1. The knob protein KAHRP assembles into a ring-shaped structure that underpins virulence complex assembly.

Abstract: Plasmodium falciparum mediates adhesion of infected red blood cells (RBCs) to blood vessel walls by assembling a multi-protein complex at the RBC surface. This virulence-mediating structure, called the knob, acts as a scaffold for the presentation of the major virulence antigen, P. falciparum Erythrocyte Membrane Protein-1 (PfEMP1). In this work we developed correlative STochastic Optical Reconstruction Microscopy-Scanning Electron Microscopy (STORM-SEM) to spatially and temporally map the delivery of the knob-associated histidine-rich protein (KAHRP) and PfEMP1 to the RBC membrane skeleton. We show that KAHRP is delivered as individual modules that assemble in situ, giving a ring-shaped fluorescence profile around a dimpled disk that can be visualized by SEM. Electron tomography of negatively-stained membranes reveals a previously observed spiral scaffold underpinning the assembled knobs. Truncation of the C-terminal region of KAHRP leads to loss of the ring structures, disruption of the raised disks and aberrant formation of the spiral scaffold, pointing to a critical role for KAHRP in assembling the physical knob structure. We show that host cell actin remodeling plays an important role in assembly of the virulence complex, with cytochalasin D blocking knob assembly. Additionally, PfEMP1 appears to be delivered to the RBC membrane, then inserted laterally into knob structures.

3. Essential role of Plasmodium perforin-like protein 4 in ookinete midgut passage.

Abstract: Pore forming proteins such as those belonging to the membrane attack/perforin (MACPF) family have important functions in many organisms. Of the five MACPF proteins found in Plasmodium parasites, three have functions in cell passage and one in host cell egress. Here we report an analysis of the perforin-like protein 4, PPLP4, in the rodent parasite Plasmodium berghei. We found that the protein is expressed only in the ookinete, the invasive stage of the parasite formed in the mosquito midgut. Transcriptional analysis revealed that expression of the pplp4 gene commences during ookinete development. The protein was detected in retorts and mature ookinetes. Using two antibodies, the protein was found localized in a dotted pattern, and 3-D SIM super-resolution microcopy revealed the protein in the periphery of the cell. Analysis of a C-terminal mCherry fusion of the protein however showed mainly cytoplasmic label. A pplp4 null mutant formed motile ookinetes, but these were unable to invade and traverse the midgut epithelium resulting in severely impaired oocyst formation and no transmission to naïve mice. However, when in vitro cultured ookinetes were injected into the thorax of the mosquito, thus by-passing midgut passage, sporozoites were formed and the mutant parasites were able to infect naïve mice. Taken together, our data show that PPLP4 is required only for ookinete invasion of the mosquito midgut. Thus PPLP4 has a similar role to the previously studied PPLP3 and PPLP5, raising the question why three proteins with MACPF domains are needed for invasion by the ookinete of the mosquito midgut epithelium.

Abstract: The apical complex is the definitive cell structure of phylum Apicomplexa, and is the focus of the events of host cell penetration and the establishment of intracellular parasitism. Despite the importance of this structure, its molecular composition is relatively poorly known and few studies have experimentally tested its functions. We have characterized a novel Toxoplasma gondii protein, RNG2, that is located at the apical polar ring--the common structural element of apical complexes. During cell division, RNG2 is first recruited to centrosomes immediately after their duplication, confirming that assembly of the new apical complex commences as one of the earliest events of cell replication. RNG2 subsequently forms a ring, with the carboxy- and amino-termini anchored to the apical polar ring and mobile conoid, respectively, linking these two structures. Super-resolution microscopy resolves these two termini, and reveals that RNG2 orientation flips during invasion when the conoid is extruded. Inducible knockdown of RNG2 strongly inhibits host cell invasion. Consistent with this, secretion of micronemes is prevented in the absence of RNG2. This block, however, can be fully or partially overcome by exogenous stimulation of calcium or cGMP signaling pathways, respectively, implicating the apical complex directly in these signaling events. RNG2 demonstrates for the first time a role for the apical complex in controlling secretion of invasion factors in this important group of parasites.

5. Apicoplast lipoic acid protein ligase B is not essential for Plasmodium falciparum.

Abstract: Lipoic acid (LA) is an essential cofactor of alpha-keto acid dehydrogenase complexes (KADHs) and the glycine cleavage system. In Plasmodium, LA is attached to the KADHs by organelle-specific lipoylation pathways. Biosynthesis of LA exclusively occurs in the apicoplast, comprising octanoyl-[acyl carrier protein]: protein N-octanoyltransferase (LipB) and LA synthase. Salvage of LA is mitochondrial and scavenged LA is ligated to the KADHs by LA protein ligase 1 (LplA1). Both pathways are entirely independent, suggesting that both are likely to be essential for parasite survival. However, disruption of the LipB gene did not negatively affect parasite growth despite a drastic loss of LA (>90%). Surprisingly, the sole, apicoplast-located pyruvate dehydrogenase still showed lipoylation, suggesting that an alternative lipoylation pathway exists in this organelle. We provide evidence that this residual lipoylation is attributable to the dual targeted, functional lipoate protein ligase 2 (LplA2). Localisation studies show that LplA2 is present in both mitochondrion and apicoplast suggesting redundancy between the lipoic acid protein ligases in the erythrocytic stages of P. falciparum.

6. Plasmodium falciparum formins are essential for invasion and sexual stage development

Abstract: Abstract The malaria parasite uses actin-based mechanisms throughout its lifecycle to control a range of biological processes including intracellular trafficking, gene regulation, parasite motility and invasion. In this work we assign functions to the Plasmodium falciparum formins 1 and 2 (FRM1 and FRM2) proteins in asexual and sexual blood stage development. We show that FRM1 is essential for merozoite invasion and FRM2 is required for efficient cell division. We also observed divergent functions for FRM1 and FRM2 in gametocyte development. Conditional deletion of FRM1 leads to a delay in gametocyte stage progression. We show that FRM2 controls the actin and microtubule cytoskeletons in developing gametocytes, with premature removal of the protein resulting in a loss of transmissible stage V gametocytes. Lastly, we show that targeting formin proteins with the small molecule inhibitor of formin homology domain 2 (SMIFH2) leads to a multistage block in asexual and sexual stage parasite development.

7. The velocity distribution of white dwarfs in Gaia EDR3

Abstract: Using a penalised maximum likelihood we estimate, for the first time, the velocity distribution of white dwarfs in the Solar neighbourhood. Our sample consists of 129 675 white dwarfs within 500 pc in Gaia Early Data Release 3 The white dwarf velocity distributions reveal a similar structure to the rest of the Solar neighbourhood stars, reflecting that white dwarfs are subjected to the same dynamical processes. In the velocity distribution for three magnitude-binned subsamples we however find a novel structure at $(U, V) = (7, -19)$ km s$^{-1}$ in fainter samples, potentially related to the Coma Berenices stream. We also see a double-peaked feature in $U$-$W$ at $U \approx -30$ km s$^{-1}$ and in $V$-$W$ at $V \approx -20$ km s$^{-1}$ for fainter samples. We determine the velocity distribution and velocity moments as a function of absolute magnitude for two samples based on the bifurcation identified in Gaia Data Release 2 in the colour-magnitude diagram. The brighter, redder sequence has a larger velocity dispersion than the fainter, bluer sequence across all magnitudes. It is hard to reconcile this kinematic difference with a bifurcation caused purely by atmospheric composition, while it fits neatly with a significant age difference between the two sequences. Our results provide novel insights into the kinematic properties of white dwarfs and demonstrate the power of analytical techniques that work for the large fraction of stars that do not have measured radial velocities in the current era of large-scale astrometric surveys., 18 pages, 10 figures. 8. Paraspeckle subnuclear bodies depend on dynamic heterodimerisation of DBHS RNA-binding proteins via their structured domains

