Summary: Cancer cachexia is a prevalent and often fatal wasting condition that cannot be fully reversed with nutritional interventions. Muscle atrophy is a central component of the syndrome, but the mechanisms whereby cancer leads to skeletal muscle atrophy are not well understood. We performed single-nucleus multi-omics on skeletal muscles from a mouse model of cancer cachexia and profiled the molecular changes in cachexic muscle. Our results revealed the activation of a denervation-dependent gene program that upregulates the transcription factor myogenin. Further studies showed that a myogenin-myostatin pathway promotes muscle atrophy in response to cancer cachexia. Short hairpin RNA inhibition of myogenin or inhibition of myostatin through overexpression of its endogenous inhibitor follistatin prevented cancer cachexia-induced muscle atrophy in mice. Our findings uncover a molecular basis of muscle atrophy associated with cancer cachexia and highlight potential therapeutic targets for this disorder.

Abstract Background Lenvatinib is a multitargeted tyrosine kinase inhibitor that is being tested in combination with immune checkpoint inhibitors to treat advanced gastric cancer; however, little data exists regarding the efficacy of lenvatinib monotherapy. Patient-derived xenografts (PDX) are established by engrafting human tumors into immunodeficient mice. The generation of PDXs may be hampered by growth of lymphomas. In this study, we compared the use of mice with different degrees of immunodeficiency to establish PDXs from a diverse cohort of Western gastric cancer patients. We then tested the efficacy of lenvatinib in this system. Methods PDXs were established by implanting gastric cancer tissue into NOD.Cg-Prkdc scid Il2rg tm1Wjl /SzJ (NSG) or Foxn1 nu (nude) mice. Tumors from multiple passages from each PDX line were compared histologically and transcriptomically. PDX-bearing mice were randomized to receive the drug delivery vehicle or lenvatinib. After 21 days, the percent tumor volume change (%Δvtumor) was calculated. Results 23 PDX models were established from Black, non-Hispanic White, Hispanic, and Asian gastric cancer patients. The engraftment rate was 17% (23/139). Tumors implanted into NSG (16%; 18/115) and nude (21%; 5/24) mice had a similar engraftment rate. El objetivo de este trabajo es describir dos eventos producidos en la provincia de Buenos Aires en los cuales Neospora caninum estuvo asociado a la ocurrencia de abortos en bovinos de cría para carne. En uno de ellos se registraron 11 abortos en 57 vaquillonas durante 45 días, en este evento fue 5 veces más probable que una vaquillona que sufrió un aborto fuera seropositiva a N. caninum que una que no lo sufrió (odds ratio [OR] = 4,9 IC 1,2-19,9) (p < 0,05). En el otro evento se registraron 14 abortos en 140 vacas, y no se observó asociación significativa entre los abortos y la seropositividad frente a N. caninum OR = 0,69 (0,06-7,31) (p > 0,05). Se analizaron dos fetos de cada evento: estos resultaron negativos a otros patógenos de la reproducción, aunque presentaron anticuerpos específicos y lesiones histopatológicas compatibles con infecciones por N. caninum. Estos resultados sugieren dos posibles modalidades de presentación de abortos en bovinos causados por N. caninum: una epidémica, como la del primer evento aquí referido, y una endémica, como la del segundo.

Easter Island, Rapa Nui or Te pito o te henua, is in the middle of the Pacific Ocean halfway between South America and Oceania, constituting one of the most isolated places on the planet. It was colonized by Polynesians at the end of the first millennium of the Christian era, thus becoming one of the extremes of the Polynesian triangle. The island is of volcanic origin, has a small surface area (166 km²) and limited resources, and gave rise to a culture that is unique in the world, recognized internationally for its numerous megalithic constructions and large moai. Just as it was discovered and colonized by sea, the development and sustainability of the island is closely related to the ocean that surrounds it. The objective of this article is to describe the sailing and fishing techniques used by the Easter Islanders, or rapanuis, and their use of marine organisms prior to contact with Europeans, demonstrating their inventiveness and adaptation to the specific characteristics of this small territory.

