Il cardo, sia selvatico che coltivato, risulta essere una pianta più tollerante agli stress per cui, essendo botanicamente vicina al carciofo, è possibile trasferire in questa specie tali caratteristiche. È stata quindi studiata l’interazione tra cardo e carciofo e l’utilizzo di eventuali resistenze a stress biotici in carciofo mediante innesti e ibridazioni. Le fasi del lavoro hanno riguardato (a) preliminarmente lo studio citogenetico a scopo conoscitivo di entrambe le specie per una stima del quantitativo di DNA e la definizione del cariotipo; (b) il reperimento del germoplasma di cardo e l’individuazione di fonti di resistenza a Verticillium dahliae; (c) l’utilizzo a breve termine del germoplasma resistente di cardo come portinnesto del carciofo e, (d) a lungo termine, il trasferimento di caratteri in carciofo mediante ibridazioni carciofo/cardo. L’obiettivo a breve termine ha mirato ad utilizzare i genotipi di cardo resistenti a V. dahliae mediante la tecnica di innesto carciofo/cardo, mettendo a punto una procedura ottimale, effettuando studi anatomico-fisiologico nel punto di unione, valutando la compatibilità e le traslocazioni dal portinnesto al nesto, ma soprattutto la risposta agronomica delle combinazioni di innesto. Il carciofo è una Asteracea (Fam. Compositae), specie Cynara cardunculus subsp. scolymus L. che, secondo i rari dati esistenti in letteratura, possiede 34 cromosomi e circa 2,22 pg di DNA (Marie D., et al., 1993) mentre il cardo (C. cardunculus var. altilis) possiede anch’esso 34 cromosomi ma un quantivo di DNA pari a 2,10-2,11 pg (Khaldi et al., 2014); in bibliografia risulta assente uno studio approfondito del cariotipo per entrambe le specie. I genotipi utilizzati per la prova di citologia sono stati: un cardo coltivato (AFGI), un cardo selvatico (AFSA) e due varietà ibride di carciofo (Romolo ed Istar). L’attività è inziata con l’individuazione del tampone ottimale per l’estrazione dei nuclei da giovani foglie da utilizzare per la stima del contenuto di DNA mediante citofluorimetro. I tamponi testati sono stati: il Tris MgCl2, l’LB01, l’Otto I e II. Successivamente è stata applicata la tecnica di estrazione dei nuclei dalle giovani radici, i cui apici sono stati fissati in formaldeide al 3,5% a diversi tempi di permanenza e successivamente a diverse velocità e tempi di frantumazione per mezzo di un mini-frullatore. Il preparato ottenuto è stato saggiato attraverso un citofluorimetro da cui si è potuto stimare e confrontare il valore di DNA con quello presente in bibliografia e constatare il quantitativo leggermente inferiore per il cardo rispetto al carciofo. Gli esperimenti hanno dimostrato che il procedimento migliore per l’ottenimento delle sospensioni di nuclei da utilizzare al citofluorimetro è la fissazione in formaldeide 3,5% delle giovani foglie di cardo e carciofo; il mais è stato utilizzato come specie di riferimento. I quantitativi stimati di DNA rispettivamente del carciofo e del cardo sono stati 2,48 e 2,29 pg. Lo studio del cariotipo del carciofo e del cardo ha inizialmente richesto l’ottimizzazione del protocollo per la sincronizzazione delle metafasi; per questo motivo sono state testate diverse metodologie, concentrazioni e tempi di permanenza degli apici radicali di carciofo e cardo in APM (amiprophos-methyl), 8-idrossichinolina, orizalina, -bromonaphtalene, paradichlorobenzene (PDB) e ghiaccio. È stato constatato che l’effetto migliore di sincronizzazione delle metafasi si ha con l’orizalina e l’8-idrossichinolina, che permettono di ottenere un 20% di metafasi e diminuiscono anche la condensazione dei cromosomi. Il ghiaccio ha invece l’effetto opposto di condensare i cromosomi. La colorazione dei cromosomi è stata quindi eseguita con DAPI e metodo Feulgen, da cui sono state ottenute le foto ed utilizzate per l’ottenimento dei primi cariotipi di cardo e carciofo sia mediante il programma Imagej sia utilizzando le immagini ottenute dalle ibridazioni in situ (FISH) effettuate. Le sonde utilizzate a riguardo sono state, sia per il carciofo che per il cardo, le seguenti: (AG)6 – (AAT)7 – (GAA)7 – (TTAGGG)5 – (AG)12 – (GACA)4 – (AG)9 – (CAT)5 – (ATT)8 – (AGG)5 – (GA)10 – (CA)10 – (AAC)5 – (AT)7 – (ATC)5 – pTa 71. Si tratta di sonde definite “oligonucleotidi”, risultate da marcatori molecolari e quindi da sequenze ripetute e piccole di DNA marcate con dei fluorofori, che si legano alle sequenze corrispettive del DNA di cardo e carciofo. I metodi FISH applicati