In amniotes, the predominant developmental strategy underlying body size evolution is thought to be adjustments to the rate of growth rather than its duration. However, most theoretical and experimental studies supporting this axiom focus on pairwise comparisons and/or lack an explicit phylogenetic framework. We present the first large-scale phylogenetic comparative analysis examining developmental strategies underlying the evolution of body size, focusing on non-avialan theropod dinosaurs. We reconstruct ancestral states of growth rate and body mass in a taxonomically rich dataset, finding that contrary to expectations, changes in the rate and duration of growth played nearly equal roles in the evolution of the vast body size disparity present in non-avialan theropods—and perhaps that of amniotes in general.

A second K/Pg boundary interval in the northern sector of the Antarctic Peninsula on Vega Island has been proposed, yet current temporal resolution of these strata prohibits direct testing of this hypothesis. To not only test for the existence of a K/Pg boundary on Vega Island but also provide increased age resolution for the associated vertebrate fauna (e.g., marine reptiles, non-avian dinosaurs, and avian dinosaurs), the Vega Island succession was intensively re-sampled. Stratigraphic investigation of the Cape Lamb Member of the Snow Hill Island Formation, and in particular, the overlying Sandwich Bluff Member of the López de Bertodano Formation, was conducted using biostratigraphy, strontium isotope stratigraphy, magnetostratigraphy, and detrital zircon geochronology. These data indicate a Late Campanian–early Maastrichtian age for the Cape Lamb Member and present three possible correlations to the global polarity time scale (GPTS) for the overlying Sandwich Bluff Member. The most plausible correlation, which is consistent with biostratigraphy, detrital zircon geochronology, sequence stratigraphy, and all but one of the Sr-isotope ages, correlates the base of the section to C31N and the top of the section with C29N, which indicates that the K/Pg boundary passes through the top of the unit. A second, less plausible option conflicts with the biostratigraphy and depends on a series of poorly defined magnetic reversals in the upper part of the stratigraphy that also correlates the section between C31N and C29R and again indicates an inclusive K/Pg boundary interval. The least likely correlation, which depends on favoring only a single Srisotope age at the top of the section over biostratigraphy, correlates the section between C31N and C30N and is inconsistent with an included K/Pg boundary interval. Although our preferred correlation is well supported, we failed to identify an Ir-anomaly, spherules/impact ejecta, or other direct evidence typically used to define the precise position of a K/Pg boundary on Vega Island. This study does, however, confirm that Vegavis, from the base of the Sandwich Bluff Member, is the oldest (69.2–68.4 Ma) phylogenetically placed representative of the avian crown clade, and that marine vertebrates and non-avian dinosaurs persisted in Antarctica up to the terminal Cretaceous.

Hyaluronan (HA) is a glycosaminoglycan polymer that often accumulates in malignancy. Megadalton complexes of HA with proteoglycans create a hydrated connective tissue matrix, which may play an important role in tumor stroma formation. Through its colloid osmotic effects, HA complexes contribute to tumor interstitial fluid pressure, limiting the effect of therapeutic molecules on malignant cells. The therapeutic potential of enzymatic remodeling of the tumor microenvironment through HA depletion was initially investigated using a recombinant human HA-degrading enzyme, rHuPH20, which removed HA-dependent tumor cell extracellular matrices in vitro. However, rHuPH20 showed a short serum half-life (t1/2 < 3 minutes), making depletion of tumor HA in vivo impractical. A pegylated variant of rHuPH20, PEGPH20, was therefore evaluated. Pegylation improved serum half-life (t1/2 = 10.3 hours), making it feasible to probe the effects of sustained HA depletion on tumor physiology. In high-HA prostate PC3 tumors, i.v. administration of PEGPH20 depleted tumor HA, decreased tumor interstitial fluid pressure by 84%, decreased water content by 7%, decompressed tumor vessels, and increased tumor vascular area >3-fold. Following repeat PEGPH20 administration, tumor growth was significantly inhibited (tumor growth inhibition, 70%). Furthermore, PEGPH20 enhanced both docetaxel and liposomal doxorubicin activity in PC3 tumors (P < 0.05) but did not significantly improve the activity of docetaxel in low-HA prostate DU145 tumors. The ability of PEGPH20 to enhance chemotherapy efficacy is likely due to increased drug perfusion combined with other tumor structural changes. Lungfish (Dipnoi) date back to the Devonian, and some fossil taxa as well as extant African lungfishes are known for their ability to aestivate, tolerating low-oxygen environments associated with seasonal drying. Extant lungfishes are separated into two families: Lepidosirenidae (Protopterus in Africa and Lepidosiren in South America) and Neoceratodontidae (Neocerotadus in Australia). African lungfishes were more geographically and phylogenetically diverse on the continent in the past than they are today, with only 5% of extinct taxa recorded from the sub-Saharan fossil record. Given the sparse record of Lepidosirenidae fossils from continental Africa, any new materials are important for understanding diversification of the clade. Here we describe new lungfish fossils cautiously referable to Protopterus annectens and Protopterus aethiopicus, which are strongly supported sister taxa based on the molecular phylogeny. Specimens were collected from the late Oligocene Nsungwe Formation in the Rukwa Rift Basin (RRB) of southwestern Tanzania. The late Oligocene Nsungwe Formation represents a sequence of continental rift-fill deposits of the Songwe sub-basin of the RRB and is subdivided into the lower Utengule and upper Songwe members. Recovery of such material from the Paleogene of Africa below the equator addresses a sizable gap in the lungfish fossil record. It also expands the Nsungwe Formation fauna that includes invertebrates, alestid fishes, ptychadenid anurans, snakes, and several clades of mammals, deepening paleoecological insights into the late Oligocene record of the continental African interior. At present, P. aethiopicus and P. dolloi have an extensive modern eastern African distribution associated with the rift lakes and a region where extant members of P. annectens are not presently known. Fossil specimens described herein document presence of the clade during Paleogene volcanic activity in the western branch of the Eastern African Rift System.

