5. Soft dimension theory.

Abstract

Classical dimension theory, when applied to preference modeling, is based upon the assumption that linear ordering is the only elemental notion for rationality. In fact, crisp preferences are in some way decomposed into basic criteria, each one being a linear order. In this paper, we propose that indeed dimension is relative to a previous idea of rationality, but such a rationality is not unique. In particular, we explore alternative approaches to dimension, based upon a more general representation and allowing different classes of orders for basic criteria. In this way, classical dimension theory is generalized. As a first consequence, we explore the existence of crisp preference representations not being based upon linear orders. 6. Searching for the dimension of valued preference relations.

Abstract

The more information a preference structure gives, the more sophisticated representation techniques are necessary, so decision makers can have a global view of data and therefore a comprehensive understanding of the problem they are faced with. In this paper we propose to explore valued preference relations by means of a search for the number of underlying criteria allowing its representation in real space. A general representation theorem for arbitrary crisp binary relations is obtained, showing the difference in representation between incomparability-related to the intersection operator-and other inconsistencies-related to the union operator. A new concept of dimension is therefore proposed, taking into account inconsistencies in source of information. Such a result is then applied to each alpha-cut of valued preference relations. Abstract

In this paper we address the problem of inconsistency in preference relations, pointing out the relevance of a meaningful representation in order to help decision maker to capture such inconsistencies. Dimension theory framework, despite its computational complexity, is considered here, pursuing in principle a decomposition of arbitrary preference relations in terms of linear orderings of alternatives. But we shall then stress that consistency should not be necessarily associated to a linear ordering. In this way, alternative decompositions of a preference relation can be proposed to decision maker, allowing an effective search for a useful representations of alternatives in terms of possible criteria. Such decompositions of our preference relations will then become the basis of a future decision aid model, always with the restricted aim of allowing the decision maker a better understanding of the problem. 8. Targeted lipidomic profiling of West Nile virus infection unveils alterations of sphingolipid metabolism in a mouse model and human patients

Abstract

West Nile virus (WNV) is a neurotropic flavivirus transmitted by the bites of infected mosquitoes. Severe forms of West Nile disease (WND) can curse with meningitis, encephalitis, or acute flaccid paralysis. A better understanding of the physiopathology associated with disease progression is mandatory to find biomarkers and effective therapies. In this scenario, blood derivatives (plasma and serum) constitute the more commonly used biofluids due to their ease of collection and high value for diagnostic purposes. Therefore, the potential impact of this virus in the circulating lipidome was addressed by combining the analysis of samples from experimentally infected mice and naturally WND patients. Our results unveil dynamic alterations in the lipidome that define specific metabolic fingerprints of different infection stages. Concomitant with neuroinvasion in mice, the lipid landscape was dominated by a metabolic reprogramming that resulted in significant elevations of circulating sphingolipids (ceramides, dihydroceramides and dihydrosphingomyelins), along with some phosphatidylethanolamines and triacylglycerols. Remarkably, patients suffering from WND also displayed an elevation of ceramides, dihydroceramides, and lactosylceramides in their sera. The dysregulation of sphingolipid metabolism by WNV may provide new therapeutic opportunities and supports the potential of certain lipids as novel peripheral biomarkers of WND progression.

Abstract

Objectives: To evaluate the in vivo efficacy of a dual carbapenem combination containing imipenem plus meropenem against carbapenem-resistant Acinetobacter baumannii producing carbapenemases OXA-23 or OXA-58.Methods: An experimental model of peritonitis using C57BL/6J female mice was developed and the minimum lethal doses were calculated for infections due to OXA-23 or OXA-58 producers of A. baumannii clinical isolates. The efficacies of the carbapenems in monotherapy and in combination were tested.Results: Meropenem was better than imipenem in mice infected with either of the carbapenem-resistant A. baumannii (CRAb) strains. The combination of meropenem plus imipenem significantly improved the clearance of CRAbs from spleen compared with non-treated groups. The carbapenem-containing combination was better than imipenem for treating mice infected with both carbapenemase producers. In blood, the carbapenem combination significantly decreased the bacterial load of the OXA-23 producers compared with imipenem or meropenem used in monotherapy.Conclusions: These results suggest that dual carbapenem combination could be an option for the treatment of infections due to carbapenemase-producing A. baumannii such as OXA-23 and OXA-58 producers.

Abstract

Immune response stimulation may be an adjuvant to antimicrobial treatment. Here, we evaluated the impact of immune response modification by lysophosphatidylcholine (LPC) combined with imipenem or ceftazidime in murine models of peritoneal sepsis (PS) and pneumonia by Pseudomonas aeruginosa. Imipenem and ceftazidime-susceptible strain (Pa39), and imipenem and ceftazidime-resistant strain (Pa238) were used. Ceftazidime pharmacokinetic and pharmacodynamic parameters were determined. Therapeutic efficacy, and TNF-α and IL-10 levels were determined in murine models of PS and pneumonia by Pa39 and Pa238 treated with LPC, imipenem or ceftazidime, alone or in combination. In PS model, LPC+ceftazidime reduced spleen and lungs Pa238 concentrations (-3.45 and -3.56 log10 CFU/g; P0.05). In pneumonia model, LPC+ceftazidime or LPC+imipenem reduced lungs Pa238 concentrations (-2.37 log10 CFU/g, P=0.1, or -1.35 log10 CFU/g, P=0.75). For Pa39 no statistically significant difference has been observed in PS and pneumonia models between combined therapy and monotherapy. Moreover, LPC+imipenem and LPC+ceftazidime decreased and increased significantly TNF-α and IL-10 levels, respectively, in comparison with untreated controls and monotherapies. These results demonstrate the impact of immune response modification by LPC plus antibiotics on the prognosis of infections by ceftazidime-resistant P. aeruginosa.