sono stati la tecnica “Drop spread”, applicata con delle modifiche (Giorgi et al., 2013), e la FISH Standard (Scwarzacher e 3 Heslop Harrison, 2000). Grazie a Levan, un plug-in di Imagej, sono state misurate le lunghezze e tutti i rapporti dei bracci di ognuno dei 34 cromosomi di cardo e carciofo, confermando l’esistenza preponderante di cromosomi sub-metacentrici e di altri più piccoli acrocentrici, dove un braccio è molto piccolo e quasi assente. Contemporaneamente allo studio del cariotipo del carciofo e del cardo è stato reperito il germoplasma di cardo selvatico e coltivato da saggiare per la resistenza a V. dahliae. In totale è stato fatto uno screening di 51 accessioni di cardo a cui sono state affiancate anche 5 varietà commerciali di carciofo come controllo. Il protocollo di inoculazione è consistito nel taglio delle radici e immersione delle piantine nella sospensione fungina con 5x105 conidi/ml. L’incubazione è stata effettuata in condizioni controllate (t=24+2°C; U.R.=60%). Sono state individuate in totale 9 accessioni di cardo più resistenti a V. dahliae. Le piante sopravvissute ai test sono state trasferite in vaso e propagate per una loro moltiplicazione. Le tolleranze individuate nelle accessioni di cardo sono state quindi utilizzate, a breve termine, mediante innesti carciofo/cardo. A riguardo, sono state applicate tre tipologie di innesto, a spacco inglese e spacco in testa con o senza i cotiledoni, per i seguenti genotipi: cardo coltivato (CC) – AFB, AFGI, Versi; cardo selvatico (WC) – AFSA, AFSI, come portainnesti e carciofo – F1 Romolo, Istar e Opal come nesti. La tecnica a spacco inglese in carciofo non presenta la problematica del ricaccio dei germogli dal portainnesto. Questa è stata osservata con lo spacco in testa dove il taglio è stato eseguito al di sopra dei cotiledoni; con l’eliminazione dei cotiledoni, si potrebbe risolvere il problema. Ha rappresentato notevole importanza lo studio anatomico-fisiologico del punto d’innesto effettuato per mezzo del SEM (Electron Scanning Microscopy) a pressione variabile sulle stesse tipologie d’innesto. Le analisi sono state effettuate a 3, 6, 10, 12, 17, 20, 30 giorni ed anche dopo due mesi dalla data d’innesto, evidenziando che la cicatrizzazione avviene in tutte le combinazioni e non è influenzata né dal genotipo, né dalla tipologia d’innesto. In particolare, la combinazione d’innesto carciofo/WC, ha manifetsato elevata affinità e, già dopo 3 gg. dall’innesto, erano visibili dei plasmodesmi tra i due bionti nel punto d’innesto e, a 6 gg., erano visibili piccoli ponti di connessione tra il tessuto indifferenziato del callo; a 12 gg., il tessuto cominciava a differenziarsi. Nella combinazione carciofo/CC, i plasmodesmi sono comparsi dopo 6 gg dall’innesto, mentre i piccoli ponti di connessione erano visibili solo dopo il decimo giorno; la differenziazione dei tessuti è avvenuta dopo il dodicesimo giorno. Lo studio dei cambiamenti anatomico/fisiologici nel punto d’innesto è stato ampliato con la valutazione della compatibilità dal punto di vista morfologico, verificando il numero dei vasi che caratterizzano i diversi genotipi utilizzati come bionti. Nell’innesto, l’uniformità di sviluppo della pianta svolge una notevole importanza nell’attecchimento, ma anche la distribuzione dei vasi ed il loro numero facilitano la cicatrizzazione dei due bionti. I genotipi utilizzati sono stati: il cardo selvatico AFSA ed il coltivato AFB mentre il carciofo Romolo è stato utilizzato come nesto. Sono state fatte delle sezioni trasversali dei fusti che, viste al microscopio ottico, hanno evidenziato come la posizione dei vasi sia concentrata al centro; le sezioni di AFB e Romolo, oltre ad essere simili per dimensione, hanno anche un numero e una simmetria dei vasi che invece non si trova nel cardo selvatico AFSA. È stato poi praticato l’innesto a spacco inglese alle stesse piantine e, dopo 26 giorni, sono state lasciate ad imbibirsi con un colorante per valutare la traslocazione dello stesso nel punto d’innesto. Praticando sezioni longitudinali del fusto nel punto d’innesto, non sono stati evidenti veri e propri vasi ma solo dei punti di connessione e di ripresa della vascolarizzazione. Da un punto di vista agronomico e di attività antiossidante, le piante innestate non hanno mostrato comportamenti significativamente diversi rispetto alle piante non innestate. Per l’obiettivo a lungo termine della tesi, le ibridazioni fino ad ora effettuate sono state:  