Extant snake faunas have their origins in the mid-Cenozoic, when colubroids replaced booid-grade snakes as the dominant species. The timing of this faunal changeover in North America and Europe based on fossils is thought to have occurred in the early Neogene, after a period of global cooling opened environments and made them suitable for more active predators. However, new fossils from the late Oligocene of Tanzania have revealed an early colubroid-dominated fauna in Africa suggesting a different pattern of faunal turnover there. Additionally, molecular divergence times suggest colubroid diversification began sometime in the Paleogene, although the exact timing and driving forces behind the diversification are not clear. Here we present the first fossil snake referred to the African clade Lamprophiinae, and the oldest fossil known of Lamprophiidae. As such, this specimen provides the only potential fossil calibration point for the African snake radiation represented by Lamprophiidae, and is the oldest snake referred to Elapoidea. A molecular clock analysis using this and other previously reported fossils as calibration points reveals colubroid diversification minimally occurred in the earliest Paleogene, although a Cretaceous origin cannot be excluded. The elapoid and colubrid lineages diverged during the period of global warming near the Paleocene-Eocene boundary, with both clades diversifying beginning in the early Eocene (proximate to the Early Eocene Climate Optimum) and continuing into the cooler Miocene. The majority of subclades diverge well before the appearance of colubroid dominance in the fossil record. These results suggest an earlier diversification of colubroids than generally previously thought, with hypothesized origins of these clades in Asia and Africa where the fossil record is relatively poorly known. Further work in these regions may provide new insights into the timing of, and environmental influences contributing to, the rise of colubroid snakes.

Postcranial skeletal pneumaticity, air-filled bones of the trunk and limbs, is exclusive to birds among extant tetrapods and exhibits significant variation in its expression among different species. Such variation is not random but exhibits relationships with both body mass and locomotor specializations. Most species-level comparative research to date has focused on aquatic-oriented taxa (e.g., Anseriformes). The lack of data from non-aquatic birds constrains our ability to characterize global (i.e., avian-wide) patterns of this trait complex. To address this gap, the study conducted herein quantified postcranial pneumaticity in Accipitrimorphae, a mostly terrestrial clade composed of species that span a range of body sizes and exhibit diverse flight/foraging behaviors. All examined species (n = 88) invariably pneumatized the postaxial through pre-caudal vertebrae, sternum, coracoid, humerus, vertebral and sternal ribs, and pelvic girdle, a pattern herein referred to as the accipitrimorph baseline. Of the 88 sampled species, 41 expanded upon this pattern, whereas 10 species exhibited a reduction. No species deviated from the accipitrimorph baseline by more than two anatomical regions. A phylogenetically-informed regression analysis failed to identify a significant relationship between body mass and pneumaticity. However, specific pneumaticity phenotypes deviating from the baseline were correlated with aspects of wing morphology, tail length, and home range size. Results from this and previous studies provide clarity on two hypotheses: (1) aquatic taxa display distinct pneumaticity expression patterns relative to non-aquatic birds, notably with reductions in the proportion of the skeleton filled with air in diving specialists and (2) contemporary comparative studies, including the one herein, that explicitly account for phylogenetic relationships consistently fail to support the oft-cited positive relationship between pneumaticity and body mass. Instead, historical relationships and functional/ecological attributes (e.g., diving, specialized flight behaviors) appear to be the primary drivers underlying patterns of variation in this trait complex.

Infections that stem from bacterial biofilms are difficult to eradicate. Within a biofilm state, bacteria are upwards of 1000-fold more resistant to conventional antibiotics, necessitating the development of alternative approaches to treat biofilm-based infections. One such approach is the development of small molecule adjuvants that can inhibit/disrupt bacterial biofilms. When such molecules are paired with conventional antibiotics, these dual treatments present a combination approach to eradicate biofilm-based infections. Previously, we have demonstrated that small molecules containing either a 2-amino pyrimidine (2-AP) or a 2-aminoimidazole (2-AI) heterocycle are potent anti-biofilm agents. Herein, we now report a scaffold hopping strategy to generate new aryl 2-AP analogs that inhibit biofilm formation by methicillin-resistant Staphylococcus aureus (MRSA). These molecules also suppress colistin resistance in colistin resistant Klebsiella pneumoniae, lowering the minimum inhibitory concentration (MIC) by 32-fold.

Mesozoic birds display considerable diversity in size, flight adaptations and feather organization1–4, but exhibit relatively conserved patterns of beak shape and development5–7. Although Neornithine (that is, crown group) birds also exhibit constraint on facial development8,9, they have comparatively diverse beak morphologies associated with a range of feeding and behavioural ecologies, in contrast to Mesozoic birds. Here we describe a crow-sized stem bird, Falcatakely forsterae gen. et sp. nov., from the Late Cretaceous epoch of Madagascar that possesses a long and deep rostrum, an expression of beak morphology that was previously unknown among Mesozoic birds and is superficially similar to that of a variety of crown-group birds (for example, toucans). The rostrum of Falcatakely is composed of an expansive edentulous maxilla and a small tooth-bearing premaxilla. Morphometric analyses of individual bony elements and three-dimensional rostrum shape reveal the development of a neornithine-like facial anatomy despite the retention of a maxilla–premaxilla organization that is similar to that of nonavialan theropods. The patterning and increased height of the rostrum in Falcatakely reveals a degree of developmental lability and increased morphological disparity that was previously unknown in early branching avialans. Expression of this phenotype (and presumed ecology) in a stem bird underscores that consolidation to the neornithine-like, premaxilla-dominated rostrum was not an evolutionary prerequisite for beak enlargement. A crow-sized stem bird, Falcatakely forsterae, possesses a long and deep rostrum—a beak morphology that was previously unknown among Mesozoic birds and is similar to that of some crown-group birds, such as toucans.

