Abstract Neurokinin 3 receptor (NK3R) is a tachykinin receptor essential for the hypothalamic-pituitary-gonadal axis. The endogenous peptide agonist neurokinin B (NKB) preferentially activates NK3R, while substance P (SP) binds preferentially to NK1R. In addition, the SP analogue senktide more potently activates NK3R than NKB and SP. However, the mechanisms of preferential binding of peptide and NK3R activation remain elusive. Herein, we determined the cryogenic electron microscopy (cryo-EM) structures of the NK3R–Gq complex bound to NKB, SP and senktide. The three NK3R–Gq/peptide complexes utilize a class of noncanonical receptor activation mechanisms. Combining the structural analysis and functional assay illustrated that the consensus C-termini of the three peptide agonists share a conserved binding mode to NK3R, while the divergent N-termini of the peptides confer the preferential binding of the agonist to NK3R. In addition, the specific interactions between the N-terminus of senktide and the N-terminus and extracellular loops (ECL2 and ECL3) of NK3R lead to the improved activation displayed by senktide compared to SP and NKB. These findings pave the way to understand tachykinin receptor subtype selectivity and provide ideas to rationally develop drugs targeting NK3R.

Profenofos is a detectable insecticide in the environment with strong toxicity to non-targeted organisms. Photodegradation is a main transformation of profenofos in the environment. Myricetin is a flavonoid that strongly scavenges free radicals. The effect of myricetin on the photodegradation of profenofos was studied. The half-lives (T1/2) of profenofos were 1.7–7.0 and 90 h under artificial light and sunlight. The photolysis rate of profenofos decreased by 1.87–4.72 and 7.62 times with the addition of 20 ratios of myricetin. Free radicals reacting with profenofos were •OH and 1O2, and the key free radical was •OH. Myricetin strongly scavenged •OH and 1O2 which rapidly reacted with profenofos. O-(2-Chlorophenyl)-O-ethyl-S-propyl phosphorothioate (M3) and O-(2-chlorophenyl)-O-ethyl phosphorothioate (M4) were major and new photoproducts of profenofos. According to the Ecological Structure Activity Relationships, photodegradation of profenofos was a detoxification process, but myricetin inhibited the photodegradation of profenofos and its photoproducts. These results highlight the implication of myricetin on the fate and potential risk of profenofos in the environment.

Abstract The endogenous cyclic tetradecapeptide SST14 was reported to stimulate all five somatostatin receptors (SSTR1–5) for hormone release, neurotransmission, cell growth arrest and cancer suppression. Two SST14-derived short cyclic SST analogues (lanreotide or octreotide) with improved stability and longer lifetime were developed as drugs to preferentially activate SSTR2 and treat acromegalia and neuroendocrine tumors. Here, cryo-EM structures of the human SSTR2–Gi complex bound with SST14, octreotide or lanreotide were determined at resolutions of 2.85 Å, 2.97 Å, and 2.87 Å, respectively. Structural and functional analysis revealed that interactions between β-turn residues in SST analogues and transmembrane SSTR2 residues in the ligand-binding pocket are crucial for receptor binding and functional stimulation of the two SST14-derived cyclic octapeptides. Additionally, Q1022.63, N2766.55, and F2947.35 could be responsible for the selectivity of lanreotide or octreotide for SSTR2 over SSTR1 or SSTR4. These results provide valuable insights into further rational development of SST analogue drugs targeting SSTR2.

Abstract Trajectory prediction is a fundamental and challenging task for numerous applications, such as autonomous driving and intelligent robots. Current works typically treat pedestrian trajectories as a series of 2D point coordinates. However, in real scenarios, the trajectory often exhibits randomness, and has its own probability distribution. Inspired by this observation and other movement characteristics of pedestrians, we propose a simple and intuitive movement description called a trajectory distribution, which maps the coordinates of the pedestrian trajectory to a 2D Gaussian distribution in space. Based on this novel description, we develop a new trajectory prediction method, which we call the social probability method. The method combines trajectory distributions and powerful convolutional recurrent neural networks. Both the input and output of our method are trajectory distributions, which provide the recurrent neural network with sufficient spatial and random information about moving pedestrians. Furthermore, the social probability method extracts spatio-temporal features directly from the new movement description to generate robust and accurate predictions. Experiments on public benchmark datasets show the effectiveness of the proposed method.

