Background Hidradenitis suppurativa (HS) is a chronic inflammatory and scarring disease with a wide spectrum of disease severity. The amount of scarring is proportional to the preceding tissue damage and poses a challenge to patients. Severe HS is most often treatment recalcitrant, but hypothetically avoidable through early biologic treatment. Early prediction of individual risk of disease progression is therefore essential for patient management. Objectives To investigate risk factors associated with disease progression and to design an algorithm capable of predicting disease -progression. Methods A prospective cohort study of 335 Hurley III-naïve patients with HS, not treated with biologics, was followed for a median of 2 years. Potential risk factors covered basic demographics, HS anamnestic factors and clinical HS factors collected during physical examination. Two separate Cox proportional hazard regression (CPHR) analyses were conducted. A summated 'progression score' was calculated and used in the predictive algorithm of severe disease. Subsequent bootstrap sampling was used to validate the predictability of the predictive algorithm. Results The CPHR analysis of Transition to severe disease found that active smoking [hazard ratio (HR) 4.01, 95% confidence interval (CI) 1.71–9.40, P = 0.001]; body mass index (BMI) points > 25 at baseline (each point: HR 1.06, 95% CI 1.02–1.09, P < 0.001); active disease in 2 (HR 4.26, 95% CI 1.23–14.84, P = 0.02) and ≥ 3 areas (HR 6.54, 95% CI 1.89–22.72, P = 0.003) all constituted substantial risk factors. Conversely, the CPHR analysis of Disease progression did not yield results of clinical relevance. A 'progression score' of 3.04 was used as a threshold in the predictive algorithm of Transition to severe disease and achieved the following test specifics: sensitivity = 0.51, specificity = 0.86, positive predictive value = 0.50, negative predictive value = 0.86. Conclusions We found a disparity between factors increasing the risk of simple Disease progression and those increasing the risk of Transition to severe disease. For the latter, active smoking, BMI points > 25, active disease in 2 or ≥ 3 areas were all shown to be the clinically relevant factors that could be used to construct an algorithm that correctly predicted progression to severe HS in more than half of all instances. To describe demographics, clinical features, and treatment outcomes of patients with highly drug-resistant tuberculosis (TB) in Ukraine, and to evaluate risk factors for an unsuccessful outcome. Data from patients with multi-, pre-extensively, or extensively drug-resistant TB were collected prospectively from TB dispensaries in 15 out of 24 Ukrainian oblasts (regions) from 2020 to 2021. Treatment outcomes were evaluated using WHO definitions. Risk factors for an unsuccessful outcome were identified using a multivariable logistic regression model. Among 1748 patients, the overall proportion of successful outcomes was 58% (95% confidence interval [95% CI] 56–60) (n = 1015/1748), ranging from 65% (95% CI: 62–69) (n = 531/814) for multidrug-resistant TB to 54% (95% CI: 49–58) (n = 301/563) for pre-extensively drug-resistant TB and 49% (95% CI: 44–55) (n = 183/371) for extensively drug-resistant TB. Results were similar across oblasts, with few exceptions. The strongest risk factors for an unsuccessful outcome were extensively drug-resistant TB (adjusted OR [aOR] 3.23; 95% CI: 1.88–5.53), total serum protein below 62 g/L in adults and below 57 g/L for children and adolescents (aOR 2.79; 95% CI: 1.93–4.04), psychiatric illness (aOR 2.79; 95% CI: 1.46–5.33), age at TB diagnosis >65 years (aOR 2.50; 95% CI: 1.42–4.42), and alcohol misuse (aOR 2.48; 95% CI: 1.89–3.26). The overall proportion of successful outcomes among Ukrainians treated for highly drug-resistant TB was 58%, notably better compared with previous years, but still low for extensively drug-resistant TB. Risk factors for unsuccessful outcomes highlight that addressing socioeconomic factors in TB management is crucial. Rice production worldwide represents a major anthropogenic source of greenhouse gas emissions. Nitrogen fertilization and irrigation practices have been fundamental to achieve optimal rice yields, but these agricultural practices together with by-products from plants and microorganisms, facilitate the production, accumulation and venting of vast amounts of CO2, CH4 and N2O. We propose that the development of elite rice varieties should target root traits enabling an effective internal O2 diffusion, via enlarged aerenchyma channels. Moreover, gas tight barriers impeding radial O2 loss in basal parts of the roots will increase O2 diffusion to the root apex where molecular O2 diffuses into the rhizosphere. These developments result in plants with roots penetrating deeper into the flooded anoxic soils, producing higher volumes of oxic conditions in the interface between roots and rhizosphere. Molecular O2 in these zones promotes CH4 oxidation into CO2 by methanotrophs and nitrification (conversion of NH4+ into NO3-), reducing greenhouse gas production and at the same time improving plant nutrition. Moreover, roots with tight barriers to radial O2 loss will have restricted diffusional entry of CH4 produced in the anoxic parts of the rhizosphere and therefore plant-mediated diffusion will be reduced. The article discusses the function and induction of the root barrier to radial oxygen loss in plants. The outer apoplastic barriers in roots serve multiple functions, including restricting the entry of soil phytotoxins and retaining water or oxygen inside the root. These barriers can be induced by environmental signals and involve the deposition of biopolymers in the cell walls of the outer part of the root. The study focuses on the role of abscisic acid (ABA) in inducing suberization of the exodermis in barley plants, which restricts radial oxygen loss. The research highlights the importance of understanding the molecular regulation mechanisms of suberin biosynthesis and the potential for using ABA to accelerate cultivar development. Since 2010, the Environment Agency of England has had a range of flexible tools at its disposal to deal with misconduct. One such sanction, enforcement undertaking, is akin to a settlement where the regulator and the offender agree to deal with the breach through a range of promises that may restore and remedy the environmental harm caused by the offence. The majority of these undertakings further require the offender to make donations to charitable environmental causes to improve the environment. However, there is little transparency around the latter promise. Consequently, this article details the findings of a pilot study interrogating the specific implementation of a subset of enforcement undertakings in the North East of England. The article argues that a more transparent enforcement process would address some of the accountability problems arising from the findings, which show that 'community benefits' operate as an offset mechanism instead of as a restorative enforcement mechanism. • Healthcare workers are at risk of occupational tuberculosis (TB). • Whole