Abstract: RNA-binding proteins of the DBHS (Drosophila Behavior Human Splicing) family, NONO, SFPQ, and PSPC1 have numerous roles in genome stability and transcriptional and posttranscriptional regulation. Critical to DBHS activity is their recruitment to distinct subnuclear locations, for example, paraspeckle condensates, where DBHS proteins bind to the long noncoding RNA NEAT1 in the first essential step in paraspeckle formation. To carry out their diverse roles, DBHS proteins form homodimers and heterodimers, but how this dimerization influences DBHS localization and function is unknown. Here, we present an inducible GFP-NONO stable cell line and use it for live-cell 3D-structured illumination microscopy, revealing paraspeckles with dynamic, twisted elongated structures. Using siRNA knockdowns, we show these labeled paraspeckles consist of GFP-NONO/endogenous SFPQ dimers and that GFP-NONO localization to paraspeckles depends on endogenous SFPQ. Using purified proteins, we confirm that partner swapping between NONO and SFPQ occurs readily in vitro. Crystallographic analysis of the NONO-SFPQ heterodimer reveals conformational differences to the other DBHS dimer structures, which may contribute to partner preference, RNA specificity, and subnuclear localization. Thus overall, our study suggests heterodimer partner availability is crucial for NONO subnuclear distribution and helps explain the complexity of both DBHS protein and paraspeckle dynamics through imaging and structural approaches.

9. New stellar velocity substructures from Gaia DR3 proper motions

Abstract: Local stellar motions are expected, and have been shown, to include signatures of the Galaxy's past dynamical evolution. These are typically divided into the disc, which shows the dynamical effects of spiral arms and the bar, and the stellar halo, with structures thought to be debris from past mergers. We use Gaia Data Release 3 to select large samples of these populations without limiting them to sources with radial velocities. We apply a penalised maximum likelihood method to these samples to determine the full 3D velocity distribution in Cartesian $(U, V, W)$ or spherical $(v_r, v_\phi, v_\theta)$ coordinates. We find that the disc population is dominated by four moving groups and also detect a new moving group at $(U, V) = (-10, -15)$ km s$^{-1}$ which we call MMH-0. For the stellar halo, we isolate the accreted component with cuts in transverse velocity and the colour-magnitude diagram. In this component we find several known structures believed to be caused by past mergers, particularly one around $(v_r, v_\phi, v_\theta) = (-150, -300, -100)$ km s$^{-1}$ appears more prominent than previously claimed. Furthermore we also identify two new structures near $(v_r, v_\phi, v_\theta) = (225, 25, 325)$ km s$^{-1}$ and $(0, 150, -125)$ km s$^{-1}$ which we refer to as MMH-1 and MMH-2 respectively. These results give new insights into local stellar motions and shows the potential of using samples that are not limited to stars with measured line-of-sight velocities, which is key to providing large samples of stars, necessary for future studies., Comment: 15 pages, 10 figures. 10. Targeting of microvillus protein Eps8 by the NleH effector kinases from enteropathogenic E. coli

Abstract: Attaching and effacing (AE) lesion formation on enterocytes by enteropathogenic Escherichia coli (EPEC) requires the EPEC type III secretion system (T3SS). Two T3SS effectors injected into the host cell during infection are the atypical kinases, NleH1 and NleH2. However, the host targets of NleH1 and NleH2 kinase activity during infection have not been reported. Here phosphoproteomics identified Ser775 in the microvillus protein Eps8 as a bona fide target of NleH1 and NleH2 phosphorylation. Both kinases interacted with Eps8 through previously unrecognized, noncanonical “proline-rich” motifs, PxxDY, that bound the Src Homology 3 (SH3) domain of Eps8. Structural analysis of the Eps8 SH3 domain bound to a peptide containing one of the proline-rich motifs from NleH showed that the N-terminal part of the peptide adopts a type II polyproline helix, and its C-terminal “DY” segment makes multiple contacts with the SH3 domain. Ser775 phosphorylation by NleH1 or NleH2 hindered Eps8 bundling activity and drove dispersal of Eps8 from the AE lesion during EPEC infection. This finding suggested that NleH1 and NleH2 altered the cellular localization of Eps8 and the cytoskeletal composition of AE lesions during EPEC infection.

Abstract: Aims. We produce a clean and well-characterised catalogue of objects within 100 pc of the Sun from the Gaia Early Data Release 3. We characterise the catalogue through comparisons to the full data release, external catalogues, and simulations. We carry out a first analysis of the science that is possible with this sample to demonstrate its potential and best practices for its use., Methods. Theselection of objects within 100 pc from the full catalogue used selected training sets, machine-learning procedures, astrometric quantities, and solution quality indicators to determine a probability that the astrometric solution is reliable. The training set construction exploited the astrometric data, quality flags, and external photometry. For all candidates we calculated distance posterior probability densities using Bayesian procedures and mock catalogues to define priors. Any object with reliable astrometry and a non-zero probability of being within 100 pc is included in the catalogue., Results. We have produced a catalogue of 331 312 objects that we estimate contains at least 92% of stars of stellar type M9 within 100 pc of the Sun. We estimate that 9% of the stars in this catalogue probably lie outside 100 pc, but when the distance probability function is used, a correct treatment of this contamination is possible. We produced luminosity functions with a high signal-to-noise ratio for the main-sequence stars, giants, and white dwarfs. We examined in detail the Hyades cluster, the white dwarf population, and wide-binary systems and produced candidate lists for all three samples. We detected local manifestations of several streams, superclusters, and halo objects, in which we identified 12 members of Gaia Enceladus. We present the first direct parallaxes of five objects in multiple systems within 10 pc of the Sun., Conclusions. We provide the community with a large, well-characterised catalogue of objects in the solar neighbourhood. This is a primary benchmark for measuring and understanding fundamental parameters and descriptive functions in astronomy.

Abstract: Hobbs, David, et al., The era of all-sky space astrometry began with the Hipparcos mission in 1989 and provided the first very accurate catalogue of apparent magnitudes, positions, parallaxes and proper motions of 120 000 bright stars at the milliarcsec (or milliarcsec per year) accuracy level. Hipparcos has now been superseded by the results of the Gaia mission. The second Gaia data release contained astrometric data for almost 1.7 billion sources with tens of microarcsec (or microarcsec per year) accuracy in a vast volume of the Milky Way and future data releases will further improve on this. Gaia has just completed its nominal 5-year mission (July 2019), but is expected to continue in operations for an extended period of an additional 5 years through to mid 2024. Its final catalogue to be released ∼ 2027, will provide astrometry for ∼ 2 billion sources, with astrometric precisions reaching 10 microarcsec. Why is accurate astrometry so important? The answer is that it provides fundamental data which underpin much of modern observational astronomy as will be detailed in this White Paper. All-sky visible and Near-InfraRed (NIR) astrometry with a wavelength cutoff in the K-band is not just focused on a single or small number of key science cases. Instead, it is extremely broad, answering key science questions in nearly every branch of astronomy while also providing a dense and accurate visible-NIR reference frame needed for future astronomy facilities., Open access funding provided by Lund University.