Abstract Rabies virus (RV) is the most widely used vector for mapping neural circuits. Previous studies have shown that the RV glycoprotein can be a target to improve the retrograde transsynaptic tracing efficiency. However, the current versions still label only a small portion of all presynaptic neurons. Here, we reshuffled the oG sequence, a chimeric glycoprotein, with positive codon pair bias score (CPBS) based on bioinformatic analysis of mouse codon pair bias, generating ooG, a further optimized glycoprotein. Our experimental data reveal that the ooG has a higher expression level than the oG in vivo, which significantly increases the tracing efficiency by up to 12.6 and 62.1-fold compared to oG and B19G, respectively. The new tool can be used for labeling neural circuits Therefore, the approach reported here provides a convenient, efficient and universal strategy to improve protein expression for various application scenarios such as trans-synaptic tracing efficiency, cell engineering, and vaccine and oncolytic virus designs.

5-Substituted benzylidene 3-acylthiotetronic acids are antifungal. A series of 3-acylthiotetronic acid derivatives with varying substitutions at the 5-position were designed, synthesized, and characterized, based on the binding pose of 3-acyl thiolactone with the protein C171Q KasA. Fungicidal activities of these compounds were screened against Valsa Mali, Curvularia lunata, Fusarium graminearum, and Fusarium oxysporum f. sp. lycopersici. Most target compounds exhibited excellent fungicidal activities against target fungi at the concentration of 50 μg·mL−1. Compounds 11c and 11i displayed the highest activity with a broad spectrum. The median effective concentration (EC50) values of 11c and 11i were 1.9–10.7 and 3.1–7.8 μg·mL−1, respectively, against the tested fungi, while the EC50 values of the fungicides azoxystrobin, carbendazim, and fluopyram were respectively 0.30, 4.22, and > 50 μg·mL−1 against V. Mali; 6.7, 41.7, and 0.18 μg·mL−1 against C. lunata; 22.4, 0.42, and 0.43 μg·mL−1 against F. graminearum; and 4.3, 0.12, and > 50 μg·mL−1 against F. oxysporum f. sp. lycopersici. The structures and activities of the target compounds against C. lunata were analyzed to obtain a statistically significant comparative molecular field analysis (CoMFA) model with high prediction abilities (q2 = 0.9816, r2 = 0.8060), and its reliability was verified. The different substituents on the benzylidene at the 5-position had significant effects on the activity, while the introduction of a halogen atom at the benzene ring of benzylidene was able to improve the activity against the tested fungi.

Emotion categories configure the basic semantic knowledge of the human cognitive structure. Previous studies with people with Williams syndrome (WS) investigated their ability to process basic emotions and the dimensions of emotional valences. However, little is known about the categorization of emotions from the subordinate perspective of lexical words in people with WS. In this study, emotion priming was used as the research paradigm. Three types of emotional valence were used as stimuli: positive, neutral, and negative. Each emotional valence was used as a prime matched to a target in one of these same three types of emotional valence. All participants were asked to judge whether the prime and the target were matched in their emotional valence. People with WS (n = 14, 11M/3F, CA = 10.49, and MA = 6.57) showed priming patterns for emotion valences like those of the typically developing controls. When positive primes were presented, accuracy was higher for positive and negative targets than neutral targets. When neutral primes were presented, accuracy was highest for negative targets. When negative primes were presented, accuracy was the lowest for negative targets. All participants showed high priming accuracy for positive emotions; however, they confused neutral with positive targets. A negative priming effect was observed when negative primes preceded negative targets. Considering previous findings that people with WS show developmental delays in the basic emotions of anger and surprise, this study concludes that people with WS responded least accurately to the classification of emotional valence. The findings regarding the categorization of emotions in people with WS not only advance our understanding of their emotion knowledge and socioemotional cognition but also confirm the superficial enrichment of lexical semantics with weak conceptual change in people with WS. This weakness may result in impaired contextual integration in people with WS.