genome sequencing (WGS) is a highly precise tool for genotyping. • WGS for TB contact investigations among healthcare workers is sparsely described. • We describe using WGS for ascertaining occupational TB among healthcare workers. • WGS reduces the efforts required for epidemiological TB contact investigations. In this case report, we present case reports for two nurses, both working in departments of respiratory medicine, who developed tuberculosis (TB). For each individual case, whole genome sequencing (WGS) revealed only one specific match within a genomic distance of <6 single-nucleotide polymorphisms. The subsequent epidemiological investigations confirmed that both nurses had relevant exposures to their corresponding match 1139 and 1704 days before presenting with TB symptoms, respectively. Twenty-two studies were identified that reported using genotyping to identify occupational transmission of Mycobacterium tuberculosis to healthcare workers. Only two studies applied WGS, both conducted in resource-rich countries, comparable to the present Danish investigation. When comparing the two WGS studies to the other studies that used older genotyping techniques, WGS provided a higher resolution and much more detailed information. Consequently, the epidemiological investigations were more straightforward. In conclusion, WGS is a powerful tool for determining whether M. tuberculosis transmission is occupational as demonstrated for the two cases in this study. [ABSTRACT FROM AUTHOR]

Background Although the vast majority of individuals succumbing to infection with severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) are elderly, infection fatality rate (IFR) estimates for the age group ≥70 years are still scarce. To this end, we assessed SARS-CoV-2 seroprevalence among retired blood donors and combined it with national coronavirus disease 2019 (COVID-19) survey data to provide reliable population-based IFR estimates for this age group. Methods We identified 60 926 retired blood donors aged ≥70 years in the rosters of 3 regionwide Danish blood banks and invited them to fill in a questionnaire on COVID-19–related symptoms and behaviors. Among 24 861 (40.8%) responders, we invited a random sample of 3200 individuals for blood testing. Overall, 1201 (37.5%) individuals were tested for SARS-CoV-2 antibodies (Wantai) and compared with 1110 active blood donors aged 17–69 years. Seroprevalence 95% confidence intervals (CIs) were adjusted for assay sensitivity and specificity. Results Among retired (aged ≥70 years) and active (aged 17–69 years) blood donors, adjusted seroprevalences were 1.4% (95% CI,.3–2.5%) and 2.5% (95% CI, 1.3–3.8%), respectively. Using available population data on COVID-19–related fatalities, IFRs for patients aged ≥70 years and for 17–69 years were estimated at 5.4% (95% CI, 2.7–6.4%) and.083% (95% CI,.054–.18%), respectively. Only 52.4% of SARS-CoV-2–seropositive retired blood donors reported having been sick since the start of the pandemic. Summary: Drought and flooding are contrasting abiotic stressors for plants. Evidence is accumulating for root anatomical traits being essential for the adaptation to drought or flooding. However, an integrated approach to comprehensively understand root anatomical traits has not yet been established.Here we analysed the root anatomical traits of 18 wild Poaceae species differing in adaptation to a range of soil water content. Regression model analyses revealed the optimal anatomical traits that were required by the plants to adapt to low or high soil water content.While the area and number of each root tissue (e.g. stele, cortex, xylem or aerenchyma) were not strongly correlated to the soil water content, the ratio of the root tissue areas (cortex to stele ratio (CSR), xylem to stele ratio (XSR) and aerenchyma to cortex ratio (ACR)) could fully explain the adaptations of the wild Poaceae species to the soil water gradients.Our results demonstrate that the optimal anatomical traits for the adaptations to soil water content can be determined by three indices (i.e. CSR, XSR and ACR), and thus we propose that these root anatomical indices can be used to improve the tolerance of crops to drought and flooding stresses. Summary: Plants typically respond to waterlogging by producing new adventitious roots with aerenchyma and many wetland plants form a root barrier to radial O2 loss (ROL), but it was not known if this was also the case for lateral roots.We tested the hypothesis that lateral roots arising from adventitious roots can form a ROL barrier, using root‐sleeving electrodes and O2 microsensors to assess ROL of Zea nicaraguensis, the maize (Zea mays ssp. mays) introgression line with a locus for ROL barrier formation (introgression line (IL) #468) from Z. nicaraguensis and a maize inbred line (Mi29).Lateral roots of Z. nicaraguensis and IL #468 both formed a ROL barrier under stagnant, deoxygenated conditions, whereas Mi29 did not. Lateral roots of Z. nicaraguensis had higher tissue O2 status than for IL #468 and Mi29. The ROL barrier was visible as suberin in the root hypodermis/exodermis. Modelling showed that laterals roots can grow to a maximum length of 74 mm with a ROL barrier, but only to 33 mm without a barrier.Presence of a ROL barrier in lateral roots requires reconsideration of the role of these roots as sites of O2 loss, which for some species now appears to be less than hitherto thought. Summary: Flooding causes oxygen deprivation in soils. Plants adapt to low soil oxygen availability by changes in root morphology, anatomy, and architecture to maintain root system functioning. Essential traits include aerenchyma formation, a barrier to radial oxygen loss, and outgrowth of adventitious roots into the soil or the floodwater. We highlight recent findings of mechanisms of constitutive aerenchyma formation and of changes in root architecture. Moreover, we use modelling of internal aeration to demonstrate the beneficial effect of increasing cortex‐to‐stele ratio on sustaining root growth in waterlogged soils. We know the genes for some of the beneficial traits, and the next step is to manipulate these genes in breeding in order to enhance the flood tolerance of our crops. Climate change litigators are increasingly relying on a range of different jurisdictional avenues and legal regimes. The recent Urgenda decision by the Dutch Supreme Court provides a surprisingly rare snapshot of the relevance of human rights law to climate change litigation. Focusing on the Supreme Court's reliance on the environmental rights case law from the ECHR, this case note argues that climate change and human rights adjudications takes the form of an adjudicatory network. This Expert View provides an update on the recent development of new microsensors, and briefly summarizes some novel applications of existing microsensors, in plant biology research. Two major topics are covered: (i) sensors for gaseous analytes (O2, CO2, and H2S); and (ii) those for measuring concentrations and fluxes of ions (macro- and micronutrients and environmental pollutants such as heavy metals). We show that application of such microsensors may