Abstract: We use Gaia DR2 astrometric and line-of-sight velocity information combined with two sets of distances obtained with a Bayesian inference method to study the 3D velocity distribution in the Milky Way disc. We search for variations in all Galactocentric cylindrical velocity components ($V_{\phi}$, $V_R$ and $V_z$) with Galactic radius, azimuth, and distance from the disc mid-plane. We confirm recent work showing that bulk vertical motions in the $R\text{-}z$ plane are consistent with a combination of breathing and bending modes. In the $x\text{-}y$ plane, we show that, although the amplitudes change, the structure produced by these modes is mostly invariant as a function of distance from the plane. Comparing to two different Galactic disc models, we demonstrate that the observed patterns can drastically change in short time intervals, showing the complexity of understanding the origin of vertical perturbations. 15. Identification and characterisation of splicing regulators in Toxoplasma gondii

Abstract: The splicing of mRNA constitutes a major source of co- and post-transcriptional regulation in metazoans. In particular, members of the serine/arginine (SR) protein family are essential splicing factors that are implicated in the regulation of gene expression and RNA metabolism. However, very little is known about these proteins in apicomplexans, a phylum that includes some of the most important global parasites. In this study, we investigated the suite of three uncharacterised SR proteins in Toxoplasma gondii and show that all three are found localised to nuclear speckles. We show, by genetic ablation, that TgSR1 is particularly important for T. gondii growth. Using RNA-seq, we also characterised the global gene expression and splicing regulation of these proteins. We find that the SR proteins regulate several types of alternative splicing of distinct but overlapping subsets of transcripts, as well as impacting transcript abundance. Most of the alternative splicing events are non-productive intron retention events that do not appear to affect transcript abundance. The splicing sites of the impacted transcripts are enriched in characteristic SR binding motifs. We also identified and conditionally knocked down two putative kinases of SR proteins. The kinases are localised to nuclear speckles and are essential to parasite survival. Their perturbation resulted in widespread changes to splicing, but the affected transcripts did not mirror the patterns seen in knockouts of individual SRs, suggesting an absence of a simple relationship between SRs and these putative kinase regulators. Overall, this study reveals a complex system of splicing factors and kinases that post-transcriptionally regulate gene expression in T. gondii.

Abstract: Porphyromonas gingivalis, a bacterial pathogen contributing to human periodontitis, exports and anchors cargo proteins to its surface, enabling the production of black pigmentation using a type IX secretion system (T9SS) and conjugation to anionic lipopolysaccharide (A-LPS). To determine whether T9SS components need to be assembled in situ for correct secretion and A-LPS modification of cargo proteins, combinations of nonpigmented mutants lacking A-LPS or a T9SS component were mixed to investigate in trans complementation. Reacquisition of pigmentation occurred only between an A-LPS mutant and a T9SS mutant, which coincided with A-LPS modification of cargo proteins detected by Western blotting and coimmunoprecipitation/quantitative mass spectrometry. Complementation also occurred using an A-LPS mutant mixed with outer membrane vesicles (OMVs) or purified A-LPS. Fluorescence experiments demonstrated that OMVs can fuse with and transfer lipid to P. gingivalis, leading to the conclusion that complementation of T9SS function occurred through A-LPS transfer between cells. None of the two-strain crosses involving only the five T9SS OM component mutants produced black pigmentation, implying that the OM proteins cannot be transferred in a manner that restores function and surface pigmentation, and hence, a more ordered temporal in situ assembly of T9SS components may be required. Our results show that LPS can be transferred between cells or between cells and OMVs to complement deficiencies in LPS biosynthesis and hemin-related pigmentation to reveal a potentially new mechanism by which the oral microbial community is modulated to produce clinical consequences in the human host. IMPORTANCEPorphyromonas gingivalis is a keystone pathogen contributing to periodontitis in humans, leading to tooth loss. The oral microbiota is essential in this pathogenic process and changes from predominantly Gram-positive (health) to predominantly Gram-negative (disease) species. P. gingivalis uses its type IX secretion system (T9SS) to secrete and conjugate virulence proteins to anionic lipopolysaccharide (A-LPS). This study investigated whether components of this secretion system could be complemented and found that it was possible for A-LPS biosynthetic mutants to be complemented in trans both by strains that had the A-LPS on the cell surface and by exogenous sources of A-LPS. This is the first known example of LPS exchange in a human bacterial pathogen which causes disease through complex microbiota-host interactions.

Abstract: Plant cell walls (PCWs) form the outer barrier of cells that give the plant strength and directly interact with the environment and other cells in the plant. PCWs are composed of several polysaccharides, of which cellulose forms the main fibrillar network. Enmeshed between these fibrils of cellulose are non-cellulosic polysaccharides (NCPs), pectins, and proteins. This study investigates the sequence, timing, patterning, and architecture of cell wall polysaccharide regeneration in suspension culture cells (SCC) of the grass species Lolium multiflorum (Lolium). Confocal, superresolution, and electron microscopies were used in combination with cytochemical labeling to investigate polysaccharide deposition in SCC after protoplasting. Cellulose was the first polysaccharide observed, followed shortly thereafter by (1,3;1,4)-β-glucan, which is also known as mixed-linkage glucan (MLG), arabinoxylan (AX), and callose. Cellulose formed fibrils with AX and produced a filamentous-like network, whereas MLG formed punctate patches. Using colocalization analysis, cellulose and AX were shown to interact during early stages of wall generation, but this interaction reduced over time as the wall matured. AX and MLG interactions increased slightly over time, but cellulose and MLG were not seen to interact. Callose initially formed patches that were randomly positioned on the protoplast surface. There was no consistency in size or location over time. The architecture observed via superresolution microscopy showed similarities to the biophysical maps produced using atomic force microscopy and can give insight into the role of polysaccharides in PCWs.

Abstract: Aims. We aim to demonstrate the scientific potential of the Gaia Early Data Release 3 (EDR3) for the study of different aspects of the Milky Way structure and evolution and we provide, at the same time, a description of several practical aspects of the data and examples of their usage., Methods. We used astrometric positions, proper motions, parallaxes, and photometry from EDR3 to select different populations and components and to calculate the distances and velocities in the direction of the anticentre. In this direction, the Gaia astrometric data alone enable the calculation of the vertical and azimuthal velocities; also, the extinction is relatively low compared to other directions in the Galactic plane. We then explore the disturbances of the current disc, the spatial and kinematical distributions of early accreted versus in situ stars, the structures in the outer parts of the disc, and the orbits of open clusters Berkeley 29 and Saurer 1., Results. With the improved astrometry and photometry of EDR3, we find that: (i) the dynamics of the Galactic disc are very complex with oscillations in the median rotation and vertical velocities as a function of radius, vertical asymmetries, and new correlations, including a bimodality with disc stars with large angular momentum moving vertically upwards from below the plane, and disc stars with slightly lower angular momentum moving preferentially downwards; (ii) we resolve the kinematic substructure (diagonal ridges) in the outer parts of the disc for the first time; (iii) the red sequence that has been associated with the proto-Galactic disc that was present at the time of the merger with Gaia-Enceladus-Sausage is currently radially concentrated up to around 14 kpc, while the blue sequence that has been associated with debris of the satellite extends beyond that; (iv) there are density structures in the outer disc, both above and below the plane, most probably related to Monoceros, the Anticentre Stream, and TriAnd, for which the Gaia data allow an exhaustive selection of candidate member stars and dynamical study; and (v) the open clusters Berkeley 29 and Saurer 1, despite being located at large distances from the Galactic centre, are on nearly circular disc-like orbits., Conclusions. Even with our simple preliminary exploration of the Gaia EDR3, we demonstrate how, once again, these data from the European Space Agency are crucial for our understanding of the different pieces of our Galaxy and their connection to its global structure and history.