Neolamarckia macrophylla (Rubiaceae) is a fast-growing tree with high economic value that is endemic to Sulawesi and Moluccas, Indonesia. Here, we presented the complete chloroplast (cp) genome sequence of N. macrophylla. The complete chloroplast genome is 155,406 bp in size and includes a large single-copy region (86,013 bp), a small single-copy region (18,063 bp), and a pair of IR regions (25,665 bp each). A total of 128 genes were predicted, including 8 rRNA, 36 tRNA, and 84 protein-coding genes. Phylogenetic analysis based on 11 plant species chloroplast genomes indicated that the genus Neolamarckia was monophyletic which clustered as a sister group to Antirhea chinensis. These complete chloroplast genomes can be subsequently used for valuable species research of Rubiaceae.

Mapping the neural circuits facilitates understanding the brain’s working mechanism. Pseudorabies virus (PRV; Bartha stain) as a tracer can infect neurons and retrogradely transport in neural circuits. To illuminate the network, tracers expressing reporter genes at a high level are needed. In this study, we optimized the expression level of reporter genes and constructed two new retrograde trans-multisynaptic tracers PRV531 and PRV724, which separately express more robust green and red fluorescent proteins than the existing retrograde tracers PRV152 and PRV614. PRV531 and PRV724 can be used for mapping the neural circuit of the central nervous system (CNS) and the peripheral nervous system (PNS). Overall, our work adds two valuable tracers to the toolbox for mapping neural circuits.

Background /Aims: The underlying mechanisms leading to focal segmental glomerulosclerosis (FSGS) are lacking. In this report, we examined the role of protease-activated receptors (PARs) subtype PAR2 and its downstream signals in regulating the pathophysiological process of FSGS. Methods: Nephropathy was induced by intravenous injections of adriamycin (ADR) in rats to study FSGS. Western Blot analysis and ELISA were employed to determine the protein expression levels of PAR2 and its downstream signal pathways as well as the levels of PICs. Results: In ADR rats, expression of PAR2, PKCε and PKA was amplified and this was accompanied with increases of pro-inflammatory cytokines (PICs) including IL-1β, IL-6 and TNF-α. Inhibition of PAR2 signal by systemic administration of FSLLRY-NH2 (FSL) attenuated amplification of PICs. Notably, FSL further influenced key molecular mediators during development of FSGS. i.e., it specifically restored the impaired nephrin and attenuated the exaggerated transforming growth factor beta 1 (TGF-β1), caspase-9 and desmin thereby improving worsened renal functions and glomerular injury. Consistent with this, in cultured podocytes FSL also largely restored downregulation of nephrin and attenuated amplifications of caspase-9 and desmin induced by TGF-β1. Conclusions: Results of this study suggest that PAR2 plays an important role in mediating renal injury induced by glomerulosclerosis. Inhibition of PAR2 signal pathway has a protective effect on FSGS mainly via PIC and TGF-β1 mechanisms. Targeting one or more of these signaling molecules may present new opportunities for treatment and management of FSGS observed in patients.

Superfine down powder was modified by formamide which acts as a plasticizer and then hot-pressed into thermoplastic down films. The influences of formamide content, hot-pressing time, pressure intensity and exposure to atmosphere on the mechanical properties of thermoplastic down films were investigated. The maximum tensile strength of thermoplastic down films can reach 28.74 Mpa after exposure to atmosphere for 40 days.

A sensitive biotinylated indirect competitive enzyme-linked immunosorbent assay (Bic-ELISA) was developed to detect acetamiprid pesticides in pollen, based on the heterogeneous coating antigen and biotinylated anti-acetamiprid monoclonal antibody. Under optimized experimental conditions, the detection limit for the Bic-ELISA was 0.17 ng/mL and the linear range was 0.25–25 ng/mL. The cross-reactivities could be regarded as negligible for the biotinylated antibodies with their analogues except for thiacloprid (1.66%). Analyte recoveries for extracts of spiked pollen (camellia pollen, lotus pollen, rape pollen) ranged from 81.1% to 108.0%, with intra-day relative standard deviations (RSDs) of 4.8% to 10.9%, and the average reproducibility was 85.4% to 110.9% with inter-assay and inter-assay RSDs of 6.1% to 11.7%. The results of Bic-ELISA methods for the Taobao’s website samples were largely consistent with HPLC-MS/MS. Therefore, the established Bic-ELISA methods would be conducive to the monitoring of acetamiprid in pollen.