The ability to accurately and reliably estimate body mass of extinct taxa is a vital tool for interpreting the physiology and even behavior of long-dead animals. For this reason, paleontologists have developed many possible methods of estimating the body mass of extinct animals, with varying degrees of success. These methods can be divided into two main categories: volumetric mass estimation and extant scaling methods. Each has advantages and disadvantages, which is why, when possible, it is best to perform both, and compare the results to determine what is most plausible within reason. Here we employ volumetric mass estimation (VME) to calculate an approximate body mass for previously described specimens of Anzu wyliei from the Carnegie Museum of Natural History. We also use extant scaling methods to try to obtain a reliable mass estimate for this taxon. In addition, we present the first digital life restoration and convex hull of the dinosaur Anzu wyliei used for mass estimation purposes. We found that the volumetric mass estimation using our digital model was 216-280kg, which falls within the range predicted by extant scaling techniques, while the mass estimate using minimum convex hulls was below the predicted range, between 159-199 kg . The VME method for Anzu wyliei strongly affirms the predictive utility of extant-based scaling. However, volumetric mass estimates are likely more precise because the models are based on comprehensive specimen anatomy rather than regressions of a phylogenetically comprehensive but disparate sample.

The Mesozoic plate tectonic and paleogeographic history of Gondwana had a profound effect on the distribution of terrestrial vertebrates. As the supercontinent fragmented into a series of large landmasses (South America, Africa-Arabia, Antarctica, Australia, New Zealand, the Indian subcontinent, and Madagascar), particularly during the Late Jurassic and Cretaceous, its terrestrial vertebrates became progressively isolated, evolving into unique faunal assemblages. We focus on four clades that, during the Mesozoic, had relatively low ability for dispersal across oceanic barriers—crocodyliforms, sauropod dinosaurs, nonavian theropod dinosaurs, and mammals. Their distributions reveal patterns that are critically important in evaluating various biogeographic hypotheses, several of which have been informed by recent discoveries from the Late Cretaceous of Madagascar. We also examine the effects of lingering, intermittent connections, or reconnections, of Gondwanan landmasses with Laurasia (through the Caribbean, Mediterranean, and Himalayan regions) on the distributions of different clades. ▪ This article reviews the biogeographic history of terrestrial vertebrates from the Mesozoic of the southern supercontinent Gondwana. ▪ Relatively large, terrestrial animals—including crocodyliforms, sauropod and nonavian theropod dinosaurs, and mammals—are the focus of this review. ▪ Most patterns related to vicariance occurred during the Late Jurassic and Cretaceous, the intervals of most active Gondwanan fragmentation. ▪ Recent discoveries of vertebrates from the Late Cretaceous of Madagascar have played a key role in formulating and testing various biogeographic hypotheses.

Complex structures, like the vertebrate skull, are composed of numerous elements or traits that must develop and evolve in a coordinated manner to achieve multiple functions. The strength of association among phenotypic traits (i.e., integration), and their organization into highly-correlated, semi-independent subunits termed modules, is a result of the pleiotropic and genetic correlations that generate traits. As such, patterns of integration and modularity are thought to be key factors constraining or facilitating the evolution of phenotypic disparity by influencing the patterns of variation upon which selection can act. It is often hypothesized that selection can reshape patterns of integration, parceling single structures into multiple modules or merging ancestrally semi-independent traits into a strongly correlated unit. However, evolutionary shifts in patterns of trait integration are seldom assessed in a unified quantitative framework. Here, we quantify patterns of evolutionary integration among regions of the archosaur skull to investigate whether patterns of cranial integration are conserved or variable across this diverse group. Using high-dimensional geometric morphometric data from 3D surface scans and computed tomography scans of modern birds (n = 352), fossil non-avian dinosaurs (n = 27), and modern and fossil mesoeucrocodylians (n = 38), we demonstrate that some aspects of cranial integration are conserved across these taxonomic groups, despite their major differences in cranial form, function, and development. All three groups are highly modular and consistently exhibit high integration within the occipital region. However, there are also substantial divergences in correlation patterns. Birds uniquely exhibit high correlation between the pterygoid and quadrate, components of the cranial kinesis apparatus, whereas the non-avian dinosaur quadrate is more closely associated with the jugal and quadratojugal. Mesoeucrocodylians exhibit a slightly more integrated facial skeleton overall than the other grades. Overall, patterns of trait integration are shown to be stable among archosaurs, which is surprising given the cranial diversity exhibited by the clade. At the same time, evolutionary innovations such as cranial kinesis that reorganize the structure and function of complex traits can result in modifications of trait correlations and modularity.

