

In general, modified starches provided a desirable feature so that they can be used in the food industry like food additive products. The awareness of eating healthy has become very evident. The food industry is willing to produce foods and food ingredients that not only maintain nutrition but also promote health and safety. Tartary buckwheat starch has been isolated to assess the effect of HMT on its digestibility, morphological structure, pasting, thermal and textural properties and compared with common buckwheat, sorghum, and broomcorn millet. HMT changed significantly the granular structure of native starches. The complete starch granular structure increased the size and changed into an irregular shape, with a rough surface. The modified starches exhibited higher onset and peak temperatures, while their amylose content, solubility, swelling power, gel hardness, and enthalpy of gelatinization were significantly reduced. In pasting properties, tartary buckwheat and sorghum modified starch had the highest thermal treatment stability with lower setback and breakdown viscosity. Moreover, after three hours of starch hydrolysis; Resistant (RS) starch was higher in tartary buckwheat followed by broomcorn millet and lastly sorghum. In this study, we reported that tartary buckwheat modified starch is more useful in the preparation of food with thermal stability combined with a high amount of resistant starch.

Cities are at the forefront of the battle against climate change. However, intercity comparisons and responsibility allocations among cities are hindered because cost- and time-effective methods to calculate the carbon footprints of global cities have yet to be developed. Here, we establish a hybrid method integrating top-down input–output analysis and bottom-up crowdsourced data to estimate the carbon footprints of global cities. Using city purchasing power as the main predictor of the carbon footprint, we estimate the carbon footprints of 465 global cities in 2020. Those cities comprise 10% of the global population but account for 18% of the global carbon emissions showing a significant concentration of carbon emissions. The Gini coefficients are applied to show that global carbon inequality is less than income inequality. In addition, the increased carbon emissions that come from high consumption lifestyles offset the carbon reduction by efficiency gains that could result from compact city design and large city scale. Large climate benefits could be obtained by achieving a low-carbon transition in a small number of global cities, emphasizing the need for leadership from globally important urban centres.

Respiratory syncytial virus (RSV) is a ubiquitous pathogen of viral bronchiolitis and pneumonia in children younger than 2 years of age, which is closely associated with recurrent wheezing and airway hyperresponsiveness (AHR). Alveolar macrophages (AMs) located on the surface of the alveoli cavity are the important innate immune barrier in the respiratory tract. AMs are recognized as recruited airspace macrophages (RecAMs) and resident airspace macrophages (RAMs) based on their origins and roaming traits. AMs are polarized in the case of RSV infection, forming two macrophage phenotypes termed as M1-like and M2-like macrophages. Both M1 macrophages and M2 macrophages are involved in the modulation of inflammatory responses, among which M1 macrophages are capable of pro-inflammatory responses and M2 macrophages are capable of anti-proinflammatory responses and repair damaged tissues in the acute and convalescent phases of RSV infection. Polarized AMs affect disease progression through the alteration of immune cell surface phenotypes as well as participate in the regulation of T lymphocyte differentiation and the type of inflammatory response, which are closely associated with long-term AHR. In recent years, some progress have been made in the regulatory mechanism of AM polarization caused by RSV infection, which participates in acute respiratory inflammatory response and mediating AHR in infants. Here we summarized the role of RSV-infection-mediated AM polarization associated with AHR in infants.

