The jaguar (Panthera onca) is the largest living cat species native to the Americas and one of few large American carnivorans to have survived into the Holocene. However, the extent to which jaguar diversity declined during the end-Pleistocene extinction event remains unclear. For example, Pleistocene jaguar fossils from North America are notably larger than the average extant jaguar, leading to hypotheses that jaguars from this continent represent a now-extinct subspecies (Panthera onca augusta) or species (Panthera augusta). Here, we used a hybridization capture approach to recover an ancient mitochondrial genome from a large, late Pleistocene jaguar from Kingston Saltpeter Cave, Georgia, United States, which we sequenced to 26-fold coverage. We then estimated the evolutionary relationship between the ancient jaguar mitogenome and those from other extinct and living large felids, including multiple jaguars sampled across the species' current range. The ancient mitogenome falls within the diversity of living jaguars. All sampled jaguar mitogenomes share a common mitochondrial ancestor ~400 thousand years ago, indicating that the lineage represented by the ancient specimen dispersed into North America from the south at least once during the late Pleistocene. While genomic data from additional and older specimens will continue to improve understanding of Pleistocene jaguar diversity in the Americas, our results suggest that this specimen falls within the variation of extant jaguars despite the relatively larger size and geographic location and does not represent a distinct taxon.

The snow leopard (Panthera uncia) is a vulnerable species native to the mountain ranges of Central and South Asia, and many snow leopards are distributed in China. A distinct genetic background and population history are critical for guiding conservation, particularly for snow leopards, but have rarely been explored. In this study, we assembled the mitochondrial genomes of 136 individuals from the Tibetan Plateau (Central group) and conducted a comprehensive genetic assessment on the basis of these mitochondrial genomes. We detected 84 polymorphic sites and 32 distinct haplotypes, with 24 haplotypes first reported in this study, revealing low genetic diversity in the snow leopard population. Our analysis suggested the possible existence of two distinct subpopulations within the Central group, with great genetic differentiation between them. Bayesian skyline plots showed different trends in the effective population dynamics over time for the two subpopulations. The findings of this study provide new genetic resources and information from a matrilineal perspective for snow leopards and are expected to aid in future planning for the conservation and protection of this species. Animal attacks are a common cause of human injuries worldwide, with human-leopard encounters frequently reported in India. Animal attack injuries are increasingly common in trauma centres due to human encroachment into wildlife habitats, leading to more human-animal interactions. These attacks, especially to the face, can cause significant functional, cosmetic, and structural damage. A 40-year-old woman was attacked by a leopard, resulting in tooth mobility, pain, and orofacial lacerations. Initial treatment and primary wound closure were performed in Ratlam, Madhya Pradesh, India. On further examination, necrosis with blackish discolouration of the left ala of the nose was observed, along with scarring on the lips. Tooth 22 exhibited luxation without alveolar bone involvement, leading to proclination. A multidisciplinary treatment approach was adopted, beginning with manual manipulation and repositioning of tooth 22, followed by stabilisation and endodontic treatment. For extraoral management, plastic surgery was performed, involving a paramedian forehead flap and alar cartilage remodelling using left auricular cartilage. The patient underwent follow-ups at one, three, six months and one year. In conclusion, managing dental trauma from animal attacks requires a multidisciplinary approach and ongoing follow-up. By fostering collaboration among healthcare professionals and ensuring vigilant care, we can significantly improve patient outcomes and recovery.

Between September and November 2021, 5 snow leopards (Panthera uncia) and 1 lion (Panthera leo) were naturally infected with severe acute respiratory coronavirus 2 (SARS-CoV-2) and developed progressive respiratory disease that resulted in death. Severe acute respiratory syndrome coronavirus 2 sequencing identified the delta variant in all cases sequenced, which was the predominant human variant at that time. The time between initial clinical signs and death ranged from 3 to 45 days. Gross lesions in all 6 cats included nasal turbinate hyperemia with purulent discharge and marked pulmonary edema. Ulcerative tracheitis and bronchitis were noted in 4 cases. Histologically, there was necrotizing and ulcerative rhinotracheitis and bronchitis with fibrinocellular exudates and fibrinosuppurative to pyogranulomatous bronchopneumonia. The 4 cats that survived longer than 8 days had fungal abscesses. Concurrent bacteria were noted in 4 cases, including those with more acute disease courses. Severe acute respiratory syndrome coronavirus 2 was detected by in situ hybridization using probes against SARS-CoV-2 spike and nucleocapsid genes and by immunohistochemistry. Viral nucleic acid and protein were variably localized to mucosal and glandular epithelial cells, pneumocytes, macrophages, and fibrinocellular debris. Based on established criteria, SARS-CoV-2 was considered a contributing cause of death in all 6 cats. While mild clinical infections are more common, these findings suggest that some SARS-CoV-2 variants may cause more severe disease and that snow leopards may be more severely affected than other felids. Individual age can be used to design more efficient and suitable management plans in both in situ and ex situ conservation programmes for targeted wildlife species. DNA methylation is a promising marker of epigenetic ageing that can accurately estimate age from small amounts of biological material, which can be collected in a minimally invasive manner. In this study, we sequenced five targeted genetic regions and used 8–23 selected CpG sites to build age estimation models using machine learning methods at only about $3–7 per sample. Blood samples of seven Felidae species were used, ranging from small to big, and domestic to endangered species: domestic cats (Felis catus, 139 samples), Tsushima leopard cats (Prionailurus bengalensis euptilurus, 84 samples) and five Panthera species (96 samples). The models achieved satisfactory accuracy, with the mean absolute error of the most accurate models recorded at 1.966, 1.348 and 1.552 years in domestic cats, Tsushima leopard cats and Panthera spp. respectively. We developed the models in domestic cats and Tsushima leopard cats, which were applicable to individuals regardless of health conditions; therefore, these models are applicable to samples collected from individuals with diverse characteristics, which is often the case in conservation. We also showed the possibility of developing universal age estimation models for the five Panthera spp. using only two of the five genetic regions. We do not recommend building a common age estimation model for all the target species using our markers, because of the degraded performance of models that included all species. This retrospective study aimed to characterize and determine the prevalence of spinal disease in nondomestic felids within a sanctuary population. A review of 304 postmortem examination reports in Panthera species from 2003 to 2021 revealed that 86/304 (28%) were diagnosed with spinal disease. Spinal lesions were categorized according to pathologic process: degenerative (78/86, 91%), developmental (8/86, 9%), inflammatory (6/86, 7%), or neoplastic (8/86, 9%). Degenerative lesions included intervertebral disk disease (IVDD; 66/78, 85%), spondylosis without concurrent IVDD (4/78, 5%), and idiopathic (noncompressive) degenerative myelopathies (8/78, 10%). Fourteen