Abstract: Aims To establish pre-clinical proof of concept that sustained subcutaneous delivery of the secretome of human cardiac stem cells (CSCs) can be achieved in vivo to produce significant cardioreparative outcomes in the setting of myocardial infarction. Methods and results Rats were subjected to permanent ligation of left anterior descending coronary artery and randomized to receive subcutaneous implantation of TheraCyte devices containing either culture media as control or 1 × 106 human W8B2+ CSCs, immediately following myocardial ischaemia. At 4 weeks following myocardial infarction, rats treated with W8B2+ CSCs encapsulated within the TheraCyte device showed preserved left ventricular ejection fraction. The preservation of cardiac function was accompanied by reduced fibrotic scar tissue, interstitial fibrosis, cardiomyocyte hypertrophy, as well as increased myocardial vascular density. Histological analysis of the TheraCyte devices harvested at 4 weeks post-implantation demonstrated survival of human W8B2+ CSCs within the devices, and the outer membrane was highly vascularized by host blood vessels. Using CSCs expressing plasma membrane reporters, extracellular vesicles of W8B2+ CSCs were found to be transferred to the heart and other organs at 4 weeks post-implantation. Furthermore, mass spectrometry-based proteomic profiling of extracellular vesicles of W8B2+ CSCs identified proteins implicated in inflammation, immunoregulation, cell survival, angiogenesis, as well as tissue remodelling and fibrosis that could mediate the cardioreparative effects of secretome of human W8B2+ CSCs. Conclusions Subcutaneous implantation of TheraCyte devices encapsulating human W8B2+ CSCs attenuated adverse cardiac remodelling and preserved cardiac function following myocardial infarction. The TheraCyte device can be employed to deliver stem cells in a minimally invasive manner for effective secretome-based cardiac therapy.

Abstract: We present the early installment of the third Gaia data release, Gaia EDR3, consisting of astrometry and photometry for 1.8 billion sources brighter than magnitude 21, complemented with the list of radial velocities from Gaia DR2. Gaia EDR3 contains celestial positions and the apparent brightness in G for approximately 1.8 billion sources. For 1.5 billion of those sources, parallaxes, proper motions, and the (G_BP-G_RP) colour are also available. The passbands for G, G_BP, and G_RP are provided as part of the release. For ease of use, the 7 million radial velocities from Gaia DR2 are included in this release, after the removal of a small number of spurious values. New radial velocities will appear as part of Gaia DR3. Finally, Gaia EDR3 represents an updated materialisation of the celestial reference frame (CRF) in the optical, the Gaia-CRF3, which is based solely on extragalactic sources. The creation of the source list for Gaia EDR3 includes enhancements that make it more robust with respect to high proper motion stars, and the disturbing effects of spurious and partially resolved sources. The source list is largely the same as that for Gaia DR2, but it does feature new sources and there are some notable changes. The source list will not change for Gaia DR3. Gaia EDR3 represents a significant advance over Gaia DR2, with parallax precisions increased by 30 percent, proper motion precisions increased by a factor of 2, and the systematic errors in the astrometry suppressed by 30--40 percent for the parallaxes and by a factor ~2.5 for the proper motions. The photometry also features increased precision, but above all much better homogeneity across colour, magnitude, and celestial position. A single passband for G, G_BP, and G_RP is valid over the entire magnitude and colour range, with no systematics above the 1 percent level.

Abstract: [Context] Gaia Early Data Release 3 (Gaia EDR3) contains results for 1.812 billion sources in the magnitude range G = 3-21 based on observations collected by the European Space Agency Gaia satellite during the first 34 months of its operational phase. [Aims] We describe the input data, the models, and the processing used for the astrometric content of Gaia EDR3, as well as the validation of these results performed within the astrometry task. [Methods] The processing broadly followed the same procedures as for Gaia DR2, but with significant improvements to the modelling of observations. For the first time in the Gaia data processing, colour-dependent calibrations of the line- and point-spread functions have been used for sources with well-determined colours from DR2. In the astrometric processing these sources obtained five-parameter solutions, whereas other sources were processed using a special calibration that allowed a pseudocolour to be estimated as the sixth astrometric parameter. Compared with DR2, the astrometric calibration models have been extended, and the spin-related distortion model includes a self-consistent determination of basic-angle variations, improving the global parallax zero point. [Results] Gaia EDR3 gives full astrometric data (positions at epoch J2016.0, parallaxes, and proper motions) for 1.468 billion sources (585 millionwith five-parameter solutions, 882 million with six parameters), and mean positions at J2016.0 for an additional 344 million.Solutions with five parameters are generally more accurate than six-parameter solutions, and are available for 93% of the sources brighter than the 17th magnitude. The median uncertainty in parallax and annual proper motion is 0.02-0.03 mas at magnitude G = 9-14, and around 0.5 mas at G = 20. Abstract: [Context] Data-driven methods play an increasingly important role in the field of astrophysics In the context of large spectroscopic surveys of stars, data-driven methods are key in deducing physical parameters for millions of spectra in a short time. Convolutional neural networks (CNNs) enable us to connect observables (e.g. spectra, stellar magnitudes) to physical properties (atmospheric parameters, chemical abundances, or labels in general)., [Aims] We test whether it is possible to transfer the labels derived from a high-resolution stellar survey to intermediate-resolution spectra of another survey by using a CNN., [Methods] We trained a CNN, adopting stellar atmospheric parameters and chemical abundances from APOGEE DR16 (resolution R = 22 500) data as training set labels. As input, we used parts of the intermediate-resolution RAVE DR6 spectra (R ∼ 7500) overlapping with the APOGEE DR16 data as well as broad-band ALL_WISE and 2MASS photometry, together with Gaia DR2 photometry and parallaxes., [Results] We derived precise atmospheric parameters Teff, log(g), and [M/H], along with the chemical abundances of [Fe/H], [α/M], [Mg/Fe], [Si/Fe], [Al/Fe], and [Ni/Fe] for 420 165 RAVE spectra. The precision typically amounts to 60 K in Teff, 0.06 in log(g) and 0.02−0.04 dex for individual chemical abundances. Incorporating photometry and astrometry as additional constraints substantially improves the results in terms of the accuracy and precision of the derived labels, as long as we operate in those parts of the parameter space that are well-covered by the training sample. Scientific validation confirms the robustness of the CNN results. We provide a catalogue of CNN-trained atmospheric parameters and abundances along with their uncertainties for 420 165 stars in the RAVE survey., [Conclusions] CNN-based methods provide a powerful way to combine spectroscopic, photometric, and astrometric data without the need to apply any priors in the form of stellar evolutionary models. The developed procedure can extend the scientific output of RAVE spectra beyond DR6 to ongoing and planned surveys such as Gaia RVS, 4MOST, and WEAVE. Abstract: The Radial Velocity Experiment (Rave) is a magnitude-limited (9 < I < 12) spectroscopic survey of Galactic stars randomly selected in Earth's southern hemisphere. The Rave medium-resolution spectra (R ∼ 7500) cover the Ca-triplet region (8410-8795 Å). The sixth and final data release (DR6) is based on 518,387 observations of 451,783 unique stars. Rave observations were taken between 2003 April 12 and 2013 April 4. Here we present the genesis, setup, and data reduction of Rave as well as wavelength-calibrated and flux-normalized spectra and error spectra for all observations in Rave DR6. Furthermore, we present derived spectral classification and radial velocities for the Rave targets, complemented by cross-matches with Gaia DR2 and other relevant catalogs. A comparison between internal error estimates, variances derived from stars with more than one observing epoch, and a comparison with radial velocities of Gaia DR2 reveals consistently that 68% of the objects have a velocity accuracy better than 1.4 km s-1, while 95% of the objects have radial velocities better than 4.0 km s-1. Stellar atmospheric parameters, abundances and distances are presented in a subsequent publication. Abstract: We present part 2 of the sixth and final Data Release (DR6) of the Radial Velocity Experiment (Rave), a magnitude-limited spectroscopic survey of Galactic stars randomly selected in Earth's southern hemisphere. The Rave medium-resolution spectra (R ∼ 7500) cover the Ca triplet region (8410-8795 Å) and span the complete time frame from the start of Rave observations on 2003 April 12 to their completion on 2013 April 4. In the second of two publications, we present the data products derived from 518,387 observations of 451,783 unique stars using a suite of advanced reduction pipelines focusing on stellar atmospheric parameters, in particular purely spectroscopically derived stellar atmospheric parameters, and the overall metallicity), enhanced stellar atmospheric parameters inferred via a Bayesian pipeline using Gaia DR2 astrometric priors, and asteroseismically calibrated stellar atmospheric parameters for giant stars based on asteroseismic observations for 699 K2 stars. In addition, we provide abundances of the elements Fe, Al, and Ni, as well as an overall [α/Fe] ratio obtained using a new pipeline based on the GAUGUIN optimization method that is able to deal with variable signal-to-noise ratios. The Rave DR6 catalogs are cross-matched with relevant astrometric and photometric catalogs, and are complemented by orbital parameters and effective temperatures based on the infrared flux method. Abstract: We study the radial migration of stars as a function of orbital action as well as the structural properties of a large suite of N-body simulations of isolated disc galaxies. Our goal is to establish a relationship between the radial migration efficiency of stars and their vertical action. We aim to describe how that relationship depends on the relative gravitational dominance between the disc and the dark matter halo. By changing the mass ratio of our disc and dark matter halo we find a relationship between disc dominance, number and strength of spiral arms, and the ensuing radial migration as a function of the vertical action. We conclude that the importance of migration at large vertical action depends on the strength of the spiral arms and therefore the dominance of the disc. Populations with more radial action undergo less radial migration, independently of disc dominance. Our results are important for the future of analytical modelling of radial migration in galaxies and furthers the understanding of radial migration which is a key component of the restructuring of galaxies, including the Milky Way., 13 pages, 12 figures, accepted for publication in MNRAS