significantly advance understanding of mechanisms of plant–environmental interaction and regulation of plant developmental and adaptive responses under adverse environmental conditions via non-destructive visualization of key analytes with high spatial and/or temporal resolution. Examples included cover a broad range of environmental situations including hypoxia, salinity, and heavy metal toxicity. We highlight the power of combining microsensor technology with other advanced biophysical (patch–clamp, voltage–clamp, and single-cell pressure probe), imaging (MRI and fluorescent dyes), and genetic techniques and approaches. We conclude that future progress in the field may be achieved by applying existing microsensors for important signalling molecules such as NO and H2O2, by improving selectivity of existing microsensors for some key analytes (e.g. Na, Mg, and Zn), and by developing new microsensors for P. The presence of naturally occurring cytokine-specific autoantibodies (c-aAb) in humans is well established, as well as associations to selected pathologies. However, the overall influence of c-aAb on immunocompetence remains largely unknown. In this paper, we performed a large-scale investigation of c-aAb association with infection risk. A cohort of healthy Danish blood donors was screened for c-aAb against IL-1α, IL-6, IL-10, IFNα, and GM-CSF using a Luminex-based multiplex assay, and results were linked to data from the Danish National Prescription Registry. The filing of an antimicrobial prescription following c-aAb measurement was used as a proxy for impaired immunocompetence. We found that c-aAb against pro-inflammatory cytokines IFNα and GM-CSF tended to associate with increased risk of prescription filings in women, whereas antibodies against anti-inflammatory IL-10 were associated with a lower predicted risk of antimicrobial prescriptions, as well as higher self-perceived health scores. We also observed an association of cumulative c-aAb presence with prescription risk. Our data show that cytokine autoantibodies in healthy individuals associate with various proxies for immunomodulation, with the exact association dependent on the pattern of pro- or anti-inflammatory cytokines targeted. This suggests that c-aAb may express cytokine-modulatory properties in healthy individuals and may be critical to further investigate as biomarkers of immunodeficiency. [ABSTRACT FROM AUTHOR]

In recent years, research on flooding stress and hypoxic responses in plants has gathered increasing attention due to climate change and the important role of O2 in metabolism and signalling. This Collection of Functional Plant Biology on ‘Flooding stress and responses to hypoxia in plants’ presents key contributions aimed at progressing our current understanding on how plants respond to low-O2 conditions, flooding stress and a combination of stresses commonly found in flooded areas. The Collection emphasises the characterisation of diverse plant responses across different developmental stages, from seed germination to fully developed plants, and under different water stress conditions ranging from waterlogging to complete submergence, or simply low-O2 conditions resulting from limited O2 diffusivity in bulky tissues. Additionally, this Collection highlights diverse approaches, including eco-physiological characterisation of plant responses, detailed descriptions of root anatomical characteristics and their surrounding microenvironments, evaluation of the seed microbiota under flooding stress, the modification of gene expression, and evaluations of diverse germplasm collections. With recent progress in active research on flooding and hypoxia/anoxia tolerance in native and agricultural crop plants, vast knowledge has been gained on both individual tolerance mechanisms and the general mechanisms of flooding tolerance in plants. Research on carbohydrate consumption, ethanolic and lactic acid fermentation, and their regulation under stress conditions has been accompanied by investigations on aerenchyma development and the emergence of the radial oxygen loss barrier in some plant species under flooded conditions. The discovery of the oxygen-sensing mechanism in plants and unravelling the intricacies of this mechanism have boosted this very international research effort. Recent studies have highlighted the importance of oxygen availability as a signalling component during plant development. The latest developments in determining actual oxygen concentrations using minute probes and molecular sensors in tissues and even within cells have provided new insights into the intracellular effects of flooding. The information amassed during recent years has been used in the breeding of new flood-tolerant crop cultivars. With the wealth of metabolic, anatomical, and genetic information, novel holistic approaches can be used to enhance crop species and their productivity under increasing stress conditions due to climate change and the subsequent changes in the environment. Background: Due to physiological demands, children and premenopausal women are at risk of developing iron deficiency. In premenopausal women, the risk may be further increased by repeated whole blood donations. Short-term consequences of iron deficiency in infancy include impaired cognitive development and lower IQ scores. This prompts concern that maternal iron deficiency before or during pregnancy may have long-term consequences for the offspring, for example, by affecting scholastic attainment. The aim of this study was to evaluate if prepregnancy donation intensity is associated with offspring scholastic attainment measured as grade averages in standardized national written examinations in Denmark.Study Design and Methods: By using the Danish personal identification number as key, we obtained information on donation intensity before pregnancy, school grade, year of graduation, age of the students, students' sex, and parental length of education and income from various nationwide registers. Linear regression analyses were performed, with grade average as outcome and maternal donation status as explanatory variable (nondonor, n = 177,078; low-frequency donor, n = 4995 [one to five donations in the 3 years before pregnancy]; high-frequency donor, n = 414 [six or more donations in the 3 years before pregnancy), and further adjusted for the covariates listed above.Results: Adjusted normalized (mean, 0; standard deviation [SD], 1) test scores were statistically significantly higher for children of active female donors compared with children of nondonors (SD, 0.104; 95% confidence interval, 0.079-0.129). We observed no differences in scholastic attainment between children of low-frequency donors and high-frequency donors.Conclusion: Prepregnancy donation intensity, as a proxy of iron stores, is not associated with subsequent offspring scholastic attainment. Background: The prevalence of iron depletion is high among premenopausal women who donate blood frequently. Studies in nondonor populations indicate that iron deficiency anemia is associated with an increased risk of low birth weight. This prompts concerns that iron deficiency induced by