individuals had lesions in more than 1 category. Developmental cases were vertebral (4/8) or spinal cord (3/8) malformations or both (1/8). Inflammatory lesions included meningitis (4/6) and meningomyelitis (2/6). Neoplasia included vertebral multiple myeloma (4/8) and others (4/8). IVDD often involved multiple disks but primarily affected the cervical (41/66, 62%) and thoracic spine (32/66, 48%). A multivariate binary logistic model predicted the diagnosis of IVDD at postmortem examination, where odds of being affected were highest for males, lions (Panthera leo), and geriatric age group (> 14 years). The spinal lesions documented in this study provide insight into high-risk signalment categories and predominant associated lesions affecting captive Panthera populations. Introduction: Felids have evolved a specialized suite of morphological adaptations for obligate carnivory. Although the musculoskeletal anatomy of the Felidae has been studied extensively, the comparative neuroanatomy of felids is relatively unexplored. Little is known about how variation in the cerebral anatomy of felids relates to species-specific differences in sociality, hunting strategy, or activity patterns. Methods: We quantitatively analyzed neuropil variation in the prefrontal, primary motor, and primary visual cortices of six species of Felidae (Panthera leo, Panthera uncia, Panthera tigris, Panthera leopardus, Acinonyx jubatus, Felis sylvestris domesticus) to investigate relationships with brain size, neuronal cell parameters, and select behavioral and ecological factors. Neuropil is the dense, intricate network of axons, dendrites, and synapses in the brain, playing a critical role in information processing and communication between neurons. Results: There were significant species and regional differences in neuropil proportions, with African lion, cheetah, and tiger having more neuropil in all three cortical regions in comparison to the other species. Based on regression analyses, we find that the increased neuropil fraction in the prefrontal cortex supports social and behavioral flexibility, while in the primary motor cortex, this facilitates the neural activity needed for hunting movements. Greater neuropil fraction in the primary visual cortex may contribute to visual requirements associated with diel activity patterns. Conclusion: These results provide a cross-species comparison of neuropil fraction variation in the Felidae, particularly the understudied Panthera, and provide evidence for convergence of the neuroanatomy of Panthera and cheetahs. Illegal trade in big cat (Panthera spp.) body parts is a prominent topic in scientific and public discourses concerning wildlife conservation. While illegal trade is generally acknowledged as a threat to big cat species, we suggest that two enabling factors have, to date, been under‐considered. To that end, we discuss the roles of human‐felid conflict, and "local" use in illegal trade in big cat body parts. Drawing examples from across species and regions, we look at generalities, contextual subtleties, ambiguities, and definitional complexities. Santiago Roth (1850–1924) was a naturalist that collected and sold fossils from the Pampean region (Argentina) in Europe. Much of the specimens collected by Roth are hosted at the Paläontologisches Institut und Museum, Universität Zürich and Muséun D’Histoire Naturelle of Geneva in Switzerland, but were never studied. In this article, we review the specimens of the order Carnivora under a taxonomical and temporal approach. We confirm the presence of 16 specimens of Carnivora collected by Roth between the provinces of Santa Fe and Buenos Aires. We identify felids (Smilodon sp., Panthera onca, and specimens related to these taxa), ursids (Arctotherium sp.), and canids (Dusicyon avus and specimens related, Lycalopex sp., and Caninae indet.). According to the information reported by S. Roth, all materials are from the Pleistocene of the Pampean Formation. Simple Summary: This study finds that the skulls and mandibles of lions and tigers in predominantly European zoos differ in shape, but not size, from lions and tigers in the wild. The nature of the shape change found indicates that the mechanical influences of diet have influenced development. The majority of captive big cats used in this study have been fed partial or whole carcasses, which better replicate the mechanical properties of wild diets than softer prepared diets. We therefore speculate that additional mechanical stresses upon the skull and mandible such as the killing bite, manipulation such as dragging, and consumption of large prey in the wild have driven differentiation between the skulls of captive and wild big cats. It is important to understand these differences to better care for captive animals, enhance their survival in reintroductions, and to understand the importance of environmental factors in influencing an animal's size or shape. Zoo animals are crucial for conserving and potentially re-introducing species to the wild, yet it is known that the morphology of captive animals differs from that of wild animals. It is important to know how and why zoo and wild animal morphology differs to better care for captive animals and enhance their survival in reintroductions, and to understand how plasticity may influence morphology, which is supposedly indicative of evolutionary relationships. Using museum collections, we took 56 morphological measurements of skulls and mandibles from 617 captive and wild lions and tigers, reflecting each species' recent historical range. Linear morphometrics were used to identify differences in size and shape. Skull size does not differ between captive and wild lions and tigers, but skull and mandible shape does. Differences occur in regions associated with biting, indicating that diet has influenced forces acting upon the skull and mandible. The diets of captive big cats used in this study predominantly consisted of whole or partial carcasses, which closely resemble the mechanical properties of wild diets. Thus, we speculate that the additional impacts of killing, manipulating and consuming large prey in the wild have driven differentiation between captive and wild big cats. [ABSTRACT FROM AUTHOR]

A skull of a large cat, found in the Zanda Formation in the northwestern Tibetan Himalaya and dated to 4.42 Ma, was originally described as Panthera blytheae and interpreted as sister species of Panthera uncia (Tseng et al. 2014). The position of this cat's infraorbital foramen, the only cranial synapomorphic and key character of the genus Panthera besides the hyoid structure, excludes however a Panthera classification. Two ridges converging to the tip at the labial side of the P4 paracone are quite unusual and were interpreted in the original description as a unique, autapomorphic feature of the Zanda felid. Indeed, such a structure is also found in Middle Pleistocene Neofelis nebulosa primigenia. On the other hand, the dentition of the Zanda cat is not specialised as in Neofelis, and a fragmentary lower molar is rather reminiscent of Middle Miocene North American Pseudaelurus. Consequently, a new genus Palaeopanthera is described for this felid, now called Palaeopanthera blytheae. The early Late Miocene pantherine species, lately classified as Miopanthera pamiri, is also attributed to this genus, now called Palaeopanthera pamiri. Both extant Neofelis species are understood as highly specialised surviving palaeopantherines. The taxonomic affiliation of the two Palaeopanthera species pamiri and blytheae inserts the last mosaic stones to design a more complete and conclusive picture of the phylogeny of the pantherine clade than has been so far possible, from its roots in the earliest Late Miocene to the seven extant species of the genera Neofelis and Panthera. Simple Summary: While people are familiar with the practice of declawing domestic cats, it is also performed on non-domesticated species, such as lions and tigers, to extend the period of safe human interaction for entertainment purposes. Although declawing (removing not only the claw itself, but also the bone from which it grows, whether in part or in