Abstract: The trafficking of the virulence antigen PfEMP1 and its presentation at the knob structures at the surface of parasite-infected RBCs are central to severe adhesion-related pathologies such as cerebral and placental malaria. This work adds to our understanding of how PfEMP1 is trafficked to the RBC membrane by defining the protein-protein interaction networks that function at the Maurer’s clefts controlling PfEMP1 loading and unloading. We characterize a protein needed for virulence protein trafficking and provide new insights into the mechanisms for host cell remodeling, parasite survival within the host, and virulence., The malaria parasite Plasmodium falciparum traffics the virulence protein P. falciparum erythrocyte membrane protein 1 (PfEMP1) to the surface of infected red blood cells (RBCs) via membranous organelles, known as the Maurer’s clefts. We developed a method for efficient enrichment of Maurer’s clefts and profiled the protein composition of this trafficking organelle. We identified 13 previously uncharacterized or poorly characterized Maurer’s cleft proteins. We generated transfectants expressing green fluorescent protein (GFP) fusions of 7 proteins and confirmed their Maurer’s cleft location. Using co-immunoprecipitation and mass spectrometry, we generated an interaction map of proteins at the Maurer’s clefts. We identified two key clusters that may function in the loading and unloading of PfEMP1 into and out of the Maurer’s clefts. We focus on a putative PfEMP1 loading complex that includes the protein GEXP07/CX3CL1-binding protein 2 (CBP2). Disruption of GEXP07 causes Maurer’s cleft fragmentation, aberrant knobs, ablation of PfEMP1 surface expression, and loss of the PfEMP1-mediated adhesion. ΔGEXP07 parasites have a growth advantage compared to wild-type parasites, and the infected RBCs are more deformable and more osmotically fragile.

Abstract: Gaia Early Data Release 3 (Gaia EDR3) gives trigonometric parallaxes for nearly 1.5 billion sources. Inspection of the EDR3 data for sources identified as quasars reveals that their parallaxes are biased, that is systematically offset from the expected distribution around zero, by a few tens of microarcsec. We attempt to map the main dependencies of the parallax bias in EDR3. In principle this could provide a recipe for correcting the EDR3 parallaxes. For faint sources the quasars provide the most direct way to estimate parallax bias. In order to extend this to brighter sources and a broader range of colours, we use differential methods based on physical pairs (binaries) and sources in the Large Magellanic Cloud. The functional forms of the dependencies are explored by mapping the systematic differences between EDR3 and DR2 parallaxes. The parallax bias is found to depend in a non-trivial way on (at least) the magnitude, colour, and ecliptic latitude of the source. Different dependencies apply to the five- and six-parameter solutions in EDR3. While it is not possible to derive a definitive recipe for the parallax correction, we give tentative expressions to be used at the researcher's discretion and point out some possible paths towards future improvements., Comment: 32 pages, 30 figures. Abstract: Context. Gaia Early Data Release 3 (Gaia EDR3) provides accurate astrometry for about 1.6 million compact (QSO-like) extragalactic sources, 1.2 million of which have the best-quality five-parameter astrometric solutions., Aims. The proper motions of QSO-like sources are used to reveal a systematic pattern due to the acceleration of the solar systembarycentre with respect to the rest frame of the Universe. Apart from being an important scientific result by itself, the acceleration measured in this way is a good quality indicator of the Gaia astrometric solution., Methods. Theeffect of the acceleration was obtained as a part of the general expansion of the vector field of proper motions in vector spherical harmonics (VSH). Various versions of the VSH fit and various subsets of the sources were tried and compared to get the most consistent result and a realistic estimate of its uncertainty. Additional tests with the Gaia astrometric solution were used to get a better idea of the possible systematic errors in the estimate., Results. Our best estimate of the acceleration based on Gaia EDR3 is (2.32 +/- 0.16) x 10(-10) m s(-2) (or 7.33 +/- 0.51 km s(-1) Myr-1) towards alpha = 269.1 degrees +/- 5.4 degrees, delta = -31.6 degrees +/- 4.1 degrees, corresponding to a proper motion amplitude of 5.05 +/- 0.35 mu as yr(-1). This is in good agreement with the acceleration expected from current models of the Galactic gravitational potential. We expect that future Gaia data releases will provide estimates of the acceleration with uncertainties substantially below 0.1 mu as yr(-1).

Abstract: The solar neighborhood contains disc stars that have recently crossed spiral arms in the Galaxy. We propose that boundaries in local velocity distributions separate stars that have recently crossed or been perturbed by a particular arm from those that haven't. Ridges in the stellar velocity distributions constructed from the second Gaia data release trace orbits that could have touched nearby spiral arms at apocentre or pericentre. Abstract: [Context] The physical processes driving the formation of Galactic spiral arms are still under debate. Studies using open clusters favour the description of the Milky Way spiral arms as long-lived structures following the classical density wave theory. Current studies comparing the Gaia DR2 field stars kinematic information of the solar neighbourhood to simulations, find a better agreement with short-lived arms with a transient behaviour. [Aims] Our aim is to provide an observational, data-driven view of the Milky Way spiral structure and its dynamics using open clusters as the main tracers, and to contrast it with simulation-based approaches. We used the most complete catalogue of Milky Way open clusters, with astrometric Gaia EDR3 updated parameters, estimated astrophysical information, and radial velocities, to revisit the nature of the spiral pattern of the Galaxy. [Methods] We used a Gaussian mixture model to detect overdensities of open clusters younger than 30 Myr that correspond to the Perseus, Local, Sagittarius, and Scutum spiral Arms, respectively. We used the birthplaces of the open cluster population younger than 80 Myr to trace the evolution of the different spiral arms and compute their pattern speed. We analysed the age-distribution of the open clusters across the spiral arms to explore the differences in the rotational velocity of stars and spiral arms. [Results] We are able to increase the range in Galactic azimuth where present-day spiral arms are described, better estimating its parameters by adding 264 young open clusters to the 84 high-mass star-forming regions used so far, thus increasing the number of tracers by 314%. We used the evolution of the open clusters from their birth positions to find that spiral arms nearly co-rotate with field stars at any given radius, discarding a common spiral pattern speed for the spiral arms explored. [Conclusions] The derivation of different spiral pattern speeds for the different spiral arms disfavours classical density waves as the main drivers for the formation of the Milky Way spiral structure, and it is in better agreement with simulation-based approaches that tend to favour transient spirals. The increase in the number of known open clusters, as well as in their derived properties, allows us to use them as effective spiral structure tracers and homogenise the view from open clusters and field stars on the nature of the Galactic spiral arms.

