A 28‐day experiment was carried out to evaluate different C/N ratios (10:1, 15:1 and 20:1) on growth performance, water quality, survival rate and gene expression related to metabolic pathways of Penaeus vannamei in a biofloc system. To maintain the C/N ratio of the feed input, different carbon sources (molasses, rice bran and wheat flour) in combination were applied after fermentation for a day with biofloc consortium. Results showed that shrimps reared in C/N 10 (630 mg) and C/N 15 (646 mg) biofloc systems showed significantly (p =.0229 and.0263) higher growth when compared to C/N 20 (528 mg) and control (374 mg). With regard to the production of extracellular enzymes, protease, lipase and xylanase were found predominantly in colonized bacteria isolated from biofloc treatments, whereas amylase was commonly found in all the treatments. The proPO activity was considerably increased in C/N 10 and 15 treatments than C/N 20 and control. The ranges of relative expression levels of potential genes entailed in metabolite pathway like hexokinase (HK‐ 6.57–8.17), pyruvate kinase (PK‐5.79–7.96), crustacean hyperglycaemic hormone (CHH‐ 6.99–7.66) and fatty acid synthase (FAS‐ 6.57–8.86), triacylglycerol lipase (TGL‐6.99–8.66) and immune gene (Toll receptor‐ 4.57–4.86) fold up‐regulated expression were observed in the biofloc treatments than that of the control. The overall findings indicate that the rearing of P. vannamei shrimp in C/N 10 and 15 biofloc systems could improve the growth performance, water quality and development of immune and metabolic responses. Abstract: Channa striatus, also called snakehead murrel, is an important freshwater teleost fish which has been widely cultured for its tasty flesh along with nutritional and medicinal values. The growth of both cultured and wild murrels is affected by various physical, chemical and biological factors. As a teleost fish, C. striatus is an intermediate organism between invertebrates and vertebrates. They have a well‐developed innate immune system than invertebrates and a primitive adaptive immune system compared to that of higher vertebrates, thus an interesting unique immune structure to explore. Studies have identified that a few external stimulants do instigate the immune system to fight against the pathogens at the time of infection in C. striatus. This review discusses the physicochemical and biological stress factors, immune system and immune molecules of C. striatus which are potentially involved in combating the stress factors. Heat shock cognate 70 (HSC70) is an important evolutionary conserved protein that plays a major role in maintaining the homeostasis and immunity of many organisms. In this study, a HSC70 from Channa striatus was identified from its cDNA library and characterized using bioinformatics and molecular biology tools. CsHSC70 cDNA was 1953 base pair (bp) in length along with an open reading frame which encoded a polypeptide of 650 amino acid residues. Tissue distribution results showed that CsHSC70 was considerably expressed in gill, to a lesser extent in head kidney, blood, spleen and liver and at low level in other tissues. Using C. striatus gill as cell model, effects of fungal, bacterial and poly I:C stimulant on the mRNA levels of CsHSC70 was examined. We also described the antimicrobial features of two peptides namely CsHSC70 A1and CsHSC70 A2 derived from the N-terminal of CsHSC70 protein. CsHSC70 A1 peptide (40 µg/ml) exhibited potent bactericidal activity against Micrococcus luteus cells. Flow cytometric analysis revealed that the M. luteus cells stained with propidium iodide, upon treated with CsHSC70 A1 at the concentration of 40 µM/ml showed 38% survival compared to its control (99.6%). It seems that CsHSC70 A1 peptide shows antimicrobial activity against M. luteus through membrane disruption. Additionally, scanning electron microscope (SEM) observation confirmed that CsHSC70 A1 peptide treatment completely damaged and destructed the M. luteus cells. Taken together, these findings suggest that CsHSC70 A1 peptide could be a safe and potential therapeutic molecule substitute to antibiotics in various clinical fields. [ABSTRACT FROM AUTHOR]

Macrobrachium rosenbergii commonly called giant freshwater prawn is a widely farmed crustacean in freshwater. Similar to other aquatic organisms, their growth and well-being is influenced by various physical, chemical and biological factors. We discuss about the critical growth limiting factors as well as disease causing agents and the potential immune molecules of M. rosenbergii that are proved to involve in preventing and/or responding to those limiting factors. Coxiella burnetii is an obligate intracellular Gram-negative bacterium that causes acute and chronic Q fever in humans. Acute Q fever is usually a flu-like, self-limiting or treatable illness, but some infections can turn into a severe and sometimes fatal chronic disease. There is currently no FDA-approved vaccine available for the prevention of human Q fever in the US, development of a safe and effective vaccine for the prevention of human Q fever remains an important goal for public health. However, there is a fundamental gap in knowledge regarding the mechanism of protective immunity against C. burnetii infection. To understand the mechanism of C. burnetii infection induced protective immunity, we examined if C. burnetii Nine Mile phase I (NMI) infection induces protection against C. burnetii reinfection in mice. Our results indicate that NMI-infected mice conferred significant protection against C. burnetii reinfection. We also found that NMI infection derived protection did not depend on the routes of infection and antibodies are required for NMI infection derived protection. In addition, NMI infection elicited a comparable level of protection in Wild type, CD4 + T cell deficient, and CD8 + T cell deficient mice, partial protection in B cell deficient mice but no protection in T cell deficient mice. These results suggest that both B cells and T cells are required for NMI-infection derived protection, but T cells may play a critical role.