Postcranial skeletal pneumaticity (i.e., epithelial-lined, air-filled bones) is a condition unique to birds among extant tetrapods. Previous research reveals extensive variation in the expression of this trait in different bird species, from taxa that pneumatize nearly the entire skeleton to others that do not pneumatize a single bone. These studies, however, have primarily focused on aquatic/semi-aquatic birds, specifically Anseriformes (screamers, ducks, geese, swans) and Aequorlitornithes (loons, gulls, penguins, storks, etc.). This is the first clade-centric study of pneumaticity in an exclusively terrestrial clade (i.e., a group without any proclivities for water), Cuculidae. Given the variation in body size and ecology exhibited by cuckoos, they represent an ideal group for evaluating previously established trends in pneumaticity patterns. Similar to previous studies, our results indicate that cuckoos do exhibit extensive postcranial skeletal pneumaticity but with much more limited variation in expression. Of the surveyed species, 30 of 41 display an identical expression pattern, pneumatizing all postaxial vertebrae, the humerus, sternum, and pelvic girdle. The remaining species (11/41) deviate from this pattern by no more than two elements (i.e., the femur or the scapula/coracoid). All variable species expand upon the basic cuckoo pattern, with five species pneumatizing the femur and the remaining six taxa pneumatizing both the scapula and coracoid. Furthermore, most variation occurs in early diverging clades, with distinct subclades associated with specific anatomical expansions in pneumaticity (e.g., pneumatic femora in Neomorphinae and pneumatic scapulae/coracoids in select members of Couinae and Centropodinae). Limited variation noted in Cuculidae may be the result of the relatively high base level of pneumaticity when compared with previously sampled groups of water-oriented birds. Additional analyses indicate a positive relationship between body mass and pneumaticity, with possible (i.e., non-quantifiable) relationships noted between the limited expansions from the basic cuckoo pattern and specific locomotor behaviors (e.g., pneumatic femora present in species with enhanced cursorial behavior). These basic trends have also been observed in other densely sampled neognath clades. Taken together, the data presented herein supports the hypothesis that changes in pneumaticity expression may be correlated with shifts in biomechanical loading regimes rather than solely as a weight saving (i.e., density-altering) mechanism.

Non-marine vertebrates, including many crocodyliform clades, remain poorly documented from uppermost Cretaceous deposits of Africa. Recent exploratory fieldwork in the Upper Cretaceous (middle Campanian) Quseir Formation exposed around Dakhla Oasis (Western Desert of Egypt) has revealed new fossils from continental and marginal marine settings that include abundant crocodyliform remains. In particular, materials of an enigmatic crocodyliform, Wahasuchus egyptensis gen. et sp. nov., represented by both cranial and postcranial remains, suggest the presence of a novel southern Tethyan crocodyliform assemblage from northern Africa during the Late Cretaceous. Materials recovered of this taxon thus far include fragmentary portions of the skull and mandible, amphicoelous dorsal vertebrae, and fragmentary appendicular remains. This form is distinguished by a number of unique features including a domed platyrostral skull; a strongly festooned lateral margin of the maxilla with two waves of tooth enlargement; a deep, sculptured fossa at the base of the postorbital bar; a jugal with an anterior process that is at least three times broader than the posterior process; an orbital margin dorsally overlapping the lateral temporal fenestra; a supraoccipital with a distinct medial tuber and associated ventral fossa; and a robust straight dentary with contribution of the splenial to the symphysis. This new form suggests a potentially unique Late Cretaceous assemblage from northern Africa, markedly different from better-known Late Cretaceous crocodyliform assemblages of South America and Madagascar, or from earlier (pre-Turonian) deposits in Africa. This pattern may reveal a distinct regional fauna along the southern Tethys, or potential Cretaceous relationships with Eurasian neosuchian-dominated assemblages.

This study uses magnetostratigraphy to help constrain the age of the paleontologically important Galula Formation (Rukwa Rift Basin, southwestern Tanzania). The formation preserves a Cretaceous vertebrate fauna, including saurischian dinosaurs, a putative gondwanatherian mammal, and notosuchian crocodyliforms. With better dating, the Galula Formation and its fossils help fill a temporal gap in our understanding of vertebrate evolution in continental Africa, enabling better evaluation of competing paleobiogeographic hypotheses concerning faunal exchange throughout Gondwana during the Cretaceous. Paleomagnetic samples for this study were collected from the Namba (higher in section) and Mtuka (lower in section) members of the Galula Formation and underwent stepwise thermal demagnetization. All samples displayed a strong normal magnetic polarity overprint, and maximum unblocking temperatures at approximately 690 °C. Three short reversed intervals were identified in the Namba Member, whereas the Mtuka Member lacked any clear reversals. Given the relatively limited existing age constraints, one interpretation correlates the Namba Member to Chron C32. An alternative correlation assigns reversals in the Namba Member to recently proposed short reversals near the end of the Cretaceous Normal Superchron (Chron C34), a time that is traditionally interpreted as having stable normal polarity. The lack of reversals in the Mtuka Member supports deposition within Chron C34. These data suggest that the Namba Member is no older than Late Cretaceous (Cenomanian-Campanian), with the Mtuka Member less well constrained to the middle Cretaceous (Aptian-Cenomanian). The paleomagnetic results are supported by the application of fold and reversal tests for paleomagnetic stability, and paleomagnetic poles for the Namba (246.4°/77.9°, α95 5.9°) and Mtuka (217.1°/72.2°, α95 11.1°) members closely matching the apparent polar wander path for Africa during the Late Cretaceous. These results confidently indicate a Late Creteceous age assignment for the Namba Member of the Galula Formation, a unit that has yielded key crocodyliform (e.g., Pakasuchus; Rukwasuchus) and dinosaur (e.g., Rukwatitan; Shingopana) fossils from eastern Africa.