frequent blood donation might impair subsequent fetal development.Study Design and Methods: The aim of this study was to assess whether prepregnancy donation intensity affects the birth weight of singletons born at term (gestational week 38 or later) to nulliparous female donors in Denmark. We identified 293,897 first live singleton births to Danish women between 1997 and 2012 with complete information on gestational age, birth weight, child sex, parental age, maternal smoking status during pregnancy, and parental education length and annual income. Linear regression analysis was applied, with birth weight as outcome, number of donations within the 3 years before pregnancy as the explanatory variable, and confounding variables as described.Results: Birth weight among children of low-intensity donors (n = 22,120) was 12.6 g (95% confidence interval, 6.7-18.6) higher than nondonors (n = 268,253) after controlling for the above-mentioned factors. The higher birth weight among low-intensity donors can be explained by the healthy donor effect. In fully adjusted analyses, birth weight among children of high-intensity donors (n = 3,524) was 20.2 g (95% confidence interval, 5.1-35.3 g) lower compared with low-intensity donors. This reduced birth weight among high-intensity donors compared to low-intensity donors may reflect blood donation-induced iron deficiency.Conclusions: Our results show that high prepregnancy donation intensity is inversely associated with birth weight of singletons born at term to nulliparous women. Using an empirical assessment of the use of enforcement undertakings by the Environment Agency and the engagement of the courts with the recently enacted sentencing guidelines for environmental offences, this article argues that the enforcement of environmental law is undergoing significant change. This change manifests itself in an increased reliance on written negotiated agreements in the form of enforcement undertakings by the Enivironment Agency and the willingness of the courts to hand down significant fines in cases against certain types of polluters. These new dynamics suggest that negotiation continues to play an important role in the enforcement of environmental law, albeit in a contractualized form. The application of the sentencing guidelines conversely suggests that environmental offences are no longer trivialized by the courts. Taken together, these emerging dynamics not only create specific incentives between agencies and offenders but also call into question established understandings and perceptions of regulatory enforcement. This article utilises the concept of policy framing to examine regulatory choices and the contestation of such choices in the context of hydraulic fracturing In the UK. The article makes no argument about the merits of fracking in the UK context, but rather explores how the institutional choices and design serve to shape the legal reasoning, thereby defining the ability of actors outside the London government to engage with its efforts to govern. Through the lens of recent judicial decisions in which claimants have sought to challenge privileged policy frames supportive of hydraulic fracturing, the article finds that the deferential approach taken by the courts serves to reinforce privileged frames, resulting in there being little scope to challenge such frames. Background and Aims Leaf tissue CO2 partial pressure (p CO2) shows contrasting dynamics over a diurnal cycle in C3 and Crassulacean Acid Metabolism (CAM) plants. However, simultaneous and continuous monitoring of p CO2 and p O2 in C3 and CAM plants under the same conditions was lacking. Our aim was to use a new CO2 microsensor and an existing O2 microsensor for non-destructive measurements of leaf p CO2 and p O2 dynamics to compare a C3 and a CAM plant in an aquatic environment. Methods A new amperometric CO2 microsensor and an O2 microsensor elucidated with high temporal resolution the dynamics in leaf p CO2 and p O2 during light–dark cycles for C3 Lobelia dortmanna and CAM Littorella uniflora aquatic plants. Underwater photosynthesis, dark respiration, tissue malate concentrations and sediment CO2 and O2 were also measured. Key Results During the dark period, for the C3 plant, p CO2 increased to approx. 3.5 kPa, whereas for the CAM plant CO2 was mostly below 0.05 kPa owing to CO2 sequestration into malate. Upon darkness, the CAM plant had an initial peak in p CO2 (approx. 0.16 kPa) which then declined to a quasi-steady state for several hours and then p CO2 increased towards the end of the dark period. The C3 plant became severely hypoxic late in the dark period, whereas the CAM plant with greater cuticle permeability did not. Upon illumination, leaf p CO2 declined and p O2 increased, although aspects of these dynamics also differed between the two plants. Conclusions The continuous measurements of p CO2 and p O2 highlighted the contrasting tissue gas compositions in submerged C3 and CAM plants. The CAM leaf p CO2 dynamics indicate an initial lag in CO2 sequestration to malate, which after several hours of malate synthesis then slows. Like the use of O2 microsensors to resolve questions related to plant aeration, deployment of the new CO2 microsensor will benefit plant ecophysiology research. [ABSTRACT FROM AUTHOR]

• Background and Aims Floating sweet-grass (Glyceria fluitans) can form aerial as well as floating leaves, and these both possess superhydrophobic cuticles, so that gas films are retained when submerged. However, only the adaxial side of the floating leaves is superhydrophobic, so the abaxial side is directly in contact with the water. The aim of this study was to assess the effect of these different gas films on underwater net photosynthesis (PN) and dark respiration (RD). • Methods Evolution of O2 was used to measure underwater PN in relation to dissolved CO2 on leaf segments with or without gas films, and O2 microelectrodes were used to assess cuticle resistance of floating leaves to O2 uptake in the dark. • Key Results The adaxial side of aerial leaves was more hydrophobic than the abaxial side and also initially retained a thicker gas film when submerged. Underwater PN vs. dissolved CO2 of aerial leaf segments with gas films had a Km of 172 mmol CO2 m-3 and a Pmax of 7.1 μmol O2 m-2 s-1, and the leaf gas films reduced the apparent resistance to CO2 uptake 12-fold. Underwater PN of floating leaves measured at 700 mmol CO2 m-3 was 1.5-fold higher than PN of aerial leaves. The floating leaves had significantly lower cuticle resistance to dark O2 uptake on the wettable abaxial side compared with the superhydrophobic adaxial side. • Conclusions Glyceria fluitans showed high rates of underwater PN and these were obtained at environmentally relevant CO2 concentrations. It appears that the floating leaves possess both aquatic and terrestrial properties and thus have 'the best of both worlds' so that floating leaves are particularly adapted to situations where the plant is partially submerged and occasionally experiences complete submergence. The world is currently experiencing dramatic increases in flood events impacting on natural vegetation and