genotipo maschiosterile MS6 ibridato con 11 genotipi di cardo coltivato – AFB, Bianco gigante, AFGI, AFFF, Cerveteri, AFM, AFN, 33, 32 e selvatico – AFSI, AFSA;  3 linee selezionate (7, 14, 21) di carciofo (portaseme) ibridati con 3 cardi (AFB, AFM) e reciproci. Preliminarmente sono state ripetute per due anni (2012 e 2013) le caratterizzazioni morfologiche delle linee parentali utilizzate di cardo; in particolare sono state valutate 3 piante per 4 ognuno degli 11 genotipi di cardo, utilizzando i descrittori UPOV specifici per il cardo proposti dalla Dott.ssa Romana Bravi del C.R.A. Svolge un ruolo importante la ricerca di piante maschiosterili che, per la segregazione dei caratteri, si manifesta in generazione F2, dopo autofecondazione degli ibridi. Pertanto, è stata valutata la vitalità pollinica di diverse F2 carciofo x cardo mediante colorazione con carminio acetico. Su un totale di 273 piante analizzate, sono state individuate 6 piante maschiosterili. In conclusione, l’attività svolta in questa ricerca ci ha permesso di stimare il quantitativo di DNA del cardo e del carciofo e di effettuare i rispettivi cariotipi mai sviluppati fino ad ora, ma vista la grande difficoltà nel sincronizzare, nello stadio giusto della mitosi, gli apici radicali e quindi nell’ottenere un numero di metafasi sostanzioso, si necessita di un ulteriore approfondimento specifico. Lo studio verrà facilitato con l’utilizzo della metodica FISH e di sonde specifiche per il carciofo che permetteranno d’identificare i singoli cromosomi. Sono state individuate 9 accessioni di cardo risultate resistenti a V. dahliae, di cui 5 di cardo selvatico e 4 di cardo coltivato; sarà importante effettuare una moltiplicazione e caratterizzazione morfologica in campo di questi genotipi. Agronomicamente l’innesto migliore è risultato lo spacco inglese perché non presenta la problematica dell’emissione dei germogli laterali di cardo; al contrario l’analisi anatomica e fisiologica ha invece evidenziato una capacità di attecchimento migliore dello spacco in testa, vista la maggiore superficie di contatto tra i bionti. Questo è stato anche confermato dalle analisi SEM con cui è stato seguito il processo di cicatrizzazione in diverse combinazioni di innesto. Il trasferimento di geni interessanti dal cardo al carciofo richiede un’attività a lungo termine, e sono stati avviati i lavori per definire i parentali ottimali di cardo (maschile) ed individuati ulteriori genotipi maschiosterili come parentali femminili. Sono state tentate alcune combinazioni di incrocio, valutate solo in parte da un punto di vista agronomico. Cultivated and wild cardoon are stress tolerant plants that, being botanically close to artichoke, can transfer this characteristic into this species. So, the PhD work aims at studying the interaction between artichoke and cardoon and at using any resistance to biotic stresses of cardoon both through graftings and hybridizations with artichoke. The phases of the PhD work have been focused on (a) a preliminary cytogenetic study for cognitive purposes of both species to estimate the amount of DNA and the definition of the karyotype; (b) the collection of cardoon germplasm and the identification of sources of resistance to Verticillium dahliae; (c) the short-term use of resistant cardoon rootstocks in graftings with artichoke, (d) the long-term transfer of verticillium resistance sources through artichoke x cardoon hybridizations. The short-term activity aimed at using the cardoon genotypes resistant to V. dahliae in artichoke / cardoon graftings, at developing the best grafting practice to apply, at conducting anatomical and physiological studies on the grafting union formation as well as at studying the rootstock/scion compatibility and translocations; the agronomic response and the antioxidant activity of grafting combinations have been reported. Artichoke is an Asteracea (Fam. Compositae), Cynara cardunculus subsp. scolymus L. that, according to the rare data existing in literature, has 34 chromosomes and about 2.22 pg of DNA (Marie D., et al., 1993); while cardoon (C. cardunculus var. altilis) has also 34 chromosomes but a DNA amount of 2.10 - 2.11 pg (Khaldi et al., 2014); bibliographic references on the karyotype studies of both species are quite absent. The genotypes used for the cytology studies were a cultivated cardoon (AFGI), a wild cardoon (AFSA) and two F1 hybrid varieties of artichoke (Romolo and Istar). The activity was started by