The Cretaceous Period is considered the archetypical greenhouse interval, yet there is mounting evidence for intermittent cooler climatic phases throughout this geologic span. Few continental climate histories exist for the Cretaceous south of the paleoequator, and fewer still for Africa during this time. The Cretaceous Galula Formation is one of the few exposed fossiliferous continental sedimentary successions from what is now subequatorial Africa that also contains paleosols and spans both the Early and Late Cretaceous series. Bulk sediment and pedogenic carbonate nodules were sampled from paleosols located throughout ~500 m of the Galula Formation stratotype section, including four from the lower Mtuka Member (Aptian-Cenomanian) and four from the upper Namba Member (Cenomanian-Campanian), to reconstruct the paleoclimate and paleoenvironment of the Rukwa Rift Basin. The Mtuka paleosols developed on channel fill deposits in a semi-arid climate amidst well-defined wet and dry seasons that predominantly generated vertic Calcisols. Geochemical and morphological climofunctions determined that mean annual precipitation (MAP) increased up-section from 454 to 860 mm/yr, whereas the δ18O values of pedogenic carbonates and Bt horizon major-element paleothermometry estimated mean annual temperature (MAT) remained relatively steady, ranging between 12.9 and 13.9 °C throughout the Mtuka Member. The paleosols of the overlying Namba Member formed on channel fill and overbank deposits proximal to the main channels, in a semi-arid to sub-humid climate with little seasonality, forming mostly calcic Argillisols. The Namba paleosols formed under slightly wetter and cooler conditions, recording MAP values between 602 and 987 mm/yr, and MAT estimates between 10.3 and 12.5 °C. These data are consistent with the Mtuka paleosols corresponding to the cool greenhouse period that spanned the Aptian-Albian, with the Namba paleosols corresponding to Late Cretaceous cooling following the Cretaceous Thermal Maximum. This work demonstrates that temperate conditions prevailed in the Rukwa Rift Basin during the mid-Late Cretaceous.

The neopterygian fish Enchodus was a widespread, speciose genus consisting of approximately 30 recognized species that were temporally distributed from the late Early Cretaceous through the Paleocene. Many Enchodus specimens are fragmentary cranial remains or isolated dental elements, as is the case for previously reported occurrences in Egypt. Here, we present the most complete specimen of Enchodus recovered from the Late Cretaceous of northeast Africa. The specimen was collected from the upper Campanian Duwi Formation, near the village of Tineida (Dakhla Oasis, Western Desert, Egypt). The new species, Enchodus tineidae sp. nov., consists of right and left dentaries, a partial ectopterygoid, and other cranial bones. The size of the specimen places it into the upper body-size range for the genus. The palatine tooth, an element often useful for diagnosing Enchodus to the species level, is not preserved, but a combination of other cranial characters supports the referral of this specimen to Enchodus. In particular, the dentary preserves three symphysial rostroventral prongs and two tooth rows, the lateral of which consists of small denticles, whereas the medial row comprises large, mediolaterally-compressed teeth. The rostral-most tooth exhibits the highest crown, whereas the rest of the teeth are of lower, variable crown heights. The eight robust, caudal-most medial-row teeth are distributed in a cluster pattern never before observed in Enchodus. Additionally, the dentary and preopercle are both without dermal ornamentation, and the mandibular sensory canal is closed. Phylogenetic analysis recovers this new species as the sister species to E. dirus from North America. Along with previously described materials from Israel, Jordan, Syria, Lebanon, Italy, Morocco, and Libya, this specimen represents a thirteenth species from the northwestern Tethyan geographic distribution of Enchodus.

This paper presents a detailed sedimentologic investigation of a newly identified, fossiliferous Late Neogene sedimentary succession in the Rukwa Rift Basin, southwestern Tanzania. This synrift deposit is a rare and significant new example of a fossiliferous succession of this age in the Western Branch of East Africa Rift System. The unit, informally termed the lower Lake Beds succession, is late Miocene to Pliocene in age based on cross-cutting relationships, preliminary biostratigraphy, and U-Pb geochronology. An angular unconformity separates the lower Lake Beds from underlying Cretaceous and Oligocene strata. Deposition was controlled by rapid generation of accommodation space and increased sediment supply associated with late Cenozoic tectonic reactivation of the Rukwa Rift and synchronous initiation of the Rungwe Volcanic Centre. The lower Lake Beds, which have thus far only been identified in three localities throughout the Rukwa Rift Basin, are characterized by two discrete lithologic members (herein A and B). The lower Member A is a volcanic-rich succession composed mostly of devitrified volcanic tuffs, and volcaniclastic mudstones and sandstones with minor conglomerates. The upper Member B is a siliciclastic-dominated succession of conglomerates, sandstones, mudstones and minor volcanic tuffs. Detailed facies analysis of the lower Lake Beds reveals various distinctive depositional environments that can be grouped into three categories: 1) alluvial fan; 2) fluvial channel; and 3) flood basin environments, characterized by volcanoclastic-filled lakes and ponds, abandoned channel-fills and pedogenically modified floodplains. Member A represents a shallow lacustrine setting filled by tuffaceous sediments, which grade up into a system of alluvial fans and high-energy, proximal gravel-bed braided rivers. An unconformity marks the contact between the two members. Member B shows an upward transition from a high-energy, gravel-bed braided river system to a sandy braided river system with increasingly abundant floodplain deposits and well-developed paleosols. Vertebrate fossils are sparse in member A, but common in member B, preserved both within pedogenic soil horizons and as isolated elements and microsites within fluvial channel facies associations. Faunal remains include fishes, turtles and crocodylians, along with well-preserved mammal cranial and post-cranial remains. In addition, freshwater gastropod shells are locally present in member A and continental trace fossils, including abundant fossilized termite nests, are present in both members.