The Chilean jagged lobster (Projasus bahamondei) is a deep-water crustacean (175-550 m) occurring in certain areas of the southeastern Pacific Ocean, including the Nazca Ridge, Desventuradas Islands, the Juan Fernandez archipelago and ridge, and the continental slope off the central coast of Chile. This review describes the taxonomic status, geographical and bathymetric distribution, some biological aspects and habitat characteristics of this species. Additionally, both artisanal and industrial exploitation attempts made within the region are detailed, as well as fishing operation results, chemical composition, different elaboration procedures and the destination of the catch. The main objectives of this review are to contribute to the knowledge of P. bahamondei as a component of the deep-sea ecosystem and to highlight its importance as a potential fishery resource.

Commercial fisheries are expanding their activities into deeper water. The life history features of these deep-water resources make them more vulnerable to exploitation than most shallow-water resources. Moreover, the apparent lack of solid information about the ecology of most deep-water species represents a major limitation for the development and implementation of management strategies. This scenario has caused great concern regarding the sustainability of these resources and the possible environmental impacts on the deep-sea ecosystem. In Latin America, commercial fisheries are going deep as well, and considering the above-mentioned concerns, we felt the need to compile the available information about the deep-water shrimp resources and the current status of their fisheries in Latin America. Focusing on Mexico, Central America, Peru, Chile and Brazil, this review describes the exploited species, and, whenever available, the fishing fleet, fishery statistics, and management strategies. A total of 17 species (10 spp. of Penaeoidea; 7 spp. of Caridea) are of commercial interest in Latin America, but deep-water shrimps are currently fished only in Costa Rica, Colombia and Chile. An implemented management plan exists in Chile and Colombia, while Brazil approved fishery regulations for the aristeid fishery, which were never implemented. Considering the lack of information about the biology of the deep-water shrimps, which hinders the development of adequate management strategies, we see the urgent need to improve the communication and collaboration between the different stakeholders in Latin American. We suggest the establishment of a searchable and constantly updated database, which may serve as a valuable source of information for researcher and decision makers. Finally, we propose the development of regional research plans aimed towards supporting measurements for a sustainable use of deep-water shrimps in Latin America.La pesca comercial está expandiendo sus actividades hacia aguas profundas. Las características del ciclo de vida de estos recursos de aguas profundas los hacen más vulnerables a la explotación que la mayoría de los recursos de aguas someras. Además, la falta de información sobre la ecología de la mayoría de las especies de aguas profundas constituye una limitación importante para el desarrollo e implementación de estrategias de manejo. Este escenario ha causado preocupación sobre la sustentabilidad de estos recursos y de los posibles impactos ambientales en los ecosistemas de aguas profundas. La pesca comercial en América Latina se extiende también hacia aguas profundas y, considerando las preocupaciones anteriormente mencionadas, se requiere la necesidad de compilar la información disponible sobre los recursos de camarones de aguas profundas y la situación actual de estas pesquerías en América Latina. Esta revisión se enfoca en México, Centro América, Perú, Chile y Brasil y describe las especies explotadas, la flota pesquera (siempre cuando sea disponible), las estadísticas pesqueras y las estrategias de manejo. Un total de 17 especies (10 spp. de Penaeoidea y 7 spp. de Caridea) son de interés comercial en América Latina; sin embargo, camarones de aguas profundas solo se pescan actualmente en Costa Rica, Colombia y Chile. Un plan de manejo implementado existe en Chile y Colombia, mientras que en Brasil se aprobaron regulaciones sobre la pesca de camarones de la familia Aristeidae, pero éstas nunca han sido implementadas. Considerando la falta de información sobre la biología de los camarones de aguas profundas, lo que dificulta el desarrollo de estrategias adecuadas de manejo, se requiere urgentemente mejorar la comunicación y colaboración entre los diferentes actores en Latinoamérica. Se sugiere establecer un banco de datos abierto para búsquedas y actualizarlo constantemente, lo que servirá como fuente valiosa de información para investigadores y quienes deben tomar decisiones. Finalmente, se propone desarrollar un plan de investigación para apoyar medidas hacia el uso sustentable de camarones de aguas profundas en América Latina.

