We investigated the impact of a rumen-bypass protein (RBP) supplement on growth performance, plasma and urinary N (UN) concentration, hepatic mitochondrial protein complexes, and hepatic mRNA expression of immune genes of beef steers with negative or positive residual feed intake (RFI) phenotype. Forty crossbred beef steers with an average body weight (BW) of 492 ± 36 kg were subjected to a generalized randomized block design over a 42-day experimental period. This study followed a 2 × 2 factorial arrangement of treatments. The factors evaluated were: 1) RFI classification (low-RFI (-2.12 kg/d) vs. high-RFI (2.02 kg/d), and 2) rumen-bypass protein supplement: RBP supplement (RBP; 227 g/steer/d) vs. control diet (CON; 0 g/d), resulting in four distinct treatments: LRFI-CON (n = 10), LRFI-RBP (n = 10), HRFI-CON (n = 10), and HRFI-RBP (n = 10). The RBP supplement (84% crude protein) is a mixture of hydrolyzed feather meal, porcine blood meal, and DL-methionine hydroxy analogue. The beef steers were stratified by BW, randomly assigned to treatments, and housed in four pens (1 treatment/pen) equipped with two GrowSafe feed bunks each to measure individual dry mater intake (DMI). Body weight was measured every 7 d. Liver tissue samples were collected on d 42 from all the beef steers. These samples were used for mRNA expression analysis of 16 immune-related genes and for evaluating the mitochondrial protein complexes I ‐ V. No significant effects due to RBP supplementation or RFI × RBP interactions (P > 0.05) were observed for average daily gain (ADG) and DMI. However, compared to high-RFI steers, low-RFI steers showed a trend towards reduced DMI (12.9 vs. 13.6 kg/d; P = 0.07) but ADG was similar for the two RFI groups. Regardless of RFI status, supplemental RBP increased blood urea nitrogen (BUN) (P = 0.01), with a lower BUN concentration in low-RFI steers compared to high-RFI ones. A tendency for interaction (P = 0.07) between RFI and RBP was detected for the UN concentrations; feeding the dietary RBP increased the UN concentration in high-RFI beef steers (209 vs. 124 mM), whereas the concentration was lower than that of the CON group for low-RFI beef steers (86 vs. 131 mM). Interactions of RBP and RFI were observed (P ≤ 0.05) for mitochondrial activities of complexes IV, V, and mRNA expressions of some immune genes such as TLR2, TLR3, and IL23A. In conclusion, while RBP supplementation did not alter growth performance, its observed effects on hepatic immune gene expression, mitochondrial protein complexes, BUN, and UN depended on the beef steers' RFI phenotype. Livestock management is challenging for resource-poor (R-P) farmers due to unavailability of quality feed, limited professional advice, and rumor-spreading about animal health condition in a herd. This research seeks to improve animal health in southern Africa by promoting sericea lespedeza (Lespedeza cuneata), a nutraceutical forage legume. An automated geospatial model for precision agriculture (PA) can identify suitable locations for its cultivation. Additionally, a novel approach of radio-frequency identifier (RFID) supported telemetry technology can track animal movement, and the analyses of data using artificial intelligence can determine sickness of small ruminants. This RFID-based system is being connected to a smartphone app (under construction) to alert farmers of potential livestock health issues in real time so they can take immediate corrective measures. An accompanying Decision Support System (DSS) site is being developed for R-P farmers to obtain all possible support on livestock production, including the designed PA and RFID-based DSS. Ticktrefoil (Desmodium) species from the USDA, ARS, Plant Genetic Resources Conservation Unit (PGRCU) germplasm collection in Griffin, GA, were evaluated for their potential livestock nutraceutical (nutritional + pharmaceutical) value in a field small plot study at the Fort Valley State University (FVSU) Agricultural Research Station, Fort Valley, GA. Horse marmalade (Desmodium discolor Vogel), greenleaf ticktrefoil (Desmodium intortum [Mill.] Urb.), panicledleaf ticktrefoil (Desmodium paniculatum [L.] DC.), Hawaii ticktrefoil (Desmodium sandwicense E. May) and twisted ticktrefoil (Desmodium tortuosum [Sw.] DC.) were established in a greenhouse and transplanted to field plots, with plant material from each species harvested, freeze-dried, ground and analyzed for nutrient content and bioactivity. Crude protein (CP) levels in D. discolor and D. tortuosum averaged 233 g/kg, whereas CP in D. intortum, D. paniculatum and D. sandwicense ranged from 99 to 157 g/kg. The