whole) clearly affects the skeletal system, the impact on the musculature has not been studied. As the mass of an animal increases cubically as a product of its volume, while the areas of its paws only increase as a square, larger cats have higher foot pressures and, therefore, the surgery may have a greater effect on larger cats. In this study, we evaluate the forearms of clawed and declawed cats to investigate the effects of declawing on muscle architecture. We found that the deep muscles that flex the digits, which are the muscles most directly affected by declawing, are significantly lighter (~73%) and less powerful (46–66%) in animals that have been declawed, while other muscles do not make up for these reductions. Thus, declawing has a substantial effect on the muscular capabilities of cats, and because these deficiencies are not compensated for in larger cats, it probably has even more functionally devastating consequences for these species. While people are familiar with the practice of declawing domestic cats, "onychectomy", as it is also known, is also performed on non-domesticated species, including pantherines, to prolong their use for entertainment purposes. Although the surgery (the partial or complete removal of the distal phalanx) has clear osteological implications, its myological effects have never been studied. As the mass of an animal increases cubically as a product of its volume, while the areas of its paws only increase as a square, larger felids have higher foot pressures and, therefore, the surgery may have particularly substantial functional effects on larger cats. In this study, we evaluate the forearms of clawed and declawed non-domestic felid specimens that spanned the body size range of the whole family to evaluate the effects of onychectomy on muscle fiber architecture. Two species of large cats found at the Tanzanian Pliocene australopithecine locality of Laetoli have been controversial since their first description in 1987. A lion-sized form was referred to Panthera onca gombaszoegensis, P. palaeosinensis, P. shawi, P. leo or to a related form, and most recently compared to P. onca. A leopard-sized species was attributed to P. pardus or to Puma pardoides. Accepting the leopard-like form as the oldest evidence of the genus Panthera to calibrate molecular dating, this site started to play an important role in understanding felid evolution. A new discussion of the extremely scanty material now revises the earlier classifications and shows that the Laetoli "lion" is not a lion and the Laetoli "leopard" is not a leopard. There seems to be no doubt that the lion-sized cat is a species of its own, representing an early stage of Panthera intrageneric evolution, the metapopulation around the first node as defined by molecular phylogenetics. Taxonomically it is introduced as Panthera principialis sp. nov. The Early Pleistocene lion-sized cat from the South African sites Bolt's Farm, Swartkrans and Kromdraai is here attributed species rank, P. shawi, and understood to represent the metapopulation around the lion-leopard evolutionary node. The leopard-sized Laetoli cat qualifies as an early African member of the genus Puma, but it is not the same as the later Early Pleistocene Swartkrans form. The latter, originally described as Panthera pardus incurva, is proposed here to have arisen by introgressive hybridization of an African puma with an early Panthera species and named Puma incurva. Body size correlates with various biological attributes. Hence, its evolution has attracted considerable attention. The family Felidae (Mammalia: Carnivora) has extensive variation in body size among extant species, with a few species being strikingly larger. The body size of extremely large species compared with phylogenetically related species may have evolved through directional selection toward increased size in a particular lineage, although lineage-specific directional selection has not been explicitly incorporated into standard models of body size evolution on a phylogeny. We used simulation-based likelihood and approximate Bayesian computation to examine the effect of lineage-specific directional selection on body mass evolution in extant felids. Our analyses revealed the occurrence of lineage-specific directional selection that favors larger body mass in the genus Panthera (lion, Panthera leo; jaguar, Panthera onca; leopard, Panthera pardus; tiger, Panthera tigris; and snow leopard, Panthera uncia), as well as in the cheetah (Acinonyx jubatus) and puma (Puma concolor). Lineage-specific directional selection was not detected in the clouded leopard (Neofelis nebulosa), which, along with the genus Panthera, diverged early from all other extant felids. The cheetah, puma, and jaguarundi (Puma yagouarondi) compose a monophyletic group in which body size may have decreased during the evolution of the jaguarundi, although our analyses revealed no lineage-specific directional selection for smaller sizes in the species. Directional selection for increased body size shown herein seems to be associated with a lifestyle dependent on relatively large prey. Panthera gombaszoegensis is a fossil pantherine from the Pleistocene of Eurasia. It has been considered to be the closest ancestor the jaguar (Panthera onca) due to dental similarities, and has even sometimes been considered to be a subspecies of jaguar. However, our knowledge of this taxon is limited by the scarcity of cranial remains, which has made it difficult to properly assess the phylogenetic affinities and possible ecological role of this taxon. Here, we describe a new cranium of P. gombaszoegensis from Belgium, and present a morphometric analysis of the cranium and dentition of extinct and extant pantherines. Whereas the lower dentition of P. gombaszoegensis is similar to that of P. onca, similarities were not recovered in other parts of the skull. Some cranial traits of P. gombaszoegensis resemble those of other pantherines, especially larger species such as the tiger (P. tigris), while some similarities to taxa such as tiger (P. tigris), lion (P. leo) and leopard (P. pardus) in the skull of P. gombaszoegensis suggest a diet adapted to a wide prey spectrum. The first ever assessment of the phylogenetic placement of P. gombaszoegensis places this taxon closer to P. tigris than to P. onca, which considerably simplifies the biogeographic history of pantherines. The great cats of the genus Panthera comprise a recent radiation whose evolutionary history is poorly understood. Their rapid diversification poses challenges to resolving their phylogeny while offering opportunities to investigate the historical dynamics of adaptive divergence. We report the sequence, de novo assembly, and annotation of the jaguar (Panthera onca) genome, a novel genome sequence for the leopard (Panthera pardus), and comparative analyses encompassing all living Panthera species. Demographic reconstructions indicated that all of these species have experienced variable episodes of population decline during the Pleistocene, ultimately leading to small effective sizes in present-day genomes. We observed pervasive genealogical discordance across Panthera genomes, caused by both incomplete lineage sorting and complex patterns of historical interspecific hybridization. We identified multiple signatures of species-specific positive selection, affecting genes involved in craniofacial and limb development, protein metabolism, hypoxia, reproduction, pigmentation, and sensory perception. There was remarkable concordance in pathways enriched in genomic segments implicated in interspecies introgression and in positive selection, suggesting that these processes were connected. We tested this hypothesis by developing exome capture probes targeting ~19,000 Panthera genes and applying them to 30 wild-caught jaguars. We found at least two genes (DOCK3 and COL4A5, both related to optic nerve development) bearing significant signatures of interspecies introgression and within-species positive selection. These findings indicate that post-speciation admixture has contributed genetic material that facilitated the adaptive evolution of big cat lineages.