Abstract: Plasmodium falciparum mediates adhesion of infected red blood cells (RBCs) to blood vessel walls by assembling a multi-protein complex at the RBC surface. This virulence-mediating structure, called the knob, acts as a scaffold for the presentation of the major virulence antigen, P. falciparum Erythrocyte Membrane Protein-1 (PfEMP1). In this work we developed correlative STochastic Optical Reconstruction Microscopy–Scanning Electron Microscopy (STORM-SEM) to spatially and temporally map the delivery of the knob-associated histidine-rich protein (KAHRP) and PfEMP1 to the RBC membrane skeleton. We show that KAHRP is delivered as individual modules that assemble in situ, giving a ring-shaped fluorescence profile around a dimpled disk that can be visualized by SEM. Electron tomography of negatively-stained membranes reveals a previously observed spiral scaffold underpinning the assembled knobs. Truncation of the C-terminal region of KAHRP leads to loss of the ring structures, disruption of the raised disks and aberrant formation of the spiral scaffold, pointing to a critical role for KAHRP in assembling the physical knob structure. We show that host cell actin remodeling plays an important role in assembly of the virulence complex, with cytochalasin D blocking knob assembly. Additionally, PfEMP1 appears to be delivered to the RBC membrane, then inserted laterally into knob structures., Author summary The human malaria parasite Plasmodium falciparum causes severe disease, which is initiated by the adhesion of parasite-infected RBCs to receptors on the walls of the host’s capillaries. Adhesion is mediated by a structure called the knob, which acts as a scaffold for the presentation of the virulence protein, P. falciparum erythrocyte membrane protein-1 (PfEMP1). In this work we investigate the assembly of this complex at different stages of parasite development using a multimodal imaging approach that combines dSTORM localization microscopy and scanning electron microscopy (STORM-SEM). We show that the knob-associated histidine-rich protein (KAHRP) is delivered to the RBC membrane skeleton as individual protein modules that assemble into a ring-shaped complex. We provide evidence that host cell remodeling, driven by association of KAHRP with spectrin and the reorganization of actin, is required for assembly of the ring complex, which in turn supports a spiral scaffold that is required for correct knob morphology. Finally, we provide evidence that PfEMP1 is delivered to the RBC membrane before associating with knob complexes.

Abstract: Summary: Increased tolerance of Plasmodium falciparum to front-line artemisinin antimalarials (ARTs) is associated with mutations in Kelch13 (K13), although the precise role of K13 remains unclear. Here, we show that K13 mutations result in decreased expression of this protein, while mislocalization of K13 mimics resistance-conferring mutations, pinpointing partial loss of function of K13 as the relevant molecular event. K13-GFP is associated with ∼170 nm diameter doughnut-shaped structures at the parasite periphery, consistent with the location and dimensions of cytostomes. Moreover, the hemoglobin-peptide profile of ring-stage parasites is reduced when K13 is mislocalized. We developed a pulse-SILAC approach to quantify protein turnover and observe less disruption to protein turnover following ART exposure when K13 is mislocalized. Our findings suggest that K13 regulates digestive vacuole biogenesis and the uptake/degradation of hemoglobin and that ART resistance is mediated by a decrease in heme-dependent drug activation, less proteotoxicity, and increased survival of parasite ring stages. : Artemisinin resistance in P. falciparum is associated with Kelch13 (K13) mutations. Yang et al. report that K13 is located in cytostome-like structures, consistent with a role in hemoglobin uptake. K13 is less abundant in mutants, and hemoglobin catabolism is impaired. Abstract: Introduction: \ud Frozen shoulder is a common, fibro-proliferative disease characterised by the insidious onset of pain and progressively restricted range of shoulder movement.\ud \ud Despite the prevalence of this disease, there is limited understanding of the molecular mechanisms underpinning the pathogenesis of this debilitating disease. Previous studies have identified increased myofibroblast differentiation and proliferation, immune cell influx and dysregulated cytokine production. We hypothesised that subpopulations within the fibroblast compartment may take on an activated phenotype, thus initiating the inflammatory processes observed in frozen shoulder. Therefore, we sought to evaluate the presence and possible pathogenic role of known stromal activation proteins in Frozen shoulder,\ud \ud Methods: \ud Shoulder capsule samples were collected from 10 patients with idiopathic frozen shoulder and 10 patients undergoing shoulder stabilisation surgery. Fibroblast activation marker expression (CD248, CD146, VCAM and PDPN, FAP) was quantified using immunohistochemistry. Control and diseased fibroblasts were cultured for in vitro studies from capsule biopsies from instability and frozen shoulder surgeries, respectively. The inflammatory profile and effects of IL-1β upon diseased and control fibroblasts was assessed using ELISA, immunohistochemistry and qPCR.\ud \ud Results: \ud Immunohistochemistry demonstrated increased expression of fibroblast activation markers CD248, CD146, VCAM and PDPN in the frozen shoulder group compared with control (p < 0.05). Fibroblasts cultured from diseased capsule produced elevated levels of inflammatory protein (IL-6, IL-8 & CCL-20) in comparison to control fibroblasts. Exposing control fibroblasts to an inflammatory stimuli, (IL-1ß) significantly increased stromal activation marker transcript and protein expression (CD248, PDPN and VCAM).\ud \ud Conclusions: \ud These results show that fibroblasts have an activated phenotype in frozen shoulder and this is associated with inflammatory cytokine dysregulation. Furthermore, it supports the hypothesis that activated fibroblasts may be involved in regulating the inflammatory and fibrotic processes involved in this disease.