In order to determine the biological parameters of the burrowing crayfish (Parastacus pugnax), we fenced in 900 m² of a humid, low-lying sector in Tiuquilemu, Bío-Bío Region, Chile. Monthly samples were taken from August 2007 to August 2008, and 3,512 specimens were caught. Records were made of their carapace length (CL), total weight (TW), sex, and number of eggs (for females). The monthly size-structures showed few juveniles between 20 and 30 mm CL in nearly all months. P. pugnax was found to carry eggs from mid-spring through late autumn; size at first maturity for females was 38.1 mm CL; and fecundity varied between three and 45 eggs per female. In terms of the global sexual proportion, males were predominant in 11 of the 12 months, although the ratio was as expected (1:1). We estimated the parameters of the growth curve using modal progression analysis, obtaining CL∞, K, and t0 values of 55.3 mm, 0.23 mm year-1, and -0.58 years, respectively. The length-weight relationship was calculated separately for males and females, and no significant differences were found between sexes. The common parameters defining the potential function of a = 0.00052 and b = 2.98 (P < 0.05) indicated isometric increments in weight.Para determinar parámetros biológicos del camarón excavador (Parastacus pugnax), se cercó un área de 900 m² de un sector de vega en Tiuquilemu, Región del Bío-Bío Chile. En dicho lugar se efectuaron muestreos mensuales entre agosto de 2007 y agosto de 2008. A un total de 3.512 especímenes se les registró la longitud cefalotorácica (CL), peso total (TW), sexo y en hembras se determinó el número total de huevos. Se construyeron las estructuras de longitudes mensuales con escasa representación de juveniles entre 20 y 30 mm de CL, en casi la mayoría de los meses. Se determinó que el período de incubación en P. pugnax se extiende entre mediados de primavera y finales de otoño, la longitud de primera madurez en las hembras se calculó en 38,1 mm de CL y la fecundidad varió entre tres y 45 huevos por hembra. En cuanto a la proporción sexual global, hubo predominio de machos en 11 de los 12 meses; no obstante, estas no difieren de la razón esperada 1:1. Se estimaron los parámetros de la curva de crecimiento mediante el método de análisis de progresión modal, obteniéndose valores de CL∞, K y t0, correspondientes a 55,3 mm, 0,23 mm-año-1 y -0,58 años, respectivamente. La relación longitud-peso se calculó en machos y hembras separadamente, no existiendo diferencias significativas entre sexos, con parámetros comunes que definen la función potencial de a = 0,00052 y b = 2,98 (P < 0,05), estableciéndose que el crecimiento en peso es de tipo isométrico.

La información obtenida en cruceros de evaluación directa de langostino amarillo (Cervimunida johní) realizados en la zona central del Chile (32°-38°S) entre 1997 y 2009, permitió modelar los cambios en la densidad anual de este recurso mediante Modelo Lineal Generalizado (MLG). El objetivo de este enfoque fue tanto minimizar el efecto de los distintos criterios y diseños de muestreo que han sido empleados en el tiempo, como también determinar los efectos que explican las variaciones en la densidad de este recurso. Lo anterior permitió estimar el valor esperado de la densidad anual para el cálculo de biomasa mediante el método de "área barrida". Los registros lance a lance de captura por unidad de área barrida (CPUA) permitieron la identificación y medición de los focos de abundancia, además de caracterizar la distribución y área que ha ocupado el recurso en el tiempo. El MLG identificó como efectos significativos el año, la zona, la profundidad, y las interacciones anuales de primer orden, explicando con ello el 43% de la devianza residual del modelo. Los principales efectos que explican las variaciones de la CPUA son el año y la interacción año-zona. Se determinó que la zona con mayor abundancia promedio corresponde a la delimitada entre 32° y 34°S, de igual manera que en el rango de profundidad menor a 250 m. Dentro del período analizado, se determinó que la población de este recurso incrementó en biomasa de manera sostenida hasta el 2006, principalmente en la zona norte del área de estudio, seguida de una fuerte reducción que al 2009 equivale al 75% respecto del máximo, reflejada en una biomasa de 11.000 ton.Changes in the annual density of the yellow squat lobster (Cervimunida johni) were modeled using a generalized linear model (GLM), using information obtained during direct stock assessment cruises carried out in central Chile (32°-38°S) between 1997 and 2009. The purpose of this approach was to minimize the effect of different sampling criteria and designs that have been used over time and to determine the factors explain in variations in the density of this resource. The above approach allowed estimating the expected value of the annual density for the biomass calculation through the "swept area" method. By using the haul-to-haul records of catch per unit of the swept area (CPUA), it was possible to identify and measure the points of abundance and characterize the distribution and area occupied by the resource over time. The significant effects identified by the GLM were the year, zone, depth, and first-order annual interactions; which explained 43% of the residual deviance in the model. The main effects that explain the CPUA variations are the year and the year-zone interaction. The zone with the greatest average abundance was located between 32° and 34°S; the same was for the depth range