crops. Flooding often results in low O2 status in root tissues during waterlogging, but sometimes also in shoot tissues when plants become completely submerged. Plants possess a suite of traits enabling tissue aeration and/or adjusted metabolism during hypoxia or even in the absence of O2. This special issue of Functional Plant Biology presents key papers for plant scientists on the quest to further address and improve flood tolerance of terrestrial plants. The papers address low O2 responses in roots, shoots or whole plants in controlled laboratory conditions or in the field situation using natural wetland plants as models as well as economically important crops, such as rice, wheat and barley. The studies advance our understanding of low O2 responses in plant tissues as caused by O2 shortage during flooding. However, in most instances, submergence not only leads to hypoxic or anoxic tissues, but inundation in water also results in accumulation of CO2 and the important plant hormone ethylene. Thus, carefully designed laboratory studies are often needed to unravel the mechanistic relationships between a combined decline in O2 followed by increases in CO2 and ethylene at tissue as well as on the cellular level. Lobelia dortmanna is an iconic keystone species of northern softwater lakes in Europe as well as North America. It has suffered a dramatic decline in distribution in recent decades and the root causes are not well‐known, although elements such as eutrophication, acidification and brownification have been suggested as underlying reasons for the decline.In order to quantify the causes of change in the occurrence of L. dortmanna in Danish lakes, we firstly mapped the historical distribution of the species and secondly collected the available recent information on the presence of L. dortmanna along with data on environmental variables and plant community composition from the lakes. We identified 168 original Lobelia lakes of which L. dortmanna had, by now, gone extinct in 48%. A subset of 80 lakes formed the backbone of this study owing to the availability of data related to environmental conditions and aquatic plant communities.Using piecewise structural equation models, we identified that only the mean height of the lake plant communities had a direct negative influence on the occurrence of L. dortmanna. The expected adverse effects of eutrophication and alkalinity were all indirect, and funnelled through the trophic affinity of the plant community, which in turn influenced the mean plant height. Direct effects of eutrophication via shading from phytoplankton, acidification or brownification were not observed.We show that eutrophication, particularly in more alkaline water, is likely to have caused the dramatic decline in occurrence of L. dortmanna, but our models indicated that the effect of eutrophication acted via interspecific competition from other larger rooted aquatic plants.Conservation efforts aimed at protecting small aquatic plant species should be prioritised in more alkaline lakes where such species are most at risk. Background: Low hemoglobin (Hb) is associated with poor general health and adverse outcomes in a wide range of diseases. However, a link between Hb levels and the risk of infection among healthy individuals has yet to be investigated.Study Design and Methods: Using data from the Scandinavian Donations and Transfusions database, 497,390 donors were followed after 5,458,499 donations in health registers. With 1,339,362 person-years of follow-up, Andersen-Gill Cox regression was used to study the association of Hb levels below deferral thresholds, very low Hb levels (in the lowest 0.1 percentile), and declining Hb levels with the risk of infection as measured by hospital or outpatient contact for infection and filling of prescription for antimicrobials, respectively, within 3 months of donation. Analyses were stratified by sex, menopausal status, and frequency of donation.Results: Hb levels below deferral thresholds were not associated with a risk of hospital contact for infection among premenopausal women (hazard ratio [HR], 1.04; 95% confidence interval [CI], 0.95-1.14), postmenopausal women (HR, 0.77; 95% CI, 0.54-1.11), or men (HR, 0.97; 95% CI, 0.81-1.16), nor was there any association with hospital contact for very low Hb levels or patterns of declining Hb levels. However, subthreshold Hb levels were associated with a reduced risk of antimicrobial prescriptions among premenopausal women (HR, 0.92; 95% CI, 0.91-0.93), postmenopausal women (HR, 0.93; 95% CI, 0.89-0.97), and men (HR, 0.91; 95% CI, 0.88-0.94).Conclusions: Neither Hb levels below deferral thresholds nor very low or declining Hb levels were associated with an increased risk of infection. Photosynthesis of most seagrass species seems to be limited by present concentrations of dissolved inorganic carbon (DIC). Therefore, the ongoing increase in atmospheric CO2 could enhance seagrass photosynthesis and internal O2 supply, and potentially change species competition through differential responses to increasing CO2 availability among species. We used short-term photosynthetic responses of nine seagrass species from the south-west of Australia to test species-specific responses to enhanced CO2 and changes in HCO3−. Net photosynthesis of all species except Zostera polychlamys were limited at pre-industrial compared to saturating CO2 levels at light saturation, suggesting that enhanced CO2 availability will enhance seagrass performance. Seven out of the nine species were efficient HCO3− users through acidification of diffusive boundary layers, production of extracellular carbonic anhydrase, or uptake and internal conversion of HCO3−. Species responded differently to near saturating CO2 implying that increasing atmospheric CO2 may change competition among seagrass species if co-occurring in mixed beds. Increasing CO2 availability also enhanced internal aeration in the one species assessed. We expect that future increases in atmospheric CO2 will have the strongest impact on seagrass recruits and sparsely vegetated beds, because densely vegetated seagrass beds are most often limited by light and not by inorganic carbon. Seagrasses grow submerged in aerated seawater but often in low O2 sediments. Elevated temperatures and low O2 are stress factors., Internal aeration was measured in two tropical seagrasses, Thalassia hemprichii and Enhalus acoroides, growing with extreme tides and diel temperature amplitudes. Temperature effects on net photosynthesis ( PN) and dark respiration ( RD) of leaves were evaluated., Daytime low tide was characterized by high pO2 (54 kPa), pH (8.8) and temperature (38°C) in shallow pools. As PN was maximum at 33°C (9.1 and 7.2 μmol O2 m−2 s−1 in T. hemprichii and E. acoroides, respectively), the high temperatures and reduced CO2 would have diminished PN, whereas RD increased (Q10 of 2.0-2.7) above that at 33°C (0.45 and 0.33 μmol O2 m−2 s−1, respectively). During night-time low tides, O2 declined resulting in shoot base anoxia in both species, but incoming water containing c. 20 kPa O2 relieved the anoxia. Shoots exposed to 40°C for 4 h showed recovery of PN and