The decline and extirpation of large carnivore populations can lead to cascading effects in natural ecosystems. An understanding of large carnivore population densities, distribution and dynamics is therefore critical for developing effective conservation strategies across landscapes. This is particularly important in island environments where species face increased extinction risk due to genetic isolation coupled with local losses of finite habitat. The Sri Lankan leopard Panthera pardus kotiya is one of two remaining island-living leopards on Earth and the only apex predator in Sri Lanka. Despite its iconic status in Sri Lanka, robust research on the species has been limited to only a handful of scientific studies, limiting meaningful scientific recommendations for the species’ conservation and management. In this study, we conducted a single season camera trap survey in Sri Lanka’s largest protected area, Wilpattu National Park (1317 km2), located in the country’s northwest. Our objective was to estimate key ecological state variables of interest (density, abundance, sex-specific movement and spatial distribution) of this leopard subspecies. Our results indicate that Wilpattu National Park supports a density of 18 individuals/100 km2 (posterior SD=1.5; 95% HPD interval=16–21) with a mean abundance of 144 (posterior SD=15) individual leopards and a healthy sex ratio (f:m=2.03:1). The estimated activity range for male leopards > 2 years old was 49.53 km2 (Posterior SD=3.43; HPD interval=43.09–56.41) and for female leopards > 2 years old was 22.04 km2 (Posterior SD=1.82; HPD interval=18.34–25.65). This density falls at the higher end of published estimates for the species anywhere in its global range, based on similar methods. Given Sri Lanka’s limited size, this national park system should be considered as a critical stronghold that maintains a source population of leopards, contributing to the long-term population viability of leopards in the larger landscape.

Background: Doublesex and mab-3 related transcription factor (DMRT) 1, commonly found in all vertebrates, regulates the transcription of genes involved in the masculinization and maintenance of gonadal somatic cells and/or germline cell development. DMRT1 has a DNA-binding domain called the DM domain and a transcription regulatory region. Unlike the former, there is little knowledge about the latter transcription regulatory region. This study aimed to identify the transcription activation regions of DMRT1 from four species: humans and mice (mammals), leopard geckos (reptiles), and medaka (teleost fish), adding perspectives on evolutionary conservation and diversity., Methods and Results: For each species, several expression plasmids of deletion mutants were constructed, and the resultant plasmid and a DMRT1-driven luciferase reporter were co-transfected into cultured cells to measure transactivation ability. The key point of this analysis is that the transactivation ability was normalized by quantifying the expression levels of DMRT1 variants using the HiBiT tag. As a result, two to three transactivation regions were suggested to exist in the C-terminal region of the DM domain in all four species. Osteoarthritis and degenerative joint disease affect many species of nondomestic felid and negatively impact quality of life in managed care settings. Previously, pain management options were limited because of the frequency of comorbidities such as renal disease and common difficulty encountered in medication administration. Solensia TM (frunevetmab) is a felinized monoclonal antibody that binds to nerve growth factor in arthritic joints, thereby inhibiting pain response cascades, and produces marked improvement in clinical signs and quality of life in domestic cats diagnosed with osteoarthritis. Protein sequence analysis was performed to inform the application of frunevetmab in four individuals from three nondomestic felid species ( Panthera tigris, Lynx rufus, and Panthera uncia ) diagnosed with degenerative joint disease to predict safety, contraindications, and likely response to treatment. Patients were then treated utilizing doses extrapolated from domestic cats and following manufacturer guidelines for administration, and clinical response was evaluated over a minimum 2-mon period. Significant improvement was noted in clinical signs in all four animals, resulting in marked improvement in mobility, lameness, activity level, demeanor, and overall quality of life. Frunevetmab presents an excellent adjunctive therapeutic alternative for nondomestic felids suffering from degenerative joint disease and may complement or decrease the need for other conventional therapeutic regimens.