Precise dating and correlation of drilled wells through continental successions is challenging for hydrocarbon exploration, especially where preservation and recovery of age-diagnostic fossils is poor. As a complement or alternative to biostratigraphic dating we demonstrate the effectiveness of U–Pb geochronology via laser ablation–inductively coupled plasma–mass spectrometry on detrital zircon from well cuttings. In basins with syndepositional volcanic input, the youngest zircons in a stratigraphic interval can refine and serve as a proxy for the age of deposition. We demonstrate the reliability of this technique when applied to hydrocarbon exploration wells by analyzing drill cuttings through a continental interval of the Galula-1 well in the Rukwa Rift Basin, East African rift system, Tanzania, which previously yielded conflicting biostratigraphy results. The lower third of the well section reveals a late Miocene to Pliocene up-hole younging trend in the youngest detrital zircon populations, which matches new radioisotopic ages on volcanic tuffs from a correlative outcrop section. This is followed by an interval with recycled young zircons, followed by a zircon-free interval, interpreted to correspond to changes in magma composition of the nearby Rungwe volcanic province. This study provides the first radioisotopic age constraints for the Lake Beds in the Rukwa rift and demonstrates that sedimentation in the basin began by 8.7 Ma, critical for burial and thermal history modeling and establishing the probability of a working hydrocarbon system. Correspondence in age and zircon preservation between well and outcrop samples from the same intervals provides strong support for applying U–Pb detrital zircon geochronology to well cuttings, as a rapid, inexpensive approach for hydrocarbon exploration.

Infections caused by multi-drug resistant (MDR) bacteria, particularly Gram-negative bacteria, are an escalating global health threat. Often clinicians are forced to administer the last resort antibiotic colistin, however colistin resistance is becoming increasingly prevalent, giving rise to the potential for a situation in which there are no treatment options for MDR Gram-negative infections. The development of adjuvants that circumvent bacterial resistance mechanisms is a promising orthogonal approach to the development of new antibiotics. We recently disclosed that the known IKK-β inhibitor IMD-0354 potently suppresses colistin resistance in several Gram-negative strains. In this report, we explore the structure-activity relationship (SAR) between the IMD-0354 scaffold and colistin resistance suppression, and identify several compounds with more potent activity than the parent against highly colistin resistant strains of Acinetobacter baumannii and Klebsiella pneumoniae.

In 2006, a partial avian femur (South Dakota School of Mines and Technology (SDSM) 78247) from the Upper Cretaceous (Maastrichtian) Sandwich Bluff Member of the López de Bertodano Formation of Sandwich Bluff on Vega Island of the northern Antarctic Peninsula was briefly reported as that of a cariamiform—a clade that includes extant and volant South American species and many extinct flightless and cursorial species. Although other authors have since rejected this taxonomic assignment, SDSM 78247 had never been the subject of a detailed description, hindering a definitive assessment of its affinities. Here we provide the first comprehensive description, illustration, and comparative study of this specimen. Comparison of characters that may be assessed in this femur with those of avian taxa scored in published character matrices refutes the inclusion of SDSM 78247 within Cariamiformes, instead supporting its assignment to a new, as-yet unnamed large-bodied species within the genus Vegavis, and therefore its referral to a clade of semiaquatic anseriforms. Important character states diagnostic of Vegavis + Polarornis include strong craniocaudal bowing of the femoral shaft, the presence of a distinct fossa just proximal to the fibular trochlea, and the broad and flat shape of the patellar sulcus. Referral to Vegavis is based on the presence of a distinctive proximocaudal fossa and distolateral scar. This genus was previously known only from Vegavis iaai, a smaller-bodied taxon from the same locality and stratigraphic unit. Our reassignment of SDSM 78247 to Vegavis sp. removes the record of cariamiform landbirds from the Antarctic Cretaceous.

The African terrestrial fossil record has been limited in its contribution to our understanding of both regional and global Cretaceous paleobiogeography, an interval of significant geologic and macroevolutionary change. A common component in Cretaceous African faunas, titanosaurian sauropods diversified into one of the most specious groups of dinosaurs worldwide. Here we describe the new titanosaurian Mnyamawamtuka moyowamkia gen. et sp. nov. from the Mtuka Member of the Galula Formation in southwest Tanzania. The new specimen preserves teeth, elements from all regions of the postcranial axial skeleton, parts of both appendicular girdles, and portions of both limbs including a complete metatarsus. Unique traits of M. moyowamkia include the lack of an interpostzygapophyseal lamina in posterior dorsal vertebrae, pronounced posterolateral expansion of middle caudal centra, and an unusually small sternal plate. Phylogenetic analyses consistently place M. moyowamkia as either a close relative to lithostrotian titanosaurians (e.g., parsimony, uncalibrated Bayesian analyses) or as a lithostrotian and sister taxon to Malawisaurus dixeyi from the nearby Aptian? Dinosaur Beds of Malawi (e.g., tip-dating Bayesian analyses). M. moyowamkia shares a few features with M. dixeyi, including semi-spatulate teeth and a median lamina between the neural canal and interpostzygapophyseal lamina in anterior dorsal vertebrae. Both comparative morphology and phylogenetic analyses support Mnyamawamtuka as a distinct and distant relative to Rukwatitan bisepultus and Shingopana songwensis from the younger Namba Member of the Galula Formation with these results largely congruent with newly constrained ages for the Mtuka Member (Aptian–Cenomanian) and Namba Member (Campanian). Coupled with recent discoveries from the Dahkla Oasis, Egypt (e.g., Mansourasaurus shahinae) and other parts of continental Afro-Arabia, the Tanzania titanosaurians refine perspectives on the development of African terrestrial faunas throughout the Cretaceous—a critical step in understanding non-marine paleobiogeographic patterns of Africa that have remained elusive until the past few years.

