Neo-sex chromosomes often originate from sex chromosome-autosome fusions and constitute an important basis for the study of gene degeneration and expression in a sex chromosomal context. Neo-sex chromosomes are known from many animal and plant lineages, but have not been reported in birds, a group in which genome organization seems particularly stable. Following indications of sex linkage and unexpected sex-biased gene expression in warblers (Sylvioidea; Passeriformes), we have conducted an extensive marker analysis targeting 31 orthologues of loci on zebra finch chromosome 4a in five species, representative of independent branches of Passerida. We identified a region of sex linkage covering approximately the first half (10 Mb) of chromosome 4a, and associated to both Z and W chromosomes, in three Sylvioidea passerine species. Linkage analysis in an extended pedigree of one species additionally confirmed the association between this part of chromosome 4a and the Z chromosome. Markers located between 10 and 21 Mb of chromosome 4a showed no signs of sex linkage, suggesting that only half of the chromosome was involved in this transition. No sex linkage was observed in non-Sylvioidea passerines, indicating that the neo-sex chromosome arose at the base of the Sylvioidea branch of the avian phylogeny, at 47.4-37.6 millions years ago (MYA), substantially later than the ancestral sex chromosomes (150 MYA). We hypothesize that the gene content of chromosome 4a might be relevant in its transition to a sex chromosome, based on the presence of genes (for example, the androgen receptor) that could offer a selective advantage when associated to Z-linked sex determination loci. Teachers in five primary schools were surveyed for their knowledge and attitudes about epilepsy. An analysis of 113 teacher responses revealed knowledge deficits, especially regarding the prevalence of epilepsy, the future of children with the disorder and E.E.G. as an aid to diagnosis. Misconceptions regarding first aid were also common. More than two-thirds of the teachers believed that children with epilepsy have academic problems more often, although about half of the respondents were in favour of normal schools for such children. Most respondents recommended a teacher's career for these children in future but considered that of a bus driver, as unsuitable. Few teachers had attended any educational programme on epilepsy. Only about one-fifth of the teachers were confident in dealing with an epiletic child. Communication about epilepsy among the teachers, parents and doctors was virtually non-existent. Purpose: Although the impact of the paternal contribution to embryo quality and blastocyst formation is a well-known phenomenon, the current literature provides insufficient evidence that hyaluronan-binding sperm selection methods improve assisted reproductive treatment outcomes. Thus, we compared the cycle outcomes of morphologically selected intracytoplasmic sperm injection (ICSI) with hyaluronan binding physiological intracytoplasmic sperm injection (PICSI) cycles., Methods: A total of 2415 ICSI and 400 PICSI procedures of 1630 patients who underwent in vitro fertilization cycles using a time-lapse monitoring system between 2014 and 2018 were analyzed retrospectively. Fertilization rate, embryo quality, clinical pregnancy rate, biochemical pregnancy rate and miscarriage rate were evaluated, differences in morphokinetic parameters and cycle outcomes were compared., Results: In total, 85.8 and 14.2% of the whole cohort were fertilized with standard ICSI and PICSI, respectively. The proportion of fertilized oocytes did not significantly differ between groups (74.53 ± 1.33 vs. 72.92 ± 2.64, p > 0.05). Similarly, the proportion of good-quality embryos according to the time-lapse parameters and the clinical pregnancy rate did not significantly differ between groups (71.93 ± 4.21 vs. 71.33 ± 2.64, p > 0.05 and 45.55 ± 2.91 vs. 44.96 ± 1.25, p > 0.05). No statistically significant differences were found between groups in clinical pregnancy rates (45.55 ± 2.91 vs. 44.96 ± 1.25, p > 0.05). Biochemical pregnancy rates (11.24 ± 2.12 vs. 10.85 ± 1.83, p > 0.05) and miscarriage rates (24.89 ± 3.74 vs. 27.91 ± 4.91, p > 0.05) were not significantly different between groups., Conclusion: The effects of the PICSI procedure on fertilization rate, biochemical pregnancy rate, miscarriage rate, embryo quality, and clinical pregnancy outcomes were not superior. Purpose: To compare the effects of microfluidic sperm sorting, density gradient and swim-up methods on the oxidative reduction potential (ORP) of split semen samples from a single patient population., Materials and Methods: A prospective controlled study was conducted to compare the effects of three different semen processing methods using split semen samples from the same population of infertile men. The primary outcome was the ORP. Secondary outcomes were the sperm concentration, progressive motility rate and total sperm motility., Results: A total of 57 split semen samples were included in this study. The ORP was significantly lower in the microfluidic group compared to the density gradient and swim-up groups (p < 0.05). The ORP/sperm concentration ratio was significantly lower in the microfluidic and density gradient groups compared to the swim-up group (p < 0.05).Total sperm concentration was significantly higher in the density gradient group than the microfluidic and swim-up groups (p < 0.05). Motility was significantly higher in the microfluidic and swim-up groups than the density gradient group (p < 0.05). The progressive motile sperm rate was significantly higher in the microfluidic and swim-up groups than the density gradient group (p < 0.05)., Conclusions: Microfluidic sperm sorting was better for selecting highly motile sperm and yielded a lower ORP than conventional sperm preparation methods.