The aim of this study is to provide an overview of ongoing research on and the development of identification tools for big cats (Panthera tigris, Panthera leo, Panthera pardus, ...). The set of tools includes a species-specific RT PCR quantitation system (nuclear and mitochondrial), STR multiplexes, a rapid system for big cat species determination, and a database solution. Context: Maintaining landscape connectivity for large felids by preserving and restoring corridors between core habitats is crucial to their long-term conservation. Tiger, leopard, and clouded leopard populations occur in isolated habitat patches across the Dawna-Tanintharyi Landscape (DTL) of Kawthoolei (all Karen National Union administrative areas in Myanmar) and Thailand. Objectives: We analyzed connectivity among 18 habitat patches in this transboundary region based on large felid presence and expert opinion of large felid dispersal requirements. Methods: Least-cost corridor and circuit theory analyses were used to identify corridors, determine corridor quality and their relative importance to connectivity in the landscape, and pinpoint bottlenecks to movement. Results: Forty-eight corridors were identified. Lower resistances to dispersal were in forested montane areas. High-quality corridors remained in the northern DTL and south of Tanintharyi Nature Reserve in Kawthoolei based on cost-weighted distance to least-cost path ratio. Pairwise current pinch point analyses revealed a possible landscape level bottleneck to movement north of Thailand's Western Forest Complex. Area corrected centrality scores indicated smaller habitat patches disproportionately contributed to landscape connectivity. Conclusions: The DTL may retain connectivity across the landscape if conservation actions are taken to protect integral habitats and corridors. Conservation efforts that expand the protected area network in Kawthoolei, either by increasing the size of current protected area habitats or by demarcating new protected areas in regions with confirmed felid presence, will aid DTL connectivity. This study investigated the exposure of jaguar populations and domestic animals to smooth Brucella, Leptospira spp. and Toxoplasma gondii in the Cerrado, Pantanal and Amazon biomes of Brazil. Between February 2000 and January 2010, serum samples from 31 jaguars (Panthera onca), 1,245 cattle (Bos taurus), 168 domestic dogs (Canis lupus familiaris) and 29 domestic cats (Felis catus) were collected and analysed by rose bengal test for smooth Brucella, microscopic agglutination test for Leptospira spp. and modified agglutination test for T. gondii. Cattle populations from all sites (9.88%) were exposed to smooth Brucella, but only one jaguar from Cerrado was exposed to this agent. Jaguars captured in the Cerrado (60.0%) and in the Pantanal (45.5%) were seropositive for different serovars of Leptospira spp., cattle (72.18%) and domestic dogs (13.1%) from the three sites and one domestic cat from Pantanal were also seropositive for the agent. The most prevalent serotype of Leptospira spp. identified in jaguars from the Cerrado (Grippotyphosa) and the Pantanal (Pomona) biomes were distinct from those found in the domestic animals sampled. Jaguars (100%), domestic dogs (38.28%) and domestic cats (82.76%) from the three areas were exposed to T. gondii. Our results show that brucellosis and leptospirosis could have been transmitted to jaguars by domestic animals; and jaguars probably play an important role in the maintenance of T. gondii in nature.

Abstract Background A century ago, pantheras were abundant across Asia. Illegal hunting and trading along with loss of habitat have resulted in the designation of Panthera as a genus of endangered species. In addition to the onslaught from humans, pantheras are also susceptible to outbreaks of several infectious diseases, including babesiosis. The latter is a hemoprotozoan disease whose causative agents are the eukaryotic parasites of the apicomplexan genus Babesia. Babesiosis affects a varied range of animals including humans (Homo sapiens), bovines (e.g. Bos taurus), pantheras (e.g. Panthera tigris, P. leo, P. pardus) and equines. Babesia spp. are transmitted by the tick vector Ixodes scapularis or ticks of domestic animals, namely Rhipicephalus (Boophilus) microplus and R. (B.) decoloratus. At the level of protein translation within these organisms, the conserved aminoacyl tRNA synthetase (aaRS) family offers an opportunity to identify the sequence and structural differences in the host (Panthera) and parasites (Babesia spp.) in order to exploit these for drug targeting Babesia spp. Methods Using computational tools we investigated the genomes of Babesia spp. and Panthera tigris so as to annotate their aaRSs. The sequences were analysed and their subcellular localizations were predicted using Target P1.1, SignalP 3.0, TMHMM v.2.0 and Deeploc 1.0 web servers. Structure-based analysis of the aaRSs from P. tigris and its protozoan pathogens Babesia spp. was performed using Phyre2 and chimera. Results We identified 33 (B. bovis), 34 (B. microti), 33 (B. bigemina) and 33 (P. tigris) aaRSs in these respective organisms. Poor sequence identity (~ 20–50%) between aaRSs from Babesia spp. and P. tigris was observed and this merits future experiments to validate new drug targets against Babesia spp. Conclusions Overall this work provides a foundation for experimental investigation of druggable aaRSs from Babesia sp. in an effort to control Babesiosis in Panthera.

Ecometabolomics could be implemented as a powerful tool in molecular ecology studies, but it is necessary to know the baseline of certain metabolites and understand how different traits could affect the metabolome of the animals. Therefore, the main objective of this study was to provide values for the nutritional metabolome profile of different diet groups and animal species, as well as to study the differences in the metabolomic profile due to the effect of diet type and species. To achieve this goal, blood samples were taken from healthy animals (n = 43) of different species: lion (Panthera leo), jaguar (Panthera onca), chimpanzee (Pan troglodytes), bison (Bison bison), gazelle (Gazella cuvieri) and fallow deer (Dama dama), and with different types of diet (carnivore, herbivore and omnivore). Each blood sample was analysed to determine nutritional metabolites. The main results this study provides are the nutritional metabolic profile of these animals based on the type of diet and the animal species. A significant effect of the dietary type was found on nutritional metabolite levels, with those metabolites related to protein metabolism (total protein and creatine) being higher in carnivores. There is also an effect of the species on nutritional metabolites, observing a metabolome differentiation between lion and jaguar. In the case of herbivores, bison showed higher levels of uric acid and cholesterol, and lower urea levels than gazelle and fallow deer. Faeces provide relevant biological information which includes, with the application of genetic techniques, the sex and identity of individuals that defecated, thus providing potentially useful data on the behaviour and ecology of individuals, as well as the dynamics and structure of populations. This paper presents estimates of the sex ratio of different felid species (jaguar, Panthera onca; puma, Puma concolor; and ocelot/margay, Leopardus pardalis/Leopardus wiedi) as observed in field collected faeces, and proposes several hypotheses that could explain the strikingly high proportion of faeces from male jaguars. The proportion of male and female faeces was estimated using a non-invasive faecal sampling method in 14 study areas in Mexico and Brazil. Faecal samples were genetically analysed to identify the species, the sex and the individual (the latter only for samples identified as belonging to jaguars). Considering the three species, 72.6% of faeces (n = 493) were from males; however, there were significant differences among them, with the proportion from males being higher for jaguars than for pumas and ocelots/margays. A male-bias was consistently observed in all study areas for jaguar faeces, but not for the other species. For jaguars the trend was the same when considering the number of individuals identified (n = 68), with an average of 4.2±0.56 faeces per male and 2.0±0.36 per female. The observed faecal marking patterns might be related to the behaviour of female jaguars directed toward protecting litters from males, and in both male and female pumas, to prevent interspecific aggressions from male jaguars. The hypothesis that there are effectively more males than females in jaguar populations cannot be discarded, which could be due to the fact that females are territorial and males are not, or a tendency for males to disperse into suboptimal areas for the species.