RD, whereas 45°C resulted in leaf damage., These seagrasses are 'living near the edge', tolerant of current diel O2 and temperature extremes, but if temperatures rise both species may be threatened in this habitat. Background and purpose The C–C chemokine receptor 5-Δ32 deletion (CCR5-Δ32) has been associated with lower levels of C-reactive protein (CRP), but the effect on cardiovascular diseases is uncertain. This study addresses the impact of CCR5-Δ32 on the risk of low-grade inflammation and hospitalization with cardiovascular diseases in a large cohort of blood donors. Methods Genotyping of 15,206 healthy participants from The Danish Blood Donor Study for CCR5-Δ32 was performed and combined with CRP measurements and questionnaire data. Cardiovascular disease diagnoses were identified by ICD-10 codes in the Danish National Patient Registry. Results CCR5-Δ32-carriers had a higher risk of hospitalization for cardiovascular diseases when compared with wild-type homozygotes (hazard ratio = 1.35, 95%-confidence interval: 1.00–1.87). CRP levels were unaffected by the CCR5-Δ32 deletion. Background: Voluntary blood donation is believed to be mostly motivated by altruism. Because studies have suggested that altruistic personality is determined by both genetic and environmental factors, we speculated that willingness to donate blood could also be governed by constitutional factors. This hypothesis was tested in a study among Danish twins.Study Design and Methods: The nationwide Danish Twin Register, which is virtually complete for all twins born since 1968, was linked with Danish portion of the Scandinavian Donation and Transfusion (SCANDAT) Database, which includes information on all active Danish blood donors from 2002 to 2012, to establish blood donor status for Danish twins, who at age 17 years became eligible for donation in 2002 or later. Casewise concordance in monozygotic (MZ) and dizygotic (DZ) twins were presented and heritability was estimated in Mx by variance component analysis in a liability threshold model.Results: A total of 274 MZ and 484 same-sex DZ twins age 17 to 27 years were identified as donors in SCANDAT. There was no difference between MZ and DZ twins with regard to age at first donation or number of donations. Casewise concordance rates were 0.61 (95% confidence interval [CI], 0.56-0.67) and 0.41 (95% CI, 0.36-0.47) in MZ and DZ twin pairs, respectively. Heritability analysis using the ACE model found that additive genetic and shared environmental effects accounted for 0.53 (95% CI, 0.33-0.73) and 0.28 (95% CI, 0.10-0.45) of the variance in the motivation to donate blood, respectively.Conclusion: Becoming a volunteer blood donor is determined by both genetic and environmental factors shared within families. Adequate tissue O2 supply is crucial for plant function. We aimed to identify the environmental conditions and plant characteristics that affect plant tissue O2 status. We extracted data and performed meta-analysis on >1500 published tissue O2 measurements from 112 species. Tissue O2 status ranged from anoxic conditions in roots to >53 kPa in submerged, photosynthesising shoots. Using information-theoretic model selection, we identified 'submergence', 'light', 'tissue type' as well as 'light × submergence' interaction as significant drivers of tissue O2 status. Median O2 status were especially low (<50% of atmospheric equilibrium) in belowground rhizomes, potato (Solanum tuberosum) tubers and root nodules. Mean shoot and root O2 were ~25% higher in light than in dark when shoots had atmospheric contact. However, light showed a significant interaction with submergence on plant O2, with a submergence-induced 44% increase in light, compared with a 42% decline in dark, relative to plants with atmospheric contact. During submergence, ambient water column O2 and shoot tissue O2 correlated stronger in darkness than in light conditions. Although use of miniaturised Clark-type O2 electrodes has enhanced understanding of plant O2 dynamics, application of non-invasive methods in plants is still lacking behind its widespread use in mammalian tissues. We collected all (>1500) published measurements of plant O2 levels ever recorded, some measured more than 100 years ago. We found that plant O2 levels depended on in which plant part (e.g. roots or leaves) O2 was measured, and that plant O2 levels become low when plants are flooded, or in darkness. In this study, we conducted a systematic review and meta-analysis to summarize and evaluate the global research potential of different circulating miRNAs as an early diagnostic biomarker for OC. A systematic literature search for relevant studies was conducted in June 2020 and followed up in November 2021. The search was conducted in English databases (PubMed, ScienceDirect). The primary search resulted in a total of 1887 articles, which were screened according to the prior established inclusion and exclusion criteria. We identified 44 relevant studies, of which 22 were eligible for the quantitative meta-analysis. Statistical analysis was performed using the Meta-package in Rstudio. Standardized mean differences (SMD) of relative levels between control subjects and OC patients were used to evaluate the differential expression. All studies were quality evaluated using a Newcastle–Ottawa Scale. Based on the meta-analysis, nine miRNAs were identified as dysregulated in OC patients compared to controls. Nine were upregulated in OC patients compared to controls (miR-21, -125, -141, -145, -205, -328, -200a, -200b, -200c). Furthermore, miR-26, -93, -106 and -200a were analyzed, but did not present an overall significant difference between OC patients and controls. These observations should be considered when performing future studies of circulating miRNAs in relation to OC: sufficient size of clinical cohorts, development of consensus guidelines for circulating miRNA measurements, and coverage of previously reported miRNAs. BACKGROUND Health-related quality of life (HRQL) represents people's subjective assessment of their mental and physical well-being. HRQL is highly predictive of future health. The effect of iron deficiency without anemia induced by blood donation on HRQL is presently unknown. The aim was to explore the relationship between iron status and self-reported mental component score (MCS; SF-12) and physical component score (PCS; SF-12) in Danish blood donors. STUDY DESIGN AND METHODS Complete relevant data, including the 12-item short-form health survey (SF-12), plasma ferritin levels, age, body mass index, smoking status, C-reactive protein levels, number of donations in the previous 3 years, and PCS and MCS, were available for 8692 men and 7683 women enrolled from March 1, 2010, to December 31, 2010. Multivariable linear and logistic (cutoff at the 10th percentile) regression analyses were used to assess the relationship between iron deficiency (ferritin < 15 ng/mL) and MCS and PCS, respectively. Analyses were performed separately for men and women. RESULTS There was no significant relationship between iron deficiency and self-reported mental or physical health. BACKGROUND It is well known that blood donors