38. Method Development & Validation of LCMS/MS for Atorvastatin and Olmesartan in Human Plasma to Trace Drug Interaction of Formulation

Abstract

Sensitive LC/MS/MS method was developed and validated to evaluate the bioavailability of cardiovascular formulation- Atorvastatin (ATSV) and Olmesartan (OLM), after administration of its combined formulation to hypertensive patients to mark the drug interaction. Blood samples (ATVS+OLM) were collected, centrifuged at 3000 rpm, at 4°C for 20min and stored at -20 °C until analysis. To the thawed 0.5 mL of sample, 50 µl IS (Rosuvastatin) solution of 50 ng/mL was added and vortex mixing was conducted for 30 sec. To 10Mm solution, about 2 μl of 10% Tris Buffer was added, then it was sonicated (1 min), extracted with adding 5mL of ethyl acetate, hand mixed (10 min) and centrifuged at 5000 rpm for 15 min. The upper organic layer (4 ml) was separated and evaporated to dryness at 45°C, using a gentle stream of N2 atmosphere. The residue was reconstituted in 200 µL of the mobile phase and was injected to the LC-MS/MS system. The developed method was validated for specificity, accuracy, precision, stability, linearity, sensitivity and recovery. It was successfully applied to detect the drug interaction of ATSV & OLM, where OLM conc. was significantly higher in its combination treatment as compared to the single OLM treatment. Thus, ATSV+OLM is least effective in combination instead of its expected synergistic activity.

Abstract

A high performance liquid chromatographic (HPLC) method was developed using liquid-liquid extraction and ultra-violet detection for quantifying pyridostigmine bromide in human plasma. Liquid chromatography was performed on a BDS Hypersil C18 coulmn (5 µm, 250×4.6 mm i.d.) using an isocratic mobile phase comprised of acetonitrile-5 mM potassium dihydrogen phosphate (40:60, v/v) and the eluent was detected at 269 nm by UV. Clonazepam was used as the internal standard. The limit of quantification was 20 ng/mL using 1 mL plasma. The intra and inter-day precision expressed as the relative standard deviation was less than 15%. The assay was applied to the analysis of samples from a pharmacokinetic study. Following the oral administration of 60 mg pyridostigmine bromide (test) to volunteers, the maximum plasma concentration (Cmax), area under the curve (AUC) and elimination half-life (t1/2) were 176.03±9.176 ng/mL, 819.999±109.64 ng hr/mL and 3.787±0.08 hr, respectively. The method was demonstrated to be highly feasible and reproducible for pharmacokinetic studies of pyridostigmine bromide in humans. [ABSTRACT FROM AUTHOR]

Abstract

Mullite–zirconia composites with 2–6 mol% yttrium oxide have been prepared from Indian coastal sillimanite beach sand, zirconia and alumina. All raw materials have been attrition milled and then isostatically pressed followed by sintering in the range of 1500–1600 °C with a 2-h soaking period. Then the sintered materials have been characterised in terms of bulk density, linear shrinkage, hot modulus of rupture, X-ray diffraction, infra red spectroscopy and microstructural features. The effect of yttrium oxide with change of sintering temperature for different properties have been discussed. [Copyright &y& Elsevier]
