The transcription factor Sox4 is vital for fetal development, as Sox4–/– homozygotes die in utero. Sox4 mRNA is expressed in the early embryonic growth plate and is regulated by parathyroid hormone, but its function in bone modeling/remodeling is unknown. We report that Sox4+/– mice exhibit significantly lower bone mass (by dual-energy X-ray absorptiometry) from an early age, and fail to obtain the peak bone mass of wild-type (WT) animals. Microcomputed tomography (μCT), histomorphometry and biomechanical testing of Sox4+/– bones show reduced trabecular and cortical thickness, growth plate width, ultimate force and stiffness compared with WT. Bone formation rate (BFR) in 3-month-old Sox4+/– mice is 64% lower than in WT. Primary calvarial osteoblasts from Sox4+/– mice demonstrate markedly inhibited proliferation, differentiation and mineralization. In these cultures, osterix (Osx) and osteocalcin (OCN) mRNA expression was reduced, whereas Runx2 mRNA was unaffected. No functional defects were found in osteoclasts. Silencing of Sox4 by siRNA in WT osteoblasts replicated the defects observed in Sox4+/– cells. We demonstrate inhibited formation and altered microarchitecture of bone in Sox4+/– mice versus WT, without apparent defects in bone resorption. Our results implicate the transcription factor Sox4 in regulation of bone formation, by acting upstream of Osx and independent of Runx2.

Sår som ikke vil gro er en av de største økonomiske utgiftene innen medisin i dag, og med en økning av den eldre befolkningen er det forventet at det vil skje en økning i pasienter med kroniske sår. Eggeskallmembranen som er et overflødig biprodukt fra eggeindustrien, har i flere hundre år blitt benyttet som en naturlig bandasje inne kinesisk medisin. Nylig er det også vitenskapelig bevist at eggeskallmembranen har fremmende effekt på sårhelingsprosessen. På grunnlag av dette er det ønskelig å utvikle nye og effektive sårhelingsprodukter som er basert på eggeskallmembran for å imøtekomme samfunnets behov. Hovedmålet med denne oppgaven er å forstå mekanismen og undersøke om industrielt oppskalert eggeskallmembranpulver (PEP) har en positiv virkning på fibroblastaktivitet som er viktig i en sårhelingsprosess. Effekten av derivater av eggeskallmembran, hydrolysert eggeskallmembranpulver (Hydrol) og karbohydrater fra eggeskallmembranpulver (Carb), med ulike sammensetninger og egenskaper ble også studert for å kunne skape et mest mulig effektivt sårhelingsprodukt. Humane dermale fibroblaster ble sådd ut i brønner hvor de etter et døgn ble stimulert med PEP, Hydrol og Carb i enten tre eller ti dager. Cellene ble deretter høstet, og ved hjelp av flere analysemetoder ble det sjekket om de ulike eggeskallmembranfraksjonene hadde påvirkning på ulike prosesser av fibroblastaktivitet. Prøvene ble analysert ved hjelp av real-time PCR, western blotting, zymografi, celleviabilitetsforsøk, proliferasjonsforsøk og fluorescens immuncytokjemi. Resultatene viser at tilsetting av PEP til cellekultur stimulerer økt grad av proliferasjon og sen differensiering av fibroblaster. PEP gir også en tidlig reorganisering av cellulære aktinfilamenter og reduksjon av cellefestepunkter, og dette tyder på effekt på migrering av fibroblaster. I tillegg gir PEP en økt aktivering av matriks degraderingsenzymet MMP-2, som spiller en viktig rolle i cellemigrering, uten å ha noen signifikant påvirkning på proteinnivå av enzymet og dens regulatorer, MMP-14 og TIMP-2. I likhet med PEP gir Hydrol en økt stimulering av proliferasjon og differensiering av fibroblaster, samt en økt aktivering av MMP-2. Til forskjell fra PEP økte også Hydrol proteinekspresjon av MMP-2 og TIMP-2. Carb har generell liten eller ingen påvirkning på disse prosessene. Carb har derimot i likhet med Hydrol, og i motsetning til PEP, en stimulerende effekt på økt produksjon av ekstracellulære matrikskomponenter. Resultatene fra denne masteroppgaven viser at eggeskallmembranpulver har en positiv effekt på fibroblastaktivitet som er viktig for sårheling, og at ESM-fraksjoner påvirker fibroblastaktiviteten forskjellig, trolig på grunn av ulike egenskaper. Delayed wound healing is one of the biggest economic expenses in medicine today, and with an increase of the elderly population, is it expected that there will be an increase in patients with chronic wounds. The eggshell membrane, a by-product of the egg industry, has been used for a hundred years as an alternative natural bandage in Chinese medicine. It has recently been scientifically demonstrated that the eggshell membrane has a beneficial effect on the wound healing process. Based on this, it is highly warrented to develop new and effective wound healing products based on eggshell membranes to meet the needs of the society. The aim of this thesis is to reveal whether industrial seperated eggshell membrane powder (PEP) intended for industrial production of a low cost wound healing product, has a positive effect on fibroblast activities important in wound healing process. The effect of derivatives of eggshell membranes, hydrolysed eggshell membrane powder (Hydrol) and carbohydrates from eggshell membrane powder (Carb), were also studied, in order for it to be able to reveal the most effective wound healing ingredient. Human dermal fibroblasts were seeded in culture wells and stimulated with PEP, Hydrol and Carb for either three or ten days. The cells were then harvested and investigated by different analytical and molecular methods whether the different eggshell membrane fractions affected fibroblast activities. Samples were analysed by real-time PCR, western blotting, zymografi, cell viability assay, proliferation assay and fluorescent immuncytochemistry. The results show that the supplement of PEP to fibroblast culture stimulates an increased rate of proliferation and late differentiation. PEP trigged also an early reorganization of cellular actin filaments and reduction of cellular attachment points, indicating an effect on fibroblast migration. In addition, PEP provides increased activation of the matrix degradation enzyme MMP-2, which is important for cell migration, without having any significant effect on the protein level of the enzyme and its regulators, MMP-14 and TIMP-2. Hydrol provides increased stimulation of proliferation and differentiation of fibroblasts, similar to PEP. Hydrol also stimulated an increased activation of MMP-2. Unlike PEP, Hydrol also increased the protein expression of MMP-2 and TIMP-2. Carb has generally little or no impact on these processes. Carb, on the other hand, like Hydrol, and unlike PEP, stimulate the increased production of extracellular matrix components. The results of this master thesis shows that PEP has a positive effect on fibroblast activity that is important for wound healing, and that ESM derived fractions affect the fibroblast activity differently, probably because of their different properties.