are at increased risk of iron deficiency and subsequent development of iron deficiency anemia. We aimed to investigate the effect of factors influencing hemoglobin (Hb) levels. STUDY DESIGN AND METHODS Initiated in 2010, the Danish Blood Donor Study is a population-based study and biobank. We performed multivariable linear regression analysis to assess the effects of donation activity, physiologic and lifestyle factors, and diet on Hb levels among 15,197 donors. We also performed multivariable logistic regression to evaluate the effects of these factors on the risk of having low Hb (defined as Hb below the 10th percentile among men and women, respectively) and of a decrease in Hb greater than 0.5 mmol/L (0.8 g/dL) between successive donations. All analyses were performed stratified for sex and smoking status. We also tested a previously used model for the prediction of Hb. RESULTS The strongest predictors of Hb and risk of low Hb were low ferritin (<15 ng/mL) and current use of iron supplementation (yes/no). No dietary factors were found to be consistently significant in multivariable models predicting Hb levels, risk of having low Hb, or risk of a decrease in Hb greater than 0.5 mmol/L. We found similar effects to previous studies of factors in the predictive model, with little additional effect of including smoking status and ferritin. CONCLUSIONS As ferritin was the strongest predictor of Hb, this study supports the implementation of regular ferritin measurement as a method of risk assessment among blood donors. Some terrestrial wetland plants, such as rice, have super-hydrophobic leaf surfaces which retain a gas film when submerged. O2 movement through the diffusive boundary layer ( DBL) of floodwater, gas film and stomata into leaf mesophyll was explored by means of a reaction-diffusion model that was solved in a three-dimensional leaf anatomy model. The anatomy and dark respiration of leaves of rice ( O ryza sativa L.) were measured and used to compute O2 fluxes and partial pressure of O2 ( pO2) in the DBL, gas film and leaf when submerged. The effects of floodwater pO2, DBL thickness, cuticle permeability, presence of gas film and stomatal opening were explored. Under O2-limiting conditions of the bulk water ( pO2 < 10 kPa), the gas film significantly increases the O2 flux into submerged leaves regardless of whether stomata are fully or partly open. With a gas film, tissue pO2 substantially increases, even for the slightest stomatal opening, but not when stomata are completely closed. The effect of gas films increases with decreasing cuticle permeability. O2 flux and tissue pO2 decrease with increasing DBL thickness. The root and shoot tissues of flood-tolerant wetland plants are highly porous to enable internal gas phase diffusion of O2 during waterlogging or submergence. In the case of only partial submergence (snorkelling), the atmosphere can act as source of O2. The aim of this study was to assess the effect of waterlogging, partial submergence and complete submergence in the dark as well as in light on O2 partial pressure ( pO2) in roots of R umex palustris (flood tolerant) and R . acetosa (flood intolerant). We used O2 microelectrodes to measure pO2 of adventitious roots during manipulations of the water level around the shoot. Root pO2 in both species declined significantly upon submergence but remained oxic also when shoots were completely submerged in the dark (0.8 and 4.6 kPa in R . acetosa and R . palustris, respectively). The snorkelling effect was substantial in R . palustris only. Submergence in light had less impact on root pO2 and the effect of snorkelling was also minor. Hence, the benefits of light (underwater photosynthesis) and air contact (snorkelling) upon growth and survival in submerged wetland plants can now be linked to enhanced internal aeration. Background: C-reactive protein (CRP) is a well-established marker of inflammation. The level of CRP is affected by several lifestyle factors. A slightly increased CRP level, also known as low-grade inflammation (LGI), is associated with increased risk of several diseases, especially cardiovascular disease. The aim of this study was to identify predictors of increased CRP levels in healthy individuals. We therefore assessed CRP in a large cohort of blood donors. Methods: We measured plasma CRP levels in 15,684 participants from the Danish Blood Donor Study. CRP was measured by a commercial assay. Furthermore, all participants completed a standard questionnaire on smoking status, alcohol consumption, physical activity, diet, and various body measurements. Female participants also reported the use of contraception, childbirth, and menopausal status. The relationship between LGI (defined here as a plasma CRP level between 3 mg/L and 10 mg/L) and predictors was explored by multivariable logistic regression analysis. Results were presented as odds ratios (OR) with 95% confidence intervals (CI). Results: We found LGI in a total of 1,561 (10.0%) participants. LGI was more frequent in women using combined oral contraception (OC) (29.9%) than in men (6.1%) and women not using OC (7.9%). Among premenopausal women, OC was the strongest predictor of LGI (odds ratio = 8.98, p<0.001). Additionally, body mass index (BMI) and waist circumference were positively associated with LGI. Conclusion: High BMI and abdominal obesity strongly predicted LGI among healthy individuals. However, the most striking finding was the high prevalence of LGI among premenopausal women who used combined oral contraception. Although the significance of CRP as a marker of inflammation is well known, the role of CRP in pathogenesis is still uncertain. The purpose of this study was to assess retrospectively complications and diagnostic yield of core needle biopsy ( CNB) of cervical lymph nodes using 14-gauge as sole needle bore. During a 10-year period ultrasound ( US)-guided CNB, using a 14-gauge Tru-Cut needle (non-advancing), was performed in 140 consecutive cases (135 patients, aged 8-88 years) when a detailed histological diagnosis was required to guide therapy. CNB findings were consistent with the final diagnosis in 129 of the 140 cases, comprising 36 benign lesions, 40 metastases, and 53 lymphomas (40 NHL and 13 HL) of which subclassification ( WHO criteria) was partial in 5 NHL, and complete in 35 (87.5%) NHL and 13 HL (100%), including all 7 lymphomatous nodes with short-axis diameter ≤1.0 cm. Two lymphomas were falsely interpreted as reactive hyperplasia. Nine samples (6.4%) were inadequate for histopathological analysis. One patient experienced pain in an arm lasting 3-4 days. No other immediate or delayed complications were diagnosed. A 14-gauge may be used safely as the sole needle bore in US-guided CNB of cervical lymph nodes, rendering samples sufficient for full subtyping in lymphoma, even in the smallest nodes. Abstract: Habitats occupied by many halophytes are not only saline, but are also prone to flooding and yet surprisingly few studies have evaluated submergence tolerance in halophytes. Sediment, floodwater, and intra-plant O2 dynamics were evaluated during tidal submergence for the leaf-succulent halophyte Suaeda