the identification of a buffer optimal for the extraction of nuclei from young leaves to be used for the DNA content estimation by flow cytometry. The buffers tested were: Tris MgCl2, the LB01, the Otto I and II. Subsequently, the extraction technique of nuclei from young roots was applied; the apices were fixed in 3.5% formaldehyde at different permanence times, speeds and times of crushing by a mini-blender,. The preparations obtained have been assessed by a flow cytometer from which it was possible to estimate and compare the DNA values with those found in literature; a quantity slightly lower for cardoon respect to artichoke was determined. The experiments have shown that the best procedure for obtaining suspensions of nuclei to be used in flow cytometry is represented by the fixation in 3.5% formaldehyde of the young leaves both of cardoon and artichoke; corn was used as the reference species. The DNA quantities estimated for artichoke and cardoon were 2.48 and 2.29 pg, respectively. Karyotype studies of artichoke and cardoon initially required to optimize the protocol for the synchronization of the metaphases. For this reason, several methods have been tested by dipping, at different concentrations and times, the root tips of both species in APM (amiprophos-methyl), 8-hydroxyquinoline, orizalina, -bromonaphtalene, paradichlorobenzene (PDB) and ice. The best effect of synchronization of metaphases was that with the orizalina and 8-hydroxyquinoline, which allow to obtain a 20% of the metaphases and also decrease the condensation of chromosomes. Ice had the opposite effect of the chromosomes condensation. Staining of the chromosomes was then performed with DAPI and Feulgen method, from which the photos were obtained and used for developing the first karyotypes of cardoon and artichoke either using the Imagej program or the images made by in situ hybridization (FISH). The probes used for both artichoke and cardoon have been as follows: (AG)6 - (AAT)7 - (GAA)7 - (TTAGGG)5 - (AG)12 - (GACA)4 - (AG)9 - (CAT)5 - (ATT)8 - (AGG)5 - (GA)10 - (CA)10 - (AAC)5 - (AT)7 - (ATC)5 - pTa71. They are defined as "oligonucleotide" probes resulting from molecular markers and then by repeated sequences and small DNA labelled with fluorophores, which bind to corresponding DNA sequences of cardoon and artichoke. The methods applied were the FISH "Drop spread" technique 3 modified by Giorgi et al. (2013) and the Standard FISH technique (Scwarzacher and Heslop Harrison, 2000). Levan, a plug-in for ImageJ program, has been used to obtain the lengths and the ratios of the arms of each of the 34 chromosomes of cardoon and artichoke, confirming the existence of predominant sub-metacentric and other smaller acrocentric chromosomes. At the same time, after a preliminary collection, the cultivated and wild cardoon germplasm has been tested for resistance to V. dahliae. In total, a screening of 51 accessions of cardoons, which have been accompanied also by 5 commercial varieties of artichoke as control, has been made. The inoculation protocol consisted in cutting the roots of seedlings and dipping them into the fungal suspension containing 5x105 conidia/ml. Incubation was carried out under controlled conditions (t = 24+2°C, RH = 60%). A total of 9 cardoon accessions have been identified as more or less resistant to V. dahliae. The plants survived after the test was transferred into the pots for their multiplication. The tolerances identified in the cardoon accessions were used in a short term activity based on artichoke/cardoon graftings. In this regard, three types of grafting, tongue and cleft grafting with or without cotyledons, have been applied onto these following genotypes: cultivated cardoon (CC) - AFB, AFGI, Versi and wild cardoon (WC) - AFSA, AFSI as rootstocks and artichoke - F1 Romolo, Istar and Opal as scions. Tongue grafting technique did not give the problem of the regrowth of shoots from the rootstock, as observed with the cleft grafting, where the cut is made above the cotyledons; elimination of the cotyledons might solve the problem. The anatomical and physiological aspects of the grafting union formation, carried out by SEM (Scanning Electron Microscopy) on the same types of grafting, were of considerable importance. The analyses were performed at 3, 6, 10, 12, 17, 20, 30 days and even two months after grafting, pointing out that the grafting union is affected neither by the genotype nor by the type of grafting. In