Foot-and-mouth disease virus (FMDV) could cause acute infection in host cells, or they could coexist with host cells to generate persistent infection. In persistent infection, the virus could survive for a long time in the host and could be transmitted between different host cells. In the case of FMDV-persistent infection cell line, there is a remarkable significant cellular heterogeneity in the FMDV-persistent infection cell line due to differences of viral load in the individual cells within the cell line. However, the mechanisms of FMDV-persistent infection are not well understood. It is now generally accepted that multiple factors contribute to the coevolution of viruses and cells during the course of persistent infection. The outcome would influence the development of persistent FMDV infection conjointly, reaching a state of equilibrium ultimately. Therefore, in order to elucidate the mechanism of cellular heterogeneity in FMDV-persistent infection cell line, single-cell sequencing was performed on BHK-Op, and pseudotime trajectory plot was draw through cell cluster. Based on the cell clusters, we predicted the development and progression of the FMDV-persistent infection. It could be well explained by the fact that, in BHK-Op cells, there are a fraction of infected cells and a fraction of virus-exposed but uninfected bystander cells. By further comparing the transcripts in cell clusters, we found that these genes were involved in changes in ribosome biogenesis, cell cycle, and intracellular signaling including the interferon signaling pathway and mitogen-activated protein kinase (MAPK) signaling pathway. Through comprehensive cross-tabulation analysis of differential expressed genes in various cluster of cells, we identified a high association of Fos, a downstream transcription factor of the MAPK/extracellular signal–regulated kinase (ERK) signaling pathway, with viral replication during the formation of FMDV-persistent infection. Through the further study of Fos, we found that downregulation of Fos facilitates viral clearance during FMDV-persistent infection. Upregulation of c-Raf, which is the upstream of the MAPK/ERK signaling pathway, could promote FMDV replication through downregulation of Fos. Our research is the first to provide insight into the mechanism of the formation FMDV-persistent infection through single-cell sequencing using persistent infection cell line. Pseudotime trajectory analysis was the first time to apply for FMDV-persistent infection cell line. Our work highlights the detailed overview of the evolution of FMDV-persistent infection. We also analyzed the differential expressed genes in the replication or elimination of FMDV within the host. We found that the MAPK/ERK signaling pathway and its downstream transcription factor Fos play an important role in FMDV-persistent infection.

In this study, 22 mung bean varieties from different producing areas in China were used as materials to provide a theoretical basis for the breeding and utilisation of special mung bean sprout varieties. Principal component analysis and cluster analysis were used to comprehensively evaluate the overall quality. Finally, 22 germinated mung bean varieties were divided into 3 edible quality grades according to their quality scores. The results showed that Lvfeng 2 and Ankang mung bean sprouts with better comprehensive characters were varieties with higher scores, which could be developed and utilised as characteristic mung bean sprouts. The content of protein, total phenol and antioxidant capacity of edible parts of Lvfeng 2 and Ankang mung bean after germination are higher than those of other varieties, so they are suitable for research and development as functional mung bean sprouts.

Abstract Background Bilateral rotator cuff tears are not uncommon and the timing of the surgical treatment of both shoulders is debated. In the present study, we aimed to compare the clinical outcomes of patients who underwent single-stage or staged bilateral arthroscopic rotator cuff repair. Methods From March 2013 to May 2018, a retrospective review on all patients who underwent bilateral arthroscopic rotator cuff repair at our department was performed. Patients were separated into 2 groups: single-stage and staged. The minimum follow-up period was 2 years. The visual analog scale (VAS), American Shoulder and Elbow Surgeons (ASES) score, University of California, Los Angeles (UCLA) score, Constant-Murley (Constant) score, the range of motion (ROM) of the shoulder and the hospitalization costs were evaluated for comparison between the two groups before and after the operation. Differences between groups were assessed using t-tests and ANOVA. Results All 51 patients completed follow-up of 2 years, single stage (n = 24) and staged group (n = 27). There was no significant difference in the VAS, ASES, UCLA and Constant scores between the single-stage group and the staged group before the operation. The present research envisaged the analysis of the dissolved oxygen fault of the water quality monitoring system using the genetic algorithm-support vector machine (GA-SVM). The real-time data collected by the dissolved oxygen sensor was classified into the fault types. The fault types were divided into complete failure fault, impact fault, and constant output fault. Based on the fault classification of the dissolved oxygen parameters, SVM fault diagnosis experiments were conducted. Experimental results show that the accuracy of dissolved oxygen was 98.53%. On comparison with the experimental results of the back propagation (BP) neural network, it was found that the diagnosis results of the dissolved oxygen parameters using SVM were better than those of the BP neural network. The genetic algorithm (GA) was used to optimize the parameters. After iteration, the optimal parameters such as C and g were selected (C is the penalty coefficient, which adjusts the weight of the two index preferences in the optimization direction, i.e., the tolerance for errors, and g is a parameter that comes with the function that implicitly determines the distribution of the data after mapping to the new feature space.). By using GA, after iteration, the optimized values of C and g was found to be 2.1649 and 5.3312, respectively. The experimental results showed that the method exhibited a good accuracy.