Desmodium species had in vitro true digestibility values of 755 to 878 g/kg. There were no detectable condensed tannins in D. discolor and D. tortuosum, whereas D. intortum, D. paniculatum and D. sandwicense averaged 88, 89 and 73 g/kg total condensed tannin, respectively. There were species differences in flavonoid content (p < .01), with the highest delphinidin and cyanidin concentrations in D. sandwicense and D. paniculatum, respectively. There were no detectable levels of delphinidin or cyanidin for D. discolor or D. tortuosum. Quercitin and kaempferol concentrations were highest in D. intortum and D. paniculatum, respectively, whereas D. discolor and D. tortuosum had the lowest values. The highest protein precipitable phenolics and total phenolic concentrations were for D. paniculatum. Aflatoxin B1 (AFB1) is a mycotoxin known to impair human and animal health. It is also believed to have a deleterious effect on ruminal nutrient digestibility under in vitro batch culture systems. The objective of this study was to evaluate the effects of increasing the dose of AFB1 on ruminal dry matter and nutrient digestibility, fermentation profile, and N flows using a dual-flow continuous culture system fed a diet formulated for lactating dairy cows. Eight fermenter vessels were used in a replicated 4 × 4 Latin square design with 10 d periods (7 d adaptation and 3 d sample collection). Treatments were randomly applied to fermenters on diet DM basis: (1) 0 μg of AFB1/kg of DM (Control); (2) 50 μg of AFB1/kg of DM (AF50); (3) 100 μg of AFB1/kg of DM (AF100); and (4) 150 μg of AFB1/kg of DM (AF150). Treatments did not affect nutrient digestibility, fermentation, and N flows. Aflatoxin B1 concentration in ruminal fluid increased with dose but decreased to undetectable levels after 4 h post-dosing. In conclusion, adding incremental doses of AFB1 did not affect ruminal fermentation, digestibility of nutrients, and N flows in a dual-flow continuous culture system fed diets formulated for lactating dairy cows. In recent times, there has been a growing focus on the well-being and healthcare of small ruminants, particularly in relation to the issue of anemia due to infection with blood-feeding gastrointestinal nema-todes, such as Haemonchus contortus. The objective of this study was to assess the hematocrit levels in blood samples obtained from small ruminants, specifically goats. Additionally, the study is an attempt to design and create a quick sensor for identifying anemia, which could be conveniently used on farms. A total of 75 mature, intact male Spanish goats were subjected to hematocrit analysis to ascertain their spectrum of hematocrit values and association with anemic conditions. Simultaneously, a unique sensor with user-friendly features was developed to promptly provide farmers with feedback on the anemic status of animals using AI-based machine learning algorithms, enabling timely intervention. The sensor utilized a semi-invasive approach with minimal blood sample requirement. 30 ^L of blood was dropped on glycerol soaked Whatman filter paper No. 1, and images were taken at 90 sec, 150 sec, and 270 sec to capture the blood pattern on filter paper soaked in glycerol using a cell phone. These images were correlated with actual hematocrit value obtained from conventional hematocrit analysis. A RMSprop with adjusted learning rate convolution neural network (RMSprop-CNN) based images classification model was developed for the classification of different patterns at different levels of hematocrit. The model was trained on a total of 1,000 images with a training and testing split of 80:20 ratio. For the optimization process, the Adam optimizer with a learning rate of 0.001 is employed. The model is compiled with a categorical cross-entropy loss function, aiming to improve its accuracy metric over training iterations. The initial studies demonstrated a detection accuracy level of 70.31 % at 10 epochs in identifying different hematocrit levels (Level 1; healthy, to Level 5; severely anemic) but improved significantly up to 94.89 % by 100 epochs, resulting in a notable reduction in the time and level of knowledge previously necessary for conducting such evaluations. The present study not only provides valuable insights into the hematological characteristics of small ruminants, but also establishes a foundation for the development of a straightforward and efficient method for detecting anemia. The factors evaluated were 1) RFI classification [low-RFI (-2.12kg/d) vs. high-RFI (2.02kg/d)], and 2) feed additive: rumen-bypass protein supplement (RCP; 227 g· steer-1· day-1) vs. Control diet (CON; 0 g/d), resulting in four treatments: Low-RFI-CON (n = 10), low-RFI-RCP (n = 9), high-RFI-CON (n = 10), and high-RFI-RCP (n = 10). The rumen-bypass protein supplement (84% CP) is a blend of hydrolyzed feather meal, porcine blood meal, and DL-methionine hydroxy analogue. Liver tissue was sampled on d 42 from all animals using a liver biopsy needle for mRNA expression analysis of 84 immune genes and analysis of mitochondrial ETC activities of complexes I - V. No effects of supplemental feed additive or RFI × feed additive interactions were observed (P > 0.05) for ADG and DMI. Compared with high-RFI, low-RFI beef steers tended to have less DMI (13.6 vs 12.9 kg/d; P = 0.09) but ADG was similar for the two RFI groups. Feed additive × RFI interactions were observed (P = 0.05) for mitochondrial activities of Complexes IV, V, and mRNA expressions of some immune genes. Supplemental RCP increased Complex IV (39.2 nmol/min/mg) activity and reduced that of Complex V (94.1 nmol· min-1· mg-1) in high-RFI beef steers whereas the activity of Complex IV (13.1 nmol· min-1· mg-1) was reduced and that of Complex V (120 nmol· min-1· mg-1) was increased in low-RFI beef steers. Supplemental RCP increased (P < 0.05) the mRNA expression of TLR2 [fold change (FC) = 1.17 vs 0.98], TLR3 (FC = 1.49 vs 0.97), NFKB1 (FC = 1.02 vs 0.90), and IL23A (FC = 1.35 vs 1.07) in high-RFI beef steers but the expression of these genes were not altered in low-RFI beef steers. In conclusion, RCP supplementation did not affect the growth performance and the observed effects on hepatic immune gene expression and activities of mitochondrial ETC complexes are dependent on the RFI phenotype of the beef steers. We applied metabolomics and 16S rRNA gene sequencing to determine the metabolic pathway and ruminal bacterial taxa associated with divergent residual body weight (BW) gain phenotype in crossbred beef steers. A group of 108 crossbred growing beef steers (average BW = 282.87 ± 30 kg) were fed a forage-based diet for a period of 56 d in a confinement dry lot equipped with GrowSafe intake nodes to determine their residual body weight gain (RADG) phenotype. After RADG identification, blood and rumen fluid samples were collected from beef steers with the greatest RADG (most efficient; n = 20; 0.76 kg/d) and least RADG (least efficient; n = 20; -0.65 kg/d). Metabolome analysis of the plasma and rumen fluid samples was conducted using chemical isotope labelling/liquid chromatography-mass spectrometry. Differentially abundant metabolites in each of the plasma and rumen fluid samples between the two groups of beef steers were determined using a false discovery rate (FDR)-adjusted P-values ≤ 0.05 and area under the curve (AUC) > 0.80. Rumen and plasma metabolic pathways that were differentially enriched or depleted (P ≤ 0.05) in beef steers with positive RADG compared with those with negative RADG were determined by the quantitative pathway enrichment analysis. A total of 1,629 metabolites were detected and identified in the plasma of the beef steers; eight metabolites including alanyl-phenylalanine, 8-hydroxyguanosine, and slaframine were differentially abundant (FDR ≤ 0.05; AUC > 0.80) in beef steers with divergent RADG; five metabolic pathways including steroid hormone biosynthesis, thiamine metabolism, propanoate metabolism, pentose phosphate pathway, and butanoate metabolism were enriched (P ≤ 0.05) in beef steers with positive RADG, relative to negative RADG steers. A total of 1908 metabolites were detected and identified in the rumen of the beef steers; results of the pathway enrichment analysis of all the metabolites revealed no metabolic pathways in the rumen were altered (P = 0.05). The rumen fluid samples were also analyzed using 16S rRNA gene sequencing to assess the bacterial community composition. We compared the rumen bacterialcommunity composition at the genus level using a linear discriminant analysis effect size (LEfSe) to identify the differentially abundant taxa between the two groups of beef steers. The LEfSe results showed greater relative abundance of Bacteroidetes_vadinHA17 and Anaerovibrio in steers with positive RADG compared with the negative RADG group, while steers in the negative RADG group had greater relative abundance of Candidatus_Amoebophilus, Clostridium_sensu_ stricto_1, Pseudomonas, Empedobacter, Enterobacter, and Klebsiella compared with the positive RADG group. Our results demonstrate that beef steers with positive or negative RADG exhibit differences in plasma metabolic profiles and some ruminal bacterial taxa which probably explain their divergent feed efficiency phenotypes.