particular, the artichoke/WC grafting combination has shown high affinity already after 3 days after the grafting; plasmodesmata were visible between the two bionts into graft junction in 6 days and small bridges were visible as connections between the undifferentiated callus tissues after 12 days, when the tissues began to differentiate. In the artichoke/CC combination, plasmodesmata have appeared 6 days after grafting, while the small bridges of connection were visible only after the tenth day; tissue differentiation took place after the twelfth day. The study on anatomical/physiological changes into graft union formation has been expanded with the assessment of the compatibility of the two bionts from the morphological point of view, checking the number of vessels that characterize the genotypes used as rootstock and scion. In the grafting, the uniformity of the plant development plays a considerable importance for the graft union, but also the distribution of the vessels and their numbers can facilitate the cicatrization of the cutting surfaces. The genotypes used were: wild cardoon AFSA and cultivated cardoon AFB as a rootstock, while Romolo was used as scion. Sections of the stems were made; by the optical microscope, they showed that the position of the vessels is concentrated in the centre; AFB and Romolo, being of similar size, also have a number and a symmetry of the vessels that instead is not present in the wild cardoon AFSA. Then, tongue grafting technique was practiced onto the same analysed seedlings, which have been left to blend for 26 days in a colorant to evaluate its translocation in the coupling point of the grafting. Longitudinal sections of the stem from the coupling point have been made; obviously, not real vessels but only connection points and recovery of the vasculature were evident. From an agronomical point of view and antioxidant activity, the grafted plants did not show any significant different behaviour respect to the non-grafted plants. For the long-term activity of the thesis, hybridizations were carried out. They were as follows: • male-sterile genotype MS6 hybridized with 11 genotypes of cultivated cardoon - AFB, Bianco Gigante, AFGI, AFFG, Cerveteri, AFM, AFN, 33, 32 and wild - AFSI, AFSA; • 3 artichoke selected lines (7, 14, 21) (seed-bearing) hybridized with 3 cardoons (AFB, AFM) and reciprocal. Preliminarily, a morphological characterization were repeated for two years (2012 and 2013) of the cardoons parental lines used; in particular, 3 plants for each of the 11 genotypes of cardoon were evaluated using the UPOV descriptors specific for cardoon, as proposed by Dr. Roman Bravi from CRA. The search for male sterile plants, occurring in the F2 generation after selfing of hybrids 4 for the segregation of characters, plays an important role in breeding activity. Pollen vitality of different F2 artichoke x cardoon populations was assessed by its staining with acetic carmine. Out of a total of 273 plants analysed, 6 new male-sterile plants were identified. In conclusion, the activities carried out in this study allowed to estimate the amount of DNA of cardoon and artichoke and to make their karyotypes never seen before. Nevertheless, considering the great difficulties in synchronizing the cells of the root tips in the right stage of mitosis, so getting a substantial number of metaphases, further specific studies are required. Use of the FISH analysis and specific probes for artichoke, that will allow to identify the individual chromosomes, could implement the data up now obtained. Nine accessions of cardoon proved to be resistant to V. dahliae, these including 5 of wild and 4 of cultivated cardoon; a multiplication and morphological characterization of these genotypes in the field is important. Agronomically, the tongue technique revealed to be the best grafting procedure used because it does not give the problematic development of side shoots from the rootstock cardoon; on the contrary, the anatomical and physiological analyses have instead showed a better engraftment capacity of the bionts by cleft grafting, due to the greater surface area of contact between rootstock and scion. Considering that the transfer of characters of interest by artichoke x cardoon hybrizations requires a long-term activity, nevertheless, some effort has been made to obtain stable parental lines by alternating selfing and sib-crosses to avoid negative effects due to inbreeding depression.