Objective To explore the relationship between health literacy and depressive symptoms in junior high school students in Chongqing, and to provide suggestions for improving adolescent mental health. Methods A total of 1 703 students were subjected by stratified cluster sampling from 4 junior high schools (2 in urban and 2 in rural areas) in a main district of Chongqing. Chi-square test and logistic regression analysis were used to study the data. Results Among 1 703 participants, the incidence of depressive symptoms was 26.8%. The overall health literacy, functional health literacy, interactive health literacy and critical health literacy were 41.8%, 44.8%, 38.4% and 60.1%, respectively. Univariate analysis showed that the overall health literacy rate of individuals without depressive symptoms was significantly higher than that of individuals with depressive symptoms (46.9% vs 27.9%, Chi-square=49.867, P < 0.001). The health literacy rates of the individuals without depressive symptoms in the 3 dimensions were also significantly higher than those of the individuals with depressive symptoms (P < 0.001). After the adjustment for gender, age, grade, school type and whether they were only children in family, logistic regression analysis indicated that scientific health concept, reasonable nutrition, problem-solving ability and health decision-making ability were protective factors for depressive symptoms (P < 0.05). Conclusion The incidence of depressive symptoms in the students with health literacy is quite low. The factors of scientific health concept, reasonable diet, problem-solving ability and health decision-making ability of health literacy are negatively correlated with the incidence of depression symptoms.

Abstract At present, there are still ambiguous reports about the perinatal infection of infants born to mothers infected with severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). The dynamic characteristics of infantile serum antibodies born to mother with SARS-CoV-2 has not been well described. In this study, we analyzed the seroconversion of 27 newborns born to 26 pregnant women infected with SARS-CoV-2. The SARS-CoV-2 IgG positive rate of parturient was 80.8%, and half of their infants obtained maternal IgG. IgG transfer rates were 18.8% and 81.8% in those infants whose mother infected less and more than 2 weeks before delivery. In the first two months of life, the IgG level of infants dropped sharply to one tenth of that at birth. These results suggest that maternal SARS-CoV-2 IgG provides limited protection for infants.

BackgroundEtiology and risk factors of peripheral artery disease (PAD) include age, smoking, and hypertension, etc. , which are shared by an abdominal aortic aneurysm (AAA). Concomitance with AAA in patients with PAD is not rare but is easily overlooked in the clinical situation, though management strategies are altered totally. This study aims to investigate diagnostic biomarkers for the prediction of AAA in patients with PAD.MethodsA total of 684 patients diagnosed with AAA and/or PAD were enrolled and analyzed retrospectively. Each patient with PAD and AAA was gender and age-matched. Demographic data, medical history, and serum laboratory test profiles were obtained. Statistical analysis was performed to determine diagnostic biomarkers of AAA in patients with PAD.ResultsFirstly, 320 patients with PAD-only and 320 patients with AAA-only were compared. Levels of bilirubin and D-Dimer were decreased, while the incidence of diabetes mellitus, levels of fibrinogen, and platelet count were increased significantly in patients with PAD-only compared with those in patients with AAA-only (P < 0.001). Next, 364 patients with PAD (44 patients with AAA) and 364 patients with AAA (44 patients with PAD) were compared. Multivariate logistic regression analysis confirmed the differential distribution of bilirubin, D-dimer, fibrinogen, and platelet count between patients with AAA and patients with PAD (P < 0.05). Receiver operator curves (ROC) showed that the area under the curve (AUC) of total bilirubin, direct bilirubin, D-dimer, fibrinogen, and platelet count was 0.6113, 0.5849, 0.7034, 0.6473, and 0.6785, respectively. Finally, to further validate the predictive efficacy of mentioned markers, a multivariable logistics regression analysis was performed between the PAD only group and the PAD with AAA group. The results suggested increased levels of D-dimer in the PAD with AAA group compared to the PAD only group (OR: 2.630, 95% CI:1.639–4.221; P < 0.001). In particular, the Youden index suggested that the cut-off value of D-dimer for predicting AAA in patients with PAD was 0.675 mg/L with a sensitivity of 76.9% and a specificity of 84.9% (AUC = 0.8673; 95% CI, 0.8106–0.9240, P < 0.001). In all 364 patients with PAD, 41.46% patients were diagnosed AAA when D-dimer is >0.675 mg/L, while only 3.55% patients were diagnosed AAA when D-dimer ≤ 0.675 mg/L.ConclusionsPAD and AAA exert different clinical and serum profiles; D-dimer (>0.675 mg/L) is a reliable biomarker for the prediction of AAA in patients with PAD.