Objective: To compare the effect of microfluiding sperm sorting chip and density gradient methods on ongoing pregnancy rates (PRs) of patients undergoing IUI., Design: Retrospective cohort study., Setting: Hospital IVF unit., Patient(s): Couples with infertility undergoing IUI cycles between 2017 and 2018., Intervention(s): Not applicable., Main Outcome Measure(s): Ongoing PRs., Result(s): A total of 265 patients were included in the study. Microfluid sperm sorting and density gradient were used to prepare sperm in 133 and 132 patients, respectively. Baseline spermiogram parameters, including volume, concentration, motility, and morphology, were similar between the two groups. Total motile sperm count was lower in the microfluiding sperm sorting group at baseline (35.96 ± 37.69 vs. 70.66 ± 61.65). After sperm preparation sperm motility was higher in the microfluid group (96.34 ± 7.29 vs. 84.42 ± 10.87). Pregnancy rates were 18.04% in the microfluid group and 15.15% in the density gradient group, and ongoing PRs were 15.03% and 9.09%, respectively. After using multivariable logistic regression and controling for confounding factors, there was a significant increase in ongoing PRs in the microfluid sperm sorting group. The adjusted odds ratio for ongoing pregnancy in the microfluid group compared with the density gradient group was 3.49 (95% confidence interval 1.12-10.89)., Conclusion(s): The microfluid sperm sorting method significantly increased the ongoing PRs compared with the density gradient group in IUI cycles., (Copyright © 2019 American Society for Reproductive Medicine. Newer parts of sex chromosomes, neo-sex chromosomes, offer unique possibilities for studying gene degeneration and sequence evolution in response to loss of recombination and population size decrease. We have recently described a neo-sex chromosome system in Sylvioidea passerines that has resulted from a fusion between the first half (10 Mb) of chromosome 4a and the ancestral sex chromosomes. In this study, we report the results of molecular analyses of neo-Z and neo-W gametologs and intronic parts of neo-Z and autosomal genes on the second half of chromosome 4a in three species within different Sylvioidea lineages (Acrocephalidea, Timaliidae, and Alaudidae). In line with hypotheses of neo-sex chromosome evolution, we observe 1) lower genetic diversity of neo-Z genes compared with autosomal genes, 2) moderate synonymous and weak nonsynonymous sequence divergence between neo-Z and neo-W gametologs, and 3) lower GC content on neo-W than neo-Z gametologs. Phylogenetic reconstruction of eight neo-Z and neo-W gametologs suggests that recombination continued after the split of Alaudidae from the rest of the Sylvioidea lineages (i.e., after ~42.2 Ma) and with some exceptions also after the split of Acrocephalidea and Timaliidae (i.e., after ~39.4 Ma). The Sylvioidea neo-sex chromosome shares classical evolutionary features with the ancestral sex chromosomes but, as expected from its more recent origin, shows weaker divergence between gametologs.

The evolution of hybrid polyploid vertebrates, their viability and their perpetuation over evolutionary time have always been questions of great interest. However, little is known about the impact of hybridization and polyploidization on the regulatory networks that guarantee the appropriate quantitative and qualitative gene expression programme. The Squalius alburnoides complex of hybrid fish is an attractive system to address these questions, as it includes a wide variety of diploid and polyploid forms, and intricate systems of genetic exchange. Through the study of genome-specific allele expression of seven housekeeping and tissue-specific genes, we found that a gene copy silencing mechanism of dosage compensation exists throughout the distribution range of the complex. Here we show that the allele-specific patterns of silencing vary within the complex, according to the geographical origin and the type of genome involved in the hybridization process. In southern populations, triploids of S. alburnoides show an overall tendency for silencing the allele from the minority genome, while northern population polyploids exhibit preferential biallelic gene expression patterns, irrespective of genomic composition. The present findings further suggest that gene copy silencing and variable expression of specific allele combinations may be important processes in vertebrate polyploid evolution.