Five cases of hemangiosarcoma were documented in three tigers and two lions during post-mortem examinations. Grossly, reddish-black coloured elevated masses were noticed with metastasis to the spleen. In one tiger, an enlarged liver with severe oozing of blood on a cut section was noticed without any elevated masses. In a white tiger, the tumour mass was located near the abdominal region. Histopathological features revealed the presence of immature plump endothelial cells forming blood filled irregular vascular spaces. Immunohistochemically, strong immunoreactivity in the cytoplasm of neoplastic endothelial cells with vascular endothelial growth factor (VEGF) was noticed. No positive reaction was noticed with the proliferative markers, i.e., Ki67 and p53, indicating VEGF is the appropriate marker to diagnose hemangiosarcoma.

Human–wildlife conflict arising from livestock-losses to large carnivores is an important challenge faced by conservation. Theory of prey–predator interactions suggests that revival of wild prey populations can reduce predator's dependence on livestock in multiple-use landscapes. We explore whether 10-years of conservation efforts to revive wild prey could reduce snow leopard's Panthera uncia consumption of livestock in the coupled human-and-natural Trans-Himalayan ecosystem of northern India. Starting in 2001, concerted conservation efforts at one site (intervention) attempted recovery of wild-prey populations by creating livestock-free reserves, accompanied with other incentives (e.g. insurance, vigilant herding). Another site, 50km away, was monitored as status quo without any interventions. Prey remains in snow leopard scats were examined periodically at five-year intervals between 2002 and 2012 to determine any temporal shift in diet at both sites to evaluate the effectiveness of conservation interventions. Consumption of livestock increased at the status quo site, while it decreased at the intervention-site. At the intervention-site, livestock-consumption reduced during 2002–2007 (by 17%, p=0.06); this effect was sustained during the next five-year interval, and it was accompanied by a persistent increase in wild prey populations. Here we also noted increased predator populations, likely due to immigration into the study area. Despite the increase in the predator population, there was no increase in livestock-consumption. In contrast, under status quo, dependence on livestock increased during both five-year intervals (by 7%, p=0.08, and by 16%, p=0.01, respectively). These contrasts between the trajectories of the two sites suggest that livestock-loss can potentially be reduced through the revival of wild prey. Conserving large carnivores that live in close proximity to people depends on a variety of socio‐economic, political and biological factors. These include local tolerance toward potentially dangerous animals, efficacy of human–carnivore conflict mitigation schemes, and identifying and then addressing the underlying causes of conflict. The Leuser Ecosystem is the largest contiguous forest habitat for the critically endangered Sumatran tiger. Its extensive forest edge is abutted by farming communities and we predict that spatial variation in human–tiger conflict (HTC) would be a function of habitat conversion, livestock abundance, and poaching of tiger and its wild prey. To investigate which of these potential drivers of conflict, as well as other biophysical factors, best explain the observed patterns, we used resource selection function (RSF) technique to develop a predictive spatially explicit model of HTC. From 148 conflict incidences recorded from 2008 to 2018 across the Leuser Ecosystem, the areas that were closer to villages and with lower occurrence of wild prey were most susceptible to tiger attacks. From 18 districts monitored, 6 stood out for having disproportionately high levels of HTC. We recommend that these areas be prioritized with increased support from conflict mitigation teams to prevent further injuries to people, livestock or tigers; district governments address one underlying cause of HTC by supporting improved animal husbandry practices, such as tiger‐proof livestock pen construction; and, an increase in ranger patrol effort to recover wild prey populations. This type of priority setting approach has wide application for better determining the required management response to reduce conflicts between people and large carnivores in both tropical and temporal landscapes. Jaguars (Panthera onca) are opportunistic predators that prey on large profitable prey items, such as sea turtles at nesting beaches. Here, we use jaguar and sea turtle track‐count surveys, combined with satellite telemetry of one jaguar, to evaluate whether jaguar hunting behavior and movements are influenced by seasonal sea turtle nesting in the Sector Santa Rosa of Área de Conservación Guanacaste in northwest Costa Rica. We used generalized linear models to evaluate the effect of moon phase and sea surface temperature on olive ridley (Lepidochelis olivacea) and green turtle (Chelonia mydas) nesting abundance, as well as the combination of these predictors on the frequency of jaguar predation activity (proximity to nesting beaches) and movements. For home‐range size and location analyses, we calculated kernel density estimates for each season at three different temporal scales. Sea turtle nesting season influenced jaguar activity patterns, as well as sea turtle abundance was related to jaguar locations and predation events, but jaguar home‐range size (88.8 km2 overall) did not differ between nesting seasons or among temporal scales. Environmental conditions influenced sea turtle nesting and, as a consequence, also influenced jaguar movements and foraging activity. Our study defined the home range of a female jaguar in the tropical dry forest and its relationship to seasonally abundant turtles. Additional information related to the effect of tourism on jaguar–sea turtle interactions would improve conservation of these species at unique nesting beaches in the area. [ABSTRACT FROM AUTHOR]