Upper Cretaceous dinosaur remains from Afro-Arabia are rare and mainly restricted to pre-Turonian horizons. Consequently, the discovery of new fossils from Upper Cretaceous deposits in the Western Desert, central Egypt is significant because it adds to the meager record of dinosaurs described from this landmass. The oases of Kharga and Dakhla, Western Desert, Egypt, expose the Quseir Formation (Campanian), with the titanosaurian sauropod Mansourasaurus shahinae as the only currently named dinosaur from strata of this age in the entire region. Numerous (~80) other non-avian dinosaur fossils have also been collected from the Quseir Formation, including material that can be referred to Somphospondyli, Titanosauriformes/Titanosauria, and non-avian Theropoda. Among the discoveries from the Dakhla Oasis are the proximal ends of an associated sauropod tibia and fibula and several sauropod caudal vertebrae representing both sub-adult and adult individuals. Middle caudal vertebrae can be referred to Titanosauria on the basis of procoelous centra and anteriorly displaced neural arches. One caudal vertebra even exhibits pneumatic internal structures rarely observed outside of the Upper Cretaceous South American saltasaurines. Dinosaur fossils recovered from the Kharga Oasis include a partial femur and partial cervical vertebra of a sauropod dinosaur and an isolated proximal fibula of a non-avian theropod dinosaur. Taken together, these findings indicate that Late Cretaceous North African ecosystems supported a diversity of non-avian dinosaurs, some demonstrating affinities with South American forms and others (e.g., Mansourasaurus) with Eurasian groups. Additional exploration of the uppermost Cretaceous units in southern Egypt offers promise for the discovery of important fossils to better characterize Afro-Arabian biotas generally while also providing important perspectives on non-avian dinosaur faunas near the close of the Mesozoic Era.

The Black Warrior Basin is one of several Carboniferous foreland basins along the Appalachian–Ouachita fold-thrust belt in the southeastern United States. Sediment dispersal within the Black Warrior Basin has been a long-debated topic because of a complex tectonic history and the potential interaction between the Appalachian and Ouachita orogenic belts, as well as far field sediment sources. Three dispersal patterns have been proposed, including dispersal routes from the craton, dispersal via the Appalachian foreland, and dispersal from the arc side of the Ouachita suture, but sediment dispersal in the Black Warrior Basin remains inconclusive. In this study, sandstone modal analysis and U–Pb detrital zircon geochronology are used to document the provenance and potential dispersal patterns for selected Mississippian sandstone units in the Black Warrior Basin, Missouri, USA. Results show that the majority of the Lewis, Evans, Sanders, and Carter sandstones are sublitharenite to mature quartzarenite and fall within the Cratonic Interior field on Q–F–L diagrams. U–Pb detrital zircon analyses of the Lewis, Sanders, and Carter sandstones show that there are four distinctive age clusters, including a prominent Paleozoic age cluster (~ 350–500 Ma), a broad Grenville age cluster (~ 900–1350 Ma), and two minor age clusters of the Granite–Rhyolite (~ 1360–1600 Ma) and the Yavapai–Mazatzal (~ 1600–1800 Ma) provinces. All Mississippian sandstones have similar age distributions except for the Lewis sandstone, which lacks zircon grains from the Superior province (>~2500 Ma). Based on the compositional maturity, similarity of age distributions, and changes of relative abundance among different age groups, we conclude that the Late Mississippian sandstone units analyzed during this study were derived from the Laurussian craton and the northern part of the Appalachian foreland through a major axial drainage that occupied the Mississippi Valley Graben.

Abelisaurid theropods were one of the most diverse groups of predatory dinosaurs in Gondwana during the Cretaceous. The group is characterized by a tall, wide skull and robust cervical region. This morphology is thought to have facilitated specialized feeding behaviors such as prolonged contact with prey. The Late Cretaceous abelisaurid Majungasaurus crenatissimus typifies this abelisaurid cranial morphotype. Recent fossil discoveries of this species include a partial growth series that allows for the first time an investigation of ontogenetic variation in cranial morphology in a representative abelisaurid. Herein we examine growth trajectories in the shape of individual cranial bones and articulated skulls of Majungasaurus using geometric morphometrics. Several major changes in skull shape were observed through ontogeny, including an increase in the height of the jugal, postorbital, and quadratojugal, an increase in the extent of the contacts between bones, and a decrease in the circumference of the orbit. The skull transitions from relatively short in the smallest individual to tall and robust in large adults, as is seen in other theropods. Such morphological change during ontogeny would likely have resulted in different biomechanical properties and feeding behaviors between small and large individuals. These findings provide a post-hatching developmental framework for understanding the evolution of the distinctive tall skull morphology seen in abelisaurids and other large-sized theropod dinosaurs.

The Campanian and Maastrichtian stages are very poorly documented time intervals in Africa's record of terrestrial vertebrate evolution. Upper Cretaceous deposits exposed in southern Egypt, near the Dakhla and Kharga Oases in the Western Desert, preserve abundant vertebrate fossils in nearshore marine environments, but have not yet been the focus of intensive collection and description. Our recent paleontological work in these areas has resulted in the discovery of numerous new vertebrate fossil-bearing localities within the middle Campanian Qusier Formation and the upper Campanian-lower Maastrichtian Duwi Formation. Fossil remains recovered from the Campanian-aged Quseir Formation include sharks, rays, actinopterygian and sarcopterygian fishes, turtles, and rare terrestrial archosaurians, including some of the only dinosaurs known from this interval on continental Africa. The upper Campanian/lower Maastrichtian Duwi Formation preserves sharks, sawfish, actinopterygians, and marine reptiles (mosasaurs and plesiosaurs). Notably absent from these collections are representatives of Mammalia and Avialae, both of which remain effectively undocumented in the Upper Cretaceous rocks of Africa and Arabia. New age constraints on the examined rock units is provided by 23 nannofossil taxa, some of which are reported from the Duwi Formation for the first time. Fossil discoveries from rock units of this age are essential for characterizing the degree of endemism that may have developed as the continent became increasingly tectonically isolated from the rest of Gondwana, not to mention for fully evaluating origin and diversification hypotheses of major modern groups of vertebrates (e.g., crown birds, placental mammals).

