Abstract

Wheat is an important food crop worldwide. Climate change, disease and abiotic stresses can decrease yield and reduce grain quality, therefore search for useful alleles in wild accessions and subspecies could improve resilience and sustainability. Two genetic resources useful for studying grain quality-related traits were analyzed and characterized in the frame of the CEREALMED project. An introgression line population obtained by crossing a durum wheat cultivar, PR22D89, and an accession of Triticum dicoccoides, MG2323, was evaluated for protein content, SDS-sedimentation volume, beta-glucans and carotenoids in two different pedoclimatic environments to find genetic loci involved in these traits. Furthermore, a collection of 59 accessions of Triticum aestivum subspecies, including cultivars, breeding materials, landraces and wilds, previously characterized by SNP markers, was evaluated for carotenoid content. The variability present in this panel was partially overlapped with that observed in durum wheat and therefore useful in enlarging the total variation for wheat. Moreover, significant SNP markers putatively associated to this trait in common wheat were identified, providing the chance to identify novel useful alleles to further improve carotenoid content in both durum and bread wheat.

Abstract

Anche nel 2020-21, pur nelle difficili circostanze determinate per il secondo anno consecutivo dalla pandemia da Covid-19, sono state condotte in Sicilia le prove della Rete nazionale di confronto varietale di frumento duro. I campi sperimentali sono stati allestiti in 5 ambienti siciliani. La sperimentazione ha riguardato 30 varieta’, di cui ben 9 al primo anno di valutazione. I risultati produttivi riscontrati nelle diverse localita’ appaiono molto differenti e fortemente influenzati dall’andamento meteorologico verificatosi nelle diverse zone dell’Isola. At 2020-21, despite the difficult circumstances determined for the second consecutive year by the Covid-19 pandemic, the tests of the national durum wheat variety comparison network were conducted in Sicily. The experimental fields were set up in 5 Sicilian environments. The experimentation involved 30 varieties, of which 9 in the first year of evaluation. The production results found in the different locations appear very different and strongly influenced by the meteorological trends that occurred in the different areas of the island.

Abstract

Per valorizzare la filiera del grano duro italiano non va trascurata la formulazione e il continuo aggiornamento di liste di orientamento varietale attraverso la Rete nazionale di confronto varietale. I potenziali ampi margini di crescita per il settore primario nazionale, al servizio e al traino delle grandi performance industriali e commerciali delle imprese della trasformazione, sono possibili se si ottimizzano stabilmente i parametri qualitativi e igienico-sanitari delle produzioni e si lavora ad una maggiore concentrazione dell’offerta. La Rete nazionale di confronto varietale del grano duro, realizzata mediante prove svolte con protocolli comuni in ambienti rappresentativi delle diverse regioni, permette di acquisire e divulgare in maniera tempestiva le caratteristiche produttive, qualitative e di sensibilità alle principali fitopatie di numerose cultivar, per valutare il loro adattamento nei diversi areali anche al variare dell’andamento climatico negli anni. Ciò assicura un’informazione indipendente e su scala nazionale utile agli agricoltori nella scelta della cultivar più idonea, di supporto al ricambio varietale e in grado di assecondare le richieste di produttori e trasformatori e quindi, alla fine, dei consumatori. Nell'articolo vengono riportati i dati quanti-qualitativi relativi all’annata agraria 2018-19 durante la quale sono stati realizzati 31 campi di confronto tra varietà di frumento duro in regime convenzionale, dislocati in 14 regioni, e 7 campi in coltura biologica, localizzati in 5 regioni del Centro-Sud. Le varietà complessivamente saggiate sono state complessivamente 38 in convenzionale e 17 in biologico.

Abstract

Leaf stripe, caused by the fungus Pyrenophora graminea Ito et Kuribayashi [anamorph Drechslera graminea (Rabenh. ex. Schlech.) Shoemaker], is an important seed-borne disease of barley (Hordeum vulgare L.). The objective of this study was to verify the reliability of a PCR-based marker (MWG2018) associated with the resistance gene Rdg2a and to assess the leaf stripe resistant phenotype in barley breeding programs. A large number of barley cultivars and accessions were thus evaluated for their reaction to a highly virulent monoconidial isolate (Dg2) of the pathogen and genotyped for the allele of the molecular marker. Several resistant genotypes were identified and four were shown to possess the same allele as the cultivar Thibaut (the resistant parent of the original mapping population in which Rdg2a was identified) at the marker locus. One of them, cv. Rebelle, is being used as a source of leaf stripe resistance in winter barley breeding programs. The allelic composition at the MWG2018 locus was verified in several resistant lines bred from five crosses, in which Rebelle acted directly or indirectly as donor of the resistance. The results showed that the resistant phenotype of the lines was always associated with the resistance allele of the molecular marker, thereby demonstrating its reliability for selecting leaf stripe resistance. Abstract