Abstract: We derive Bayesian distances for all stars in the RV sample of Gaia DR2, and use the statistical method of Schoenrich, Binney & Asplund(2012) to validate the distances and test the Gaia parallaxes. In contrast to other methods, which rely on special sources, our method directly tests the distances to all stars in our sample. We find clear evidence for a near-linear trend of distance bias f with distance s, proving a parallax offset delta p. On average, we find delta p = -0.054 mas (parallaxes in Gaia DR2 need to be increased) when accounting for the parallax uncertainty under-estimate in the Gaia set (delta p = -0.048 mas on the raw parallax errors) with negligible formal error and a systematic uncertainty of about 0.006 mas. The value is in concordance with results from asteroseismic measurements, but differs from the much lower bias found on quasar samples. We further use our method to compile a comprehensive set of quality cuts in colour, apparent magnitude, and astrometric parameters. Last, we find that for this sample delta p appears to strongly depend on the parallax uncertainty sigma_p (when including the additional 0.043 mas) with a statistical confidence far in excess of 10\sigma and a proportionality factor close to 1, though the dependence varies somewhat with sigma_p. Correcting for the sigma_p dependence also resolves otherwise unexplained correlations of the offset with the number of observation periods n_{vis} and ecliptic latitude. Every study using Gaia DR2 parallaxes/distances should investigate the sensitivity of their results on the parallax biases described here and - for fainter samples - in the DR2 astrometry paper., Comment: 14 pages, 13 figures, accepted in MNRAS. Abstract: We present evidence from the RAdial Velocity Experiment (RAVE) survey of chemically separated, kinematically distinct disc components in the solar neighbourhood. We apply probabilistic chemical selection criteria to separate our sample into $\alpha$-low (`thin disc') and $\alpha$-high (`thick disc') sequences. Using newly derived distances, which will be utilized in the upcoming RAVE DR5, we explore the kinematic trends as a function of metallicity for each of the disc components. For our $\alpha$-low disc, we find a negative trend in the mean rotational velocity ($V_{\mathrm{\phi}}$) as a function of iron abundance ([Fe/H]). We measure a positive gradient $\partial V_{\mathrm{\phi}}$/$\partial$[Fe/H] for the $\alpha$-high disc, consistent with results from high-resolution surveys. We also find differences between the $\alpha$-low and $\alpha$-high discs in all three components of velocity dispersion. We discuss the implications of an $\alpha$-low, metal-rich population originating from the inner Galaxy, where the orbits of these stars have been significantly altered by radial mixing mechanisms in order to bring them into the solar neighbourhood. The probabilistic separation we propose can be extended to other data sets for which the accuracy in [$\alpha$/Fe] is not sufficient to disentangle the chemical disc components a priori. For such datasets which will also have significant overlap with Gaia DR1, we can therefore make full use of the improved parallax and proper motion data as it becomes available to investigate kinematic trends in these chemical disc components., Comment: 11 pages, 7 figures, accepted for publication in MNRAS

Abstract: IntroductionFrozen shoulder is a common, fibro-proliferative disease characterised by the insidious onset of pain and progressively restricted range of shoulder movement. Despite the prevalence of this disease, there is limited understanding of the molecular mechanisms underpinning the pathogenesis of this debilitating disease. Previous studies have identified increased myofibroblast differentiation and proliferation, immune cell influx and dysregulated cytokine production. We hypothesised that subpopulations within the stromal compartment may take on an activated phenotype, thus initiating the inflammatory processes observed in frozen shoulder. Therefore, we sought to evaluate the presence and possible pathogenic role of known stromal activation proteins in Frozen shoulder,MethodsShoulder capsule samples were collected from 10 patients with idiopathic frozen shoulder and 10 patients undergoing shoulder stabilisation surgery. Stromal activation marker expression (CD248, CD146, VCAM and PDPN, FAP) was quantified using immunohistochemistry. Control and diseased fibroblasts were cultured for in vitro studies from capsule biopsies from instability and frozen shoulder surgeries, respectively. The inflammatory profile and effects of IL-1β upon diseased and control fibroblasts was assessed using ELISA, immunohistochemistry and qPCR.ResultsImmunohistochemistry demonstrated increased expression of stromal activation markers CD248, CD146, VCAM and PDPN in the frozen shoulder group compared with control (p < 0.05). Fibroblasts cultured from diseased capsule produced elevated levels of inflammatory protein (IL-6, IL-8 & CCL-20) in comparison to control fibroblasts. Exposing control fibroblasts to an inflammatory stimuli, (IL-1ß) significantly increased stromal activation marker transcript and protein expression (CD248, PDPN and VCAM).ConclusionsThese results show that stromal fibroblasts have an activated phenotype in frozen shoulder and this is associated with inflammatory cytokine dysregulation. Furthermore, it supports the hypothesis that activated stromal fibroblasts may be involved in regulating the inflammatory and fibrotic processes involved in this disease.

Abstract: We combine parallaxes from the first Gaia data release with the spectrophotometric distance estimation framework for stars in the fifth RAVE survey data release. The combined distance estimates are more accurate than either determination in isolation - uncertainties are on average two times smaller than for RAVE-only distances (three times smaller for dwarfs), and 1.4 times smaller than TGAS parallax uncertainties (two times smaller for giants). We are also able to compare the estimates from spectrophotometry to those from Gaia, and use this to assess the reliability of both catalogues and improve our distance estimates. We find that the distances to the lowest log g stars are, on average, overestimated and caution that they may not be reliable. We also find that it is likely that the Gaia random uncertainties are smaller than the reported values. As a byproduct we derive ages for the RAVE stars, many with relative uncertainties less than 20 percent. These results for 219566 RAVE sources have been made publicly available, and we encourage their use for studies that combine the radial velocities provided by RAVE with the proper motions provided by Gaia. A sample that we believe to be reliable can be found by taking only the stars with the flag notification flag_any=0, MNRAS accepted. Updated to final version, reflecting referee reports. Abstract: We explore the connections between stellar age, chemistry, and kinematics across a Galactocentric distance of $7.5 < R\,(\mathrm{kpc}) < 9.0$, using a sample of $\sim 12\,000$ intermediate-mass (FGK) turnoff stars observed with the RAdial Velocity Experiment (RAVE) survey. The kinematics of this sample are determined using radial velocity measurements from RAVE, and parallax and proper motion measurements from the Tycho-Gaia Astrometric Solution (TGAS). In addition, ages for RAVE stars are determined using a Bayesian method, taking TGAS parallaxes as a prior. We divide our sample into young ($0 < ��< 3$ Gyr) and old ($8 < ��< 13$ Gyr) populations, and then consider different metallicity bins for each of these age groups. We find significant differences in kinematic trends of young and old, metal-poor and metal-rich, stellar populations. In particular, we find a strong metallicity dependence in the mean Galactocentric radial velocity as a function of radius ($\partial {V_{\rm R}}/\partial R$) for young stars, with metal-rich stars having a much steeper gradient than metal-poor stars. For $\partial {V_��}/\partial R$, young, metal-rich stars significantly lag the LSR with a slightly positive gradient, while metal-poor stars show a negative gradient above the LSR. We interpret these findings as correlations between metallicity and the relative contributions of the non-axisymmetries in the Galactic gravitational potential (the spiral arms and the bar) to perturb stellar orbits., 14 pages, 13 figures, accepted for publication in MNRAS