Identifying factors promoting jaguar (Panthera onca) occupancy is crucial for planning effective conservation and management actions that can contribute to long-term population viability. We used camera-trapping and modeled factors affecting detection and occupancy for jaguars in Sonora at 149 sites during August–November 2008 and 2009. We measured 24 covariates presumed to affect detection (n = 6) and occupancy (n = 18) at camera sites, including sampling period and various habitat characteristics. We obtained 29 jaguar detections at 19 sites (naïve occupancy = 0.12) in 5455 trap-days of effort. Jaguar detectability (p = 0.16 ± 0.05) was negatively affected by human presence and varied by study year and sampling period. Jaguar occupancy (ψ = 0.30 ± 0.03) increased as prey richness, the abundance of calves of domestic cattle, and the proportion of subtropical vegetation increased. Jaguar occupancy was lower with increased abundance of adult cattle and peccary, and higher levels of fragmentation. Jaguar occupancy in Sonora was hence driven by a diverse combination of factors which should be considered when planning conservation actions. We suggest that managers mitigate the impact of livestock, especially on subtropical habitats, by employing appropriate fencing. Fencing may increase wildlife prey for jaguars, thereby reducing calf depredation by jaguars, while concurrently protecting habitat from overgrazing. Furthermore, providing financial support for ecotourism and compensation for depredation to offset potential jaguar impacts should be considered for jaguar conservation in Sonora. Felid material from the middle Early Pleistocene Haro River quarry, northern Pakistan, is described. Most of the material is referred to the Eurasian jaguar, Panthera gombaszogensis georgica, representing the first record of the jaguar in southern Asia. A systematic review of dental morphology of all living species of Pantherinae is made to provide a better background for studying the evolution of pantherine cats. A renewed dispersal scenario of the jaguar in Eurasia is provided. Panthera gombaszogensis georgica is suggested to be a distinct subspecies distributed in central, western and southern Asia during the Early Pleistocene, characterized by relatively small body size and slender dentition. The Eurasian jaguar does not possess the derived dental characters of the living jaguar Panthera onca (e.g. weak or no canine vertical groove, robust premolars), and presumably did not specialize on hard‐shelled prey, and therefore should be viewed as a distinct species from the living P. onca. Two additional felids from the Haro River Quarry, the large sabre‐toothed felid? Megantereon sp. and a small undetermined feline, are also reported. The Felidae faunal composition of southern Asia during the Early Pleistocene is generally similar to that of northern Eurasia, and the paleoenvironment is probably slightly more closed. Mammals play a central role in the cycle of several zoonoses, the study of their prevalence and distribution is extremely important to prevent outbreaks and create prophylaxis mechanisms. Wild feline species have increasingly in contact with humans, exposing them to possible overflow or sharing of various pathogens and participating in the cycle of numerous zoonoses. Our objective was to investigate the participation of Brazilian feline species in zoonotic cycles, through secondary data analyses. We found records of 19 zoonoses for 10 feline species, including two caused by viruses, five by nematodes, four by protozoa, and eight by bacteria. The zoonoses with highest prevalence caused by viruses was rabies, protozoa was toxoplasmosis, bacteria were brucellosis and leptospirosis, while by nematodes were hookworm and toxocariasis. The species Leopardus pardalis, Leopardus tigrinus, Panthera onca, and Puma concolor presented highest number of records of pathogens and wider distribution of records through the Brazilian regions. Records of these pathogens occurred mainly in the Central and Southeastern Brazil. The role of the members of the family Felidae as reservoirs of agents of several lethal zoonoses is observed, although leptospirosis presents a register of different serovars, including some non-pathogenic to the humans. These results bring to light the importance of preserving and maintaining the natural habitats of these species as a public health measure, in order to prevent the proliferation of these zoonoses, both for humans and animals. The preservation of feline natural environments could minimize possible pathogen exchanges among these species and domestic animals and, consequently, with humans, as well as reducing the likelihood of their direct contact with felines. Human-wildlife conflict (HWC) is a pressing issue worldwide but varies by species over time and place. One of the most prevalent forms of HWC in the mid-hills of Nepal is human-common-leopard conflict (HLC). Leopard attacks, especially in forested areas, can severely impact villagers and their livestock. Information on HLC in the Gorkha district was scarce, thus making it an ideal location to identify high-risk zones and landscape variables associated with such events. Registered cases were collected and reviewed from the Division Forest Office (DFO) during 2019-2021. Claims from DFO records were confirmed with herders and villagers via eight focus group discussions. To enhance modeling success, researchers identified a total of 163 leopard attack locations on livestock, ensuring a minimum distance of at least 100 meters between locations. Using maximum entropy (MaxEnt) and considering 13 environmental variables, we mapped common leopard attack risk zones. True Skill Statistics (TSS) and area under receiver-operator curve (AUC) were used to evaluate and validate the Output. Furthermore, 10 replications, 1,000 maximum iterations, and 1000 background points were employed during modeling. The average AUC value for the model, which was 0.726 ± 0.021, revealed good accuracy. The model performed well, as indicated by a TSS value of 0.61 ± 0.03. Of the total research area (27.92 km 2 ), about 74% was designated as a low-risk area, 19% as a medium-risk area, and 7% as a high-risk area. Of the 13 environmental variables, distance to water (25.2%) was the most significant predictor of risk, followed by distance to road (16.2%) and elevation (10.7%). According to response curves, the risk of common leopard is highest in the areas between 1.5 to 2 km distances from the water sources, followed by the closest distance from a road and an elevation of 700 to 800 m. Results suggest that managers and local governments should employ intervention strategies immediately to safeguard rural livelihoods in high-risk areas. Improvements include better design of livestock corrals, insurance, and total compensation of livestock losses. Settlements near roads and water sources should improve the design and construction of pens and cages to prevent livestock loss. More studies on the characteristics of victims are suggested to enhance understanding of common leopard attacks, in addition to landscape variables. Unsustainable trade in big cats affects all species in the genus, Panthera, and is one of the foremost threats to their conservation. To provide further insight into the impact of policy interventions intended to address this issue, we examine the case study of the Republic of Korea (South Korea), which in the early 1990s was one of the world's largest importers of tiger (Panthera tigris) bone and a major manufacturer of tiger-derived medicinal products. In 1993, South Korea became a Party to the Convention on International Trade in Endangered Species (CITES) and introduced a ban on commercial trade in CITES Appendix I-listed big cats a year later. We used an expert-based questionnaire survey and an exploration of the CITES trade database to investigate what has since happened to big cat trade in South Korea. Expert opinion suggested that big cat trade has likely substantially reduced since the early 1990s, as a result of the trade ban and broad socioeconomic