maritima (L.) Dum. For S. maritima growing in soil just above the mud flat in a UK salt marsh, the soil was only moderately hypoxic just prior to tidal inundation, presumably owing to drainage and O2 entry facilitated by frequent, large cracks. O2 declined to very low levels following high tide. By contrast, mud flat sediment remained waterlogged, lacked cracks, and was anoxic. Plant O2 dynamics were investigated using field-collected plants in sediment blocks transported to a controlled-submergence system in a glasshouse. Submergence during night-time resulted in anoxia within leaves, whereas during day-time O2 was produced by underwater photosynthesis. The thin lateral roots of S. maritima presumably access some O2 from hypoxic sediments, but could also experience transient episodes of severe hypoxia/anoxia, especially as any internal O2 movement from shoots would be small owing to the low gas-filled porosity in roots. Fermentative metabolism to lactate, producing some ATP in the absence of O2, might contribute to tolerance of transient O2 deficits. Lactate was high in root tissues, whereas ethanol production (tissue and incubation medium contents) was low, both in comparison with values reported for other species. Our findings demonstrate the importance of tolerance to transient waterlogging and submergence for the halophyte S. maritima growing in a tidal salt marsh. [Copyright &y& Elsevier]

Aim: The present study aims to estimate the relative importance of genetic and environmental factors for health-related quality of life (HRQL) measured by the 12-item Short-Form Health Survey (SF-12). Methods: The study was based on two Danish twin cohorts (46,417 twin individuals) originating from the nationwide, population-based Danish Twin Registry. The twins were approached by a mailed-out questionnaire in 2002. The questionnaire included the SF-12, information on demographic factors, and questions on a variety of specific diseases. Heritability of the SF-12 includes the physical component summary (PCS) and the mental component summary (MCS); and etiologically important variance components were estimated using multivariate biometric models. The respondents were stratified into six groups, based on age and sex. Results: A total of 33,794 (73%) individual twins responded to the survey. The SF-12 was completed by 29,619 individuals, which included 9,120 complete twin pairs. Overall, the best-fitting model explaining the variance of HRQL was the ACE model. The estimated heritability of the SF-12 was between 11% and 35%, whereas between 65% and 89% could be explained by unique environmental or stochastic factors in the different sex and age groups. The highest heritability was seen among older twins. In addition, the genetic correlation between MCS and PCS scores was low (0.07 and 0.23 for males and females, respectively) among younger and high (0.26 and 0.45 for males and females, respectively) in the oldest age group. Both the largest genetic influence on HRQL and the largest genetic overlap between the scores were seen in the oldest age group, which consisted of twins older than 55. The unique environmental correlation between MCS and PCS were generally negative. Conclusion: The heritability of HRQL differs between different age groups. Enforcement undertakings have recently been added to the Environment Agency's list of mechanisms available for the enforcement of certain environmental offences. An enforcement undertaking is a written agreement between the regulator and the offender in which the offender proposes to undertake particular actions in relation to the relevant non-compliant activity. This article examines the use of enforcement undertakings by the Environment Agency, arguing that the undertakings have significant regulatory implications and give rise to a series of possible problems. Notwithstanding this, the article concludes that enforcement undertakings possess a number of potentially positive features, making them a useful alternative to existing enforcement mechanisms. In situ microelectrode studies of the wetland plant Meionectes brownii showed that O2 dynamics in the submerged stems and aquatic roots of partially submerged plants were similar to those of completely submerged plants, with internal O2 concentrations in both organs dropping to less than 5 kPa by dawn regardless of submergence level. The anatomy at the nodes and the relationship between tissue porosity and rates of O2 diffusion through stems were studied. Stem internodes contained aerenchyma and had mean gas space area of 17.7% per cross section, whereas nodes had 8.2%, but nodal porosity was highly variable, some nodes had very low porosity or were completely occluded (ca. 23% of nodes sampled). The cumulative effect of these low porosity nodes would have impeded internal O2 movement down stems. Therefore, regardless of the presence of an aerial connection, the deeper portions of submerged organs sourced most of their O2 via inwards diffusion from the water column during the night, and endogenous production in underwater photosynthesis during the daytime. Insects, spiders and plants risk drowning in their wetland habitats. The slow diffusion of O2 can cause asphyxiation when underwater, as O2 supply cannot meet respiratory demands. Some animals and plants have found a common solution to the major challenge: how to breathe underwater with respiratory systems evolved for use in air? Hydrophobic surfaces on their bodies possess gas films that act as a 'physical gill' to collect O2 when underwater and thus sustain respiration. In aquatic Insects, this feature/process has been termed 'plastron respiration'. Here, we demonstrate the similarities in function between underwater respiration of insect (Aphelocheirus aestivalis) plastrons and gas films on leaves of wetland plants (Phalaris arundinacea) and also show the importance of these physical gills by the resulting changes upon their removal. The gas films provide an enlarged gas-water interface to enhance O2 uptake underwater that is above that if only spiracles (insects) or stomata (plants) provided the gas-phase contact with the water. Background There is a 5- to 8-fold increased risk of mortality during the first 3 months after a hip fracture. Several risk factors are known. We studied the predictive value (for mortality) of routine blood tests taken on admission. Methods 792 hip fracture patients were included prospectively; blood tests were taken on admission. Follow-up data on mortality were obtained from the civil registration system. Patients were divided into 2 groups based on whether they had survived at least 90 days after the hip fracture. To estimate which laboratory tests could be used to predict outcome, we used receiver operation characteristic (ROC) curves. Results 3-month mortality could be predicted with 69% accuracy from the level of plasma creatinine in standard admission blood tests. The mortality in patients with elevated levels of creatinine was almost 3-fold that of the patients with normal creatinine. Mortality was also associated with age, low blood hemoglobin, high plasma potassium, and low plasma albumin levels.