Due to the overlap of many disciplines and the availability of novel technologies, modern agriculture has become a wide, interdisciplinary endeavor, especially in Precision Agriculture. The adoption of a standard format for reporting field experiments can help researchers to focus on the data rather than on re-formatting and understanding the structure of the data. This paper describes how a European consortium plans to: i) create a “handbook” of protocols for reporting definitions, methodologies and Parameters measured/calculated; and ii) how a data-template for field data was created and will be linked to the “handbook”. The overall goal of the EU-funded project Solutions for Solutions for improving Agroecosystem and Crop Efficiency for water and nutrient use (SolACE) is to help European agriculture face major challenges, such as increased rainfall variability and reduced use of N and P fertilizers in order to satisfy both economic and ecological goals. The “Handbook of Protocols” and the “Data Template” have been created to achieve a flexible, standard, and clear documentation linked with the data itself to facilitate interchange of data among project’s partners and any statistical analysis and modelling of different datasets.

Abstract

Background: Frost tolerance is a key trait with economic and agronomic importance in barley because it is a major component of winter hardiness, and therefore limits the geographical distribution of the crop and the effective transfer of quality traits between spring and winter crop types. Three main frost tolerance QTL (Fr-H1, Fr-H2 and Fr-H3) have been identified from bi-parental genetic mapping but it can be argued that those mapping populations only capture a portion of the genetic diversity of the species. A genetically broad dataset consisting of 184 genotypes, representative of the barley gene pool cultivated in the Mediterranean basin over an extended time period, was genotyped with 1536 SNP markers. Frost tolerance phenotype scores were collected from two trial sites, Foradada (Spain) and Fiorenzuola (Italy) and combined with the genotypic data in genome wide association analyses (GWAS) using Eigenstrat and kinship approaches to account for population structure. Results: GWAS analyses identified twelve and seven positive SNP associations at Foradada and Fiorenzuola, respectively, using Eigenstrat and six and four, respectively, using kinship. Linkage disequilibrium analyses of the significant SNP associations showed they are genetically independent. In the kinship analysis, two of the significant SNP associations were tightly linked to the Fr-H2 and HvBmy loci on chromosomes 5H and 4HL, respectively. The other significant kinship associations were located in genomic regions that have not previously been associated with cold stress Conclusions: Haplotype analysis revealed that most of the significant SNP loci are fixed in the winter or facultative types, while they are freely segregating within the un-adapted spring barley genepool. Abstract

The determinants of barley grain yield in drought-prone Mediterranean environments have been studied in the Nure x Tremois (NT) population. A large set of yield and other morpho-physiological data were recorded in 118 doubled-haploid lines of the population, in multi-environment field trials (18 site-year combination). Agrometeorological variables have been recorded and calculated at each site too. Four main periods of barley development were considered, vegetative, reproductive early and late grain filling phases, to dissect the effect on yield traits of the growth phases. Relationships between agrometeorological variables, grain yield (GY) and its main components (GN and GW) were also investigated by correlation. Results firstly gave a clear indication of the involvement of water consumption in determining GY and GW (r2=0.616, P=0.007 and r2=0.703, P=0.005, respectively) calculated from sowing to the early grain filling period, while GN showed its highest correlation with the total photothermal quotient (PQ) calculated for the same period (r2=0.646, P=0.013). With the only exception of total PQ calculated during the vegetative period, all significant correlations with GY were associated to water-dependent agrometeorological parameters. As a second result, the NT segregating population allowed us to weight the amount of interaction due to genotypes over environments or to environments in relation to genotypes by a GGE analysis; 47.67% of G+GE sum of squares was explained by the first two principal components. Then, the introduction of genomic information at major barley genes regulating the length of growth cycle allowed us to explain patterns of adaptation of different groups of NT lines according to the variants (alleles) harbored at venalization (Vrn-H1) in combination with earliness (Eam6) genes. Abstract

An association panel consisting of 185 accessions representative of the barley germplasm cultivated in the Mediterranean basin was used to localise quantitative trait loci (QTL) controlling grain yield and yield related traits. The germplasm set was genotyped with 1,536 SNP markers and tested for associations with phenotypic data gathered over 2 years for a total of 24 year × location combinations under a broad range of environmental conditions. Analysis of multi-environmental trial (MET) data by fitting a mixed model with kinship estimates detected from two to seven QTL for the major components of yield including 1000 kernel weight, grains per spike and spikes per m2, as well as heading date, harvest index and plant height. Several of the associations involved SNPs tightly linked to known major genes determining spike morphology in barley (vrs1 and int-c). Similarly, the largest QTL for heading date co-locates with SNPs linked with eam6, a major locus for heading date in barley for autumn sown conditions. Co-localization of several QTL related to yield components traits suggest that major developmental loci may be linked to most of the associations. This study highlights the potential of association genetics to identify genetic variants controlling complex traits.