The rate constant of the reaction of Cl atoms with methacrolein (k(1)) has been measured relative to that of Cl with propane (k(2)) or cyclohexane (k(6)) at ambient temperature and pressures varying from 1-950 Torr. The experiments were carried out by irradiation (350 nm) of Cl(2)/methacrolein/propane or cyclohexane mixtures in N(2) or N(2)/O(2) diluent at ambient temperature in a spherical (500 cm(3)) Pyrex reactor (GC/FID analyses) or a 140 L FTIR smog chamber. The measured relative rate constant ratios in the GC/FID experiments were k(1)/k(2) = 1.464 +/- 0.015(2sigma) in N(2) and k(1)/k(2) = 1.68 +/- 0.03(2sigma) in N(2)/O(2) diluent (O(2) > 20,000 ppm). No pressure dependence was observed over the range studied in N(2) (120-950 Torr) using the GC/FID. In the FTIR/smog chamber experiments values of k(1)/k(6) = 0.645 +/- 0.032, 0.626 +/- 0.037, 0.586 +/- 0.026, and 0.479 +/- 0.024 were measured in 700, 100, 10, and 1 Torr, respectively, of N(2) diluent. Using k(2) = (1.4 +/- 0.2) x 10(-10) and k(6) = (3.3 +/- 0.5) x 10(-10) high pressure limiting rate constants of k(1) = (2.05 +/- 0.3) x 10(-10) [GC/FID] and (2.13 +/- 0.34) x 10(-10) [FTIR] cm(3) molecule(-1) s(-1) were determined. In experiments using the GC/FID reactor with N(2) diluent the following products (molar yields) were observed: 2,3-dichloro-2-methylpropanal [(47.2 +/- 8)% excluding error in calibration]; methacryloyl chloride [(22.9 +/- 2)%]; and 2-chloromethylacrolein [(2.3 +/- 0.8)%]. Addition of 200 ppm O(2) (with Cl(2) = 5000 ppm) resulted in a sharp reduction of the 2,3-dichloro-2-methylpropanal yield (to approximately 2%) with an accompanying appearance of chloroacetone [yield = (55 +/- 7)% decreasing to (44 +/- 7)% in air diluent]. The methacryloyl chloride yield was 23% for [O(2)]/[Cl(2)] ratios from 0 to 0.2 but decreased to near zero as the O(2)/Cl(2) ratio was increased to approximately 400. The variation in methacryloyl chloride yield with the O(2)/Cl(2) ratio in the initial mixture allowed an approximate measurement of the rate constant for the reaction of the methacryloyl radical with O(2) relative to that with Cl(2) (k(O(2))/k(Cl(2)) = 0.066 +/- 0.02). In experiments using the FTIR reactor in 700 Torr of N(2) diluent, methacryloyl chloride [(26 +/- 3)%] and HCl [(27 +/- 3)%] were observed as products. In 700 Torr of air diluent, the observed products were: chloroacetone [(44 +/- 5)%], CO(2) [(27 +/- 3)%], HCl [(21 +/- 3)%], and HCHO [(14 +/- 2)%], and CH(3)C(O)CH(2)OH (3-4%). The observation of CH(3)C(O)CH(2)OH indicates the presence of OH radicals in the system. At atmospheric pressure and 297 K, the title reaction proceeds [(24.5 +/- 5)%] via abstraction of the aldehydic hydrogen atom, [(2.3 +/- 0.8)%] via abstraction from the -CH(3) group, and approximately [(47 +/- 8) %] via addition to the CH(2)=C < double bond with most of the addition occurring at the terminal carbon atom (uncertainties represent statistical 2sigma). The results are discussed with respect to the literature data.

Background: Sex determination processes vary widely among different vertebrate taxa, but no group offers as much diversity for the study of the evolution of sex determination as teleost fish. However, the knowledge about sex determination gene cascades is scarce in this species-rich group and further difficulties arise when considering hybrid fish taxa, in which mechanisms exhibited by parental species are often disrupted. Even though hybridisation is frequent among teleosts, gene based approaches on sex determination have seldom been conducted in hybrid fish. The hybrid polyploid complex of Squalius alburnoides was used as a model to address this question., Methodology/principal Findings: We have initiated the isolation and characterization of regulatory elements (dmrt1, wt1, dax1 and figla) potentially involved in sex determination in S. alburnoides and in the parental species S. pyrenaicus and analysed their expression patterns by in situ hybridisation. In adults, an overall conservation in the cellular localization of the gene transcripts was observed between the hybrids and parental species. Some novel features emerged, such as dmrt1 expression in adult ovaries, and the non-dimorphic expression of figla, an ovarian marker in other species, in gonads of both sexes in S. alburnoides and S. pyrenaicus. The potential contribution of each gene to the sex determination process was assessed based on the timing and location of expression. Dmrt1 and wt1 transcripts were found at early stages of male development in S. alburnoides and are most likely implicated in the process of gonad development., Conclusions/significance: For the first time in the study of this hybrid complex, it was possible to directly compare the gene expression patterns between the bisexual parental species and the various hybrid forms, for an extended set of genes. The contribution of these genes to gonad integrity maintenance and functionality is apparently unaltered in the hybrids, suggesting that no abrupt shifts in gene expression occurred as a result of hybridisation.