Research in the Upper Cretaceous (Maastrichtian) Maevarano Formation of northwestern Madagascar has yielded numerous individuals of the abelisaurid theropod Majungasaurus crenatissimus. The number of individuals and quality of preservation allow for a detailed assessment of pathology in this species. A previous survey of Majungasaurus material collected prior to 2005 indicated assorted types of potential infectious and traumatic etiologies. Ongoing research on materials recovered since 2005 provide many additional opportunities to better characterize the anatomy and paleopathology in Majungasaurus. In particular, an almost complete skeleton of a single individual (FMNH PR 2836) preserves evidence of pre-mortem pathologies on multiple bones, including elements of the face and lower jaw, vertebrae, forelimb, ribs and gastralia. Analyses of the pathologies suggest infection of the jugal and quadratojugal, healed fractures and hypertrophic bone growth on several dorsal ribs and a gastralium, septic arthritis in the left forelimb, and evidence of healed/partially healed tooth marks on one cervical vertebra. The injuries appear to be the result of multiple non-fatal events experienced during the life of the individual, rather than a single traumatic incident. Our survey of Majungasaurus indicates most well preserved individuals lack observable pathology, whereas select individuals possess multiple pathologies, a general pattern also noted in other large, nonavian theropods. Such a pattern may result from a snowball effect wherein one injury or infection increases the likelihood of additional maladies due to functional impairment or a compromised immune system in an individual once an initial injury has occurred.

Although knowledge of Mesozoic marine reptiles from Antarctica has improved considerably in recent years, associated and well-preserved skeletal material of these animals remains uncommon. Here we describe a largely complete, closely associated plesiosaur pelvic girdle recovered from the uppermost Cretaceous (Maastrichtian) Sandwich Bluff Member of the Lopez de Bertodano Formation of Vega Island, in the James Ross Basin of the northernmost Antarctic Peninsula. The new specimen exhibits characters that allow its referral to Elasmosauridae, but its incompleteness precludes a more precise taxonomic determination. Ontogenetically variable and systematically useful features of the elasmosaurid pelvis are reviewed and discussed. The new specimen improves knowledge of Southern Hemisphere elasmosaurids just prior to the K/Pg extinction event.

We here establish a new mammaliaform genus and species, Galulatherium jenkinsi (Mammalia), from the Upper Cretaceous Galula Formation in the Rukwa Rift Basin of southwestern Tanzania. This represents the first named taxon of a mammaliaform from the entire Late Cretaceous of continental Afro-Arabia, an interval of 34 million years. Preliminary study of the holotypic and only known specimen (a partial dentary) resulted in tentative assignation to the Gondwanatheria, a poorly known, enigmatic clade of Late Cretaceous-Paleogene Gondwanan mammals (Krause et al. 2003). The application of advanced imaging (mu CT) and visualization techniques permits a more detailed understanding of key anatomical features of the new taxon. It reveals that the lower dentition consisted of a large, procumbent lower incisor and four cheek teeth, all of which were evergrowing (hypselodont). Importantly, all of the teeth appear devoid of enamel. Comparisons conducted with a range of Mesozoic and selected Cenozoic mammaliaform groups document a number of features (e.g., columnar, enamel-less and evergrowing teeth, with relatively simple occlusal morphology) expressed in Galulatherium that are reminiscent of several distantly related groups, making taxonomic assignment difficult at this time. Herein we retain the provisional referral of Galulatherium (RRBP 02067) to Gondwanatheria; it is most similar to sudamericids such as Lavanify and Bharratherium from the Late Cretaceous of Madagascar and India, respectively, in exhibiting relatively simple, high-crowned, columnar cheek teeth. Other features (e.g., enamel-less dentition) are shared with disparate forms such as the Late Jurassic Fruitafossor and toothed xenarthrans (e.g., sloths), here attributed to convergence. Revised analyses of the depositional context for the holotype place it as having lived sometime between the late Turonian and latest Campanian (roughly 91-72 million years ago). This enhanced geochronological context helps to refine the palaeobiogeographical significance of Galulatherium among Cretaceous mammals in general and those from Gondwanan landmasses specifically.

Cape Marsh, located on the eastern end of Robertson Island to the east of the Antarctic Peninsula, exposes an isolated outcrop of Upper Cretaceous sedimentary strata. The outcrop is approximately 120 km southwest of the much better-studied exposures of similar age on and around James Ross Island (JRI); as such, its remoteness has complicated logistical access to the site and hindered geologic correlations on a regional scale. Here we present the results of fieldwork conducted in 2016 that yielded a more diverse invertebrate fossil assemblage than had been previously recognized, in addition to new U–Pb detrital zircon and magnetostratigraphic data. The invertebrate fauna, particularly the ammonites and inoceramids, support a biostratigraphic correlation of the upper Cape Marsh strata to Ammonite Assemblage 7 previously established on JRI. Detrital zircon U–Pb analysis conducted on a sandstone sample from the same strata indicates a maximum depositional age of 74.2 ± 1.1 Ma, and magnetostratigraphic interpretation of the lower strata suggest a normal magnetochron. These results are all consistent with a Campanian age for the deposition of the upper strata at Cape Marsh, and deposition during magnetochron C33N for the lower layers. However, a slight age inconsistency between the biostratigraphic correlation and the detrital zircon-derived maximum depositional age may imply that the fossils are reworked. Regardless, these new data allow us to correlate the strata at Cape Marsh to the Santa Marta and Rabot formations (or possibly the lower part of the Snow Hill Island Formation) in the northern part of the James Ross Basin.