Para determinar el crecimiento en el camarón excavador (Parastacus pugnax) en la zona centro sur de Chile se utilizó un marbete tipo cinturón. Los parámetros longitud cefalotorácica asintótica (Lc m) y la velocidad de incremento en longitud y peso (K), se establecieron mediante el método de Gulland & Holt (1959). El parámetro t0 se determinó mediante la ecuación inversa del modelo de von Bertalanffy, estableciéndose que las curvas de crecimiento en longitud y peso fueron definidas por los parámetros K = 0,35 mm año-1, t0 = -0,38 años, Lc∞ = 55, 9 mm y W∞ = 83,8 g, valores similares a los de Samastacus spinifrons, especie chilena de alto potencial de cultivo, y de otros parastácidos sudamericanos, tales como P. brasiliensis y P. deffosus.A modified tag belt type was used in the burrowing crayfish (Parastacus pugnax), in the central-south of Chile. The parameters asymptotic carapace length (Lc0) and rate of increase (K) were determined through the Gulland & Holt (1959) method. The parameter t0 was determined by the inverse equation of von Bertalanffy model, which allowed to establish that the growth curves in length and weight are defined by the parameters K = 0.35 mm yr-1, t0 = -0.38 years, Lc∞ = 55. 9 mm and W∞= 83.8 g. These values were similar to those of Samastacus spinifrons, Chilean species with high potential for aquaculture, and similar to those of other South American parastacids such as P. brasiliensis and P. deffosus.

Se analiza la información mensual de composición de tamaño recopilada en el monitoreo de la pesca artesanal sobre cangrejo dorado (Chaceon chilensis) realizado entre julio de 2005 y mayo de 2006, para evaluar los parámetros de crecimiento, mortalidad natural y puntos biológicos de referencia de los machos sobre los cuales se basa esta pesquería. Se estableció una longevidad promedio de 20 años y una mortalidad natural en torno a M = 0,27 año-1 . La talla crítica se determinó a los 110 mm de longitud cefalotorácica (Lc), que es levemente inferior a la talla de primera captura de 114 mm de Lc. De acuerdo al análisis efectuado, se explotan individuos entre 4 y 10 años de vida. Mediante análisis de equilibrio se determina que la población se encuentra en 82% de la condición virginal, que se refleja en una talla promedio en las capturas de 128 mm de Lc. Una eventual reducción de la población a un límite del 40% de la condición original, se consigue aumentando en tres veces el nivel actual de desembarques, lo que se traduciría en una talla media en las capturas de 118 mm de Lc. Finalmente, se recomiendan distintos puntos biológicos de referencia para garantizar una explotación sustentable en el tiempo.Monthly information on the size composition of golden crab (Chaceon chilensis) catches compiled while monitoring the artisanal fishery (July 2005 through May 2006) is analyzed in order to evalúate the growth parameters, natural mortality, and biological reference points in male specimens, the basis of the fishery. Average longevity was found to be 20 years and natural mortality around M = 0.27. The critical size was determined to be around 110 mm carapace length (Lc), slightly lower than that of the first catch (114 mm Lc). According to this analysis, individuals between 4 and 10 years of age are exploited. A balance analysis revealed that 82%) of the population is virginal, as reflected in an average size-at-catch of about 128 mm Lc. Tripling the current level of landings would lead to an eventual reduction to a limit of 40%> of the original population; this would result in an average catch size of 118 mm Lc. Finally, several biological reference points are recommended for guaranteeing sustainable exploitation over time.
