BACKGROUND: Necroptosis is a caspase-independent regulated necrotic cell death modality that elicits strong adaptive immune responses, and has the potential to activate antitumor immunity. Long non-coding RNAs (lncRNAs) have critical effects on oral squamous cell carcinoma (OSCC), which are closely associated with the prognosis and immune regulation of OSCC patients. OBJECTIVE: This study aimed to identify a novel necroptosis-related lncRNAs signature to predict the prognosis and immune response of OSCC patients and provide patients with anti-tumor drug selection through bioinformatics analysis and in vitro experiments. METHODS: A series of analyses, including differential lncRNA screening, survival analysis, Cox regression analysis, ROC analysis, nomogram prediction, enrichment analysis, tumor-infiltrating immune cells, drug sensitivity analysis, and consensus cluster analysis, were performed to determine and validate the prognostic value of necroptosis-associated lncRNAs signature in OSCC. And real-time quantitative polymerase chain reaction (RT-qPCR) was used to determine the expression levels of these lncRNAs. RESULTS: This signature including 5 lncRNAs (AC099850.3, StarD4-AS1, AC011978.1, LINC01503, CDKN2A-DT) in OSCC associated with necroptosis were established and verified by bioinformatics. Further, ROC, K-M, univariate/multivariate Cox regression, and nomogram analysis were used to evaluate the model's features for OSCC prognosis. Using multiple bioinformatics techniques, the levels of tumor-infiltrating immune cells, immune checkpoints and semi-inhibitory concentrations showed significant differences across risk subtypes. By consensus cluster analysis, there were significant differences between clusters in survival, immune checkpoint expression, clinicopathological correlation, and tumor immunity. RT-qPCR showed that AC099850.3, AC011978.1, LINC01503 were up-regulated, STARD4-AS1 and CDKN2A-DT were down-regulated in OSCC cell lines compared with human normal oral keratinoid cell line. CONCLUSION: We established 5-NRLs markers, which is useful for assessing OSCC immune response and prognosis, recommending personalized antitumor drugs. The expression level of 5-NRLs in OSCC was identified in vitro, and the results preliminarily verified this model. And this study would generate new insights for future experimental research. This study aimed to assess whether EA could ameliorate intestinal barrier damage that had been reversed in a mouse model of obesity induced by a high-fat diet (HFD) and whether this repair is correlated with ferroptosis and gut microbiota enhancement.Methods: To assess the potential of EA to prevent obesity and restore the intestinal barrier, we divided in C57BL/6J mice into two groups; one was fed with HFD and another one with a normal diet. Samples of stool, blood, fat, and intestinal epithelium were then evaluated, along with body weight.Results: Following EA, we observed a significant reduction in body weight, fat accumulation, and serum triglyceride (TG), total cholesterol (TC), and low-density lipoprotein cholesterol (LDL-C) levels; an increase was seen in high-density lipoprotein cholesterol (HDL-C) levels. EA also activated the Nrf2 signaling pathway; upregulated the expression of GPX4, FTH1, and SLC7A11; and downregulated the expression of TFR1. In addition, the administration of EA resulted in a notable modification of the gut microbiota composition, characterized by a decrease in the Firmicutes to Bacteroidetes ratio.Conclusion: EA had beneficial effects on weight loss and showed potential ability to repair the intestinal barrier by activating the Nrf2 signaling pathway, inhibiting intestinal inflammation and ferroptosis, and regulating the intestinal microbiota to treat IBD caused by HFD-induced obesity. Nanoporous ZnMn2O4 nanorods have been successfully synthesized by calcining β‐MnO2/ZIF‐8 precursors (ZIF‐8 is a type of metal–organic framework). If measured as an anode material for lithium‐ion batteries, the ZnMn2O4 nanorods exhibit an initial discharge capacity of 1792 mA h g−1 at 200 mA g−1, and an excellent reversible capacity of 1399.8 mA h g−1 after 150 cycles (78.1 % retention of the initial discharge capacity). Even at 1000 mA g−1, the reversible capacity is still as high as 998.7 mA h g−1 after 300 cycles. The remarkable lithium‐storage performance is attributed to the one‐dimensional nanoporous structure. The nanoporous architecture not only allows more lithium ions to be stored, which provides additional interfacial lithium‐storage capacity, but also buffers the volume changes, to a certain degree, during the Li+ insertion/extraction process. The results demonstrate that nanoporous ZnMn2O4 nanorods with superior lithium‐storage performance have the potential to be candidates for commercial anode materials in lithium‐ion batteries. Unique hierarchical mosaic‐structure SiOx/C@CoO nanosheets were successfully synthesized from SiOx/C nanospheres embedded in CoO nanosheets through a high‐energy ball milling and hydrothermal self‐assembly process. The SiOx/C@CoO material exhibited good cyclic performance with a reversible discharge capacity of 1079 mAh g−1 over 250 cycles at a current density of 1000 mA g−1. It also displayed an excellent rate capability (with an average specific reversible discharge capacity of 777.8 mAh g−1 at 2000 mA g−1 and 480 mAh g−1 at 5000 mA g−1). The stable skeleton structure of SiOx/C‐embedded CoO nanosheets and the synergistic effect between the two materials should be responsible for the improvement of the lithium storage. This work proposes a feasible approach to promote CoO application in lithium‐ion batteries. In the mill: Unique hierarchical mosaic‐structure SiOx/C@CoO nanosheets, with both SiOx and graphite particles embedded in a CoO layered structure, are successfully fabricated through a high‐energy ball‐milling process and through a one‐step hydrothermal process to form a 3D hierarchical mosaic structure. The unique combination of the CoO and SiOx/C can accommodate the large volume changes and retain the structural integrity of the nanosheets, maintaining the cycling stability and rate capability of SiOx/C@CoO electrode materials.