changes. However, illegal trade has not been eradicated entirely and we were able to confirm that products reportedly derived from big cats were still publicly available for sale on a range of Korean online marketplaces, sometimes openly. The items most commonly reported by respondents from post-1994 trade and supported by expert-led evidence were tiger and leopard (Panthera pardus) skins and tiger bone wine. Although South Korea may provide a useful case study of a historically significant consumer country for tiger which has made strong progress in addressing unsustainable levels of big cat trade within a short period of time, there remains a need to address recalcitrant small-scale, illegal trade. Protected areas are traditionally the foundation of conservation strategy, but land not formally protected is of particular importance for the conservation of large carnivores because of their typically wide-ranging nature. In South Africa, leopard (Panthera pardus) population decreases are thought to be occurring in areas of human development and intense negative interactions, but research is biased towards protected areas, with quantitative information on population sizes and trends in non-protected areas severely lacking. Using Spatially Explicit Capture-Recapture and occupancy techniques including 10 environmental and anthropogenic covariates, we analysed camera trap data from commercial farmland in South Africa where negative human-wildlife interactions are reported to be high. Our findings demonstrate that leopards persist at a moderate density (2.21 /100 km 2 ) and exhibit signs of avoidance from areas where lethal control measures are implemented. This suggests leopards have the potential to navigate mixed mosaic landscapes effectively, enhancing their chances of long-term survival and coexistence with humans. Mixed mosaics of agriculture that include crops, game and livestock farming should be encouraged and, providing lethal control is not ubiquitous in the landscape, chains of safer spaces should permit vital landscape connectivity. Hair is a commonly encountered trace evidence in wildlife crimes involving mammals and can be used for species identification which is essential for subsequent judicial proceedings. This proof of concept study aims, to distinguish the black guard hair of three wild cat species belonging to the genus Panthera i.e. Royal Bengal Tiger (Panthera tigris tigris), Indian Leopard (Panthera pardus fusca), and Snow Leopard (Panthera uncia) using a rapid and non-destructive ATR-FTIR spectroscopic technique in combination with chemometrics. A training dataset including 72 black guard hair samples of three species (24 samples from each species) was used to construct chemometric models. A PLS2-DA model successfully classified these three species into distinct classes with R-Square values of 0.9985 (calibration) and 0.8989 (validation). VIP score was also computed, and a new PLS2DA-V model was constructed using variables with a VIP score ≥ 1. External validation was performed using a validation dataset including 18 black guard hair samples (6 samples per species) to validate the constructed PLS2-DA model. It was observed that PLS2-DA model provides greater accuracy and precision compared to the PLS2DA-V model during cross-validation and external validation. The developed PLS2-DA model was also successful in differentiating human and non-human hair with R-Square values of 0.99 and 0.91 for calibration and validation, respectively. Apart from this, a blind test was also carried out using 10 unknown hair samples which were correctly classified into their respective classes providing 100 % accuracy. The Bengal cat breed was developed from intercrosses between the Asian leopard cat, Prionailurus bengalensis, and the domestic cat, Felis catus, with a last common ancestor approximately 6 million years ago. Predicted to derive ∼94% of their genome from domestic cats, regions of the leopard cat genome are thought to account for the unique pelage traits and ornate color patterns of the Bengal breed, which are similar to those of ocelots and jaguars. We explore ancestry distribution and selection signatures in the Bengal breed by using reduced representation and whole-genome sequencing from 947 cats. The mean proportion of leopard cat DNA in the Bengal breed is 3.48%, lower than predicted from breed history, and is broadly distributed, covering 93% of the Bengal genome. Overall, leopard cat introgressions do not show strong signatures of selection across the Bengal breed. However, two popular color traits in Bengal cats, charcoal and pheomelanin intensity, are explained by selection of leopard cat genes whose expression is reduced in a domestic cat background, consistent with genetic incompatibility resulting from hybridization. We characterize several selective sweeps in the Bengal genome that harbor candidate genes for pelage and color pattern and that are associated with domestic, rather than leopard, cat haplotypes. The jaguar (Panthera onca) is a charismatic species considered Vulnerable in Colombia but yet largely unknown in the country. The species is mostly threatened by the continuous decline in its habitats, mostly derived from deforestation and habitat loss, additional to hunting and conflicts with humans. Thus, the future of jaguars in Colombia depends on protecting and recovering existing habitats. The aims of this study were to 1) evaluate jaguar distribution and identify the remnant patches of habitat in Colombia, 2) define an ecological connectivity network within the country, and 3) propose a priority areas portfolio for the conservation and recovery of jaguars. We used a presence background model for estimating species potential distribution and subsequently identified remaining habitat patches across the country based on land cover and species-specific ecological attributes. We then created an ecological connectivity network based on circuit theory and following a multi-criteria approach identified jaguar priority areas for conservation (JPCA) and recovery (JPRA). Jaguar potential distribution comprises 1103122.43 km2, from which 56.71% maintain suitable patches of potential habitat. We identified 960 corridors between remnant patches of natural or semi-natural vegetation. Based on the criteria, JPCAs with greater importance were identified in each of the five Colombian regions. JPRAs were located mainly towards the Andean and Caribbean regions. Leopard attacks on human beings is common in Uttarakhand which is an area of national conservation. The objective of this study is to examine the pattern of injuries and circumstances to encourage ways of reducing such conflicts. There were eight deaths from leopard attacks brought for autopsy to the mortuary between January 2019 and August 2020. Incidents occurred mostly in a forest close to human settlements. One tourist was among the male victims, the remainder of whom were women carrying out household duties near the forest. Leopard attack injuries present in the head and neck were ante-mortem, with abdomen, buttocks and limbs post-mortem.