Abstract: The early stages of a galaxy's evolution leave an imprint on its metallicity distribution. We discuss the origins and evolution of radial metallicity gradients in discs of spiral galaxies using an analytical chemical evolution model. We explain how radial metallicity gradients in stellar populations are determined by three factors: the radial metallicity profile of the star-forming ISM, radial changes in the star-formation history (in particular inside-out formation), and radial mixing of stars. Under reasonable assumptions, inside-out formation steepens the negative ISM metallicity gradient, but contributes positively to the stellar metallicity gradient, up to inverting the metallicity profile to a positive d[Fe/H]/dR. This reconciles steep negative d[Fe/H]/dR in some high redshift galaxies to generally flatter gradients in local observations. We discuss the evidence for inverse radial metallicity gradients (positive d[X/H]/dR) at high redshifts and the inverse relationship between azimuthal velocity and the metallicity (positive dV_{\phi}/d[Fe/H]) of stars for the Milky Way's thick disc. The former can be achieved by high central gas-loss rates and re-distribution processes, e.g. re-accretion of enriched material in conjunction with inside-out formation, and near-disc galactic fountaining. For the Milky Way thick disc, we show that the positive dV_{\phi}/d[Fe/H] correlation points to comparable timescales for inside-out formation, initial metal enrichment and SNIa enrichment. We argue that the original ISM metallicity gradient could be inferred with better data from the high-metallicity tail of the alpha enhanced population. Abstract: We use data from the Radial Velocity Experiment (RAVE) and the Tycho-Gaia astrometric solution catalogue (TGAS) to compute the velocity fields yielded by the radial (VR), azimuthal (Vphi) and vertical (Vz) components of associated Galactocentric velocity. We search in particular for variation in all three velocity components with distance above and below the disc midplane, as well as how each component of Vz (line-of-sight and tangential velocity projections) modifies the obtained vertical structure. To study the dependence of velocity on proper motion and distance we use two main samples: a RAVE sample including proper motions from the Tycho-2, PPMXL and UCAC4 catalogues, and a RAVE-TGAS sample with inferred distances and proper motions from the TGAS and UCAC5 catalogues. In both samples, we identify asymmetries in VR and Vz. Below the plane we find the largest radial gradient to be dVR / dR = -7.01+- 0.61 km\s kpc, in agreement with recent studies. Above the plane we find a similar gradient with dVR / dR= -9.42+- 1.77 km\s kpc. By comparing our results with previous studies, we find that the structure in Vz is strongly dependent on the adopted proper motions. Using the Galaxia Milky Way model, we demonstrate that distance uncertainties can create artificial wave-like patterns. Abstract: Pore forming proteins such as those belonging to the membrane attack/perforin (MACPF) family have important functions in many organisms. Of the five MACPF proteins found in Plasmodium parasites, three have functions in cell passage and one in host cell egress. Here we report an analysis of the perforin-like protein 4, PPLP4, in the rodent parasite Plasmodium berghei. We found that the protein is expressed only in the ookinete, the invasive stage of the parasite formed in the mosquito midgut. Transcriptional analysis revealed that expression of the pplp4 gene commences during ookinete development. The protein was detected in retorts and mature ookinetes. Using two antibodies, the protein was found localized in a dotted pattern, and 3-D SIM super-resolution microcopy revealed the protein in the periphery of the cell. Analysis of a C-terminal mCherry fusion of the protein however showed mainly cytoplasmic label. A pplp4 null mutant formed motile ookinetes, but these were unable to invade and traverse the midgut epithelium resulting in severely impaired oocyst formation and no transmission to naïve mice. However, when in vitro cultured ookinetes were injected into the thorax of the mosquito, thus by-passing midgut passage, sporozoites were formed and the mutant parasites were able to infect naïve mice. Taken together, our data show that PPLP4 is required only for ookinete invasion of the mosquito midgut. Thus PPLP4 has a similar role to the previously studied PPLP3 and PPLP5, raising the question why three proteins with MACPF domains are needed for invasion by the ookinete of the mosquito midgut epithelium.

Abstract: Gaia Data Release 2 (Gaia DR2) contains results for 1693 million sources in the magnitude range 3 to 21 based on observations collected by the European Space Agency Gaia satellite during the first 22 months of its operational phase. We describe the input data, models, and processing used for the astrometric content of Gaia DR2, and the validation of these results performed within the astrometry task. Some 320 billion centroid positions from the pre-processed astrometric CCD observations were used to estimate the five astrometric parameters (positions, parallaxes, and proper motions) for 1332 million sources, and approximate positions at the reference epoch J2015.5 for an additional 361 million mostly faint sources. Special validation solutions were used to characterise the random and systematic errors in parallax and proper motion. For the sources with five-parameter astrometric solutions, the median uncertainty in parallax and position at the reference epoch J2015.5 is about 0.04 mas for bright (G, 25 pages, 29 figures. Abstract: We present a freely downloadable software package for modelling the dynamics of galaxies, which we call the Torus Mapper (TM). The package is based around `torus mapping', which is a non-perturbative technique for creating orbital tori for specified values of the action integrals. Given an orbital torus and a star's position at a reference time, one can compute its position at any other time, no matter how remote. One can also compute the velocities with which the star will pass through any given point and the contribution it will make to the time-averaged density there. A system of angle-action coordinates for the given potential can be created by foliating phase space with orbital tori. Such a foliation is facilitated by the ability of TM to create tori by interpolating on a grid of tori. We summarise the advantages of using TM rather than a standard time-stepper to create orbits, and give segments of code that illustrate applications of TM in several contexts, including setting up initial conditions for an N-body simulation. Abstract: The malaria parasite Plasmodium falciparum dramatically remodels its host red blood cell to enhance its own survival, using a secretory membrane system that it establishes outside its own cell. Cisternal organelles, called Maurer's clefts, act as a staging point for the forward trafficking of virulence proteins to the red blood cell (RBC) membrane. The Ring-EXported Protein-1 (REX1) is a Maurer's cleft resident protein. We show that inducible knockdown of REX1 causes stacking of Maurer's cleft cisternae without disrupting the organization of the knob-associated histidine-rich protein at the RBC membrane. Genetic dissection of the REX1 sequence shows that loss of a repeat sequence domain results in the formation of giant Maurer's cleft stacks. The stacked Maurer's clefts are decorated with tether-like structures and retain the ability to dock onto the RBC membrane skeleton. The REX1 mutant parasites show deficient export of the major virulence protein, PfEMP1, to the red blood cell surface and markedly reduced binding to the endothelial cell receptor, CD36. REX1 is predicted to form a largely α-helical structure, with a repetitive charge pattern in the repeat sequence domain, providing potential insights into the role of REX1 in Maurer's cleft sculpting.

Abstract: We present the identification of potential members of nearby Galactic globular clusters using radial velocities from the RAdial Velocity Experiment Data Release 4 (RAVE-DR4) survey database. Our identifications are based on three globular clusters -- NGC 3201, NGC 5139 ($\omega$ Cen) and NGC 362 -- all of which are shown to have |RV|>100 km/s. The identification of globular cluster stars in RAVE DR4 data offers a unique opportunity to test the precision and accuracy of the stellar parameters determined with the currently available Stellar Parameter Pipelines (SPPs) used in the survey, as globular clusters are ideal testbeds for the validation of stellar atmospheric parameters, abundances, distances and ages. For both NGC 3201 and $\omega$ Cen, there is compelling evidence for numerous members (> 10) in the RAVE database; in the case of NGC 362 the evidence is more ambiguous, and there may be significant foreground and/or background contamination in our kinematically-selected sample. A comparison of the RAVE-derived stellar parameters and abundances with published values for each cluster and with BASTI isochrones for ages and metallicities from the literature reveals overall good agreement, with the exception of the apparent underestimation of surface gravities for giants, in particular for the most metal-poor stars. Moreover, if the selected members are part of the main body of each cluster our results would also suggest that the distances from Binney et al. 2013, where only isochrones more metal-rich than -0.9 dex were used, are typically underestimated by ~ 40% with respect to the published distances for the clusters, while the distances from Zwitter et al. 2010 show stars ranging from 1 to ~ 6.5 kpc -- with indications of a trend toward higher distances at lower metallicities -- for the three clusters analysed in this study., Comment: Accepted for publication MNRAS, 3 April 2015

Abstract: We apply the twin method to determine parallaxes to 232,545 stars of the RAVE survey using the parallaxes of Gaia DR1 as a reference. To search for twins in this large dataset, we apply the t-stochastic neighbour embedding t-SNE projection which distributes the data according to their spectral morphology on a two dimensional map. From this map we choose the twin candidates for which we calculate a chi^2 to select the best sets of twins. Our results show a competitive performance when compared to other model-dependent methods relying on stellar parameters and isochrones. The power of the method is shown by finding that the accuracy of our results is not significantly affected if the stars are normal or peculiar since the method is model free. We find twins for 60% of the RAVE sample which is not contained in TGAS or that have TGAS uncertainties which are larger than 20%. We could determine parallaxes with typical errors of 28%. We provide a complementary dataset for the RAVE stars not covered by TGAS, or that have TGAS uncertainties which are larger than 20%, with model-free parallaxes scaled to the Gaia measurements., MNRAS accepted