In mammals, the increase in gene dosage, in the form of polyploidy or involving chromosomal fragments, has deleterious effects [1]. Regulation of appropriate gene product amounts has to be warranted by complex dosage-compensation mechanisms. Lower vertebrates, on the other hand, cope very well with ploidy increase [2-4], implying either effective compensation or a lack of necessity for such mechanisms. Unfortunately, nothing is known about the genetic and molecular mechanisms underlying this phenomenon. For an experimental approach, we have studied gene expression in the allotriploid form of Squalius alburnoides. In these organisms, different genomes are joined through hybridization; thus, sequence differences can be used to follow expression of different alleles [5, 6]. We found that a compensation mechanism exists, reducing transcript levels to the diploid state. Our data suggest a silencing of one of the three alleles. Unexpectedly, it is not a whole haplome that is inactivated. The allelic expression patterns differ between genes and between different tissues for one and the same gene. Our data provide the first evidence of a regulation mechanism involving gene-copy silencing in a triploid vertebrate.

The reactions of Cl atoms with cis- and trans-2-butene have been studied using FTIR and GC analyses. The rate constant of the reaction was measured using the relative rate technique. Rate constants for the cis and trans isomers are indistinguishable over the pressure range 10-900 Torr of N2 or air and agree well with previous measurements at 760 Torr. Product yields for the reaction of cis-2-butene with Cl in N2 at 700 Torr are meso-2,3-dichlorobutane (47%), DL-2,3-dichlorobutane (18%), 3-chloro-1-butene (13%), cis-1-chloro-2-butene (13%), trans-1-chloro-2-butene (2%), and trans-2-butene (8%). The yields of these products depend on the total pressure. For trans-2-butene, the product yields are as follows: meso-2,3-dichlorobutane (48%), dl-2,3-dichlorobutane (17%), 3-chloro-1-butene (12%), cis-1-chloro-2-butene (2%), trans-1-chloro-2-butene (16%), and cis-2-butene (2%). The products are formed via addition, addition-elimination from a chemically activated adduct, and abstraction reactions. These reactions form (1) the stabilized 3-chloro-2-butyl radical, (2) the chemically activated 3-chloro-2-butyl radical, and (3) the methylallyl radical. These radicals subsequently react with Cl2 to form the products via a proposed chemical mechanism, which is discussed herein. This is the first detailed study of stereochemical effects on the products of a gas-phase Cl+olefin reaction. FTIR spectra (0.25 cm(-1) resolution) of meso- and DL-2,3-dichlorobutane are presented. The relative rate technique was used (at 900 Torr and 297 K) to measure: k(Cl + 3-chloro-1-butene) = (2.1 +/- 0.4) x 10(-10), k(Cl + 1-chloro-2-butene) = (2.2 +/- 0.4) x 10(-10), and k(Cl + 2,3-dichlorobutane) = (1.1 +/- 0.2) x 10(-11) cm3 molecule(-1) s(-1).

In mammals, the anti-Müllerian hormone (Amh) is responsible for the regression of the Müllerian ducts; therefore, Amh is an important factor of male sex differentiation. The amh gene has been cloned in various vertebrates, as well as in several teleost species. To date, all described species show a sexually dimorphic expression of amh during sex differentiation or at least in differentiated juvenile gonads. We have identified the medaka amh ortholog and examined its expression pattern. Medaka amh shows no sexually dimorphic expression pattern. It is expressed in both developing XY male and XX female gonads. In adult testes, amh is expressed in the Sertoli cells and in adult ovaries in granulosa cells surrounding the oocytes, like in mammals. To better understand the function of amh, we cloned the anti-Müllerian hormone receptor type II (amhrII) ortholog and compared its expression pattern with amh, aromatase (cyp19a1), and scp3. During gonad development, amhrII is coexpressed with medaka amh in somatic cells of the gonads and shows no sexually dimorphic expression. Only the expression level of the Amh type II receptor gene was decreased noticeably in adult female gonads. These results suggest that medaka Amh and AmhrII are involved in gonad formation and maintenance in both sexes.

Riboflavin binding protein, purified from egg white, binds copper(II) under dialysis conditions in an approximately 1:1 molar ratio. Results further indicate a small, but not negligible, amount of copper is present in the protein as purified from egg white. Electron paramagnetic resonance indicates a single type II copper site present in the protein. These results suggest the possibility of a previously unknown function of riboflavin binding protein in the storage or transport of copper.

Primary or idiopathic segmental infarction of the greater omentum is a rare surgical condition. We describe a case of omental torsion in an adult patient who was diagnosed preoperatively by contrast-enhanced computed tomography and managed by laparoscopy.