Potassium (K+) is one of the indispensable elements in plant growth and development. The Shaker K+ channel protein family is involved in plant K+ uptake and distribution. Foxtail millet (Setaria italica), as an important crop, has strong tolerance and adaptability to abiotic stresses. However, no systematic study focused on the Shaker K+ channel family in foxtail millet. Here, ten Shaker K+ channel genes in foxtail millet were identified and divided into five groups through phylogenetic analysis. Gene structures, chromosome locations, cis-acting regulatory elements in promoter, and post-translation modification sites of Shaker K+ channels were analyzed. In silico analysis of transcript level demonstrated that the expression of Shaker K+ channel genes was tissue or developmental stage specific. The transcription levels of Shaker K+ channel genes in foxtail millet under different abiotic stresses (cold, heat, NaCl, and PEG) and phytohormones (6-BA, BR, MJ, IAA, NAA, GA3, SA, and ABA) treatments at 0, 12, and 24 h were detected by qRT-PCR. The results showed that SiAKT1, SiKAT3, SiGORK, and SiSKOR were worth further research due to their significant responses after most treatments. The yeast complementation assay verified the inward K+ transport activities of detectable Shaker K+ channels. Finally, we found interactions between SiKAT2 and SiSNARE proteins. Compared to research in Arabidopsis, our results showed a difference in SYP121 related Shaker K+ channel regulation mechanism in foxtail millet. Our results indicate that Shaker K+ channels play important roles in foxtail millet and provide theoretical support for further exploring the K+ absorption mechanism of foxtail millet under abiotic stress.

Abstract Purpose In the present study, we aimed to determine whether decompression of suprascapular nerve (SSN) at the spinoglenoid notch could lead to a better functional outcome for the patients who underwent repairment of rotator cuff due to posterosupeior massive rotator cuff tear (MRCT) and suprascapular neuropathy. Methods A total of 20 patients with posterosuperior MRCT and suprascapular neuropathy were analyzed in the present work. The preoperative magnetic resonance imaging (MRI) showed rotator cuff tear in supraspinatus and infraspinatus. All patients underwent arthroscopic rotator cuff repair. Patients were divided into two groups (group A: non-releasing, group B: releasing) according to whether the SSN at the spinoglenoid notch was decompressed. The modified University of California at Los Angeles shoulder rating scale (UCLA) and visual analog scale (VAS) questionnaire were adopted to assess the function of the affected shoulder preoperatively and 12 months after the operation. Electromyography (EMG) and nerve conduction study (NCS) were used to evaluate the nerve condition. Patients underwent MRI and EMG/NCS at 6 months after operation and last follow-up. Results All patients were satisfied with the treatment. MRI showed that it was well-healed in 19 patients at 6 months after the operation. However, the fatty infiltration of supraspinatus and infraspinatus was not reversed. Only one patient in the non-releasing group showed the retear. The retear rate of group A and group B were 30% (3/10) and 20% (2/10) respectively at 12 months after the operation. One patient undergoing SSN decompression complained of discomfort in the infraspinatus area. His follow-up EMG after 6 months showed fibrillation potentials (1+) and positive sharp waves (1+) in the infraspinatus. The other patients’ EMG results showed no abnormality. The postoperative UCLA and VAS scores were improved in both groups, and there was no significant difference in the follow-up outcomes between the two groups. Conclusions Patients with postersuperior MRCT and suprascapular neuropathy, decompression of suprascapular nerve at spinoglenoid notch didn’t lead to a better functional outcome with the repairment of rotator cuff. Arthroscopic rotator cuff repair could reverse the suprascapular neuropathy. Level of evidence Level III.

The Chinese white wax scale insect, Ericerus pela Chavannes (Hemiptera: Coccidae), is one of the scale insects with great economic value and has been dispersed and reared in China for over one thousand years. Its mitochondrial genome provides essential information for the molecular identification and genetic study of this species. We assembled the complete mitochondrial genome of E. pela based on PacBio sequencing and analyzed its genomic features. The genome was 17,766 bp in length with 13 protein-coding genes, 22 tRNAs, and two rRNA genes. The analysis results showed E. pela had significant gene rearrangements involving tRNAs compared with other Coccoidea species. Furthermore, E. pela’s nine tRNAs were identified to have obvious truncated structures. The phylogenetic tree compiled of the species showed a long branch of the Coccoidea lineage, which indicated the high evolutionary rate in this group. Our study revealed the mitochondrial characteristics of E. pela and enriched the mitochondrial genetic information on Coccoidea species. It also determined the occurrence of gene rearrangement for the species in this superfamily.
